Abstract Migraine is a debilitating neurological disorder impacting millions worldwide. Calcitonin Gene-Related Peptide (CGRP) has emerged as a key player in migraine pathophysiology, leading to the development of targeted therapies. This study reviews novel CGRP-targeted treatments, including monoclonal antibodies small molecule inhibitors/nutraceuticals and introduces Asarinin as a potential modulator of the pathway. Asarinin, a natural compound found in various plants, is examined for its pharmacological potential in migraine management. Pharmacokinetic assessments, toxicological modelling, molecular property analysis, and network pharmacology were conducted. Molecular docking and dynamics studies with CGRP reveal potential interactions, providing a foundation for understanding Asarinin's therapeutic effects. Asarinin's favourable pharmacokinetics, safety profile, and bioactivity, supporting its candidacy as a therapeutic agent. In-depth molecular docking studies with the CGRP receptor (PDB: 6ZHO) demonstrate strong binding affinity (− 10.3kcal/mol), while molecular dynamics simulations unveil the dynamic behavior of the Asarinin-CGRP complex, (− 10.53 kcal/mol) for Atogepant-CGRP complex. Network analysis highlights key proteins in migraine pathology, indicating Asarinin's potential efficacy. The groundwork for future investigations, suggests Asarinin as a promising candidate for migraine management by targeting OPRM1 pathway. The integration of diverse assessments provides a comprehensive understanding of Asarinin's potential and paves the way for further preclinical and clinical research.

The hepatic manifestation of metabolic syndrome, associated with various metabolic diseases such as type 2 diabetes, insulin resistance, and high cholesterol, is called non-alcoholic fatty liver disease (NAFLD). Despite several research efforts, no approved medicine is currently available for the treatment of this illness. Swiss Target Prediction was used to screen phytochemicals. To examine potential targets, the protein-protein interaction (PPI) network was developed. Cytoscape was used to create the component-target-pathway (C-T-P) network, and AutoDock was used to assess molecular docking. Antioxidant and anti-inflammatory qualities were tested in vitro. Naringenin, ellagic acid, and cyanidin were found to be the main active components. As important targets, PPARA, PPARG, and AKT1 were selected. Through enrichment analysis, a total of 20 crucial signaling pathways, including insulin resistance (IR), NAFLD, relaxin, PI3K-Akt, HIF-1, AGE-RAGE, and MAPK, were identified. The in silico computational techniques predicted the molecular pathway for the active ingredients and the disease targets, thus helping to further research.

BackgroundMigraines affect one billion individuals globally, with a higher occurrence among young adults and women. A significant survey in the United States indicated that 17.1% of women and 5.6% of men suffer from migraines. This study seeks to investigate the potential connection between NLRP3 and MMP9 in migraine pathology.MethodsThe research involved searching databases such as PubMed, Scopus, Science Direct, Google Scholar, and Proquest, with the search concluding on March 31, 2024. Following PRISMA guidelines, PICO data were collected, focusing exclusively on animal models induced by Nitroglycerine (10 mg/kg), while excluding clinical studies.ResultsThe study, originally registered in Prospero Reg. No. CRD42022355893, conducted bias analysis using SYRCLE’s RoB tool and evaluated author consensus using GraphPad v9.5.1. Out of 7,359 search results, 22 papers met the inclusion criteria. Inter-rater reliability among reviewers was assessed using Cohen’s kappa statistics.ConclusionThis review summarizes 22 preclinical studies on Nitroglycerin (NTG), NLRP3, MMP9, and related biomarkers in migraine. Background: Non-healing diabetic wounds represent a significant clinical challenge globally, necessitating innovative approaches in drug delivery to enhance wound healing. Understanding the pathogenesis of these wounds is crucial for developing effective treatments. Bioactive dressings and polymeric nanofibers have emerged as promising modalities, with silk biomaterials gaining attention for their unique properties in diabetic wound healing. Purpose of Review: The purpose of this review is to examine the challenges and innovations in treating non-healing diabetic wounds, emphasizing the global burden and the need for effective solutions. This review explores the complex mechanisms of wound healing in diabetes and evaluates the therapeutic potential of bioactive dressings and polymeric nanofibers. Special focus is given to the application of silk biomaterials, particularly silk fibroin, for wound healing, detailing their properties, mechanisms, and clinical translation. This review also describes various nanofiber fabrication methods, especially electrospinning technology, and presents existing evidence on the effectiveness of electrospun silk fibroin formulations. Recent Findings: Recent advancements highlight the potential of silk biomaterials in diabetic wound healing, owing to their biocompatibility, mechanical strength, and controlled drug release properties. Electrospun silk fibroin-based formulations have shown promising results in preclinical and clinical studies, demonstrating accelerated wound closure and tissue regeneration. Summary: Non-healing diabetic wounds present a significant healthcare burden globally, necessitating innovative therapeutic strategies. Bioactive dressings and polymeric nanofibers, particularly silk-based formulations fabricated through electrospinning, offer promising avenues for enhancing diabetic wound healing. Background: Polycystic ovarian syndrome is a metabolic disorder majorly caused by the hormonal fluctuations in female and the current scenario explains that adolescents remain predominantly affected with this disorder. Bauhinia variegata assist in the treatment of various ailments and are used as an ingredient in targeting uterine disorders, yet the exact constituent that contributes on activity remains unknown. Aim: The main aim of our study is to determine the potent phytoconstituent of Bauhinia variegata to fight against the symptoms of PCOS through computational techniques. Materials and Methods: Five in silico techniques like Molecular docking analysis, Pharmacokinetics, toxicity prediction of the compounds, Biological activity and Molecular dynamics simulation studies were performed to identify the potent phytoconstituent. Results and Discussion: Molecular docking studies show that the major constituent lupeol had a good binding interaction and high docking score of -10.31 Kcal/mol and -11.52 Kcal/mol with both the proteins 3RUK and 1E3K. Pharmacokinetics, toxicity and biological activity studies reveal that it had ideal drug likeliness properties with proper biological activity values and were found to non-toxic in the analysed parameters. Lupeol complex was found to be potentially stable throughout the molecular dynamic’s simulation for 100 ns. Conclusion: Thus, through in silico analysis it is evident that from the list of phytoconstituents of Bauhinia variegata, lupeol possess potent activities in mitigating the symptoms of PCOS.
