Abstract: DNA viruses have a major influence on the ecology and evolution of cellular organisms, but their overall diversity and evolutionary trajectories remain elusive. Here we carried out a phylogeny-guided genome-resolved metagenomic survey of the sunlit oceans and discovered plankton-infecting relatives of herpesviruses that form a putative new phylum dubbed Mirusviricota. The virion morphogenesis module of this large monophyletic clade is typical of viruses from the realm Duplodnaviria, with multiple components strongly indicating a common ancestry with animal-infecting Herpesvirales. Yet, a substantial fraction of mirusvirus genes, including hallmark transcription machinery genes missing in herpesviruses, are closely related homologues of giant eukaryotic DNA viruses from another viral realm, Varidnaviria. These remarkable chimaeric attributes connecting Mirusviricota to herpesviruses and giant eukaryotic viruses are supported by more than 100 environmental mirusvirus genomes, including a near-complete contiguous genome of 432 kilobases. Moreover, mirusviruses are among the most abundant and active eukaryotic viruses characterized in the sunlit oceans, encoding a diverse array of functions used during the infection of microbial eukaryotes from pole to pole. Abstract: The family of Malacoherpesviridae is currently represented by only two viruses infecting molluscs, Ostreid herpesvirus 1 (OsHV-1) and Haliotid herpesvirus 1 (HaHV-1), both causing detrimental infections in aquaculture species. Malacoherpesvirus-like sequences were also detected through genome sequencing projects in amphioxus (Branchiostoma species) and annelid worm (Capitella teleta), suggesting the existence of a hidden diversity of malacoherpesviruses in aquatic animals. Here, to extend the knowledge on malacoherpesvirus diversity, we searched for the presence of malacoherpesvirus relatives in genomic, transcriptomic and metagenomic datasets, including from the Tara Oceans expedition, and report 4 novel malacoherpesvirus-like genomes (MalacoHV1-4). Genomic analysis suggested gastropods and bivalves as the most probable hosts for these new malacoherpesviruses. Phylogenetic analysis based on the family B DNA polymerase placed the novel MalacoHV1 and MalacoHV3 as sister lineages of OsHV-1 and HaHV-1, respectively, whereas MalacoHV2 and MalacoHV4 showed higher divergence. The viral genome found associated with amphioxus together with MalacoHV4 formed a sister clade to the mollusc and annelid malacoherpesviruses, suggesting an early divergence of the two virus assemblages. In conclusion, although relatively rare in the available sequence databases, the previously undescribed malacoherpesviruses, MalacoHV1-4, circulate in aquatic ecosystems and should be considered as possible emerging viruses under changing environmental conditions.

Abstract: Horizontal gene transfer is a powerful source of innovations in prokaryotes that can affect almost any cellular system, including microbial organelles. The formation of magnetosomes, one of the most sophisticated microbial mineral-containing organelles synthesized by magnetotactic bacteria for magnetic navigation in the environment, was also shown to be a horizontally transferrable trait. However, the mechanisms determining the fate of such genes in new hosts are not well understood, since non-adaptive gene acquisitions are typically rapidly lost and become unavailable for observation. This likely explains why gene clusters encoding magnetosome biosynthesis have never been observed in non-magnetotactic bacteria. Here, we report the first discovery of a horizontally inherited dormant gene clusters encoding biosynthesis of magnetosomes in a non-magnetotactic phototrophic bacterium Rhodovastum atsumiense. We show that these clusters were inactivated through transcriptional silencing and antisense RNA regulation, but retain functionality, as several genes were able to complement the orthologous deletions in a remotely related magnetotactic bacterium. The laboratory transfer of foreign magnetosome genes to R. atsumiense was found to endow the strain with magnetosome biosynthesis, but strong negative selection led to rapid loss of this trait upon subcultivation, highlighting the trait instability in this organism. Our results provide insight into the horizontal dissemination of gene clusters encoding complex prokaryotic organelles and illuminate the potential mechanisms of their genomic preservation in a dormant state.

