Currently, submerged cultivation of the Bacillus anthracis STI-1 strain for live anthrax vaccine production requires liquid nutrient media, which have disadvantages of a short shelf life (no more than one month) and a narrow range of storage temperatures (2–8 °С). Dry media, in contrast, have a number of indisputable advantages: such media are transportable and easy to use, have a standard capability to retain properties, and can be stored without preservatives at 2–30 °С for 2–5 years. The aim of this work was to develop a technology for the preparation of a dry nutrient medium for anthrax vaccine production. Materials and methods: The study used the Bacillus anthracis STI-1 vaccine strain and a nutrient medium for its cultivation, containing a 70:30 mixture of an enzymatic digest of casein and a pre-processed corn extract solution. Drying of the nutrient medium was carried out on a spray-drying unit. The authors evaluated physicochemical parameters of experimental medium batches. The shelf life was determined by an accelerated stability study. The dry nutrient medium was used to produce a live anthrax vaccine. Quality attributes of the vaccine were assessed for compliance with regulatory requirements. Results: The authors developed the dry media production technology. According to it, the liquid nutrient medium is fed to the drying unit at a rate of 20–25 dm3/h. The spray air pressure is 0.02 MPa. Temperatures at the drying chamber inlet and outlet are 118–122 °С and 85–90 °С, respectively. The technology was used to obtain 3 experimental batches of the dry medium. The study results demonstrate that the technology is reproducible, and the tested quality attributes of experimental medium batches are consistent with the requirements. According to the accelerated stability study, the shelf life of the dry nutrient medium at 2–30 °С is at least 3 years. Experiments demonstrated the possibility of using the dry nutrient medium for live anthrax vaccine production. Critical quality attributes of the vaccine obtained with the medium met regulatory requirements. Conclusions: The developed technology allows for the production of a standard dry nutrient medium with a prolonged shelf life, which is convenient for live anthrax vaccine production.

We studied the age-related characteristics of the response of stem cells and liver in male Wistar rats to administration of carbon tetrachloride (CCl4) and ethanol. It was shown that modeling of liver cirrhosis caused inflammation, fibrosis, damage to sinusoidal capillaries, necrosis, and disturbances in the functional activity of hepatocytes in young rats. These processes were accompanied by mobilization of profibrotic mesenchymal stem cells (MSC), proinflammatory hematopoietic stem cells (HSC) and lymphocytes (CD45hiCD133+) from the bone marrow into the blood and migration to the liver. On the other hand, the number of hepatocyte precursors expressing Sox9 (cells of Hering's canal), immature cholangiocytes, Ito cells, oval cells, and endothelial cells of the liver sinusoids) sharply increased in the liver. In young rats, mobilization and migration of MSC, HSC, and hepatocyte precursors against the background of liver cirrhosis were more intensive than in old animals. The higher resistance of old rats to exposure is associated with age-related changes in the niches as well as in mobilization and migration of cells.

The aim of the study was to improve the procedure for the Yersinia pestis EV strain cell concentration using the system for tangential flow microfiltration with the ASF-020 filter support unit.Materials and methods. The study used the vaccine Y. pestis EV strain derived from NIIEG cell line. Submerged cultivation of the native culture was performed using BIOR-0.25 reactor with automated control system. Microbial suspension concentrate was produced through microfiltration applying (Adaptive filtration system) AFS-009 and AFS-020 installations. The content of live microbial cells was determined by cytorefractometry. Assessment of the resistance of Y. pestis EV strain cells to technological factors was performed by photometric registration of changes in the optical density of bacterial suspension during the lytic response of cells to sodium dodecyl sulfate. Physical-chemical and immunobiological properties of the dry live plague vaccine were determined in accordance with the pharmacopoeial item.3.3.1.0021.15 of the State Pharmacopoeia of the Russian Federation, 14th edition.Results and discussion. The design features of the equipment introduced made it possible to carry out membrane filtration of microbial suspension, using BIOR-0.25 reactor as an intermediate storage unit, thereby excluding three technological stages. The total concentration of microbes in the suspension obtained by routine and improved methods was more than 150 billion microbial cells per ml. A comparative study of the effect of various hydrodynamic regimes in the working cavities of AFS-009 and AFS-020 filter units did not significantly affect the morphometric properties and resistance of microbial cultures to extreme (technological) factors. Based on the experimental data, the mass balance of the membrane filtration process has been determined. The optimized technology gave 0.13 liter yield of concentrate from 1 liter of native culture, and the process duration was reduced to 5 hours, the yield of the finished product in one production cycle was increased by 3 times. Thus, the process of concentrating Y. pestis EV strain cells during the production of the tablet form of live plague vaccine has been enhanced. A comparative study of the morphometric properties and resistance of plague microbe cultures to technological factors in the process of their concentration using optimized technology did not reveal any significant differences as compared to the routine one. Technological stage of concentrating has been reduced to 5 h term with a three-fold increase in the yield of finished product.

