Abstract

Light/dark (LD) cycles are responsible for oscillations in gene expression, which modulate several aspects of plant physiology. Those oscillations can persist under constant conditions due to regulation by the circadian oscillator. The response of the transcriptome to light regimes is dynamic and allows plants to adapt rapidly to changing environmental conditions. We compared the transcriptome of Arabidopsis under LD and constant light (LL) for 3 days and identified different gene co‐expression networks in the two light regimes. Our studies yielded unforeseen insights into circadian regulation. Intuitively, we anticipated that gene clusters regulated by the circadian oscillator would display oscillations under LD cycles. However, we found transcripts encoding components of the flavonoid metabolism pathway that were rhythmic in LL but not in LD. We also discovered that the expressions of many stress‐related genes were significantly increased during the dark period in LD relative to the subjective night in LL, whereas the expression of these genes in the light period was similar. The nocturnal pattern of these stress‐related gene expressions suggested a form of "skotoprotection." The transcriptomics data were made available in a web application named Cyclath, which we believe will be a useful tool to contribute to a better understanding of the impact of light regimes on plants. Abstract

The species Arabidopsis halleri, an emerging model for the study of heavy metal tolerance and accumulation in plants, has evolved a high level of constitutive zinc tolerance. Mapping of quantitative trait loci (QTL) was used to investigate the genetic architecture of zinc tolerance in this species. A first generation backcross progeny of A. halleri ssp. halleri from a highly contaminated industrial site and its nontolerant relative A. lyrata ssp. petraea was produced and used for QTL mapping of zinc tolerance. A genetic map covering most of the A. halleri genome was constructed using 85 markers. Among these markers, 65 were anchored in A. thaliana and revealed high synteny with other Arabidopsis genomes. Three QTL of comparable magnitude on three different linkage groups were identified. At all QTL positions zinc tolerance was enhanced by A. halleri alleles, indicating directional selection for higher zinc tolerance in this species. The two-LOD support intervals associated with these QTL cover 24, 4, and 13 cM. The importance of each of these three regions is emphasized by their colocalization with HMA4, MTP1-A, and MTP1-B, respectively, three genes well known to be involved in metal homeostasis and tolerance in plants.

Abstract

Essential trace metals function as structural components or cofactors in many proteins involved in a wide range of physiological processes in plants. Hence, trace metal deficiency can significantly hamper plant growth and development. On the other hand, excess concentrations of trace metals can also induce phytotoxicity, for example via an enhanced production of reactive oxygen species. Besides their roles in plant growth under favourable environmental conditions, trace metals also contribute to plant responses to biotic and abiotic stresses. Heat is a stress factor that will become more prevalent due to increasing climate change and is known to negatively affect crop yield and quality, posing a severe threat to food security for future generations. Gaining insight into heat stress responses is essential to develop strategies to optimize plant growth and quality under unfavourable temperatures. In this context, trace metals deserve particular attention as they contribute to defence responses and are important determinants of plant nutritional value. Here, we provide an overview of heat-induced effects on plant trace metal homeostasis and the involvement of trace metals and trace metal-dependent enzymes in plant responses to heat stress. Furthermore, avenues for future research on the interactions between heat stress and trace metals are discussed. [ABSTRACT FROM AUTHOR]
