To remove the hazardous organic adulterants from the wastewater, iron tungstate nanoparticles were prepared by a simple wet chemical route. These nanoparticles characterized to be polydispersed, polycrystalline with irregular spherical morphologies without any notable impurities were used as a catalyst for heterogeneous Fenton reaction, an advanced oxidation process. Their catalytic properties, degradation efficiency, and recyclability were analyzed for methyl orange degradation, a common -reactive azo dye in an acidic aqueous. The rapid fall of absorption peak exhibits the complete degradation of dye within 50 minutes of reaction with discoloration efficiency of ~97% without any additional external exposures such as light or ultrasonic waves. In addition to this, the durability of the FeWO4 nanoparticles catalyst is proved to be maintained over five catalyst reuse cycles, which strongly suggests that at this present scenario of ecological deterioration, the chosen nanomaterial is one of the apt candidates for azo dye removal by heterogeneous dark Fenton process.

The SIN3 transcriptional coregulator influences gene expression through multiple interactions that include histone deacetylases (HDACs). Haploinsufficiency and mutations in SIN3 are the underlying cause of Witteveen-Kolk syndrome and related intellectual disability (ID)/autism syndromes, emphasizing its key role in development. However, little is known about the diversity of its interactions and functions in developmental processes. Here we show that loss of SIN-3, the single SIN3 homologue inCaenorhabditis elegans, results in maternal effect sterility associated with deregulation of the germline transcriptome, including desilencing of X-linked genes. We identify at least two distinct SIN3 complexes containing specific HDACs, and show that they differentially contribute to fertility. Single cell smFISH reveals that insin-3mutants, the X chromosome becomes re-expressed prematurely and in a stochastic manner in individual germ cells. Furthermore, we identify histone residues whose acetylation increases in the absence of SIN3. Together, this work provides a powerful framework for thein vivostudy of SIN3 and associated proteins.

The subphylum Saccharomycotina is a lineage in the fungal phylum Ascomycota that exhibits levels of genomic diversity similar to those of plants and animals. The Saccharomycotina consist of more than 1 200 known species currently divided into 16 families, one order, and one class. Species in this subphylum are ecologically and metabolically diverse and include important opportunistic human pathogens, as well as species important in biotechnological applications. Many traits of biotechnological interest are found in closely related species and often restricted to single phylogenetic clades. However, the biotechnological potential of most yeast species remains unexplored. Although the subphylum Saccharomycotina has much higher rates of genome sequence evolution than its sister subphylum, Pezizomycotina, it contains only one class compared to the 16 classes in Pezizomycotina. The third subphylum of Ascomycota, the Taphrinomycotina, consists of six classes and has approximately 10 times fewer species than the Saccharomycotina. These data indicate that the current classification of all these yeasts into a single class and a single order is an underappreciation of their diversity. Our previous genome-scale phylogenetic analyses showed that the Saccharomycotina contains 12 major and robustly supported phylogenetic clades; seven of these are current families (Lipomycetaceae, Trigonopsidaceae, Alloascoideaceae, Pichiaceae, Phaffomycetaceae, Saccharomycodaceae, and Saccharomycetaceae), one comprises two current families (Dipodascaceae and Trichomonascaceae), one represents the genus Sporopachydermia, and three represent lineages that differ in their translation of the CUG codon (CUG-Ala, CUG-Ser1, and CUG-Ser2). Using these analyses in combination with relative evolutionary divergence and genome content analyses, we propose an updated classification for the Saccharomycotina, including seven classes and 12 orders that can be diagnosed by genome content. This updated classification is consistent with the high levels of genomic diversity within this subphylum and is necessary to make the higher rank classification of the Saccharomycotina more comparable to that of other fungi, as well as to communicate efficiently on lineages that are not yet formally named.

