2. Cardioinotropic effects in subchronic intoxication of rats with lead and/or cadmium oxide nanoparticles

Abstract

Subchronic intoxication was induced in outbred male rats by repeated intraperitoneal injections with lead oxide (PbO) and/or cadmium oxide (CdO) nanoparticles (NPs) 3 times a week during 6 weeks for the purpose of examining its effects on the contractile characteristics of isolated right ventricle trabeculae and papillary muscles in isometric and afterload contractions. Isolated and combined intoxication with these NPs was observed to reduce the mechanical work produced by both types of myocardial preparation. Using the in vitro motility assay, we showed that the sliding velocity of regulated thin filaments drops under both isolated and combined intoxication with CdO– NP and PbO–NP. These results correlate with a shift in the expression of myosin heavy chain (MHC) isoforms towards slowly cycling β–MHC. The type of CdO–NP + PbO–NP combined cardiotoxicity depends on the effect of the toxic impact, the extent of this effect, the ratio of toxicant doses, and the degree of stretching of cardiomyocytes and muscle type studied. Some indices of combined Pb–NP and CdO–NP cardiotoxicity and general toxicity (genotoxicity included) became fully or partly normalized if intoxication developed against background administration of a bioprotective complex. © 2021 by the authors. 13. Organism's responses to a long-term inhalation of silica-containing submicron particles of an industrial aerosol

Abstract

Female white rats were exposed during up to 6 months 5 times a week, 4 hr per day in a "nose only" inhalation device to an aerosol containing predominantly submicron (nanoscale included) particles of amorphous silica in the concentration 2.6±0.6 or 10.6±2.1 mg/m3. In an auxiliary experiment with a single-shot intratracheal instillation of these particles, it was shown that they induced a pulmonary cell response comparable with that to highly cytotoxic and fibrogenic standard quartz powder DQ12. However, in the long-term inhalation test, the studied aerosol proved to be of very low systemic toxicity and fibrogenicity. This paradox may be explained by low SiO2 retention in lungs and other organs due to a relatively high in vivo solubility of these nanoparticles. Nevertheless, their genotoxic action and transnasal penetration into the brain should make one give a cautious overall assessment of this aerosol as an occupational or environmental hazard. © Published under licence by IOP Publishing Ltd.

Abstract

Rats were exposed to nickel oxide nanoparticles (NiO-NP) inhalation at 0.23 ± 0.01 mg/m3 for 4 h a day 5 times a week for up to 10 months. The rat organism responded to this impact with changes in cytological and some biochemical characteristics of the bronchoalveolar lavage fluid along with a paradoxically little pronounced pulmonary pathology associated with a rather low chronic retention of nanoparticles in the lungs. There were various manifestations of systemic toxicity, including damage to the liver and kidneys; a likely allergic syndrome as indicated by some cytological signs; transient stimulation of erythropoiesis; and penetration of nickel into the brain from the nasal mucous membrane along the olfactory pathway. Against a picture of mild to moderate chronic toxicity of nickel, its in vivo genotoxic effect assessed by the degree of DNA fragmentation in nucleated blood cells (the RAPD test) was pronounced, tending to increasing with the length of the exposure period. When rats were given orally, in parallel with the toxic exposure, a set of innocuous substances with differing mechanisms of expected bioprotective action, the genotoxic effect of NiO-NPs was found to be substantially attenuated. © 2019 by the authors. Abstract

Morphological criteria of diagnosis of dust lesions of lungs in tumors on resection material are considered in the article. It is shown that the complex application of various research methods allows the identification of dust particles in the lung and lymph node tissues, and contributes to the improvement of the diagnosis of dust lesions of the respiratory system. Morphological characteristics of bauxite pneumoconiosis in primary lung cancer on resection material are presented.

Abstract

With taking into account the biological toxicity of metallic and metal oxide nanoparticles to be a well-established fact, the widespread use of nanotechnologies and the significant proportion of nanoscale particles in industrial aerosols released during many pyrometallurgical and electric welding processes determine the importance of searching for approaches to increase the organism's resistance to them. Results of a number of experiments show the application of combinations of some bioactive agents composed in accordance with sound theoretical premises and used in innocuous doses to be potent of the significant attenuation of the integral and specific toxicity of metallic nanoparticles and even their genotoxicity. Further research followed by practical application of the results is a promising trend in the occupational risk management strategy. © СOLLECTIVE OF AUTHORS, 2017.

