- Abstract
This paper presents an optoelectronic design for high-definition imaging in low-light underwater environments. By integrating a high-performance camera with a custom-designed electronic system, we obtain a photogrammetry system capable of being integrated into an ROV or operated by divers to obtain high quality images of marine habitats and species. These images are then processed to obtain a three-dimensional reconstruction of the environment and allow detailed monitoring., Peer Reviewed

- Abstract
Monitoring threatened species is essential for setting conservation priorities and measures to prevent species extinction. The endemic species Pinna nobilis is now classified on the IUCN Red List as a critically endangered species, with the only possibility to observe entire populations in very specific coastal areas, and a few resistant individuals in open waters. Thus, monitoring and management of these populations are crucial to avoid global species extinction. The present study aims to monitor P. nobilis Mar Menor's coastal lagoon population and to evaluate its possible resilience to recuperate from several environmental catastrophes and human mediated impacts that drastically decimated the population between 2016 and 2019. To do so, between 2019 and 2022, we assessed its population maintenance and its reproductive success. First, we monitored changes in overall densities and shell growth, and we installed between 23 and 45 larvae collectors to assess reproductive success. We observed stabilization of population densities and positive shell growth of individuals. However, no incorporation of recruits into this ageing population was observed. Our results highlighted the lack of species recovery and the first signs of probable local population extinction. Therefore, urgent management and conservation actions focused on restoring the ecosystem and protecting P. nobilis individuals are needed to avoid this population extinction and loss of one of the last reservoirs of this emblematic species.

- Abstract
The development of tourism and intensification of agriculture has released large amounts of nitrogen and phosphorus into the Mar Menor coastal lagoon in South-east Spain, resulting in a phytoplankton bloom in 2016. This bloom turned the clear and transparent waters turbid and greenish, and killed approximately 85% of benthic macrophytes. Nutrient bioextraction by flat oysters, Ostrea edulis, has been proposed for remediation of these eutrophication events and water quality recovery. This research aims to quantify the clearance rate and investigate the genetic origin of Mar Menor oysters under eutrophized conditions for potential applications to bioremediation projects. Oligotrophic and eutrophic conditions were replicated in the laboratory, and oyster feeding behaviour (i.e. clearance rates, ingestion rates, absorption efficiency and absorption rates) were studied using a flow-through system. The genetic characterization of oysters showed no significant difference between individuals from the Mar Menor and individuals collected from a nearby Mediterranean bed (Tabarca Island). Based on the physiological results observed, oysters were grouped into high-feeder (HF) and low-feeder (LF) categories according to their clearance rate, that was 3-fold higher in the HF group. Different responses in feeding behaviour were observed under eutrophic conditions in both oyster groups. Constraints in the absorption capacity of LF oysters seemed to be related to their reduced filtering activity. Lower body condition of LF oysters was evidenced by their negative scope for growth value. From this work, several conclusions can be drawn for future restoration/bioextraction actions: (i) the recovery of half of the oyster population that existed in the past would act as an effective top-down control on the phytoplankton community; (ii) using clearance rate measurements is recommended to select oysters as broodstock; and (iii) Mediterranean oysters could be used as broodstock in the event

- Abstract
The Mar Menor is a hypersaline coastal lagoon representing one of the most iconic natural areas in Southeastern Spain. However, it has suffered several catastrophic environmental events causing an exceptional eutrophication status. A study was performed to assess the recruitment of oysters in the Mar Menor, determine their taxonomic allocation and evaluate functionality in filtration and growth A total of 45 spat collectors were installed in June 2020 and posteriorly retrieved in October 2020. In total, 3431 oyster recruits were counted. Two species of oyster Ostrea edulis and O. stentina, inhabit the Mar Menor, but they are indistinguishable at the morphological level when they are small. Thus, a genetic tool was developed to identify both species. From a total of 90 oyster recruits, 95% of them corresponded to O. stentina. Finally, growth rate and clearance measurements were conducted on 661 oysters over 6 months under laboratory conditions to explore their possible use in the bioremediation of the lagoon.

