The article focuses on the diversity and distribution of snake species in and around Solapur district, Maharashtra, India based on data collected over two years (2018-2020). It identifies 20 species of snakes, including venomous, semi-venomous, and non-venomous types from six families. Key findings include the occurrence of both common and rare species, such as the Albino Spectacled Cobra in residential areas.

The conventional field technique of castration of bull's using Burdizzo's castrator induces immense pain responses including vocalization, defecation, struggling, violent kicking, restlessness, elevation in vital clinical parameters and post-operative stress. In this study 10 bulls of different age group were castrated, out of them five were castrated under effect of combination of Xylazine-Butorphanol-Ketamine (XBK) and another five bulls were castrated without use of combination of XBK, the analysis of vital parameters and pain symptoms was carried out to ascertain the efficacy of this combination. No significant changes in heart rate, respiration rate and rectal temperature, no pain signs were observed just after crushing of chord by Burdizzo's clamp under effect of combination of XBK while significant increase in heart rate, respiration rate and rectal temperature observed along with vocalization, defecation, struggling in all five bulls who were castrated without use of XBK. Intravenous Ketamine recumbent stun technique using combination of Xylazine hydrochloride, Butorphanol Tartrate and Ketamine hydrochloride, is found to produce commendable analgesia and anaesthesia which help in alleviation of pain signs during castration of bulls. The paper reports a case of spindle cell tumor in a dog of nondescript breed and its surgical management. Histopathological evaluation confirmed it as spindle cell tumour which was characterised by cellular tumour composed of elongated spindle cells in groups, sheets and nests and have mostly vesicular nuclei. Studies were taken up on 25 mm thick samples ofTectona grandisto understand its drying behavior under vacuum press drying. Five different vacuum level–temperature combinations were used in the study to observe the moisture reductions achieved. The results were analyzed to understand which combination gave maximum moisture content reduction from two moisture content ranges: one above and the other below the fiber saturation point of the species. It was found that the samples could be dried to less than 13% moisture content levels from about 25% moisture content levels effectively in just two runs approximately lasting 7 h each. More interestingly, the moisture removal by this method was found more effective when samples were seasoned from initial moisture content levels below the fiber saturation point. Optical filtering is an indispensable part of fluorescence microscopy for selectively highlighting molecules labeled with a specific fluorophore and suppressing background noise. However, the utilization of optical filtering sets increases the complexity, size, and cost of microscopic systems, making them less suitable for multifluorescence channel, high-speed imaging. Here, we present filter-free fluorescence microscopic imaging enabled with deep learning-based digital spectral filtering. This approach allows for automatic fluorescence channel selection after image acquisition and accurate prediction of fluorescence by computing color changes due to spectral shifts with the presence of excitation scattering. Fluorescence prediction for cells and tissues labeled with various fluorophores was demonstrated under different magnification powers. The technique offers accurate identification of labeling with robust sensitivity and specificity, achieving consistent results with the reference standard. Beyond fluorescence microscopy, the deep learning-enabled spectral filtering strategy has the potential to drive the development of other biomedical applications, including cytometry and endoscopy.

Acoustic manipulation has emerged as a valuable tool for precision controls and dynamic programming of cells and particles. However, conventional acoustic manipulation approaches lack the finesse necessary to form intricate, configurable, continuous, and 3D patterning of particles. Here, this study reports acoustography by Beam Engineering and Acoustic Control Node (BEACON), which delivers intricate, configurable patterns by guiding particles along custom paths with independent phase modulation. Leveraging analytical methods of orbital angular momentum beam via iterative Wirtinger hologram algorithm, this study accomplish acoustography by facilitating orbital angular momentum traps, enabling continuous 2D and 3D acoustic manipulation of microparticles in any desired geometry, with phase modulation independent of intensity. Utilizing on-chip acoustography, the BEACON platform markedly increases the space-bandwidth product to 31 000 while attaining an enhanced resolution with a pixel size of ≈25 µm, surpassing the typical resolution of over 200 µm in previous holographic particle manipulation methods. The capabilities of BEACON are demonstrated in creating intricate triple helical tracing structures using microdroplets (20 µm in diameter) and those carrying DNA to validate the effectiveness of the acoustography and phase control methods. Laboratory automation technologies have revolutionized biomedical research. However, the availability of automation solutions at the single-cell level remains scarce, primarily owing to the inherent challenges of handling cells with such small dimensions in a precise, biocompatible manner. Here, we present a single-cell-level laboratory automation solution that configures various experiments onto standardized, microscale test-tube matrices via our precise ultrasonic liquid sample ejection technology, known as PULSE. PULSE enables the transformation of titer plates into microdroplet arrays by printing nanodrops and single cells acoustically in a programmable, scalable, and biocompatible manner. Unlike pipetting robots, PULSE enables researchers to conduct biological experiments using single cells as anchoring points (e.g., 1 cell vs. 1000 cells per "tube"), achieving higher resolution and potentially more