We studied the formation of injuries in lung endothelium and the response of angiogenesis cells during modeling of pulmonary emphysema in male and female C57BL/6 mice with metabolic disorders. Hemodynamic disturbances and reduction in the area of the microvasculature caused by combined pathology in male mice were more pronounced than in females. Mobilization and migration of angiogenic precursors were impaired in both male and female mice. In males, activity of recruiting endothelial progenitor cells, vascular smooth muscle cells, luminal cells of nascent vessels and pericytes into the lungs was additionally reduced. In females, accumulation of endothelial progenitor cells (CD45-CD31+CD34+), vascular smooth muscle cells, and pericytes in the lungs was observed, which indicated activation of endothelial regeneration. Sex differences in the reaction of the lung endothelium and angiogenesis cells can be explained by genetic factors of lipid and glucose metabolism.

We studied the effects of spiperone, a selective blocker of dopamine D2 receptors, on the model of pulmonary emphysema provoked by administration of elastase and D-galactosamine hydrochloride to female C57BL/6 mice and characterized by activation of proteases in the lungs and systemic deficiency of its inhibitor α1-antitrypsin. In this model, spiperone prevented the development of inflammatory reaction and reduced the area of emphysematous expanded alveolar tissue. The expression of angiogenic marker CD31 in the lungs increased under these conditions. Regeneration of the damaged microvascular bed under the action of spiperone resulted from recruiting of Notch1+ endothelial progenitor cells (CD45-CD31+CD34+) into the lungs and blockade of the inhibitory effect of dopamine on phosphorylation of VEGF-2 receptors in endothelial cells of different maturity. In addition, spiperone produced a protective effect on hepatocytes and restored the production and secretion of α1-antitrypsin by these cells.

The phenomenon of improved energy confinement during radiative cooling at the plasma edge was studied experimentally in the T-10 tokamak. It was shown that the effect is independent on the kind of radiating gas. No substantial differences were observed using Ne, which radiates at two-thirds of the plasma radius, or He, which radiates at the very edge. This phenomenon is explained in frames of nonequilibrium thermodynamics. In a self-organized plasma, the energy balance is described by a Smoluchowski-type equation, where the plasma thermal conductivity and its functional dependence on the intensity of the heat flux, perturbing the pressure profile, is determined from experiment.

Various stem cells were studied in female BALB/c mice at the early terms after administration of N-methyl-N-nitrosourea to search early diagnostic markers and therapeutic targets. Bladder cancer relapses are associated with diffuse pattern of neoplastic changes in the mucous layer of the bladder; treatment efficacy depends on the degree of structural alterations in the bladder wall.Materials and methods: for evaluation of microcirculation characteristics and structural alterations of mucous layer of the bladder we have used laser analyzer of capillary blood circulation («ЛАКК-01» produced by НПП «ЛАЗМА», Russia) and compact mobile optic tomograph (ИПФ РАН, Russia).Results: conducted researches have shown alterations affecting both epithelial and submucous layers with their microcirculatory bed in perifocal to bladder tumor tissues.Conclusions: complimentary application of laser Doppler flowmeter and compact mobile optic tomograph allows defining the type and extent of pathologic process in the bladder wall in superficial tumors and choosing optimal method of correction.

Introduction. The effectiveness of methods for treating chronic prostatitis, which are aimed at improving the hemodynamics of the pelvic organs, has been proven by numerous studies conducted over more than thirty years. At the same time, pelvic myofascial syndrome can lead to impaired pelvis' hemodynamics. Purpose of research. This study aimed to determine the effect of myofascial syndromes on the blood circulation in the prostate in patients with chronic bacterial prostatitis, as well as an assessment of the dynamics of changes in blood flow in the prostate on the treatment of concomitant myofascial syndrome. Materials and methods. 59 men 34-52 years old with various clinical manifestations of myofascial syndrome (patients of the neurological clinic) were examined to assess the dynamics of blood circulation in the prostate. In addition, 127 men aged 28-52 years (mean age 38 years) with typical clinical manifestations of chronic prostatitis, who repeatedly and unsuccessfully received treatment for this disease, evaluated the effect of treatment of concomitant myofascial syndrome on prostate hemodynamics Results. The results suggest violations of microcirculation in the prostate in patients with chronic pelvic pain with myofascial syndrome. At the same time, hemodynamic impairment in the prostate is directly associated with impaired pelvic hemodynamics in general. The state of microcirculation in the prostate depends on the severity of pain symptoms in nonspecific chronic pelvic pain, and the treatment of nonspecific chronic pelvic pain leads to a reduction in the main symptom of the disease (pain) and to the normalization of microcirculation in the prostate. Treatment of concomitant myofascial syndrome in patients with chronic bacterial prostatitis allowed to achieve the disappearance of clinical manifestations of the disease and normalization of hemodynamics in the prostate, as well as a decrease in laboratory signs of inflammation. Conclusions. Concomitant myofascial syndrome in patients with chronic pelvic pain is a cause of pelvic hemodynamic disorders and because of the development of the inflammatory process. The severity of hemodynamic disturbances directly depends on the intensity of painful manifestations of the myofascial syndrome. Treatment of myofascial syndrome in patients with chronic bacterial prostatitis is accompanied by the disappearance of disease's clinical symptoms and the restoration of prostate's blood circulation, as well as the reduction or disappearance of inflammation's signs.

