Abstract

The purpose of this work is to improve the predictive ability of the geological model of coastal formations when assessing hydrocarbon reserves and laying production wells. The article presents the results of work on the analysis of the geometry of sand bodies of the formations of the TP group of the Tanopchinskaya suite. To determine the morphology of sandstones, the authors used an integrated approach that takes into account: the results of sedimentological core studies, well logging data, 3D seismic materials. The authors studied in detail the conditions for the formation of these rocks on the basis of actual material (core), and presented photographs with characteristic structural and textural features of the rocks, which make it possible to establish the conditions for their formation. Spectral decomposition sections were analyzed in detail in the studied interval at several license areas of NOVATEK.The reasons for changes in the geometry of sand bodies from the perspective of sequence stratigraphy have been established and explained. In addition, an idea has been formed about the vertical evolution of the coastal zone under conditions of sea transgression. The filtration and reservoir properties of rocks were also analyzed based on measurements of porosity and permeability on more than 700 core samples formed in different sedimentation environments. Recommendations are given for the practical use of established patterns of changes in reservoir properties and the geometry of sand bodies when constructing a geological model of hydrocarbon deposits, which will certainly improve the efficiency of production drilling.

Abstract

Based on the complex geological and geophysical information and results of laboratory core studies, the detailed structure of sand sediments of the underwater part of the delta system has been studied. This has allowed to explain the distribution of hydrocarbons in the deposits due to morphological features of sedimentological objects.The work used the results of spectral decomposition of seismic data, interpretation of which allows to reconstruct with high detail morphology and structure of studied sediments. Sedimentological analysis of core was performed. The conclusions were compared with actual data of geological and geophysical well studies. The work identified two classes of sandstones with different filtration characteristics, which, taking into account seismic facies analysis, formed the basis for a conceptual geological model of the progradation of the sea delta coast with a predominance of wave processes.The presented conceptual model explains the distribution of sand material, its filtration characteristics, takes into account the physical processes of formation of the sediment profiles and allows to more accurately predict the distribution of the productive part of the formation, which helps to increase the success of drilling horizontal wells.The presented conceptual model explains the distribution of sandy material, its filtration characteristics, takes into account the physical processes of formation of the sediment profile of the sea coast and makes it possible to more accurately predict the distribution of the productive part of the formation, which helps to increase the success of drilling horizontal wells.

3. The Influence of Sandstones with a High Concentration of Heavy Minerals on the Distribution of Hydrocarbons in a Reservoir Using the Example of a Field in Western Siberia

Abstract

The article presents the results of special core studies in sandstone intervals with abnormally high values of natural radioactivity. Combining the obtained data with sedimentological analysis and actual field information made it possible to explain the position of the fluid contact and, accordingly, the distribution of hydrocarbons in the reservoir.In two wells, 16 samples were selected for lithological and mineralogical study in places characterized by anomalous values of natural radioactivity according to gamma ray logging data. Laboratory studies of the core were carried out: profile spectral gamma ray, X-ray phase analysis of the mineral composition of the rocks, petrographic description of thin sections, study by scanning electron microscopy.The studied rocks are represented by sandstones. Their texture is due to numerous layers of enrichment in heavy minerals, the minimums and maximums of which correspond to the minimums and maximums of profile radioactivity on the core. Among the accessory minerals, the main ones associated with an increased content of radioactive elements are zircon, thorite, fluorapatite and orthite. Their content in the rock can reach 15–40%.As a result of the work performed, a conceptual model was built that describes the mechanisms of formation of layers with high concentrations of heavy accessory minerals. From the point of view of sedimentation, such layers are a marker of transgression and have low filtration and capacitance properties according to laboratory data. This explains the different positions of fluid contacts in closely spaced wells and clarifies the internal structure of the productive formation.

Abstract

Aim. The article is devoted to the analysis of personalized training through the lens of the mentoring system at a medical university.Material and methods. The study involved 211 full-time 1st-6th year students of the Volgograd State Medical University. A sociological survey was used in the form of a questionnaire.Results. The article reveals the role of mentoring within federal innovation platform System of Personalized Training in a Higher Education Establishment. The authors substantiate the need to introduce a mentoring model to optimize the educational process and unlock the potential of students, residents, and teachers. The article examines the conceptual foundations, goals and objectives of mentoring, as well as implementation arrangements within the federal innovation platform. Additionally, the mentoring model benefits presented, including improved academic performance, increased student motivation, professional skill development, and career counseling. It also emphasizes the importance of monitoring and evaluating the effectiveness of the mentoring model to ensure its continuous modernization.Conclusion. The article emphasizes that the introduction of a mentoring model in the federal innovation platform System of Personalized Training in a Higher Education Establishment is an important step to achieve the goals of personalized training. The authors recommend future research on the implementation and evaluation of mentoring models in different higher education contexts.

5. Folklore Motifs in Contemporary Short Fiction of Mozambique

Abstract

The article explores the role of folklore tradition in the short prose of modern Mozambique. The symbiosis of the European cultural matrix, inseparable from the language in which the work is written, and the oral heritage of the Mozambican people is analyzed on the material of one of Aldino Muianga’s short stories. The story «Rosa Maria» is included in the author’s debut collection «Xitala Mati», the main material for which became the folklore «scary stories» about witchcraft and the living dead. And while in his later works Muianga focuses on acute social plots of the colonial past and post-colonial present, in the world of «Xitala Mati» there is practically no place for white people: it is not colonizers and the colonized that interact there, but it is people and spirits. The text of «Rosa Maria» is filled with numerous «conventional signs» by means of which, in the paradigm of the traditional culture of the peoples of Mozambique, the world of the dead communicates with the world of the living. These signs are intuitive to the African reader, while the European must identify and decipher them. At the same time, «Rosa Maria» shows motifs and imagery characteristic of the Gothic novel, albeit reinterpreted and modified to suit African realities. Such components of the poetics as the encounter with the infernal in the image of a beautiful woman, the gloomy colouring supported by the landscape and ominous animals, and the interest in the emotional reactions of the characters, typical of Gothic literature, allow us to see in «Rosa Maria» a hybrid genre, the roots of which should be sought both in the European literary tradition and in the folk tales of Mozambique.