relevant data for modeling and downstream analyses. We demonstrate the ability of PULSE to perform biofabrication, precision gating, and deterministic array barcoding via preallocated droplet-addressable primers. Single cells can be gently printed at a speed range of 5-20 cell⋅s -1 with an accuracy of 90.5-97.7%, which can then adhere to the substrate and grow for up to 72 h while preserving cell integrity. In the deterministic barcoding experiment, 95.6% barcoding accuracy and 2.7% barcode hopping were observed by comparing the phenotypic data with known genotypic data from two types of single cells. Contactless microscale tweezers are highly effective tools for manipulating, patterning, and assembling bioparticles. However, current tweezers are limited in their ability to comprehensively manipulate bioparticles, providing only partial control over the six fundamental motions (three translational and three rotational motions). This study presents a joint subarray acoustic tweezers platform that leverages acoustic radiation force and viscous torque to control the six fundamental motions of single bioparticles. This breakthrough is significant as our manipulation mechanism allows for controlling the three translational and three rotational motions of single cells, as well as enabling complex manipulation that combines controlled translational and rotational motions. Moreover, our tweezers can gradually increase the load on an acoustically trapped cell to achieve controllable cell deformation critical for characterizing cell mechanical properties. Furthermore, our platform allows for three-dimensional (3D) imaging of bioparticles without using complex confocal microscopy by rotating bioparticles with acoustic tweezers and taking images of each orientation using a standard microscope. Therapeutic apheresis aims to selectively remove pathogenic substances, such as antibodies that trigger various symptoms and diseases. Unfortunately, current apheresis devices cannot handle small blood volumes in infants or small animals, hindering the testing of animal model advancements. This limitation restricts our ability to provide treatment options for particularly susceptible infants and children with limited therapeutic alternatives. Here, we report our solution to these challenges through an acoustofluidic-based therapeutic apheresis system designed for processing small blood volumes. Our design integrates an acoustofluidic device with a fluidic stabilizer array on a chip, separating blood components from minimal extracorporeal volumes. We carried out plasma apheresis in mouse models, each with a blood volume of just 280 μL. Additionally, we achieved successful plasmapheresis in a sensitized mouse, significantly lowering preformed donor-specific antibodies and enabling desensitization in a transplantation model. Separating plasma from whole blood is an important sample processing technique required for fundamental biomedical research, medical diagnostics, and therapeutic applications. Traditional protocols for plasma isolation require multiple centrifugation steps or multiunit microfluidic processing to sequentially remove large red blood cells (RBCs) and white blood cells (WBCs), followed by the removal of small platelets. Here, we present an acoustofluidic platform capable of efficiently removing RBCs, WBCs, and platelets from whole blood in a single step. By leveraging differences in the acoustic impedances of fluids, our device generates significantly greater forces on suspended particles than conventional microfluidic approaches, enabling the removal of both large blood cells and smaller platelets in a single unit. As a result, undiluted human whole blood can be processed by our device to remove both blood cells and platelets (>90%) at low voltages (25 Vpp). The ability to successfully remove blood cells and platelets from plasma without altering the properties of the proteins and antibodies present creates numerous potential applications for our platform in biomedical research, as well as plasma-based diagnostics and therapeutics. Large-field nanoscale fluorescence imaging is invaluable for many applications, such as imaging subcellular structures, visualizing protein interactions, and high-resolution tissue imaging. Unfortunately, conventional fluorescence microscopy requires a trade-off between resolution and field of view due to the nature of the optics used to form the image. To overcome this barrier, we developed an acoustofluidic scanning fluorescence nanoscope that simultaneously achieves superior resolution, a large field of view, and strong fluorescent signals. The acoustofluidic scanning fluorescence nanoscope utilizes the superresolution capabilities of microspheres that are controlled by a programmable acoustofluidic device for rapid fluorescence enhancement and imaging. The acoustofluidic scanning fluorescence nanoscope resolves structures that cannot be resolved with conventional fluorescence microscopes with the same objective lens and enhances the fluorescent signal by a factor of ~5 without altering the field of view of the image. Techniques to resolve images beyond the diffraction limit of light with a large field of view (FOV) are necessary to foster progress in various fields such as cell and molecular biology, biophysics, and nanotechnology, where nanoscale resolution is crucial for understanding the intricate details of large-scale molecular interactions. Although several means of achieving super-resolutions exist, they are often hindered by factors such as high costs, significant complexity, lengthy processing times, and the classical tradeoff between image resolution and FOV. Microsphere-based super-resolution imaging has emerged as a promising approach to address these limitations. In this review, we delve into the theoretical underpinnings of microsphere-based imaging and the associated photonic nanojet. This is followed by a comprehensive exploration of various microsphere-based imaging techniques, encompassing static imaging, mechanical scanning, optical scanning, and acoustofluidic scanning methodologies. This review concludes with a forward-looking perspective on the potential applications and future scientific directions of this innovative technology.

