The prevalence of proximate risk factors for active tuberculosis (TB) in areas of high prevalence of latent tuberculosis infection (LTBI) is not clearly understood. We aimed at assessing the prevalence of non-communicable multi-morbidity focusing on diabetes mellitus (DM), malnutrition, and hypertension (HTN) as common risk factors of LTBI progressing to active TB. In a cross-sectional study, 2,351 adults (45% male and 55% female) from villages in the Kancheepuram district of South India were enrolled between 2013 and 2020. DM was defined as HbA1c >6.4%, undernutrition was defined as low body mass index (LBMI) 130 mmHg, and LTBI was defined as positive (≥ 0.35 international units/ml) by QuantiFERON Gold In-Tube assay. A total of 1,226 individuals (52%) were positive for LTBI out of 2351 tested individuals. The prevalence of DM and pre-diabetes mellitus (PDM) was 21 and 35%, respectively, HTN was 15% in latent tuberculosis (LTB)-infected individuals. The association of DM [odds ratio (OR)]; adjusted odds ratio (aOR) (OR = 1.26, 95% CI: 1.13–1.65; aOR = 1.19, 95% CI: 1.10–1.58), PDM (OR = 1.11, 95% CI: 1.0–1.35), and HTN (OR = 1.28, 95% CI: 1.11–1.62; aOR = 1.18, 95% CI: 1.0–1.56) poses as risk factors of LTBI progression to active TB. The prevalence of LBMI 9% (OR = 1.07, 95% CI: 0.78–1.48) and obesity 42% (OR = 0.85, 95% CI: 0.70–1.03) did not show any statistically significant association with LTB-infected individuals. The present evidence of a high burden of multi-morbidity suggests that proximate risk factors of active TB in LTBI can be managed by nutrition and lifestyle modification.

BackgroundThe prevalence of Strongyloides stercoralis infection is estimated to be 30-100 million worldwide, although this an underestimate. Most cases remain undiagnosed due to the asymptomatic nature of the infection. We wanted to estimate the seroprevalence of S. stercoralis infection in a South Indian adult population.MethodsTo this end, we performed community-based screening of 2351 individuals (aged 18-65) in Kanchipuram District of Tamil Nadu between 2013 and 2020. Serological testing for S. stercoralis was performed using the NIE ELISA.ResultsOur data shows a seroprevalence of 33% (768/2351) for S. stercoralis infection which had a higher prevalence among males 36% (386/1069) than among females 29.8% (382/1282). Adults aged ≥55 (aOR = 1.65, 95% CI: 1.25-2.18) showed higher adjusted odds of association compared with other age groups. Eosinophil levels (39%) (aOR = 1.43, 95% CI: 1.19-1.74) and hemoglobin levels (24%) (aOR = 1.25, 95% CI: 1.11-1.53) were significantly associated with S. stercoralis infection. In contrast, low BMI (aOR = 1.15, 95% CI: 0.82-1.61) or the presence of diabetes mellitus (OR = 1.18, 95% CI: 0.83-1.69) was not associated with S. stercoralis seropositivity.ConclusionsOur study provides evidence for a very high baseline prevalence of S. stercoralis infection in South Indian communities and this information could provide realistic and concrete planning of control measures.