Abstract: PurposeThis article of exploratory research provides a critical perspective on accountability, focusing on three characteristics: transparency, asymmetry and individual agency. An experimental method is developed, calling for an ethics of accountability.Design/methodology/approachFour entrepreneurs have given accounts of themselves and their projects in life cycle interviews. This article applies Devereux's approach (1967), which allows for opacity (the “unconscious”) to oneself and to others with symmetry between analysts and analysed, and a lack of demarcation between the observer and the observed.FindingsA tragic entrepreneurial accountability trap of continuous self-justification was discovered, which pertains both to the entrepreneurs and the researchers. Nonetheless, the researchers as inspired by Devereux's method were able to realize a form of accounterability.Social implicationsThis article shows that the demands for transparent, asymmetrical and agentive accountability call for ethical reflection. The request for accounts, as resulting in the accounts given and the research conducted into accountability, are all sources of constraints. Differing the accountability situation may lessen the constraints.Originality/valueThis study introduces Devereux's method as an investigative tool in accountability research, opening up new perspectives on communication and analysis. This article shows the researcher as situated both inside and outside of the accountability mechanisms. This article explores a singular form of accountability; that of entrepreneurs who seemingly only account for the future, thereby disconnecting them from others.

Abstract: We introduce the Human Development Index (HDI) in a growth model to show that if households cared about human capital, even if infinitesimally, then human capital would increase forever protecting economies indirectly. Here, human capital will have an additional positive effect on social welfare through the quality of individual health and education. In a simple economy with a Cobb–Douglas technology and logarithmic preferences, we provide the explicit trajectories for human capital, consumption, and the HDI, which correspond to the balanced growth path (BGP). Using a two-step maximization strategy, we compute the optimal initial value of the control variable, in this case, the initial optimal labour supply. In other words, we prove the optimality of the BGP. We highlight a HDI crossing property: the propensity to consume has a positive effect on the HDI in the short run, but negative in the long run.

Abstract: Myotonic dystrophy type I (DM1) is one the most frequent muscular dystrophies in adults. Although DM1 has long been considered mainly a muscle disorder, growing evidence suggests the involvement of peripheral nerves in the pathogenicity of DM1 raising the question of whether motoneurons (MNs) actively contribute to neuromuscular defects in DM1.By using micropatterned 96-well plates as a co-culture platform, we generated a functional neuromuscular model combining DM1 and MBNL-knock-out human induced pluripotent stem cells-derived MNs and human healthy skeletal muscle cells.This approach led to the identification of pre-synaptic defects which affect the formation or stability of the neuromuscular junction at an early developmental stage. These neuropathological defects could be reproduced by the loss of RNA-binding MBNL proteins, whose loss of function in vivo is associated with muscular defects associated with DM1. These experiments indicate that the functional defects associated with MNs can be directly attributed to MBNL family proteins. Comparative transcriptomic analyses also revealed specific neuronal-related processes regulated by these proteins that are commonly misregulated in DM1.Beyond the application to DM1, our approach to generating a robust and reliable human neuromuscular system should facilitate disease modelling studies and drug screening assays.

Abstract: SulfAtlas (https://sulfatlas.sb-roscoff.fr/) is a knowledge-based resource dedicated to a sequence-based classification of sulfatases. Currently four sulfatase families exist (S1–S4) and the largest family (S1, formylglycine-dependent sulfatases) is divided into subfamilies by a phylogenetic approach, each subfamily corresponding to either a single characterized specificity (or few specificities in some cases) or to unknown substrates. Sequences are linked to their biochemical and structural information according to an expert scrutiny of the available literature. Database browsing was initially made possible both through a keyword search engine and a specific sequence similarity (BLAST) server. In this article, we will briefly summarize the experimental progresses in the sulfatase field in the last 6 years. To improve and speed up the (sub)family assignment of sulfatases in (meta)genomic data, we have developed a new, freely-accessible search engine using Hidden Markov model (HMM) for each (sub)family. This new tool (SulfAtlas HMM) is also a key part of the internal pipeline used to regularly update the database. SulfAtlas resource has indeed significantly grown since its creation in 2016, from 4550 sequences to 162 430 sequences in August 2022.