To date, the technology of live plague vaccine production uses a combined method of concentrating microbial cells which consists of three operations with a total duration of 18 hours. The procedure of obtaining concentrate, which is used in the current vaccine production technology, has a number of disadvantages, namely: multiple operations, high energy consumption, long duration, and, as a consequence, low yield of concentrated suspension (0.04 l from 1 l of native culture). The aim of the study was to optimise the procedure of Yersinia pestis ЕV microbial cell concentration using the system for tangential flow microfiltration with the ASF-020 filter support unit. Materials and methods: the vaccine strain used in the study was Yersinia pestis ЕV derived from NIIEG cell line (the strain of the Research Institute of Epidemiology and Hygiene). Submerged cultivation of the native culture was performed using the BIOR-0.25 reactor. The content of live microbial cells was determined by cytorefractometry. Oxidative metabolism was assessed using the chronoamperometric method. Physico-chemical and immunobiological properties of the dry live plague vaccine were assessed according to the monograph FS.3.3.1.0022.15 of the State Pharmacopoeia of the Russian Federation, 14 edition. Results: the equipment’s design features made it possible to carry out membrane filtration of the microbial suspension using the BIOR-0.25 reactor as an intermediate storage unit, thereby excluding three technological stages. The total concentration of microbes in the suspension obtained by the routine and the optimised methods was not less than 120 billion microbial cells/ml. A comparative study of the effect of various hydrodynamic regimes in the working cavities of ASF-009 and ASF020 filter units did not significantly affect the morphometric and physiological properties of microbial cultures. Experimental data helped to determine the process mass balance of membrane filtration. The optimised technology gave 0.17 l yield of the concentrate from 1 l of native culture, and the process duration was reduced to 4 hours. Conclusions: the process of concentrating Y. pestis EV microbial cells during production of plague vaccines was optimised. A comparative study of morphometric and physiological properties of plague microbe cultures that was carried out during their concentration using the optimised technology did not reveal any significant differences as compared to the routine one.

Ion heat conductivity in deuterium plasma of the circular limiter tokamak T-10 is analyzed in more than 100 Ohmic shots. Four data scans: plasma density, effective charge, plasma current, and toroidal magnetic field are presented. Each scan is formed with the preservation of other parameters. It is shown that at the plasma periphery, [Formula: see text], the ion heat conductivity exceeds neoclassical values up to three to eight times. The anomalous part [Formula: see text] increases with plasma density and effective charge, while it decreases with plasma current and does not depend on the toroidal magnetic field. The highest value of [Formula: see text] is observed in high-density low-current regimes. More negative values of plasma electric potential correlate with higher values of [Formula: see text]. The empirical scaling of [Formula: see text] is derived. The transport model based on this scaling allows us to describe experimental profiles of ion temperature [Formula: see text] within experimental error [Formula: see text].

Metabolic syndrome can lead to several challenging complications including degeneration of the pancreas and hypogonadism. Recently, we have shown that Bisamide Derivative of Dicarboxylic Acid (BDDA) can contribute to pancreatic restoration in mice with metabolic disorders via its positive effects on lipid and glucose metabolism, and by increasing the numbers of pancreatic stem cells. In the present study, we hypothesized that BDDA might also be effective in restoring hypogonadism caused by metabolic syndrome. Experiments were performed on male C57BL/6 mice with hypogonadism, where metabolic disorders have been introduced by a combination of streptozotocin treatment and high fat diet. Using a combination of histological and biochemical methods along with a flow cytometric analysis of stem and progenitor cell markers, we evaluated the biological effects of BDDA on testicular tissue, germ cells, spermatogonial stem cells in vitro and in vivo, as well as on fertility. We demonstrate that in mice with metabolic disorders, BDDA has positive effects on spermatogenesis and restores fertility. We also show that BDDA exerts its therapeutic effects by reducing inflammation and by modulating spermatogonial stem cells. Thus, our results suggest that BDDA could represent a promising lead compound for the development of novel therapeutics able to stimulate regeneration of the testicular tissue and to restore fertility in hypogonadism resulting from complications of metabolic syndrome.

