The aim of this study was to investigate the increasing resistance to COL and in particular its coexistence with ESBL-producing E. coli recovered from pig farms in China.Materials and methods: E. coli were isolated from 14 pig farms in Jiangsu China. Susceptibility testing was identified by micro-dilution method. PCR assay and nucleotide sequencing were used to detect COL-resistant genes, mcr-1 to −5, as well as ESBL genes, blaCTX-M, blaSHV and blaTEM. Conjugation experiment, plasmid replicon typing of the multidrug resistance (MDR), S1-PFGE and DNA southern hybridization were performed to study the transferability of these genes.Results: Overall, 275 E. coli isolates were recovered from a total of 432 cloacal and nasal swabs. More than 90% of the isolates were MDR, of which 70.18% were resistant to COL. Of these 275 isolates, mcr-1 was identified as the most predominant gene carried by 71.63% (197/275) of isolates, 39.59% (78/197) of the isolates were harboring both mcr-1 and ESBL genes (blaCTX-M, blaSHV and blaTEM). ESBL genotyping showed that blaCTX-M was the most predominant ESBL (68.49%) followed by blaSHV (16.4%) and blaTEM (15%). Sequencing revealed that the most common variants of blaCTX-M identified were, blaCTX-M-15 (69%), blaCTX-M-55 (29%) and blaCTX-M-1 (1.8%). IncHI2, IncFIB, IncFIC, IncN and IncX4 were found to be the most common Inc-types found both in donors and in transconjugants and were associated with the transfer of the mcr-1 and ESBL encoding genes. Six strains carried a total of five different plasmids: approximately 97-, 130-, 160-, 227- and 242-kb plasmids.Conclusion: The coexistence of the mcr-1- and blaCTX-M-15-carrying isolates displaying high MDR, recovered from E. coli of pig origin, is a major concern for both humans and veterinary medicine.Keywords: E. coli, colistin, mcr-1, ESBL, coexistence

This study investigates the impact of different educational activities on students' academic performance in a problem-based learning curriculum. Methods: This is a retrospective cohort study conducted on the cardiology block at the College of Medicine, King Saud Bin Abdulaziz University for Health Sciences, Riyadh, Saudi Arabia. All students who undertook the cardiology block during the academic year 2011–2012 were included. The students' attendance was measured using their overall attendance percentage. This percentage is a product of their attendance of many activities throughout the block. The students' performance was assessed by the final mark obtained, which is a product of many assessment elements. Statistical correlation between students' attendance and performance was established. Results: A total of 127 students were included. The average lecture attendance rate for the medical students in this study was found to be 86%. The -application of nanoparticles for accurate diagnosis and ample treatment to various human diseases. Despite the use of diverse polymers in nanoparticle synthesis, biosynthesis of these particles in viable, metal-tolerant microorganisms is preferred over all other prevalent procedures including chemical methods, because of its cost-effectiveness, and eco-friendliness. Diabetes is the most fifth growing trend prevalent diseases worldwide due to the lifestyle changes, physical inactivity, aging, nutrition and lack of diet nutrition. The disease spread is influenced by numerous hereditary facts, environmental causes, temporal and additional behaviour. The study purpose by using in-silico approach identify inhibitors binding with a-glucosidase against DM. The a-glucosidase protein sequence from Saccharomycescerevisiae was retrieved using NCBI and utilizing software molecular docking and AdmetSAR were perormed. AdmetSAR utilized for the identification of properties of (ligand) MgONPs associate brain blood 0.9827% barrier and human intestinal 0.9848% absorption. MgONPs biodegradation 0.77% percentage was calculated with acute oral toxicity 0.589%. Total positive and negative charged residues Arg + Lys:73, Asp + Glu: 87 were also calculated. The presence of various physiological barriers, primarily the blood–brain barrier (BBB) limits the accessibility of the brain and hinders the efficacy of various drug therapies. Hence, drug targeting to the brain, particularly to the diseased cells by circumventing the physiological barriers is essential to develop a promising therapy for the treatment of brain disorders. Presently, the investigations emphasize the role of different nanocarrier systems or surface modified target specific novel carrier system to improve the efficiency and reduce the side effects of the brain therapeutics. Such approaches supposed to circumvent