The early death, which is more common in acute promyelocytic leukemia (APL) patients rather than other types of acute myelocytic leukemia (AML) highlights the importance of appropriate diagnostic method for early detection of this disease. Background: It is increasingly evident that interactions between leukemic cells and their niches can have profound effects on clinical outcomes and have been contributed to the failure in treatment and drug shortage in the eradication of minimal residual disease, at least in part, through moving the cells from proliferative state to quiescent state. Objectives: We, therefore, investigated the effects of different bone marrow stromal cells (BMSCs) on the induction of quiescence and tested the advantage of pan-HDAC inhibitor panobinostat in the induction of apoptosis and targeting the quiescence cells of APL-derived (NB4) and CML-derived (K562) cell lines. Methods: We firstly evaluated the effect of BMSCs including mesenchymal stem cell (MSC), osteoblast, and macrophage on the induction of NB4 and K562 cells quiescence in co-culture models. Next, the alterations in mRNA expression of quiescence-related genes and leukemia-driver oncogenes were evaluated in different models. Finally, the anti-leukemic effects of panobinostat were evaluated, using MTT assay and evaluation of apoptosis and G0 population. Results: Upon 10 days of co-culture with stromal cells, we found that leukemic cells significantly accumulated in the G0 phase. The co-cultured cells also depictured an overall overexpression of most quiescence promoter genes. The oncogenes were underexpressed in the majority of co-cultured models. The results also showed that although panobinostat could induce apoptosis in co-cultured cells, its effect on the reduction of the G0 population was more striking. Conclusions: These data propose that leukemia cells' quiescence state induced by stromal cells is reversible by HDAC inhibition and panobinostat could be a potentiate drug for eradication of treatment-resistance quiescence leukemic cells.

Background: Cancer is a multi-faceted diseases caused cell proliferation in an out of control manner due to accumulation of defects and mutation in their DNA and with an impendence to invade or spread to parts of the body. During last decades new compounds with natural roots have emerged as a new paradigm for effective anti-cancer treatment. Objective: The aim of this study is to evaluate the potential of ergolide a sesquiterpene lactone with multi-functional history to induce apoptosis. Methods: Ergolide from Inula-Oculus-Christi a sesquiterpene lactone from Asteraceae plant was extracted by Traditional Medicine Research Center of Shahid Beheshti University of Medical Sciences. The cytotoxic effects of Ergolide on the acute lymphoblastic leukemia (MOLT4) and PBMC (normal cell line) were investigated at different doses for 48 hours. In this study, MTT assay and acridine orange staining were used. Also, annexin V-PI assays was utilized for further evaluation. In addition the gene expression level of BAX and BCL2 were analyzed by q-RealTime-PCR (quantitive RT-PCR). Results: The results of MTT assay demonstrated the induction of apoptosis and reduction in proliferation of MOLT4 cells treated by ergolide. (P < 0.001). Interestingly Ergolide could be less toxic in normal cells (PBMCs). AO staining and Flow cytometry analysis confirmed a significantly high percentage of autophagic and apoptotic cells compared with control groups respectively (P < 0.05). Conclusion: According to the results, ergolide demonstrated cytotoxic effects on MOLT-4, but further studies are needed to confirm its effectiveness as a complementary agent in the treatment of acute lymphoblastic leukemia.