Idiopathic pulmonary fibrosis (IPF) is a chronic progressive disease characterized by interstitial fibrosis and progressive respiratory failure. Pirfenidone and nintedanib slow down but do not stop the progression of IPF. Thus, new compounds with high antifibrotic activity and simultaneously regenerative activity are an unmet clinical need. Recently, we showed that Treamid can help restoring the pancreas and testicular tissue in mice with metabolic disorders. We hypothesized that Treamid may be effective in antifibrotic therapy and regeneration of damaged lung tissue in pulmonary fibrosis. In this study, experiments were performed on male C57BL/6 mice with bleomycin-induced pulmonary fibrosis. We applied histological and immunohistochemical methods, ELISA, and assessed the expression of markers of endothelial and epithelial cells in primary cultures of CD31+ and CD326+ lung cells. Finally, we evaluated esterase activity and apoptosis of lung cells in vitro. Our data indicate that Treamid exhibits antifibrotic activity in mice with pulmonary fibrosis and has a positive effect on capillaries of the lungs. Treamid also increases the number of endothelial progenitor cells in the lungs of animals with pulmonary fibrosis. Lastly, Treamid increases esterase activity and decreases apoptosis of CD31+ lung cells in vitro. Based on these findings, we suggest that Treamid may represent a promising compound for the development of new antifibrotic agents, which are capable of stimulating regeneration of lung endothelium in IPF patients.

This paper is devoted to the studies of the T-10 plasma impurity composition during experimental campaigns of 2014–2016, when a carbon limiter was replaced by a tungsten one and a movable lithium limiter was installed. In these experiments, the effective charge of the plasma was determined from the bremsstrahlung intensity and from CXRS measurements. The total concentration of tungsten ions in the column was estimated from the intensity of the radiation detected by AXUV. The source of light impurities (C, N, O, Li), as well as of tungsten atoms, was estimated from absolute measurements of line emission profiles. The experiment showed that the use of lithium as a limiter material can significantly reduce the flow of light impurities into the discharge and also reduce the dispersion of the W diaphragm. At the same time, the concentration of lithium nuclei in the column does not reach values measurable by CXRS diagnostics of the T-10 (0.3–0.5% of the plasma density).

The results of numerical modeling of impurity concentration profiles of tungsten ions for discharges with ohmic heating (OH) on the T-10 tokamak with W limiter (for Zeff ≈ 1) are described. It is shown that in OH discharges the maximum values of relative concentrations for each tungsten ion state are located on their radii of coronal equilibrium. Fulfillment of coronal equilibrium on the T-10 tokamak makes it possible to determine the full radiation intensity from the total concentration of tungsten ions. The results show that modeling of the Prad(r) profile for tungsten using currently accepted scaling of anomalous transport coefficients of light and moderate impurities for T-10 plasma does not make it possible to obtain complete alignment with the experimental data. The dependences of tungsten radiation intensity profiles on different published data on excitation, ionization, recombination rates, and anomalous transport coefficients are shown in the paper. A conclusion about the value of the anomalous diffusion coefficient and its radial distribution for heavy impurity (tungsten) in OH discharges at the T-10 tokamak is made.

We studied the effects of elastase, cigarette smoke extract, D-galactosamine hydrochloride, and tyrosine kinase inhibitor SU5416 on endothelial progenitor cells and angiogenesis precursors, as well as on Notch-1 expression by immature endothelial cells. Simultaneously with pulmonary emphysema, different damaging factors with diverse mechanisms of action caused pathological changes in the microvascular network of the lungs and destroyed the alveolar endothelium in female C57Bl/6 mice. D-galactosamine hydrochloride disturbed mobilization of endothelial progenitor cells expressing VEGFR (CD45-CD309+) and angiogenesis progenitors (CD45-CD309+CD117+) and their migration into emphysema expanded lungs. Elastase inhibited VEGFR-expressing endothelial progenitor cells, while cigarette smoke extract inhibited cells with CD45-CD31+CD34+ phenotype. In pulmonary emphysema provoked by elastase or D-galactosamine hydrochloride, angiogenesis was provided by endothelial cells with CD45-CD31+CD34+ phenotype, whereas in emphysema modeled with SU5416 or cigarette smoke extract, it was provided by the endothelial VEGFR-expressing cells and mature CD31+ endothelial cells, respectively. Replenishment of immature endothelial cells damaged by elastase and SU5416 involved Notch-1+ angiogenesis precursors and Notch-1+ endothelial progenitor cells with VEGFR.