the BBB or have the ability to cross the barrier function and thus increases the drug concentration in the brain. Although the efficacy of novel carrier system depends upon various physiological factors like active efflux transport, protein corona of the brain, stability, and toxicity of the nanocarrier, physicochemical properties, patient-related factors and many more. Hence, to develop a promising carrier system, it is essential to understand the physiology of the brain and BBB and also the other associated factors. Along with this, some alternative route like direct nose-to-brain drug delivery can also offer a better means to access the brain without exposure of the BBB. In this review, we have discussed the role of various physiological barriers including the BBB and blood-cerebrospinal fluid barrier (BCSFB) on the drug therapy and the mechanism of drug transport across the BBB. Further, we discussed different novel strategies for brain targeting of drug including, polymeric nanoparticles, lipidic nanoparticles, inorganic nanoparticles, liposomes, nanogels, nanoemulsions, dendrimers, quantum dots, etc. along with the intranasal drug delivery to the brain. This article investigates the rapid expansion of private schooling in the Northern Areas of Pakistan during the period 1993-2003. Our observations are mainly based on a survey of about 40% of the total number of private schools in the Northern Areas in which we attested trends that are characteristic of many private schools elsewhere, both nationally and internationally. For instance, most of the parents are by no means wealthy, most think that private schools provide better quality than government schools, English-medium instruction is a key requirement, and all have pragmatic expectations of the benefits for their children both from private schooling and from English-medium education.

Proteases have received a lot of attention in recent years thanks to their use in a variety of industries, including the food, feed, leather, textile, and pharmaceutical sectors. The soil sample from a cold climate was used in the current experimental study of protease production. In order to identify the protease-producing bacterial species, morphological, physiological, and biochemical analyses were carried out, and the isolated bacterial strain's ability to produce proteases on skim milk agar was assessed. As a result of casein hydrolysis, a zone formed surrounding the bacterial colony suggested a protease-positive strain. To increase the production of protease by isolated Bacillus species, various sources of nitrogen and carbon were examined. Yeast extract (309.32 IU/ml) was the nitrogen source that supported growth the best among the other sources studied, and fructose (269.4 IU/ml) was the carbon source that produced the most protease activity. The fermentation time of 72 h (393.7033 IU/ml) was anticipated to be the best setting for protease synthesis. When 2% (w/v) soybean meal was employed, protease activity was measured at 494.92 IU/ml. The influence of pH was investigated, and it was discovered that 7.5 was the best and optimal pH for Bacillus species to produce 511 IU/ml of protease. Silicon (Si) application can boost plant growth and physiology under heavy metal toxicity. The article aimed to investigate how the foliar and root application of silicon (Si) affects the uptake, translocation, and concentration of cadmium (Cd) for winter wheat (Triticum aestivum L.), and in turn the plant's growth, physiology, and antioxidant enzyme activity, using a complete randomized block design (CRBD)? For this purpose, we used twelve treatments consisting of two levels of Si (0 and 3 mM Na2SiO3) against four levels of Cd (0, 50, 100, and 200 µM CdCl2) with three replications. Results showed that all levels of Cd significantly enhanced membrane permeability measured as malonyldialdehyde (MDA) content, osmotic stress measured as hydrogen peroxide (H2O2) content, and triggered the enzymatic and non-enzymatic antioxidant system, resulting in a severe reduction in the growth of wheat seedlings. Besides, Si application either as root (SiR3) or foliar spray (SiF3) remarkably encountered Cd toxicity by further improving antioxidative enzymes activities, and by hindering Cd uptake, but more significant results were recorded for SiR3. Therefore, it might be established that Si root supplementation inhibited Cd translocation from root to shoot more effectively than foliar application, thereby lowering the potential health risk of Cd pollution. The present study is designed to make a comprehensive understanding of the attitude of the urban consumers and explore the factors