Background Mycobacterium fortuitum (M. fortuitum) is a bacterium, which can cause infections in many anatomical regions of the body, including the skin, lymph nodes, and joints. This bacterium, which belongs to a group of bacteria known as nontuberculous mycobacteria, is regarded as an important nosocomial pathogen worldwide owing to its increasing antibiotic resistance. Recently, the antimicrobial effects of carbon nanotubes have been reported in numerous studies. These nanotubes can be very useful in drug delivery; besides, they exhibit unique properties against multidrug-resistant bacterial infections. This study aimed to investigate the antimicrobial effects of carboxyl-functionalized multi-walled carbon nanotubes (MWCNT-COOH) to reduce antibiotic resistance. Methods In this study, antibacterial effects of nanofluids containing functionalized MWCNTs at initial concentration of 2 mg/mL and serial dilutions of 54, 28.5, 14.25, 7.12, 3.5 µg/mL, antibiotics alone and combination of nanofluids with antibiotics were investigated. Standard and resistant strains of M. fortuitum were obtained from the microbial bank of the Department of Mycobacteriology and Pulmonary Research, Pasteur Institute of Iran. Results It was observed that nanofluid containing MWCNT-COOH can exert antimicrobial effects on M. fortuitum and significantly reduce bacterial resistance to antibiotics including kanamycin and streptomycin. In the presence of antibiotics and nanofluids containing MWCNT-COOH at a dose of 28.5 µg/mL, no growth was observed. Conclusion One of the main antimicrobial mechanisms of MWCNT-COOH is penetration into the bacterial cell wall. In this study, by using the nanofluid containing MWCNT-COOH with increased stability, the antibiotic resistance of M. fortuitum was significantly reduced at lower dilutions compared to the antibiotic alone.

We aimed to evaluate the predictive value of circulatory microRNAs in extracellular vesicles (EVs) and plasma in platelet and neutrophil engraftment. Circulatory miR-125b, mir-126, miR-150, and miR-155 expression was assessed in isolated EVs and plasma in samples collected from AHSCT candidates. Several studies show that childhood and adulthood asthma and its symptoms can be modulated through epigenetic modifications. Epigenetic changes are inheritable modifications that can modify the gene expression without changing the DNA sequence. The most common epigenetic alternations consist of DNA methylation and histone modifications. How these changes lead to asthmatic phenotype or promote the asthma features, in particular by immune pathways regulation, is an understudied topic. Since external effects, like exposure to tobacco smoke, air pollution, and drugs, influence both asthma development and the epigenome, elucidating the role of epigenetic changes in asthma is of great importance. This review presents available evidence on the epigenetic process that drives asthma genes and pathways, with a particular focus on DNA methylation, histone methylation, and acetylation. We gathered and assessed studies conducted in this field over the past two decades. Our study examined asthma in different aspects and also shed light on the limitations and the important factors involved in the outcomes of the studies. To date, most of the studies in this area have been carried out on DNA methylation. Therefore, the need for diagnostic and therapeutic applications through this molecular process calls for more research on the histone modifications in this disease.

Vanadium pentoxide (V2O5) is a transition metal oxide with features such as high availability, good catalytic activity, unique electrical properties and high conductivity which are appropriate for gas sensing applications. In this review, we discuss different gas sensing aspects of V2O5 in pristine, doped, decorated and composite forms. Depending on its synthesis procedure, morphology, sensing temperature and surface conditions, the V2O5-based gas sensors show different responses to target gases. Herein, we have discussed the behavior of V2O5-based gas sensors to different gases and associated sensing mechanisms. This review paper can be a useful reference for the researchers who works in the field of gas sensors.

Background: Acute graft versus host disease (aGVHD) is a common complication following allogeneic hematopoietic stem cell transplantation (AHSCT) caused by cellular and inflammatory factors, including those arising from monocytes and dendritic cells as integral parts of the immune system. Long non-coding RNAs (lncRNA) have recently emerged as potential regulators of the immune responses and it is supported that their dysregulation can develop various immune disorders. As an intergenic lncRNA, the lnc-DC was shown to regulate the human monocytes differentiation and antigen presenting cells (APCs) activation during immune responses. It is also shown that lnc-DC knockdown reduces T-cell activation and cytokine release. Objectives: The aim of this study was to assess whether the lnc-DC plays a role in patients with aGVHD by measuring its expression levels compared to non-aGVHD patients on specific time intervals following transplantation. Methods: Participants included 38 patients who underwent primary allogeneic bone marrow transplantation. Peripheral blood mononuclear cells (PBMCs) were isolated by Ficoll-Hypaque gradient from the blood samples collected at days 0, 7, 14, 28, and final day of transplantation. The qRT-PCR was used to quantify the lnc-DC levels. Results: Findings revealed a significant increase in the lnc-DC levels on day 28 and the final day after transplantation in patients with aGVHD compared to non-GVHD patients (CI = 95%, P < 0.03 on day 28 and P < 0.01 on the final day). Furthermore, the receiver operating characteristic (ROC) curve analysis showed an acceptable total area under the curve for the lnc-DC gene expression data, suggesting a fair diagnostic value for lnc-DC. Conclusions: Taken together, data of the present study supported a strong correlation between lncRNA-DC expression and aGVHD occurrence. As a result, lnc-DC may be considered as a new molecular marker for the aGVHD prognosis.