The applicability of the CXSFIT code to process experimental data from Charge-eXchange Recombination Spectroscopy (CXRS) diagnostics at the T-10 tokamak is studied with a view to its further use for processing experimental data at the ITER facility. The design and operating principle of the CXRS diagnostics are described. The main methods for processing the CXRS spectra of the 5291-A line of C5+ ions at the T-10 tokamak (with and without subtraction of parasitic emission from the edge plasma) are analyzed. The method of averaging the CXRS spectra over several shots, which is used at the T-10 tokamak to increase the signal-to-noise ratio, is described. The approximation of the spectrum by a set of Gaussian components is used to identify the active CXRS line in the measured spectrum. Using the CXSFIT code, the ion temperature in ohmic discharges and discharges with auxiliary electron cyclotron resonance heating (ECRH) at the T-10 tokamak is calculated from the CXRS spectra of the 5291-A line. The time behavior of the ion temperature profile in different ohmic heating modes is studied. The temperature profile dependence on the ECRH power is measured, and the dynamics of ECR removal of carbon nuclei from the T-10 plasma is described. Experimental data from the CXRS diagnostics at T-10 substantially contribute to the implementation of physical programs of studies on heat and particle transport in tokamak plasmas and investigation of geodesic acoustic mode properties.

Biological activity of a new pegylated form of an of glucagon-like peptide-1 (GLP-1) analogue pegGLP-1 was studied in C57Bl/6 mice under normal conditions and during modeling of streptozotocin-induced type I diabetes mellitus. pegGLP-1 differs from GLP-1 (7-37) by polyethylene glycol residue covalently bound to His7, Lys26, and Lys34 of the GLP-1 molecule. It was shown that single intragastrical administration of pegGLP-1 induced an increase in GLP-1 level in blood serum of healthy mice. The maximum level of this parameter was observed in 4-8 h. pegGLP-1 elimination half-time was 8.5 h and mean retention time was 15 h. Administration of pegGLP-1 to animals with modeled type I diabetes mellitus was followed by an increase in the levels of GLP-1 and insulin in blood serum, produced a hypoglycemic effect, and improved the parameters of glucose-tolerance test. Biological activity of pegGLP-1 was higher than activity of GLP-1.

In clinical practice, the metabolic syndrome can lead to multiple complications, including diabetes. It remains unclear which component of the metabolic syndrome (obesity, inflammation, hyperglycemia, or insulin resistance) has the strongest inhibitory effect on stem cells involved in beta cell regeneration. This makes it challenging to develop effective treatment options for complications such as diabetes. In our study, experiments were performed on male C57BL/6 mice where metabolic disorders have been introduced experimentally by a combination of streptozotocin-treatment and a high-fat diet. We evaluated the biological effects of Bisamide Derivative of Dicarboxylic Acid (BDDA) and its impact on pancreatic stem cells in vivo. To assess the impact of BDDA, we applied a combination of histological and biochemical methods along with a cytometric analysis of stem cell and progenitor cell markers. We show that in mice with metabolic disorders, BDDA has a positive effect on lipid and glucose metabolism. The pancreatic restoration was associated with a decrease of the inhibitory effects of inflammation and obesity factors on pancreatic stem cells. Our data shows that BDDA increases the number of pancreatic stem cells. Thus, BDDA could be used as a new compound for treating complication of the metabolic syndrome such as diabetes.

In clinical practice, the metabolic syndrome (MetS) is often associated with chronic obstructive pulmonary disease (COPD). Although gender differences in MetS are well documented, little is known about sex-specific differences in the pathogenesis of COPD, especially when combined with MetS. Consequently, it is not clear whether the same treatment regime has comparable efficacy in men and women diagnosed with MetS and COPD. In the present study, using sodium glutamate, lipopolysaccharide, and cigarette smoke extract, we simulated lipid metabolism disorders, obesity, hyperglycemia, and pulmonary emphysema (comorbidity) in male and female C57BL/6 mice. We assessed the gender-specific impact of lipid metabolism disorders and pulmonary emphysema on angiogenic precursor cells (endothelial progenitor cells (EPC), pericytes, vascular smooth muscle cells, cells of the lumen of the nascent vessel), as well as the biological effects of pegylated glucagon-like peptide 1 (pegGLP-1) in this experimental paradigm. Simulation of MetS/COPD comorbidity caused an accumulation of EPC (CD45&minus, CD31+CD34+), pericytes, and vascular smooth muscle cells in the lungs of female mice. In contrast, the number of cells involved in the angiogenesis decreased in the lungs of male animals. PegGLP-1 had a positive effect on lipids and area under the curve (AUC), obesity, and prevented the development of pulmonary emphysema. The severity of these effects was stronger in males than in females. Furthermore, PegGLP-1 stimulated regeneration of pulmonary endothelium. At the same time, PegGLP-1 administration caused a mobilization of EPC (CD45&minus, CD31+CD34+) into the bloodstream in females and migration of precursors of angiogenesis and vascular smooth muscle cells to the lungs in male animals. Gender differences in stimulatory action of pegGLP-1 on CD31+ endothelial lung cells in vitro were not observed. Based on these findings, we postulated that the cellular mechanism of in vivo regeneration of lung epithelium was at least partly gender-specific. Thus, we concluded that a pegGLP-1-based treatment regime for metabolic disorder and COPD should be further developed primarily for male patients.

