1. Plasma Exosome Gene Signature Differentiates Colon Cancer from Healthy Controls.

Abstract

BackgroundLiquid biopsies have become an integral part of cancer management as minimally invasive options to detect molecular and genetic changes. However, current options show poor sensitivity in peritoneal carcinomatosis (PC). Novel exosome-based liquid biopsies may provide critical information on these challenging tumors. In this initial feasibility analysis, we identified an exosome gene signature of 445 genes (ExoSig445) from colon cancer patients, including those with PC, that is distinct from healthy controls.MethodsPlasma exosomes from 42 patients with metastatic and non-metastatic colon cancer and 10 healthy controls were isolated and verified. RNAseq analysis of exosomal RNA was performed and differentially expressed genes (DEGs) were identified by the DESeq2 algorithm. The ability of RNA transcripts to discriminate control and cancer cases was assessed by principal component analysis (PCA) and Bayesian compound covariate predictor classification. An exosomal gene signature was compared with tumor expression profiles of The Cancer Genome Atlas.ResultsUnsupervised PCA using exosomal genes with greatest expression variance showed stark separation between controls and patient samples. Using separate training and test sets, gene classifiers were constructed capable of discriminating control and patient samples with 100% accuracy. Using a stringent statistical threshold, 445 DEGs fully delineated control from cancer samples. Furthermore, 58 of these exosomal DEGs were found to be overexpressed in colon tumors.ConclusionsPlasma exosomal RNAs can robustly discriminate colon cancer patients, including patients with PC, from healthy controls. ExoSig445 can potentially be developed as a highly sensitive liquid biopsy test in colon cancer.

Cytoreductive surgery (CRS) combined with hyperthermic intraperitoneal chemotherapy (HIPEC) is a treatment approach for select patients. The use of curcumin, a natural compound with antitumor properties, in HIPEC is of interest due to its lower side effects compared to conventional drugs and potential for increased efficacy through direct delivery to the peritoneal cavity.Methods: An in vitro hyperthermic model was developed to simulate clinical HIPEC conditions. Three colon cancer cell lines (SK-CO-1, COLO205, SNU-C1) representing different genetic mutations (p53, KRAS, BRAF) were treated with either curcumin (25 μM) or mitomycin-C (1 μM) for 1, 2, or 3 hours. Post-treatment, cells were incubated at 37°C (normothermia) or 42°C (hyperthermia). Abstract

Introduction: Chemotherapy resistance remains a significant challenge in the treatment of pancreatic adenocarcinoma (PDAC), particularly in relation to gemcitabine (Gem), a commonly used chemotherapeutic agent. MicroRNAs (miRNAs) are known to influence cancer progression and chemoresistance. This study investigates the association between miRNA expression profiles and gemcitabine resistance in PDAC. Methods: The miRNA expression profiles of a gemcitabine-sensitive (GS) PDAC cell line, MIA PaCa-2, and its gemcitabine-resistant (GR) progeny, MIA PaCa-2 GR, were analyzed. miRNA sequencing (miRNA-seq) was employed to identify miRNAs expressed in these cell lines. Differential expression analysis was performed, and Ingenuity Pathway Analysis (IPA) was utilized to elucidate the biological functions of the differentially expressed miRNAs. Results: A total of 1867 miRNAs were detected across both cell lines. Among these, 97 (5.2%) miRNAs showed significant differential expression between the GR and GS cell lines, with 65 (3.5%) miRNAs upregulated and 32 (1.7%) miRNAs downregulated in the GR line. The most notably altered miRNAs were implicated in key biological processes such as cell proliferation, migration, invasion, chemosensitization, alternative splicing, apoptosis, and angiogenesis. A subset of these miRNAs was further analyzed in patient samples to identify potential markers for recurrent tumors. Discussion: The differential miRNA expression profiles identified in this study highlight the complex regulatory roles of miRNAs in gemcitabine resistance in PDAC. These findings suggest potential targets for improving prognosis and tailoring treatment strategies in PDAC patients, particularly those showing resistance to gemcitabine. Future research should focus on validating these miRNAs as biomarkers for resistance and exploring their therapeutic potential in overcoming chemoresistance. [ABSTRACT FROM AUTHOR]

