Abstract

Den siste tiden har inhibitorer av reopptakstransportører for nevrotransmittere blitt viet stor oppmerksomhet. Det er gjort mange forsøk på å finne nye forbindelser, ettersom denne typen inhibitorer kan ha interessante farmakologiske egenskaper. Bicifadine har antagonistisk effekt på både serotonin- og norepinefrin-reopptakstransportørene og ble benyttet som ”lead compound” i utviklingen av en serie analoger i dette prosjektet. En fire-trinns synstese ble utviklet, og åtte analoger ble fremstilt. Utbyttene spenner fra moderat til godt, og det enantiomere overskudd var stort for alle analogene.

Abstract

The structures of the title dipeptides, C9H18N2O4·0.33H2O, C12H16N2O4 and C8H16N2O4S·0.34H2O, complete a series of investigations focused onl-Xaa-l-serine peptides, where Xaa is a hydro­phobic residue. All three structures are divided into hydro­philic and hydro­phobic layers. The hydro­philic layers are thin forl-phenyl­alanyl-l-serine, rendered possible by an unusual peptide conformation, and thick forl-isoleucyl-l-serine andl-methionyl-l-serine, which include cocrystallized water mol­ecules on the twofold axes. 12. New PhenylglycineDerived Primary Amine Organocatalysts for the Preparation of Optically Active Warfarin

Abstract

In this work we present new, fully synthetic phenylglycinederived primary amine organocatalysts useful for the onestep preparation of optically active warfarin, an important anticoagulant. Both enantiomeric forms of the catalysts are equally available and can be prepared by robust procedures without recourse to chromatographic purification. Together with a cocatalyst, particularly acetic acid or 2,4dinitrophenol, they can furnish warfarin in approximately 80  eeand represent inexpensive alternatives to other primary amine organocatalysts such as the chiral diamines and Cinchonaderived primary amines.© WileyVCH Verlag GmbH & Co. Abstract

The absolute structure has been determined by X-ray analysis for the title compound, C(11)H(8)Cl(2)O(2). The five-membered ring of the mol-ecule is best described as a flattened envelope conformation with the methyl-ene C atom located 0.208 (2) Å below the plane formed by the other four atoms. A weak intermolecular C-H⋯O hydrogen bond is present in the crystal structure.

The absolute structure of the title compound, C(11)H(13)FN(+)·Cl(-), has been determined. The five-membered ring has an envelope conformation with the N atom at the flap position. In the crystal structure, the Cl(-) anion links with the organic cation via N-H⋯Cl hydrogen bonding.
