Ayocote beans (Phaseolus coccineusL.); 2n= 2x= 22) is a legume native to the highlands of Mexico, it is important in human nutrition and detection of improvement characteristics, it shows resistance to pathogens and certain abiotic factors that cause losses in the bean crop. With the premise of identifying Phaseolusgermplasm with multiple resistance to biotic and abiotic adverse factors, this work was carried out with the objective of this work was to determine the resistance or susceptibility to two important pathogens: common blight [Xanthomonas axonopodispv. phaseoli(Smith)] and anthracnose [Colletotrichum lindemutianumSacc. & Magn (anamorph Glomerella lindemuthianaShear)] based on the identification of sequences characterized by amplified regions (SCARs). The study included 117 accessions collected at different sites in Puebla, Mexico (region called Carso Huasteco), which were analyzed with 10 SCAR markers (six markers for common blight: SAP6, BAC6, SU91, LG5, R7313, R4865, four for anthracnose:SAS13, SBB14, SAB3, SH18) at the Center for Genomic Biotechnology in Reynosa, Tamaulipas. The SAS13 and SBB14 anthracnose resistance markers are present more frequently (89 and 74%), followed by the common blight resistance markers BAC6 and SU91 (74 and 42%).The germplasm of Zacapoaxtla and Tlatlauquitepec had a higher frequency of SCARs (Zacapoaxtla: 90% and 100% for SAS13 and SBB14;Tlatlauquitepec: 94% and 56%). Accessions with five SCARs could be used as a source of resistance to diseases in Phaseolusand early and late genotypes to flowering and physiological maturity, as well as variables in testa color, pod size and seed, but no association was detected between the presence of SCARs with the morphology of ayocote beans, El frijol ayocote (Phaseolus coccineus L.); 2n= 2x= 22) es una leguminosa de las zonas altas de México, tiene importancia en la alimentación humana y detección de características de mejoramiento, muestra resistencia a patógenos y a ciertos factores abióticos que ocasionan pérdidas en el cultivo de frijol. Con la premisa de identificar el germoplasma de Phaseolus con resistencia a factores adversos bióticos y abióticos, el objetivo fue determinar la resistencia a dos patógenos importantes: tizón común [Xanthomonas axonopodis pv. phaseoli (Smith)] y antracnosis [Colletotrichum lindemutianum Sacc. & Magn (anamorfo G. lindemuthiana Shear)] con base a la identificación de secuencias caracterizadas de regiones amplificadas (SCARs). El estudio incluyó 117 accesiones colectadas en diferentes sitios de Puebla (región Carso Huasteco), mismas que se analizaron con 10 marcadores SCAR (seis marcadores para tizón común: SAP6, BAC6, SU91, LG5, R7313, R4865, cuatro para antracnosis: SAS13, SBB14, SAB3, SH18) en el Centro de Biotecnología Genómica en Reynosa, Tamaulipas. Los marcadores de resistencia a antracnosis SAS13 y SBB14 están presentes con mayor frecuencia (89 y 74%), seguidos por los marcadores de resistencia al tizón común BAC6 y SU91 (74 y 42%). El germoplasma de Zacapoaxtla y Tlatlauquitepec tuvo mayor frecuencia de SCARs (Zacapoaxtla: 90% y 100% para SAS13 y SBB14; Tlatlauquitepec: 94% y 56%). Las accesiones con cinco SCARs podrían aprovecharse como fuente de resistencia a enfermedades en Phaseolus y genotipos precoces y tardíos a floración y madurez fisiológica, así como variables en color de testa, tamaño de vaina y semilla, pero no se detectó asociación entre la presencia de SCARs con la morfología del frijol ayocote.

Ayocote beans (Phaseolus coccineus L., 2n=2x=22) are a legume native from Mexican highlands; this is one species that has been poorly studied, despite its importance for human feeding and its potential for Phaseolus breeding of disease resistance as well as against some abiotic factors as cold tolerance. The aim of this study was to determine the population structure of 117 collections of P. coccineus from 15 locations of the Huasteco Karst from Puebla, Mexico, plus five controls in order to to know the degree of genetic variability present in Mexican native germplasm materials of P. coccineus by using the Amplified Fragment Length Polymorphism (AFLP) molecular marker strategy. The optimum genetic structure indicated a delta K value = 3. The polymorphism was 87.4% and the AMOVA confirmed the low genetic differentiation among accessions (Fst = 0.067), the genetic variance explained among and within populations was 6.7 and 93.3%, respectively. Cluster analysis did not show groupings based on germplasm origins., RESUMEN El frijol ayocote (Phaseolus coccineus L.; 2n=2x=22) es una leguminosa originaria de las zonas altas de México; es una especie que ha sido poco estudiada a pesar de su importancia para consumo humano y el uso potencial en el mejoramiento de la resistencia de Phaseolus a enfermedades y factores abióticos (tolerancia a bajas temperaturas). El objetivo del presente estudio fue determinar la estructura poblacional de 117 colectas de P. coccineus correspondientes a 15 localidades del Carso Huasteco del estado de Puebla, México, más cinco testigos para conocer el grado de variablidad genética existente en germoplasma criollo mexicano de P. coccineus mediante la técnica de polimorfismos en longitud de fragmentos amplificados (AFLP). La estructura poblacional indicó un valor delta K = 3. El polimorfismo fue del 87.4% y el AMOVA confirmó la baja diferenciación genética entre accesiones (Fst = 0.067), la varianza explicada entre y dentro de poblaciones fue 6.7 y 93.3%, respectivamente. El análisis de conglomerados no mostró agrupamientos definidos con base en el origen del germoplasma.

