Abstract: Large aggregate (LA) fraction of the pulmonary surfactant (PS) isolated from five different animals of the vertebrate group, lungfish, chicken, crocodile, stumpie lizard and guinea pig were isolated and characterized. Active pulmonary surfactant components were obtained by chloroform-methanol extraction of the saline suspended LA fraction. Total phospholipid (PL) and protein content were estimated biochemically by standard enzymatic methods. A systematic progression in the PL and protein content was noticed with the developmental sequence of the animals, except the crocodile, which could be due to the difficulty in the PS isolation procedure. In vitro functionality of the solvent spread film was carried out in a Langmuir surface balance by way of surface pressure (Ï€)-area (A) measurements. PS from all the species exhibited reversible compression and expansion cycles. A clear correlation between the maximum attainable surface pressure (Ï€max), also known as the collapse pressure (Ï€c) and the developmental sequence, with some exceptions, could have been established. Langmuir-Blodgett deposits, transferred onto freshly cleaved mica, were imaged by atomic force microscopy for the five different species. DPPC enriched domains showed different dimensions for the five different species. The comprehensive set of studies shed light on the composition, film functionality and structure of the pulmonary surfactants of the vertebrates where a correlation with the evolution sequence is observed.

2. Understanding GP Attitudes Towards A Data Amalgamating Health Informatics System

Abstract: This paper reports on the attitudes of 20 practitioners in South Australian General Practice towards adopting an unspecified data amalgamating Health Informatics (HI) system. Although aimed at improving the overall quality and management of healthcare, HI system adoption may require a change in the General Practitioner’s (GP) approach to the way they perform their healthcare delivery role. This qualitative study found HI adoption was primarily influenced by the perceived potential for change in the professional’s value and role. While GPs were generally reluctant to consider technological innovation that was not perceived to demonstrate potential for improvement in patient health outcomes, increased exposure to HI systems positively influenced perceptions of both the importance and the certainty of potential implementation outcomes. It was concluded that GP attitudes could be characterised by four different perspectives of HI systems use in general practice medicine delivery.

Abstract: PURPOSE: Triple negative breast cancers (estrogen, progesterone and human epidermal growth factor 2 (HER2) receptor-negative) are among the most aggressive forms of cancers with limited treatment options. Doxorubicin is one of the agents found in many of the current cancer treatment protocols, although its use is limited by dose-dependent cardiotoxicity. This work investigates one of the ways to suppress cancer growth by inhibiting tumor cell ability to remove acid accumulated during its metabolism by proton pump inhibitor esomeprazole (a drug with extensive clinical use) which could serve as an addition to doxorubicin therapy. METHODS: In this work, we have investigated growth suppression of triple-negative breast cancer cells MDA-MB-468 by esomeprazole and doxorubicin by trypan blue exclusion assay. Measurement of acidification of treated cancer cells was performed using intracellular pH-sensitive probe, BCECF-AM. Finally, expression of gastric type proton pump (H+/K+ ATPase, a target for esomeprazole) on MDA-MB-468 cells was detected by immunofluorescence and Western blotting. RESULTS: We have found that esomeprazole suppresses growth of triple-negative breast cancer cell in vitro in a dose-dependent manner through increase in their intracellular acidification. In contrast, esomeprazole did not have significant effect on non-cancerous breast epithelial MCF-10A cells. Esomeprazole increases doxorubicin effects suggesting that dual treatments might be possible. In addition, response of MDA-MB-468 cells to esomeprazole could be mediated by gastric type proton pump (H+/K+ ATPase) in cancer cells contrary to previous beliefs that this proton pump expression is restricted to parietal cells of the stomach epithelia. CONCLUSION: This study provides first evidence that adjunct use of esomeprazole in breast cancer treatment might be a possible to combat adverse effects of doxorubicin and increase its effectiveness. Abstract: Although faecal microbiota transplantation (FMT) has a well-established role in the treatment of recurrent Clostridioides difficile infection (CDI), its widespread dissemination is limited by several obstacles, including lack of dedicated centres, difficulties with donor recruitment and complexities related to regulation and safety monitoring. Given the considerable burden of CDI on global healthcare systems, FMT should be widely available to most centres.Stool banks may guarantee reliable, timely and equitable access to FMT for patients and a traceable workflow that ensures safety and quality of procedures. In this consensus project, FMT experts from Europe, North America and Australia gathered and released statements on the following issues related to the stool banking: general principles, objectives and organisation of the stool bank; selection and screening of donors; collection, preparation and storage of faeces; services and clients; registries, monitoring of outcomes and ethical issues; and the evolving role of FMT in clinical practice,Consensus on each statement was achieved through a Delphi process and then in a plenary face-to-face meeting. For each key issue, the best available evidence was assessed, with the aim of providing guidance for the development of stool banks in order to promote accessibility to FMT in clinical practice.

Abstract: International audience; In some visible light communication (VLC) systems, transmitters are light sources which support not only illumination but also information broadcast and positioning. Since transmit signals in these systems inevitably interfere with each other at the receiver side, extra devices may be added to synchronize the transmitters, but this impairs an appealing advantage in VLC systems that existing lighting infrastructure can be simply reused. This paper proposes a novel multiple access scheme to realize decentralized asynchronous VLC systems such that both the transmitters and receivers are asynchronous. For a system with N transmitters, two unique codewords of L chips are allocated to each transmitter where L is a prime number larger than 4N. Those codewords are designed to help maintain a constant transmitted average power and enhance channel estimation, as well as conveying data. Our scheme enables the receiver to obtain one channel estimate and one data symbol per L chips. Simulation results show that the proposed scheme significantly outperforms other schemes in the literature in terms of bit error rate and system throughput.

Abstract: International audience; We present a novel, web-based visual eye-tracking analytics tool called Gazealytics. Our open-source toolkit features a unified combination of gaze analytics features that support flexible exploratory analysis, along with annotation of areas of interest (AOI) and filter options based on multiple criteria to visually analyse eye tracking data across time and space. Gazealytics features coordinated views unifying spatiotemporal exploration of fixations and scanpaths for various analytical tasks. A novel matrix representation allows analysis of relationships between such spatial or temporal features. Data can be grouped across samples, user-defined AOIs or time windows of interest (TWIs) to support aggregate or filtered analysis of gaze activity. This approach exceeds the capabilities of existing systems by supporting flexible comparison between and within subjects, hypothesis generation, data analysis and communication of insights. We demonstrate in a walkthrough that Gazealytics supports multiple types of eye tracking datasets and analytical tasks.

Abstract: Coordinated changes of cellular plasticity and identity are critical for pluripotent reprogramming and oncogenic transformation. However, the sequences of events that orchestrate these intermingled modifications have never been comparatively dissected. Here, we deconvolute the cellular trajectories of reprogramming (via Oct4/Sox2/Klf4/c-Myc) and transformation (via Ras/c-Myc) at the single-cell resolution and reveal how the two processes intersect before they bifurcate. This approach led us to identify the transcription factor Bcl11b as a broad-range regulator of cell fate changes, as well as a pertinent marker to capture early cellular intermediates that emerge simultaneously during reprogramming and transformation. Multiomics characterization of these intermediates unveiled a c-Myc/Atoh8/Sfrp1 regulatory axis that constrains reprogramming, transformation and transdifferentiation. Mechanistically, we found that Atoh8 restrains cellular plasticity, independent of cellular identity, by binding a specific enhancer network. This study provides insights into the partitioned control of cellular plasticity and identity for both regenerative and cancer biology.