The study of molecular mechanisms at the single-cell level holds immense potential for enhancing immunotherapy and understanding neuroinflammation and neurodegenerative diseases by identifying previously concealed pathways within a diverse range of paired cells. However, existing single-cell pairing platforms have limitations in low pairing efficiency, complex manual operation procedures, and single-use functionality. Here, we report a multiparametric cellular immunity analysis by a modular acoustofluidic platform: CIAMAP. This platform enables users to efficiently sort and collect effector-target (i.e., NK92-K562) cell pairs and monitor the real-time dynamics of immunological response formation. Furthermore, we conducted transcriptional and protein expression analyses to evaluate the pathways that mediate effector cytotoxicity toward target cells, as well as the synergistic effect of doxorubicin on the cellular immune response. Our CIAMAP can provide promising building blocks for high-throughput quantitative single-cell level coculture to understand intercellular communication while also empowering immunotherapy by precision analysis of immunological synapses.

The intrinsic biophysical properties of cells, such as mechanical, acoustic, and electrical properties, are valuable indicators of a cell's function and state. However, traditional single-cell biophysical characterization methods are hindered by limited measurable properties, time-consuming procedures, and complex system setups. This study presents acousto-dielectric tweezers that leverage the balance between controllable acoustophoretic and dielectrophoretic forces applied on cells through surface acoustic waves and alternating current electric fields, respectively. Particularly, the balanced acoustophoretic and dielectrophoretic forces can trap cells at equilibrium positions independent of the cell size to differentiate between various cell-intrinsic mechanical, acoustic, and electrical properties. Experimental results show our mechanism has the potential for applications in single-cell analysis, size-insensitive cell separation, and cell phenotyping, which are all primarily based on cells' intrinsic biophysical properties. Our results also show the measured equilibrium position of a cell can inversely determine multiple biophysical properties, including membrane capacitance, cytoplasm conductivity, and acoustic contrast factor. Background: Treatment and prevention of wound infection continues to be a challenging issue in clinical settings of Nepal especially in the context of globally growing problem of antimicrobial resistance. Study on opportunistic pathogens and sensitivity to commonly prescribed local antimicrobial agents are cardinal to reduce the disease burden of wound infections. The aim of this study was to determine the prevalence and antimicrobial susceptibility pattern of methicillin resistant Staphylococcus aureus (MRSA) and extended spectrum β-lactamase (ESBL) producing bacteria from wound infections of patients at a tertiary care hospital in Nepal., Methods: Pus specimens were processed using standard microbiological procedures. Antimicrobial susceptibility test was performed following the modified Kirby Bauer disc diffusion technique. Clinical information of patients was obtained from preformed questionnaire and hospital record., Results: One hundred eighty two pus specimens from wounds of different body parts: leg, hand, backside, abdominal part, foot, breast and chest, head and neck region were collected and analyzed; 113 bacterial isolates were isolated showing the overall bacterial growth rate of 62%, where the highest rate was among patients of ≤10 years age group (82.1%). A higher rate (68.5%) of bacterial isolates were from inpatients ( p  < 0.05). Among 116 bacterial isolates, Staphylococcus aureus was the most predominant bacteria (56.9%) followed by Escherichia coli (8.6%), coagulase negative staphylococci (7.8%), Acinetobacter spp. (5.2%), Klebsiella pneumoniae (5.2%), Pseudomonas aeruginosa (4.3%), Enterococcus spp. (4.3%), Citrobacter freundii (2.6%), Proteus vulgaris (1.6%) and P. mirabilis (0.9%). Both Gram positive (73.3%) and negative (78.8%) isolates showed high frequency of sensitive to gentamycin., Conclusion: Among S. aureus isolates , 60.6% were MRSA strains, whereas 40% of K. pneumoniae and 33.3% of C. freundii were ESBL producing bacteria followed by E. coli (25%). Protective role of diet supplements (Spirulina, tamarind fruit pulp and their combination) on a freshwater fish G. affinis exposed at sublethal concentration of fluoride (F-) (10 ppm), Al(+3) (3 ppm) and aluminum fluoride (AlF3) (35.4 ppm) in the microcosms (15 L sized) for 30-60 days in winter (90 days in summer) has been reported. Toxic effects of chemicals were manifested as higher fish mortality (4-50%) and acid (approximately -30%) and alkaline phosphatase (25-50%) contents, but reduction in RBC counts (5-55%) and protein content (approximately -29%) compared with controls. Alterations in values of these parameters were found maximum in aluminum exposed fish suggesting it as the most toxic among the tested chemicals. Diet supplements reduced toxicity of tested chemicals, especially when Spirulina and tamarind were given together.