Abstract Background In order to increase understanding of how infused cells work, it becomes important to track their initial movement, localization, and engraftment efficiency following transplantation. However, the available in vivo cell tracking techniques are suboptimal. The study objective was to determine the biodistribution of intravenously administered Indium-111 (In-111) oxine labeled human umbilical tissue-derived cells (hUTC) in a rat model of transient middle cerebral occlusion (tMCAo) using single photon emission computed tomography (SPECT). Methods Rats received 3 million In-111 labeled hUTC (i.v.) 48 hrs after tMCAo. Following the administration of either hUTC or equivalent dose of In-111-oxine (18.5 MBq), animals underwent SPECT imaging on days 0, 1, and 3. Radioactivity in various organs as well as in the stroke area and contralateral hemisphere was determined, decay corrected and normalized to the total (whole body including head) radioactivity on day 0. Immunohistochemical analysis was also performed to confirm the beneficial effects of hUTC on vascular and synaptic density, and apoptosis. Results Most of the radioactivity (43.36±23.07% on day 0) trafficked to the lungs immediately following IV administration of In-111 labeled hUTC (day 0) and decreased drastically to 8.81±7.75 and 4.01±4.52% on days 1 and 3 post-injection, respectively. In contrast, radioactivity measured in the lung of animals that received In-111-oxine alone remained relatively unchanged from day 0 to day 1 (18.38±5.45% at day 0 to 12.59±5.94%) and decreased to 8.34±4.25% on day 3. Significantly higher radioactivity was observed in stroke areas of animals that received In-111 labeled hUTC indicating the presence of cells at the site of injury representing approximately 1% of total administered dose. In addition, there was significant increase in vascular and synaptophysin immunoreactivity in stroke areas of rats that received In-111 labeled hUTC. Conclusions The present studies showed the tracking of In-111 labeled hUTC to the sites of stroke in a rat model of tMCAo using SPECT. Animals treated with In-111 labeled hUTC showed histological improvements, with higher vascular and synaptic densities observed in the ischemic boundary zone (IBZ).

Abstract Background The current method to determine the efficacy of chemoprevention in TRAMP mouse model of carcinoma of prostate (CaP) is by extracting and weighing the prostate at different time points or by immunohistochemistry analysis. Non-invasive determination of volumes of prostate glands and seminal vesicles before, during and after treatment would be valuable in investigating the efficacy of newer chemopreventive agents in CaP. The purpose of this study was to determine whether in vivo magnetic resonance imaging (MRI) using a 3 tesla clinical MRI system can be used to follow the effect of chemoprevention in TRAMP model of mouse CaP. Methods Mice were randomized into control and treated groups. The animals in treated group received 10 µmol/kg of CDDO, 5 days a week for 20 weeks. Animals underwent in vivo MRI of prostate gland and seminal vesicles by a clinical 3 Tesla MRI system just before (at 5 weeks), during and at the end of treatment, at 25 weeks. T1-weighted and fat saturation (FATSAT) multiecho fast spin echo T2- weighted images (T2WI) were acquired. Volume of the prostate glands and seminal vesicles was determined from MR images. T2 signal intensity changes in the seminal vesicles were determined by subtracting higher echo time (TE) from lower TE T2WI. Following treatments all animals were sacrificed, prostate and seminal vesicles collected, and the tissues prepared for histological staining. All data were expressed as mean ± 1 standard deviation. Two-way or multivariate analysis of variance followed by post-hoc test was applied to determine the significant differences. A p-value of Results Histological analysis indicated tumor in 100% of control mice, whereas 10% of the treated mice showed tumor in prostate gland. Both MRI and measured prostate weights showed higher volume/weight in control mouse group. MRI showed significantly higher volume of seminal vesicles in control animals and T2 signal intensity changes in seminal vesicles of control mice indicating higher number of tumor foci, which was also proven by histology. Conclusions In vivo MRI is helpful in determining the efficacy of chemoprevention of prostate cancer in TRAMP mice.

