The invasive tetraploid Cirsium vulgare hybridizes with both Cirsium and Lophiolepis. Its conflicted position in molecular phylogenies, and its peculiar combination of morphological, anatomical, and genomic features that are alternatively shared with representatives of Cirsium or Lophiolepis, strongly suggest its intergeneric hybrid origin.Genetic relationships of C. vulgare (8 samples) with genus Lophiolepis (11 species) and other representatives of genus Cirsium (12 species) were evaluated using restriction site‐associated DNA sequencing (RADseq) and examined using analytical and imaging approaches, such as NeighborNet, Heatmap, and STRUCTURE, to identify nuclear genomes admixture. Estimation of the intensity of spontaneous hybridization within and between Cirsium and Lophiolepis was based on herbarium revisions and published data for all reported hybrids pertinent to taxa currently included in Cirsium or Lophiolepis.The genome of any examined Cirsium species is more similar to C. vulgare than to any Lophiolepis species, and vice versa. The nuclear genome of the tetraploid C. vulgare is composed of two equivalent parts, each attributable either to Lophiolepis or to Cirsium; the organellar RADseq data clustered C. vulgare with the genus Cirsium. Spontaneous hybridization between Cirsium and Lophiolepis is significantly less intensive than within these genera.Our analyses provide compelling evidence that the invasive species C. vulgare has an allotetraploid intergeneric origin, with the maternal parent from Cirsium and the paternal from Lophiolepis. For the purpose of delimiting monophyletic genera, we propose keeping Lophiolepis separate from Cirsium and segregating C. vulgare into the hybridogenous genus Ascalea. [ABSTRACT FROM AUTHOR]

Purpose: This case study refers to a case of complete androgen insensitivity syndrome, which presented with amenorrhea back in the past. Case: After conformation of the diagnosis, parents decided not to reveal the condition’s specifics to the patient. At the age of 34 years, she attended our hospital with abdominal swelling, and the following CT scan showed an abdominal mass arising from the ovary of 13 cm x 14 cm size. A laparotomy was done, and a mixture of solid and cystic mass of 20 cm in diameter was found and removed with the ovary. Histopathology of the tumor showed features of testicular seminoma. Conclusion: The aim of this case report is to outline the difficulties in choosing whether and when to do the gonadectomy and whether and how to explain the condition to the patient.

BackgroundMR-Linac allows for daily online treatment adaptation to the observed geometry of tumor targets and organs at risk (OARs). Manual delineation for head and neck cancer (HNC) patients takes 45-75 minutes, making it unsuitable for online adaptive radiotherapy. This study aims to clinically and dosimetrically validate an in-house developed algorithm which automatically delineates the elective target volume and OARs for HNC patients in under a minute.MethodsAuto-contours were generated by an in-house model with 2D U-Net architecture trained and tested on 52 MRI scans via leave-one-out cross-validation. A randomized selection of 684 automated and manual contours (split half-and-half) was presented to an oncologist to perform a blind test and determine the clinical acceptability. The dosimetric impact was investigated for 13 patients evaluating the differences in dosage for all structures.ResultsAutomated contours were generated in 8 seconds per MRI scan. The blind test concluded that 114 (33%) of auto-contours required adjustments with 85 only minor and 15 (4.4%) of manual contours required adjustments with 12 only minor. Dosimetric analysis showed negligible dosimetric differences between clinically acceptable structures and structures requiring minor changes. The Dice Similarity coefficients for the auto-contours ranged from 0.66 ± 0.11 to 0.88 ± 0.06 across all structures.ConclusionMajority of auto-contours were clinically acceptable and could be used without any adjustments. Majority of structures requiring minor adjustments did not lead to significant dosimetric differences, hence manual adjustments were needed only for structures requiring major changes, which takes no longer than 10 minutes per patient.