We herein aimed to compare the expression patterns of a histone modifiers panel consisted of SUV39H1, PRDM16, UHRF2, KDM2B, and KDM3C between acute myeloid leukemia(AML) cells and normal cells and to assess the correlation of these genes with the expression of vital tumor suppressor genes, including p16INK4A and p53.Materials and methods Bone marrow or peripheral blood samples of 50 AML patients at diagnosis and also 18 subjects with a normal hematopoietic system as a control group were obtained after informed consent. Then, qRT-PCR was performed to determine the expression levels of the aforementioned genes. Results We found a broad alteration in the expression signature of five out of seven studied genes in AML patients as compared with the control group. UHRF2 and p53 were remarkably downregulated in AML patients (P < 0.001), while SUV39H1, PRDM16, and KDM3C significantly overexpressed (P< 0.01). Based on the Spearman rank correlation, SUV39H1, KDM2B, and age of the patients negatively regulated both p16INK4A and p53 expression. Conclusion Taken together, our findings provided preliminary evidence regarding the pervasive mRNA perturbation of histone modifiers and their plausible influences on critical tumor suppressor genes. Future studies in this area would be required to assist in establishing these results in the clinical practice of AML patients.

Acetone is a well-known volatile organic compound that is widely used in different industrial and domestic areas. However, it can have dangerous effects on human life and health. Thus, the realization of sensitive and selective sensors for recognition of acetone is highly important. Among different gas sensors, resistive gas sensors based on nanostructured metal oxide with high surface area, have been widely reported for successful detection of acetone gas, owing to their high sensitivity, fast dynamics, high stability, and low price. Herein, we discuss different aspects of metal oxide-based acetone gas sensors in pristine, composite, doped, and noble metal functionalized forms. Gas sensing mechanisms are also discussed. This review is an informative document for those who are working in the field of gas sensors.

BACKGROUND: The recent outbreak by a novel coronavirus originated from Wuhan, China in 2019, and is progressively spreading to other countries. Timely diagnosis of the coronavirus disease 2019 (COVID-19) improves the survival of the patients and also prevents the transmission of the infection. In this study, we reviewed the applicable and available methods for the diagnosis of COVID-19. METHODS: For the review, we systematically searched Web of Science, PubMed, and Iranian articles that were published about COVID-19 diagnostic methods with a combination of the key terms: laboratory, radiological, tests, coronavirus. RESULTS: Although the current gold standard diagnostic test for this virus is real-time reverse-transcriptase polymerase chain reaction (RT-PCR), the occasional false-negative and the low sensitivity of the test should not be underestimated. A chest computed tomography (CT) scan is another diagnostic test for COVID-19, with higher sensitivity but low specificity. A combination of sensitive RT-PCR with a chest CT scan together with the clinical features are highly recommended for the proper diagnosis. Notably, there are some other sensitive and low-cost tests for evaluation of COVID-19 infection, but their validation should be approved. CONCLUSION: Since early and accurate diagnosis of the viral disease could improve the survival rate of the patients, and halt the transmission chain, it is not surprising that tremendous attempts should be made to reduce the limitations of the tests leading to the false-negative results and to find a rapid test for the diagnosis of COVID-19. © 2020 The Author(s). Introduction: Gene mutation is an infrequent cause of tumor suppressor gene (TSG) defect in de novo AML patients. Instead, it seems that leukemic cells employ epigenetic tricks to attenuate the negative impacts of intact TSGs. Ordinarily, critical TSGs, such as p16INK4A, is hyper-methylated in AML blasts under the impact of master epigenetic regulators, such as UHRF1. In this study, we investigated the correlation between UHRF1 and p16INK4A gene expression levels in newly diagnosed AML patients. Methods: Bone marrow and peripheral blood samples were obtained from 50 newly diagnosed AML patients and 18 healthy normal control subjects. Gene expression levels of UHRF1 and P16INK4A were surveyed using SYBR Green Quantitative Real-time PCR. Statistical analyses were done using SPSS statistical software 21.0. Results: P16INK4A gene expression showed reduced levels in 80.64% of patients above 45 years of age, while only 32% of patients below 45 years had reduced expression levels. The Spearman correlation test also demonstrated a significant negative correlation between UHRF1 and p16INK4A gene expression levels in AML patients, which was not observed in the control group (r=0.343 and P= 0.015). Conclusion: Regarding the age-related patterns of UHRF1 and p16INK4A gene expression, and also the presence of negative correlation between them, we conclude that UHRF1 may potentially be involved in p16INK4A down-regulation in elderly AML patients, which may subsequently facilitate the progression of AML in older ages.

