Objective: To determine how biochanin-A (BCA) affects highfat diet and streptozotocin-induced pathological changes in lipid metabolism and antioxidant status in diabetic rats. Methods: Diabetic rats were orally administered BCA (10 mg/kg body weight) for 30 days to investigate its effects on lipid profiles and oxidative stress markers in the liver and kidney. In addition, the mRNA expression of antioxidant and lipid metabolism enzymes in the liver was examined. Results: BCA attenuated hyperlipidemia by regulating mRNA expressions of HMG-CoA reductase, fatty acid synthase, carnitine palmitoyl transferase, and acetyl-CoA carboxylase. Additionally, BCA reduced high-fat diet and streptozotocin-induced oxidative stress by suppressing lipid peroxidation, improving superoxide dismutase, catalase, and glutathione peroxidase levels, and upregulating mRNA expressions of these enzymes. Conclusions: BCA may be a promising nutraceutical for the treatment of dyslipidemia and oxidative stress associated with diabetes. [ABSTRACT FROM AUTHOR]

In the present study, we evaluated the effect of biochanin A (BCA) on high-fat diet (HFD)-induced obesity cardiomyopathy. BCA (10 mg/kg body weight) was administered to HFD-induced obese rats for 30 days, and its effect on anthropometrical, morphological, plasma cardiac, and inflammatory biomarkers, along with cardiac lipid profiles was assessed. Supplementation of HFD to rats significantly increased body mass index, obesity index parameters, and cardiac lipid profile along with notable oxidative stress and inflammation. Additionally, BCA treatment in obese rats demonstrated a superior therapeutic action by restoring the altered parameters to almost normal levels. Simultaneously, BCA increased the activities and mRNA expressions of enzymatic antioxidants by upregulating the Nrf-2 pathway and inhibiting the NF-κB cascade. BCA may attenuate obesity and its associated cardiomyopathy by suppressing oxidative stress and inflammation through activation of the Nrf-2 pathway and inhibition of NF-κB activation. In the current study, we evaluated the ameliorative effect of S-allylcysteine (SAC) against streptozotocin (STZ)-nicotinamide (NAD)-induced diabetic nephropathy (DN) in rats and also an attempt was made to establish the molecular mechanism of SAC. DN rats were orally supplemented with SAC (150 mg/kg body weight) for a period of 45 days and the effect of SAC on urinary albumin excretion, metabolic parameters, and tubular injury biomarkers by ELISA, total levels and phosphorylation of MEK1/2, ERK1/2, and RSK2 by western blotting analysis in control and experimental rats were assessed. From this study, we observed that SAC considerably decreased polydipsia, poly urea, polyphagia, albuminuria and the levels of urinary N-acetyl-beta-D-glucosaminidase, neutrophil gelatinase-associated lipocalin, transforming growth factor-β1 and SAC effectively altered the pathological changes in DN rats. SAC also reserved renal cortical phosphorylation of MEK1/2, ERK1/2 and RSK2. Hence this study recommended that SAC can successfully protect the DN through regulation of MEK1/2-ERK1/2-RSK2 signalling. In the current study, we evaluated the effects of Asiatic acid (AA) on lipid metabolic markers in HFD-induced obese Sprague-Dawley rat model. AA (20 mg/kg BW) was administered orally to HFD-fed rats for 42 days. Changes in body composition, glucose, insulin resistance (IR) and lipid profiles of tissues, plasma and the pattern of gene expression of peroxisome proliferator-activated receptor-γ (PPAR-γ) and its target genes fatty-acid synthase (FAS), adipocyte protein-2 (aP2) and uncoupling protein-2 (UCP-2) and pro-inflammatory factor tumor necrosis factor (TNF)-α were observed in experimental rats. Oral administration of AA exerts therapeutic effects similar to orlistat in attenuating body weight gain, glucose, IR, plasma and tissue lipids and mRNA levels of PPAR-γ, FAS, aP2 and inflammatory factor TNF-α and increasing UCP-2 expression in HFD-fed rats. Hence, these findings concluded that AA attenuate HFD-induced obesity by modulating PPAR-γ and its target genes and regulate lipid metabolism, suggesting their possible antiobesity effects. Abstract: