20. Ethylene production during Alternaria infections on potato plants and its antagonistic role in virulence of different Alternaria species.

Abstract

Alternaria species are notorious pathogens of solanaceous crops and are known to produce a plethora of toxins. Nevertheless, the involvement of toxins or other virulence factors in Alternaria infections on potato plants has never been investigated. In view of this, we analysed whether different Alternaria species produced host‐ and non‐host‐specific toxins in vitro and in vivo. Secondly, we assessed if ethylene (ETH) is involved in potato plant infections, as was previously demonstrated in tomato. Although many toxins were detected in vitro, no toxins were present before symptom appearance in potato leaves. Isolates that made large conidia (A. solani) produced up to six times more ETH in vitro than isolates with small conidia (A. arborescens) in the presence of the ETH precursor α‐keto‐γ‐methylthiobutyric acid (KMBA). In contrast, on potato leaf discs, an inverse relation was found between ETH emission and conidia size after correcting the data according to fungal DNA content, suggesting a role for ETH in symptom development rather than initiation. Moreover, application of a plant ETH biosynthesis inhibitor, 2‐aminoethoxyvinyl glycine (AVG), resulted in a 60% reduction in necrosis after inoculation with a large‐conidia isolate, whereas a 35% increase in necrosis was observed after inoculation with a small‐conidia isolate. Finally, it was concluded that toxin production is not essential for symptom development of Alternaria on potato and that ETH (either from the plant and/or the fungus) is a crucial factor in symptom development by A. solani, whereas its role was found to be antagonistic for A. arborescens. Abstract

Background: Dietitians play a critical role in the public's relationship with food and are often overlooked as an important stakeholder group in the general debate about sustainable food. Genetically modified organisms (GMOs) are one type of modern food source that could contribute to a more sustainable food system. This case study is the first to examine the knowledge, perception and willingness‐to‐recommend (WTR) genetically modified (GM) foods by dietitians in Europe. Methods: An online survey was addressed to all members of the Flemish Association of Dietitians (Belgium) in 2021, resulting in a sample of 98 valid responses. Multivariate linear regression included sociodemographic, knowledge, and attitudinal factors as the independent variables to explain dietitians' WTR. Results: Flemish dietitians had limited knowledge of GMOs; only about half of the GM questions were answered correctly. Most dietitians (53%–76%) would recommend GMOs with positive effects on human nutrition or sustainability, whereas few dietitians (19%–27%) would recommend other GMO applications. Trust in GMO information sources and perceived GM benefits significantly influenced a positive WTR of GM foods. Predominant negative information about GM foods was significantly associated with dietitians' low trust and WTR such foods. Conclusions: Countering the predominantly negative portrayal with more neutral and factual information could improve trust, which in turn could positively influence dietitians' perceptions towards GMOs. By further examining the knowledge and perception of dietitians worldwide GMOs and gene‐edited products, new insights could be could gathered into the positioning of this underexposed stakeholder group. Abstract

The two principal growth regulators cytokinins and ethylene are known to interact in the regulation of plant growth. However, information about underlying molecular mechanism and positional specificity of the cytokinin/ethylene crosstalk in root growth control is scarce. We have identified spatial specificity of cytokinin-regulated root elongation and root apical meristem (RAM) size, both of which we demonstrate to be ethylene biosynthesis-dependent. Upregulation of the cytokinin biosynthetic gene ISOPENTENYLTRANSFERASE(IPT) in proximal and peripheral tissues leads to both root and RAM shortening. In contrast, IPTactivation in distal and inner tissues reduces RAM size while leaving the root length comparable to mock-treated controls. We show that cytokinins regulate two steps specific to ethylene biosynthesis, the production of ACC by ACC SYNTHASEs (ACSs), and its conversion to ethylene by ACC OXIDASEs (ACOs). We describe cytokinin- and ethylene-specific regulation controlling the activity of ACSs and ACOs that are spatially discrete along both proximo/distal and radial root axes. Using direct ethylene measurements, we identifyACO2, ACO3andACO4as being responsible for ethylene biosynthesis and the ethylene-regulated root and RAM shortening in cytokinin-treated Arabidopsis. Finally, we describe the tight cooperation between cytokinin and ethylene signaling in cytokinin-induced, ethylene-regulated control ofACO4due to the direct interaction between ARABIDOPSIS RESPONSE REGULATOR 2 (ARR2), a member of the multistep phosphorelay cascade and the C-terminal portion of ETHYLENE INSENSITIVE 2 (EIN2-C), a key regulator of canonical ethylene signaling.