This research paper focused on investigating the influence of various factors on steady Couette flow beneath a permeable bed. Factors studied included aligned magnetic field, thermal radiation, temperature gradient heat source, viscous dissipation, and Joules dissipation. The momentum and energy equations governing flow and heat transmission were analytically solved to predict the generation of entropy. The findings were visually presented through graphical representations, which effectively demonstrated the effect of these factors on the fluid's velocity, temperature, entropy production, and Bejan number. Summary: To increase rice yields and feed billions of people, it is essential to enhance genetic gains. However, the development of new varieties is hindered by longer generation times and seasonal constraints. To address these limitations, a speed breeding facility has been established and a robust speed breeding protocol, SpeedFlower is developed that allows growing 4–5 generations of indica and/or japonica rice in a year. Our findings reveal that a high red‐to‐blue (2R > 1B) spectrum ratio, followed by green, yellow and far‐red (FR) light, along with a 24‐h long day (LD) photoperiod for the initial 15 days of the vegetative phase, facilitated early flowering. This is further enhanced by 10‐h short day (SD) photoperiod in the later stage and day and night temperatures of 32/30 °C, along with 65% humidity facilitated early flowering ranging from 52 to 60 days at high light intensity (800 μmol m−2 s−1). Additionally, the use of prematurely harvested seeds and gibberellic acid treatment reduced the maturity duration by 50%. Further, SpeedFlower was validated on a diverse subset of 198 rice accessions from 3K RGP panel encompassing all 12 distinct groups of Oryza sativa L. classes. Our results confirmed that using SpeedFlower one generation can be achieved within 58–71 days resulting in 5.1–6.3 generations per year across the 12 sub‐groups. This breakthrough enables us to enhance genetic gain, which could feed half of the world's population dependent on rice. We've developed a robust SpeedFlower protocol for rapid rice breeding, enabling 4–5 generations of indica and/or japonica rice annually. The SpeedFlower protocol was validated on 198 diverse rice accessions, representing all 12 sub‐groups of Oryza sativa L. With a timeframe of 58–71 days per generation, we achieved 5.1–6.3 generations per year. Introduction: Success in endodontics depends on canal preparation, disinfection, and obturation. Cleaning and shaping play a pivotal role in successful endodontic therapy. An important factor in preventing procedural mishaps and further complications is the quantity of dentine removed during instrumentation because the fracture resistance of the root is directly influenced by the amount of dentine that remains after instrumentation. Aim: To evaluate and compare the remaining dentine thickness of the root canal after instrumentation with three different rotary file systems using Cone Beam Computed Tomography (CBCT) imaging. Materials and Methods: Thirty permanent extracted, singlerooted teeth were collected. The samples were decoronated at the level of the Cementoenamel Junction (CEJ). A preinstrumentation CBCT scan was taken. Access opening and working length were determined. The samples were randomly assigned to three groups: Group I - Edge Endo X7, Group II-Hero Gold, Group III - Neo-endo flex files. Biomechanical preparation was carried out using the respective files. To evaluate the remaining dentine thickness, a comparison was made with the pre-instrumentation CBCT scan. Statistical analysis was performed using One-way ANOVA. Results: Upon inter-group comparison of the pre-instrumentation group, no significant difference (p=0.05) was observed. Postinstrumentation, it was noted that the remaining dentine thickness in Group I (p=0.05) at the apical third was significantly less than in Group II (p=0.007) and Group III (p=0.042). Maximum preservation of dentine was seen with Hero gold files, whereas the minimum preservation of dentine was seen with Edge Endo X7. Conclusion: Hero gold files exhibited less removal of dentine apically, whereas Edge Endo X7 showed more removal of dentine apically. [ABSTRACT FROM AUTHOR]

Aim and objective: The aim and objective of present study are to prepare and characterize Pluronic F-127 coated colloidal silver nanoparticles and to evaluate its cytocompatibility and anti-microbial efficacy in comparison to 5.25% NaOCl. Materials and Methodology: The cytocompatibility was evaluated on L929 human fibroblast cell line by MTT assay at five different concentrations of Pluronic F-127 coated colloidal silver nanoparticles [0.01μg/mL; 0.1μg/mL; 1μg/mL; 10μg/mL; 100μg/mL] and antimicrobial activity was evaluated by Agar well diffusion method on Enterococcus faecalis (ATCC 29212) using 100μL Pluronic F-127 coated colloidal silver nanoparticles [as test solution], 100μL 5.25%NaOCl [as standard solution], 100μL water [as control]. Statistical Analysis: Statistical analysis was performed using one way analysis of variance (ANOVA), followed by Dunnett's multiple comparison test (Graph Pad Prism, Version 6, Graph Pad Software Inc., La Jolla, USA). Results: The values with p < 0.05 were considered significant. MTT assay showed Pluronic F-127 coated colloidal silver nanoparticles are more cytocompatible on healthy fibroblast cells. In antimicrobial activity there is more significant difference among groups. Conclusion: Pluronic F-127 coated colloidal silver nanoparticles showed more zone of inhibition compared to NaOCl. Pluronic F-127 coated colloidal silver nanoparticles proved to be more cytocompatible and has highest antimicrobial activity compared to NaOCl.

