Anemia in β-thalassemia is related to ineffective erythropoiesis and reduced red cell survival. Excess free heme and accumulation of unpaired α-globin chains impose substantial oxidative stress on β-thalassemic erythroblasts and erythrocytes, impacting cell metabolism. We hypothesized that increased pyruvate kinase activity induced by mitapivat (AG-348) in the Hbbth3/+ mouse model for β-thalassemia would reduce chronic hemolysis and ineffective erythropoiesis through stimulation of red cell glycolytic metabolism. Oral mitapivat administration ameliorated ineffective erythropoiesis and anemia in Hbbth3/+ mice. Increased ATP, reduced reactive oxygen species production, and reduced markers of mitochondrial dysfunction associated with improved mitochondrial clearance suggested enhanced metabolism following mitapivat administration in β-thalassemia. The amelioration of responsiveness to erythropoietin resulted in reduced soluble erythroferrone, increased liver Hamp expression, and diminished liver iron overload. Mitapivat reduced duodenal Dmt1 expression potentially by activating the pyruvate kinase M2-HIF2α axis, representing a mechanism additional to Hamp in controlling iron absorption and preventing β-thalassemia-related liver iron overload. In ex vivo studies on erythroid precursors from patients with β-thalassemia, mitapivat enhanced erythropoiesis, promoted erythroid maturation, and decreased apoptosis. Overall, pyruvate kinase activation as a treatment modality for β-thalassemia in preclinical model systems had multiple beneficial effects in the erythropoietic compartment and beyond, providing a strong scientific basis for further clinical trials.

13. Substance Use among Transgender and Cisgender High School Students

- Abstract
Using data from a regional census survey of high school students, we compared substance use patterns between transgender and cisgender students, and students who did not know if they are transgender (DKT). Data from more than 23,000 youth from 26 high schools were used to compare alcohol-related behaviors, marijuana use, and non-medical use of prescription drugs (NMUPD) between these three groups. We also examined differences in substance use behaviors between these groups by grade. Results show that transgender students have significantly higher odds of engaging in most types of substance misuse behaviors including problem drinking, current marijuana use, and NMUPD, compared to cisgender students. DKT students have significantly higher odds of engaging in problem drinking, and NMUPD compared to cisgender students. Furthermore, differences in some substance use behaviors between transgender and cisgender students are greater among students in earlier grades compared to later grades; though this was not the case when comparing cisgender students to DKT students. Results highlight the elevated risk of substance use among transgender youth--especially during early high school years, and students who are not sure about their gender identity. Implications about the need for targeted and early prevention efforts for this vulnerable population are discussed.

This paper deals with the optimal scheduling of the resources of a renewable energy community, whose coordination is aimed at providing flexibility services to the electrical distribution network. The available resources are renewable generation units, battery energy storage systems, dispatchable loads, and power-to-hydrogen systems. The main purposes behind the proposed strategy are enhancement of self-consumption and hydrogen production from local resources and the maximization of the economic benefits derived from both the selling of hydrogen and the subsidies given to the community for the shared energy. The proposed approach is formulated as an economic problem accounting for the perspectives of both community members and the distribution system operator. In more detail, a mixed-integer constrained non-linear optimization problem is formulated. Technical constraints related to the resources and the power flows in the electrical grid are considered. Numerical applications allow for verifying the effectiveness of the procedure. The results show that it is possible to increase self-consumption and the production of green hydrogen while providing flexibility services through the exploitation of community resources in terms of active and reactive power support. More specifically, the application of the proposed strategy to different case studies showed that daily revenues of up to EUR 1000 for each MW of renewable energy generation installed can be obtained. The decarbonization of the electricity grid is one of the actions that can help reduce fossil fuel emissions, and thus their impact on global warming in the future. This is also going to be supported by the shift from the paradigm of production–transmission–distribution, where electricity production oversees large-size power plants, to renewable-based distributed/diffused production, where electricity is generated very close or even by the same (group of) user(s) (or prosumers in the latter case). The number of mid-/small-size installations based on renewable energy technologies will therefore increase substantially, and the related renewable generation will be dominant against that from large-size power plants. Unfortunately, this will very likely reduce the reliability of the grid, unless appropriate countermeasures are taken/implemented, hopefully at the same time that the paradigm shift is being achieved. To this aim, it is important to identify the anomalies and main fault causes that might possibly affect some of the central renewable (wind, PV, hydrogen) and ancillary technologies that will be used to establish future renewable-based power systems. Accordingly, this paper presents a literature survey, also extending the focus to related datasets that can be used for deeper investigation. It is highlighted that the gaps mainly refer to a lack of a common taxonomy that prevents the establishment of structured knowledge in the scope of renewable-based power systems, a lack of contributions to anomalies/faults specific to wind turbines, and a lack of datasets related to electrolyzers, fuel cells, DC/x conversion, and monitoring and communication systems. Further, in the case of monitoring and communication systems, the scientific literature is both very dated, therefore not considering possible new aspects that would be currently worthy of investigation, and not oriented toward the particular domain addressed, thus considering peculiar aspects that are left out. Accurate predictions of photovoltaic generation are essential for effectively managing power system resources, particularly in the face of high variability in solar radiation. This is especially crucial in microgrids and grids, where the proper operation of generation, load, and storage resources is necessary to avoid grid imbalance conditions. Therefore, the availability of reliable prediction models is of utmost importance. Authors address this issue investigating the potential benefits of a machine learning approach in combination with photovoltaic power forecasts generated using weather models. Several machine learning methods have been tested for the combined approach (linear model, Long Short-Term Memory, eXtreme Gradient Boosting, and the Light Gradient Boosting Machine). Among them, the linear models were demonstrated to be the most effective with at least an RMSE improvement of 3.7% in photovoltaic production forecasting, with respect to two numerical weather prediction based baseline methods. The conducted analysis shows how machine learning models can be used to refine the prediction of an already established PV generation forecast model and highlights the efficacy of linear models, even in a low-data regime as in the case of recently established plants. Overexpression of the Runt-related transcription factor 2 (RUNX2) has been reported in several cancer types, and the C-X-C motif chemokine receptor 4 (CXCR4) has an important role in tumour progression. However, the interplay between CXCR4 and RUNX2 in melanoma cells remains poorly understood. In the present study, we used melanoma cells and a RUNX2 knockout (RUNX2-KO) in vitro model to assess the influence of RUNX2 on CXCR4 protein levels along with its effects on markers associated with cell invasion and autophagy. Osteotropism was assessed using a 3D microfluidic model. Moreover, we assessed the impact of CXCR4 on the cellular levels of key cellular signalling proteins involved in autophagy. We observed that melanoma cells express both RUNX2 and CXCR4. Restored RUNX2 expression in RUNX2 KO cells increased the expression levels of CXCR4 and proteins associated with the metastatic process. The protein markers of autophagy LC3 and beclin were upregulated in response to increased CXCR4 levels. The CXCR4 inhibitor WZ811 reduced osteotropism and activated the mTOR and p70-S6 cell signalling proteins. Our data indicate that the RUNX2 transcription factor promotes the expression of the CXCR4 chemokine receptor on melanoma cells, which in turn promotes autophagy, cell invasiveness, and osteotropism, through the inhibition of the mTOR signalling pathway. Our data suggest that RUNX2 promotes melanoma progression by upregulating CXCR4, and we identify the latter as a key player in melanoma-related osteotropism. [ABSTRACT FROM AUTHOR]