The investigations of tungsten transport in T-10 plasma are presented in the current paper. The process of tungsten accumulation in OH discharges and its dependence on parameters ne, Ip, and Zeff are discussed. Peaked tungsten density profiles nW(r) are observed in plasmas with Zeff ≈ 1–1.5. In plasmas with Zeff ≈ 3–4, W accumulation is mainly integral with no profile peaking with regard to electron density ne(r). Lower densities of W are observed in discharges with a lithized T-10 vacuum vessel in comparison with plasma contaminated with light impurities. The disposal of W accumulation is produced by auxiliary ECRH input. During the central ECRH, the tungsten density profile flattens with the central density drop off by ~10 times together with W source increase up to 4 times.

The regenerative potential of stem and progenitor cells from ischemic testes of C57Bl/6 mice was studied in vitro (cell culture) and in vivo (mouse model of busulfan-induced suppression of spermatogenesis). Spermatogonial stem cells with phenotypes CD117-CD90+ and CD51-CD24+CD52+ from ischemic testes demonstrated 33-fold and 7-fold increments of cell mass and generated colonies in vitro. Epithelial (CD45-CD31-Sca-1+CD49f+) and endothelial (CD45-CD31+) precursors exhibited lower self-renewal capacity. On day 30 after injection of stem and progenitor cells from ischemic testes to the rete testis zone of the testes of busulfantreated animals, an increase in the count of CD117-CD90+ spermatogonial stem cells, total count, and mobile sperm count in the testes of recipient mice was observed. In addition, we observed an increase in Sca-1+ cell count, recovery of the spermatogenic epithelium in the seminiferous tubules, and appearance of immature Leydig cells in "busulfan" testes; the level of tissue testosterone and fertility index also increased.

The paper describes the scanning spectroscopic diagnostics designed for measurement of line integrated plasma radiation in two visible spectral ranges. This diagnostic system is aimed at measuring the bremsstrahlung absolute values and profile with high spatial resolution. The bremsstrahlung absolute values are used to determine the value and radial distribution of effective plasma ion charge Zeff(r) in T-10 discharges. The importance of Zeff measurement is due to its strong influence on plasma heating, confinement, and stability. The spatial distribution of emission for one of the chosen spectral lines is measured simultaneously with bremsstrahlung. The spatial resolution of measurements is ~1 cm, and the temporal resolution is up to 10 ms. The spectral equipment and methods for its calibration are described. Examples of line integrated brightness distribution in a “continuum window” of 5236 ± 6 A and brightness of the lines C5+ (5291 A), He1+ (4686 A), and Dβ (4861 A) are given. Flattening of the bremsstrahlung brightness profile in the central region of the plasma column in some discharges with sawtooth oscillations in the T-10 is observed. The measured effective ion charge profiles in ohmic discharges with high plasma density and low discharge currents demonstrate accumulation of light impurities at the column axis; as a consequence, quenching of sawtooth oscillations in some discharges is observed. The developed diagnostics provides necessary data for investigation of heat, particle, and current transport in the plasma of the T-10. Successful application of the obtained data on Zeff(r) for investigation of geodesic acoustic modes of plasma oscillations in the T-10 should be specially noted.

This article presented a rare clinical case in Urology - Dirofilariasis of the penis. Filaria are biohelminths, which are most common in tropical countries. Dirofilariasis is the only human filariasis, found in the territory of the Russian Federation. Mosquitoes are carriers of the disease. Changing climate conditions in the direction of warming, the availability of international travel, non-compliance with preventive measures lead to an increase in the number of cases of infection by helminths, including rare species.

Ureteral resection for the mid-ureter urothelial carcinoma is the operation of choice in patients with low-grade tumors. However, extensive excision within normal tissues may lead to difficulty in appropriate apposition of the ends of the ureter, while incomplete resection increases the risk of oncological progression. This article describes the first experience with laparoscopic segmental ureteral resection with the ileal - ureter substitution for mid-ureter urothelial carcinoma. In this case, a short non-reconfigurated segment of the ileum was interposed between the distal and proximal ends of the resected ureter. Operative time was 230 min, and blood loss was less than 100 ml. No complications were observed. The patients postoperative hospital stay was seven days. Follow-up examination 12 months after surgery showed no evidence of the disease progression and preserved normal renal function. The proposed method may be considered as an alternative treatment for carefully selected patients.