Abstract

Stable suspensions of nanogold (NG) and nanosilver (NS) with mean particle diameter 50 and 49 nm, respectively, were prepared by laser ablation of metals in water. To assess rat's pulmonary phagocytosis response to a single intratracheal instillation of these suspensions, we used optical, transmission electron, and semi-contact atomic force microscopy. NG and NS were also repeatedly injected intraperitoneally into rats at a dose of 10 mg/kg (0.5 mg per mL of deionized water) three times a week, up to 20 injections. A group of rats was thus injected with NS after oral administration of a "bioprotective complex" (BPC) comprised of pectin, multivitamins, some amino acids, calcium, selenium, and omega-3 PUFA. After the termination of the injections, many functional and biochemical indices and histopathological features of the spleen, kidneys and liver were evaluated for signs of toxicity, and accumulation of NG or NS in these organs was measured. From the same rats, we obtained cell suspensions of different tissues for performing the RAPD test. It was demonstrated that, although both nanometals were adversely bioactive in all respects considered in this study, NS was more noxious as compared with NG, and that the BPC tested by us attenuated both the toxicity and genotoxicity of NS. © 2013 by the authors; licensee MDPI, Basel, Switzerland.

Abstract

Sodium fluoride solution was injected i.p. to rats at a dose equivalent to 0.1 LD50 three times a week up to 18 injections. Two thirds of these rats and of the sham-injected ones were exposed to the whole body impact of a 25 mT static magnetic field for 2 or 4 h a day, 5 times a week. For mathematical analysis of the effects they produced in combination, we used a response surface model. This analysis demonstrated that (like in combined toxicity) the combined adverse action of a chemical plus a physical agent was characterized by a diversity of types depending not only on particular effects these types were assessed for but on their level as well. From this point of view, the indices for which at least one statistically significant effect was observed could be classified as identifying (1) single-factor action; (2) additivity; (3) synergism; (4) antagonism (both subadditive unidirectional action and all variants of contradirectional action). Although the classes (2) and (3) taken together encompass a smaller part of the indices, the biological importance of some of them renders the combination of agents studied as posing a higher health risk than that associated with each them acting alone., (Copyright © 2016 Elsevier Ltd. Abstract

Stable suspensions of NiO and/or Mn304 nanoparticles with average diameter 16,7?8,2 nm and 18,4?5,4 nm respectively, obtained via laser ablation of the metals with 99,99% purification in deionized water, were injected intraperitoneally into rats in dose of 0,5 mg or 0,25 mg three times per week up to 18 times separately or in various dose combinations. A group of rats received combined injections of nanoparticles in the highest dose or merely water with oral <> containing pectin, vitamins A, C and E, glutamate, glycine, N-acetylcysteine, selenium, iodine and polyunsaturated fatty acids of omega-3 class. Intoxication development was assessed through multiple functional parameters and histologic changes in liver, spleen, kidneys and brain. Nickel and manganese accumulation in these organs was measured-via various methods. Both types of metallic oxide nanoparticles appeared to be hazardous for body, but Mn304 caused more harm according to major nonspecific toxicity manifestations. Moreover, they caused more intense injury to caudate nucleus and hippocamp neurons - that can be considered as an experimental model of manganese parkinsonism. Mathematic analysis based on response pattern revealed ambiguity of the combined toxicity type, depending on the effects assessed and on its level. Due to the bioprotector complex, organic and systemic toxicity and genotoxicity of Mn304 and NiO nanoparticles combined were diminished.

Abstract

Extremely high toxicity of metal-containing nanoparticles necessitates search of methods to increase body resistance against its harmful effects. The authors' experiments summarized in the article demonstrated that some combinations of certain biologically active substances selected according to sound theoretic background and prescribed in harmless doses can significantly decrease integral and specific manifestations of organ and system toxicity and even genotoxicity of such nanoparticles. Further development of this research direction should be recommended for practical implementation.

Abstract

Subchronic intoxications in rats induced by repeated intraperitoneal injections of stable water suspensions of silver or copper oxide nanoparticles in low dosage were manifested by adverse shifts in some functional and biochemical indices, by development of histo-structural changes in different tissues and by poly-organ fragmentation of DNA. All these manifestations of toxicity were substantially attenuated against the background of parallel oral administration of bioprotective complexes comprising vitamins, trace elements, pectin, some amino acids and a fish oil preparation rich in omega-3 fee fatty acids, this composition has been adjusted to mechanisms of action of this or that nanomaterial.

Abstract

Aim: To specify the cytokine profile in chronic glomerulonephritis (CGN) with aggravating renal function, to assess prognostic value of some cytokines in development of CGN in these patients., Material and Methods: Indices of the cytokine profile were followed up in 56 CGN patients with nephrotic syndrome and arterial hypertension and compared in those patients who developed CGN for 2 years (group 2) versus patients with intact renal function (group 1)., Results: Serum levels of IL-1beta and TNF-alpha were maximal in CGN patients with aggravating renal function. Such patients had a low concentration of serum IL-1beta receptors antagonists. Severity of proteinurea correlated with serum level of TNF-alpha. If the latter was higher than 120 pg/ml, there was a high probability of CRF development within 2 years., Conclusion: Pathogenesis of CRF in CGN is associated with proinflammatory cytokines, especially TNF-alpha. Its serum concentration can be used for evaluating risk of renal function deterioration.
