Abstract: PLATO (PLAnetary Transits and Oscillations of stars) is ESA's M3 mission designed to detect and characterise extrasolar planets and perform asteroseismic monitoring of a large number of stars. PLATO will detect small planets (down to <2 R_(Earth)) around bright stars (<11 mag), including terrestrial planets in the habitable zone of solar-like stars. With the complement of radial velocity observations from the ground, planets will be characterised for their radius, mass, and age with high accuracy (5 %, 10 %, 10 % for an Earth-Sun combination respectively). PLATO will provide us with a large-scale catalogue of well-characterised small planets up to intermediate orbital periods, relevant for a meaningful comparison to planet formation theories and to better understand planet evolution. It will make possible comparative exoplanetology to place our Solar System planets in a broader context. In parallel, PLATO will study (host) stars using asteroseismology, allowing us to determine the stellar properties with high accuracy, substantially enhancing our knowledge of stellar structure and evolution. The payload instrument consists of 26 cameras with 12cm aperture each. For at least four years, the mission will perform high-precision photometric measurements. Here we review the science objectives, present PLATO's target samples and fields, provide an overview of expected core science performance as well as a description of the instrument and the mission profile at the beginning of the serial production of the flight cameras. PLATO is scheduled for a launch date end 2026. This overview therefore provides a summary of the mission to the community in preparation of the upcoming operational phases.

Abstract: This Letter reports the first observational estimate of the heating rate in the slowly expanding solar corona. The analysis exploits the simultaneous remote and local observations of the same coronal plasma volume with the Solar Orbiter/Metis and the Parker Solar Probe instruments, respectively, and relies on the basic solar wind magnetohydrodynamic equations. As expected, energy losses are a minor fraction of the solar wind energy flux, since most of the energy dissipation that feeds the heating and acceleration of the coronal flow occurs much closer to the Sun than the heights probed in the present study, which range from 6.3 to 13.3 solar radii. The energy deposited to the supersonic wind is then used to explain the observed slight residual wind acceleration and to maintain the plasma in a non-adiabatic state. As derived in the Wentzel-Kramers-Brillouin limit, the present energy transfer rate estimates provide a lower limit, which can be very useful in refining the turbulence-based modeling of coronal heating and subsequent solar wind acceleration.

Abstract: Solar flares are occasionally responsible for severe Space Weather events, which can affect space-borne and ground-based infrastructures, endangering anthropic technological activities and even human health and safety. Thus, an essential activity in the framework of Space Weather monitoring is devoted to the observation of the activity level of the Sun. In this context, the acquisition system of the Catania Solar Telescope has been recently upgraded in order to improve its contribution to the European Space Agency (ESA) - Space Weather Service Network through the ESA Portal, which represents the main asset for Space Weather in Europe. Here, we describe the hardware and software upgrades of the Catania Solar Telescope and the main data products provided by this facility, which include full-disc images of the photosphere and chromosphere, together with a detailed characterization of the sunspot groups. As a showcase of the observational capabilities of the revamped Catania Solar Telescope, we report the analysis of a B5.4 class flare occurred on 2020 December 7, simultaneously observed by the IRIS and SDO satellites.

