The main function of the mammalian skin is to protect the organism against various deleterious environmental factors such as UVR. The skin contains specialized pigment producing cells, known as melanocytes, to protect against UVR. However, when melanocytes acquire mutations from excessive exposure to UVR or other factors, they transform into melanoma. Melanoma is the deadliest type of skin cancer that invades neighboring tissue during early tumor development. The BRAF oncogene is mutated in over 50% of human melanoma cases and therapies that target the BRAF signaling are effective at inducing tumor regression. Unfortunately, melanoma tumors acquire resistance and become unresponsive to BRAF therapy because of alterations in various signaling networks. Genes and pathways that allow cells to cope with oncogene-induced stress represent selective cancer therapeutic targets that remain largely undiscovered. In addition, melanomas accumulate a high burden of mutations that could potentially generate neoantigens, yet somehow suppress the immune response to facilitate continued growth. In order to better understand the signaling networks that drive BRAF mutant melanoma, we identified a RhoJ signaling pathway that is a selective therapeutic target for BRAF mutant tumors. RhoJ is a small GTPase that cycles between an active-GTP bound state and an inactive-GDP bound state that regulates various physiological mechanisms. Previously we demonstrated that RhoJ is involved in chemoresistance in melanoma and in suppressing ATR activity. Using an autochthonous mouse model of melanoma, we determined that loss of RhoJ significantly prolonged the life of mice bearing BRAFV600E mutant melanomas by delaying tumor initiation and reducing metastasis. Transcriptome analysis revealed that RhoJ deletion in BRAF mutant tumors modulates the expression of the pro-apoptotic protein BAD as well as genes involved in cellular metabolism, generating tumors that grow more slowly and metastasize less avidly. PAK inhibitors induce apoptosis in melanoma cells in vitro via a BAD-dependent mechanism and induce the regression of BRAF mutant melanoma tumors in vivo. We also demonstrate that ATR mutant tumors accumulate multiple mutations and alter the immune system by decreasing T-cells to encourage tumor growth. Taken together, these studies identify the RhoJ-BAD signaling network as a therapeutic vulnerability for BRAF mutant tumors. Additionally, these studies also identify a novel mechanism by which melanoma cells modulate the tumor microenvironment to promote survival.

Maestría en Estadística Aplicada El propósito principal de la investigación fue comprobar si el rendimiento académico de los estudiantes en el curso de Estadística General de la Universidad Nacional Agraria La Molina (UNALM) es explicado a través de un modelo propuesto de Ecuación Estructural de tres factores. El primero denominado “desempeño docente”, medido por las variables: planificación del curso, dominio del curso, métodos y recursos de instrucción, obligaciones docentes, método evaluativo, y motivación e interacción con los alumnos; el segundo llamado “autoconcepto”, medido por las variables: académico/laboral, social, emocional, familiar y físico y finalmente el tercero “rendimiento pasado”, medido a través del promedio ponderado acumulado. Los datos utilizados corresponden a las notas de los alumnos matriculados en el ciclo académico 2014-I en el curso de Estadística General, al promedio ponderado acumulado; y los valores se registraron en la escala de Likert de 1 al 10 de las encuestas de desempeño docente y autoconcepto. Estos dos instrumentos, cumplen con los requisitos de confiabilidad y validez al registrar en ambos casos indicadores por encima de los mínimos aceptables. El modelo de ecuación estructural propuesto fue reespecificado (mejorado) mediante la inclusión de una nueva relación de interdependencia, el rendimiento pasado como predictor del autoconcepto. Se verificó el ajuste del modelo de ecuación estructural reespecificado a través de los principales indicadores de ajuste absoluto e incremental. Entre los resultados más importantes de la investigación se verificó que el factor rendimiento pasado es el mejor predictor del factor rendimiento académico de los estudiantes en el curso de Estadística General y que los factores desempeño docente y rendimiento pasado explican al factor autoconcepto. The principal goal of this research is if the academic performance score into general statistics course of the National Agrarian University La Molina (UNALM) is explained through Structural Equation Modeling with three factors. The first factor professor´s performance has measured by variables like: Plan targeted lessons, course´s knowledge, instructions ´s method, the duties of professor, the evaluate criteria, teacher's motivation and interactions in class. The second factor called self concept was measured by: academic/occupational, social, emotional, family and, physical health. Finally, the three factor is "last performance score" was measured by cumulative grade point average. Datasets are student's score, who have studied 2014-I academic semester in statistics course. The registered values was likert scale from 1 to 10 through the surveys of the professor´s performance and, self concept. These two instruments have followed with requirements like reliability and validity when the indicator have been showing highest values over