International audience; Fungal laccases are oxidoreductases with low-specificity for substrates. The characterization of laccase’s surface is a prerequisite used to obtain hybrid catalysts with new properties. Surface-exposed lysine residues are targets in immobilization reactions. In this work, LAC3-K0, an enzyme devoid of lysine, was used as a platform to detect potential surface-exposed sites suitable for replacement with a lysine residue. Seven sites were selected from a LAC3-K0 3-D model, and single lysine mutants (UNIKn, n = residue number) were obtained by site-directed mutagenesis. All mutants were expressed in Saccharomyces cerevisiae W303-1A and detected as functional secreted proteins by their ability to oxidize guaiacol or 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) on agar plates. All variants were active at acidic pH but presented no activity at neutral pH, as expected. Likewise, variants were stable a temperature between 15‒55°C, and were completely inactivated at 70°C. Oxidation assays revealed that the replacement of one or two surface residues with lysine greatly affected enzyme activity and substrate specificity. The catalytic parameters (KappM and kappcat) determined with ABTS were found to be different among the variants; the Vappmax was 1.5‒2 fold higher in UNIK269 and triple mutant, with a KappM of 0.27 and 0.30, respectively; kappcat was 30.25 in UNIK238 and 32.34 in the triple mutant. The role of hydrophobic patches detected on the surface of LAC3-K0 was determined to be a favorable factor to be considered in the interaction of hybrid materials. All variants with uniquely surface located lysine created in this work can be in demand for obtaining laccases with a certain substrate specificity in the design of hybrid materials.

PURPOSE: Preclinical data suggest the combination of an anti-programmed death receptor 1 antibody plus dabrafenib and trametinib to have superior antitumor activity compared with dabrafenib plus trametinib alone. These observations are supported by translational evidence suggesting that immune checkpoint inhibitors plus targeted therapy may improve treatment outcomes in patients with BRAF V600-mutant metastatic melanoma. COMBI-i is a phase III trial evaluating spartalizumab, an anti-programmed death receptor 1 antibody, in combination with dabrafenib and trametinib (sparta-DabTram), versus placebo plus dabrafenib and trametinib (placebo-DabTram) in patients with BRAF V600-mutant unresectable or metastatic melanoma. METHODS: Patients received spartalizumab 400 mg intravenously every 4 weeks plus dabrafenib 150 mg orally twice daily and trametinib 2 mg orally once daily or placebo-DabTram. Participants were age ≥ 18 years with unresectable or metastatic BRAF V600-mutant melanoma. The primary end point was investigator-assessed progression-free survival. Overall survival was a key secondary end point (ClinicalTrials.gov identifier: NCT02967692). RESULTS: At data cutoff (July 1, 2020), the median progression-free survival was 16.2 months (95\% CI, 12.7 to 23.9 months) in the sparta-DabTram arm versus 12.0 months (95\% CI, 10.2 to 15.4 months) in the placebo-DabTram arm (hazard ratio, 0.82 [95\% CI, 0.66 to 1.03]; P = .042 [one-sided; nonsignificant]). The objective response rates were 69\% (183 of 267 patients) versus 64\% (170 of 265 patients), respectively. Grade ≥ 3 treatment-related adverse events occurred in 55\% (146 of 267) of patients in the sparta-DabTram arm and 33\% (88 of 264) in the placebo-DabTram arm. CONCLUSION: The study did not meet its primary end point; broad first-line use of sparta-DabTram is not supported by these results. Further biomarker-driven investigation may identify patient subpopulations who could benefit from checkpoint inhibitor plus targeted therapy combinations.