Abstract: International audience; Refugia can facilitate the persistence of species under long-term environmental change, but it is not clear if Pleistocene refugia will remain functional as anthropogenic climate change progresses. Dieback in populations restricted to refugia therefore raises concerns about their long-term persistence. Using repeat field surveys, we investigate dieback in an isolated population of Eucalyptus macrorhyncha during two droughts and discuss prospects for its continued persistence in a Pleistocene refugium. We first confirm that the Clare Valley in South Australia has constituted a long-term refugium for the species, with the population being genetically highly distinct from other conspecific populations. However, the population lost >40 % of individuals and biomass through the droughts, with mortality being just below 20 % after the Millennium Drought (2000–2009) and almost 25 % after the Big Dry (2017–2019). The best predictors of mortality differed after each drought. While north-facing aspect of a sampling location was significant positive predictor after both droughts, biomass density and slope were significant negative predictors only after the Millennium Drought, and distance to the north-west corner of the population, which intercepts hot, dry winds, was a significant positive predictor after the Big Dry only. This suggests that more marginal sites with low biomass and sites located on flat plateaus were more vulnerable initially, but that heat-stress was an important driver of dieback during the Big Dry. Therefore, the causative drivers of dieback may change during population decline. Regeneration occurred predominantly on southern and eastern aspects, which would receive the least solar radiation. While this refugial population is experiencing severe decline, some gullies with lower solar radiation appear to support relatively healthy, regenerating stands of red stringybark, providing hope for persistence in small pockets. Monitoring and managing these pockets during future droughts will be essential to ensure the persistence of this isolated and genetically unique population.

Abstract: International audience; Despite increased awareness of the lack of gender equity in academia and a growing number of initiatives to address issues of diversity, change is slow, and inequalities remain. Abstract: Accurate lesion segmentation is critical in stroke rehabilitation research for the quantification of lesion burden and accurate image processing. Current automated lesion segmentation methods for T1-weighted (T1w) MRIs, commonly used in stroke research, lack accuracy and reliability. Manual segmentation remains the gold standard, but it is time-consuming, subjective, and requires neuroanatomical expertise. We previously released an open-source dataset of stroke T1w MRIs and manually-segmented lesion masks (ATLAS v1.2, N = 304) to encourage the development of better algorithms. However, many methods developed with ATLAS v1.2 report low accuracy, are not publicly accessible or are improperly validated, limiting their utility to the field. Here we present ATLAS v2.0 (N = 1271), a larger dataset of T1w MRIs and manually segmented lesion masks that includes training (n = 655), test (hidden masks, n = 300), and generalizability (hidden MRIs and masks, n = 316) datasets. Algorithm development using this larger sample should lead to more robust solutions; the hidden datasets allow for unbiased performance evaluation via segmentation challenges. We anticipate that ATLAS v2.0 will lead to improved algorithms, facilitating large-scale stroke research.

Abstract: Background: Many children worldwide do not eat recommended amounts of vegetables. Disliking vegetables is a key factor associated with low intake. Objective: This umbrella review synthesized systematic reviews to determine the effectiveness of sensory and behavioral strategies to facilitate liking of vegetables (primary outcome) in young children up to 5 y of age, as key predictors of vegetable intake (secondary outcome). Methods: Nine databases were searched up to May 2019 (updated in September 2020). Two reviewers independently conducted study screening and selection, data extraction, and assessment of methodological quality using AMSTAR 2 (A MeaSurement Tool to Assess systematic Reviews). Eleven reviews (n = 1 rated strong quality, n = 4 moderate quality, n = 6 low/critically low quality)examining 85 primary studies met the review criteria: systematic reviews and meta-analyses of primary studies (any quantitative design) that examined sensory or behavioral strategies on vegetable liking or intake (outcomes reported separately for children ≤5y). Strategy effectiveness was synthesized into 3 categories based on evidence strength: 1) promising (large and consistent body of moderate quality evidence), 2) emerging (small to moderate body of mixed consistency and quality evidence), and 3) limited (small body of limited consistency and quality evidence). Results: Promising evidence was identified for repeated exposure to a single or a variety of vegetables. Emerging evidence was identified for several strategies that increase familiarity with vegetable flavors(e.g., via exposure in utero and through breast milk, and a “vegetable first” approach to complementary feeding) and/or willingness to try vegetables (e.g., via parental role modeling, nonfood rewards, and vegetable-based story books). Conclusions: Current evidence supports incorporation of tailored advice into guideline documents for parents and carers to repeatedly expose their children to a variety of vegetables to increase vegetable intake. Abstract: The physical and mechanical processes rooted in the hydrated, serpentinized mantle above subduction zones remain insufficiently explored despite fundamental implications for our understanding of rheology and fluid recycling along subduction interfaces. Through a field-based investigation, serpentinized peridotites and jadeitite samples from a fossil forearc mantle in the Polar Urals (Russia) are studied here to document fluid–rock interaction processes in the high-P field, as well as the long-term evolution of the base of the mantle wedge. Petrographic, geochemical and microstructural observations reveal a complex, protracted evolution of the jadeitite-forming fluid pathway throughout the gradual cooling of the forearc mantle and increasing serpentinization of the host. It is shown that the jadeitite lenses in the studied locality (a) derive for a large part from a trondhjemitic dyke earlier emplaced in a warm subduction environment, and (b) record the cooling of the subduction hangingwall under high-P conditions associated with increasing host serpentinization. In the studied locality, the majority of the jadeitites formed at relatively high temperatures (>600°C) by the influx of Na–Al-rich, slab-derived metamorphic fluids that were drained along the base of the mantle wedge, parallel to the subduction interface. Changes in bulk-rock geochemical signatures and in paragenetic sequences also constrain the compositional evolution of the fluid channelized along this drainage, with an increasing sedimentary component. The phlogopite-bearing walls of the dyke exhibit Rb–Sr and Ar–Ar ages ranging between c. 405 and c. 390 Ma, a range partly overlapping within uncertainty with the previously dated zircons from the jadeitite core (410–⁠400 Ma; U–Pb). Abstract: International audience; We propose TimeTables, a novel prototype system that aims to support data exploration, using embodiment with space-time cubes in virtual reality. TimeTables uses multiple space-time cubes on virtual tabletops, which users can manipulate by extracting time layers or individual buildings to create new tabletop views. The surrounding environment includes a large space for multiple linked tabletops and a storage wall. TimeTables presents information at different time scales by stretching layers to drill down in time. Users can also jump into tabletops to inspect data from an egocentric perspective. We present a use case scenario of energy consumption displayed on a university campus to demonstrate how our system could support data exploration and analysis over space and time. From our experience and analysis we believe the system has a high potential in assisting spatio-temporal data exploration and analysis.