Abstract

In recent years there is a positive trend in the development of preventive medicine, in particular, primary prevention of diseases. However, in most cases, patients seek help from a doctor after the manifestation of the disease, and therefore, early identification of risk factors (RF) remains relevant. Conduction of a large number of studies that are aimed at studying modifiable RF associated with the development of cardiovascular diseases (CVD), allowed the experts of the American Heart Association to develop recommendations “Life's Simple 7”, which makes it possible to structure methods of primary prevention of CVD and minimize the risk of their development. In 2019, experts from the American College of Cardiology presented a simplified version of these recommendations, to improve approaches to primary prevention and their effectiveness not only for doctors but also for patients. Thus, by involving the patient in the decision-making process about follow-up treatment, doctors can achieve a high level of compliance, which is essential for improving the prognosis. The “ABCDE” recommendations, in name of which are reflected the first letters of the leading CVD RF, include such paragraphs as RF assessment, the use of antiaggregating therapy, correction of blood pressure, cholesterol levels, smoking elimination, correction of high glucose levels and diabetes treatment, weight loss, assessment of social and economic factors affecting the morbidity in a particular patient. Despite the undoubted benefit of the “ABCDE” recommendations, some problems of primary prevention currently cannot be solved: the inability to accurately assess social and economic RF; the imperfection of the used CVD risk scales. The updated version of the recommendations allows not only to assess the existing RF of the patient, but also to effectively correct them. In addition, the patient himself can read the recommendations, which improves understanding of the primary prevention importance.

Abstract

Ticagrelor is known to prefer clopidigrel except in cases of high risk of bleeding, but according to the literature, clopidogrel still remains the most frequent second component of dual antiplatelet therapy (DAPT) in the world. The aim of the study was to conduct comparative description of patient groups with the acid acetilsalicilic (ACS), taken depending on a prescription of clopidogrel or ticagrelor as the second component of DAPT according to data of the year’s work of the Pirogov City Clinical Hospital №1. Materials and methods: clinical data of 854 patients with ACS who undergone treatment in of Pirogov City Clinical Hospital №1 in 2017 were analyzed. Clopidogrel was prescribed to 623 patients (73%) – the I group, ticagrelor 231 (27%) – the II group. Patients in the I group compared to the II group were significantly older (70 and 62 years accordingly), women accounted for 43% in I group and 27% in II group. Arterial hypertension (96 and 89%), diabetes (34 and 26%), post-acute myocardial infarction (38 and 19%), chronic kidney disease (26 and 12%), anemia (15 and 7%). Among patients of II group final diagnosis of ST-elevation myocardial infarction (STEMI) was more often (64 and 31%), coronary angiography (CAG) and percutaneous coronary intervention (PCI) were more frequent – 98/94% and 88/75%, accordingly. Clopidogrel is prescribed to the patients more often in comparison with ticagrelor. Doctors make a choice in favor of clopidogrel for elderly patients, more often women and more comorbid patients. The presence of STEMI, as well as the performance of CAG / PCI in any definitive diagnosis, is associated with a relatively more frequent prescription of ticagrelor, and in elderly patients, the implementation of PCI is the only factor that significantly influences the choice of more active antiaggregant in DAPT.

Abstract

Aim. Revealing of the associations of various cardiovascular psychosocial risk factors prevalence — vital exhaustion (VE) and hostility — with arterial hypertension (AH) in open male population, age 25-64 y. o., of moderately urbanized Siberian city.Material and methods. One stage epidemiological study was conducted on representative selection, shaped from election lists of males 25-64 y. o., from one of Tymen city districts. For the analysis of AH the data from cardiological screening was used, and surveying by psychosocial methods in algorithms of MONICA-MOPSY.Results. The prevalence of psychocosical risk factors was found as following: VI — 54,5%, hostility — 46,4%. With the age, higher levels of VE and hostility become more prevalent, achieving maximum at 55-64 y. o. Populational variation of systolic and diastolic blood pressure in males 25-64 y. o. is shifted to the right, with high prevalence of AH. In AH groups there are low and moderate levels of VE and high hostility.Conclusion. The data points on high demand on AH prevention in economically active citizens that have psychosocial risk factors; this requires interventions from the healthcare institutions of the region.

Abstract

Osteoarthritis, both idiopathic and post-traumatic, is currently the most significant problem in orthopedics. It is particularly difficult to treat patient with comorbidities, when it is necessary to decide on surgery for a late-stage joint disease. An operation is associated with a great risk in such patients. Even if the patient does not have serious somatic disorders, the main task of doctors (and this is a multidisciplinary problem) is to prolong joint functioning and maintain the patient’s quality of life.In this article, we report difficult cases, when a complex of non-pharmacological (therapeutic exercise, physiotherapy, orthobiology – PRP therapy) and pharmacological (nonsteroidal anti-inflammatory drugs, chondroprotectors, vitamins, calcium, tissue repair stimulants) treatments ensured a good effect, thereby maintaining the joint function and adequate quality of life without surgery. We also found that the use of Ambene® Bio (a chondroprotector) increased treatment efficacy.