involved in dealing with the perishable food of certain kinds. The rise of the middle class stipulates the enhancement of the shopping environment; hence witnessing a substantial increase of the number of the supermarkets in developing countries like Bangladesh will not be surprising. A number of urban supermarkets in recent times start selling perishable foods that were once available in Bangladesh only in flea markets (Kaccha Bazaar). However, due to the lack of proper infrastructure, agro-based perishable food reaches the urban market via a long process of chain mediations and raises concerns about quality and price for both retailers and consumers. Very often the attitudes of consumers regarding perishable foods are unknown and their preferences remain unidentified. This high level of uncertainty regarding the attitude of consumers and the unpopularity regarding overall food quality need to be resolved to ensure the continuity of the business and guarantee the quality of the products. This has made the study of the consumers' attitude towards perishable food, especially relevant for emerging economies like Bangladesh. The data is collected from one hundred (100) consumers, who buy food regularly from both super-shops and flea markets in Dhaka city. This study was designed to evaluate the impact of ethnicity on left ventricular (LV) mass, and relative wall thickness in 527 patients (57% men, mean age 60 +/- 7 years) with mild to moderate high blood pressure. There were 63% Caucasians, 21% African-Americans, and 16% Hispanics. LV mass was indexed according to body surface area, height, and height to the allometric power of 2.7. Relative wall thickness included the 4 widely recognized patterns: normal, concentric remodeling, eccentric hypertrophy, and concentric hypertrophy. LV mass indexed to body surface area was similar among all 3 ethnic groups (Caucasians 117.1 g/m2, African-Americans 119.2 g/m2, Hispanics 122.7 g/m2); however, when indexed to height and height to the power of 2.7, Hispanics had slightly larger masses than the other 2 groups (Hispanics 168.1 and 73.3 g/m2.7 vs Caucasians 159.8 and 64.4 g/m2.7 [p = NS and p < 0.005]; and vs African-Americans 164.8 and 69.2 g/m2.7 [p = NS for both]). Using body surface area, the concentric remodeling was the predominant form of cardiac adaptation in Caucasians (36%) and African-Americans (42%), whereas the concentric hypertrophy pattern was 38% in Hispanics. Using indexing for both height and height to the power of 2.7, the concentric hypertrophy pattern predominated in all 3 ethnic groups (Caucasians 48% and 51%; African-Americans 68% and 66%; Hispanics 59% and 65%). In conclusion, because of the independent impact of weight on high blood pressure, LV mass adjusted to height or to height at the power of 2.7 should be reported in population studies. The concentric hypertrophy pattern--classic LV response to pressure overload conditions--is better represented when LV mass is indexed to height or to height to the allometric power of 2.7 than to body surface area. To combat climate change, a country needs to take part in the development of energy sources and the renovation of its energy infrastructure. Since, green energy production is frequently costly and dangerous, especially in its early stages, capital is one of the barriers to the energy revolution. The aims of the study to analyze the non-linear relationship between energy consumption, financial development, and technology innovation on green economic growth, and environmental pollution indicators including ecological footprint and carbon dioxide emission in the E-7 countries over the period of 1995 to 2022. Using a new panel non-linear autoregressive distribution model (NLPARDL) approach, the results confirm that carbon dioxide emissions, green economic growth, and ecological footprint have a positive and strong long-term correlation with the positive component of the energy use. Conversely, negative shocks are negative and significant with ecological footprint but positive and significant with carbon dioxide emissions and green economic growth. Furthermore, financial development has a positive and substantial relationship with ecological footprint in addition to having a long-term negative and large impact on carbon dioxide emissions and a negative but small impact on green economic growth in a positive shock. Similar to this, financial development negative shock coefficients are significant and negative over the long term when it comes to carbon dioxide emissions and green economic growth, and they are positively significant when it comes to ecological footprint negative component. In the