Background: COMBI-AD demonstrated long-term benefit of adjuvant dabrafenib plus trametinib in patients with resected stage III BRAF V600E/K–mutant melanoma; however, 9% of patients permanently discontinued therapy due to pyrexia. COMBI-APlus evaluated whether an adapted pyrexia management algorithm reduces high-grade pyrexia and pyrexia-related adverse outcomes. Methods: COMBI-APlus is an open-label, phase IIIb trial evaluating an adapted pyrexia management algorithm in patients with high-risk resected stage III BRAF V600E/K–mutant melanoma treated with up to 12 months of adjuvant dabrafenib plus trametinib. Both drugs were interrupted for pyrexia (temperature ≥38°C) or the occurrence of pyrexia syndrome for suspected recurrent pyrexia. Treatment was restarted at the same dose once patients were symptom free for ≥24 h. The primary endpoint was the composite rate of grade 3/4 pyrexia, hospitalisation due to pyrexia, or permanent discontinuation due to pyrexia versus historical COMBI-AD control (20.0%; 95% confidence interval [CI], 16.3%–24.1%). Results: At data cutoff (5 October 2020), COMBI-APlus met its primary endpoint of significant improvement in the composite rate of pyrexia (8.0% [95% CI, 5.9%–10.6%]), with rates of 3.8% for grade 3/4 pyrexia, 4.3% for hospitalisation due to pyrexia, and 2.4% for discontinuation due to pyrexia. Estimated 12-month relapse-free survival was 91.8% (95% CI, 89.0%–93.9%). The most common adverse events were consistent with those in COMBI-AD, and 14.7% of patients permanently discontinued treatment due to adverse events. Conclusions: The adapted pyrexia management algorithm appears to reduce the incidence of severe pyrexia outcomes, enables patients to manage pyrexia at home, and helps patients remain on treatment. International audience; The volcanic-hydrothermal geo-diversity of the Basse-Terre Island of Guadeloupe archipelago (Eastern Caribbean, France) is a major asset of the Caribbean bio-geoheritage. In this paper, we use Guadeloupe as a representative of many small island developing states (SIDS), to show that the volcanic-hydrothermal geodiversity is a major resource and strategic thread for resilience and sustainability. These latter are related to the specific richness of Guadeloupe’s volcanic-geothermal diversity, which is de facto inalienable even in the wake of climate change and natural risks that are responsible for this diversity, i.e., volcanic eruptions. We propose the interweaving the specificity of volcanic-geothermal diversity into planning initiatives for resilience and sustainability. Among these initiatives research and development programs focused on the knowledge of geodiversity, biodiversity and related resources and risks are central for the long-term management of the water resource, lato sensu. Such a management should include a comprehensive scientific observatory for the characterization, exploration, and sustainable exploitation of the volcanic-hydrothermal geodiversity alongside planning for and mitigating geophysical risks related to sudden volcanic-induced phenomena and long-term systemic drifts due to climate change. The results of this exercise for Guadeloupe could typify innovative paths for similar SIDS around their own volcanic-hydrothermal geodiversity.

The geriatric population, often polymedicated, is exposed to the risk of adverse drug events. Medication reconciliation (MR), which is an interactive and pluriprofessional process, helps ensure continuity of care. The objective of this study was to analyze and to define relevant prioritization criteria for MR in older patients in order to avoid a maximum of medication errors.A clinical audit of MR at the transition points of patient admission and discharge was conducted prospectively for 10 months. Patients were selected on the basis of a prioritization procedure already established in our structure, that is the presence of at least one of the three following criteria: originating from an hospital department, severe renal failure and prescription of at-risk drugs.The cohort of patients reconciled at admission included 136 patients. A total of 63 unintentional discrepancies (UDs) were identified, the majority of which (76.2%) involved drug omissions. Three criteria were identified as independent predictors of UDs risk on the entry prescription compared to the optimized drug assessment: rheumatological history, originating from an hospital department and hyponatremia. Hyponatremia was found in the present study to be the most relevant criterion that significantly increased the risk of having an UD on the patient's prescription, particularly a risk of treatment omission at admission.This study will allow to improve the prioritization criteria on the healthcare establishment's procedure and to implement MR in geriatric day hospitalization in order to strengthen the city-hospital link.

Since July 2014, the ESA Gaia mission has been surveying the entire sky down to magnitude 20.7 in the visible. In addition to the millions of stars, thousands of Solar System Objects (SSOs) are observed daily. By comparing their positions to those of known objects, a daily processing pipeline filters known objects from potential discoveries. However, owing to Gaia's specific scanning law designed for stars, potential newly discovered moving objects are characterized by very few observations, acquired over a limited time. This aspect was recognized early in the design of the Gaia data processing. A daily processing pipeline dedicated to these candidate discoveries was set up to release calls for observations to a network of ground-based telescopes. Their aim is to acquire follow-up astrometry and to characterize these objects. From the astrometry measured by Gaia, preliminary orbital solutions are determined, allowing to predict the position of these potentially new discovered objects in the sky accounting for the large parallax between Gaia and the Earth (separated by 0.01 au). A specific task within the Gaia Consortium has been responsible for the distribution of requests for follow-up observations of potential Gaia SSO discoveries. Since late 2016, these calls for observations (called alerts) are published daily via a Web interface, freely available to anyone world-wide. Between November 2016 and July 2020, over 1700 alerts have been published, leading to the successful recovery of more than 200 objects. Among those, six have provisional designation assigned with the Gaia observations, the others being previously known objects with poorly characterized orbits, precluding identification at the time of Gaia observations. There is a clear trend for objects with a high inclination to be unidentified, revealing a clear bias in the current census of SSOs against high inclination populations., Submitted to Astronomy & Astrophysics