The present study evaluated the effects of asiatic acid (AA), a pentacyclic triterpenoid from Centella asiatica on lipid metabolism parameters in a rat model of obesity induced using a high fat diet (HFD) for 42 days. AA (20 mg/kg body weight [BW]) was administered orally once daily for 42 days, and an orlistat-treated group of rats (10 mg/kg BW) was included for comparison. Changes in BW, blood glucose levels, insulin resistance and leptin, adiponectin, amylase, and lipase levels in the blood; lipid profiles of plasma; liver antioxidants levels; and acetyl CoA carboxylase(ACC), uncoupling protein-2 (UCP2), and carnitine palmitoyltransferase-1 (CPT1) mRNA expression were observed in the experimental rats. Our results revealed that AA (20 mg/kg BW), similar to orlistat, reduced the increase in BW; increased bone mineral contents and bone mineral densities; reduced blood glucose levels, insulin resistance, leptin, plasma lipid levels; increased adiponectin, amylase, lipase levels in the blood; showed antioxidant activity; and altered mRNA expression of lipid metabolism-related genes, including ACC, UCP 2, and CPT 1, in the HFD-fed rats. From these results, we concluded that AA possesses significant anti-obesity potential through the suppression of BW gain, lipid lowering action, development of insulin and leptin sensitivity, antioxidant activity, and increased mRNA expression of lipid metabolism-related genes.Graphical Abstract: [ABSTRACT FROM AUTHOR]

Purpose: The aim of this paper is to examine the relationship between bi-direction of work family enrichment and work related outcomes as job satisfaction and family related outcomes such as family satisfaction and life satisfaction. Design/methodology/approach: An exploratory research design was adopted using the sample of women faculty members working in private and government colleges in Vellore District. Structural equation modelling- PLS was used to find the relationship between bi-direction of work family enrichment and work and family related outcomes. Findings: The result shows that the direction W2FE is positively significant with job satisfaction and F2WE is not significant, W2FE is negatively related with family satisfaction but F2WE is positively related with family satisfaction and finally W2FE is not significant with life satisfaction and positively significant with life satisfaction. Practical implications: The organization has to provide work and family life policies like flexible working schedules, family-friendly programs to motivate women faculty members so that high involvement in work and family leads to job, family and life satisfaction. Originality/value: This study offers useful guidelines to educational institutions to improve the performance of the faculty members and also helps for individual satisfaction which provides good service to the society. In the present study, we assessed the therapeutic potential of Biochanin-A (BCA) (10 mg/kg BW/day) pretreatment for 30 days on lipid metabolic abnormalities, proinflammatory cytokines and matrix metalloproteinase expression in isoproterenol (ISO)-induced myocardial infarction (MI) in rats. We measured the potential role of BCA on tissue and circulatory lipid profiles as well as on lipid metabolic enzymes: serum inflammatory cytokines (TNF-α, IL-1α, IL-1β, IL-6 and MCP1) and serum Matrix Metalloproteinases (particularly, MMP-2 and MMP-9) together with mRNA expressions of TNF-α, IL-6, MMP-2 and MMP-9 by RT-PCR analysis. Administration of ISO to rats significantly distorted their lipid metabolism and augmented inflammatory process, MMP expression and proteolytic activity. In addition, pretreatment with BCA of ISO-induced MI rats significantly reestablished the altered lipid metabolism and concealed the inflammation of cytokines. BCA suppressed the expressions of proinflammatory cytokines and MMPs in ISO-induced MI in rats when compared to normal untreated MI rats. Hence, these results established that BCA could improve the pathological processes of myocardial remodeling which was confirmed by histopathology of heart in MI rats and might be an effective beneficial ingredient for the management of heart failure disorders.