Nanotechnology is rapidly advancing towards the development of applications for sustainable plant growth and photosynthesis optimization. The nanomaterial/plant interaction has been intensively investigated; however, there is still a gap in knowledge regarding their effect on crop seed development and photosynthetic performance. In the present work, we apply a priming procedure with 10 and 50 mg/L Pluronic-P85-grafted single-walled carbon nanotubes (P85-SWCNT) on garden pea seeds and examine the germination, development, and photosynthetic activity of young seedlings grown on soil substrate. The applied treatments result in a distorted topology of the seed surface and suppressed (by 10–19%) shoot emergence. No priming-induced alterations in the structural and functional features of the photosynthetic apparatus in 14-day-old plants are found. However, photosynthetic gas exchange measurements reveal reduced stomatal conductance (by up to 15%) and increased intrinsic water use efficiency (by 12–15%), as compared to hydro-primed variants, suggesting the better ability of plants to cope with drought stress—an assumption that needs further verification. Our study prompts further research on the stomatal behavior and dark reactions of photosynthesis in order to gain new insights into the effect of carbon nanotubes on plant performance.

Extracts from medicinal plants are widely used in the treatment and prevention of different diseases. Micromeria frivaldszkyana is a Balkan endemic species with reported antioxidant and antimicrobial characteristics; however, its phytochemical composition is not well defined. Here, we examined the metabolome of M. frivaldszkyana by chromatography–mass spectrometry (GC-MS), ultra-performance liquid chromatography–mass spectrometry (UPLC-MS-MS), and inductively coupled plasma mass spectrometry (ICP-MS). Amino acids, organic acids, sugars, and sugar alcohols were the primary metabolites with the highest levels in the plant extract. Detailed analysis of the sugar content identified high levels of sucrose, glucose, mannose, and fructose. Lipids are primary plant metabolites, and the analysis revealed triacylglycerols as the most abundant lipid group. Potassium (K), magnesium (Mg), zinc (Zn), and calcium (Ca) were the elements with the highest content. The results showed linarin, 3-caffeoil-quinic acid, and rosmarinic acid, as well as a number of polyphenols, as the most abundant secondary metabolites. Among the flavonoids and polyphenols with a high presence were eupatorin, kaempferol, and apigenin—compounds widely known for their bioactive properties. Further, the acute toxicity and potential anti-inflammatory activity of the methanolic extract were evaluated in Wistar rats. No toxic effects were registered after a single oral application of the extract in doses of between 200 and 5000 mg/kg bw. A fourteen-day pre-treatment with methanolic extract of M. frivaldszkyana in doses of 250, 400, and 500 mg/kg bw induced anti-inflammatory activity in the 1st, 2nd, and 3rd hours after carrageenan injection in a model of rat paw edema. This effect was also present in the 4th hour only in the group treated with a dose of 500 mg/kg. In conclusion, M. frivaldszkyana extract is particularly rich in linarin, rosmarinic acid, and flavonoids (eupatorin, kaempferol, and apigenin). Its methanolic extract induced no toxicity in male Wistar rats after oral application in doses of up to 5000 mg/kg bw. Additionally, treatment with the methanolic extract for 14 days revealed anti-inflammatory potential in a model of rat paw edema on the 1st, 2nd, and 3rd hours after the carrageenan injection. These results show the anti-inflammatory potential of the plant, which might be considered for further exploration and eventual application as a phytotherapeutic agent.

Fucoidans are sulfated polysaccharides detected mainly in the cell walls of brown seaweeds. Here, we examined the effects of single doses of fucoidan derived from Ericaria crinita (formerly Cystoseira crinita) on carrageenan-induced paw inflammation in rats. The serum levels of TNF-α, IL-1β, IL-6, and IL-10 of rats with LPS-induced systemic inflammation after 14 days of treatment were also evaluated. Subchronic treatment with fucoidan from E. crinita attenuated the inflammation during the late phase of the degraded carrageenan-induced paw edema (3rd to 5th hour after carrageenan injection) with peak activity at the 3rd hour after the application. Both doses of fucoidan from E. crinita (25 and 50 mg/kg bw) significantly decreased the levels of all tested pro-inflammatory cytokines (IL-1β, TNF-α, and IL-6) in the serum of rats with a model of system inflammation but had no effect on the anti-inflammatory cytokine IL-10. The results showed that the repeated application of fucoidan has a more prominent effect on the levels of some pro-inflammatory cytokines in serum in comparison to a single dose of the sulfated polysaccharide. This reveals the potential of E. crinita fucoidan as an anti-inflammatory agent. Furthermore, E. crinita fucoidan exhibited in vitro antioxidant capacity, determined by 2,2-diphenyl-1-picryl-hydrazyl radical scavenging and ferric reducing antioxidant power assays as follows: IC50 = 412 µg/mL and 118.72 μM Trolox equivalent/g, respectively.

