Abstract

Dark matter elastic scattering off nuclei can result in the excitation and ionization of the recoiling atom through the so-called Migdal effect. The energy deposition from the ionization electron adds to the energy deposited by the recoiling nuclear system and allows for the detection of interactions of sub-GeV/c2 mass dark matter. We present new constraints for sub-GeV/c2 dark matter using the dual-phase liquid argon time projection chamber of the DarkSide-50 experiment with an exposure of (12 306±184) kg d. The analysis is based on the ionization signal alone and significantly enhances the sensitivity of DarkSide-50, enabling sensitivity to dark matter with masses down to 40 MeV/c2. Furthermore, it sets the most stringent upper limit on the spin independent dark matter nucleon cross section for masses below 3.6 GeV/c2.

Abstract

We report on the search for dark matter WIMPs in the mass range below 10 GeV/c$^2$, from the analysis of the entire dataset acquired with a low-radioactivity argon target by the DarkSide-50 experiment at LNGS. The new analysis benefits from more accurate calibration of the detector response, improved background model, and better determination of systematic uncertainties, allowing us to accurately model the background rate and spectra down to 0.06 keV$_{er}$. A 90% C.L. exclusion limit for the spin-independent cross section of 3 GeV/c$^2$ mass WIMP on nucleons is set at 6$\times$10$^{-43}$ cm$^2$, about a factor 10 better than the previous DarkSide-50 limit. This analysis extends the exclusion region for spin-independent dark matter interactions below the current experimental constraints in the $[1.2, 3.6]$ GeV/c$^2$ WIMP mass range., 11 pages, 12 figures

Abstract

We present a search for dark matter particles with sub-GeV/$c^2$ masses whose interactions have final state electrons using the DarkSide-50 experiment's (12306 $\pm$ 184) kg d low-radioactivity liquid argon exposure. By analyzing the ionization signals, we exclude new parameter space for the dark matter-electron cross section $\bar{\sigma}_e$, the axioelectric coupling constant $g_{Ae}$, and the dark photon kinetic mixing parameter $\kappa$. We also set the first dark matter direct-detection constraints on the mixing angle $\left|U_{e4}\right|^2$ for keV sterile neutrinos., Comment: 6 pages, 2 figures

Abstract

The role of seasonality is indisputable in climate and ecosystem dynamics. Seasonal temperature and precipitation variability are of vital importance for the availability of food, water, shelter, migration routes, and raw materials. Thus, understanding past climatic and environmental changes at seasonal scale is equally important for unearthing the history and for predicting the future of human societies under global warming scenarios. Alas, in palaeoenvironmental research, the term ‘seasonality change’ is often used liberally without scrutiny or explanation as to which seasonal parameter has changed and how.\ud \ud Here we provide fundamentals of climate seasonality and break it down into external (insolation changes) and internal (atmospheric CO2 concentration) forcing, and regional and local and modulating factors (continentality, altitude, large-scale atmospheric circulation patterns). Further, we present a brief overview of the archives with potentially annual/seasonal resolution (historical and instrumental records, marine invertebrate growth increments, stalagmites, tree rings, lake sediments, permafrost, cave ice, and ice cores) and discuss archive-specific challenges and opportunities, and how these limit or foster the use of specific archives in archaeological research.\ud \ud Next, we address the need for adequate data-quality checks, involving both archive-specific nature (e.g., limited sampling resolution or seasonal sampling bias) and analytical uncertainties. To this end, we present a broad spectrum of carefully selected statistical methods which can be applied to analyze annually- and seasonally-resolved time series. We close the manuscript by proposing a framework for transparent communication of seasonality-related research across different communities.

Abstract

International audience; We recently showed that the DFT+U approach with a linear-response U yields adiabatic energy differences biased toward high spin [Mariano et al. J. Chem. Theory Comput. 2020, 16, 6755–6762]. Such bias is removed here by employing a density-corrected DFT approach where the PBE functional is evaluated on the Hubbard U-corrected density. The adiabatic energy differences of six Fe(II) molecular complexes computed using this approach, named PBE[U] here, are in excellent agreement with coupled cluster-corrected CASPT2 values for both weak- and strong-field ligands resulting in a mean absolute error (MAE) of 0.44 eV, smaller than that of the recently proposed Hartree–Fock density-corrected DFT (1.22 eV) and any other tested functional, including the best performer TPSSh (0.49 eV). We take advantage of the computational efficiency of this approach and compute the adiabatic energy differences of five molecular crystals using PBE[U] with periodic boundary conditions. The results show, again, an excellent agreement (MAE = 0.07 eV) with experimentally extracted values and a superior performance compared with the best performers M06-L (MAE = 0.08 eV) and TPSSh (MAE = 0.31 eV) computed on molecular fragments.