Abstract: In a simple discrete-time version of the Lucas (1988) model, we prove, first, that the Balanced Growth Path (BGP) is optimal and, second, that the optimal solution is unique. After briefly discussing the results obtained in some of the continuous-time versions of the Lucas model, we address the issues of existence and uniqueness of the optimal solution in discrete time. Making use of the supermodularity of the value function, we prove that the optimal solution must be monotonically increasing. This feature does indeed suit the BGP, although nothing can ensure yet its optimality. After providing the exact expression of the BGP, we do prove both its optimality and its uniqueness.

Abstract: Background Extended-spectrum β-lactamase–producing Enterobacterales (ESBL-E) represent a major threat to public health. Little is known on their potential for sexual transmission. Methods We recruited individuals at a sexually transmitted infection and human immunodeficiency virus (HIV) outpatient clinic in Paris, France, in whom we evaluated the prevalence of ESBL-E intestinal carriage and, among those testing positive, the proportion with clearance 6 months thereafter. We compared carriage prevalence between groups using logistic regression adjusted for age, geographic origin, travel outside Europe, and antibiotic use in the past 6 months. Results A total of 2157 individuals participated, of whom 226 (10.5%) were ESBL-E carriers. The proportions of ESBL-E carriers varied across sexual groups and were as follows: HIV-negative men who have sex with men (MSM) and who were on preexposure prophylaxis (PrEP), 16.3% (41 of 251); HIV-negative MSM not on PrEP, 9.7% (47 of 487); HIV-positive MSM, 12.2% (61 of 500); HIV-negative men who have sex exclusively with women, 10.0% (44 of 439); and HIV-negative women who have sex with men, 6.9% (n = 33 of 480). After adjustment, ESBL-E prevalence was significantly higher in HIV-negative MSM on PrEP (P Conclusion ESBL-E carriage is more frequent in MSM undergoing PrEP or living with HIV and with increasing number of sexual partners. More research is warranted to understand the consequences of ESBL-E carriage in these populations and how transmission can be reduced.

Abstract: Cancer patients at advanced stages experience a severe depletion of skeletal muscle compartment together with a decrease in muscle function, known as cancer cachexia. Cachexia contributes to reducing quality of life, treatment efficiency, and lifespan of cancer patients. However, the systemic nature of the syndrome is poorly documented. Here, we hypothesize that glucocorticoids would be important systemic mediators of cancer cachexia.To explore the role of glucocorticoids during cancer cachexia, biomolecular analyses were performed on several tissues (adrenal glands, blood, hypothalamus, liver, and skeletal muscle) collected from ApcTwenty-three-week-old Apc mice recapitulated important features of cancer cachexia including body weight loss (-16%, P 0.0001), muscle atrophy (gastrocnemius muscle: -53%, P 0.0001), and weakness (-50% in tibialis anterior muscle force, P 0.0001), increased expression of atrogens (7-fold increase in MuRF1 transcript level, P 0.0001) and down-regulation of Akt-mTOR pathway (3.3-fold increase in 4EBP1 protein content, P 0.0001), together with a marked transcriptional rewiring of hepatic metabolism toward an increased expression of gluconeogenic genes (Pcx: +90%, Pck1: +85%), and decreased expression of glycolytic (Slc2a2: -40%, Gk: -30%, Pklr: -60%), ketogenic (Hmgcs2: -55%, Bdh1: -80%), lipolytic/fatty oxidation (Lipe: -50%, Mgll: -60%, Cpt2: -60%, Hadh: -30%), and lipogenic (Acly: -30%, Acacb: -70%, Fasn: -45%) genes. The hypothalamic pituitary-adrenal axis was activated, as evidenced by the increase in the transcript levels of genes encoding corticotropin-releasing hormone in the hypothalamus (2-fold increase, P 0.01), adrenocorticotropic hormone receptor (3.4-fold increase, P 0.001), and steroid biosynthesis enzymes (Cyp21a1, P 0.0001, and Cyp11b1, P 0.01) in the adrenal glands, as well as by the increase in corticosterone level in the serum (+73%, P 0.05), skeletal muscle (+17%, P 0.001), and liver (+24%, P 0.05) of cachectic 23-week-old Apc mice. A comparative transcriptional analysis with dexamethasone-treated C57BL/6J mice indicated that the activation of the hypothalamic-pituitary-adrenal axis in 23-week-old ApcThese findings highlight the role of the hypothalamic-pituitary-adrenal-glucocorticoid pathway in the transcriptional regulation of skeletal muscle catabolism and hepatic metabolism during cancer cachexia. They also provide the paradigm for the design of new therapeutic strategies.