Etifoxine is an anxiolytic drug with a dual mechanism of action. In contrast to conventional benzodiazepine anxiolytics, which induce cognitive dysfunction and myorelaxation, no memory impairment nor a decrease in motor activity is observed with etifoxine. This study aims to evaluate the effects of etifoxine on locomotor activity and passive learning in rats with diazepam-induced memory deficit. Male Wistar rats were treated intraperitoneally for 7 days with: (1) saline; (2) diazepam 2.5 mg/kg bw or (3) diazepam 2.5 mg/kg bw and etifoxine in a dose of 50 mg/kg bw. Activity cage test was used for evaluation of locomotor activity, and step-through and step-down tests were performed to study the passive learning. Etifoxine increased the number of horizontal movements on the 7th and 14th days of the experiment. The drug exhibits anti-amnesic effect in a model of diazepam-induced anterograde amnesia by enhancing long-term memory in passive learning tests. The data obtained suggest that etifoxine can reduce the benzodiazepine-induced cognitive deficit. Moreover, such a combination can alleviate the negative influence of benzodiazepines on locomotor activity. However, additional studies are necessary to translate these results into clinical practice.

The aim of this study was to identify the chemical composition and sequential structure of alginate isolated from C. crinita harvested in the Bulgarian Black Sea, as well as its effects in histamine-induced paw inflammation in rats. The serum levels of TNF-α, IL-1β, IL-6, and IL-10 in rats with systemic inflammation, and the levels of TNF-α in a model of acute peritonitis in rats were also investigated. The structural characterization of the polysaccharide was obtained by FTIR, SEC-MALS, and 1H NMR. The extracted alginate had an M/G ratio of 1.018, a molecular weight of 7.31 × 104 g/mol, and a polydispersity index of 1.38. C. crinita alginate in doses of 25 and 100 mg/kg showed well-defined anti-inflammatory activity in the model of paw edema. A significant decrease in serum levels of IL-1β was observed only in animals treated with C. crinita alginate in a dose of 25 mg/kg bw. The concentrations of TNF-α and IL-6 in serum were significantly reduced in rats treated with both doses of the polysaccharide, but no statistical significance was observed in the levels of the anti-inflammatory cytokine IL-10. A single dose of alginate did not significantly alter the levels of the pro-inflammatory cytokine TNF-α in the peritoneal fluid of rats with a model of peritonitis.

The aim of this study was to evaluate the effects of fucoidan isolated from C. crinita on histamine-induced paw inflammation in rats, and on the serum levels of TNF-α, IL-1β, IL-6, and IL-10 in rats during systemic inflammation response. The levels of TNF-α in a model of acute peritonitis in rats were also investigated. The isolated crude fucoidan was identified as a sulfated xylogalactofucan with high, medium, and low molecular weight fractions and a content of fucose of 39.74%, xylose of 20.75%, and galactose of 15.51%. Fucoidan from C. crinita showed better anti-inflammatory effects in the rat paw edema model, and this effect was present during all stages of the experiment. When compared to controls, a commercial fucoidan from F. vesiculosus, the results also displayed anti-inflammatory activity on the 60th, 90th, and 120th minute of the experiment. A significant decrease in serum levels of IL-1β in rats treated with both doses of C. crinita fucoidan was observed in comparison to controls, whereas TNF-α concentrations were reduced only in the group treated with fucoidan from C. crinita at the dose of 25 mg/kg bw. In the model of carrageenan-induced peritonitis, we observed a tendency of decrease in the levels of the pro-inflammatory cytokine TNF-α in peritoneal fluid after a single dose of C. crinita fucoidan, but this did not reach the statistical significance margin. Single doses of C. crinita fucoidan did not alter serum levels of the anti-inflammatory cytokine IL-10 in animals with lipopolysaccharide-induced systemic inflammation.