Abstract: Background The optimal dosing of antibiotics in critically ill patients receiving renal replacement therapy (RRT) remains unclear. In this study, we describe the variability in RRT techniques and antibiotic dosing in critically ill patients receiving RRT and relate observed trough antibiotic concentrations to optimal targets. Methods We performed a prospective, observational, multinational, pharmacokinetic study in 29 intensive care units from 14 countries. We collected demographic, clinical, and RRT data. We measured trough antibiotic concentrations of meropenem, piperacillin-tazobactam, and vancomycin and related them to high- and low-target trough concentrations. Results We studied 381 patients and obtained 508 trough antibiotic concentrations. There was wide variability (4–8-fold) in antibiotic dosing regimens, RRT prescription, and estimated endogenous renal function. The overall median estimated total renal clearance (eTRCL) was 50 mL/minute (interquartile range [IQR], 35–65) and higher eTRCL was associated with lower trough concentrations for all antibiotics (P < .05). The median (IQR) trough concentration for meropenem was 12.1 mg/L (7.9–18.8), piperacillin was 78.6 mg/L (49.5–127.3), tazobactam was 9.5 mg/L (6.3–14.2), and vancomycin was 14.3 mg/L (11.6–21.8). Trough concentrations failed to meet optimal higher limits in 26%, 36%, and 72% and optimal lower limits in 4%, 4%, and 55% of patients for meropenem, piperacillin, and vancomycin, respectively. Conclusions In critically ill patients treated with RRT, antibiotic dosing regimens, RRT prescription, and eTRCL varied markedly and resulted in highly variable antibiotic concentrations that failed to meet therapeutic targets in many patients.

We address sustainability and describe the processes used to engage the EOSC-Life funded projects, as well as future work.

Abstract: International audience; This research investigates the health impacts and access to health services by children who are engaged in waste collection in Dhaka, the capital city of Bangladesh. The relevant qualitative data were collected through expert interviews and personal observations, while quantitative data were gathered through a face-to-face questionnaire survey given to 50 street children who collected waste at the landfill site located in Dhaka city's Matuail area. The results indicate that 94% of these children have suffered from many health problems, such as fever and fatigue due to tiredness, dizziness, and vomiting. Consequently, a significant portion of their daily income is spent on medical treatment. This study suggests that the waste collection system must integrate modern technological, health and environmental resources so that: firstly, they do not harm waste collectors; and secondly, rehabilitate the street children and give them better access to acceptable basic amenities. This is a priority the city authorities.

Abstract: International audience; We propose a deep reinforcement learning-based solution for the 3D reconstruction of objects of complex topologies from a single RGB image. We use a template-based approach. However, unlike previous template-based methods, which are limited to the reconstruction of 3D objects of fixed topology, our approach learns simultaneously the geometry and topology of the target 3D shape in the input image. To this end, we propose a neural network that learns to deform a template to fit the geometry of the target object. Our key contribution is a novel reinforcement learning framework that enables the network to also learn how to adjust, using pruning operations, the topology of the template to best fit the topology of the target object. We train the network in a supervised manner using a loss function that enforces smoothness and penalizes long edges in order to ensure high visual plausibility of the reconstructed 3D meshes. We evaluate the proposed approach on standard benchmarks such as ShapeNet, and in-the-wild using unseen real-world images. We show that the proposed approach outperforms the state-of-the-art in terms of the visual quality of the reconstructed 3D meshes, and also generalizes well to out-of-category images.

Abstract: International audience; Context: The Dyspnoea-12 (D-12) and Multidimensional Dyspnea Profile (MDP) were specifically developed for assessment of multiple sensations of breathlessness.Objectives: This systematic review aimed to identify the use and measurement properties of the D-12 and MDP across populations, settings and languages.Methods: Electronic databases were searched for primary studies (2008-2020) reporting use of the D-12 or MDP in adults. Two independent reviewers completed screening and data extraction. Study and participant characteristics, instrument use, reported scores and minimal clinical important differences (MCID) were evaluated. Data on internal consistency (Cronbach's α) and test-retest reliability (intraclass correlation coefficient, ICC) were pooled using random effects models between settings and languages.Results: A total 75 publications reported use of D-12 (n = 35), MDP (n = 37) or both (n = 3), reflecting 16 chronic conditions. Synthesis confirmed two factor structure, internal consistency (Cronbach's α mean, 95% CI: D-12 Total = 0.93, 0.91-0.94; MDP Immediate Perception [IP] = 0.88, 0.85-0.90; MDP Emotional Response [ER] = 0.86, 0.82-0.89) and 14 day test-rest reliability (ICC: D-12 Total = 0.91, 0.88-0.94; MDP IP = 0.85, 0.70-0.93; MDP ER = 0.84, 0.73-0.90) across settings and languages. MCID estimates for clinical interventions ranged between -3 and -6 points (D-12 Total) with small variability in scores over 2 weeks (D-12 Total 2.8 (95% CI: 2.0 to 3.7), MDP-A1 0.8 (0.6 to 1.1) and six months (D-12 Total 2.9 (2.0 to 3.7), MDP-A1 0.8 (0.6 to 1.1)).Conclusion: D-12 and MDP are widely used, reliable, valid and responsive across various chronic conditions, settings and languages, and could be considered standard instruments for measuring dimensions of breathlessness in international trials.

Abstract: BackgroundResponse to lithium in patients with bipolar disorder is associated with clinical and transdiagnostic genetic factors. The predictive combination of these variables might help clinicians better predict which patients will respond to lithium treatment.AimsTo use a combination of transdiagnostic genetic and clinical factors to predict lithium response in patients with bipolar disorder.MethodThis study utilised genetic and clinical data (n = 1034) collected as part of the International Consortium on Lithium Genetics (ConLi+Gen) project. Polygenic risk scores (PRS) were computed for schizophrenia and major depressive disorder, and then combined with clinical variables using a cross-validated machine-learning regression approach. Unimodal, multimodal and genetically stratified models were trained and validated using ridge, elastic net and random forest regression on 692 patients with bipolar disorder from ten study sites using leave-site-out cross-validation. All models were then tested on an independent test set of 342 patients. The best performing models were then tested in a classification framework.ResultsThe best performing linear model explained 5.1% (P = 0.0001) of variance in lithium response and was composed of clinical variables, PRS variables and interaction terms between them. The best performing non-linear model used only clinical variables and explained 8.1% (P = 0.0001) of variance in lithium response. A priori genomic stratification improved non-linear model performance to 13.7% (P = 0.0001) and improved the binary classification of lithium response. This model stratified patients based on their meta-polygenic loadings for major depressive disorder and schizophrenia and was then trained using clinical data.ConclusionsUsing PRS to first stratify patients genetically and then train machine-learning models with clinical predictors led to large improvements in lithium response prediction. When used with other PRS and biological markers in the future this approach may help inform which patients are most likely to respond to lithium treatment.