Abstract: International audience; Karrikins, of which KAR1 and KAR2 are representatives, are a class of abiotic plant growth regulators produced from the partial combustion of plant material (Flematti et al., 2004; Flematti et al., 2009). Released into the soil following wildfires, karrikins can promote seed germination and seedling photomorphogenesis, and thereby facilitate the revegetation process (Nelson et al., 2009; Nelson et al., 2010; Stevens et al., 2007). The butenolide moiety of karrikins is also an essential structural feature of strigolactones (SLs), which are carotenoid-derived plant hormones that regulate a range of developmental processes in plants, and also serve as a rhizosphere signal for arbuscular mycorrhizal fungi and root parasitic weeds (Waters et al., 2016; Bürger and Chory, 2020). Collectively, these butenolides possess wide-ranging biological activities with considerable agricultural and environmental significance.

Abstract: International audience; Nous avons examiné ici comment les acteurs.rices de laboratoires étudiant l’impression 3D métallique s’organisent face aux risques de cette technologie émergente, en prêtant attention aux négociations spécifiques auxquelles cette période donne lieu entre normes juridiques, technologiques et normes scientifiques ainsi qu’aux interactions entre ces différents registres normatifs. Travaillant sur des machines expérimentales, les acteurs.rices font face à une situation qu’ils décrivent comme « hors-norme » : les imprimantes 3D qu’ils.elles étudient ne sont pas stabilisées ni standardisées, leurs risques sont mal caractérisés. Les usages de ces imprimantes à des fins de recherche, donnant lieu à du « bricolage » et les pannes associées, renforcent la prise de risque – confirmant la spécificité de la recherche en matière de santé au travail. Dans ce contexte, les logiques de précaution et de prévention identifiées par la littérature autour d’autres risques professionnels incertains permettent de décrire la manière dont ont été prises les principales décisions en matière de sécurité et dont s’articulent, dans le quotidien des acteurs.rices, les normativités concernées. La logique de précaution, dont l’interprétation s’est opérée en amont de la recherche entre un nombre réduit d’acteurs.rices, a donné lieu au choix de séparer l’imprimante 3D du reste du laboratoire par un caisson aussi hermétique que possible. Elle isole, ce faisant, les acteurs.rices les plus précaires et les plus exposé.e.s du reste du laboratoire et acte une répartition spatiale et sociale des risques. La logique de prévention oblige l’employeur à former, informer, et protéger ses salariés. Si cette seconde modalité est complexe face à un risque émergent, la comparaison s’est révélée heuristique pour mettre en évidence des différences importantes entre deux des laboratoires étudiés, qui s’expliquent par les mobilisations inégales des « ingénieurs de terrain » dans la négociation de normativités en cours de construction. Les négociations auxquelles donne lieu la gestion des risques de l’impression 3D métallique permettent d’appréhender, dans le quotidien des acteurs.rices et dans leurs pratiques, des processus originaux de coproduction du droit, des sciences et des techniques. Si ces processus ne permettent pas de conclure quant à la qualité des mesures qui seront finalement adoptées pour protéger la santé et la sécurité des opérateurs.rices, ils confirment l’intérêt de la phase d’émergence technologique pour mieux comprendre la façon dont les normativités en jeu interagissent et dont les acteurs.rices, consciemment ou pas, se positionnent à leur égard.