meantime, the long-term positive shock of technology innovation has a negative significant correlation with ecological footprint, ecological footprint a positive and negligible correlation with green economic growth, and a positive and significant correlation with carbon dioxide emission. Similarly, technology innovation long-term negative shock coefficients for carbon dioxide emissions and green economic growth are both negative and significant; on the other hand, technology innovations long-term negative shock coefficients for the negative component of ecological footprint are positively significant. Based on the results, E-7 nations need to invest in projects that utilize energy and technology innovation to reduce environmental degradation and boost green economic growth, such as investing in energy projects and reduce the dependency on fossil fuels. Neuropathy is a disturbance of function or a pathological change in nerves causing poor health and quality of life. A proportion of chronic pain patients in the community suffer persistent neuropathic pain symptoms because current drug therapies may be suboptimal so there is a need for new therapeutic modalities. This study investigated the neuroprotective flavonoid, 6-methoxyflavone (6MF), as a potential therapeutic agent and gabapentin as the standard comparator, against neuropathic models. Thus, neuropathic-like states were induced in Sprague-Dawley rats using sciatic nerve chronic constriction injury (CCI) mononeuropathy and systemic administration of streptozotocin (STZ) to induce polyneuropathy. Subsequent behaviors reflecting allodynia, hyperalgesia, and vulvodynia were assessed and any possible motoric side-effects were evaluated including locomotor activity, as well as rotarod discoordination and gait disruption. 6MF (25-75 mg/kg) antagonized neuropathic-like nociceptive behaviors including static- (pressure) and dynamic- (light brushing) hindpaw allodynia plus heat/cold and pressure hyperalgesia in the CCI and STZ models. 6MF also reduced static and dynamic components of vulvodynia in the STZ induced polyneuropathy model. Additionally, 6MF reversed CCI and STZ suppression of locomotor activity and rotarod discoordination, suggesting a beneficial activity on motor side effects, in contrast to gabapentin. Globally, there is a growing concern about tree mortality due to harsh climates and changes in pest and disease patterns. However, experimental studies on the interactions between biotic and abiotic stresses in plants are relatively scarce. In this study, we investigated the interaction between Fusarium solani and water-stressed Dalbergia sissoo saplings. We postulated that under drought conditions, sissoo plants would become more susceptible to dieback infestation. Five fungi, including Fusarium oxysporum, Curvularia lunata, Cladophialophora carrionii, Alternaria alternaria, and Fusarium solani, were isolated from an old shisham tree showing advanced symptoms of dieback infestation. These fungi were identified based on their ITS sequence homology and spore characteristics. Dieback development was more pronounced in plants experiencing water stress, regardless of their predisposition or whether it occurred simultaneously. Lesions were more noticeable and longer in predisposed saplings (3.8cm), followed by simultaneous (2.4cm) and much smaller lesions in seedlings that were inoculated and well-watered (0.24cm). Progressive browning of the upper leaves, which lowers sapling height in predisposed, simultaneous, and well-watered inoculated saplings to 8.09 inches, 5.93 inches, and 17.42 inches, are typical dieback symptoms. Water stress causes the loss of chlorophyll a, b, and carotenoids, which reduces stomatal conductance, transpiration rate, and photosynthetic activity, leading to poor development and mortality. Similarly, predisposed, simultaneous, and well-watered inoculated seedlings expressed increased activity of CAT (22.57, 18.148, and 9.714 U/mg) and POD (3.0, 4.848, 1.246 U/mg), to reduce the damage caused by elevated levels of H2O2 expression. It is concluded that water stress is the main cause of dieback in shisham saplings that subsequently disposed of infected seedlings to secondary agents such as fungi and insects in the advanced stages of the dieback with prolonged drought stress. The lack of dieback in native populations is attributed to the absence of several ecological stresses, including water stress, extended droughts, waterlogging, and salinity. This study emphasizes the need for additional research into the effects of abiotic factors linked with fungal diseases on the long-term production and management of D. sissoo in Pakistan.