The Pinna nobilis, a Mediterranean mollusc, has suffered population declines due to a massive mortality event associated with various factors including the parasite Haplosporidium pinnae. Some populations show resilience, possibly due to local environmental conditions. In this study, a molecular multiplex PCR method was developed using species‐specific primers targeting Internal Transcribed Spacer (ITS) regions of P. nobilis and P. rudis, allowing accurate species identification and hybrid detection. Samples from Mediterranean areas were analysed, including putative hybrids and individuals from five other bivalve species. DNA was isolated, ITS regions were amplified and sequenced, and phylogenetic analyses confirmed species differentiation and primer specificity. The multiplex‐PCR successfully identified P. nobilis, P. rudis, and their hybrids based on distinct amplicon patterns. This study highlights the value of molecular tools in species conservation, especially for monitoring and managing hybridization, supporting effective biodiversity conservation strategies. - Abstract
A mass mortality event (MME) affecting the fan mussel Pinna nobilis was first detected in Spain in autumn 2016 and spread north- and eastward through the Mediterranean Sea. Various pathogens have been blamed for contributing to the MME, with emphasis in Haplosporidium pinnae, Mycobacterium sp. and Vibrio spp. In this study, samples from 762 fan mussels (necropsies from 263 individuals, mantle biopsies from 499) of various health conditions, with wide geographic and age range, taken before and during the MME spread from various environments along Mediterranean Sea, were used to assess the role of pathogens in the MME. The number of samples processed by both histological and molecular methods was 83. The most important factor playing a main role on the onset of the mass mortality of P. nobilis throughout the Mediterranean Sea was the infection by H. pinnae. It was the only non-detected pathogen before the MME while, during MME spreading, its prevalence was higher in sick and dead individuals than in asymptomatic ones, in MME-affected areas than in non-affected sites, and it was not associated with host size, infecting both juveniles and adults. Conversely, infection with mycobacteria was independent from the period (before or during MME), from the affection of the area by MME and from the host health condition, and it was associated with host size. Gram (-) bacteria neither appeared associated with MME.

- Abstract
Studies of non-indigenous species (NIS) often tend to focus on medium and large-sized taxa with potential for remarkable ecological and/or economic impact, whereas the early detection of small invertebrates is often delayed due to taxonomic challenge, lack of consistent, standardised monitoring efforts and limited funding. This study represents the first records of the marine amphipod Laticorophium baconi (Shoemaker, 1934) in Morocco, Tunisia, Corsica (France), Italy, Greece, Egypt, Saudi Arabia and New Caledonia (France). Furthermore, it reports an expansion of its known distribution in Portugal (earliest and northernmost record for the country and first record for Macaronesia), Spain (earliest record for Atlantic and Mediterranean European waters), and Australia (first record for Indian Ocean). Recreational boating and commercial shipping, mainly through hull fouling and secondarily ballast waters, are proposed as vectors for introduction and secondary spread of L. baconi. The following traits, analysed during the present study, could contribute to its invasive potential: (i) quick and extensive spread of the species worldwide, (ii) high densities in marinas, harbours, hull fouling and other artificial habitats, including aquaculture facilities and floating debris, (iii) high ability for short-term colonisation of empty artificial niches, (iv) diet based on detritus suggesting an opportunistic feeding behaviour, and (v) population survival during seasonal fluctuations in different regions. Taxonomic expertise and scientific collaboration, based on multidisciplinary networks of experts, are crucial for the early detection, distribution updates, and risk assessment of small and overlooked stowaways in marine environments.

- Abstract
The fan mussel Pinna nobilis is experiencing a mass mortality event throughout the Mediterranean Sea. The survival of the remaining isolated populations and the maintenance and reproduction of individuals in captivity could determine the future of the species. This paper examines the clearance rate (CR) and oxygen consumption (VO) of fan mussel individuals measured under 5 different temperatures (8.5, 14, 18, 23 and 28ºC). The measurements of both physiological variables revealed a threshold limit of thermal tolerance at both extreme temperatures, indicating the critical situation of the remaining populations, which are located in coastal lagoons and shallow bays where these extreme temperatures usually occur. Besides, the high clearance rates observed highlight the significant ecosystem service provided by the species in terms of water filtration, especially in confined waters with low renewal rates. Routine control of the clearance rate as an early warning system is proposed for detecting ailing individuals. Such a system could also be used to improve captivity conditions.