Abstract

Chronic venous insufficiency, being the outcome and complication of most diseases of the veins of the lower extremities, is the fundamental cause of the formation of trophic ulcers. Over the past decade, in solving the problem of treating venous trophic ulcers, certain successes have been achieved, both in theoretical and practical terms. The modern concept of curation of patients with trophic ulcers of venous origin involves a reasonable combination of the following methods of treatment: local, including compression, as well as medical, surgical, minimally invasive and additional methods (physiotherapy, photodynamic therapy, etc.). The main goal of the treatment is to eliminate the ulcer, after which measures are taken to correct the underlying cause (chronic venous insufficiency) and prevent relapse. However, the tactics of the most successful treatment of patients with trophic ulcers of venous etiology is still not defined and requires further improvement. In particular, the method of photodynamic therapy, which includes the usage of special drugs - photosensitizers, also deserves to be studied and expanded for clinical use. The mechanism of photodynamic action is based on the ability of photosensitizers to accumulate in microbial and pathological cells. Under local light exposure, the photosensitizer enters an excited state and charges oxygen, which is associated with erythrocyte hemoglobin. Thus, in cells under the influence, a photochemical reaction is triggered, which ensures the formation of singlet oxygen and its free radicals. Reactive oxygen species, in turn, have a damaging effect on pathogenic flora and necrotic tissues. The main advantages of the method are relative simplicity, no risk of developing antibiotic resistance, acceleration of reparative and regenerative processes, and prevention of secondary infection. Thus, photodynamic therapy allows non-invasive, safe and effective optimization of local treatment and preoperative preparation of patients with venous trophic ulcers.

11. Selection of equipment for oil tank repair in northern conditions

Abstract

Introduction. The oil industry is a key component of the economy of the Russian Federation. A large number of tanks are used to store oil, each of which needs timely repair. According to the results of some research, almost half of the accidents are caused by excess uneven settlement of the tank, which causes premature failure. To restore the serviceable and operable condition of the tanks, it is necessary to carry out repair work, the quality and efficiency of which depends on the repair equipment used. An urgent task is to justify the choice of equipment for the repair of tanks, in particular, steel tanks of a vertical type.Methods and materials. A review of the different types of tanks was carried out and showed that vertical steel tanks are the most widely used. One of the ways to eliminate the uneven settlement of the tank is to raise it with the help of jacks and form a new base with the elimination of the slope.Results. The repair process for a 10,000 m3 steel vertical tank was considered. The values of the mass of the tank were obtained when filling with liquid petroleum products at 1% of the maximum occupancy. The value of the load on one lifting device is obtained. The design of the device structure and the strength calculation in the Compass 3D program were completed.Сonclusions. The most efficient type of the lift from the existing ones was determined, and its design was modernized. The strength calculation showed that the hydraulic lift is efficient under the given conditions.

18. Dynamics of cytokine production during immunization with polio vaccine with DTP in vivo

Abstract

Immunization with whole-cell adsorbed diphtheria-tetanus-pertussis vaccine (DTP) can cause various undesirable effects. The most common complications are febrile seizures, neuromyalgic syndrome and, in more severe cases, various encephalopathies. The listed complications are quite dangerous, especially in childhood, when primary DTP immunization is carried out. Many studies indicate that the development of these pathological processes is often associated with the action of various cytokines produced in response to vaccination. There are specific pro- and anti-inflammatory cytokines, the high levels of which are associated with the development of neurological complications after DTP vaccination. For example, IL-6 and IL-1 are often associated with the development of febrile seizures and encephalopathies. On the other hand, there are clinical data indicating a decrease in the incidence of complications after concomitant administration of vaccines. Thus, it is of particular interest to study the cytokine profile after the combined administration of DTP with another vaccine, which in some cases leads to a decrease in the number of complications and better tolerance of the vaccine. The vaccine against polio infection is currently one of the safest, but its effect on the level of cytokines is extremely poorly understood. Moreover, due to the fact that these drugs have been used for a long time and the interest in them is not as high as before, the number of new experimental works specifically on the cytokine profiles of many vaccines is limited. Basically, all existing work is aimed at studying various pathological processes associated with the introduction of a vaccine preparation. This leads to the fact that the mechanism of the formation of the immune response remains not fully understood. The aim of this work was to study the effect of combined vaccine administration on the cytokine profile. Results were obtained for the following cytokines: IL-2, RANTES, Eotaxin, MIP-1β, IL-12p40, IL-4, IL-6, IL-1α, and G-CSF determined in murine serum samples after combined administration of DTP and polio vaccine to the experimental animals. The cytokine profile was determined using Bio-Rad MAGPIX fluorescence reader. The study revealed and described the patterns of changes in the cytokine profile, both with the administration of the poliomyelitis vaccine alone, or in combination with the DTP vaccine. The results obtained in this work may be further used for more detailed studies on the mechanism of the immune response formation upon combined administration of vaccines and further improvement of existing drugs.