Heavy metals have been recognized as a prominent hazard in today's world, causing pollution in the air environment. Woody tree species can play a significant role in the extraction and remediation of metal pollutants from the air, therefore promoting the air quality index. This study investigated the potential of four species of the Ficus genus (F. benjamina, F. microcarpa, F. religiosa, and F. virens) to remediate varying levels of heavy metal contamination in industrial, residential, and highway areas of Faisalabad City, Pakistan. For this purpose, six heavy metals (cadmium, chromium, copper, lead, zinc, and manganese) were assessed in young leaves (YL) as well as old leaves (OL) of subjected tree species at selected study sites. Eight fully expanded leaves were selected from each tree species: two from each cardinal direction from the shoot of the current year (young leaves, YL), as well as from the shoot of the previous year (old leaves, OL). The results showed that the same genus has different capabilities to accumulate different heavy metals, and the overall trend was in the following order: F. virens > F. religiosa > F. benjamina > F. microcarpa at all study sites. The heavy metal contents in both YL and OL of selected tree species decreased in the order of Manganese (Mn)> Zinc (Zn)> Copper (Cu) > Chromium (Cr) > Lead (Pb) > Cadmium (Cd) at all study sites. The metal accumulation index (MAI) values ranged between 2.14-5.42 for F. benjamina, 2.09-3.89 for F. microcarpa, 3.61-7.01 for F. religiosa and 4.77-6.48 for F. virens across all study sites. Among the studied  tree species, it has been determined that F. virens and F. religiosa are well-suited for urban areas with significant heavy metal contamination and can be strategically planted in barrier areas to effectively combat atmospheric pollution.

In this study, eleven novel acyl hydrazides derivative of polyhydroquinoline were synthesized, characterized and screened for their in vitro anti-diabetic and anti-glycating activities. Seven compounds 2a, 2d, 2i, 2 h, 2j, 2f, and 2 g exhibited notable α-amylase inhibitory activity having IC 50 values from 3.51 ± 2.13 to 11.92 ± 2.30 µM. Similarly, six compounds 2d, 2f, 2 h, 2i, 2j, and 2 g displayed potent α-glucosidase inhibitory activity compared to the standard acarbose. Moreover, eight derivatives 2d, 2 g, 2f, 2j, 2a, 2i, 2 g, and 2e showed excellent anti-glycating activity with IC 50 values from 6.91 ± 2.66 to 15.80 ± 1.87 µM when compared them with the standard rutin (IC 50  = 22.5 ± 0.90 µM). Molecular docking was carried out to predict the binding modes of all the compounds with α-amylase and α-glucosidase. The docking analysis revealed that most of the compounds established strong interactions with α-amylase and α-glucosidase. All compounds fitted well into the binding pockets of α-amylase and α-glucosidase. Among all compounds 2a and 2f were most potent based on docking score -8.2515 and -7.3949 against α-amylase and α-glucosidase respectively. Belief and plausibility functions based on evidence theory (ET) have been widely used in managing uncertainty. Various generalizations of ET to fuzzy sets (FSs) have been reported in the literature, but no generalization of ET to q-rung orthopair fuzzy sets (q-ROFSs) has been made yet. Therefore, this paper proposes a novel, simple, and intuitive approach to distance and similarity measures for q-ROFSs based on belief and plausibility functions within the framework of ET. This research addresses a significant research gap by introducing a comprehensive framework for handling uncertainty in q-ROFSs using ET. Furthermore, it acknowledges the limitations inherent in the current state of research, notably the absence of generalizations of ET to q-ROFSs and the challenges in extending belief and plausibility measures to certain aggregation operators and other generalizations including Hesitant fuzzy sets, Bipolar fuzzy sets, Fuzzy soft sets etc. Our contribution lies in the proposal of a novel approach to distance and similarity measures for q-ROFSs under ET, utilizing Orthopairian belief and plausibility intervals (OBPIs). We establish new similarity measures within the generalized ET framework and demonstrate the reasonability of our method through useful numerical examples. Additionally, we construct Orthopairian belief and plausibility GRA (OBP-GRA) for managing daily life complex issues, particularly in multicriteria decision-making scenarios. Thirteen novel hydrazone-Schiff bases (3-15) of fexofenadine were succesfully synthesized, structurally