Abstract: Controlled dielectric breakdown (CDB) is gaining popularity for fabricating solid-state nanopores in situ with size control in a simple, low-cost, and scalable way. This technique could be used for a broad type of applications in the field of nucleic acid analysis and even for protein studies. In this work, we studied the entry and transport of double-stranded DNAs using a solid-state nanopore fabricated by CDB as a function of applied voltage for two different DNA lengths. We showed that the blockade rate increases exponentially with voltage up to 120 mV. The energy barrier depends on the chain length, and the dwell times decrease with applied voltage up to 120 mV. Moreover, no matter the chain length, it is possible to differentiate two families of blockade amplitudes, high and low ones, due to DNA folding.

Abstract: The entire world is nowadays undergoing major ecological crises with the current mass extinction of a wide range of living forms. Indeed, between 20% to 50% of all living species may extinct in the 21st century. To understand these phenomenons, some ecologists use formal methods to model ecosystems and analyse their possible trajectories [Gaucherel, Pommereau 2019]. A major difficulty in this approach is the combinatorics that leads to models with huge state spaces. Symbolic techniques like decisions diagrams allow to efficiently compute these state spaces and analyse them automatically [Thierry-Mieg 2016]. However, this does not help with the understanding of the models by the ecologists because such large state spaces remain opaque to manual exploration. The techniques developed so far to explore such large state spaces are based on their presentation into topological components (strongly connected components, deadlocks, basins of attraction), but they do not scale to models with a large number of components, or with very large components, which is the case of the models that require symbolic techniques. This thesis aims at developing analysis techniques for these large state spaces relying on question of ecological nature, that should lead to splits of the state space into macro-trajectories interpretable by an ecologist. To do so, it is expected to start from, on the one hand, an existing modelling language [Gaucherel, Pommereau 2019] to formulate queries in the terms used by the modeller, and, on the other hand, on temporal logics to allow the expression of trajectories the modeller wants to study in particular (or conversely, those she wants to ignore for her exploration space). These requests should address both the states and the transitions in the system, which are the two basic elements handled by the modeller. These requests will have to be complemented with analysis techniques of the components allowing to present some of their significant characteristics in order to guide the modeller in the refinement of his requests. On the long term, the goal is this to propose efficient exploration methods of large state spaces, guided by ecological queries that an ecologist modeller is able to express. Finally, the methods developed during the thesis will have to be applied on cases studies about ecosystems studied in Cédric Gaucherel's team.; Le monde subit actuellement des crises écologiques majeures avec l'extinction de masse d'un très grand nombre d'espèces vivantes. De l'ordre de 20 à 50% des espèces vivant au 21ème siècle devrait s'éteindre au cours du siècle. Pour comprendre ces phénomènes, des écologues utilisent des méthodes formelles pour modéliser les écosystèmes et analyser leurs trajectoires possibles [Gaucherel, Pommereau 2019]. Les techniques développées jusqu'à présent pour permettre une telle exploration de grands espaces d'états se basent sur une présentation des espaces d'états selon ses composantes topologiques (composantes fortement connexes, blocages, bassins d'attractions), mais elle ne passent pas à l'échelle de modèles ayant un grand nombre de composantes, ou des composantes de grande taille, ce qui est le tas des modèles nécessitant des techniques symboliques L'objectif de la thèse est de développer des techniques d'analyse de ces grands espaces d'états en s'appuyant sur des questions d'ordre écologique, qui devront se traduire par des découpages de l'espace d'états en macro-trajectoires interprétables par des écologues. Pour cela, il faudra se baser d'une part sur le langage de modélisation existant [Gaucherel, Pommereau 2019] pour formuler des requêtes dans les termes utilisés par le modélisateur, et d'autre part sur des logiques temporelles permettant d'exprimer des trajectoires sur lesquelles le modélisateur souhaite porter son attention (ou au contraire qu'il souhaite éliminer de son espace d'exploration). Ces requêtes devront pouvoir porter à la fois sur les états et sur les transitions du système, qui sont les deux éléments de base manipulés par le modélisateur. Ces requêtes devront être complétés par des méthodes d'analyse des composantes symboliques permettant d'en présenter des caractéristiques pertinentes pour le modélisateur afin de le guider dans le raffinement de ses requêtes. Il s'agit donc à terme de proposer des méthodes d'exploration efficaces de grands espaces d'états, guidées par des questions écologiques formulables par un modélisateur écologue. Enfin, les méthodes développées devront être appliquées sur des études de cas portant sur des écosystèmes étudiés dans l'équipe de Cédric Gaucherel.