20. Effect of Xymedon and Mexidol in Combination with Antineoplastic Drugs on Spermatogenesis Indicators and Functional State of Spermatozoa in Rats with Walker-256 Carcinoma

Abstract

We compared the effect of Xymedon (100 mg/kg), Mexidol (50 mg/kg), and their combination on spermatogenesis indicators and functional state of spermatozoa in rats with Walker-256 carcinoma treated with doxorubicin (4 mg/kg) and cyclophosphamide (45 mg/kg) (once intraperitoneally on day 11 after tumor cells transplantation). Xymedon and Mexidol were injected intramuscularly for 10 days starting from day 11 of the experiment. The studied parameters were evaluated on experimental days 14 and 21. We have established that gonadoprotective effect of Xymedon developed gradually and persisted longer than that of Mexidol. It manifested in an increase in the number of epithelial spermatogenesis cells (spermatogonia by 3.2 times, early spermatids by 2.2 times, late spermatids by 2.9 times, and Leydig cells by 4 times) in the testes and also the proportion of viable progressively and non-progressively motile epididymal spermatozoa (by 2 times). The combination of Xymedon and Mexidol stimulated spermatogenesis (with restoration of the initial level of spermatocytes, an increase in the number of early spermatids by 65.5 and 99% in comparison with Xymedon alone and Mexidol alone, respectively) and increased the number of viable epididymal spermatozoa more effectively than Xymedon and Mexidol alone by 54 and 60%, respectively.

Abstract

Objectives. The objectives of this work are the development of a multi-primer system based on the polymerase chain reaction (PCR) aimed at the simultaneous detection of six bacterial pathogens that cause human pneumonia and the determination of the parameters important for the optimization of this multi-primer system, including solid-phase PCR systems (biological microarrays).Methods. To determine the optimal parameters of the system, PCR methods were used in monoplex and multiplex formats.Results. Primers for Staphylococcus aureus, Pseudomonas aeruginosa, Haemophilus influenza, Legionella pneumophila, Klebsiella pneumoniae, and Streptococcus pneumoniae detection were designed, and the PCR cycling conditions were optimized. The patterns of primer design for solidphase PCR were revealed.Conclusions. The developed prototype of a system specifically identifies six clinically significant bacterial pathogens. It could be expanded for the analysis of viral and fungal pathogens and used in clinical diagnostics. A prototype of a system for pathogenic agent detection in the immobilized phase (biological microarray) was created.

Abstract

The thermoelectric properties of thin Cr0.33Si0.67 films in amorphous and crystalline states, as well as at different annealing stages in the temperature range 100–900 K have been studied. The crystallization of amorphous films at a temperature of ~550 K is accompanied by an increase in the resistivity and thermoelectric power. Amorphous films crystallize with the formation of CrSi2 nanocrystals with an average grain size of 10–20 nm. Abstract

Purpose of the study. Determination of the level and structure of population immunity to SARS-CoV-2 among the population of the Murmansk region against the background of the incidence of COVID-19.Materials and methods. The study was carried out according to a unified method for determining the seroprevalence of the population, developed by Rospotrebnadzor with the participation of the St. Petersburg Pasteur Institute. A total of 3117 volunteers were examined, distributed into 7 age groups. The content of antibodies to SARS-CoV-2 was determined by ELISA using a set of reagents for analyzing human serum or plasma for the presence of specific immunoglobulins of class G to the nucleocapsid of the SARS-CoV-2 virus produced by the FBUN GNCPMiB Rospotrebnadzor (Obolensk) in accordance with the instructions for application.Results. The level of population immunity in the Murmansk region as a whole was 31,2%. The highest seroprevalence was found among children aged 1–6 and 7–13 years (35.6% and 44,1%, respectively), the lowest among the elderly population aged 60–69, as well as 70 and more years (20,4% and 20,9%, respectively). In the cities of the Murmansk region. the herd level of immunity varied from 19,6% (Kola city) to 46,1% (Kandalaksha city). It was not possible to find a reliable relationship between the incidence of the population of the cities of the Murmansk region and seroprevalence. The highest level of seroprevalence was noted among office workers (38,0%), the lowest among transport workers (19,5%). In the presence of contacts with patients with COVID-19, seropositivity increased 1,4 times compared to the average population value. The level of specific humoral immunity in convalescents after COVID-19 is 64,1%, which is 2,2 times higher than the average for half-day. The proportion of asymptomatic forms among seroprevalence volunteers was 89%.

Abstract

Introduction.The use of an electromechanical transmission in the design of a tracked vehicle allows an increase in the complex indicator of mobility, an increase in the range, fuel efficiency, maximum speed, a decrease in acceleration time, etc. The improvement of these indicators is achieved mainly due to the different performance characteristics of the internal combustion engine and the energy characteristics of electrical machines. The latter fact makes it possible to ensure the operation of the power plant of the tracked vehicle in such a way as to avoid unfavorable operating modes of both the internal combustion engine and the elements of the electromechanical transmission (a generator, a traction electric motor, an energy storage) from the point of view of energy efficiency, and to realize the high efficiency of the entire system.Research methods. To improve the mobility and implement a rational strategy for electromechanical transmission control, it is necessary to have an idea of the effective modes of operation of the main elements of the power plant. As a way to solve this problem it is proposed to study the energy characteristics of the main elements of an electromechanical transmission using the developed mathematical model for various modes of movement of a tracked vehicle.Results.Modeling the motion of a tracked vehicle with an electromechanical transmission makes it possible, in addition to determining the transmission parameters, to formulate preliminary requirements for its characteristics.Discussion and conclusion. To solve these problems, it is necessary to simulate the process of movement of a tracked vehicle, taking into account the initial data that are adequate to real operating conditions.