Each of these physical behaviours has been investigated in epidemiological studies, yet their codependency and interactions need to be further explored and accounted for in data analysis. Modern accelerometers capture continuous movement through the day, which presents the challenge of how to best use the richness of these data. In recent years, analytical approaches first applied in other scientific fields have been applied to physical behaviour epidemiology (eg, isotemporal substitution models, compositional data analysis, multivariate pattern analysis, functional data analysis and machine learning). A comprehensive description, discussion, and consensus on the strengths and limitations of these analytical approaches will help researchers decide which approach to use in different situations. In this context, a scientific workshop and meeting were held in Granada to discuss: (1) analytical approaches currently used in the scientific literature on physical behaviour, highlighting strengths and limitations, providing practical recommendations on their use and including a decision tree for assisting researchers’ decision-making; and (2) current gaps and future research directions around the analysis and use of accelerometer data. Abstract: We introduce an evaluation methodology for visual question answering (VQA) to better diagnose cases of shortcut learning. These cases happen when a model exploits spurious statistical regularities to produce correct answers but does not actually deploy the desired behavior. There is a need to identify possible shortcuts in a dataset and assess their use before deploying a model in the real world. The research community in VQA has focused exclusively on question-based shortcuts, where a model might, for example, answer "What is the color of the sky" with "blue" by relying mostly on the question-conditional training prior and give little weight to visual evidence. We go a step further and consider multimodal shortcuts that involve both questions and images. We first identify potential shortcuts in the popular VQA v2 training set by mining trivial predictive rules such as co-occurrences of words and visual elements. We then introduce VQA-CounterExamples (VQA-CE), an evaluation protocol based on our subset of CounterExamples i.e. image-question-answer triplets where our rules lead to incorrect answers. We use this new evaluation in a large-scale study of existing approaches for VQA. We demonstrate that even state-of-the-art models perform poorly and that existing techniques to reduce biases are largely ineffective in this context. Our findings suggest that past work on question-based biases in VQA has only addressed one facet of a complex issue. Abstract: International audience; In the animal kingdom, skin pigmentation is highly variable between species, and it contributes to phenotypes. In humans, skin pigmentation plays a part in sun protection. Skin pigmentation depends on the ratio of the two pigments pheomelanin and eumelanin, both synthesized by a specialized cell population, the melanocytes. In this review, we explore one important factor in pigmentation: the tyrosinase-related protein 1 (TYRP1) gene which is involved in eumelanin synthesis via the TYRP1 protein. Counterintuitively, high TYRP1 mRNA expression is associated with a poor clinical outcome for patients with metastatic melanomas. Recently, we were able to explain this unexpected TYRP1 function by demonstrating that TYRP1 mRNA sequesters microRNA-16, a tumor suppressor miRNA. Here, we focus on actors influencing TYRP1 mRNA abundance, particularly transcription factors, single nucleotide polymorphisms (SNPs), and miRNAs, as they all dictate the indirect oncogenic activity of TYRP1.

Abstract: During the last decade, numerous solar forecasting tools have been developed to predict the energy generation of photovoltaic (PV) farms. The quality of solar forecasts is assessed by comparing predictions with measured solar data. However, this methodology does not consider the added value of the forecasts for their applications. As a consequence, what value could be given to the improvement of forecasts considering this evaluation framework? To answer this question, this work compares the value of different operational solar forecasts for a specific application. The aim is to look for relationships between the economic value and the error metrics defined to evaluate the forecast quality. A new generation of large-scale PV plants integrates ESS. The aim is to add flexibility to the injection of the production into the grid and thus to maximize the profit by taking advantage of the possibilities offered by the electricity market, such as energy arbitrage. To optimize the operation of these specific ESS, forecasting of the solar production is of paramount importance. The study case considered in this work is a large-scale PV farm of several megawatts associated with Li-ion batteries in the Australian energy market context. For this specific case study, the results show that the metrics used to evaluate the forecast quality based on the mean absolute error (MAE) have an almost linear relationship with the economic gain brought by applying the forecast. More precisely, an improvement of 1% point in MAE results approximately in an increase of 2% points in economical gain. Refereed/Peer-reviewed

Abstract: Uniparentally-inherited markers on mitochondrial DNA (mtDNA) and the non-recombining regions of the Y chromosome (NRY), have been used for the past 30 years to investigate the history of humans from a maternal and paternal perspective. Researchers have preferred mtDNA due to its abundance in the cells, and comparatively high substitution rate. Conversely, the NRY is less susceptible to back mutations and saturation, and is potentially more informative than mtDNA owing to its longer sequence length. However, due to comparatively poor NRY coverage via shotgun sequencing, and the relatively low and biased representation of Y-chromosome variants on capture assays such as the 1240 k, ancient DNA studies often fail to utilize the unique perspective that the NRY can yield. Here we introduce a new DNA enrichment assay, coined YMCA (Y-mappable capture assay), that targets the "mappable" regions of the NRY. We show that compared to low-coverage shotgun sequencing and 1240 k capture, YMCA significantly improves the mean coverage and number of sites covered on the NRY, increasing the number of Y-haplogroup informative SNPs, and allowing for the identification of previously undiscovered variants. To illustrate the power of YMCA, we show that the analysis of ancient Y-chromosome lineages can help to resolve Y-chromosomal haplogroups. As a case study, we focus on H2, a haplogroup associated with a critical event in European human history: the Neolithic transition. By disentangling the evolutionary history of this haplogroup, we further elucidate the two separate paths by which early farmers expanded from Anatolia and the Near East to western Europe., Open Access funding enabled and organized by Projekt DEAL. Abstract: PurposeCompanies develop and implement environmental initiatives in particular national governance and institutional contexts. The purpose of this paper is to study how the background governance conditions of legal systems, economic policies and national culture enable or impede the relationship between corporate environmental performance (CEP) and lagged corporate financial performance (CFP).Design/methodology/approachThis is an empirical study of 427 MNCs headquartered in 22 different countries. The authors merged data from the SiRi database (generally known as SustainAnalytics now), which contains ratings of stakeholder relations for 427 large corporations with publicly available data from Datastream.FindingsDrawing on the new institutionalism in economics and sociology, the authors show that common-law systems and high economic freedom in a company’s home country tend to strengthen the CEP-CFP link. In addition, the home-country cultural variables of uncertainty avoidance, long-term orientation, and (to a lesser extent) masculinity may impede the deployment of CEP for maximum financial gain at the organizational level. The macrolevel analysis starts to move the field toward an understanding of the particular national governance configurations that provide the most supportive conditions for any CEP-CFP links.Originality/valueOne of the central questions in the field of organizations and the natural environment is about the background conditions that may incentivize and reward firms to be more environmentally responsive. The paper addresses this issue through a nation-level investigation of the background governance conditions that may help or hinder the relationship between CEP and CFP.