Abstract: International audience; This chapter examines the impact of directors’ home regions on employee human rights policy (EHRP) disclosure in the French context. We classify foreign directors into four home regions, namely, Anglo-American, European, Asian, and Middle Eastern regions. Logistic regression analyses are conducted, referring to a sample of 73 French firms that were listed on the Euronext Paris Stock Exchange and members of the SBF 120 index over the period from 2008 to 2018. EHRP disclosure data are collected from the Bloomberg database and indicate whether the firm has disclosed EHRP information in its annual report or CSR statement. Results provide evidence that board home region diversity is positively and significantly associated with EHRP disclosure. Results also show that the presence of Anglo-American and Asian directors is positively associated with EHRP disclosure. Surprisingly, European directors are less concerned about EHRP disclosure. Furthermore, we provide evidence that there is no relationship between the presence of directors from the Middle East and EHRP disclosure. The chapter highlights the need to reconsider the composition of French boards by promoting the presence of directors from different home regions. Specifically, the authors support the recommendations of the AFEP-MEDEF (2020) regarding the need to intensify international recruitment when selecting new board members.

Abstract: The smallest phytoplankton species are key actors in oceans biogeochemical cycling and their abundance and distribution are affected with global environmental changes. Among them, algae of the Pelagophyceae class encompass coastal species causative of harmful algal blooms while others are cosmopolitan and abundant. The lack of genomic reference in this lineage is a main limitation to study its ecological importance. Here, we analysed Pelagomonas calceolata relative abundance, ecological niche and potential for the adaptation in all oceans using a complete chromosome-scale assembled genome sequence. Our results show that P. calceolata is one of the most abundant eukaryotic species in the oceans with a relative abundance favoured by high temperature, low-light and iron-poor conditions. Climate change projections based on its relative abundance suggest an extension of the P. calceolata habitat toward the poles at the end of this century. Finally, we observed a specific gene repertoire and expression level variations potentially explaining its ecological success in low-iron and low-nitrate environments. Collectively, these findings reveal the ecological importance of P. calceolata and lay the foundation for a global scale analysis of the adaptation and acclimation strategies of this small phytoplankton in a changing environment.

Abstract: International audience; Anion recognition represents an intense area of research in supramolecular chemistry. The last two decades have been marked by spectacular advances in the design of new anion receptors. Moreover, the development of approaches combining experimental and theoretical studies has proved to be particularly relevant, allowing a better understanding and rationalization of the phenomena involved in anion complexation processes. In this context, the combination of weak interactions within the same receptor and their synergistic effects, called cooperativity, has attracted increasing interest within the scientific community. This review focuses on the combination of anion-π and hydrogen bonds and the emerging concept of cooperativity. The most relevant anion-π donor families are presented. The concept of cooperativity is illustrated using the most recent examples present in current literature.