We report on a new straightforward magnetic celllabeling approach that combines three US Food and Drug Administration (FDA)-approved drugs--ferumoxytol, heparin and protamine--in serum-free medium to form self-assembling nanocomplexes that effectively label cells for in vivo magnetic resonance imaging (MRI). We observed that the ferumoxytol-heparin-protamine (HPF) nanocomplexes were stable in serum-free cell culture medium. HPF nanocomplexes show a threefold increase in T2 relaxivity compared to ferumoxytol. Electron microscopy showed internalized HPF in endosomes, which we confirmed by Prussian blue staining of labeled cells. There was no longterm effect or toxicity on cellular physiology or function of HPF-labeled hematopoietic stem cells, bone marrow stromal cells, neural stem cells or T cells when compared to controls. In vivo MRI detected 1,000 HPF-labeled cells implanted in rat brains. Introduction: The ultrasound-guided Transversus Abdominis Plane (TAP) block is an analgesic technique which involves injection of a local anesthetic between the internal oblique abdominal and the transversus abdominis muscle planes. It provides analgesia to the cranial branches of T10-L1 nerve roots. It has been a practice to provide analgesia in patients following various surgical procedures including the gynecological procedures like hysterectomy and caesarean section. Aim: To assess the analgesic efficacy of dexamethasone and dexmedetomidine as an adjuvant to 2% lignocaine adrenaline and 0.5% bupivacaine in TAP block following caesarean delivery. Materials and Methods: This was a cross-sectional study conducted at Konaseema Institute of Medical Sciences and Research Foundation (KIMS&RF), Amalapuram, Andhra Pradesh, India for a period of one year from January 2021 to December 2021. One hundred patients with an American Society of Anaesthesiologists (ASA) physical scores of I-II, who underwent caesarean section under the Pfannenstiel incision method under subarachnoid anaesthesia with 0.5% heavy bupivacaine, were enrolled in the study. Group I consisted of patients that received an ultrasound-guided bilateral TAP block immediately following surgery with 10 mL of 0.5% bupivacaine, 10 mL of 2% lignocaine with adrenaline, and 20 mcg dexmedetomidine. Group II included patients who received ultrasound-guided bilateral TAP block immediately following surgery with 10 mL of 0.5% bupivacaine, 10 mL of 2% lignocaine adrenaline, and 8 mg dexamethasone. The Electrocardiogram (ECG) recordings and blood pressure were recorded during the block in each patient. This approach enabled a more precise analgesic approach for each individual patient. The patients were assessed for pain up to 12 hours of surgery based on a Visual Analog Scale (VAS), where 0 represented no pain and 10 represented 'the worst pain ever possible. Results: The patients who received ultrasound-guided TAP block with dexmedetomidine were significantly less on-demand of tramadol (p-value=0.005) and had significantly lower VAS scores at 3 hours (p-value=0,000014), 6 hours (p=0.015) and 12 hours (p-value=0.006), postoperatively. Conclusion: This study proved that dexmedetomidine was more effective when compared to dexamethasone, when added as an adjuvant to 2% lignocaine adrenaline and 0.5% bupivacaine.