Cucurbita maxima, is a therapeutic plant spread all over the world. The seed of C. maxima constitutes a large amount of alkaloids, phenolic compounds, Vitamin E, and other secondary metabolites. The experiment was performed in four setups (Control, Only H 2 O 2 , H 2 O 2  + 25 mg of alkaloid, H 2 O 2  + 50 mg of alkaloids). The enzymatic and nonenzymatic antioxidants and lipid peroxidation (LPO) were determined to analyze the antioxidant effect of partially purified alkaloids. The results showed that partially purified alkaloids from C. maxima seed reacts effectively on the erythrocytes and leukocytes antioxidant enzyme system when compared to the hydrogen peroxide group. The present results indicate that the alkaloids present in this plant can be used as a natural antioxidant for the pharmacological purposes. PRACTICAL APPLICATIONS: The C. maxima seed constitutes rich source of alkaloids, phenolic compounds, Vitamin E, and other secondary metabolites. The results obtained revealed that the purified alkaloids from C. maxima seed acts as natural antioxidants, which enhanced the potential to scavenge H 2 O 2 and aids in protecting the RBC and WBC cells. The presence of 27 alkaloids of which seven major alkaloids possessing higher medicinal properties like antioxidant, antimicrobial, anti-inflammatory, antitumor, anti-aging, anti-diabetic, anticancer, anti-malarial, analgesic, nematicide, pesticide, and hemolytic activity were determined from the GC-MS analysis of the extract. Alkaloids acts as major constituent in phytotheraphy and has wide range of clinical application in humans and animals. In this study, we made an attempt to evaluate the potential of Cucurbita maxima seeds oil (CSO) against high-fat diet (HFD)-induced obesity in rats. We investigated the effect of CSO (100 mg/kg body weight) supplementation over 30 days on the changes of HFD-induced obese rats in body weight, biochemical parameters and lipid profile as well as investigated the effects of CSO on the histopathological changes. Oral administration with CSO revealed significant diminution in body weight gain, glucose and insulin levels, which altered the activity of lipid profile and restored the pathological alterations. It demonstrated that CSO had considerably altered these parameters when evaluated with HFD control rats. In conclusion, this study established that CSO prevents the HFD-induced obesity by altering the markers important to lipid metabolism.

Unlabelled: Ripe and unripe exotic pepino fruit were evaluated for antioxidant activity, total phenols, and flavonoid content. The antioxidant potency was investigated by employing various established in vitro systems, such as 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2-2'-azinobis(3-ethylbenthiazoline-6-sulphonic acid (ABTS), hydroxyl radical scavenging, reducing power, ferrous ion chelation, ferric reducing antioxidant power (FRAP), and lipid peroxidation. The EC(50) values of ripe ethanolic extract on DPPH radical, reducing power, ferrous ion chelation, ABTS radical, FRAP, hydroxyl radical, lipid peroxidation (brain), and lipid peroxidation (liver) were obtained to be 2.20, 2.81, <5.00, 34.06, 8.53, 1.30, 1.75, and 0.51 mg/mL, respectively. However, the EC(50) values for unripe fruit extract were noted to be 3.75, 3.40, 11.25, 40.12, 9.75, 0.80, 1.91, and 0.63 mg/mL, respectively. Ripe fruit exhibited the highest values of antioxidant activity in all the scavenging assays except for hydroxyl radical scavenging assay. Ripe pepino had higher total phenol and flavonoid content than unripe fruit. This study suggests that possible mechanism of the biological activities may be due to free radical scavenging and antioxidant characteristics, which may be due to the presence of polyphenols in the fruit extracts., Practical Application: The ripe and unripe pepino fruit have excellent antioxidant properties, so the results obtained in this study clearly indicate that pepino fruit has a significant potential to use as a natural antioxidant agent and possibly as a food supplement., (© 2012 Institute of Food Technologists®)