Abstract: Since the onset of the `space revolution' of high-precision high-cadence photometry, asteroseismology has been demonstrated as a powerful tool for informing Galactic archaeology investigations. The launch of the NASA TESS mission has enabled seismic-based inferences to go full sky -- providing a clear advantage for large ensemble studies of the different Milky Way components. Here we demonstrate its potential for investigating the Galaxy by carrying out the first asteroseismic ensemble study of red giant stars observed by TESS. We use a sample of 25 stars for which we measure their global asteroseimic observables and estimate their fundamental stellar properties, such as radius, mass, and age. Significant improvements are seen in the uncertainties of our estimates when combining seismic observables from TESS with astrometric measurements from the Gaia mission compared to when the seismology and astrometry are applied separately. Specifically, when combined we show that stellar radii can be determined to a precision of a few percent, masses to 5-10% and ages to the 20% level. Abstract: Metis is the first solar coronagraph designed for a space mission capable of performing simultaneous imaging of the off-limb solar corona in both visible and UV light. The observations obtained with Metis aboard the Solar Orbiter ESA-NASA observatory will enable us to diagnose, with unprecedented temporal coverage and spatial resolution, the structures and dynamics of the full corona from 1.7 $R_\odot$ to about 9 $R_\odot$. Due to the uniqueness of the Solar Orbiter mission profile, Metis will be able to observe the solar corona from a close vantage point (down to 0.28 AU), achieving out-of-ecliptic views with the increase of the orbit inclination over time. Moreover, observations near perihelion, during the phase of lower rotational velocity of the solar surface relative to the spacecraft, will allow longer-term studies of the coronal features. Thanks to a novel occultation design and a combination of a UV interference coating of the mirrors and a spectral bandpass filter, Metis images the solar corona simultaneously in the visible light band, between 580 and 640 nm, and in the UV H I Lyman-{\alpha} line at 121.6 nm. The coronal images in both the UV Lyman-{\alpha} and polarised visible light are obtained at high spatial resolution with a spatial scale down to about 2000 km and 15000 km at perihelion, in the cases of the visible and UV light, respectively. A temporal resolution down to 1 second can be achieved when observing coronal fluctuations in visible light. The Metis measurements will allow for complete characterisation of the main physical parameters and dynamics of the electron and neutral hydrogen/proton plasma components of the corona in the region where the solar wind undergoes acceleration and where the onset and initial propagation of coronal mass ejections take place, thus significantly improving our understanding of the region connecting the Sun to the heliosphere.

Abstract: In patients with ischaemic stroke, a carotid free-floating thrombus (CFFT) raises diagnostic and therapeutic challenges. We describe two women, each taking tamoxifen for invasive non- metastatic breast cancer, who developed large-vessel occlusion ischaemic strokes. The first had a CFFT 24 hours after receiving intravenous thrombolysis and mechanical thrombectomy; the thrombus completely resolved after 1 week of therapeutic anticoagulation. The second had a tandem occlusion with a CFFT at admission; her neurological deficits rapidly improved after intravenous thrombolysis without needing a mechanical thrombectomy. However, subsequently, under therapeutic anticoagulation, distal migration of the CFFT caused a recurrent large vessel occlusion ischaemic stroke, requiring mechanical thrombectomy. The CTFF in both cases appeared to relate to a cancer-related prothrombotic state. Both received long-term oral anticoagulation and their tamoxifen was switched to anastrozole. Abstract: Introduction: Cardiology and cardiothoracic surgery are among the specialties that most commonly require neurology inpatient consultations. We aimed to study the neurology referrals by the cardiovascular-specialized hospital included in our tertiary hospital center. Methods: Retrospective study of consecutive patients referred for neurology inpatient consultation between January 1, 2020, and December 31, 2022. We analyzed referrals, patients' characteristics, and the approach taken. A detailed subanalysis was performed for patients diagnosed with acute ischemic stroke (AIS). Results: 143 patients were observed (mean age 67.3 years, 46 [32.2%] females). Most frequent referral reasons were suspected AIS deficits (39.2%), altered mental status (19.6%), suspected seizures (13.3%), and neuroprognostication (11.9%). Mean referral-to-consult time was 2.7 days, and 117 (81.8%) consults were in-person. Additional investigation, treatment changes, and outpatient clinic referral were proposed, respectively, in 79.7%, 60.1%, and 19.6% of patients. Most common diagnoses were AIS (45.5%), hypoxic-ischemic encephalopathy (18.9%), and delirium (7.0%). Regarding patients with AIS (n = 62), most common stroke causes were post-cardiac procedure (44.6%), infective endocarditis (18.5%), aortic dissection (10.8%), acute myocardial infarction (10.8%), and anticoagulant withdrawal in patients with atrial fibrillation (6.2%). Thirty-four AIS patients were diagnosed less than 24 h since last seen well, of which four (6.2%) were treated (three with thrombolysis and one with mechanical thrombectomy). Conclusion: AIS is the most common reason for referral in our cardiovascular hospital. Our results highlight the importance of the availability of a neurologist/neurohospitalist with stroke expertise for consultation of inpatients admitted in a specialized cardiovascular hospital. 24. Epileptogenic focal lesions in Dravet syndrome: A warning to investigators

