The world collection of sugarcane (Saccharum hybrids. and related grasses (WCSRG) is an important source of genes for sugarcane and energycane breeding. The core collection or diversity panel of the WCSRG was created in Canal Point with 300 accession and 10 checks to evaluate its diversity. The fiber components of the species and accessions within the WCSRG are unknown, so a shredded and dried fiber sample was taken from each accession and sent for fiber analysis. The acetyl groups, acid insoluble lignin, acid soluble lignin, arabinan, glucan, holocellulose, total lignin, structural ash, and xylan were quantified on a % fiber basis and nonstructural ash on a % total basis. There were significant, but not large differences between species for holocellulose, lignin, acetyl, acid soluble lignin, nonstructural ash, and glucan. For each trait, Saccharum spontaneum had significantly more holocellulose, glucan, lignin, and nonstructural ash and less acetyl and acid soluble lignin than other species. In all populations, glucan and holocellulose were positively correlated and glucan and lignin were negatively correlated. In hybrids, internode length correlated positively with holocellulose and nonstructural ash and negatively with lignin. Principal component analysis indicated that a large amount of diversity exists within each of the species. The results suggest that strategic use of the WCSRG could provide both positive and negative selection for fiber-related traits could be made within all species of the collection.

Alfalfa (Medicago sativa L.) is an internationally significant forage crop. Forage yield, lodging resistance and spring vigor are important agronomic traits conditioned by quantitative genetic and environmental effects. The objective of this study was to identify quantitative trait loci (QTL) and molecular markers associated with increased forage yield, resistance to lodging, and spring vigor. A backcross population composed of 128 progeny was developed by crossing the breeding parents DW000577 (lodging susceptible) and NL002724 (lodging-resistant) and back-crossing an individual F1 plant to the maternal parent (i.e. DW000577). A linkage map of NL002724 was developed based upon the segregation of 236 AFLP, SRAP, and SSR markers among the backcross progeny. The markers were distributed among 14 linkage groups, covering an estimated recombination distance of 1497.6 centiMorgans (cM). Replicated clones of both parents and backcross progeny were evaluated in the field for estimated forage yield, lodging, and spring vigor in Washington and Wisconsin during 2007 and 2008. Significant QTL were found for all three traits. In particular, two QTL for lodging resistance were identified that explained ≥14 % of trait variation, and were significant in all years and locations. Major QTL explaining over 25 % of trait variation for forage yield were detected in multiple environments at two separate locations on chromosome III. Several QTL for spring vigor were located in the same or similar positions as QTL for forage yield, possibly explaining the significant correlation between these traits. Molecular markers associated with the aforementioned QTL were also identified.

Environmental stresses such as chilling temperatures can decrease germination, emergence, flower and fruit development, marketable yield, and postharvest fruit storage longevity in cucumber (Cucumis sativus L.). While response to chilling injury in cucumber is controlled by simple plastidic (maternal) and nuclear (paternal) factors, no chilling tolerant U.S. processing varieties are commercially available. Furthermore, even though three single nucleotide polymorphic sites have been identified as plastid components associated with chilling tolerance in cucumber, it is not known how these factors interact with nuclear factors controlling economically important traits. Therefore, an experiment was designed to evaluate the rate of recovery of the chilling susceptible (cytoplasm) genotype during introgression backcrossing (IB), where it was used as a recurrent parent after the initial mating to a line possessing chilling tolerant cytoplasm (donor parent). Phenotypic yield and quality trait data were collected on processing type backcross progeny (BC1–5 and BC2S3) derived from an initial ‘Chipper’ (tolerant) × line M 29 (susceptible) mating, and rate of progression to the recurrent parent was determined by simple sequence repeat marker and morphological trait analyses. Substantial degrees of the recurrent parent phenotype and nuclear genome were recovered by the BC2 generation (P = 0.001), with nearly complete recovery of recurrent parental traits and its nuclear genome occurring by the BC3. General combining ability (GCA) of derived BC2S3 lines was significant for yield, yield/plant, length (L), diameter (D), and L:D ratios. The BC2S3 line GCA and rate of progression towards the recurrent parent for economically important traits suggests that elite chilling tolerant cucumber germplasm can be developed rapidly through IB and marker genotyping.

