WOS: 000362666900010, Probiotic yeast, Cryptococcus humicola M5-2 strain, which has high cholesterol assimilation feature and isolated from traditionally produced cheese, was investigated in vivo. This study was conducted to determine cholesterol assimilation ability of C. humicola M5-2 in vivo and specify the effect of the strain on serum total cholesterol, HDL/LDL cholesterol and triglycerides levels in rats fed on cholesterol-enriched diet. C. humicola had the ability to assimilate cholesterol at the rate of 73.33% in media. The strain was used with two different concentrations in animal feed (high and low doses containing 2% and 0.1% lyophilized strains, respectively). When the treatment groups were compared, low dose feeding group had the positive results in terms of testing values. According to the results of serum analysis, triglyceride and total cholesterol level were decreased by 25% and 1.34% respectively. Especially, decreasing the percentage rate of triglyceride has not obtained in other in vivo studies. We described heterozygous deletion of tumor protein 53 (TP53) gene in 11 patients including 2 patients with non-malignant diseases (pneumonia) and 9 patients with malignant diseases [including small cell lung cancer (n = 3), non-small cell lung carcinoma (n = 4), non-Hodgkin%u2019s lymphoma (n=1), and gastric carcinoma (n=1)]. Chromosomal aberrant status was analyzed by fluorescence in situ hybridization with centromere specific and 17p13.1 locus specific probes. In 3 of 9 cancer patients we did not find malignant pleural effusion with histological examination and/or closed pleural biopsy. Heterozygous deletion of TP53 gene was found to be significantly higher in patients with malignant disease when compared to the patients with benign pleural fluid. As a result, we suggest that heterozygous deletion of TP53 may have indicator value for malignancy; however further studies are warranted to confirm this suggestion in large patient cohorts.

Amaç: Yaban mersini olarak bilinen ve halk arasında diyabet başta olmak üzere birçok hastalıkta tedavi edici olarak kullanılan Vaccinium myrtillus, Türkiye’de yaygın bir yetişme alanına sahiptir. Bu araştırmada, Vaccinium myrtillus’un, streptozotosin (STZ) ile diyabet oluşturulmuş sıçanlarda, lipit peroksidasyonu, antioksidan sistem ve plazma glikoz düzeyi üzerine etkilerinin incelenmesi amaçlandı. Materyal ve Metot: 32 adet Sprague-Dawley yetişkin erkek sıçan, her grupta 8’er adet olmak üzere 4 gruba ayrıldı. Gruplar: Kontrol (K), streptozotosin (STZ), Vaccinium myrtillus (VM), ve Vaccinium myrtillus + streptozotosin (VM + STZ) verilecek şekilde belirlendi. STZ sitratlı tampon (0,1 M, pH 4,5) içerisinde 55 mg/kg olacak şekilde hazırlandıktan sonra sıçanlara tek doz ve intraperitoneal uygulandı. Hazırlanan ekstre hayvanlara gruplarına göre 10 gün boyunca her gün gavaj olarak verildi (100 mg/kg). 11. gün hayvan gruplarına göre streptozosin ile diyabet oluşturuldu (55 mg/kg). Kuyruktan alınan kan ile glikoz ölçüldü. Objectives: This study aims to investigate the genetic association between single nucleotide mutation in mitochondrial manganese superoxide dismutase and a Behçet's disease (BD) population by using molecular techniques.Patients and methods: Ninety-three BD patients (45 males, 48 females; mean age 33.15±8.99 years; range 17 to 65 years) and 125 controls (58 males, 67 females; mean age 28.33±7.31 years; range 18 to 62 years) were genotyped by polymerase chain reaction-restriction fragment length polymorphism method. The genotypic distributions in BD patients and controls were consistent with the Hardy-Weinberg equilibrium.Results: Significant differences were observed between BD patients and controls in terms of genotypic distribution. Frequencies of alanine (Ala)/Ala, Ala/valine (Val), and Val/Val were 14.0% (n=13), 45.2% (n=42), and 40.9% (n=38) in BD patients and 21.6% (n=27), 53.6% (n=67), and 24.8% (n=31) in controls, respectively (p=0.033).Conclusion: The Val/Val genotype of the manganese superoxide dismutase gene is associated with the physiopathology of BD in a group of Turkish patients.

