An ESIPT based light activated hydrogen sulfide (H2S) donor using a p-hydroxyphenacyl phototrigger has been developed. The unique feature of our H2S donor system is that it provides real-time monitoring of H2S release by a non-invasive fluorescence colour change approach. [ABSTRACT FROM AUTHOR]

The design, synthesis, and in vitro biological studies of a biotin-carbazole-dicyanovinyl-chlorambucil conjugate (Bio-CBZ-DCV-CBL; 6) are reported. This conjugate ( 6) is a multifunctional single-molecule appliance composed of a thiol-sensor DCV functionality, a CBZ-derived phototrigger as well as fluorescent reporter, and CBL as the anticancer drug, and Bio as the cancer-targeting ligand. In conjugate 6, the DCV bond undergoes a thiol-ene click reaction at pH<7 with intracellular thiols, thereby shutting down internal charge transfer between the donor CBZ and acceptor DCV units, resulting in a change of the fluorescence color from green to blue, and thereby, sensing the tumor microenvironment. Molecular rotor-based fluorophores (RBFs) that are target-selective and sensitive to both polarity and viscosity are valuable for diverse biological applications. Here, we have designed next-generation RBFs based on the underexplored bimane fluorophore through either changing in aryl substitution or varying π-linkages between the rotatable electron donors and acceptors to produce red-shifted fluorescence emissions with large Stokes shifts. RBFs exhibit a twisted intramolecular charge transfer mechanism that enables control of polarity and viscosity sensitivity, as well as target selectivity. These features enable their application in: (1) turn-on fluorescent detection of α-synuclein (αS) fibrils, a hallmark of Parkinson's disease (PD), including amplified fibrils from patient samples; (2) monitoring early misfolding and oligomer formation during αS aggregation; and (3) selective imaging of αS condensates formed by liquid-liquid phase separation (LLPS). In all three cases, we show that our probes have high levels of selectivity for αS versus other aggregating proteins. Certain volatile organic compounds (VOCs), such as formaldehyde, acetone, and ethanol, are overexpressed in some terminal diseases like cancer, diabetes, Alzheimer's, etc. Therefore, high-precision detection and quantification of VOCs is imperative for early diagnosis of such detrimental diseases. Non-invasive and accurate fluorescence-based detection of such analytes has garnered widespread attention. The inherent luminescent properties of covalent organic frameworks (COFs), resulting from their extensive π-conjugation, have made them suitable for sensing applications. Structural tunability and strong covalent linkers facilitate sensing by COFs. Appropriate choices of linker and skeletal units of the COF can help detect various biologically important analytes selectively. The most common linkers used in this regard is the imine linker, which can undergo excellent hydrogen bonding with different protic VOCs e. g., ethanol, methanol, etc. Besides imine detection, hydrogen bonding also proved useful for detection of aldehydes. Suitable combinations of donors and acceptors enable the COFs to have specific charge transfer interactions with many electron-rich and electron-poor VOCs. In this review, we have highlighted the syntheses of selective COFs incorporating linkers designed for sensing cancer-inducing VOCs. One- and two-photon activated sulfur dioxide donors based on a 4,5-dimethoxy-2-nitrobenzyl phototrigger have been developed. The designed donors have the ability to release not only SO2 but also a hydroxy-compound in a simultaneous manner. Furthermore, we demonstrated their application in combinatorial therapy by the dual release of SO2 and an active drug, i.e. ferulic acid ethyl ester (FAEE) with self-monitoring ability. Next, we investigated the in vitro cellular uptake and the capability of SO2 generation from the donors using a well-known coumarin-hemicyanine fluorescent probe. Finally, we evaluated the antibacterial activity of the designed donors (5a, 5b and 6) by broth dilution and agar well diffusion methods on E. cloacae cells (MTCC 509). The results show that the donor 5a exhibits enhanced antibacterial efficacy compared to 5b and 6, due to the synergetic effect of dually released SO2 and FAEE.

The direct hydroxylation of 2-arylpyridines and 2-arylbenzothiazoles via the merger of organic photoredox and metal catalysis is reported where 4CzIPN is used as the visible-light photocatalyst and Pd(OAc) 2 as the metal catalyst. This method has been employed to synthesize organic molecules exhibiting excited-state intramolecular proton transfer properties for generating tunable luminescence responses.