- Abstract
In this study, we investigated the presence of the parasite Haplosporidium pinnae, which is a pathogen for the bivalve Pinna nobilis, in water samples from different environments. Fifteen mantle samples of P. nobilis infected by H. pinnae were used to characterize the ribosomal unit of this parasite. The obtained sequences were employed to develop a method for eDNA detection of H. pinnae. We collected 56 water samples (from aquaria, open sea and sanctuaries) for testing the methodology. In this work, we developed three different PCRs generating amplicons of different lengths to determine the level of degradation of the DNA, since the status of H. pinnae in water and, therefore, its infectious capacity are unknown. The results showed the ability of the method to detect H. pinnae in sea waters from different areas persistent in the environment but with different degrees of DNA fragmentation. This developed method offers a new tool for preventive analysis for monitoring areas and to better understand the life cycle and the spread of this parasite.

- Abstract
Pinna nobilis (Linnaeus, 1758) populations have been severely damaged in the last few decades, and since early autumn 2016, a mass mortality event (MME) has drastically impacted populations in the Mediterranean Sea. Accordingly, the aim of the present study was to improve the knowledge on the status of P. nobilis populations in the Bizerte Lagoon (Tunisia) between 2016 and 2022. Before the MME, P. nobilis was found in the lagoon at depths from 1.5 to 6 m, with a density ranging from 2 to 30 ind/100 m². After the MME, mortality reached 100% in the monitored area, except in the eastern part of the Bizerte Lagoon near the Menzel Jemil site, where some living specimens were detected. Moreover, in 2022, successful recruitment was observed in the lagoon. Additionally, our results revealed the presence of Haplosporidium pinnae and Mycobacterium sp. in some living specimens sampled in the Bizerte Lagoon after the MME. This finding constitutes the first detection of both pathogens in Tunisia and in the entire Southern Mediterranean Sea.

- Abstract
Studies of non-indigenous species (NIS) often tend to focus on medium and large-sized taxa with potential for remarkable ecological and/or economic impact, whereas the early detection of small invertebrates is often delayed due to taxonomic chal-lenge, lack of consistent, standardised monitoring efforts and limited funding. This study represents the first records of the marine amphipod Laticorophium baconi (Shoemaker, 1934) in Morocco, Tunisia, Corsica (France), Italy, Greece, Egypt, Saudi Arabia and New Caledonia (France). Furthermore, it reports an expansion of its known distribution in Portugal (earliest and northernmost record for the country and first record for Macaronesia), Spain (earliest record for Atlantic and Mediterranean European waters), and Australia (first record for Indian Ocean). Recreational boating and commercial shipping, mainly through hull fouling and sec-ondarily ballast waters, are proposed as vectors for introduction and secondary spread of L. baconi. The following traits, analysed during the present study, could contribute to its invasive potential: (i) quick and extensive spread of the species worldwide, (ii) high densities in marinas, harbours, hull fouling and other artificial habitats, including aquaculture facilities and floating debris, (iii) high ability for short-term colonisation of empty artificial niches, (iv) diet based on detritus suggesting an opportunistic feed-ing behaviour, and (v) population survival during seasonal fluctuations in different regions. Taxonomic expertise and scientific collaboration, based on multidisciplinary networks of experts, are crucial for the early detection, distribution updates, and risk assessment of small and overlooked stowaways in marine environments.

- Abstract
The fan mussel Pinna nobilis is currently on the brink of extinction due to a multifactorial disease mainly caused to the highly pathogenic parasite Haplosporidium pinnae, meaning that the selection pressure outweighs the adaptive potential of the species. Hopefully, rare individuals have been observed somehow resistant to the parasite, stretching the need to identify the traits underlying this better fitness. Among the candidate to explore at first intention are fast-evolving immune genes, of which toll-like receptor (TLR). In this study, we examined the genetic diversity at 14 TLR loci across P. nobilis, Pinna rudis and P. nobilis × P. rudis hybrid genomes, collected at four physically distant regions, that were found to be either resistant or sensitive to the parasite H. pinnae. We report a high genetic diversity, mainly observed at cell surface TLRs compared with that of endosomal TLRs. However, the endosomal TLR-7 exhibited unexpected level of diversity and haplotype phylogeny. The lack of population structure, associated with a high genetic diversity and elevated dN/dS ratio, was interpreted as balancing selection, though both directional and purifying selection were detected. Interestingly, roughly 40% of the P. nobilis identified as resistant to H. pinnae were introgressed with P. rudis TLR. Specifically, they all carried a TLR-7 of P. rudis origin, whereas sensitive P. nobilis were not introgressed, at least at TLR loci. Small contributions of TLR-6 and TLR-4 single-nucleotide polymorphisms to the clustering of resistant and susceptible individuals could be detected, but their specific role in resistance remains highly speculative. This study provides new information on the diversity of TLR genes within the P. nobilis species after MME and additional insights into adaptation to H. pinnae that should contribute to the conservation of this Mediterranean endemic species.