AIMThe harmful effects of smoking on body are known to be caused by oxidative stress in tissues. With the many harmful substances in smoke it serve as a good model of free radical damage could be monitored. The agents preventing free radical induced oxidation, capable of trapping and stabilizing free radicals are called 'antioxidants". Resveratrol is a powerful antioxidant that has anti-inflammatory, analgesic and osteogenic effects. In this study we evaluated the effects of smoking and resveratrol on oxidative stress.MATERIAL VE METHODFor 4 weeks rats were inhalated cigarette smoke (6 cigarette/day) and after that time 20 mg/kg/day resveratrol was given via oral gavage during 28 days together with cigarette smoke. At the end of 8 weeks total exposure to cigarette smoke, rats were sacrified and bloods and tongue samples were collected. Biochemically serum and tissue total antioxidant / oxidant capacity (TAC / TOC) were evaluated.RESULTSThere were no statistical difference between groups in terms of TAC and TOC serum. TAC tongue is higher in cigarette+resveratrol and cigarette groups. CONCLUSIONIn the experiment performed with 6 cigarettes per day for 8 weeks, toxic effect of cigarette induced antioxidant system in the organism to protect the body and that is more effective than antioxidant effect of resveratrol., AMAÇSigaranın vücutta hasara neden olan etkilerinin dokularda oluşturduğu oksidatif strese bağlı olduğu bilinmektedir. Sigara içerdiği çok sayıdaki zararlı madde ile serbest radikal hasarının izlenebileceği bir model oluşturmaktadır. Serbest radikallerin neden olduğu oksidasyonları önleyen, serbest radikalleri yakalama ve stabilizasyon yeteneğine sahip maddelere 'antioksidan' adı verilir. Resveratrol antienflamatuar, osteojenik ve analjezik etkili güçlü bir antioksidandır. Bu çalışmada sigaranın neden olduğu oksidatif stres üzerine resvaratrolün etkisi araştırılmıştır.MATERYAL VE METODRatlar 4 hafta süresince sigara dumanına (6 sigara/gün) maruz bırakıldıktan sonra 28 gün boyunca 20 mg/kg/gün resveratrol intragastrik gavaj yoluyla verilmiştir. Sekiz haftalık deney süresi sonunda ratlar sakrifiye edilerek kan ve dil dokuları alınmıştır. Biyokimyasal olarak serum ve dokuda Total antioksidan/oksidan kapasite (TAK/TOK) değerlendirilmiştir. BULGULARSerum TAK ve TOK açısından gruplar arasında anlamlı fark bulunmamıştır. TAK ve TOK dil sigara+resveratrol grubunda yüksek bulunmuştur. SONUÇ 8 hafta süresince günde 6 adet sigarayla yapılan deneyde, sigaranın toksik etkisine karşı vücudun kendini korumak için aktive olan antioksidan sistemin etkisinin, resveratrolün antioksidan kapasitesinden daha yüksek olduğu sonucuna varılmıştır.

Aim: This study was performed to determine the effect of caffeic acid phenethyl ester (CAPE) treatment on levels of malondialdehyde (MDA) and nitric oxide (NO) and activities of antioxidant enzymes in retinas of streptozotocin (STZ)-induced diabetic rats. Materials and Methods: Twenty-seven rats were enrolled and divided into three groups: group 1: non-diabetic rats as control (n = 9); group 2: STZ-induced, untreated diabetic rats (n = 8); and group 3: STZ-induced, CAPE-treated diabetic rats (n = 10). Levels of MDA and NO and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in retinal tissues were analyzed. Results: In retinas of untreated diabetic rats, levels of MDA and NO and the activity of GSH-Px were significantly increased (P = 0.001, P = 0.0001, P = 0.0001, respectively), while the SOD activity was significantly decreased (P = 0.021) when compared to the control group. GSH-Px activity in the CAPE-treated diabetic group was markedly higher than that of the control group (P = 0.0001). MDA and NO levels were lower (P = 0.005, P = 0.001, respectively) and SOD activity was higher in the CAPE-treated diabetic group compared to the untreated diabetic group (P = 0.016). Conclusions: It can be concluded that nitrosative and oxidative stress is increased in the diabetic retina. CAPE treatment inhibits lipid peroxidation and NO overproduction and regulates SOD enzyme activities in diabetic rat retinas while it has no effects on GSH-Px activity.

Aim: This study aimed to determine the possible chromosomal abnormalities in leukemia patients who were diagnosed and treated in Hematology Clinics of Internal Medicine Department and Pediatric Hematology Clinic of Pediatry Deparment in Süleyman Demirel University Research Hospital.Material and Method: In study 29 patients of whom 12 in post-chemotherapeutic period, 17 in remission periods with any stage of treatment in leukemia group and 30 age-and gender-matched control person were evaluated cytogenetically. Results: Of the leukemia group; 15 had acute lymphoblastic leukemia, 5 had chronic lymphoblastic leukemia, 5 had chronic myeloid leukemia and 4 had acute myeloid leukemia. Four out of 15 patients with acute lymphoblastic leukemia, 1 of 5 with chronic lymphoblastic leukemia and 1 of 5 with chronic myeloid leukemia had different cytogenetic aberrations. t(9;22)(q34;q11) was detected in two patients, one with chronic myeloid leukemia and the other with chronic lymphoblastic leukemia.; del(17q), inv(9), inv(1), and t(12;15) are found in four ALL cases respectively. Conventional cytogenetic analysis of peripheral blood is fundamental for advanced genetic study.Key words: Leukemia, GTG banding, Chemotherapy, Chromosome, Peripheric blood., Amaç: Çalışmada Süleyman Demirel Üniversitesi Araştırma Uygulama Hastanesi Dahiliye Anabilim Dalı Hematoloji Kliniği ve Pediatri Anabilim Dalı Çocuk Hematolojisi Kliniğinde tedavisi ve takibi yapılan lösemi tanısı almış vakalarda olabilecek muhtemel kromozom anomalilerinin gözlenmesi amaçlandı. Gereç ve Yöntem: Çalışmada 12 post-kemoterapik, 17 remisyonu sağlanmış ve tedavisinin herhangi bir basamağında olan 29 lösemi vakası ile yaş ve cinsiyet olarak uyumlu 30 kontrol birey sitogenetik olarak değerlendirildi. Bulgular: Lösemi grubununu 15'i akut lenfoblastik lösemi, 5'i kronik lenfoblastik lösemi, 5'i kronik myeloid lösemi ve 4'ü akut myeloid lösemi idi. 15 akut lenfoblastik lösemi vakasının 4'ünde, 5 kronik lenfositik lösemi ve kronik myeloid lösemi vakalarının birer tanesinde anomali tespit edilmiştir. Kronik myeloid lösemi vakalarının birinde t(9;22)(q34;q11), 4 akut lenfoblastik lösemi vakasında sırasıyla; del(17q), inv(9), inv(1) ve t(12;15), kronik lenfositik lösemi vakalarından birinde ise t(9;22)(q34;q11) tespit edilmiştir.