Abstract: Most patients with Dravet syndrome (DS) have unremarkable neuroimaging studies. However, a small number of patients exhibit focal abnormalities that may modify the epilepsy phenotype. We report a case series of DS patients carrying SCN1Avariants concurrent with additional focal brain lesions, aiming to provide details regarding their clinical course, electrographic findings, and imaging features. We reviewed the electronic medical records of patients with developmental and epileptic encephalopathies in our center, from January 2000 to December 2022, identifying 90 patients with DS resulting from SCN1Avariants. Of these, patients displaying focal brain lesions were eligible. Five patients (4 males and 1 female), with median age of 26 years, were included. All exhibited clinical and electroencephalographic features consistent with the DS spectrum. Sequencing analysis of the SCN1Agene identified pathogenic variants. Magnetic resonance imaging (MRI) revealed focal cortical dysplasia (FCD) in two patients, while the remaining three had cystic lesions. Three patients had previously undergone resective epilepsy surgery in other centers, with no improvement in seizure frequency. Neuropathology studies revealed the presence of FCD type IIA, intracranial teratomas, and dysembryoplastic neuroepithelial tumor (DNET). When an individual with an established diagnosis of genetic epilepsy and a focal lesion on MRI is undergoing preoperative evaluation, it is crucial to conduct a comprehensive analysis to understand the relevance of the focal finding for the patient's phenotype and thus anticipate potential surgical outcomes. In instances where epilepsy in DS patients is influenced by a specific focal structural lesion, resective surgery should be carefully considered after precise pharmacological treatment, acknowledging the persistent influence of an SCN1Avariant on expected outcomes.

Abstract: The solar wind measured in situ by Parker Solar Probe in the very inner heliosphere is studied in combination with the remote-sensing observation of the coronal source region provided by the METIS coronagraph aboard Solar Orbiter. The coronal outflows observed near the ecliptic by Metis on 2021 January 17 at 16:30 UT, between 3.5 and 6.3 R (circle dot) above the eastern solar limb, can be associated with the streams sampled by PSP at 0.11 and 0.26 au from the Sun, in two time intervals almost 5 days apart. The two plasma flows come from two distinct source regions, characterized by different magnetic field polarity and intensity at the coronal base. It follows that both the global and local properties of the two streams are different. Specifically, the solar wind emanating from the stronger magnetic field region has a lower bulk flux density, as expected, and is in a state of well-developed Alfvenic turbulence, with low intermittency. This is interpreted in terms of slab turbulence in the context of nearly incompressible magnetohydrodynamics. Conversely, the highly intermittent and poorly developed turbulent behavior of the solar wind from the weaker magnetic field region is presumably due to large magnetic deflections most likely attributed to the presence of switchbacks of interchange reconnection origin.