Abstract

International audience; Lead-halide perovskites are the promising materials in optoelectronic devices for their unique properties including direct band gap, strong light absorption, high carrier mobility and low fabrication cost. Here, by using ab inito molecular dynamics and electronic structure calculations, we report a systematic study on the broadening density of states (DOS) deep in the valence bands that has been experimentally observed but absent in static calculations. We quantify the broadening DOS reduction from cubic phase to lower temperature tetragonal and orthorhombic phases and attribute observed effects to the molecular vibrations and the anharmonicity of iodine atoms motion. Specifically, the MA cations’ vibrations are strongly linked to the moderate C−N stretch and CH3 bend, as well as the strong CH3 and NH+ 3 stretches. These results present a theoretical perspective on the structural dynamics in lead halide perovskites which may be valuable for future studies toward desired functionalities in perovskite-based optoelectronic devices.

Abstract

International audience; A novel gas sensing mechanism exploiting lanthanide luminescence modulation upon NO2 adsorption is demonstrated here. Two isostructural lanthanide-based metal–organic frameworks (MOFs) are used, including an amino group as the sensitive recognition center for NO2 molecules. The transfer of energy from the organic ligands to Ln is strongly dependent on the presence of NO2, resulting in an unprecedented photoluminescent sensing scheme. Thereby, NO2 exposition triggers either a reversible enhancement or a decrease in the luminescence intensity, depending on the lanthanide ion (Eu or Tb). Our experimental studies combined with density functional theory and complete active space self-consistent field calculations provide an understanding of the nature and effects of NO2 interactions within the MOFs and the signal transduction mechanism.

Abstract

We here present the first detailed study of the specimen KNM-RU 18340 from Rusinga Island (Kenya), the only known complete early Miocene chameleon skull, using micro-CT. This specimen represents one of the oldest chameleon fossils ever recovered. For the first time, the skull bone internal surfaces, their sutures, and elements contained inside the rocky matrix are observed. Our morphological comparisons and phylogenetic analyses place this specimen confidently in the genus Calumma and a new species, Calumma benovskyi sp. nov., is erected for it. Since all species of this genus are endemic to Madagascar, this fossil uniquely demonstrates the existence of Calumma on continental Africa in the past. Our results challenge the long-held view that chameleons originated on Madagascar and dispersed over water to Africa, and provide a strong evidence of an African origin for some Malagasy lineages. The Oligocene–early Miocene dispersal to Madagascar, using oceanic currents that favoured eastward dispersal at that time, is a highly supported scenario matching the suggested dispersal of lemurs to this island. This is consistent with a previously suggested hypothesis based on molecular data.

Abstract

International audience; Adolescent Idiopathic Scoliosis (AIS) is a spinal deformity that affects approximately 3 percent of human adolescents. Although the etiology and molecular basis of AIS is unclear, several genes such as POC5 have been identified as possible causes of the condition. In order to understand the role of POC5 in the pathogenesis of AIS, we investigated the subcellular localization of POC5 in cilia of cells over-expressing either the wild type (wt) or an AIS-related POC5 variant POC5A429V. Mutation of POC5 was found to alter its subcellular localization and to induce ciliary retraction. Furthermore, we observed an impaired cell-cycle progression with the accumulation of cells in the S-phase in cells expressing POC5A429V. Using immunoprecipitation coupled to mass spectrometry, we identified specific protein interaction partners of POC5, most of which were components of cilia and cytoskeleton. Several of these interactions were altered upon mutation of POC5. Altogether, our results demonstrate major cellular alterations, disturbances in centrosome protein interactions and cilia retraction in cells expressing an AIS-related POC5 mutation. Our study suggests that defects in centrosomes and cilia may underlie AIS pathogenesis.

Abstract

International audience; The bacterial adenylate cyclase two-hybrid system (BACTH) is a genetic approach used to test protein interactions in vivo in E. coli. This system takes advantage of the two catalytic domains of Bordetella pertussis adenylate cyclase (CyaA) toxin, which can be fused separately to proteins of interest. If the proteins of interest interact, then the adenylate cyclase domains will be brought in close proximity to each other, reconstituting cyclic AMP (cAMP) production. Interacting proteins can be both qualitatively and quantitatively assessed by the expression of chromosomal genes of the E. coli lac or mal operon, which are positively regulated by cAMP production. Because cAMP is diffusible, the proteins of interest do not need to interact near the transcriptional machinery. Consequently, both cytosolic and membrane protein-protein interactions can be tested. The BACTH system has recently been modified to be compatible with Gateway® recombinational cloning, BACTHGW. This chapter explains the principle of the BACTH, its Gateway® modified system, and details of the general procedure.