BackgroundA typical pathological feature of Alzheimer's disease (AD) is the appearance in the brain of senile plaques made up of β-amyloid (Aβ) and neurofibrillary tangles. AD is also associated with an abnormal accumulation of some metal ions, and we have recently shown that one of these, aluminum (Al), plays a relevant role in affecting Aβ aggregation and neurotoxicity.MethodologyIn this study, employing a microarray analysis of 35,129 genes, we investigated the effects induced by the exposure to the Aβ1–42-Al (Aβ-Al) complex on the gene expression profile of the neuronal-like cell line, SH-SY5Y.Principal FindingsThe microarray assay indicated that, compared to Aβ or Al alone, exposure to Aβ-Al complex produced selective changes in gene expression. Some of the genes selectively over or underexpressed are directly related to AD. A further evaluation performed with Ingenuity Pathway analysis revealed that these genes are nodes of networks and pathways that are involved in the modulation of Ca2+ homeostasis as well as in the regulation of glutamatergic transmission and synaptic plasticity.Conclusions and SignificanceAβ-Al appears to be largely involved in the molecular machinery that regulates neuronal as well as synaptic dysfunction and loss. Aβ-Al seems critical in modulating key AD-related pathways such as glutamatergic transmission, Ca2+ homeostasis, oxidative stress, inflammation, and neuronal apoptosis.

Anemia in [beta]-thalassemia is related to ineffective erythropoiesis and reduced red cell survival. Excess free heme and accumulation of unpaired [alpha]-globin chains impose substantial oxidative stress on [beta]-thalassemic erythroblasts and erythrocytes, impacting cell metabolism. We hypothesized that increased pyruvate kinase activity induced by mitapivat (AG-348) in the [Hbb.sup.th3/+] mouse model for [beta]-thalassemia would reduce chronic hemolysis and ineffective erythropoiesis through stimulation of red cell glycolytic metabolism. Oral mitapivat administration ameliorated ineffective erythropoiesis and anemia in [Hbb.sup.th3/+] mice. Increased ATP, reduced reactive oxygen species production, and reduced markers of mitochondrial dysfunction associated with improved mitochondrial clearance suggested enhanced metabolism following mitapivat administration in [beta]-thalassemia. The amelioration of responsiveness to erythropoietin resulted in reduced soluble erythroferrone, increased liver Hamp expression, and diminished liver iron overload. Mitapivat reduced duodenal Dmt1 expression potentially by activating the pyruvate kinase M2-HIF2[alpha] axis, representing a mechanism additional to Hamp in controlling iron absorption and preventing [beta]-thalassemia-related liver iron overload. In ex vivo studies on erythroid precursors from patients with [beta]-thalassemia, mitapivat enhanced erythropoiesis, promoted erythroid maturation, and decreased apoptosis. Overall, pyruvate kinase activation as a treatment modality for [beta]-thalassemia in preclinical model systems had multiple beneficial effects in the erythropoietic compartment and beyond, providing a strong scientific basis for further clinical trials., Introduction Ineffective erythropoiesis and reduced red cell survival are key determinants of the anemia and other comorbidities in [beta]-thalassemia (1-3).