The model of streptozotocin-induced diabetes mellitus in C57Bl/6 mice was employed to study the role of precursors of insulin-producing β-cells, hematopoietic stem cells, and progenitor hematopoietic cells in inflammation. In addition to provoking hyperglycemia, streptozotocin elevated serum levels of IL-1β and hyaluronic acid, induced edema in the pancreatic insular tissue and its infiltration by inflammatory cells (neutrophils, lymphocytes, and macrophages) and fibroblasts. Inflammation in pancreatic islets was accompanied by necrotic processes and decreasing counts of multipotent progenitor β-cells (CD45(-), TER119(-), c-kit-1(-), and Flk-1(-)), oligopotent progenitor β-cells (CD45(-), TER119(-), CD133(+), and CD49f(low)), and insulinproducing β-cells (Pdx1(+)). Pancreatic infl ammation was preceded by elevation of the number of short-term hematopoietic stem cells (Lin-Sca-1(+)c-kit(+)CD34(+)) relative to long-term cells (Lin(-)Sca-1(+)c-kit(+)CD34(-)) in the bone marrow as well as recruitment of hematopoietic stem and progenitor cells into circulation. Transplantation of bone marrow hematopoietic stem and progenitor cells from diabetic C57Bl/6 donor mice to recipient CBA mice with 5-fluorouracilinduced leukopenia accelerated regeneration of granulocytopoiesis in recipient mice.

The experiments on recording molecular emission in the edge plasma of the T-10 tokamak are described. To obtain reliable spectra with sufficient spectral, temporal, and spatial resolution, the optical circuit is optimized for various experimental conditions. Typical spectra measured in two sections of the tokamak are shown. It is shown that, upon varying the parameters of the discharge, the molecular spectrum not only changes significantly in intensity but also undergoes a qualitative change in the rotational and vibrational structure. For a detailed analysis, we use the Fulcher-α system (d3Πu–a3Σ g + ) of deuterium in the wavelength range from 590 to 640 nm. The rotational temperatures of ground state X1Σ g + and upper excited state d3Πu are estimated by the measured spectra.

This work presents the results of modernization of the CXRS (charge exchange recombination spectroscopy) diagnostics [1] at the T-10 tokamak. The relevance of this work is due to the importance of measurements of the ion temperature and nuclei density of the working gas and impurities for analysis of transport processes in the plasma ion component. Measurements of radial profiles of the ion temperature are extremely important for investigating the geodesic acoustic mode behavior which is conducted at the T-10 [2]. The modernized scheme of CXRS measurements, as well as the design and operational features of the spectrometer created for the new diagnostics, is described. Principles of data recording and further processing are considered in detail; attention is given to the problem of calibration of the whole complex of equipment. The performed changes in diagnostics allow the measurements to be taken simultaneously in three spectral intervals: in the region of the beam line Hα, the CXRS line of carbon ion C5+, and the CXRS line of one of the hydrogen-like ions: He1+, Li2+, N6+, O7+ or Ne9+. This makes it possible to measure the density profiles of two plasma impurities simultaneously, as well as the ion temperature from CXRS lines of different elements. The modernized diagnostics significantly broadens the possibilities of studying the physics of transport processes and quasi-coherent modes of plasma oscillations at the T-10.

The report addresses a remote molecular diagnostics system for studies of the edge plasma inT-10 tokamak at National Research Centre Kurchatov Institute. Web control of a monochromator and a CCD camera based on Internet technologies was used to monitor radiation. The structure and communications chart of the software components implemented in the system are described. A special web application has been developed to visualize, identify and analyze protium and deuterium molecular spectra. It is accessible to remote users through their private accounts during and after a remote control session. The system allows researchers located far away from the unique high-temperature plasma test facility to take an active part in experiments.

Course administration streptozotocin to male C57Bl/6 mice induces a complex of symptoms typical of type 1 diabetes mellitus: hyperglycemia and insulin deficiency, focal inflammatory infiltration of the pancreas, destructive changes in the Langerhans islets, damage to the insular apparatus (reduced number of PDX1+ cells and insulin expression by the secreting cells). Male reproductive disorder are serious complications of type 1 diabetes mellitus. In "diabetic" mice, interstitial edema with inflammatory infiltration and microvascular disorders in the testicular tissue are observed, the number of endothelial precursors (CD45-/CD31+) and the total number and percentage of motile spermatozoa decreased, immature spermatogenic epithelium cells are desquamated of into the lumen of the tubules. Disturbances in the proliferation and differentiation of various spermatogonial stem cell populations (c-kit-/CD90+, c-kit+/CD90+, and CD51-/CD24+/CD52+) in diabetes can be explained by the inhibitory influence of inflammatory factors on testosterone-producing Leydig cells.

Genitourinary tract is the second most common site where extrapulmonary tuberculosis (TB) occurs. Genitourinary TB is notable for a latent clinical course and difficult diagnosis. The paper presents clinical observations of two patients treated in a urology department of a general public hospital. One of them was diagnosed with tuberculosis of the prostate, MTB+. In the other, TB of the prostate was suspected based on pathologic assessment of the surgical specimen after surgery for prostate cancer.

