45. Phylogenetic relationship of the genus Gilbertellaand related genera within the order Mucorales based on 5.8 S ribosomal DNA sequences

Abstract

The complete ITS (internal transcribed spacer) region coding the ITS1, the ITS2 and the 5.8S rDNA was amplified by polymerase chain reaction from two strains of Gilbertella persicaria, six strains in the Mucoraceae(Mucor piriformis, M. rouxii, M. circinelloides, Rhizomucor miehei, R. pusillus andR. tauricus) and four strains representing three species of the Choanephoraceae(Blakeslea trispora, Choanephora infundibulifera andPoitrasia circinans).Sequences of the amplified DNA fragments were determined and analysed. G. persicariabelongs to the monogeneric family (Gilbertellaceae), however, originally it was described as Choanephora persicaria. The goal of this study was to reveal the phylogenetic relationship among fungi belonging to Gilbertellaceae, Choanephoraceae andMucoraceae.Our results support that the “intermediate” position of this family is between Choanephoraceaeand Mucoraceae.

46. Secretion of a trypsin‐like thiol protease by a new keratinolytic strain of Bacillus licheniformis

Abstract

When cultured in feather‐containing broth with a growth optimum of pH 7.0 and 47°C, a Bacillus licheniformisstrain exhibited a high chicken feather‐degrading activity. A trypsin‐like protease was isolated from its ferment broth and was partially characterized. The enzyme was constitutively secreted and was highly active towards N‐benzoyl‐Phe‐Val‐Arg‐p‐nitroanilide as chromogenic substrate. Its pH optimum was 8.5 and it exhibited the highest activity at 52°C. Fractionation on Sephadex G‐100 column revealed that its molecular mass was about 42 kDa. The enzyme, which is new for the genus Bacillus, is a thiol protease, as tosyl‐l‐phenylalanine chloromethyl ketone, tosyl‐l‐lysine chloromethyl ketone, phenylmethylsulfonyl fluoride and ethylenediamine tetraacetate did not inhibit it, while HgCl2and para‐chloromercuribenzoate lowered its activity.

47. Variation of Isoenzyme and Rapd Patterns in Candida AlbicansMorphological Mutants with Altered Colony Ultrastructure

Abstract

Molecular typing methods were applied to characterize four stable morphological mutants [1] isolated from a UV-induced unstable mutant colony of Candida albicans. The wild-type strain (ATCC 64385), the intermediate unstable mutant and its four morphologically altered derivatives revealed the same elec-trophoretic karyotypes. Of the five isoenzymes tested (catalase, malate dehydrogenase, glutamate dehy-drogenase, acid phosphatase and ß-glucosidase), glutamate dehydrogenase displayed a different enzyme pattern (with an extra band of lower mobility) in the morphological mutants. In contrast, the random amplification DNA polymorphism patterns of the mutant strains differed in all cases from that of the parental strain. Different primers revealed various degrees of DNA polymorphism; one of them (OPC-8) proved to be useful for differentiation between all examined strains. Differences in genetic alterations between spontaneous and induced mutants, and the applicability of different molecular markers to analyse the consequences of induced mutagenesis in C. albicansare discussed.

Nuclei were isolated from protoplasts of a haploid auxotrophic Aspergillus nidulans strain. Transformation of protoplasts prepared from a complementary haploid auxotrophic strain with these purified nuclei resulted in both heterokaryotic and diploid colonies. Abstract

Summary: A comprehensive study of the degradation of thiamethoxam (THIA) and metoprolol (MET) was conducted by using UV-induced photolysis (λ =254nm), ozonation, and a combination of these methods. In order to investigate how molecular structure of the substrate influences the rate of its degradation, we compared these three processes for the insecticide THIA and the drug MET (a β1-blocker). Of the three treatments applied, the UV photolysis and the combination of UV/O3 were found to be most effective in the degradation of THIA, while the UV/O3 process appeared to be the most efficient in terms of MET decay. The degradation kinetics was monitored by LC–DAD, and spectrophotometry, while the mineralization of the substrates was studied by TOC analysis. Reaction intermediates were studied in detail and a number of them were identified using LC–MS (ESI+/ESI−). Both parent compounds showed slight toxic effects towards algae Pseudokirchneriella subcapitata and bacteria Vibrio fischeri. However, the toxicity of the solutions containing also the degradation intermediates appeared to be much higher for all the test organisms. The inhibition/mortality rates were reduced most efficiently by the UV/O3 procedure. Ames test and Comet assay were used to follow the genotoxicity during the degradation of the studied compounds. Genotoxic intermediates were frequently detected in the case of MET in the UV treatment alone or in the presence of ozone. Treatments of THIA samples resulted less frequently in genotoxic intermediates. To our best knowledge, this work is the first genotoxicological investigation dealing with the photolytic degradation process of the studied compounds. [Copyright &y& Elsevier]