Abstract: A technologocal tool is used to measure the effectiveness of students’ learning in the subjects os Statistics I adn Statistics II of the Administration course, ande in the Statistic subjects of the Production Engeneering and Accounting Sciences courses of a university center in Zona dmore interesa Mata Mineira; this tool has proven to be useful and agile for real feedback on class learning. The tool in study is called Plickers, being classified as Classroom Response. In mentioned subjects, the tool was integrated into the classes using the Peer Instruction Methodology (PI), in which students learn frim their peers. The tool was well accepted by students who were instantly more interested in classes and new technology, as shown by the results. The fact that each studant has a presponse card, for exclusive use, makes the students’ omission, when answering a question proposed by the teacher, non-existent. This way, this is no longer a problem in the classroom, the teacher has, quickly, the real leaning diagnosis of his students, this allows measures to be taken, when learning can be considered unsatisfactory. The tool allows dynamism during classes, allowing the teacher to make decisions and paths that can revesse unsatisfactory situations. Se utiliza una herramienta tecnológica para medir la efectividad del aprendizaje de los estudiantes, en las asignaturas de Estadística I y Estadística II del curso de Administración, y en las asignaturas de Estadística de los cursos de Ingeniería de Producción y Ciencias Contables de un centro universitário en la Zona da Mata Mineira. Esta herramienta há demonstrado ser útil y ágil para obtener comentários reales sobre el aprendizaje en classe. La herramienta en cuestión se llama Plickers, y se classifica como una respuesta en la clase. En las matérias antes mencionadas, la herramienta se integró en las clases utilizando la metodogía de instrucción entre pares (PI), en la que los estudiantes aprenden de sus compañeros. La herramienta fue bien aceptada por los estudiantes, que instantaneamente se mostraron más interessados en las clases y las nuevas tecnologías, como lo demuestran los resultados. El hecho de que cada estudiante tenga una carta de respuesta, para uso exclusivo, hace que la omisión de los estudiantes, al responder una pregunta propuesta por el maestro, sea inexistente. De esta manera, esto ya no es un problema en la clase, el maestro tiene, rapidamente, el diagnóstico real de aprendizaje de sus alumnos, esto permite tomar medidas cuando el aprendizaje puede considerarse insatisfactorio. La herramienta permite dinamismo durante las clases, lo que permite al maestro tomar decisiones y caminos que pueden revertir situaciones insatisfactorias. O Plickers® é uma ferramenta tecnológica utilizada para medir a efetividade do aprendizado dos alunos, sendo classificada como um Classroom Response. Por ser considerada útil e ágil para medir o real feedback do aprendizado, foi utilizada nas turmas nas disciplinas de Estatística I e Estatística II do curso de Administração e nas disciplinas de Estatística dos cursos de Engenharia de Produção e Ciências Contábeis de um Centro Universitário da Zona da Mata Mineira. Nas disciplinas citadas, a ferramenta foi integrada às aulas por meio da metodologia Peer Instruction (PI), na qual os alunos aprendem com seus pares. Desta forma o objetivo desta pesquisa foi investigar a utilização do software Plickers® como ferramenta para medir a efetividade do aprendizado durante as aulas de estatística, verificando sua real aplicabilidade e aceitação pelos alunos. Os dados coletados e analisados são de natureza qualitativa e quantitativa, se tratando de uma pesquisa de campo. A ferramenta foi bem aceita pelos alunos, que instantaneamente se mostraram mais interessados pelas aulas e pela nova tecnologia, como demonstram os resultados. O fato de cada aluno possuir um cartão resposta, de uso exclusivo, faz com que a omissão dos alunos, no momento de responder a uma questão proposta pelo professor seja inexistente; dessa forma, isso deixa de ser um problema na sala de aula, o professor tem, de forma rápida, o real diagnóstico de aprendizado de seus alunos, isso permite que medidas possam ser tomadas, quando o aprendizado possa ser considerado insatisfatório. A ferramenta permite a dinamicidade durante as aulas, permitindo que o professor tome decisões e caminhos que possam reverter situações insatisfatórias.