One cellular mechanism associated with chemoresistance is alternative splicing dysfunction, a process through which nascent mRNA is spliced into different isoforms. Survivin (Baculoviral IAP Repeat-Containing Protein 5 (BIRC5)), a member of the inhibitor of apoptosis (IAP) protein family and a cell cycle-associated oncoprotein, is overexpressed in most cancers and undergoes alternative splicing (AS) to generate six different splicing isoforms.Methods: To determine if survivin splice variants (SSV) could be involved in PDAC chemoresistance, a Gemcitabine (Gem) resistant (GR) cell line, MIA PaCa-2 GR, was created and assessed for its SSV levels and their potential association with GR. Cross-resistance was assessed in MIA-PaCa-2 GR cells to FIRINOX (5-fluorouracil (5-FU), irinotecan, and oxaliplatin). Once chemoresistance was confirmed, RT-qPCR was used to assess the expression of survivin splice variants (SSVs) in PDAC cell lines. To confirm the effect of SSVs on chemoresistance, we used siRNA to knockdown all SSVs or SSV 2β.Results: The MIA PaCa-2 GR cell line was 40 times more resistant to Gem and revealed increased resistance to FIRINOX (5-fluorouracil (5-FU), irinotecan, and oxaliplatin); when compared to the parental MIA-PaCa-2 cells. Abstract

Integrating the plasma core performance with an edge and scrape-off layer (SOL) that leads to tolerable heat and particle loads on the wall is a major challenge. The new European medium size tokamak task force (EU-MST) coordinates research on ASDEX Upgrade (AUG), MAST and TCV. This multi-machine approach within EU-MST, covering a wide parameter range, is instrumental to progress in the field, as ITER and DEMO core/pedestal and SOL parameters are not achievable simultaneously in present day devices. A two prong approach is adopted. On the one hand, scenarios with tolerable transient heat and particle loads, including active edge localised mode (ELM) control are developed. On the other hand, divertor solutions including advanced magnetic configurations are studied. Considerable progress has been made on both approaches, in particular in the fields of: ELM control with resonant magnetic perturbations (RMP), small ELM regimes, detachment onset and control, as well as filamentary scrape-off-layer transport. For example full ELM suppression has now been achieved on AUG at low collisionality with n = 2 RMP maintaining good confinement HH(98,y2)≈0.95 Advances have been made with respect to detachment onset and control. Studies in advanced divertor configurations (Snowflake, Super-X and X-point target divertor) shed new light on SOL physics. Cross field filamentary transport has been characterised in a wide parameter regime on AUG, MAST and TCV progressing the theoretical and experimental understanding crucial for predicting first wall loads in ITER and DEMO. Conditions in the SOL also play a crucial role for ELM stability and access to small ELM regimes.

Abstract

Consulta en la Biblioteca ETSI Industriales (9209), [CA] L'estudi de la locomoció bípeda de robots tracta d'assolir la generació de moviments semblants als que realitza l'ésser humà en la seua caminada, per aplicar-los a robots de morfologia humana amb la combinació de les articulacions necessàries. En el present projecte es desenvolupa el disseny d'un algoritme de control per a la caminada d'un robot bípede dissenyat i construït al laboratori de robòtica de la Universidad de Chile. La tècnica emprada per al disseny de l'algoritme es basa en els principis de la caminada passiva i en el treball realitzat pel personal investigador de dita universitat. En primer lloc, s'han analitzat les característiques qualitatives i cinemàtiques de la caminada humana, ja que ens serveixen com a model a imitar pel nostre robot i ens donen una idea de la complexitat d'aquest tipus de moviments. A continuació, hem realitzat un estudi sobre les tècniques de locomoció de robots bípedes més rellevants que existeixen en l'actualitat, i com es generen els algoritmes de control necessaris per implementar-les. Així, hem pogut comprendre els avantatges de cadascuna d'elles i ens han servit d'inspiració per a la generació del nostre propi algoritme de control. Abans, però, de començar a dissenyar el nostre algoritme, cal conèixer la morfologia del robot bípede que es controlarà: aquest, ha estat dissenyat i construït amb unes característiques determinades quant a distribució d'articulacions i masses, que li atribueixen un comportament dinàmic que cal analitzar. A més compta amb actuadors i sensors de diferents tipus que hem hagut de conèixer per dissenyar l'algoritme de control adequat. A banda d'això, s'han efectuat una sèrie de càlculs i simulacions sobre el model dinàmic del nostre robot que ens han permés conèixer les trajectòries de les articulacions i les forces i moments que hi actuen. Abstract