Background: Sulfur mustard (SM) is a chemical blistering warfare that affects different organs especially hematopoietic system. Prevalence of acute myeloblastic and lymphoblastic leukemia is increased by sulfur mustard exposure. FLT3-ITD mutation can be effective on leukemogenesis. Therefore, the aim of this study was to verify the frequency of FLT3-ITD mutation in the patients who exposed to SM. Methods: This study was implemented on 42 people poisoned by SM during Iraq-Iran war about three decades ago and is now resident in Mashhad, Iran. The control group included 30 healthy males that are relatives of the patients with first-degree. After DNA extraction, PCR was performed for FLT3-ITD analysis. Results: By analysis of PCR products, no FLT3-ITD mutation was detected in the patient or control groups. There was no significant difference in hematological factors between the two groups. Conclusion: Other mechanisms can lead to leukemia in SM exposed persons. Elapsed time after exposure to sulfur mustard can be effective on leukemogenesis, then future more study may be beneficial for early diagnosis of leukemia in SM exposed veterans.

Purpose of the study Acute graft versus host disease (aGVHD) is an immune-mediated reaction that results in impaired immune and body function after allogeneic hematopoietic stem cell transplantation (allo-HSCT). lncRNAs have been discovered as particular T cell regulators, and alloreactive T cells have been known as a critical factor in aGVHD. As a result, we investigated the importance of lnc-MAF4 and IFNG-AS1 expression levels in aGVHD patients versus non-aGVHD patients. Material and Methods This research included 38 patients with hematological disorders who were undergoing primary allo-HSCT. Human identical siblings or unrelated donors were used to collect stem cell. Samples were taken within days 0, 7, 14, 28, and 52±8 after transplantation. The expression of lncRNA levels was measured using the QRT-PCR technique. And the data were analyzed using GraphPad Prism 6 Results Our data revealed that LncRNA MAF4 and INFG-AS1 expression levels in aGVHD were not significantly different compared to the non-GVHD group immediately after transplantation, nor at day 7 or 14. However, the aGVHD group showed an overt up-regulation of the two lncRNAs on samples taken at day 28 and 52±8 compared to non-GVHD patients. Discussion Since the intracellular pathway of these lncRNAs shows a direct relationship with the IFNγ cytokine production resulting in differentiation to TH1 cells and inhibition of differentiation to TH2 cells, they can be, therefore, considered as suitable molecular candidates for the prediction of aGVHD in patients receiving HSCT.

In this paper a novel triple band compact microstrip slot antenna is designed, utilizing fractal Koch shape based on metamaterial electric-LC element coupling load. The antenna is fed by CPW line, and it has triple band resonances operating at frequencies of about 1.5, 3.5 and 5.4 GHz for GPS, WiMAX and WLAN applications. The bandwidth of antenna at aforementioned frequencies is about 1.9%, 14% and 5% with the gains of −4.5, 3.75 and 5.3 dBi respectively. Also it has property of circular polarization for WiMAX application at 3.5 GHz. Furthermore metamaterial ELC element based on the metasurface theory, and making variation of the current distribution for circular polarization, made a compact small antenna at 1.5 GHz, where the antenna size is reduced to λ/5 × λ/5 through coupling on ELC element. The total size of the fractal antenna is assumed 40 × 40 mm, and is designed and fabricated on FR-4. The simulation results are extracted by HFSS commercial software, and compared with experimental results.