Chilling damage can cause major reductions in cucumber (Cucumis sativus L.) yield. Cucumber plants can withstand a chilling event (i.e., tolerance and susceptibility), in which response is dictated primarily by maternally inherited plastid genomes or by the biparental contribution of a nuclear factor. To examine the modes of inheritance, exact reciprocal backcross cucumber populations (BC1–5), were created by crossing ‘Chipper’ (chilling-tolerant plastid, susceptible nucleus) and line North Carolina State University (NCSU) M29 (chilling-susceptible plastid, susceptible nucleus). These progeny and their parents were subjected to chilling stress [5.5 h at 4 °C in 270 μmol·m−2·s−1 photosynthetic photon flux (PPF) cool white lighting] at the first true-leaf stage. The chilling response of individuals possessing either NCSU M29 or ‘Chipper’ cytoplasm in any generational comparison was not significantly different (P > 0.05) from that of the maternal plastid source (susceptible or tolerant). Moreover, lines within a plastid type did not differ significantly (P > 0.05) in chilling response despite unequal nuclear contributions demonstrating the absence of nuclear additive or dosage effects originating in ‘Chipper’ or NCSU M29. Additionally, line NC-76, previously identified as a nuclear source of chilling tolerance, performed intermediate to ‘Chipper’ and NCSU M29 in chilling response under these stress conditions. The F1 progeny derived from crossing both BC5 plastidic response types (susceptible and tolerant) with NC-76 (paternal parent) performed comparable to their plastid donors and were significantly different (P < 0.0001) from one another despite their heterozygous nuclear nature resulting from the contribution of the nuclear chilling-tolerant factor contributed by NC-76. The response of tolerant and susceptible BC5 lines (i.e., ‘Chipper’ plastid in the NCSU M29 background and NCSU M29 plastid in ‘Chipper’ background, respectively) was reversible by crossing BC progeny with an alternate chilling-response plastid type. It is concluded that under these chilling conditions, plastid effects determine tolerance or susceptibility in the cucumber germplasms examined.

Chilling injury in cucumber (Cucumis sativus L.) is conditioned by maternal factors, and the sequencing of its chloroplast genome could lead to the identification of economically important candidate genes. Complete sequencing of cucumber chloroplast (cp)DNA was facilitated by the development of 414 consensus chloroplast sequencing primers (CCSPs) from conserved cpDNA sequences of Arabidopsis (Arabidopsis thaliana L.), spinach (Spinacia oleracea L.), and tobacco (Nicotiana tabacum L.) cpDNAs, using degenerative primer technologies. Genomic sequence analysis led to the construction of 301 CCSPs and 72 cucumber chloroplast-specific sequencing primers (CSSPs), which were used for the complete sequencing of cpDNA of Gy14 (155 525 bp) and ‘Chipper’ (155 524 bp) cucumber lines, which are, respectively, susceptible and tolerant to chilling injury (4 °C for 5.5 h) in the first leaf stage. Comparative cpDNA sequence analyses revealed that 1 sequence span (located between genes trnK and rps16) and 2 nucleotides (located in genes atpB and ycf1) differed between chilling-susceptible and -tolerant lines. These sequence differences correspond to previously reported maternally inherited differences in chilling response between reciprocal F1progeny derived from these lines. Sequence differences at these 3 cpDNA sites were also detected in a genetically diverse array of cucumber germplasm with different chilling responses. These and previously reported results suggest that 1 or several of these sequences could be responsible for the observed response to chilling injury in cucumber. The comprehensive sequencing of cpDNA of cucumber by CCSPs and CSSPs indicates that these primers have immediate applications in the analysis of cpDNAs from other dicotyledonous species and the investigation of evolutionary relationships.