The development of research in robotics in a developing country is a challenging task. Factors such as low research funds, low trust from local companies and the government, and a small number of qualified researchers hinder the development of strong, local research groups. In this article, and as a case of study, we present our research group in robotics at the Advanced Mining Technology Center of the Universidad de Chile, and the way in which we have addressed these challenges. In 2008, we decided to focus our research efforts in mining, which is the main industry in Chile. We observed that this industry has needs in terms of safety, productivity, operational continuity, and environmental care. All these needs could be addressed with robotics and automation technology. In a first stage, we concentrate ourselves in building capabilities in field robotics, starting with the automation of a commercial vehicle. An important outcome of this project was the earn of the local mining industry confidence. Then, in a second stage started in 2012, we began working with the local mining industry in technological projects. In this article, we describe three of the technological projects that we have developed with industry support: (i) an autonomous vehicle for mining environments without global positioning system coverage; (ii) the inspection of the irrigation flow in heap leach piles using unmanned aerial vehicles and thermal cameras; and (iii) an enhanced vision system for vehicle teleoperation in adverse climatic conditions. Abstract

The aim of this paper is to propose an automated refereeing and analysis tool for robot soccer. This computer vision based tool can be applied for diverse tasks such as: (i) automated game refereeing, (ii) computer-based analysis of the game, and derivation of game statistics, (iii) automated annotations and semantic descriptions of the game, which could be used for the automatic generation of training data for learning complex high-level behaviors, and (iv) automatic acquisition of real game data to be used in robot soccer simulators. The most attractive application of the tool is automated refereeing. In this case, the refereeing system is built using a processing unit (standard PC) and some static and/or moving video cameras. The system can interact with the robot players and with the game controller using wireless data communication, and with the human spectators and human second referees by speech synthesis mechanisms or using visual displays. We present a refereeing and analysis system for the RoboCup Four-Legged League. This system is composed by three modules: object perception, tracking, and action analysis. 41. Motion Detection and Tracking for an AIBO Robot Using Camera Motion Compensation and Kalman Filtering.

Abstract

Motion detection and tracking while moving is a desired ability for any soccer player. For instance, this ability allows the determination of the ball trajectory when the player is moving himself or when he is moving his head, for making or planning a soccer-play. If a robot soccer player should have a similar functionality, then it requires an algorithm for real-time movement analysis and tracking that performs well when the camera is moving. The aim of this paper is to propose such an algorithm for an AIBO robot. The proposed algorithm uses motion compensation for having a stabilized background, where the movement is detected, and Kalman Filtering for a robust tracking of the moving objects. The algorithm can be adapted for almost any kind of mobile robot. Results of the motion detection and tracking algorithm, working in real-world video sequences, are shown. [ABSTRACT FROM AUTHOR]

Abstract

The extracellular environment exhibits a potent effect on cellular growth and development. Exosomes secreted into this milieu carry functional proteins and nucleic acids from the cell of origin to recipient cells, facilitating intercellular communication. This interaction is particularly influential in the tumor microenvironment, transporting oncogenes and oncoproteins within a tumor and to distant sites. The mechanisms by which cells internalize exosomes vary greatly and the factors dictating this process are still unknown. Most cancers show evidence of exosomal transfer of material, but differences in cell type can dictate the effectiveness and extent of the process. Improving therapeutics requires addressing specific cellular functions, illustrating the need to better understand the forces involved in exosome-cell interactions. This review summarizes what is known about the different types of cells that play a role in exosome internalization.

Abstract

Purpose: Peritoneal carcinomatosis (PC), metastasized from colorectal cancer (CRC), remains a highly lethal disease. Outcomes of PC is significantly influenced by the amount of intra-abdominal tumor burden and therefore diagnostic tests that facilitate earlier diagnosis could improve PC treatment and patient outcomes., Experimental Design: Using mass-spectrometry-based proteomics, we characterized the protein features of circulating exosomes in the context of CRC PC, CRC with liver metastasis, and primary CRC limited to the colon. We profiled exosomes isolated from patient plasma to identify exosome-associated protein cargoes released by these cancer types., Results: Analysis of the resulting data identified metastasis-specific exosome protein signatures. Bioinformatic analyses confirmed enrichment of proteins annotated to vesicle-associated processes and intracellular compartments, as well as representation of cancer hallmark functions and processes., Conclusion and Clinical Relevance: This research yielded distinct protein profiles for the CRC patient groups and suggests the utility of plasma exosome proteomic analysis for a better understanding of PC development and metastasis., (© 2022 The Authors.
