A study relating to research on the structure and properties of dendrimers was conducted to investigate the influence of solvents on the hydrodynamical volume of dendrimers. Specifically, a fluorophore-labeled dendrimer with a rubicene derivative as a core was covalently linked to the dendrons and fluoresence depolarization was used to derive the hydrodynamical volume of this labeled dendrimer. The experiment and its results are discussed.

Spherical micellar aggregates have been obtained in chloroform by mixing poly(styrene)-block-poly(4-vinylpyridine) (PS-b-P4VP) diblock copolymers with perfluorinated surfactants (FS) bearing a carboxylic acid head. These micellar aggregates are resulting from the self-assembly of the insoluble P4VP/fluorinated complexes into a core surrounded by the soluble PS coronal chains. Their characteristic features have been studied as a function of various parameters including the composition of the PS-b-P4VP copolymer, the tail length of the fluorinated surfactant, the 4VP/FS molar ratio, the number of carboxylic acid group (1 or 2) on the surfactant, the presence of the PS block and of the fluorine atoms on the surfactant. Dilution of these initial micellar aggregates triggers a morphological reorganization resulting in the formation of more stable vesicles. The extent of this morphological transition is related to the solubility of the P4VP/fluorinated complexes during the dilution process. This transition is complete for short P4VP/FS complexes, incomplete for long P4VP/FS complexes, and not observed whenever an alpha,omega-difunctional FS is used in P4VP/FS complexes, leading to a cross-linked core. Finally, the spheres-to-vesicles transition has been advantageously used in order to encapsulate molecules, as demonstrated by confocal fluorescence microscopy.

Wide-field imaging of individual multichromophoric molecules and successive photobleaching were used to determine, accurately, the relative position of the chromophores in such systems. First, a polyphenylene dendrimer with well-defined geometry was used to establish the accuracy in localization that can be obtained by this methodology. For a signal-to-noise ratio of 20, interchromophoric distances could be measured with 4 nm accuracy. Next, the method was used to determine the end-to-end distribution of an end-capped polyfluorene polymer. From comparison between the experimental and simulated distributions, information on the conformation of the polymer could be deduced. It was found that the polymer has a nonlinear conformation. A conjugation length of six monomer units gave the best fit of the experimental data to the proposed model.

Reversible photoswitching of individual molecules has been demonstrated for a number of mutants of the green fluorescent protein (GFP). To date, however, a limited number of switching events with slow response to light have been achieved at the single-molecule level. Here, we report reversible photoswitching characteristics observed in individual molecules of Dronpa, a mutant of a GFP-like fluorescent protein that was cloned from a coral Pectiniidae. Ensemble spectroscopy shows that intense irradiation at 488 nm changes Dronpa to a dim protonated form, but even weak irradiation at 405 nm restores it to the bright deprotonated form. Although Dronpa exists in an acid-base equilibrium, only the photoinduced protonated form shows the switching behavior. At the single-molecule level, 488- and 405-nm lights can be used to drive the molecule back and forth between the bright and dim states. Such reversible photoswitching could be repeated >100 times. The response speed to irradiation depends almost linearly on the irradiation power, with the response time being in the order of milliseconds. The perfect reversibility of the Dronpa photoswitching allows us to propose a detailed model, which quantitatively describes interconversion among the various states. The fast response of Dronpa to light holds great promise for following fast diffusion or transport of signaling molecules in live cells.

The differences in the fluorescence behavior of a polyphenylene dendrimer with eight peryleneimides chromophores (1) and a single hexaphenylperyleneimide chromophore have been investigated at a single-molecule level through the combination of ultrasensitive fluorescence detection and microscopy.