In 2020 a modernized tokamak T-15MD (R=1.5 m, a=0.67 m, κ=1.8, Ipl≤2 MA, Bt≤2 T, discharge duration ≤10 s, Paux≤24 MW) will be launched in the National Research Center "Kurchatov Institute". For this installation, a modern Charge eXchange Recombination Spectroscopy (CXRS-diagnostics) has been developed. It can function both on a diagnostic injector (EDNB=60 keV) and heating injector (ENBI=80 keV) . CXRS-diagnostics of T-15MD can be used to measure local values of the following plasma parameters with high spatial (≈1 cm) and temporal (≈1 ms) resolutions: ion temperature (Ti), density of main gas and light impurities nuclei (nz), toroidal and poloidal rotation velocity (Vpol, Vtor) and the fast-ion spectrum. The composition of the CXRS-diagnostic complex and its schematic is presented. Its main components are the DNB (H, 60 keV, 150 kW), four spectrometers (two of them based on Echelle gratings, one based on holographic grating and another utilizes both types of gratings), and EMCCD-cameras. The attenuation of the diagnostic beam in the plasma is calculated considering its ionization and charge exchange by the main gas and impurities (C, O), as well as electron impact ionization. The intensities of active and passive components of λ=5291 Aline of C5+ ion are calculated considering the line fine structure, Doppler, and Zeeman effects. The active and passive signals emitted by C5+ ion are estimated for toroidal and poloidal registration systems. It is established that the acquisition time of 1 – 3 ms is enough to achieve the valid signal level to measure Ti with a statistical error ≈ 3 %. The calculated ratio between the active and passive CXRS-signal in the plasma center is 2.2 – 3.7. Measurement error is estimated for λ=5291 ACX-line and calculations show low measurement errors for Ti and nz (around 2 – 5 %), Vpol and Vtor (around 2 km/s). This indicates that CXRS-diagnostics have potential to provide reliable and accurate measurements of the named plasma parameters on T-15MD.

The article represents the study of quality of life in patients with muscle-invasive bladder cancer stages T2b and T3a after surgery, depending on the operation type. The study was conducted using questionnaires EORTC QLQ-30 and FACT-BL. The best levels of quality of life were observed in patients after organ-saving treatment of muscle-invasive bladder cancer.

The authors determined the detection rates of prostatic intraepithelial neoplasia (PIN) in 2317 patients with benign prostatic hyperplasia (BPH) and prostate cancer (PC); and those of chronic prostatitis and fibrosis in patients with PIN, BPH, or PC. There was no difference in median age between the groups. PC was found to be more concurrent with PIN 2 than with BPH. The severer inflammation or fibrosis is, more likely there is a concomitance with PIN 2 or PC. There is evidence for the theory of inflammation is a factor of carcinogenesis. Prostatic fibrosis may also initiate carcinogenesis.

The investigation examined the feasibility of cross-polarization optical cohe-rence tomography (CP OCT) to detect early urinary bladder cancer (UBC). Studies were performed in 376 patients; 5290 images were obtained using an OCT 133-U optical coherence tomograph. To acquire and compare intrared-light scattering images in baseline and orthogonal polarizations is the basis of CP OCT; their analysis makes it possible to judge from the state of the epithelium/connective tissue system and to obtain information on changes in tissue depolarizing components, collagen in particular. The authors elaborated criteria as determinants of the nature of CP OCT changes in direct and orthogonal polarizations in health, inflammatory changes, and UBC at its early stage - urothelial dysplasia and carcinoma in situ in flat suspected areas.

Whether cross-polarization (CP) optical coherence tomography (OCT) could be used to detect early bladder cancer was ascertained; it was compared with traditional OCT within the framework of blind (closed) clinical statistical studies. One hundred and sixteen patients with local nonexophytic (flat) pathological processes of the bladder were examined; 360 CP OCT images were obtained and analyzed. The study used an OCT 1300-U CP optical coherence tomographer. CP OCT showed a high (94%) sensitivity and a high (84%) specificity in the identification of suspected nonexophytic areas in the urinary bladder.

The antifibrotic properties of spiperone and its effect on stem and progenitor cells were studied on the model of reversible bleomycin-induced pulmonary fibrosis in C57Bl/6 mice. Spiperone reduced infiltration of the alveolar interstitium and alveolar ducts with inflammatory cells and prevented the growth of the connective tissue in the parenchyma of bleomycin lungs. Apart from anti-inflammatory effect, spiperone suppressed bone marrow hemopoietic cells (CD3, CD45R (B220), Ly6C, Ly6G (Gr1), CD11b (Mac1), TER-119)-, Sca-1+, c-Kit+, CD34- and progenitor hemopoietic cells (granulocyte-erythroid-macrophage-megakaryocytic and granulocyte CFU). Spiperone-induced disturbances of fi brogenesis were paralleled by restoration of endothelial cells in the lung parenchyma, reduction of the number of circulating bone marrow cells and lung mesenchymopoietic cells (mesenchymal multipotent stromal cells (CD31-, CD34-, CD45-, CD44+, CD73+, CD90+, CD106+) and progenitor fi broblast cells), and suppression of multilineage differentiation of multipotent mesenchymal stromal cells (including fi broblast-lineage cells).