The collection of native varieties of Phaseolus coccineus L. of the National Research Institute of Forestry, Agriculture and Livestock has 798 sheltered ex situ accessions. Representing varieties of the centre and south of the country and are still needed to include those the northern territory. The plant P. coccineus L. has a mixed mating system with predominance of crosslinking so that a population can have multiple colours of seeds and the plants display different phenology. The genebank received a collection of the Ejido San José de las Corrientes, Vicente Guerrero, Durango, Mexico. Seed colour variants were defined and plated on June 25, 2010 and May 17, 2011 in Chapingo, Mexico. We recorded: photoperiod, days and accumulated temperature of seedling emergence at flowering, and days and temperature required to start flowering at the beginning of maturity. Short-cycle variants were identified, 123-144 days and long cycle of 165 days. In May plantings, the plant bloomed for 26-36 days with 13.17 and 13.19 hours of light; and plantings on June 40-56 days to 12.33 and 12.49 hours of light due to natural photoperiod decreased over time. When the growing season increases, so does the accumulation of temperature due to the increase occurring in spring and summer at this latitude. When planting in May accumulated in plants growing season, 468-719 °C, and in June 713-990 °C, La colección de variedades nativas de Phaseolus coccineus L. del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias tiene 798 accesiones resguardadas ex situ. Están representadas las variedades del centro y sur del país y hace falta introducir las del norte del territorio. La planta de P. coccineus L. tiene sistema de apareamiento mixto con predominancia del entrecruzamiento por lo que una población puede tener varios colores de semilla y las plantas mostrar diferente fenología. El banco de germoplasma recibió una colecta del Ejido San José de las Corrientes, Municipio de Vicente Guerrero, Durango, México. Se definieron variantes por colores de semilla y se sembraron 25 de junio de 2010 y el 17 de mayo 2011 en Chapingo, México. Se registró: fotoperiodo, días y temperatura acumulada de emergencia de plántula a inicio de floración, y días y temperatura requerida de floración a inicio de madurez. Se identificaron variantes de ciclo corto, 123-144 días, y de ciclo largo de 165 días. En siembras de mayo la planta floreció a los 26- 36 días con 13.17 y 13.19 horas luz; y en siembras de junio a los 40-56 días con 12.33 y 12.49 horas luz debido a la disminución de fotoperíodo natural a través del tiempo. Al alargarse el período vegetativo aumentó la acumulación de temperatura debido al incremento que ocurre en primavera-verano en esta latitud. Al sembrar en mayo las plantas acumularon en período vegetativo, 468-719 °C, y al sembrar en junio 713-990 °C

The runner bean is native to the temperate regions of Mexico. The physiographic provinces of the country, most represented in the collection of native varieties of runner bean germplasm bank are Neovolcanic and the Sierra Madre Oriental. In the Spring-Summer 2009, cycle 231 native varieties were cultivated in the Experimental Field Valle de Mexico in Chapingo, State of Mexico. Two rows were planted on April 30, 2009 on time with seven plants in each row of 4 m. There were days to flowering and days to beginning of maturity. In October-November and harvested seed was determined in eight replicates of 100 seeds weight, in grams. We used a principal component analysis and a cluster with the method "complete". The first three components explained 73% of the variability contained in 231 varieties. The first one was related to the elevation and minimum and maximum average annual temperatures in the collection sites, the second to days to flowering and maturity of the plant, and the third with 100 seed weight, days to flowering, and precipitation of collection sites. The dendrogram associated four groups, according to their geographical origin: the first accessions of Chichonquiaco physiographic sub-province and the remaining three of the Neovolcanic sub-provinces Plains and Sierras de Querétaro and Hidalgo, Puebla Southern and Tarasca Neovolcánica. The runner beans of the Sierra Madre Oriental, Chiconquiaco subprovince, differed from those of Neovolcanic; its shorter growing cycle and small seed. The Neovolcanic runner beans of crop cycles were longer with larger seeds. In turn, the runner beans from the Neovolcanic region were different in their phenological plasticity: some of them when extending their growing season; they shortened the reproductive period, thus reducing the size of its seed. Instead, others, despite