deduced and finally assessed their capability to inhibit urease enzyme (in vitro). In the series, six compounds 12 (IC 50 =10.19±0.16 μM), 11 (IC 50 =15.05±1.11 μM), 10 (IC 50 =17.01±1.23 μM), 9 (IC 50 =17.22±0.81 μM), 13 (IC 50 =19.31±0.18 μM), and 14 (IC 50 =19.62±0.21 μM) displayed strong inhibitory action better than the standard thiourea (IC 50 =21.14±0.24 μM), while the remaining compounds displayed significant to less inhibition. LUMO and HOMO showed the transferring of charges from molecules to biological transfer and MEP map showed the chemically reactive zone appropriate for drug action are calculated using DFT. Heavy metal ions pose significant risks to human health, pelagic, and several other life forms due to perniciousness, tendency to accumulate, and resistance to biodegradation. Waste bio-materials extend a budding alternative as low-cost adsorbent to address the removal of noxious pollutants from wastewater on account of being cost-effective and exhibiting exceptional adsorption capacities. The current exploration was accomplished to gauge the performance of raw and modified human hair concerning lead scavenging in a down-flow fixed bed column. The appraisal of column performance under varying operational parameters encompassing bed height (15-45 cm), influent metal ion concentration (60-140 mg L -1 ), and a solution flow rate (20-40 mL min -1 ) was performed by breakthrough curve analysis. The consequences acquired were evaluated using the Yoon Nelson, Thomas, Adam-Bohart, and Bed Depth Service Time (BDST) model. Among these employed models, Bed Depth Service Time (BDST) and Thomas models exhibited the highest R-squared value compared to the Yoon Nelson and Adam-Bohart's model for most cases. Pollution in the environment is today the biggest issue facing the globe and the main factor in the development of many fatal diseases. The main objective of the study to investigate green investments, economic growth and financial development on environmental pollution in the G-7 countries. This study used annual penal data from 1997 to 2021. The panel NARDL (Non-linear autoregressive distributed lag) results affirm that the positive change of green investment and negative shock in green investment have a significant and positive association with environment pollution in G-7 nations. Our findings provide more evidence for the long-term asymmetry between financial development and environmental performance. However, the findings confirm that a positive modification in financial development has a positive and significant effect on environment pollution. Whereas negative shock in financial development is negative and insignificant relationship with environment pollution. Moreover, the outcomes of the study reveal that both positive shock in gross domestic product growth and negative shock of economic growth have a significant and positive link with environment pollution in G-7 countries. According to the findings, by lowering carbon dioxide emissions, green investments reduced environmental pollution in the G-7 nations over the long and short term. Moreover, it is an innovative research effort that provides light on the connection between green investments, financial development, and the environment while making mention to the EKC in G-7 countries. Climate change and cadmium (Cd) contamination pose severe threats to rice production and food security. Biochar (BC) has emerged as a promising soil amendment for mitigating these challenges. To investigate the BC effects on paddy soil upon GHG emissions, Cd bioavailability, and its accumulation, a meta-analysis of published data from 2000 to 2023 was performed. Data Manager 5.3 and GetData plot Digitizer software were used to obtain and process the data for selected parameters. Our results showed a significant increase of 18% in soil pH with sewage sludge BC application, while 9% increase in soil organic carbon (SOC) using bamboo chips BC. There was a significant reduction in soil bulk density (8%), but no significant effects were observed for soil porosity, except for wheat straw BC which reduced the soil porosity by 6%. Sewage sludge and bamboo chips BC significantly reduced carbon dioxide (CO 2 ) by 7-8% while municipal biowaste reduced methane (CH 4 ) emissions by 2%. In the case of heavy metals, sunflower seedshells-derived materials and rice husk BC significantly reduced the bioavailable Cd in paddy soils by 24% and 12%, respectively. Cd uptake by rice roots was lowered considerably by the addition of kitchen waste (22%), peanut