The morphology of scales from four flank regions in two species of Scorpaeniforme fishes of the family Platycephalidae was studied by applying light and scanning electron microscopy, and the age-dependent variability of scale structures was discussed. The scales of four flank regions from three size classes in both species were ctenoid. The posterior margin of all the ctenoid scales was formed by two rows of cteni, while the three complete rows of cteni were not observed in the examined scales. The cteni of both rows were similar in morphology. Some scale characteristics showed alteration during the fish growth, including the shape of the anterior region of scale in Platycephalus indicus, the presence of lepidonts in Grammoplites suppositus, and the general shape of scale as well as the indices SCL.SCW and JSW.SL in both species. Also, the number of cteni in posterior margin of scales was increased during fish development, while their general morphology did not change significantly. The number of cteni in the scales of small and large fishes in G. suppositus was found to be obviously lower than P. indicus. By considering the function of lepidonts in improving the hydrodynamic efficiency of swimming, it can be assumed that G. suppositus is not probably very active in swimming as P. indicus. As a conclusion, modification in the ornamentations of the posterior region are subject to alteration during the growth of the fish. Besides its developmental changes, some scale characters in the adult forms have taxonomic significance, including the square-shaped scales in P. indicus vs. the oval-shaped scale in G. suppositus; a total of 26–31 radii in the anterior field in P. indicus vs. 11–12 radii in G. suppositus; the presence of many (54–58) cteni in the posterior part in P. indicus vs. a moderate (19–23) cteni in G. suppositus. These characters could be used as additional morphological data to be used for studying systematics and even phylogeny of the flathead fishes.

By the advent of the effective therapies for many coagulation diseases and hereditary spherocytosis (HS), patient's survival has been improved significantly; however, if patients are diagnosed late or left untreated, both diseases could ominously be life threatening. Concurrent occurring of factor VII (FVII) deficiency and HS is extremely rare and there is no literature report that explain this condition, thus far. In this study, we confronted a 9-year-old female patient diagnosed with HS and enlarged spleen as a result of this blood disorder. Given to her sever signs and symptoms of splenomegaly, she was candidate for emergent splenectomy. However, assessment of coagulation tests revealed a prolonged prothrombin time, suggesting the moderate FVII deficiency. With a multidisciplinary consultation, we decided to performed total splenectomy with prophylaxis administration of totally 6 doses of active recombinant FVII, initiated 1 hour before surgery and followed until 30 hours postoperation. As a result of cautious undertaken in Mofid Children's Hospital, the patient did not experience any hemostatic defect. Patient is now 14-year-old, generally well-being under regular surveillance of FVII deficiency.

Pre-eclampsia is a common pregnancy disorder syndrome whose molecular mechanism is not clear. Nitric oxide (NO) is a key regulator of placentation. Reduction of NO has previously been associated with endothelial dysfunction in pre-eclamptic women. Therefore, we measured expression and methylation of some placental genes that were involved in NO pathway like named ARG II, PRMT1 and DDAH2 in pre-eclampsia and normal pregnancies in order to determine whether impairment of expression of these genes in the pre-eclamptic placenta could contribute to development of disease. ARG II, PRMT1 expressions as well as DDAH2 expression and methylation, in placentas collected from 59 patients with preeclampsia and 40 normotensive pregnancies were measured using real-time PCR and methylation specific PCR, respectively. The relationship among ARG II, PRMT1 and DDAH2 expressions was analyzed statistically. ARG II expression was increased, PRMT1 expression was not significantly changed. DDAH2 expression was decreased and qualitative methylation patterns were 32/59 and 21/40 in placentas from patients with pre-eclampsia compared with control group, respectively. The alterations in ARG II and DDAH2 expressions in pre-eclampsia patients maybe correlated with decreased eNOS expression. These findings indicate that ARG II and DDAH2 may be involved in pre-eclampsia pathogenesis and could be potential therapeutic targets for this disease.

