4. Structure of diagnostics horizons and humus classification

Abstract

The classification of the main humus forms is generally based on the morpho-genetic characters of the A and OH diagnostic horizons. This is the case in the new European key of classification presented in Freiburg on September 2004 (Eurosoil Congress). Among the morpho-genetic characters, the soil structure covers a very important role. In this work, the structure of the diagnostic A and OH horizons has been analysed in terms of aggregation force, diameter and composition of the soil lumps (peds). In order to study the aggregation force, two disaggregating tools have been conceived and used. The diameter of the lumps has been measured by sieving the soil samples with standardised webs. Observing the samples thanks to a binocular magnifying 10X and 50X, the organic or/and mineral composition of the soil aggregates has been determined, data being investigated with ANOVA and Factorial Analysis. The article examines the argument from two points of view: crashing tools for estimating the soil structure (part 1) and the dimensions of the peds given in European key of humus forms classification (part 2). The categories of soil peds diameter and composition seem to be linked to the main humus forms. For instance, aggregates having a diamater larger than 1 mm and well amalgamate organo-mineral composition are more present in the A horizons of the Mull forms than in which of the other forms; contrary to the OH horizon of the Moder or Mor, the OH horizon of the Amphi forms shows an important percent of small organic lumps. Some propositions have been given in order to improve the European key of humus forms classification.

5. Uncoordinated transcription and compromised muscle function in the lmna-null mouse model of Emery- Emery-Dreyfuss muscular dystrophy.

Abstract

LMNA encodes both lamin A and C: major components of the nuclear lamina. Mutations in LMNA underlie a range of tissue-specific degenerative diseases, including those that affect skeletal muscle, such as autosomal-Emery-Dreifuss muscular dystrophy (A-EDMD) and limb girdle muscular dystrophy 1B. Here, we examine the morphology and transcriptional activity of myonuclei, the structure of the myotendinous junction and the muscle contraction dynamics in the lmna-null mouse model of A-EDMD. We found that there were fewer myonuclei in lmna-null mice, of which ∼50% had morphological abnormalities. Assaying transcriptional activity by examining acetylated histone H3 and PABPN1 levels indicated that there was a lack of coordinated transcription between myonuclei lacking lamin A/C. Myonuclei with abnormal morphology and transcriptional activity were distributed along the length of the myofibre, but accumulated at the myotendinous junction. Indeed, in addition to the presence of abnormal myonuclei, the structure of the myotendinous junction was perturbed, with disorganised sarcomeres and reduced interdigitation with the tendon, together with lipid and collagen deposition. Functionally, muscle contraction became severely affected within weeks of birth, with specific force generation dropping as low as ∼65% and ∼27% of control values in the extensor digitorum longus and soleus muscles respectively. These observations illustrate the importance of lamin A/C for correct myonuclear function, which likely acts synergistically with myotendinous junction disorganisation in the development of A-EDMD, and the consequential reduction in force generation and muscle wasting.

6. Integrated functions of Pax3 and Pax7 in the regulation of proliferation, cell size and myogenic differentiation.

Abstract

Pax3 and Pax7 are paired-box transcription factors with roles in developmental and adult regenerative myogenesis. Pax3 and Pax7 are expressed by postnatal satellite cells or their progeny but are down regulated during myogenic differentiation. We now show that constitutive expression of Pax3 or Pax7 in either satellite cells or C2C12 myoblasts results in an increased proliferative rate and decreased cell size. Conversely, expression of dominant-negative constructs leads to slowing of cell division, a dramatic increase in cell size and altered morphology. Similarly to the effects of Pax7, retroviral expression of Pax3 increases levels of Myf5 mRNA and MyoD protein, but does not result in sustained inhibition of myogenic differentiation. However, expression of Pax3 or Pax7 dominant-negative constructs inhibits expression of Myf5, MyoD and myogenin, and prevents differentiation from proceeding. In fibroblasts, expression of Pax3 or Pax7, or dominant-negative inhibition of these factors, reproduce the effects on cell size, morphology and proliferation seen in myoblasts. Our results show that in muscle progenitor cells, Pax3 and Pax7 function to maintain expression of myogenic regulatory factors, and promote population expansion, but are also required for myogenic differentiation to proceed.