Abstract

The seasonal variability of the physicochemical parameters of milk of both ordinary goats and goats producers of a biosimilar human lactoferrin has been established. The average values of the mass fraction of fat, protein, lactose, nonfat milk solids, density index in the second and third quarters were lower than in the first and fourth quarters. At the same time, the difference between the spring-summer and autumn-winter periods in the mass fraction of fat was 25%. In the mass fraction of protein, lactose, nonfat milk solids, density it was 5–7%. Comparative analysis of milk from goats-producers of a biosimilar human lactoferrin of various lactations revealed a number of changes in the physicochemical composition of raw materials in comparison with non-transgenic animals: an increase in the mass fraction of protein by 4–6% (P˂0.05), of lactose by 2–6 (P˂0.05), nonfat milk solids by 1–6, density by 2–5% (P˂0.05), freezing temperature by 5 and a decrease in the mass fraction of fat by 5–8% (P˂0, 05). At the same time, the indicators of active and titratable acidity of milk were similar in all groups of animals. At the same time, the concentration of human lactoferrin in the milk of producers of the second and third lactation had almost identical seasonal changes in the increase and decrease in the synthesis of lactoferrin in the mammary gland during the year: in the first quarter – 2.88 and 2.97 g/l, in the second – 4.76 and 4.63, in the third – 7.44 and 7.55 and in the fourth – 7.97 and 6.72 with an average annual value of 5.84 and 5.72 g/l, respectively.

Abstract

As it is known, allergic rhinitis is one of the most pressing issues of modern rhinology. At present, special attention is directed to the study of the problem of year-round allergic rhinitis, since in recent years its incidence in economically developed countries has been steadily growing. Often the only complaint of patients is the constant stuffy nose with mucous discharge, so both patients and doctors often underestimate the importance of timely diagnosis and treatment of the disease. According to research, year-round allergic rhinitis can contribute to the development of pathology of the nasopharynx, middle ear and auditory tube, nasal cavity and paranasal sinuses, which often take on a chronic or recurrent form. The treatment proposed in the clinical recommendations, carried out in three main areas, does not always lead to persistent remission of the atopic process. This article covers the course of ENT diseases against the background of respiratory chlamydia and mycoplasmic intracellular infection. The problem of year-round allergic rhinitis associated with respiratory intracellular infection has not been elucidated to date, but the chronization of the process, long uncontrolled course, and the tendency to complications of this pathology of the upper respiratory tract may be related to it.

Abstract

We present a study of the sensitivity of a hybridization microarray analysis of DNA labeled with 2'‑deoxyuridine-5'-triphosphate derivatives that contain the Cy5 and Cy7 cyanine fluorophore dyes. Two Cy5-dUTP and two Cy7-dUTP derivatives that can be effectively incorporated into a newly synthesized DNA strand during PCR were chosen for this study. PCR products fluorescently labeled with different dyes were hybridized with a matrix of oligonucleotide probes immobilized in the microchip hydrogel cells. Fluorescent signals were recorded on a digital fluorescence microscope in the red (for Cy5-dUTP) and near-infrared (for Cy7-dUTP) range. Using the serial dilution method, the minimum DNA concentration was evaluated, which is necessary for genotyping by the specific interaction between the fluorescently labeled PCR product and oligonucleotide probes on the microarray. It has been shown that the fluorescently labeled Cy7-dUTP, unlike Cy5-dUTP, provides greater sensitivity of the hybridization analysis.

Replicative strand slippage is a biological phenomenon, ubiquitous among different organisms. However, slippage events are also relevant to non-natural replication models utilizing synthetic polymerase substrates. Strand slippage may notably affect the outcome of the primer extension reaction with repetitive templates in the presence of non-natural nucleoside triphosphates. In the current paper, we studied the ability of Taq, Vent (exo-), and Deep Vent (exo-) polymerases to produce truncated, full size, or expanded modified strands utilizing non-natural 2′-deoxyuridine nucleotide analogues and different variants of the homopolymer template. Our data suggest that the slippage of the primer strand is dependent on the duplex fluttering, incorporation efficiency for a particular polymerase-dNTP pair, rate of non-templated base addition, and presence of competing nucleotides.

30. Investigation of Tantalum Capacitors Using the Accelerated Quality Assessment Technique STRESS TEST

Abstract

The paper discusses the possibility of conducting accelerated tests to assess the quality of tantalum bulk-porous capacitors according to the developed STRESS TEST technique instead of outdated methods that require costly resources for testing and do not provide proper rejection of capacitors (detection of defects in the oxide layer). It is known that the grown amorphous pentoxide is able to form a uniform layer on the surface of tantalum, but due to various factors (due to surface roughness, porosity, uneven distribution of the electrolyte during the oxidation process and the electric contact of the carrier-anode), a surface is created with "problem areas", "incomplete formation", or, in other words, a "weak spot", the rejection of which is most important for modern tantalum capacitors. A comparative analysis of the application of the standard method and the STRESS TEST technique for accelerated evaluation of the quality of tantalum bulk-porous capacitors will prove the effectiveness of the developed technique. The developed STRESS TEST technique is based on cyclic (10 cycles) application of an increased voltage to the capacitor (corresponding to the voltage during the formation of an oxide layer on a tantalum anode) for the duration of each cycle for 5 minutes, which is proved by calculating this process based on the Zhurkov reliability acceleration model. The technique makes it possible to identify low-quality capacitors in advance with defects in the structure of the oxide layer. The technique makes it possible to evaluate the quality of capacitors in an accelerated time, which is confirmed by the obtained regression model of equivalent series resistance in comparison with the regression model of the standard type of tests for long-term reliability for 24,000 hours. The application of the STRESS TEST technique for tantalum volumetrically porous capacitors will reduce the testing time by approximately two and a half years.