Abstract: Properties of rubberized cement-based composites have been investigated and the up-to-date research findings have demonstrated the practical feasibility of using the materials for large surface applications. Despite the detrimental effect in some mechanical properties, beneficial characteristics known as improved strain capacity, low propensity for restrained shrinkage cracking, high freeze-thaw resistance, better sound insulation, etc. make such composites relatively interesting for pavements such as road dedicated for low traffic volume, base or sub-base courses, or overlays. Abstract: International audience; Exploring aspects of how innovative methods can truly attain a sustainable society is necessary for the future of our planet. This study focuses on serious games, and how users can increase their understanding of sustainability issues and their familiarity with sustainable development strategies. “Users” of serious games consist of all possible target groups that are interested in attaining knowledge of sustainability through the use of games that are designed for a purpose beyond entertainment, in this case for sustainability education. This paper follows the systematic literature review method to deliver a study of serious games featuring sustainable development practices and policies. In order to provide a thorough analysis of their dynamic features, 77 games were explored in this study. The findings show the growing number of serious games that seek to educate in sustainability and the categorization of these games according to the triple-bottom line of sustainability, giving clarification hints to users wishing to select the relevant tool that offers an understanding of specific sustainability issues. The purpose of such research is to reveal the contribution of serious games as effective tools in facilitating sustainability education and to group them according to their nature and direction in relation to sustainability. Limitations in their effectiveness are also identified and a research agenda for new, relevant serious games is proposed that will enhance holistic knowledge and make it easier to clarify their pedagogical basis. The recipients of the findings will be all those future users and trainers who are interested in accessing sustainability education patterns through the use of serious games. This study will enable them to select the serious game that best serves their needs.

Abstract: Hypothesis: The formation of polyion complexes (PICs) comprising thermoresponsive polymers is intended to result in the formation of aggregates that undergo significant structural changes with temperature. Moreover the observed modifications might be critically affected by polymer structure and PICs composition. Experiments: Different block copolymers based on cationic poly(3-acrylamidopropyltrimethylammonium chloride) and thermoresponsive poly(N-isopropylacrylamide) were synthesized by aqueous RAFT/MADIX polymerization at room temperature. Addition of poly(acrylic acid) in a controlled fashion led to the formation of PICs aggregates. The structural changes induced by temperature were characterized by differential scanning calorimetry, Nuclear Magnetic Resonance spectroscopy and scattering methods. Findings: Thermoresponsive PICs undergo significant structural changes when increasing temperature above the cloud point of the thermoresponsive block. The reversibility of these phenomena depends strongly on the structural parameters of the block copolymers and on PICs composition. Refereed/Peer-reviewed

Abstract: International audience; Sensitivity analysis (SA) is en route to becoming an integral part of mathematical modeling. The tremendous potential benefits of SA are, however, yet to be fully realized, both for advancing mechanistic and data-driven modeling of human and natural systems, and in support of decision making. In this perspective paper, a multidisciplinary group of researchers and practitioners revisit the current status of SA, and outline research challenges in regard to both theoretical frameworks and their applications to solve real-world problems. Six areas are discussed that warrant further attention, including (1) structuring and standardizing SA as a discipline, (2) realizing the untapped potential of SA for systems modeling, (3) addressing the computational burden of SA, (4) progressing SA in the context of machine learning, (5) clarifying the relationship and role of SA to uncertainty quantification, and (6) evolving the use of SA in support of decision making. An outlook for the future of SA is provided that underlines how SA must underpin a wide variety of activities to better serve science and society.

Abstract: International audience; This paper addresses the challenge of embedded computing resources required by future autonomous Unmanned Aircraft Systems (UAS). Based on an analysis of the required onboard functions that will lead to higher levels of autonomy, we look at most common UAS tasks to first propose a classification of UAS tasks considering categories such as flight, navigation, safety, mission and executing entities such as human, offline machine, embedded system. We then analyse how a given combination of tasks can lead to higher levels of autonomy by defining an autonomy level. We link UAS applications, the tasks required by those applications, the autonomy level and the implications on computing resources to achieve that autonomy level. We provide insights on how to define a given autonomy level for a given application based on a number of tasks. Our study relies on the state-of-the-art hardware and software implementations of the most common tasks currently used by UAS, also expected tasks according to the nature of their future missions. We conclude that current computing architectures are unlikely to meet the autonomy requirements of future UAS. Our proposed approach is based on dynamically reconfigurable hardware that offers benefits in computational performance and energy usage. We believe that UAS designers must now consider the embedded system as a masterpiece of the system.

Abstract: International audience; Multi-task learning by robots poses the challenge of the domain knowledge: complexity of tasks, complexity of the actions required, relationship between tasks for transfer learning. We demonstrate that this domain knowledge can be learned to address the challenges in life-long learning. Specifically, the hierarchy between tasks of various complexities is key to infer a curriculum from simple to composite tasks. We propose a framework for robots to learn sequences of actions of unbounded complexity in order to achieve multiple control tasks of various complexity. Our hierarchical reinforcement learning framework, named SGIM-SAHT, offers a new direction of research, and tries to unify partial implementations on robot arms and mobile robots. We outline our contributions to enable robots to map multiple control tasks to sequences of actions: representations of task dependencies, an intrinsically motivated exploration to learn task hierarchies, and active imitation learning. While learning the hierarchy of tasks, it infers its curriculum by deciding which tasks to explore first, how to transfer knowledge, and when, how and whom to imitate.

Abstract: International audience; Green buildings are synonymous with environmental sustainability; however, it is unclear what role its occupants, specifically employees, play in promoting sustainability in green office buildings. This paper proposes that Green Human Resource Management (HRM), underpinned by Social Identity Theory (SIT), can maximize the potential of green behaviors to improve employees' outcomes (e.g., job satisfaction and work-related flow) in green office buildings by creating positive workplace behaviors (i.e., green behaviors). We collected multisource data from 549 employees and 91 managers working in 17 organizations in green office buildings across Australia. We confirm the double mediation effects of Green HRM, green behaviors and organizational identification on the relationships between organizational readiness, job satisfaction and work-related flow. The paper makes theoretical contributions by advancing the concept of Green HRM and green behaviors within the realm of SIT, thus taking a multidisciplinary stance in the built environment and sustainability literatures.