7. Further characterisation of the molecular signature of quiescent and activated mouse muscle satellite cells.

Abstract

Satellite cells are the resident stem cells of adult skeletal muscle. To date though, there is a paucity of native markers that can be used to easily identify quiescent satellite cells, with Pax7 probably being the best that is currently available. Here we have further characterized a number of recently described satellite cell markers, and also describe novel ones. Caveolin-1, integrin alpha7 and the calcitonin receptor proved reliable markers for quiescent satellite cells, being expressed by all satellite cells identified with Pax7. These three markers remained expressed as satellite cells were activated and underwent proliferation. The nuclear envelope proteins lamin A/C and emerin, mutations in which underlie Emery-Dreifuss muscular dystrophy, were also expressed in both quiescent and proliferating satellite cells. Conversely, Jagged-1, a Notch ligand, was not expressed in quiescent satellite cells but was induced upon activation. These findings further contribute to defining the molecular signature of muscle satellite cells.

8. Illegal Self-Emancipation in the Urban Upper South, 1800-1860

Abstract

Contrary to common assumptions that self-emancipation by flight was only possible to regions outside the southern states, this article argues that many slaves actively took and preserved their freedom by hiding amongst free African American populations in urban areas. In the late eighteenth and early nineteenth centuries, free black communities within the slaveholding southern states emerged or were bolstered as a result of an increase in manumissions. For many African-American slaves, it was an age of emancipation. Yet for most enslaved people living in the US South it was a period of intensification and expansion of human bondage. The developments of the time provided more slaves with new opportunities to escape slavery by fleeing to free black communities.The concept of illegal freedom will be applied, which stands in contrast to the legal freedom that could be obtained on free soil. Few scholars have examined the various strategies employed by self-emancipators to remain concealed from the authorities. This article concentrates on runaway slaves in Baltimore and Richmond, two cities which had large African-American populations. The case will be made that enslaved African Americans carved new spaces of freedom within southern cities, with the assistance of free black communities. It will demonstrate that urban centers within the Upper South were important spaces of illegal freedom for slave refugees, largely because of their numerous free black populations, and that the two group’s experiences were deeply interwoven.

Abstract

Edible electronics is an emerging research field targeting electronic devices that can be safely ingested and directly digested or metabolized by the human body. As such, it paves the way to a whole new family of applications, ranging from ingestible medical devices and biosensors to smart labelling for food quality monitoring and anti-counterfeiting. Being a newborn research field, many challenges need to be addressed to realize fully edible electronic components. In particular, an extended library of edible electronic materials is required, with suitable electronic properties depending on the target device and compatible with large-area printing processes, to allow scalable and cost-effective manufacturing. In this work, we propose a platform for future low-voltage edible transistors and circuits that comprises an edible chitosan gating medium and inkjet-printed inert gold electrodes, compatible with low thermal budget edible substrates, such as ethylcellulose. We report the compatibility of the platform, characterized by critical channel features as low as 10 mu m, with different inkjet-printed carbon-based semiconductors, including biocompatible polymers present in the picogram range per device. A complementary organic inverter is also demonstrated with the same platform as a proof-of-principle logic gate. The presented results offer a promising approach to future low-voltage edible active circuitry, as well as a testbed for non-toxic printable semiconductors.

10. A n-type, Stable Electrolyte Gated Organic Transistor Based on a Printed Polymer

Abstract

Electrolyte-gated organic transistors (EGOTs) are promising and versatile devices for next-generation biosensors, neuromorphic systems, and low-voltage electronics. They are particularly indicated for applications where stable operation in aqueous environment and cost-effective manufacturing are required. Indeed, EGOTs can be fabricated through low-cost, large area, and scalable techniques, such as printing, from a large portfolio of solution processable organic materials, which are often able to stably operate in water or physiological solutions. Despite a large number of solution processable EGOTs have been reported in the literature so far, only a few are based on printed semiconductors, with no examples of digitally printed, i.e., inkjet printed, n-type devices, which would easily enable complementary architectures. In this work, we propose the first example of a n-type electrolyte gated organic transistor based on an inkjet printed polymer. The proposed device shows a high stability when operated in water and requires only 3 hours of conditioning to produce a stable response, a much faster dynamic than in the case of printed polymers currently tested for p-type EGOTs. As a proof-of-concept, the proposed printed n-type EGOT is successfully integrated with a printed single-walled carbon-nanotubes based p-type device in a logic inverter, demonstrating the possibility to build simple water-gated digital electronic circuits.