Abstract

Today, increasing volumes of household solid waste (HSW) pose a serious problem throughout the world. The solution to this problem is involvement of secondary raw materials and waste in production. The disposal of HSW includes laborious stages of its collection and transportation. Aggregates of the garbage truck work inefficiently, only pressing HSW into a shapeless mass, not subject to sorting and processing. The suggested re-equipment of a special vehicle with aggregates with the combined functions of «loading–forwarding–grinding–pressing– briquetting» will significantly reduce the energy consumption of the transportation process by providing simultaneous processing and briquetting of garbage at the time of its transportation. A scientific and technical problem arises in development of a technique for technical re-equipment of high-performance special equipment with given nominal energy and power characteristics of the machines. It was proposed to solve this problem by combining recuperative systems with reuse of the energy of gravity of the own mass of garbage. Thus, the objective of the work is to develop a rotor-inertial device with reduced energy intensity. Methods of analytical and statistical research of the model range of special equipment with an analysis of its technical characteristics have been applied. To solve the problem of developing a kinematic diagram of a briquetting device, a calculation was performed based on the method of modeling the structure of composite aggregates. Modeling was performed in SolidWorks program in Simulation application package. The developed kinematic diagrams of units and aggregates for briquetting and pressing garbage operate at rated power characteristics of hydraulic equipment achieved due to distribution of drive power among the most energy- loaded operations. The article presents the rationale for effectiveness of the developed rotary- inertia device for briquetting HSW. The originality of the design lies in the structural arrangement of the briquetting unit and the grinding mechanism. The use of the principles of inertial moments and gravity of own mass of garbage allowed us to significantly (by 25 %) reduce the energy consumption of the pressing process and formation of the finished briquette. The use of mechanical energy of the conveyor belt tensioners and of the mass of the roll increased with briquetting under its own weight, allowed to reduce the work spent to form compacted HSW from 48000 kJ to 11970 kJ, to reduce the volume of the pressed roll, to increase the utilization rate of load carrying capacity, to reduce the energy intensity of the process.

Abstract

The effects of long-period climate change and fluctuations of climate factors in the recent decades on the Scots pine (Pinus sylvestris) growing in the Kerzshenskii State Nature Biosphere Reserve (the Nizhny Novgorod region) are considered. It is found that the amount of precipitation forming phytocenoses water availability is the most important climatic factor limiting the pine growth and development. A competition between the linear and radial pine increments for the water resource, which directly depends on the amount of precipitation, is identified. The direct response of the radial to linear increment is found, because these are the height tree growth and the crown development that induce the successful stem diameter formation. It is noted that due to the widespread occurrence and variety of environmental impact signals, Pinus sylvestris is an optimum object for the environmental monitoring of forest ecosystems, which makes its further investigation in a changing climate essential.

Abstract

Introduction. The implementation of production and technological processes of machines on the vast territory of the Russian Federation is related to the necessity to operate them in low temperatures conditions. The article presents the results of the applied scientific research of the factors that have a negative impact on the operation process of internal combustion engines at low ambient temperatures. The use of the modern methods and means of diagnostics of the cylinder-piston group and the crank- engine mechanism make it possible to reliably establish the causes of the internal combustion engines failures.Materials and methods. In the course of the experimental studies the non-destructive testing, organoleptic, measuring and expert methods were used.Results. As a result of a comprehensive assessment of the engine failures causes, it was found that the breakdowns in the use of inappropriate operating materials at low temperatures, measures to prepare for the launch of diesel generator sets were the main reason for untimely start-up and the faults in cylinder-piston group and the crank mechanism elements, which led to emergency failure of the internal combustion engine.Discussion and conclusion. The proposed research results will make it possible to specificate the development prospects in the field of providing the reliability of internal combustion engines operation.

Abstract

The efficiency of DNA labeling was assessed for 2'-deoxyuridine 5'-triphosphate (dUTP) derivatives containing the Cy7 cyanine dye as a fluorophore. Two fluorescent Cy7-labeled dUTP analogs differed in the chemical structure of the linker between the fluorophore and nucleotide moieties. The efficiency of the polymerase chain reaction (PCR) and inhibition with modified nucleotides were estimated by real-time PCR. The efficiency of labeled nucleotide incorporation in PCR products was measured by quantitative electrophoresis. The efficiency of target DNA labeling was evaluated by binding the fluorescently labeled PCR products to a microarray of oligonucleotide probes immobilized in hydrogel drops (a biochip). The near-infrared hybridization signal was detected by digital luminescence microscopy. An increase in linker length was found to provide more efficient incorporation of the labeled nucleotide. Both of the compounds provided high sensitivity and high specificity of DNA testing via allele-specific hybridization on a biochip.

Abstract

Particular features of the simultaneous incorporation into the growing DNA chain of modified Cy5-deoxyuridines (Cy5-dU) and deoxycytidines (Cy5-dC) and Taq DNA polymerase in the process of PCR amplification were studied. The studies were carried out for the pairwise incorporation of nucleotides modified with fluorophores charged positively, negatively, or neutral. A portion of each of the modified dNTP (Cy5-dUTP and Cy5-dCTP) in respect to natural analogs (dTTP and dCTP) was varied from 0 to 100%, i.e., until the complete substitution. The results were compared with the individual incorporation of the corresponding modified derivatives. The (Cy5-dU + Cy5-dC) pairs were used to amplify of the bacterial genome fragments of 126, 283, and 370 bp in length. Under the amplification conditions with Cy5-modified nucleotides the yield of the product decreased with an increase in the length of the amplified DNA fragment. Electronegative Cy5-dNTPs showed the lowest inhibitory effect on the PCR procedure, whereas electroneutral nucleotides provided a greater density of label incorporation into the growing DNA chain.