Abstract: The main objective of this study is to analyse the rationales, challenges and prospects of Dhaka City being split up, for the purposes of creating a sustainable city grounded in relevant theories and widely used and accepted standards. We cited the opinions of urban planners and politicians and referred to relevant theories and standards to achieve this objective. An assessment of Dhaka being divided in accordance with the concept of City Development Strategies reveals considerable deficiencies in most aspects of public goods and services provisions. Whilst splitting Dhaka into separate sections supports the World City Hypothesis it is not without criticisms, for instance those raised by urban planners, experts and politicians. The lack of resources and oversight to address those deficiencies and problems and the administrative, allocative, economic and social inefficiencies makes it very difficult for Dhakas authorities to achieve sustainable urbanisation. For this reason, stringent decisions and appropriate strategies must be implemented by the Bangladeshi government to resolve these problems, inefficiencies and mismanagement in order for the city to be liveable sustainable. Abstract: International audience; Advances in plasma medicine, and particularly those concerning dermatology and microbial decontamination of biological surfaces, have recently led several laboratories to take an interest in the applications of atmospheric pressure cold plasmas to cosmetics (e.g. [1][2][3]), a field in which the use of "physical" principles (e.g. weak current, light, ultrasound, lasers) is more widespread. At the same time, there are already plasma-based systems for skin treatments on the market, many of which use thermal effect mediated by plasmas whose action, linked to local energy delivery and increase in temperature [4], is fundamentally different from that of non-equilibrium cold plasmas. In addition, the treatment using thermal plasmas, usually involve damages to healthy skin (mostly erythema and micro-burns) which should not be sought in the case of daily cosmetic treatments.Considering the above, it appeared important to discuss what could be transferred from achievements in cold plasma medicine to cosmetics, to clarify the contours of what "plasma cosmetics" can be and to appreciate what are the market needs in this sector. That also extends to decontamination and to packaging for cosmetic products as used plasma medicine sources can also serve to induce surface modifications of interest in this domain. To this end, the first meeting on Plasma Cosmetic Science (IMPCS1) was organized in Orléans, France, in November 2019. In this presentation, after recalling the general context, we will address the main themes dealt with during this meeting and we will set out the main lessons learned from the presentations and discussions concerning the future opportunities for Plasma Cosmetics.IMPCS1 was supported by Le Studium and Région Centre-Val de Loire. Abstract: With an increase in life expectancy, the incidence of chronic degenerative pathologies such as dementia has progressively risen. Cognitive impairment leads to the gradual loss of skills, which results in substantial personal and financial cost at the individual and societal levels. Grapes and wines are rich in healthy compounds, which may help to maintain homeostasis and reduce the risk of several chronic illnesses, including dementia. This review analyzed papers that were systematically searched in PubMed, MEDLINE, Embase, and CAB-Abstract, using the association between grapes (or their derivatives) and their effects on cognitive functions in humans. Analysis was restricted to epidemiological and randomized-controlled studies. Consumption of grape juice (200-500 mL/day) and/or light-to-moderate wine (one to four glasses/day) was generally associated with improved cognitive performance, while the results for other alcoholic beverages were controversial and inconclusive. Bioactive molecules contained in grapes and wine were also considered, with particular attention paid to resveratrol. Due to the relatively high doses required (150-1000 mg/day) for bioactivity coupled with its low bioavailability, resveratrol is only one of the possible grape-derived compounds that may partly underpin the beneficial effects of grapes on the central nervous system.

Abstract: International audience; Gas chromatography-olfactometry-mass spectrometry (GC-O-MS) has been very useful in identifying aroma compounds from within the complex matrix of wine. Supplementary separation can be required to overcome co-elution of volatiles or other sensory-directed chromatographic strategies are needed, including multidimensional chromatography and preparative fraction collection coupled to GC. Studies investigating ‘overripe orange’ aroma in sweet Sauternes wine and the similar ‘apricot’ aroma in Viognier wine were conducted. Wines with the targeted aroma attributes were selected and concentrated wine extracts prepared. GC-O found no individual aroma compounds with the targeted aroma attribute. Semi-preparative HPLC was used to obtain less complex fractions of the wine extracts. The fractions were eluted in water/ethanol and, therefore, could be smelled directly. Fractions with the targeted aroma character were further resolved by GC-preparative fraction collection (GC-PFC). Recombinational GC-PFC demonstrated the importance of the components within a 4 min preparative GC fraction to the ‘overripe orange’ aroma of typical Bordeaux dessert wine. In Viognier wine, monoterpenes linalool, α-terpineol and geraniol as well as benzaldehyde were found to be associated with the ‘apricot’ character. Thus, several wine aroma compounds interact for these specific aromas to be perceived. This sensory-led combination of separation techniques is a powerful tool for the identification of key compounds responsible for specific aromas across the wine and beverage industries.

Abstract: The World Health Organization Global Action Plan recommends integrated surveillance programs as crucial strategies for monitoring antibiotic resistance. Although several national surveillance programs are in place for clinical and veterinary settings, no such schemes exist for monitoring antibiotic-resistant bacteria in the environment. In this transnational study, we developed, validated, and tested a low-cost surveillance and easy to implement approach to evaluate antibiotic resistance in wastewater treatment plants (WWTPs) by targeting cefotaxime-resistant (CTX-R) coliforms as indicators. The rationale for this approach was: i) coliform quantification methods are internationally accepted as indicators of fecal contamination in recreational waters and are therefore routinely applied in analytical labs; ii) CTX-R coliforms are clinically relevant, associated with extended-spectrum β-lactamases (ESBLs), and are rare in pristine environments. Abstract: The field of energy forecasting has attracted many researchers from different fields (e.g., meteorology, data sciences, mechanical or electrical engineering) over the last decade. Solar forecasting is a fast-growing subdomain of energy forecasting. Despite several previous attempts, the methods and measures used for verification of deterministic (also known as single-valued or point) solar forecasts are still far from being standardized, making forecast analysis and comparison difficult. To analyze and compare solar forecasts, the well-established Murphy–Winkler framework for distribution-oriented forecast verification is recommended as a standard practice. This framework examines aspects of forecast quality, such as reliability, resolution, association, or discrimination, and analyzes the joint distribution of forecasts and observations, which contains all time-independent information relevant to verification. To verify forecasts, one can use any graphical display or mathematical/statistical measure to provide insights and summarize the aspects of forecast quality. The majority of graphical methods and accuracy measures known to solar forecasters are specific methods under this general framework. Additionally, measuring the overall skillfulness of forecasters is also of general interest. The use of the root mean square error (RMSE) skill score based on the optimal convex combination of climatology and persistence methods is highly recommended. By standardizing the accuracy measure and reference forecasting method, the RMSE skill score allows—with appropriate caveats—comparison of forecasts made using different models, across different locations and time periods. Abstract: International audience; Since 1988, alcohol has been classified as a Group 1 carcinogen, the highest level of risk, by the International Agency for Research on Cancer (IARC). In fact, alcohol consumption is the third leading risk factor for disease and mortality in Europe. It accounts for 4.65 % of the global burden of both injury and disease, making it one of the most preventable causes of injury and death. Tissues in closest contact with alcohol when it is ingested, such as those of the oral cavity, pharynx, esophagus and larynx, have at greater risk of becoming cancerous than other body tissues. The consumption of alcohol is also associated with an increased risk of stomach, colon, rectum, liver, female breast and ovarian cancers. Conversely, recent studies suggest that red wine components inhibit colony formation of human breast cancer and esophageal carcinoma cells, suggesting that wine-derived phenolic compounds may be inhibitory, in contrast to the alcohol component of wine. Because of a lack of systematic studies dealing with the different types of cancer and alcoholic beverages and wine in particular, in this narrative review we summarize the general risk of cancer linked to the consumption of alcoholic beverages, including wine, according to type of cancer, with 140 extracted relevant references from 1966 to 2020. Mostly epidemiological studies concerning large cohorts have been selected. For the cancers of the upper aerodigestive tract, liver, colorectum, breast cancer, pancreatic, prostate, an excessive consumption and/or misuse of alcoholic beverages is correlated with increased risk. Conversely a probable decreased risk has been found for renal/kidney cancers, as well as for Non-Hodgkin lymphomas, such as thyroid lymphomas, associated with the moderate consumption of alcoholic beverages. There is no evidence of ovarian, gastric, head and neck, and lung cancer being linked to the moderate consumption of alcoholic beverages. Cancer is a multifactorial disease, and many factors contribute to effects on health status, usually being both genetic and environmental. Habits (smoking, dietary/lifestyle pattern/ habits, physical activity), should also be taken into account when defining appropriate consumption frequencies for different types of alcoholic drink (wine, beer, spirits). Further research is needed related to wine consumption in the context of a healthy dietary and lifestyle pattern given health-promoting constituents of wine and its effects on cancer incidence.