Mosquitoes (Diptera, Culicidae) form a family of insects of considerable public health importance. Mention of their presence/absence was tackled in the literature and by specialized websites for eight African countries: Cape Verde, Mauritania, Senegal, Gambia, Mali, Burkina Faso, Niger and Chad. A significant portion of the disproportionally high chemical weathering flux in volcanic island arcs may originate from hydrothermal fluid-rock interaction, thereby compromising the accurate estimate of atmospheric CO2 consumption rates. The objective of this study is to evaluate how the riverine Ge/Si ratio and Si isotopes, two well-established tracers of weathering, respond to hydrothermal inputs. The work took place in Basse-Terre, Guadeloupe, a tropical volcanic island with a dense river network, high chemical weathering fluxes and various hydrothermal surface manifestations. We characterized the Ge/Si ratio and δ30Si of 15 thermal springs, nine non-impacted (NI) rivers and 13 hydrothermally-impacted (HI) rivers. The soil solution from a highly weathered soil profile (Ferralsol) and a clayey-rock corresponding to the material exposed in an extinct hydrothermal system were also measured. A new purification method was successfully developed in order to allow the reliable measurement of Si isotopes in SO42−- and Cl−-rich thermal spring and HI river waters by mass spectrometry. Basse-Terre's thermal springs have variable Ge/Si ratios (0.05–21.03 μmol.mol−1) and δ30Si (0.71–1.50‰), but with no apparent relationship to the water compositional type. The Ge/Si ratio (0.15–2.57 μmol.mol−1) and Si isotope composition (0.26–1.21‰) values of the NI rivers reveal differences in the watersheds' weathering degree. Dissolution of Ge- and 28Si-rich secondary minerals explains the high Ge/Si and isotopically light composition of the northern NI rivers draining strongly weathered terranes. The Ge/Si ratio and δ30Si values measured for the NI and HI rivers overlap, implying that they cannot be used to diagnose hydrothermal contributions to river basins unambiguously. However, when combined with the Cl− and SO42− concentrations, the analysis of Ge and Si in the HI rivers suggests that water seeping through an extinct hydrothermal system produces SO4-rich drainages with distinctively lower Ge/Si ratios than those inferred for watersheds receiving thermal spring discharges associated with an active hydrothermal system. Overall, our results provide new constraints for applying and interpreting Ge/Si and Si isotope measurements to study weathering in volcanic environments.

CTD S2P mediates nutrient-dependent development., Serine 2 phosphorylation (S2P) within the CTD of RNA polymerase II is considered a Cdk9/Cdk12-dependent mark required for 3′-end processing. However, the relevance of CTD S2P in metazoan development is unknown. We show that cdk-12 lesions or a full-length CTD S2A substitution results in an identical phenotype in Caenorhabditis elegans. Embryogenesis occurs in the complete absence of S2P, but the hatched larvae arrest development, mimicking the diapause induced when hatching occurs in the absence of food. Genome-wide analyses indicate that when CTD S2P is inhibited, only a subset of growth-related genes is not properly expressed. These genes correspond to SL2 trans-spliced mRNAs located in position 2 and over within operons. We show that CDK-12 is required for maximal occupancy of cleavage stimulatory factor necessary for SL2 trans-splicing. We propose that CTD S2P functions as a gene-specific signaling mark ensuring the nutritional control of the C. elegans developmental program.