Abstract

A method of thermal dissociation of DNA–protein conjugates was developed for the selection of aptamers to human alpha-fetoprotein. The method is based on the identification of the DNA oligonucleotides from the combinatorial DNA libraries with the highest specificity to targets, which remained within the conjugate after two stages: a) washing of unbound sequences; and b) removal of the sequences bound to the DNA but displaying a low affinity to it. The undestroyed most stable complexes were subjected to the thermal dissociation with the involvement of the corresponding oligonucleotides in the next rounds of selection. A comparison of the proposed method with the standard washing demonstrated a more intensive enrichment of the library. Due to the tighter conditions for washing of the target-bound DNA oligonucleotides the use of thermal dissociation can increase the efficiency of selection of highly specific aptamers to their molecular targets.

3-Phenoxybenzoic acid derivatives were synthesized. Their pharmacological activity was studied. Several compounds, in particular 2-cyanoprop-2-yl 3-phenoxybenzoate, were found to exhibit peroxisome proliferator-activated receptor γ agonist activity and to be capable of activating glucokinase and inhibiting protein glycation. The studied compounds did not influence dipeptidyl peptidase-4 activity.

Abstract

Chronic noninfectious diseases (cardiovascular, bronchopulmonary, oncological diseases and diabetes mellitus) are presently the most common cause of death worldwide, with cardiovascular diseases (CVD) being predominant. For this reason, the key goal of a physician is not only to treat but also to prevent diseases. Acetylsalicylic acid (ASA) is considered one of the most effective drugs for secondary prevention of CVD. However, the use of ASA for primary prevention is still debated. Results of many studies of ASA are inconsistent. Some studies have suggested that using ASA in patients aged 40-70 with a high 10-year risk of CVD and a low risk of bleeding may reduce the incidence of CVD. Administration of ASA to patients with a high or medium risk of CVD is also considered.

Abstract

Background. The epidemiology of hypertension (HTN) and antihypertensive treatment (AHT) in the Russian Federation was repeatedly estimated in epidemiology trials with various methodology, but situation in Moscow region has not been fully explored.Objective. To examine the rate, age and gender features of HTN and AHT in unorganized Moscow cohort.Design and methods. 532 participants older than 16 years old were interviewed in Moscow park after signing informed consent. Information about sex, age, rate of diagnosed HTN, AHT and its effectiveness was collected based on a questionnaire. Arterial blood pressure (BP) was measured consequently three times.Results. Median and interquartile range of participants age was 57 (38–66) years, 71,1 % were women. Women were significantly older than men (59 (46–67) and 48 (31–61) years respectively, p = 0,0001). 42,7 % reported previously diagnosed HTN, 62,5 % received AHT. 79,7 % participants receiving AHT (49,8 % HTN patients) considered it effective. Home BP self-monitoring was performed by 43,7 % respondents (sex- and age-adjusted rates). While measured, 29,7 % participants had optimal BP level, 22,6 % — normal and 15,2 % — high normal. BP > 140 and/or 90 mm Hg was measured in 32,6 % of participants (rates are sex and age adjusted). BP > 140 / 90 mm Hg at assessment was found in 48,8 % HTN patients and 16,4 % participants without previously diagnosed HTN (8,7 % in entire studied cohort). There were no significant differences in the rate increased BP level at assessment and in HTN patients who received AHT or not (49,5 and 46,9 %). HTN rate was 39,6 % in participants who considered their AHT effective, and 75,5 % — ineffective, p < 0,0001. 63,4 % participants taking AHT could specify the drugs (65,3 % received monotherapy, 27,1 % — 2 drugs and 7,6 % — 3 drugs; fixed combinations received 17 %). BP levels and HTN did not differ in HTN patients who received one or more drugs.Conclusions. In unorganized Moscow cohort age and gender adjusted rate of HTN at BP measurement was 32,6 %. In 8,7 % participants high BP was detected at first time. Only 62,5 % patients with previously diagnosed HTN received AHT. 39,6 % participants who considered AHT effective demonstrate high BP at assessment. AHT was not associated with the lower HTN rate or BP level, which may be attributed to excessive use of monotherapy and insufficient application of fixed combinations.

Abstract

The competitiveness of the country in the modern world mainly depends on the ability of the national education system to provide training for the professional elite, to involve the broad strata of society in the process of lifelong learning and to create conditions to meet the various cultural and socioeconomic needs of the population. The Chinese dominant national idea − the revival of greater China − in the most important documents is supplemented recently by new content: “the creation a powerful country with a strong education.” Education in China’s strategy is considered as a main factor of social stability and internal ethnic integration, and at the same time as an engine of innovative development. China provides the world with an example of how to increase the potential of human capital in a country where illiteracy and poverty reigned 30 years ago. The Chinese higher education system not only demonstrates its ability to compete with the world’s leading universities, but also promotes its technologies, standards and values in the global educational space, fi rst of all, strengthening its infl uence on the countries included in the “One Road ‒ One Road” project.