Abstract: International audience; Hardware in the loop simulation has become a fundamental tool for the safe and rapid development of embedded systems. Dynamically and partially reconfigurable FPGA provide an energy efficient solution for high performance computing in embedded systems, such as computer vision, with limited resources. Finally 3D simulation with realistic physics simulation is required by designers of Unmanned Aerial Vehicle (UAV) and related missions. The combination of the three techniques are required to design UAV with reconfigurable HW/SW embedded systems that can self-adapt to different mission phases according to environment changes. But they require different complex and specific skills from separated communities and so are not considered simultaneously. In this paper we demonstrate a complete framework that we apply to a UAV case simulated with the well adopted Gazebo 3D simulation tool including the Ardupilot model. According to usual practices in Robotics, we use Robot Operating System (ROS) middleware over Linux that we implement on a separated Intel Cyclone V FPGA board including HW/SW interfaces. As a convincing case study we implement, besides software classical navigation tasks, a vision-based emergency-landing security task and a detect and tracking classic mission application (TLD) that can run in different HW and SW versions dynamically configured on the FPGA according to mission steps simulated with Gazebo.

Abstract: Accurate prediction of aircraft arrival times is one of the fundamental elements for air traffic controllers to manage an optimal arrival and departure sequencing on the runway, reduce flight delays, and achieve a good collaboration with airports and airlines. In this work, we analyze the feature engineering problem to predict Aircraft Landing Time (LDT) in Extended-TMA with machine learning models. Two main contributions are highlighted in this work. First, the impact of different features in LDT prediction is investigated. Second, a machine learning prediction model is presented to predict LDT. Our case of study is the E-TMA of Singapore Changi Airport (WSSS) with a radius of $100$NM. Firstly, data analysis is conducted to check the availability of different resource data, as well as cleaning the raw trajectory data. Then, feature construction and extraction are discussed in details, machine learning prediction models are proposed to address the LDT prediction. The experimental results show that 4 sets of features play a significant impact on LDT prediction for primary runway-in-use, they are: (1) Control intent: traffic demand, current traffic density, and adjacent flow; (2) Weather: surface wind; (3) Trajectory: the position of aircraft; (4) Seasonality: parts of a day and a week. Moreover, comparing three Machine Learning algorithms, in our study case, Extra-Trees is the best prediction algorithm compared with other machine learning models in terms of Root Mean Square Error (RMSE) and Mean Absolute Error (MAE). It is also found that Machine learning models perform much better than the current operational system. In summary, two main conclusions are drawn from the present work. First, predicting the aircraft LDT is strongly correlated with the TMA density at the flight operation time. Second, feature selection with domain knowledge and expert opinions is very important, and with good features, the model is less sensitive to the choice of machine learning algorithm. Abstract: International audience; This study investigates whether the US and Brazilian ethanol markets are globalized or regionalized (i.e., whether they are interdependent or independent) using weekly frequency data from July 2006 to December 2017. The empirical results indicate that the US ethanol market is unlinked to the Brazilian one in the short-run, however in the long-run they are globalized (interdependent) and co-influence each other. Asymmetric effects are identified between both ethanol markets and the Brazilian ethanol market Granger causes the US ethanol market. The findings help us understand the asymmetric dependence of the US ethanol market.

Abstract: International audience; Augmented Reality (AR) is a rapidly growing field in information and communication technologies, drawing increasing numbers of professionals. Higher education institutions, however, are struggling to keep abreast of its development and to train specialists quickly, providing few courses which sufficiently align with the needs of industry. In addition to this, the field is developing so rapidly that existing courses struggle to keep pace. They also often focus too narrowly on specifics to allow for the building of the formative foundations of AR education. This paper aims to address this need by proposing a blueprint curriculum in Computer Science Education for teaching AR in universities at two levels, foundations and advanced. To begin, we survey the state of the art, identifying common needs and problems in existing courses which focus on AR. We then detail a skills framework comprised of 12 groups of skills suitable to meet industry needs, and built upon it two model lesson plans for a foundation and an advanced course. We conclude with a discussion of assessment techniques and curricular design options of embedding such coursework into existing academic programs and a forecast of the future of this academic field.