minimal acceptable. Structural equation model was improved by respecification of structural model through a new interdependent relationship, last performance score was predictor of the self concept. Respecification of structural model was proved by principal indicators of absolute index and incremental. The main results of this investigation proved that last performance score is the best predictor of the students performance score in statistics course so, professor´s performance and last performance score have explained the self concept factor. The increased activity of aquaculture in Chile involves cultivation of salmonids, oysters mussels and other species such, and to a lesser extent species such as red abalone (Haliotis rufescens) and Japanese abalone (Haliotis discus hannai). The aim of this study was to evaluate the growth rate of Haliotis rufescens and Haliotis discus hannai fed with different pellet based diets with Macrocystis sp. and Ulva sp., grown in ponds for 13 months. The results for both species denoted that there was an increase in length and biomass during experimental period, existing low growth rates during the austral winter (July-September) and increase during the austral summer (December-January). Results are consistent with descriptions of literature that there is high rate of growth during the summer and using diet of brown algae. From the economic standpoint abalone farming would be an economically viable activity for local aquaculture, considering the water quality and food requirements. El incremento de las actividades de acuicultura en Chile involucra principalmente cultivo de salmónidos, ostiones, algas, mitílidos y otras especies, y en menor escala especies como los abalones rojo (Haliotis rufescens) y japonés (Haliotis discus hannai). El objetivo del presente estudio fue evaluar el crecimiento de Haliotis rufescens y Haliotis discus hannai alimentados con diferentes dietas a base de pellet Macrocystis sp. y Ulva sp., cultivados en estanques durante 13 meses. Los resultados para ambas especies denotaron que hubo incremento en longitud y biomasa durante el periodo experimental, existiendo bajas tasas de crecimiento durante invierno austral (julio-septiembre) e incremento durante el verano austral (diciembre-enero). Los resultados concuerdan con las descripciones de la literatura en que hay altas tasas de crecimiento durante el verano usando dieta a base de algas pardas. Desde el punto de vista económico el cultivo de abalones sería una actividad económicamente viable para la acuicultura local, si se considera los requerimientos de calidad de agua y alimentación.

Actualmente la tendencia de la acuicultura mundial está orientada hacia la diversificación de los cultivos, principalmente de especies nativas. El congrio colorado Genypterus chilensis es un pez nativo de alta demanda gastronómica y explotación estacional que lo proyecta como candidato para el desarrollo de su tecnología de cultivo. El objetivo de este trabajo fue evaluar la eficiencia de dos sistemas de incubación de masas de huevos de G. chilensis, uno con circuito cerrado de agua (SICC) y el otro con circuito abierto (SICA); su eficiencia fue medida por el porcentaje de eclosión de huevos. Dos ensayos fueron realizados midiendo y comparando los porcentajes de fecundación y eclosión de huevos en cuatro réplicas entre los dos sistemas, encontrándose solo diferencias significativas entre los porcentajes de fecundación del bioensayo 2. El bioensayo 1 tuvo un 81% promedio de fecundación de los huevos y el porcentaje promedio de eclosión para el SICC fue 42,9 ± 34,5% y para el SICA fue 0,0 ± 0,0%. El bioensayo 2 tuvo un porcentaje promedio de fecundación de los huevos de 87,3 ± 2,6% para el SICC y 79,8 ± 3,2% para el SICA y el porcentaje promedio de eclosión para el SICC fue 27,9 ± 33,7% y para el SICA fue 4,8 ± 5,6%. Se discuten los parámetros de incubación para obtener una máxima eclosión y se entrega una proposición para mejorar el sistema SICC. El sistema de incubación con circuito cerrado de agua generó mayores sobrevivencias en los huevos de G. chilensis. The current trend in world aquaculture is towards the diversification of cultures, mainly native species. The red cusk eel Genypterus chilensis is a native Chilean species of high gastronomic demand and seasonal exploitation that is projected as a candidate for the development of farming technology. The objective of this study was to test the efficiency of two incubation systems for G. chilensis egg masses, one with a closed water circuit (SICC) and the other with an open water circuit (SICA). Efficiency was measured by the percentage of eggs hatched. Two assays were performed, measuring and comparing the percentages of egg fecundation and hatching in four replicates between the two incubation systems. Significant differences were only found between the percentages of fecundation of bioassay 2. On average, in bioassay 1, egg fecundation reached 81%, and the average of hatching for the SICC was 42.9 ± 34.5% and for the SICA was 0.0 ± 0.0%. Bioassay 2 had an average fecundation of 87.3 ± 2.6% for the SICC and 79.8 ± 3.2% for the SICA, and the average for hatching was 27.9 ± 33.7% for the SICC and 4.8 ± 5.6% for the SICA. Incubation parameters for maximum hatching were discussed and a proposal was made for improving the SICC system. The incubation system with a closed water circuit generated greater survival of G. chilensis eggs.