Abstract: Partial resistance in plants generally exerts a low selective pressure on pathogens, and thus ensuring their durability in agrosystems. However, little is known about the effect of partial resistance on the molecular mechanisms of pathogenicity, a knowledge that could advance plant breeding for sustainable plant health. Here we investigate the gene expression of Phytophthora capsici during infection of pepper (Capsicum annuum L.), where only partial genetic resistance is reported, using Illumina RNA-seq. Comparison of transcriptomes of P. capsici infecting susceptible and partially resistant peppers identified a small number of genes that redirected its own resources into lipid biosynthesis to subsist on partially resistant plants. The adapted and non-adapted isolates of P. capsici differed in expression of genes involved in nucleic acid synthesis and transporters. Transient ectopic expression of the RxLR effector genes CUST_2407 and CUST_16519 in pepper lines differing in resistance levels revealed specific host-isolate interactions that either triggered local necrotic lesions (hypersensitive response or HR) or elicited leave abscission (extreme resistance or ER), preventing the spread of the pathogen to healthy tissue. Although these effectors did not unequivocally explain the quantitative host resistance, our findings highlight the importance of plant genes limiting nutrient resources to select pepper cultivars with sustainable resistance to P. capsici.

Abstract: Marine plankton mitigate anthropogenic greenhouse gases, modulate biogeochemical cycles, and provide fishery resources. Plankton is distributed across a stratified ecosystem of sunlit surface waters and a vast, though understudied, mesopelagic ‘dark ocean’. In this study, we mapped viruses, prokaryotes, and pico-eukaryotes across 32 globally-distributed cross-depth samples collected during the Tara Oceans Expedition, and assessed their ecologies. Based on depth and O2 measurements, we divided the marine habitat into epipelagic, oxic mesopelagic, and oxygen minimum zone (OMZ) eco-regions. We identified specific communities associated with each marine habitat, and pinpoint environmental drivers of dark ocean communities. Our results indicate that water masses primarily control mesopelagic community composition. Through co-occurrence network inference and analysis, we identified signature communities strongly associated with OMZ eco-regions. Mesopelagic communities appear to be constrained by a combination of factors compared to epipelagic communities. Thus, variations in a given abiotic factor may cause different responses in sunlit and dark ocean communities. This study expands our knowledge about the ecology of planktonic organisms inhabiting the mesopelagic zone.

Mobilisant 2 200 salariés, la grève dure sept semaines et remet en cause, pour une période brève, la puissance et l’autorité des patrons, de l’État, voire des militants communistes à la tête de la mairie de Douarnenez et en position d’intermédiaires syndicaux dans la grève. Durant le conflit, les revendications, la place des femmes, la souveraineté des assemblées plénières de grévistes, les prérogatives du comité de grève contrôlant l’activité productive du port, la forme des manifestations, les blocages de routes expriment la profondeur de la révolte. L’effectivité de la participation ouvrière dans la direction du mouvement contribue à définir la nature du conflit, dont les dimensions politiques se conjuguent avec une organisation consciente de la grève. At the end of November 1924, a strike broke out among the women of the Douarnenez canning industry in Finistère. Mobilizing 2200 workers, the strike lasted seven weeks and challenged, for a brief period, the power and authority of the bosses, the state and even the Communist militants at the head of the Douarnenez town hall and in the position of union intermediaries in the strike. During the conflict, the demands, the place of women, the sovereignty of the plenary assemblies of strikers, the prerogatives of the strike committee controlling the productive activity of the port, the form of the demonstrations and the road blockades all expressed the depth of the revolt. The effectiveness of the workers' participation in the direction of the movement contributes to define the nature of the conflict whose political dimensions were combined with a conscious organization of the strike.

