Abstract

A study to verify if previous results on the haplotype frequencies of normal individuals are confirmed for beta(super 0)39-thalassemia carriers and to analyze the phylogenetic position of Sardinians among the Mediterranean population using the haplotype frequencies associated with mutations in the beta-globin genes is conducted. It concluded that the haplotype frequencies of the globin cluster in heterozygotes carrying the beta(super 0) mutation have underlined the genetic distinctiveness.

Abstract

An informative set of biallelic polymorphisms was used to study the structure of Y-chromosome variability in a sample from the Mediterranean islands of Corsica and Sicily, and compared with data on Sardinia to gain insights into the ethnogenesis of these island populations. The results were interpreted in a broader Mediterranean context by including in the analysis neighboring populations previously studied with the same methodology. All samples studied were enclosed in the comparable spectrum of European Y-chromosome variability. Pronounced differences were observed between the islands as well as in the percentages of haplotypes previously shown to have distinctive patterns of continental phylogeography. Approximately 60% of the Sicilian haplotypes are also prevalent in Southern Italy and Greece. Conversely, the Corsican sample had elevated levels of alternative haplotypes common in Northern Italy. Sardinia showed a haplotype ratio similar to that observed in Corsica, but with a remarkable difference in the presence of a lineage defined by marker M26, which approaches 35% in Sardinia but seems absent in Corsica. Although geographically adjacent, the data suggest different colonization histories and a minimal amount of recent gene flow between them. Our results identify possible ancestral continental sources of the various island populations and underscore the influence of founder effect and genetic drill The Y-chromosome data are consistent with comparable mtDNA data at the RFLP haplogroup level of resolution, as well as linguistic and historic knowledge. Abstract

Distribution of seven plasma proteins and nine red cell enzymes has been studied in a sample of unrelated persons from Corsica for the purpose of studying genetic structure of the area. Additionally, it was a goal to gain knowledge of microgeographic variability of polymorphisms in Corsica. Some differentiation was found and there was genetic heterogeneity between Corsica and other European/Mediterranean groups. Genetic differences look to be smaller between Corsicans and Sardinians than between Corsicans and other populations for which data are available.

Abstract

International audience; Volatile components of Teucrium marum from Corsica were investigated using a combination of analytical techniques. Analysis of essential oils obtained by hydrodistillation was performed using capillary GC-RI, GC-MS and 13C NMR spectroscopy after fractionation on column chromatography. The emitted volatile fractions from full aerial parts and separated organs of fresh T. marum were investigated using HS-SPME/GC-RI and HS-SPME/GC-MS. The intra-species variations of the chemical compositions of essential oils and volatile fractions from 25 Corsican sample locations were investigated using statistical analysis. The main essential oil components were caryophyllene oxide (0.4-20.5%), (E)-b-caryophyllene (0.9-18.9%), (E)-a-bergamotene (1.0-21.5%), (3Z,6E,10E)-a-springene (1.1-17.8%), b-bisabolene (0.4-14.7%), b-sesquiphellandrene (0.9-10.9%) and estragole (0.5-7.0%), while dolichodial (20.0-83.4%), (E)-a-bergamotene (0.2-20.6%), (E)-b-caryophyllene (0.3-23.1%), dolicholactone (0.4-20.9%), b-bisabolene (0.2-10.0%), estragole (1.1-10.5%) and b-sesquiphellandrene (0.2-8.3%)were the main volatiles emitted by the plant. The 13C-NMR data of (3Z,6E,10E)-a-springene and its occurrence in T. marum essential oils are reported for the first time. The importance of the sample preparation step is discussed through the absence/occurrence of dolichodial in both liquid and gaseous matrices studied, respectively.

Abstract

International audience; C. creticus subsp. eriocephalus and C. creticus subsp. creticus from the Corsican-Sardinian continuum were differentiated by botanical characteristics (absence or presence of glandular trichomes) linked with essential oil production and composition of the volatile fraction (limonene or 13-epi-manoyl oxide, respectively, as the major component). Moreover, data analysis of the HS-SPME volatile fraction confirms the clustering observed using polymorphism of the 65 ISSR loci. The Mantel test also confirms this result and shows a significant correlation (r = 0.9215, p < 0.020) between the two matrices (volatile composition and ISSR markers). These results indicate that the observed chemical variability was mainly due to genetic diversity. The concordance between the two data sets is in accordance with the findings from other studies using terpenic and genetic markers [1, 2, 3 and 4]. These data suggest that the basis of variation in the volatile composition of the two C. creticus subspecies depends more on the genetic background and less on geographical factors. In the two examined subspecies of C. creticus, the distribution of leaf trichomes and the difference in essential oil production may be considered distinctive characters at the subspecies level. This is consistent with the taxonomic classification. The HS-SPME volatile fractions of samples were allotted to two main groups; the first group (C. creticus subsp. corsicus) had a high content of components belonging to the labdane biosynthetic pathway, while the second (C. creticus subsp. eriocephalus) was characterized by limonene and nonterpenic compounds. In addition, the groups deducted from the volatile fraction composition were remarkably congruent with the groups inferred from genetic data obtained from ISSR markers. Such information would be useful for the development of conservation programs and for the selection of populations in order to consider a potential utilization of these oils.

Abstract

International audience; The chemical composition of fatty acids and unsaponifiable fraction of leaves, stems and roots of Arbutus unedo L. were determined using gas chromatography and gas chromatography-Mass Spectrometry. Then fatty acid fractions of the leaves, stems and roots contained 38.5%, 31.32% and 14.1% palmitic acid respectively, along with other long chain fatty acids (up to C22). The chemical composition of the unsaponifiable fractions differed: the leaf and stem fractions contained high levels of aliphatic (32.1% and 62.6%, respectively) and terpenic compounds (49.6% and 25.7%, respectively), and the root fraction mainly contained esters, of which the most aboundant was benzyl cinnamate (36.6%). The antimicrobial activities of the unsaponifiable fractions against nine species of microorganismes were assessed. The unsaponifiable leaf and stem extracts inhibited the growth of Klebsiella pneumoniae, Enteroccocus faecalis and Candida albicans.

International audience; Corsica and Sardinia Islands are considered as a biodiversity hotspot and provides a model system to address the impact of long-term isolation on genetic diversity and gene flow. The scope of our study, based on two cpDNA noncoding sequences, is to elucidate the evolution of Cistus creticus diversity in both Islands. The populations of C. creticus are separated into two distinct groups. The ancestral origin of these groups appears to be located in Central Corsica. The low gene flow and significant genetic diversity between these two groups seems to be correlated with geological events that have characterized these two islands.

Abstract

International audience; Cistus creticus L., an aromatic species from the Mediterranean area, contains various diterpenes bearing the labdane skeleton. The production of essential oil from this species has potential economic value, but so far, it has not been optimized. In order to contribute to a better knowledge of this species and to its differentiation, the morphological characters, volatile chemical composition and genetic data of two subspecies (C. creticus subsp. eriocephalus and C. creticus subsp. corsicus) were investigated. The leaf trichomes were studied using scanning electron microscopy. The chemical composition of Corsican essential oil (C. creticus subsp. corsicus) has been reported using GC, GC/MS and 13C NMR; the main constituents were oxygenated labdane diterpenes (33.9%) such as 13-epi-manoyl oxide (18.5%). Using plant material (54 samples) collected from 18 geographically distinct areas of the islands of Corsica and Sardinia, the basis of variation in the headspace solid-phase microextraction volatile fraction and an inter-simple sequence repeat genetic analysis were also examined. It was shown that the two subspecies of C. creticus differed in morphology, essential oil production, volatile fraction composition and genetic data.