hulls (21%), and corn cob (15%) based BC. Similarly, cotton sticks, kitchen waste, peanut hulls, and rice husk BC restricted Cd translocation from rice roots to shoots by 22%, 27%, 20%, and 19%, respectively, while sawdust and rice husk-based BC were effective for reducing Cd accumulation in rice grains by 25% and 13%. Regarding rice yield, cotton sticks-based BC significantly increased the yield by 37% in Cd-contaminated paddy soil. Lead (Pb) is a highly toxic contaminant that is ubiquitously present in the ecosystem and poses severe environmental issues, including hazards to soil-plant systems. This review focuses on the uptake, accumulation, and translocation of Pb metallic ions and their toxicological effects on plant morpho-physiological and biochemical attributes. We highlight that the uptake of Pb metal is controlled by cation exchange capacity, pH, size of soil particles, root nature, and other physio-chemical limitations. Pb toxicity obstructs seed germination, root/shoot length, plant growth, and final crop-yield. Pb disrupts the nutrient uptake through roots, alters plasma membrane permeability, and disturbs chloroplast ultrastructure that triggers changes in respiration as well as transpiration activities, creates the reactive oxygen species (ROS), and activates some enzymatic and non-enzymatic antioxidants. Pb also impairs photosynthesis, disrupts water balance and mineral nutrients, changes hormonal status, and alters membrane structure and permeability. This review provides consolidated information concentrating on the current studies associated with Pb-induced oxidative stress and toxic conditions in various plants, highlighting the roles of different antioxidants in plants mitigating Pb-stress. Additionally, we discussed detoxification and tolerance responses in plants by regulating different gene expressions, protein, and glutathione metabolisms to resist Pb-induced phytotoxicity. Peritoneal fibrosis together with increased capillaries is the primary cause of peritoneal dialysis failure. Mesothelial cell loss is an initiating event for peritoneal fibrosis. We find that the elevated glucose concentrations in peritoneal dialysate drive mesothelial cell pyroptosis in a manner dependent on caspase-3 and Gasdermin E, driving downstream inflammatory responses, including the activation of macrophages. Moreover, pyroptosis is associated with elevated vascular endothelial growth factor A and C, two key factors in vascular angiogenesis and lymphatic vessel formation. GSDME deficiency mice are protected from high glucose induced peritoneal fibrosis and ultrafiltration failure. Application of melatonin abrogates mesothelial cell pyroptosis through a MT1R-mediated action, and successfully reduces peritoneal fibrosis and angiogenesis in an animal model while preserving dialysis efficacy. Mechanistically, melatonin treatment maintains mitochondrial integrity in mesothelial cells, meanwhile activating mTOR signaling through an increase in the glycolysis product dihydroxyacetone phosphate. These effects together with quenching free radicals by melatonin help mesothelial cells maintain a relatively stable internal environment in the face of high-glucose stress. Aluminum (Al), a non-essential metal for plant growth, exerts significant phytotoxic effects, particularly on root growth. Anthropogenic activities would intensify Al's toxic effects by releasing Al 3+ into the soil solution, especially in acidic soils with a pH lower than 5.5 and rich mineral content. The severity of Al-induced phytotoxicity varies based on factors such as Al concentration, ionic form, plant species, and growth stages. Al toxicity leads to inhibited root and shoot growth, reduced plant biomass, disrupted water uptake causing nutritional imbalance, and adverse alterations in physiological, biochemical, and molecular processes. These effects collectively lead to diminished plant yield and quality, along with reduced soil fertility. Plants employ various mechanisms to counter Al toxicity under stress conditions, including sequestering Al in vacuoles, exuding organic acids (OAs) like citrate, oxalate, and malate from root tip cells to form Al-complexes, activating antioxidative enzymes, and overexpressing Al-stress regulatory genes. Recent advancements focus on enhancing the exudation of OAs to prevent Al from entering the plant, and developing Al-tolerant varieties. Gene transporter families, such as ATP-Binding Cassette (ABC), Aluminum-activated Malate Transporter (ALMT), Natural resistance-associated macrophage