Abstract

Ambient energy harvesting has great potential to contribute to sustainable development and address growing environmental challenges. Converting waste energy from energy-intensive processes and systems (e.g. combustion engines and furnaces) is crucial to reducing their environmental impact and achieving net-zero emissions. Compact energy harvesters will also be key to powering the exponentially growing smart devices ecosystem that is part of the Internet of Things, thus enabling futuristic applications that can improve our quality of life (e.g. smart homes, smart cities, smart manufacturing, and smart healthcare). To achieve these goals, innovative materials are needed to efficiently convert ambient energy into electricity through various physical mechanisms, such as the photovoltaic effect, thermoelectricity, piezoelectricity, triboelectricity, and radiofrequency wireless power transfer. By bringing together the perspectives of experts in various types of energy harvesting materials, this Roadmap provides extensive insights into recent advances and present challenges in the field. Additionally, the Roadmap analyses the key performance metrics of these technologies in relation to their ultimate energy conversion limits. Building on these insights, the Roadmap outlines promising directions for future research to fully harness the potential of energy harvesting materials for green energy anytime, anywhere.

The objective of this paper is to illustrate an alternative control algorithm for power converters which have the task of introducing the energy generated by systems based on the use of renewable sources such as photovoltaic or wind power, into small-sized networks, microgrids. Virtual Synchronous Generator can operate both in parallel to the main network as well as in isolated and autonomous conditions (islanding-mode). Abstract

The increasing penetration of Renewable Energy (RE) into the electrical market is desirable in terms of sustainability. Nevertheless, it is a challenge that all the interested actors shall address from both the technical and economical points of view. This paper provides an overview of the main challenges and solutions towards the technological transition to an electrical system with 100% renewable energy sources in terms of innovations and operative limits of the traditional systems. INTRODUCTION The purpose was to compare two flexible regimens of intravitreal aflibercept (IVT-AFL) with fixed dosing every 8 weeks, beyond the first year of treatment, in patients with diabetic macular edema (DME). VIOLET was a 100-week, randomized, Phase IIIb, non-inferiority study in patients with center-involving DME previously treated with IVT-AFL for ≥ 1 year according to the European label. METHODS Patients received an initial dose of IVT-AFL at study baseline and were randomly assigned (1:1:1) to treat-and-extend (T&E), pro re nata (PRN), or fixed regimens. The primary endpoint was mean change in best-corrected visual acuity (BCVA) from baseline (randomization) to Week 52. RESULTS Full analysis set comprised 458 patients (baseline mean BCVA: 72.5, 71.0, and 72.7 letters in the T&E, PRN, and fixed-dose groups, respectively). Patients received a mean (min-max) of 10.0 (2-14; T&E), 11.5 (1-25; PRN), and 12.3 (3-13; fixed) injections over 100 weeks, with 13.3 (4-23), 25.0 (3-29), and 16.1 (5-25) clinic visits, respectively. Abstract

Green roofs are widely recognized as powerful nature-based solutions, which can provide multiple benefits for the sustainable development of urban areas (e.g., pluvial flood mitigation, thermal building insulation, biodiversity increase, air pollution reduction, etc.). Although the potential benefits of these tool have been widely investigated, the social awareness and citizens’ willingness to pay for green roof, on both private and public buildings, have not been assessed yet, especially in the Mediterranean areas. Citizens’ perception and opinion are particularly relevant to support the decisions of policy makers and urban planners and could determine the actual implementation of green roofs in the urban environment. In this work, we investigated the interest and willingness to pay for green roofs in the region of Sardinia (Italy), analyzing the outcomes of an anonymous online survey, distributed via the mailing list of the University of Cagliari and social media. Results highlights a realistic perception of the citizens regarding the most common environmental issues (i.e., urban flood, increase of temperature, energy consumption, air pollution and lack of green spaces) and the potential benefits of green roof installation. Sardinian citizens showed high interest in having green roofs on public buildings, with an average willingness to pay of about 35 euro per person every year. On the other hand, the interest and willingness to pay for green roof installation on private buildings is limited, due to the high construction and maintenance costs and to the preference for photovoltaic systems as roof coverage., The 28th IUGG General Assembly (IUGG2023) (Berlin 2023)