- Abstract
The critically endangered fan mussel Pinna nobilis, endemic to the Mediterranean Sea, faces a drastic population decline all over the Mediterranean Sea owing to an infectious disease. Coastal lagoons represent one of this species’ last reservoirs; thus, knowledge of the population conservation status in these ecosystems is essential. An extensive monitoring programme was conducted during 2014, 2017 and 2019 in the Mar Menor lagoon, a highly eutrophicated lagoon from Spain. On average, 32,535 m2 have been surveyed each year to determine population densities, distributions and size structures. The cumulative impacts produced by intensive agriculture activity around the lagoon, together with a catastrophic meteorological phenomenon that occurred in 2019, have triggered a significant decline in fan mussel population densities and in population mean shell sizes over the years. Additionally, some fan mussels were infected by the protozoan Haplosporidium pinnae. Therefore, the current P. nobilis population from the Mar Menor is mainly composed of young individuals in specific areas that are facing an extreme situation. Urgent management and conservation actions focused on restoring the ecosystem and protecting P. nobilis individuals are urgently needed, together with reinforcement projects to increase the species’ stock in one of its last reservoirs.

Climate change is causing an increase in the frequency and intensity of marine heatwaves (MHWs) and mass mortality events (MMEs) of marine organisms are one of their main ecological impacts. Here, we show that during the 2015–2019 period, the Mediterranean Sea has experienced exceptional thermal conditions resulting in the onset of five consecutive years of widespread MMEs across the basin. These MMEs affected thousands of kilometers of coastline from the surface to 45 m, across a range of marine habitats and taxa (50 taxa across 8 phyla). Significant relationships were found between the incidence of MMEs and the heat exposure associated with MHWs observed both at the surface and across depths. Our findings reveal that the Mediterranean Sea is experiencing an acceleration of the ecological impacts of MHWs which poses an unprecedented threat to its ecosystems' health and functioning. Overall, we show that increasing the resolution of empirical observation is critical to enhancing our ability to more effectively understand and manage the consequences of climate change.

- Abstract
Background: The measurement of the energy available for growth (scope of growth, SFG) can be used in bivalves to make a long-term prediction in a short-term experiment of the condition of the individual. In order to tackle the best conditions for captive maintenance of Mediterranean Pinnids, a SFG study was conducted using Pinna rudis as a model species. Three diets were examined to test the viability of live microalgae and commercial products: i) a control diet using 100% of live microalgae based on the species Isochrysis galbana (t-ISO), ii) a 100% of commercial microalgae diet based on the product Shellfish Diet 1800®, and iii) a 50/50% mix diet of I. galbana (t-ISO) and Shellfish Diet 1800®. Results: SFG results showed significant differences among diets in the physiological functions measured and suggested lower acceptability and digestibility of the commercial product. Negative SFG values were obtained for the commercial diet which indicates that it should be rejected for both Pinnid maintenance. The mixed diet showed improved physiological performance compared to the commercial diet, resulting in a higher SFG that had no significant differences with the control diet. However, in the long-term, the lower digestibility of the mixed diet compared to the control diet could lead to a deterioration of individuals’ conditions and should be considered cautiously. Conclusions: This work represents the first case study of SFG in Pinna spp. and provides fundamental data on dietary needs for the critically endangered species, P. nobilis.

- Abstract
The endemic species Pinna nobilis is the most endangered Mediterranean bivalve, facing nearly extinction all over the Mediterranean Sea, hosting its last reservoirs in highly impacted coastal lagoons. Thus, knowledge about the populations' conservation status in these ecosystems is essential. In 2019, the Mar Menor lagoon’s population was considered as a highly vulnerable population (Nebot-Colomer et al., 2021) due to several ecological disasters. The present study represents a continuation of the previous work, which aims to evaluate the resilience of the population, by assessing its reproductive success and maintenance of the population. To do so, between 2019 to 2022, we installed between 23-45 larvae collectors, monitored 13 permanent individual’s plots, and conducted visual searches and censuses. Overall, densities remained stable over years, although the number of individuals alive monitored in permanent plots decrease each year. Moreover, none of the methodologies carried out detected the incorporation of recruits in the population. Our results pointed out to opposite signs of recovery of the species, increasing its vulnerability to future disturbances. Therefore, urgent management and conservation actions focused on restoring the ecosystem and protecting P. nobilis individuals are needed to avoid this population extinction.