Abstract

The physiological role and the regulation of ADGRG7 are not yet elucidated. The functional involvement of this receptor was linked with different physiological process such as reduced body weight, gastrointestinal function and recently, a gene variant in ADGRG7 was observed in patients with adolescent idiopathic scoliosis. Here, we identify the ADGRG7 as an estrogen-responsive gene under the regulation of estrogen receptor ERα in scoliotic osteoblasts and other cells lines. We found that ADGRG7 expression was upregulated in response to estrogen (E2) in adolescent idiopathic scoliosis (AIS) cells. ADGRG7 promoter studies indicate the presence of an ERα response half site in close vicinity of a specificity protein 1 (SP1) binding site. Mutation of the SP1 site completely abrogated the response to E2, indicating its essential requirement. ChIP confirmed the binding of SP1 and ERα to the ADGRG7 promoter. Our results identify the ADGRG7 gene as an estrogen-responsive gene under the control of ERα and SP1 tethered actions, suggesting a possible role of estrogens in the regulation of ADGRG7. Abstract

The bacterial two-hybrid (BACTH, for "Bacterial Adenylate Cyclase-based Two-Hybrid") technique is a simple and fast genetic approach to analyze protein-protein interactions in vivo. In this system, the proteins of interest are genetically fused to two complementary fragments from the catalytic domain of Bordetella pertussis adenylate cyclase and co-expressed in strains of Escherichia coli deficient in adenylate cyclase. Association of the hybrid proteins restores synthesis of cyclic AMP (cAMP), which then triggers the expression of catabolic operons such as the lactose operon or the maltose regulon. As BACTH uses a cAMP second messenger, the association between the chimeric proteins can take place at a distance from the transcription machinery. This technique is therefore particularly appropriate for studying interactions involving integral-membrane or membrane-associated proteins that may not be soluble in the cytoplasm, and/or that may only associate in the plane of the membrane. This unit describes the basic procedures to characterize protein-protein interactions with the BACTH genetic system and to search for potential partners of known proteins. © 2017 by John Wiley & Sons, Inc.

International audience; The bacterial two-hybrid (BACTH, for "Bacterial Adenylate Cyclase-Based Two-Hybrid") system is a simple and fast genetic approach to detecting and characterizing protein-protein interactions in vivo. This system is based on the interaction-mediated reconstitution of a cyclic adenosine monophosphate (cAMP) signaling cascade in Escherichia coli. As BACTH uses a diffusible cAMP messenger molecule, the physical association between the two interacting chimeric proteins can be spatially separated from the transcription activation readout, and therefore it is possible to analyze protein-protein interactions that occur either in the cytosol or at the inner membrane level as well as those that involve DNA-binding proteins. Moreover, proteins of bacterial origin can be studied in an environment similar (or identical) to their native one. The BACTH system may thus permit a simultaneous functional analysis of proteins of interest-provided the hybrid proteins retain their activity and their association state. This chapter describes the principle of the BACTH genetic system and the general procedures to study protein-protein interactions in vivo in E. coli.

Abstract

International audience; Proximity-dependent biotin identification (BioID) is a method for identifying protein associations that occur in vivo. By fusing a promiscuous biotin ligase to a protein of interest expressed in living cells, BioID permits the labeling of proximate proteins during a defined labeling period. In this study we used BioID to study the human nuclear pore complex (NPC), one of the largest macromolecular assemblies in eukaryotes. Anchored within the nuclear envelope, NPCs mediate the nucleocytoplasmic trafficking of numerous cellular components. We applied BioID to constituents of the Nup107-160 complex and the Nup93 complex, two conserved NPC subcomplexes. A strikingly different set of NPC constituents was detected depending on the position of these BioID-fusion proteins within the NPC. By applying BioID to several constituents located throughout the extremely stable Nup107-160 subcomplex, we refined our understanding of this highly conserved subcomplex, in part by demonstrating a direct interaction of Nup43 with Nup85. Furthermore, by using the extremely stable Nup107-160 structure as a molecular ruler, we defined the practical labeling radius of BioID. These studies further our understanding of human NPC organization and demonstrate that BioID is a valuable tool for exploring the constituency and organization of large protein assemblies in living cells.