Abstract: The central theme of the article is the different learning participative methodologies, based on Bloom Revised Taxonomy, applied in many courses at the UNIFACIG University Center. The justification for the use of Taxonomy theoretical support focuses on the point of reflective, critical and creative development of students. The objective of the study is to present a systematic revision of papers published concerning to active methodologies developed at UNIFACIG, seeking out to verify the contribution of thematic development of a critical, reflective and collaborative professional and the ability to apply the learned content and also the state of the art in which the theme is applied as a didactic strategy in different courses. As a methodological design, it was opted a systematic review of studies published in different Forums conducted by Sthem-Brasil Consortium totaling 21 works. In view of analyses carried out in the various works of the institution, it is clear that application of active methodologies in teaching and learning process are preponderant aspects of a better content development. Whether using simulated practices or real experiences; communication, interaction between participants, restlessness and involvement are intrinsic parts of the process, which leads to more meaningful and lasting learning. It is noticed that the desired result was achieved, because, in general, studies show extremely positive results: a more critical, reflective and instigating university education, with more collaborative and creative students and, specially, with social and responsible conscience. El tema central del artículo son las diferentes metodologías participativas de aprendizaje, basadas en la Taxonomía Revisada de Bloom, aplicadas en diferentes cursos en el Centro Universitario UNIFACIG. La justificación para el uso de Taxonomía cómo apoyo teórico se centra en el punto de desarrollo reflexivo, crítico y creativo de los estudiantes. El objectivo del estudio se centra en presentar una revisión sistemática de los trabajos publicados sobre metodología activa desarrollados en UNIFACIG, buscando verificar la contribución del tema al desarrollo de un profesional crítico, reflexivo, colaborativo y capaz de aplicar el contenido aprendido y, también, el estado del arte en el cual el tema se aplica cómo estrategia didáctica en los diferentes cursos. Cómo diseño metodológico, se fue elegida una revisión sistemática de los estudios publicados en los diferentes Foros realizados por el Consorcio Sthem-Brasil en la totalidad de 21 obras. En vista de los análisis realizados en los distintos trabajos de la Institución, queda claro que la aplicación de las metodologías activas en el proceso de enseñanza-aprendizaje son aspectos preponderantes de un mejor desarrollo de los contenidos. Sea utilizando prácticas simuladas o experiencias reales, la comunicación, la interacción entre los participantes, la inquietud y la participación son partes intrínsecas del proceso, lo que lleva a un aprendizaje más significativo y duradero. Se observa que se logró el resultado deseado, porque, en general, los estudios muestran resultados extremadamente positivos: una educación universitaria más crítica, reflexiva y estimulante con estudiantes más colaborativos y creativos y, principalmente, con conciencia social y responsabilidad. O artigo possui como temática central as diferentes metodologias participativas de aprendizagem, fundamentadas na Taxionomia Revisada de Bloom, aplicadas em diferentes cursos do Centro Universitário UNIFACIG. A justificativa para a utilização da Taxionomia como suporte teórico centra-se no ponto do desenvolvimento reflexivo, crítico e criativo dos discentes. O objetivo do estudo centra-se em apresentar uma revisão sistemática dos trabalhados publicados sobre metodologia ativa desenvolvidos no UNIFACIG, buscando verificar a contribuição da temática ao desenvolvimento de um profissional crítico, reflexivo, colaborativo e com capacidade de aplicar o conteúdo aprendido e, também, o estado da arte em que a temática se encontra aplicada como estratégia didática nos diferentes cursos. Como desenho metodológico, optou-se por uma revisão sistemática dos estudos publicados nos diferentes Fóruns realizados pelo Consórcio Sthem-Brasil totalizando 21 artigos. Frente às análises realizadas nos diversos trabalhos da Instituição, percebe-se que a aplicação das metodologias ativas no processo ensino e aprendizagem são aspectos preponderantes de um melhor desenvolvimento dos conteúdos. Seja utilizando de práticas simuladas ou experiências reais a comunicação, a interação entre os participantes, a inquietação e o envolvimento são partes intrínsecas do processo, o que conduz a uma aprendizagem mais significativa e duradoura. Percebe-se que o resultado almejado foi alcançado, pois, de uma forma geral, os estudos apontam resultados extremamente positivos: uma formação universitária mais crítica, reflexiva e instigante com discentes mais colaborativos e criativos e, principalmente, com consciência social e responsável.