Volcanic gas emissions from the main low-temperature fumaroles (around 100°C) of one of the most dangerous volcanoes in the Lesser Antilles, La Soufrière de Guadeloupe, are measured routinely using direct sampling with NaOH bottles (for H2O, CO2, H2Stot, H2, CH4, CO, N2, He, Ar, O2), with P2O5 bottles (for H2O, CO2, H2S, SO2, H2) and MULTIGAS (for H2O, CO2, H2S, SO2, H2). This allows us to perform a study about an intercomparison of volcanic gas monitoring methologies. The results show some good overlap and similar temporal evolution between the dataset of the different methologies, which is of utmost of importance for monitoring purposes. However, there is no good agreement between the three methodologies for the same chemical ratio. The differences could be explained by a timedependant diffusive phenomenon that affects the concentrations of sampled gases for two methods of the three (P2O5 bottles and MULTIGAS). P2O5-based analyses are affected because effusion on rapid decompression and fast sampling affects the gas volume in the pipeline connecting the vent outlet to the sampling. Although MULTIGAS is a continuous, flow-through, sampling method, results are likely affected because of the sampling architecture involving filter and pump. Moreover, sulphur reaction (precipitation of elemental S upon exit from the fumarole, conversion of H2S to SO2, chemical reaction with the P2O5 powder that release H2S) and water condensation can also explain some discrepancies.

Deposition of histone H3 lysine 4 (H3K4) methylation at promoters by SET1/COMPASS is associated with context-dependent effects on gene expression and local changes in chromatin organization. Whether SET1/COMPASS also contributes to higher-order chromosome structure has not been investigated. Here, we address this question by quantitative FRET (Förster resonance energy transfer)-based fluorescence lifetime imaging microscopy (FLIM) onC. elegansgerm cells expressing histones H2B-eGFP and H2B-mCherry. We find that SET1/COMPASS subunits strongly influence meiotic chromosome organization, with marked effects on the close proximity between nucleosomes. We further show that inactivation of SET-2, theC. eleganshomologue of SET1, or CFP-1, the chromatin targeting subunit of COMPASS, strongly enhance chromosome organization defects and loss of fertility resulting from depletion of condensin-II. Defects in chromosome morphology resulting from conditional inactivation of topoisomerase II, another structural component of chromosomes, were also aggravated in the absence of SET-2. Combined, ourin vivofindings suggest a model in which the SET1/COMPASS histone methyltransferase complex plays a role in shaping meiotic chromosome in cooperation with the non-histone proteins condensin-II and topoisomerase.

In thirty years, bed nets treated with pyrethroids have been established as a means of prevention against malaria for personal protection and in public health. One of the actors of these discoveries gives an overview of insecticidetreated bed nets (ITNs) and brings his testimony, more particularly on the circumstances and frames of mind which prevailed in the 1980's, a crucial decade for these breakthroughs which served humanity. In 1983, the first experiment testing the effect of bed nets treated with pyrethroid insecticide was performed and revealed the impact of this method on the reduction of Plasmodium transmission. This discovery was made in an experimental field station in Burkina Faso showing that each of the many entomological parameters measured led to a clear decrease in mosquito-human contact. A few years later, still in Burkina Faso, it was shown that ITNs, massively used in a village, reduced the longevity and vector capacity of mosquito populations, to such an extent that ITNs acted as a means of vector control. These researches were all led within the Centre Muraz of the Organization of Coordination and Cooperation for the Control of Great Endemies (OCCGE) by researchers belonging to the Office of Overseas Scientific and Technical Research (ORSTOM, which later became French National Research Institute for Sustainable Development), and benefited from the scientific and financial support of the Division of Vector Biology and Control from the World Health Organization (WHO).En une trentaine d’années, les moustiquaires imprégnées de pyrétrinoïdes se sont imposées en protection personnelle et en santé publique comme un moyen de prévention du paludisme. Un des acteurs de ces découvertes présente un panorama des moustiquaires imprégnées d’insecticide et apporte ici son témoignage, se fondant particulièrement sur les circonstances et les états d’esprit qui ont prévalu dans les années 1980, décennie cruciale pour ces avancées qui ont servi l’humanité. C’est en 1983 qu’a été réalisée la première expérimentation testant l’effet des moustiquaires imprégnées de pyréthrinoïdes et qui a mis en évidence l’impact de cette méthode sur la réduction de la transmission du Plasmodium. Cette découverte a été faite dans une station expérimentale de terrain au Burkina Faso montrant que chacun des nombreux paramètres entomologiques mesurés allait dans le sens d’une nette réduction du contact moustique–humain. Quelques années plus tard, toujours au Burkina Faso, il a été montré que les moustiquaires imprégnées, utilisées massivement dans un village, réduisaient la longévité et la capacité vectrice de la population de moustiques, à tel point que ces moustiquaires agissaient comme un moyen de lutte antivectorielle. Ces recherches ont toutes été effectuées par des chercheurs de l’Office de la recherche scientifique et technique outre-mer, devenu Institut de recherche pour le développement, dans le cadre du centre Muraz de l’Organisation de coordination et de coopération pour la lutte contre les grandes endémies, en bénéficiant de l’appui scientifique et financier de la Division de la biologie et de la lutte contre les vecteurs de l’Organisation mondiale de la santé.