coming from very high and very cold places, maintained their long growing season and possibly seed size by the capacity of the root of "sprout" after frost, El frijol ayocote es originario de las zonas templadas del territorio mexicano. Las provincias fisiográficas del territorio nacional, más representadas en la colección de variedades nativas de frijol ayocote del banco de germoplasma del INIFAP son el Eje Neovolcánico y La Sierra Madre Oriental. En el ciclo Primavera-Verano 2009 se cultivaron 231 variedades nativas de ayocote en el Campo Experimental Valle de México en Chapingo, Estado de México. Se sembraron dos surcos el 30 de abril de 2009 bajo temporal con siete matas dentro de cada surco de 4 m. Se registraron los días a inicio de floración y días a inicio de madurez. En octubre- noviembre se cosechó la semilla y se determinó en ocho repeticiones el peso de 100 semillas en gramos. Se corrió un análisis de componentes principales y otro de agrupamientos con el método "complete". Los tres primeros componentes explicaron 73% de la variabilidad contenida en las 231 variedades. El primero se relacionó con la altitud y temperaturas mínima y máxima media anual de los sitios de colecta, el segundo con los días a floración y a madurez de la planta, y el tercero con el peso de 100 semillas, días a floración, y precipitación de los sitios de colecta. El dendograma asociació cuatro grandes grupos de acuerdo a su origen geográfico: el primero con accesiones de la subprovincia fisiográfica Chichonquiaco y los tres restantes de las subprovincias Eje Neovolcánico, Llanuras y Sierras de Querétaro e Hidalgo, Sur de Puebla, y Neovolcánica Tarasca. Los ayocotes de la Sierra Madre Oriental, subprovincia Chiconquiaco, se diferenciaron de los del Eje Neovolcánico por su ciclo de cultivo más corto y semilla pequeña. Los ayocotes del Eje Neovolcánico tuvieron ciclos de cultivo más largos con semilla de mayor tamaño. A su vez, los ayocotes del Eje Neovolcánico fueron diferentes en su plasticidad fenológica: algunos al alargar su período vegetativo, acortaron el período reproductivo disminuyendo así el tamaño de su semilla. The conservation and regeneration of seed collection of runner bean of the germplasm bank from INIFAP requires knowing the weather sites of domestication of the species. In 2010, at the Experimental Station of Valle de Mexico, were registered the passport data, locality, state, altitude, geographical coordinates, precipitation, minimum and maximum temperature and annual average of such sites. For this data and maps from Garcia and CONABIO were used (1998) 1:1 000 000 scale, the "software" GIS ARC-INFO, version 8.0 and the "hardware" SPARCstation, SUN OS, Version 5.5.1 from the Laboratory of Geographic Information Systems from INIFAP. The 798 accessions in the collection were domesticated in 140 sites in 13 states of the country, 80% at altitudes from 1 500 to 2 500 masl, precipitation from 500 to 1 000 mm annually. The wide range of minimum and maximum temperature separated sites into: very cold, -5 to -1 °C at night and 21 to 31 °C during the day, cold 3 to 5 °C at night and 25 to 33 °C during the day, and semi cold 6 to 9 °C at night and 25 to 37 °C in the day. The distribution of native varieties of the species are concentrated in high and humid areas of the country, but also adapted to semi-arid with little rainfall and very warm., La conservación y regeneración de semilla de la colección de frijol ayocote del banco de germoplasma del INIFAP requiere conocer el clima de los sitios de domesticación de la especie. En 2010, en el Campo Experimental Valle de México, se registraron los datos de pasaporte, localidad, estado, altitud, coordenadas geográficas, precipitación, y temperatura mínima y máxima media anual de dichos sitios. Para ello se utilizaron datos y mapas de García y CONABIO (1998) escala 1:1000000; el "software" SIG ARC-INFO, versión 8.0 y el "hardware" SPARCstation, SUN OS, versión 5.5.1 del laboratorio de Sistemas de Información Geográfica del instituto. Las 798 accesiones de la colección se domesticaron en 140 sitios de 13 estados del territorio nacional, 80% en altitudes de 1 500 a 2 500 msnm precipitación de 500 a 1 000 mm anuales. El amplio rango de temperatura mínima y temperatura máxima separó los sitios en: muy fríos, de -5 a 1 °C en la noche y de 21 a 31 °C en el día; fríos de 3 a 5 °C en la noche y de 25 a 33 °C en el día; y semifríos de 6 a 9 °C en la noche y de 25 a 37 °C en el día. La distribución de variedades criollas de la especie se concentra sobre todo en zonas altas y húmedas del territorio nacional, aunque también se adapta a zonas semiáridas con poca precipitación y muy cálidas.