The European Union adopted a decision to achieve by 2020 at least 20% reduction in greenhouse gas emissions. To fulfill this task, the EU proposed in 2020 binding targets - further improve energy efficiency by 20%, achieving a 20% share of renewable energy and a 10% share of biofuels in the fuel market. One of the most widely used biofuel in the automotive industry is bioethanol. Bioethanol can be used on low-percentage blending into petrol, which is governed by European Directive 2003/30/EC, and on high-blend bioethanol mixture, particularly E85 consists of 85% bioethanol and 15% petrol BA95. But in recent years, increasingly demonstrating opinions, that biofuels do not produce nearly as much effect as was propagated. This paper deals with the concentration of water in the engine oil with using biofuel E85. The water in the engine oil significantly affects its parameters (especially viscosity). How measurement results demonstrated the use of bioethanol in the fuel increases the concentration of water in the engine oil. [ABSTRACT FROM AUTHOR]

Combining the selective targeting of tumor cells through antigen-directed recognition and potent cell-killing by cytotoxic payloads, antibody-drug conjugates (ADCs) have emerged in recent years as an efficient therapeutic approach for the treatment of various cancers. Besides a number of approved drugs already on the market, there is a formidable follow-up of ADC candidates in clinical development. While selection of the appropriate antibody (A) and drug payload (D) is dictated by the pharmacology of the targeted disease, one has a broader choice of the conjugating linker (C). In the present paper, we review the chemistry of ADCs with a particular emphasis on the medicinal chemistry perspective, focusing on the chemical methods that enable the efficient assembly of the ADC from its three components and the controlled release of the drug payload.

Research background: The People’s Republic of China in the 21st century can be described as an economy with high growth rates and great ambitions. Some statistics even indicate that China will become the world’s new hegemon by 2040. The People’s Republic of China is not only one of the largest exporters but is increasingly speaking to the world economy and international relations. Since the beginning of the 1990s, mutual relations between the People’s Republic of China and the Czech Republic can be described as very good and constantly deepening, which can be evidenced, among other things, by the number of trade agreements. Purpose of the article: Purpose of the article is to evaluate a development of mutual relations between Czech and Chinese economies in order to predict their future development. Emphasis will be placed on the development of mutual trade through the evaluation of absolute and relative indicators and growth rates. Methods: The paper is based on secondary data from the database of the Czech Statistical Office. The key methods used in the article are the analysis of time series of real products of the Czech Republic and China and their foreign trade. The analysis focuses on the trend, deviations and development of absolute and relative indicators. Findings & Value added: The results of the analysis proved that the Chinese economy is growing on average more than twice as fast as the Czech economy. Because of that, the economic/living standards of the population of both countries are converging. The growth rate of the People’s Republic of China, together with its high spending on science and research, means that (if this trend is maintained) China is likely to become the world’s new hegemon in the near future.

Research background: The economy with the strongest influence on world affairs, international politics and world economy is undoubtedly the economy of the United States of America. In the Czech Republic, after the revolution, the USA quickly came to the forefront of interest and mutual foreign trade grew significantly. However, their global influence was significantly reflected not only in economic aspects, but also in sociological aspects. In recent years, the so-called “Americanization” has been taking place in the Czech Republic. American terms were often adopted in the commonly used Czech language, and the demand for American goods increased significantly. Purpose of the article: The objective of the presented text is to evaluate development of the influence of the USA on the economy of the Czech Republic by analysing the development of key macroeconomic quantities, in particular gross domestic product, gross national product, balance of payments and foreign trade. Based on the results of this analysis, the future development of the Czech economy, more precisely opportunities and impacts arising from mutual trade relations between the American and Czech economies, is predicated. Methods: Descriptive statistic. Findings & Value added: The results of the research showed a gradual expansion of the influence of the American economy in the Czech Republic, which began after 1989 and continues to this day. This influence manifested itself in several aspects. It was reflected in the structure of mutual foreign trade, and also in the Czech culture and the Czech language (adoption of English terms into the Czech language). The further potential of mutual trade is highlighted out by comparing the structure of export and import from the USA in total and export and import from the USA to the Czech Republic.