The observed line-of-sight velocity dispersion $\sigma_{los}$ of the ultra diffuse galaxy Dragonfly 44 (DF44) requires a Newtonian dynamical mass-to-light ratio of $M_{dyn}/L_I=26^{+7}_{-6}$ Solar units. This is well outside the acceptable limits of our stellar population synthesis (SPS) models, which we construct using the integrated galactic initial mass function (IGIMF) theory. Assuming DF44 is in isolation and using Jeans analysis, we calculate $\sigma_{los}$ profiles of DF44 in Milgromian dynamics (MOND) and modified gravity (MOG) theories without invoking dark matter. Comparing with the observed kinematics, the best-fitting MOND model has $M_{dyn}/L_I = 3.6_{-1.2}^{+1.6}$ and a constant orbital anisotropy of $\beta=-0.5_{-1.6}^{+0.4}$. In MOG, we first fix its two theoretical parameters $\alpha$ and $\mu$ based on previous fits to the observed rotation curve data of The HI Nearby Galaxy Survey (THINGS). The DF44 $\sigma_{los}$ profile is best fit with $M_{dyn}/L_I=7.4_{-1.4}^{+1.5}$, larger than plausible SPS values. MOG produces a $\sigma_{los}$ profile for DF44 with acceptable $M_{dyn}/L_I$ and isotropic orbits if $\alpha$ and $\mu$ are allowed to vary. MOND with the canonical $a_0$ can explain DF44 at the $2.40 \sigma$ confidence level (1.66\%) if considering both its observed kinematics and typical star formation histories in an IGIMF context. It has been well documented that preeclampsia (PE) has a common etiological background, but little is known about its linkage at the molecular level.Non- coding RNAs are critical posttranscriptional regulators ofgene expression. This study was performed to determine whether PE is associated with alterations in placental non-coding RNAs expression. MicroRNA (miR)-155-5p and long non-coding RNA (lnc)sONE expression, in placentas collected sequentially from 59 patients with PE and 40 normotensive pregnancies were measured using real-time PCR.The relationship between miR-155-5p and lncsONE expressions was analyzed statistically. miR-155-5p expression was increased (fold change =1.6, P=0.04), while lncsONE expression was not significantly changed (fold change =1.1, P=0.68), in placentas from patients compared with control group.miR-155-5p was upregulated in placentas from patients with PE and may have influenced eNOS expression. These findings indicate that miRNA-155-5p may be involved in PE pathogenesis and could be a potential biomarker for this disease.

Regarding the increasing tendency to travel in recent years, many cities offer more amusement plans to have the tourists visit them again. The importance of these plans is due to maintaining the current tourists, but not attracting the new ones. Hence, recognizing loyal tourists can reduce the expenses and flourish the tourism. Accordingly, in this research, we have attempted to develop a new model based on data mining techniques to reconnoiter loyal tourists more accurately. The presented model is named DFR and is based on the development of the well-known model RFM. In addition to DFR model, the Imperialist Competitive Algorithm (ICA) has been used to cluster the tourists. The DFR model was implemented on 1100 tourists visiting Shiraz between 2013 and 2015, show that this model has a higher accuracy than other existing models and has identified loyal tourists more precisely.

In the present era, marketing based on some environmental and social considerations is raised as one of the most important activity of good and service production companies, and green marketing is one of the marketing areas that in environmental issues, many topics have been allocated to it. The purpose of this study is to develop a structural model for investigating the relationship between market orientation and environmental performance and green supply chain management strategies. The research method is a descriptive of type of survey and target population is employees and managers of Khalessazan Rooy Company in Zanjan. A questionnaire was used to collect data and was distributed among 215 employees and managers of the different levels of the company through random stratified sampling. In this research, the modeling of structural equations and partial least squares (PLS) were used to test the hypotheses and fit the model. The results showed that market orientation was effective on the supply chain management strategies. But its impact on environmental performance was rejected. On the other hand, the indirect effect of market orientation has been confirmed through the intermediate variable of supply chain management strategies. Finally, it was revealed that supply chain management approaches have a positive and significant effect on environmental performance.