Abstract

Recently, sensor networks effectively provide many applications in different fields. The deployment of mobile sensors, unmanned aerial vehicles is to reduce the burden of energy consumption for sensor nodes. However, saving energy in such networks is still a critical issue. This paper investigates the performance of a hybrid RF-Solar harvesting circuit for remote sensing devices. The harvesting circuit can simultaneously harvest power from solar and radio frequency (RF) sources readily available in the surrounding environment. The proposed work builds RF and solar harvester circuits to create hybrid harvesting circuits for all elements in the combined sensing networks. The stand-alone RF harvester circuit is a dual-band multi-stage harvester that is designed to work at 2.4GHz, Wi-Fi/WLAN bands. The standalone solar harvester circuit comprises a solar panel with a maximum power point tracking (MPPT) algorithm. The whole hybrid system can produce a maximum power of up to 133.25W with a boost current element in the charging system. Since each node is equipped with a rechargeable battery, all the batteries are charged by the harvested power from the proposed circuits. This approach supports the operation of the networks safe and continuous even when a shortage of harvested power happens due to bad conditions such as cloudy or rainy days. This work shows promise and is applicable.

Abstract

Partial discharges (PD) are localized breakdowns in the insulation within high-voltage (HV) equipment and can be a warning sign of potential future failure. Regular PD monitoring through preventive maintenance is essential to the reliability of many expensive HV apparatuses. Hence, the purpose of this work is to assess the performance of a novel class of multi-turn inductive loop sensors based on printed circuit board (PCB) to detect PD in free space. Based on their topology, three distinct inductive sensors, named meander, non-spiral, and spiral, are evaluated towards detecting in-lab corona, internal, and surface PDs. Through simulations and measurements, the low-frequency lumped-element circuit model for each of the three magnetic field probes was extracted. Experimental PRPD patterns showed that the three evaluated inductive loops could be effectively applied for online monitoring and recognition of PDs. The experimental results show that the single PD pulses detected by the three sensors have a typical PD pulse shape that is a damped sinusoidal function with frequency spectra spanning from 14.84 MHz to 46 MHz. The spiral sensor yielded the maximum sensitivity, while the meander topology produced the lowest.

Abstract

The CHBMI performance optimization can be reached both with hardware and software solutions. Generally, asymmetrical configurations allow the extension of the voltage levels number and the implementation of an innovative modulation strategy allows for improving the performance in terms of harmonic distortion and conversion efficiency. This work is devoted to the development of a modulation strategy that allows improving the performance of an asymmetrical single-phase five-level Cascaded H-Bridge Inverter generating a 7-level voltage waveform. Thus, a digital approach and corresponding digital functions are presented. Subsequently, the impact of the dead time and the influence on the voltage harmonic distortion have been discussed. Through simulation analysis, in the Matlab environment, the main benefits and drawbacks of the proposed approach have been discussed.