46. Bifunctional CTP:inositol-1-phosphate cytidylyltransferase/CDP-inositol: inositol-1-phosphate transferase, the key enzyme for di-myo-inositol-phosphate synthesis in several (hyper)thermophiles

Abstract: The pathway for the synthesis of di-myo-inositol-phosphate (DIP) was recently elucidated on the basis of the detection of the relevant activities in cell extracts of Archaeoglobus fulgidus and structural characterization of products by nuclear magnetic resonance (NMR) (N. Borges, L. G. Goncalves, M. V. Rodrigues, F. Siopa, R. Ventura, C. Maycock, P. Lamosa, and H. Santos, J. Bacteriol. 188:8128-8135, 2006). Here, a genomic approach was used to identify the genes involved in the synthesis of DIP. Cloning and expression in Escherichia coli of the putative genes for CTP:L-myo-inositol-1-phosphate cytidylyltransferase and DIPP (di-myo-inositol-1,3'-phosphate-1'-phosphate, a phosphorylated form of DIP) synthase from several (hyper)thermophiles (A. fulgidus, Pyrococcus furiosus, Thermococcus kodakaraensis, Aquifex aeolicus, and Rubrobacter xylanophilus) confirmed the presence of those activities in the gene products. The DIPP synthase activity was part of a bifunctional enzyme that catalyzed the condensation of CTP and L-myo-inositol-1-phosphate into CDP-L-myo-inositol, as well as the synthesis of DIPP from CDP-L-myo-inositol and L-myo-inositol-1-phosphate. The cytidylyltransferase was absolutely specific for CTP and L-myo-inositol-1-P; the DIPP synthase domain used only L-myoinositol-1-phosphate as an alcohol acceptor, but CDP-glycerol, as well as CDP-L-myo-inositol and CDP-D-myoinositol, were recognized as alcohol donors. Genome analysis showed homologous genes in all organisms known to accumulate DIP and for which genome sequences were available. In most cases, the two activities (L-myo-inositol-1-P cytidylyitransferase and DIPP synthase) were fused in a single gene product, but separate genes were predicted in Aeropyrum pernix, Thermotoga maritima, and Hyperthermus butylicus. Additionally, using L-myo-inositol-1-phosphate labeled on C-1 with carbon 13, the stereochemical configuration of all the metabolites involved in DIP synthesis was established by NMR analysis. The two inositol moieties in DIP had different stereochemical configurations, in contradiction of previous reports. The use of the designation di-myo-inositol-1,3'-phosphate is recommended to facilitate tracing individual carbon atoms through metabolic pathways.

Abstract: Since the onset of the "space revolution" of high-precision high-cadence photometry, asteroseismology has been demonstrated as a powerful tool for informing Galactic archeology investigations. The launch of the NASA Transiting Exoplanet Survey Satellite (TESS) mission has enabled seismic-based inferences to go full sky-providing a clear advantage for large ensemble studies of the different Milky Way components. Here we demonstrate its potential for investigating the Galaxy by carrying out the first asteroseismic ensemble study of red giant stars observed by TESS. We use a sample of 25 stars for which we measure their global asteroseimic observables and estimate their fundamental stellar properties, such as radius, mass, and age. Significant improvements are seen in the uncertainties of our estimates when combining seismic observables from TESS with astrometric measurements from the Gaia mission compared to when the seismology and astrometry are applied separately. Specifically, when combined we show that stellar radii can be determined to a precision of a few percent, masses to 5%-10%, and ages to the 20% level. This is comparable to the precision typically obtained using end-of-mission Kepler data. © 2020. The American Astronomical Society. All rights reserved.

Abstract: Since the onset of the "space revolution" of high-precision high-cadence photometry, asteroseismology has been demonstrated as a powerful tool for informing Galactic archeology investigations. The launch of the NASA Transiting Exoplanet Survey Satellite (TESS) mission has enabled seismic-based inferences to go full sky-providing a clear advantage for large ensemble studies of the different Milky Way components. Here we demonstrate its potential for investigating the Galaxy by carrying out the first asteroseismic ensemble study of red giant stars observed by TESS. We use a sample of 25 stars for which we measure their global asteroseimic observables and estimate their fundamental stellar properties, such as radius, mass, and age. Significant improvements are seen in the uncertainties of our estimates when combining seismic observables from TESS with astrometric measurements from the Gaia mission compared to when the seismology and astrometry are applied separately. Specifically, when combined we show that stellar radii can be determined to a precision of a few percent, masses to 5%-10%, and ages to the 20% level. This is comparable to the precision typically obtained using end-of-mission Kepler data.