Research background: Even though we live in the 21st century, where society is no longer expected to treat women and men differently, the reality is unfortunately different. Gender differences can be observed in virtually every country in the world. In some aspects of gender inequality, the Czech Republic is doing relatively well, for example, in terms of equal access to education, but in other areas it is doing much worse. In the Czech Republic, gender inequality is manifested in particular in the gender pay gap and on the labour market situation, where female unemployment is higher than male unemployment. Purpose of the article: This article analyses gender inequality in the Czech Republic on the labour market. The aim of this article is to evaluate the state, development and estimate the future direction of the labour market with an emphasis on gender inequality. Methods: Using time-series analysis of the number of applicants at the labour offices, it assesses the status and development of gender inequality in the labour market. The analysis also includes a prediction of future developments, where the author seeks an answer to the question whether the gender gap in this area is decreasing or increasing over time. Findings & Value added: The analysis showed a persistent trend of higher unemployment among women compared to men. However, this gap is decreasing in the long term.

Inflammation is the initial response of the immune system to potentially harmful stimuli (e.g., injury, stress, and infections). The process involves activation of macrophages and neutrophils, which produce mediators, such as nitric oxide (NO), prostaglandin E2 (PGE2), pro-inflammatory and anti-inflammatory cytokines. The pro-inflammatory cytokines interleukin-1β (IL-1β), interleukin 6 (IL-6), and tumor necrosis factor-α (TNF-α) are considered as biomarkers of inflammation. Even though it occurs as a physiological defense mechanism, its involvement in the pathogenesis of various diseases is reported. Rheumatoid arthritis, inflammatory bowel disease, Alzheimer’s disease, and cardiovascular diseases are only a part of the diseases, in which pathogenesis the chronic inflammation is involved. Fucoidans are complex polysaccharides from brown seaweeds and some marine invertebrates, composed mainly of l-fucose and sulfate ester groups and minor amounts of neutral monosaccharides and uronic acids. Algae-derived fucoidans are studied intensively during the last years regarding their multiple biological activities and possible therapeutic potential. However, the source, species, molecular weight, composition, and structure of the polysaccharides, as well as the route of administration of fucoidans, could be crucial for their effects. Fucoidan is reported to act on different stages of the inflammatory process: (i) blocking of lymphocyte adhesion and invasion, (ii) inhibition of multiple enzymes, and (iii) induction of apoptosis. In this review, we focused on the immunemodulating and anti-inflammatory effects of fucoidans derived from macroalgae and the models used for their evaluation. Additional insights on the molecular structure of the compound are included.

Hematological effects of the herbicide Avalon (GAL-57), a mixture of bentazon and dicamba,were tested on rats. Avalon was administered by gavage at three and four dose levels(250, 500, 1000 and 1250 mg/kg weight/day) for 28 and 90 days.Hematological parameters, number of erythrocytes, leukocytes and platelets, hemoglobinconcentration, hematocrit and erythrocyte indexes (MCV, MCH and MCHC) weremonitored.The results showed that the herbicide Avalon caused decrease in the values of hemoglobin,hematocrit and erythrocyte indexes (both males and females). The changes(mostly) correlated with the doses administered and, in most cases, a lower susceptibilityof females than males was observed. The herbicide GAL-57 had no adverse effect on thenumber of leukocytes, erythrocytes and thrombocytes (both sexes, all doses tested).The results showed that the herbicide Avalon causes weak anemia to the animals tested.Reversibility was apparent during the recovery period of 28 days.

A micronucleus test was run to investigate mutagenic potential of the herbicide GAL-57, a formulated mixture of bentazone and dicamba.The test was applied to mice of both sexes (strain: CRL: NMRI BR) and the herbicide (product) was administered by gavage at 2000 mg/kg rate, twice within 24 hs. Cyclophosphamide (positive control) was administered at 60 mg/kg, while distilled water as a solvent was negative control. The animals were sacrificed 24 hs after second treatment, their bone marrow cells isolated from femur, and effects evaluated.The data acquired showed that repeated treatment of mice with GAL-57 caused neither biological nor significant statistical increase in the number of micronuclei in treated animals. At the same time, the number of micronucleated polychromatic erythrocytes in the bone marrow of animals treated with cyclophosphamide (positive control) showed a significant statistical increase. The results suggest that the herbicide product tested did not show any mutagenic activity under the conditions of mouse micronucleus test.