Abstract

BACKGROUND: Faricimab is a bispecific antibody that acts through dual inhibition of both angiopoietin-2 and vascular endothelial growth factor A. We report primary results of two phase 3 trials evaluating intravitreal faricimab with extension up to every 16 weeks for neovascular age-related macular degeneration (nAMD). METHODS: TENAYA and LUCERNE were randomised, double-masked, non-inferiority trials across 271 sites worldwide. Treatment-naive patients with nAMD aged 50 years or older were randomly assigned (1:1) to intravitreal faricimab 6·0 mg up to every 16 weeks, based on protocol-defined disease activity assessments at weeks 20 and 24, or aflibercept 2·0 mg every 8 weeks. Randomisation was performed through an interactive voice or web-based response system using a stratified permuted block randomisation method. Patients, investigators, those assessing outcomes, and the funder were masked to group assignments. The primary endpoint was mean change in best-corrected visual acuity (BCVA) from baseline averaged over weeks 40, 44, and 48 (prespecified non-inferiority margin of four letters), in the intention-to-treat population. Safety analyses included patients who received at least one dose of study treatment. These trials are registered with ClinicalTrials.gov (TENAYA NCT03823287 and LUCERNE NCT03823300). FINDINGS: Across the two trials, 1329 patients were randomly assigned between Feb 19 and Nov 19, 2019 (TENAYA n=334 faricimab and n=337 aflibercept), and between March 11 and Nov 1, 2019 (LUCERNE n=331 faricimab and n=327 aflibercept). BCVA change from baseline with faricimab was non-inferior to aflibercept in both TENAYA (adjusted mean change 5·8 letters [95% CI 4·6 to 7·1] and 5·1 letters [3·9 to 6·4]; treatment difference 0·7 letters [-1·1 to 2·5]) and LUCERNE (6·6 letters [5·3 to 7·8] and 6·6 letters [5·3 to 7·8]; treatment difference 0·0 letters [-1·7 to 1·8]). Abstract

Background/objectives: To compare the number of eye surgical procedures performed in Italy in the 2 months following the beginning of lockdown (study period) because of COVID-19 epidemic with those performed in the two earlier months of the same year (intra-year control) and in the period of 2019 corresponding to the lockdown (inter-year control). Methods: Retrospective analysis of surgical procedures carried out at 39 Academic hospitals. A distinction was made between elective and urgent procedures. Intravitreal injections were also considered. Percentages for all surgical procedures and incidence rate ratios (IRR) for rhegmatogenous retinal detachment (RRD) events were calculated. A p value <0.05 was considered significant. Results: A total of 20,886 versus 55,259 and 56,640 patients underwent surgery during the lockdown versus intra-and inter-year control periods, respectively. During the lockdown, only 70% of patients for whom an operation/intravitreal injection was recommended, finally underwent surgery; the remaining patients did not attend because afraid of getting infected at the hospital (23%), taking public transportation (6.5%), or unavailable swabs (0.5%). Elective surgeries were reduced by 96.2% and 96.4%, urgent surgeries by 49.7% and 50.2%, and intravitreal injections by 48.5% and 48.6% in the lockdown period in comparison to intra-year and inter-year control periods, respectively. IRRs for RRDs during lockdown dropped significantly in comparison with intra- and inter-year control periods (CI: 0.65–0.80 and 0.61–0.75, respectively, p < 0.001 for both). Conclusion: This study provides a quantitative analysis of the reduction of eye surgical procedures performed in Italy because of the COVID-19 epidemic.

Abstract

Abstract Background Lung adenocarcinoma (LUAD) has been observed to have significant sex differences in incidence, prognosis, and response to therapy. However, the molecular mechanisms responsible for these disparities have not been investigated extensively. Methods Sample-specific gene regulatory network methods were used to analyze RNA sequencing data from non-cancerous human lung samples from The Genotype Tissue Expression Project (GTEx) and lung adenocarcinoma primary tumor samples from The Cancer Genome Atlas (TCGA); results were validated on independent data. Results We found that genes associated with key biological pathways including cell proliferation, immune response and drug metabolism are differentially regulated between males and females in both healthy lung tissue and tumor, and that these regulatory differences are further perturbed by tobacco smoking. We also discovered significant sex bias in transcription factor targeting patterns of clinically actionable oncogenes and tumor suppressor genes, including AKT2 and KRAS. Using differentially regulated genes between healthy and tumor samples in conjunction with a drug repurposing tool, we identified several small-molecule drugs that might have sex-biased efficacy as cancer therapeutics and further validated this observation using an independent cell line database. Conclusions These findings underscore the importance of including sex as a biological variable and considering gene regulatory processes in developing strategies for disease prevention and management.