49. Biosynthetic pathways of inositol and glycerol phosphodiesters used by the hyperthermophile Archaeoglobus fulgidus in stress adaptation

Abstract: Archaeoglobus fulgidus accumulates di-myo-inositol phosphate (DIP) and diglycerol phosphate (DGP) in response to heat and osmotic stresses, respectively, and the level of glycero-phospho-myo-inositol (GPI) increases primarily when the two stresses are combined. In this work, the pathways for the biosynthesis of these three compatible solutes were established based on the detection of the relevant enzymatic activities and characterization of the intermediate metabolites by nuclear magnetic resonance analysis. The synthesis of DIP proceeds from glucose-6-phosphate via four steps: (i) glucose-6-phosphate was converted into L-myo-inositol l-phosphate by L-myo-inositol 1-phosphate synthase; (ii) L-myo-inositol 1-phosphate was activated to CDP-inositol at the expense of CTP; this is the first demonstration of CDP-inositol synthesis in a biological system; (iii) CDP-inositol was coupled with L-myo-inositol 1-phosphate to yield a phosphorylated intermediate, 1,1'di-myo-inosityl phosphate 3-phosphate (DIPP); (iv) finally, DIPP was dephosphorylated into DIP by the action of a phosphatase. The synthesis of the two other polyol-phosphodiesters, DGP and GPI, proceeds via the condensation of CDP-glycerol with the respective phosphorylated polyol, glycerol 3-phosphate for DGP and L-myo-inositol 1-phosphate for GPI, yielding the respective phosphorylated intermediates, 1X, 1'X-diglyceryl phosphate 3-phosphate (DGPP) and 1-(1X-glyceryl) myo-inosityl phosphate 3-phosphate (GPIP), which are subsequently dephosphorylated to form the final products. The results disclosed here represent an important step toward the elucidation of the regulatory mechanisms underlying the differential accumulation of these compounds in response to heat and osmotic stresses.

50. Effect of pyruvate kinase overproduction on glucose metabolism of Lactococcus lactis

Abstract: Lactococcus lactis strain NZ9000(pNZpyk), which overproduces pyruvate kinase (PK), was constructed. The pNZpyk plasmid carries the [P.sub.nisA]-pyk transcriptional fusion, and the overexpression of its pyk gene was accomplished by using the nisin-inducible expression system of the NZ9000 strain. In vivo [sup.13]C- and [sup.31]P-NMR spectroscopy was used to evaluate the effect of this modification on the metabolism of glucose in non-growing cells. A detailed description of the kinetics of glucose, end products, glycolytic intermediates, NA[D.sup.+] and NADH was obtained. A 15-fold increase in the level of PK did not increase the overall glycolytic flux, which, on the contrary, was slightly reduced. Significant differences were observed in (i) the level of 3-phosphoglycerate (3-PGA) and phosphoenolpyruvate (PEP), metabolites associated with starvation; (ii) the rate of fructose 1,6-bisphosphate (FBP) depletion upon glucose exhaustion; and (iii) the NA[D.sup.+]/NADH ratio during glucose catabolism. In the mutant, the rate of FBP consumption after glucose depletion was notably accelerated under anaerobic conditions, whereas 3-PGA and PEP decreased to undetectable levels. Furthermore, the level of NA[D.sup.+] decreased steadily during the utilization of glucose, probably due to the unanticipated reduction in the lactate dehydrogenase activity in comparison with the control strain, NZ9000(pNZ8020). The results show that PK is an important bottleneck to carbon flux only when glucose becomes limiting; in the overproducer this constriction was no longer present, as evidenced by the faster FBP consumption and lack of accumulation of 3-PGA and PEP in anaerobic as well as aerobic conditions. Despite these clear changes, the PK-overproducing strain showed typical homolactic metabolism under anaerobic conditions, as did the strain harbouring the vector plasmid without the pyk insert. However, under an oxygen atmosphere, there was increased channelling of carbon to the production of acetate and acetoin, to the detriment of lactate production.