Abstract: Agile heterogeneous embedded computing refers to the consideration of platform architectures as a first class design variable in deploying applications on heterogeneous platforms, which constitute one or more of the following compute engines: FPGA, GPU and CPU. Design flows for agile heterogeneous computing are in the early phases of development. Few state of the art design flows that consider variable platform architectures have the following drawbacks: (1) restricted template-based representations, which cannot express all possible architectural topologies, (2) limited support for concurrent tasks on all three compute engines and (3) design space exploration approaches indirectly consider variable platform architectures, because they are derived from fixed platform design flows, where different platform architectures are iteratively evaluated without closely considering the application's computational needs. This research explores solutions to these three major issues that have yet to be addressed in agile heterogeneous computing design flows. The solutions to these issues are encapsulated into a new design flow called agile heterogeneous computing flow (AhcFlow) that can support both design space exploration and deployment. In order to realise AhcFlow, a new representation to express variable platform architectures called parameterised platform graph (PPG) is conceived, a new intermediate data structure called augmented synchronous dataflow (ArcSDF) is created and a new design space exploration algorithm called agile mapping and scheduling algorithm (AMS) is developed. PPG is a constraint based representation, which unlike template-based representations has higher expressive capabilities to represent the different topologies of agile heterogeneous computing. ArcSDF is a new dataflow based intermediate data structure, which is built on the well-known synchronous dataflow graphs (SDF) to express the architectural decisions together with the application. ArcSDF extends the capabilities of the dataflow paradigm to incorporate computation resource analysis and capital cost analysis. Due to these extra analysis capabilities, ArcSDF is used within design space exploration. The design space exploration algorithm is AMS that considers the variable platform architecture in a fully integrated way together with mapping and scheduling decisions. A prototype of the new design flow (AhcFlow) has been created and shown to be valid both for an exhaustive number of synthetic test cases and for a large real life embedded multi-object visual tracking application. The new analysis capabilities of ArcSDF have been validated through its integration within the design flow prototype. The prototype also consists of a deployment module, which is used to validate the design space exploration predictions with the actual deployment results. The results from the real life tracking application show that design space exploration estimates closely match the deployment results. The new mapping and scheduling algorithm, that also makes architecture topology decisions, has shown to be competitive with published results of applications implemented manually to hand crafted architectures., L'informatique embarquée hétérogène agile fait référence à la prise en compte des architectures de plate-forme en tant que variable de conception de première classe dans le déploiement d'applications sur des plates-formes hétérogènes, qui constituent un ou plusieurs des moteurs de calcul suivants: FPGA, GPU et CPU. Les flux de conception pour l'informatique hétérogène agile en sont aux premières phases de développement. Abstract: BackgroundThe World Health Organization (WHO) and the International Labour Organization (ILO) are developing joint estimates of the work-related burden of disease and injury (WHO/ILO Joint Estimates), with contributions from a large network of individual experts. Evidence from mechanistic data and prior studies suggests that exposure to long working hours may cause stroke. In this paper, we present a systematic review and meta-analysis of parameters for estimating the number of deaths and disability-adjusted life years from stroke that are attributable to exposure to long working hours, for the development of the WHO/ILO Joint Estimates.ObjectivesWe aimed to systematically review and meta-analyse estimates of the effect of exposure to long working hours (three categories: 41–48, 49–54 and ≥55 h/week), compared with exposure to standard working hours (35–40 h/week), on stroke (three outcomes: prevalence, incidence, and mortality).Data sourcesA protocol was developed and published, applying the Navigation Guide to systematic reviews as an organizing framework where feasible. We searched electronic databases for potentially relevant records from published and unpublished studies, including Ovid MEDLINE, PubMed, EMBASE, Scopus, Web of Science, CISDOC, PsycINFO, and WHO ICTRP. We also searched grey literature databases, Internet search engines, and organizational websites; hand-searched reference lists of previous systematic reviews; and consulted additional experts.Study eligibility and criteriaWe included working-age (≥15 years) individuals in the formal and informal economy in any WHO and/or ILO Member State but excluded children (aged < 15 years) and unpaid domestic workers. We included randomized controlled trials, cohort studies, case-control studies and other non-randomized intervention studies with an estimate of the effect of exposure to long working hours (41–48, 49–54 and ≥55 h/week), compared with exposure to standard working hours (35–40 h/week), on stroke (prevalence, incidence or mortality).Study appraisal and synthesis methodsAt least two review authors independently screened titles and abstracts against the eligibility criteria at a first review stage and full texts of potentially eligible records at a second stage, followed by extraction of data from qualifying studies. Missing data were requested from principal study authors. We combined relative risks using random-effects meta-analysis. Two or more review authors assessed the risk of bias, quality of evidence and strength of evidence, using the Navigation Guide and GRADE tools and approaches adapted to this project.ResultsTwenty-two studies (20 cohort studies, 2 case-control studies) met the inclusion criteria, comprising a total of 839,680 participants (364,616 females) in eight countries from three WHO regions (Americas, Europe, and Western Pacific). The exposure was measured using self-reports in all studies, and the outcome was assessed with administrative health records (13 studies), self-reported physician diagnosis (7 studies), direct diagnosis by a physician (1 study) or during a medical interview (1 study). The outcome was defined as an incident non-fatal stroke event in nine studies (7 cohort studies, 2 case-control studies), incident fatal stroke event in one cohort study and incident non-fatal or fatal (“mixed”) event in 12 studies (all cohort studies). Cohort studies were judged to have a relatively low risk of bias; therefore, we prioritized evidence from these studies, but synthesised evidence from case-control studies as supporting evidence. For the bodies of evidence for both outcomes with any eligible studies (i.e. stroke incidence and mortality), we did not have serious concerns for risk of bias (at least for the cohort studies).Eligible studies were found on the effects of long working hours on stroke incidence and mortality, but not prevalence. Compared with working 35–40 h/week, we were uncertain about the effect on incidence of stroke due to working 41–48 h/week (relative risk (RR) 1.04, 95% confidence interval (CI) 0.94–1.14, 18 studies, 277,202 participants, I2 0%, low quality of evidence). There may have been an increased risk for acquiring stroke when working 49–54 h/week compared with 35–40 h/week (RR 1.13, 95% CI 1.00–1.28, 17 studies, 275,181participants, I2 0%, p 0.04, moderate quality of evidence). Compared with working 35–40 h/week, working ≥55 h/week may have led to a moderate, clinically meaningful increase in the risk of acquiring stroke, when followed up between one year and 20 years (RR 1.35, 95% CI 1.13 to 1.61, 7 studies, 162,644 participants, I2 3%, moderate quality of evidence).Compared with working 35–40 h/week, we were very uncertain about the effect on dying (mortality) of stroke due to working 41–48 h/week (RR 1.01, 95% CI 0.91–1.12, 12 studies, 265,937 participants, I2 0%, low quality of evidence), 49–54 h/week (RR 1.13, 95% CI 0.99–1.29, 11 studies, 256,129 participants, I2 0%, low quality of evidence) and 55 h/week (RR 1.08, 95% CI 0.89–1.31, 10 studies, 664,647 participants, I2 20%, low quality of evidence).Subgroup analyses found no evidence for differences by WHO region, age, sex, socioeconomic status and type of stroke. Sensitivity analyses found no differences by outcome definition (exclusively non-fatal or fatal versus “mixed”) except for the comparison working ≥55 h/week versus 35–40 h/week for stroke incidence (p for subgroup differences: 0.05), risk of bias (“high”/“probably high” ratings in any domain versus “low”/“probably low” in all domains), effect estimate measures (risk versus hazard versus odds ratios) and comparator (exact versus approximate definition).ConclusionsWe judged the existing bodies of evidence for human evidence as “inadequate evidence for harmfulness” for all exposure categories for stroke prevalence and mortality and for exposure to 41–48 h/week for stroke incidence. Evidence on exposure to 48–54 h/week and ≥55 h/week was judged as “limited evidence for harmfulness” and “sufficient evidence for harmfulness” for stroke incidence, respectively. Producing estimates for the burden of stroke attributable to exposures to working 48–54 and ≥55 h/week appears evidence-based, and the pooled effect estimates presented in this systematic review could be used as input data for the WHO/ILO Joint Estimates.

Abstract: The population dynamics for the replicator equation has been well studied in continuous time, but there is less work that explicitly considers the evolution in discrete time. The discrete-time dynamics can often be justified indirectly by establishing the relevant evolutionary dynamics for the corresponding continuous-time system, and then appealing to an appropriate approximation property. In this paper we study the discrete-time system directly, and establish basic stability results for the evolution of a population defined by a positive definite system matrix, where the population is disrupted by random perturbations to the genotype distribution either through migration or mutation, in each successive generation.

Abstract: Contains fulltext : 218274.pdf (Publisher’s version ) (Closed access) Genetic syndromes frequently present with overlapping clinical features and inconclusive or ambiguous genetic findings which can confound accurate diagnosis and clinical management. An expanding number of genetic syndromes have been shown to have unique genomic DNA methylation patterns (called "episignatures"). Peripheral blood episignatures can be used for diagnostic testing as well as for the interpretation of ambiguous genetic test results. We present here an approach to episignature mapping in 42 genetic syndromes, which has allowed the identification of 34 robust disease-specific episignatures. We examine emerging patterns of overlap, as well as similarities and hierarchical relationships across these episignatures, to highlight their key features as they are related to genetic heterogeneity, dosage effect, unaffected carrier status, and incomplete penetrance. We demonstrate the necessity of multiclass modeling for accurate genetic variant classification and show how disease classification using a single episignature at a time can sometimes lead to classification errors in closely related episignatures. We demonstrate the utility of this tool in resolving ambiguous clinical cases and identification of previously undiagnosed cases through mass screening of a large cohort of subjects with developmental delays and congenital anomalies. This study more than doubles the number of published syndromes with DNA methylation episignatures and, most significantly, opens new avenues for accurate diagnosis and clinical assessment in individuals affected by these disorders.