protein (Nramp), Multidrug and Toxic compounds Extrusion (MATE), and aquaporin, play a crucial role in regulating Al toxicity. This comprehensive review examined recent progress in understanding the cytotoxic impact of Al on plants at the cellular and molecular levels. Nanotechnology offers many potential solutions for sustainable agroecosystem, including improvement in nutrient use efficiency, efficacy of pest management, and minimizing the adverse environmental effects of agricultural production. Herein, we first highlighted the integrated application of nanotechnology and precision agriculture for sustainable productivity. Application of nanoparticle mediated material and advanced biosensors in precision agriculture is only possible by nanochips or nanosensors. Nanosensors offers the measurement of various stresses, soil quality parameters and detection of heavy metals along with the enhanced data collection, enabling precise decision-making and resource management in agricultural systems. Nanoencapsulation of conventional chemical fertilizers (known as nanofertilizers), and pesticides (known as nanopesticides) helps in sustained and slow release of chemicals to soils and results in precise dosage to plants. Further, nano-based disease detection kits are popular tools for early and speedy detection of viral diseases. Many other innovative approaches including biosynthesized nanoparticles have been evaluated and proposed at various scales, but in fact there are some barriers for practical application of nanotechnology in soil-plant system, including safety and regulatory concerns, efficient delivery at field levels, and consumer acceptance. King grass has been recognized as a potential phytoremediation plant species due to its high biomass and resistance to heavy metals (HMs). However, the possible impacts of cadmium (Cd) contamination on rhizocompartments' microbial activities in association with king grass have not been extensively explored. The utilization of 16S rRNA gene and ITS sequencing was carried out to examine alterations in the bacterial and fungal communities in the rhizosphere and rhizoplane of king grass in response to low and high Cd stress. Results demonstrated that both bacterial and fungal communities' diversity and richness were negatively impacted by Cd stress, regardless of its concentration. However, evenness did not exhibit any significant response to either of the concentrations. Additionally, nonmetric multidimensional scaling (NMDS) ordination demonstrated a significant difference (p < 0.001) in microbial communities under different treatments. The abundance of bacterial taxa such as Steroibacter, Nitrospira, Pseudoxanthomonas, Cellvirio, Phenylobacterium, Mycobacterium, Pirellula and Aquicella was adversely affected under Cd stress while Flavobacterium, Gemmata, Thiobacillus and Gemmatimonas showed no prominent response, indicating their resistance to Cd stress. Like that, certain fungal taxa for instance, Cladosporium, Cercophora, Acremonium, Mortierella, Aspergillus, Penicillium, Glomus and Sebacina were also highly reduced by low and high Cd stress. In contrast, Fusarium, Thanatephorus, Botrytis and Curvularia did not show any response to Cd stress. The anti-inflammatory and analgesic drugs currently used are associated with several adverse effects and found to be highly unsafe for long-term use. Currently, nineteen novel bis-Schiff base derivatives (1-19) of flurbiprofen have been designed, prepared and assessed for in-vivo analgesic, anti-inflammatory and in vivo acute toxicity evaluation. The structures of the acquired compounds were deduced through modern spectroscopic techniques including HR-ESI-MS, 13 C-, and 1 H NMR. Amongst the series, compounds 7, 9, and 10 attributed potent activities with 93.89, 92.50, and 90.47% decreased edema, respectively compared to flurbiprofen (90.01%), however, compounds 11 and 15 exhibited significant activity of 90.00% decrease. Out of them, fourteen compounds (1-6, 8, 12-14, and 16-19) displayed good activity in the range of 68.96-86.95%. In case of an analgesic study, all the derivatives significantly (p 0.001) increased the pain threshold time particularly compound 7 had the best analgesic effect (24 ± 2.08 s) in comparison with flurbiprofen (21.66 ± 2.02 s) using hot plate test. Similarly, in the acetic acid-induced writhing test, compound 7 determined a potent inhibitory effect (60.47 %) close to flurbiprofen (59.28%). All the synthesized derivatives were found safe up to the dose of 30 mg/kg, in acute toxicity study.