Abstract

Introduction.The problem of increasing the efficiency of power machines is important to develop a new design of the main elements or drive with the possibility of widespread use of hybrid drives. To preserve the power characteristics of the drive and its technical and operational indicators, the authors consider a hybrid with the available electric and hydraulic drive parts that form a complex synergistic mechanism as an alternative. The proposed combined tracking system with the specified parameters allows for the efficient operation of the drive of many machines.Materials and methods.The paper presented the results of an analytical study, the prospects for the development of mechatronic systems in engineering and technological complexes. The research also identified the industry area with a high development potential for mechatronic and hybrid drives. The authors formulated the problem of designing and developing hybrid drives due to the fact that there was no single method for substantiating the energy efficiency indicators of a power hybrid drive that ensured the interaction of two dissimilar systems (electric drive and hydraulic drive with fundamentally different parameters).Results. Therefore, the authors conducted the analysis of the structural, technological and operational parameters of the hybrid drive systems and proposed a combined servo drive system.Discussion and conclusions. The authors prove and justify the extensive possibilities of using hybrid drives as an alternative to pneumatic and hydraulic drives The results of the research allow specifying the directions and prospects of using hybrid synergetic drives and systems for the functioning of working equipment, machines and complexes.Financial transparency: the authors have no financial interest in the presented materials or methods. Abstract

In metal forming using high power current pulses, it becomes necessary to control both reproduction frequency and pulse amplitude. Description of a generator of high power current pulses with controlled thyristor converter is provided as a power source of charging device (charger) for regulating voltage (pulse amplitude) of capacitor charge. Faults of the generators associated with inrush current in capacitor charge modes are revealed, which reduces quality of supply network. To reduce time of transient processes while lowering voltage across capacitors, application of reverse thyristor converter is applied as a power source. Structural diagram of generator is considered, which includes reversible thyristor converter with separate control, power unit, capacitor recharge device, charger parameters automatic control system and capacitor charge process control system. Calculation of parameters of automatic control system regulators is presented. To obtain optimal transients, standard methodology for setting regulators to a modular optimum was used. In order to reduce overshoot at time of disturbances appearance, which can reach 100 % and higher, socalled logical device was introduced into the automatic control system. It blocks control pulses on thyristors of converter and simultaneously reduces signal at the output of current regulator to zero. Simulation model of high power current pulse generator in MatLab – Simulink environment was synthesized. Analysis of the model was carried out, and graphs are given that explain principle of device operation and transition processes under various operating modes. Generator application will allow user to adjust amplitude of current pulses with high speed and to obtain sufficiently high-quality transient processes of capacitors charge (discharge), which will have beneficial effect on supply network. Application of better converters will significantly increase frequency of reproduction of current pulses.

Abstract

A measurement of the inclusive jet production in proton-proton collisions at the LHC at root s = 13TeV is presented. The double-differential cross sections are measured as a function of the jet transverse momentum p(T) and the absolute jet rapidity vertical bar y vertical bar. The anti- k(T) clustering algorithm is used with distance parameter of 0.4 (0.7) in a phase space region with jet p(T) from 97 GeV up to 3.1TeV and vertical bar y vertical bar < 2.0. Data collected with the CMS detector are used, corresponding to an integrated luminosity of 36.3 fb(-1) (33.5 fb(-1)). The measurement is used in a comprehensive QCD analysis at next-to-next-to-leading order, which results in significant improvement in the accuracy of the parton distributions in the proton. Simultaneously, the value of the strong coupling constant at the Z boson mass is extracted as alpha(S)(m(Z)) = 0.1170 +/- 0.0019. For the first time, these data are used in a standard model effective field theory analysis at next-to-leading order, where parton distributions and the QCD parameters are extracted simultaneously with imposed constraints on the Wilson coefficient c(1) of 4-quark contact interactions.

Abstract

A search for flavor-changing neutral current interactions of the top quark (t) and the Higgs boson (H) is presented. The search is based on a data sample corresponding to an integrated luminosity of 137 fb(-1) recorded by the CMS experiment at the LHC in proton-proton collisions at root s = 13TeV. Events containing exactly one lepton (muon or electron) and at least three jets, among which at least two are identified as originating from the hadronization of a bottom quark, are analyzed. A set of deep neural networks is used for kinematic event reconstruction, while boosted decision trees distinguish the signal from the background events. No significant excess over the background predictions is observed, and upper limits on the signal production cross sections are extracted. These limits are interpreted in terms of top quark decay branching fractions (B) to the Higgs boson and an up (u) or a charm quark (c). Assuming one nonvanishing extra coupling at a time, the observed (expected) upper limits at 95% confidence level are B(t -> Hu) < 0.079 (0.11)% and B(t -> Hc) < 0.094 (0.086)%.

Abstract

The state of the hemostasis system was studied in 9 patients of the middle age group (44 ± 9.94 years) who received thermal trauma on an area of more than 32% (49.4 ± 18.3) of the body surface, accompanied by the development of burn shock. The standard therapy for burn injury was supplemented with HBO sessions. Treatment with hyperbaric oxygen was carried out in pressure chambers BLKS-307, BLKS-307/1. The state of the coagulation, anticoagulant and fibrinolytic links of the hemostasis system, as well as the viscoelastic properties of the blood, were assessed immediately before the HBO session and immediately after it. The total number of comparison pairs was 45. Abstract

A novel electroneutral rigid meso-sulfophenyloxy substituted heptamethine dye was synthesized in six steps. Selective derivatization of one sulfonamide group with octanedioic acid introduced the carboxy end group attached through the hexamethylene linker, which provided the dye solubility in water. Absorbance of the dye in the near infrared region makes it promising for covalent labeling of biomolecules.

Abstract

The substrate efficiency of modified derivatives of 2'-deoxyuridine-(mod-dUTP) and 2'-deoxycytidine-5'-triphosphates (mod-dCTP) was studied in the regime of simultaneous incorporation using the primer elongation reaction (PEX) and PCR with complete replacement of natural deoxytimidine (dT) and deoxycytidine (dC). Polymerases with missing 3'–5' exonuclease activity were used: Taq and Vent (exo-), as well as natural and constructed DNA templates. DNA fragments modified simultaneously by two types of nucleotides with similar and different functional groups were obtained. Research is important for obtaining aptamers with extended properties.
