4. A new and promising C-phycocyanin-producing cyanobacterial strain, Cyanobium sp. MMK01: practical strategy towards developing a methodology to achieve C-phycocyanin with ultra-high purity.

Abstract

Selecting a suitable cyanobacterial strain and developing easy-to-afford purification processes are two crucial aspects impacting the optimal production yield and appropriate purity of C-phycocyanin (C-PC). Cyanobium sp. MMK01, a highly efficient C-PC-producing bacterium, was identified among four cyanobacterial isolates using morphological characteristics and 16S rRNA gene sequencing. The purification process of C-PC began with ammonium sulfate precipitation, leading to a purity index (PI) of 4.04. Subsequent purification through ion exchange chromatography ultimately resulted in an ultra-highly purified form of C-PC with a significant PI of 5.82. SDS-PAGE analysis of purified C-PC showed the presence of two distinct bands, α (13 kDa) and β (15 kDa). Significantly effective at scavenging free radicals, C-PC also inhibits the viability of human lung cancer cells (Calu-6). Antibacterial, anti-inflammatory, antioxidant, and cancer-preventive compounds were detected in the MMK01 cells' methanolic extract following GC–MS analysis. The promising results indicate that Cyanobium sp. MMK01 has a great deal of potential for producing C-PC that is on par with strains found in the market, and the tried-and-true two-step purification process proved to work well to achieve an ultra-highly purified form of C-PC. Abstract

An extracellular matrix protein, fibronectin (Fn), was covalently immobilized on 316L stainless steel, L605 cobalt chromium (CoCr), and nickel titanium (NiTi) surfaces through an 11-mercaptoundecanoic acid (MUA) self-assembled monolayer (SAM) pre-formed on these surfaces. Polarization modulation infrared reflection adsorption spectroscopy (PM-IRRAS) confirmed the presence of Fn on the surfaces. The Fn monolayer attached to the SAM was found to be stable under fluid shear stress. Deconvolution of the Fn amide I band indicated that the secondary structure of Fn changes significantly upon immobilization to the SAM-functionalized metal substrate. Scanning electron microscopy and energy dispersive X-ray analysis revealed that the spacing between Fn molecules on a modified commercial stent surface is approximately 66 nm, which has been reported to be the most appropriate spacing for cell/surface interactions. Abstract

Model genetic systems are invaluable, but limit us to understanding only a few organisms in detail, missing the variations in biological processes that are performed by related organisms. One such diverse process is the formation of magnetosome organelles by magnetotactic bacteria. Studies of model magnetotactic α-proteobacteria have demonstrated that magnetosomes are cubo-octahedral magnetite crystals that are synthesized within pre-existing membrane compartments derived from the inner membrane and orchestrated by a specific set of genes encoded within a genomic island. However, this model cannot explain all magnetosome formation, which is phenotypically and genetically diverse. For example, Desulfovibrio magneticus RS-1, a δ-proteobacterium for which we lack genetic tools, produces tooth-shaped magnetite crystals that may or may not be encased by a membrane with a magnetosome gene island that diverges significantly from those of the α-proteobacteria. To probe the functional diversity of magnetosome formation, we used modern sequencing technology to identify hits in RS-1 mutated with UV or chemical mutagens. We isolated and characterized mutant alleles of 10 magnetosome genes in RS-1, 7 of which are not found in the α-proteobacterial models. These findings have implications for our understanding of magnetosome formation in general and demonstrate the feasibility of applying a modern genetic approach to an organism for which classic genetic tools are not available.

Abstract

The biological identity of nanoparticles (NPs) is established by their interactions with a wide range of biomolecules around their surfaces after exposure to biological media. Understanding the true nature of the biomolecular corona (BC) in its native state is, therefore, essential for its safe and efficient application in clinical settings. The fundamental challenge is to visualize the biomolecules within the corona and their relationship/association to the surface of the NPs. Using a synergistic application of cryo-electron microscopy, cryo-electron tomography, and three-dimensional reconstruction, we revealed the unique morphological details of the biomolecules and their distribution/association with the surface of polystyrene NPs at a nanoscale resolution. The analysis of the BC at a single NP level and its variability among NPs in the same sample, and the discovery of the presence of nonspecific biomolecules in plasma residues, enable more precise characterization of NPs, improving predictions of their safety and efficacies.

Abstract

The phagocytosis of apoptotic cells and associated vesicles (efferocytosis) by DCs is an important mechanism for both self tolerance and host defense. Although some of the engulfment ligands involved in efferocytosis have been identified and studied in vitro, the contributions of these ligands in vivo remain ill defined. Here, we determined that during Mycobacterium tuberculosis (Mtb) infection, the engulfment ligand annexin1 is an important mediator in DC cross-presentation that increases efferocytosis in DCs and intrinsically enhances the capacity of the DC antigen-presenting machinery. Annexin1-deficient mice were highly susceptible to Mtb infection and showed an impaired Mtb antigen-specific [CD8.sup.+] T cell response. Importantly, annexin1 expression was greatly downregulated in Mtb-infected human blood monocyte-derived DCs, indicating that reduction of annexin1 is a critical mechanism for immune evasion by Mtb.