- Abstract
Research in marine Spanish waters are focused on several actions to achieve an effectively management on protected areas, with the active participation of the stakeholders and research as basic tools for decision-making. Among these actions, there is one about the knowledge and control on NIS. One of its objectives is the creation of NIS factsheets, which are going to be added to the National Marine Biodiversity Geographical System (GIS) providing complementary information about taxonomic classification, common names, taxonomic synonyms, species illustrations, identification morphological characters, habitat in the native and introduced regions, biological and ecological traits, GenBank DNA sequences, world distribution, first record and evolution in the introduced areas, likely pathways of introduction, effects in the habitats and interaction with native species, and potential management measures to apply. The database will also provide data for (1) the European online platforms, (2) the environmental assessment for the Descriptor 2 (D2-NIS) of the EU Marine Strategy Framework Directive (MSFD), as well as (3) supporting decisions made by stakeholders. It is the result of extensive collaboration among scientist, manager’s and citizen science in the Spanish North-Atlantic, South-Atlantic, Gibraltar Strait-Alboran, Levantine-Balearic and Canary Islands marine divisions, providing an updated overview of the spatial distribution of relevant extended and invasive NIS of recent and established NIS introduced by maritime transport and aquaculture pathways, as well as on cryptogenic or native species in expansion due to the climatic water warming trend.

- Abstract
A mass mortality event (MME) affecting the fan mussel Pinna nobilis was first detected in Spain in autumn 2016 and spread north- and eastward through the Mediterranean Sea. Various pathogens have been blamed for contributing to the MME, with emphasis in Haplosporidium pinnae, Mycobacterium sp. and Vibrio spp. In this study, samples from 762 fan mussels (necropsies from 263 individuals, mantle biopsies from 499) of various health conditions, with wide geographic and age range, taken before and during the MME spread from various environments along Mediterranean Sea, were used to assess the role of pathogens in the MME. The number of samples processed by both histological and molecular methods was 83. The most important factor playing a main role on the onset of the mass mortality of P. nobilis throughout the Mediterranean Sea was the infection by H. pinnae. It was the only non-detected pathogen before the MME while, during MME spreading, its prevalence was higher in sick and dead individuals than in asymptomatic ones, in MME-affected areas than in non-affected sites, and it was not associated with host size, infecting both juveniles and adults. Conversely, infection with mycobacteria was independent from the period (before or during MME), from the affection of the area by MME and from the host health condition, and it was associated with host size. Gram (-) bacteria neither appeared associated with MME.

Knowledge of the genetic structure and dispersal patterns for keystone and vulnerable species is essential to the establishment of conservation strategies. Since autumn 2016, the Critically Endangered Pinna nobilis bivalve has suffered mass mortality events throughout the Mediterranean Sea, causing mortality of up to 100% in affected populations. To propose appropriate reintroduction programs for its recovery, the present study sought to determine the genetic structure and local dispersal patterns from a well-documented population of P. nobilis. Using 19 microsatellite markers, we obtained genotypic information for 771 individuals from 9 localities within Cabrera National Park (CNP). The CNP population of P. nobilis was a single and homogeneous population, with nearly half of the sampled individuals being related through 333 halfsib and 14 full-sib relationships. The siblings belonged to 126 different family clusters composed of 2−8 individuals recruited during several recruitment events from up to 4 different localities. No evidence was found to suggest that the population was self-sustaining, since no parent−offspring dyad was found. However, the fine-scale dispersal patterns observed in Santa Maria Bay highlight the importance of this locality for the sustainment of the population as a whole. These findings suggest that the CNP could be a good choice for future reintroduction programs. Future studies that compile data from this and other studies conducted in CNP should be considered when modeling for reintroduction.

The presence of Pinctada imbricata radiata has been explored in the Bay of Palma (Balearic Archipelago, NW Mediterranean Sea) by means of Rapid Assessment Surveys (RAS). Forty three specimens were found in rocky substrates from recreational marinas and neighbouring natural habitats, including Cabrera National Park. The main occurrence of the exotic oyster in marinas, and also faraway in Cabrera, points to maritime transport as the primary introduction vector; whereas records in the adjacent natural habitats suggest secondary spread by natural dispersal. Considering the populations of P. imbricata radiata documented in the Balearic Archipelago, the bivalve seems to be well established in the area.