After 25 years of gradual increase, volcanic unrest at La Soufriere of Guadeloupe reached its highest seismic energy level on 27 April 2018, with the largest felt volcano-tectonic (VT) earthquake (M-L 4.1 or M-W 3.7) recorded since the 1976-1977 phreatic eruptive crisis. This event marked the onset of a seismic swarm (180 events, 2 felt) occurring after three previous swarms on 3-6 January (70 events), 1 st February (30 events, 1 felt) and 16-17 April (140 events, 1 felt). Many events were hybrid VI's with long-period codas, located 2-4 km below the volcano summit and clustered within 2 km along a regional NW-SE fault cross-cutting La Soufriere. Elastic energy release increased with each swarm whereas inter-event time shortened. At the same time, summit fractures continued to open and thermal anomalies to extend. Summit fumarolic activity increased significantly until 20 April, with a maximum temperature of 111.4 degrees C and gas exit velocity of 80 m/s, before declining to similar to 95 degrees C and similar to 33 m/s on 25 April. Gas compositions revealed increasing C/S and CO2/CH4 ratios and indicate hydrothermal P-T conditions that reached the critical point of pure water. Repeated MultiGAS analysis of fumarolic plumes showed increased CO2/H2S ratios and SO2 contents associated with the reactivation of degassing fractures (T = 93 degrees C, H2S/SO2 approximate to 1). While no direct evidence of upward magma migration was detected, we attribute the above phenomena to an increased supply of deep magmatic fluids that heated and pressurized the La Soufriere hydrothermal system, triggering seismogenic hydro-fracturing, and probable changes in deep hydraulic properties (permeability) and drainage pathways, which ultimately allowed the fumarolic fluxes to lower. Although this magmatic fluid injection was modulated by the hydrothermal system, the unprecedented seismic energy release and the critical point conditions of hydrothermal fluids suggest that the 2018 sequence of events can be regarded as a failed phreatic eruption. Should a similar sequence repeat, we warn that phreatic explosive activity could result from disruption of the shallow hydrothermal system that is currently responsible for 3-9 mm/y of nearly radial horizontal displacements within 1 km from the dome. Another potential hazard is partial collapse of the dome's SW flank, already affected by basal spreading above a detachment surface inherited from past collapses. Finally, the increased magmatic fluid supply evidenced by geochemical indicators in 2018 is compatible with magma replenishment of the 6-7 km deep crustal reservoir feeding La Soufriere and, therefore, with a potential evolution of the volcano's activity towards magmatic conditions.

Designing sol-gel synthesis of stable oxide nanostructures are one among the few key steps in simplifying the synthesis strategies for mass scale production of such nanomaterials. Herein, low temperature, CTAB based sol-gel technique has been adopted for the development of iron and cobalt tungstate nanostructures as they are one among the potential oxide nanostructures. As synthesized nanoparticles possess fine particle size in polycrystalline nature with good thermal stability and magnetic properties. The procedure adopted produced near-pure materials with no noticeable binary oxide formation and other impurities. These results may lead to promising applications

PPy-FeWO4 composites were synthesized by polymerization of pyrrole in presence of varying weight percentage of FeWO4 nanoparticles using FeCl3 as both catalyst and dopant. The synthesized polymer composites were characterized by FTIR, XRD, FESEM-EDAX, TEM and TGA analysis. The XRD and FTIR analysis confirmed showing near pure samples were achieved. The FESEM-EDAX and TEM studies showed the presence of FeWO4 nanoparticles which were well dispersed in the polymer matrix. The thermal degradation studies showed similar stability of polymer matrix in the presence of nanoparticles. Interfacial solar water evaporation devices were fabricated by depositing the polymer composite on floatable PU sponges. The studies revealed that 50-50 wt% PPy-FeWO4 showed maximum solar to vapour conversion up to 86% with an evaporation rate up to 1.2 kg m−2 hr−1.