Abstract: International audience; Depuis les travaux de Mayer, Davis et Schoorman (1995), la recherche s’accorde à reconnaître qu’une personne est digne de confiance lorsqu’elle est capable, intègre et bienveillante. Si la question de la capacité, et des moyens de la renforcer, a très largement travaillée par les ressources humaines, et si le coeur du concept d’intégrité a été éclairé récemment (Jacquinot, 2021), celui de bienveillance est souvent réduit à un affect et la complémentarité entre intégrité et bienveillance fait encore l’objet d’un large questionnement. Le présent travail apporte des réponses en mobilisant la pensée d’Aristote transcrite dans les livres V et VIII de l’Ethique à Nicomaque.

Abstract: PurposeThe current study aims to investigate the relation between corporate social responsibility (CSR) and information asymmetry, as well as the moderating effect of board characteristics (gender diversity, size and independence) on this relationship.Design/methodology/approachThis paper uses a panel data regression analysis with the system generalized method of moments (SGMM) estimator of nonfinancial French firms included in the SBF 120 index. The environmental and social disclosure scores are collected from the Bloomberg database, while financial data are collected from the FactSet database.FindingsThe empirical results demonstrate that environmental disclosure has a positive impact on the level of information asymmetry, while social disclosure has no effect on the information environment. Gender diversity and board independence negatively impact the opacity index, while board size has a positive effect. The presence of women in board composition has a substitution effect on the relationship between environmental disclosure and information asymmetry. There is no moderating effect of board size on the association between CSR disclosure and information asymmetry. However, the proportion of independent female directors and board independence operates as substitutes to social disclosure on reducing information asymmetry.Research limitations/implicationsAlthough the models include the most common control variables used in the literature, they omit some variables. Second, the results should be interpreted with caution and should not be generalized to the entire stock market since the sample is based on large French companies.Practical implicationsThe results of this study may be of interest to managers, investors and French market authorities since France is characterized by highly developed laws and reforms in the area of CSR. In addition, the paper leads to a better understanding of how women on the board, in particular, independent female directors, affect the relationship between CSR disclosure and information asymmetry. This could be of interest to French authorities, which has encouraged the appointment of women through the adoption of the Copé–Zimmermann law.Originality/valueFirst, to the best of the authors' knowledge, this is the first study to explore the moderating effect of board characteristics on the relationship between CSR and information asymmetry. Second, unlike previous studies using individual proxies to measure information asymmetry, the authors favor the opacity index of Andersonet al.(2009). They calculate this index by including a fifth individual measure, namely, share price volatility. The opacity index better describes the information environment of companies than individual measures since it reflects the perceptions of investors and analysts together.

Abstract: International audience; We introduce binacox, a prognostic method to deal with the problem of detecting multiple cut-points per feature in a multivariate setting where a large number of continuous features are available. The method is based on the Cox model and combines one-hot encoding with the binarsity penalty, which uses total-variation regularization together with an extra linear constraint, and enables feature selection. Original nonasymptotic oracle inequalities for prediction (in terms of Kullback-Leibler divergence) and estimation with a fast rate of convergence are established. The statistical performance of the method is examined in an extensive Monte Carlo simulation study, and then illustrated on three publicly available genetic cancer data sets. On these high-dimensional data sets, our proposed method outperforms state-of-the-art survival models regarding risk prediction in terms of the C-index, with a computing time orders of magnitude faster. In addition, it provides powerful interpretability from a clinical perspective by automatically pinpointing significant cut-points in relevant variables.

Abstract: International audience; Phosphoglycerate mutases (PGAMs) catalyse the reversible isomerisation of 3-phosphoglycerate and 2-phosphoglycerate, a step of glycolysis. PGAMs can be sub-divided into 2,3-biphosphoglycerate dependent (dPGAM) and independent (iPGAM) enzymes. In plants, phosphoglycerate isomerisation is carried out by cytosolic iPGAM. Despite its crucial role in catabolism, little is known about post-translational modifications of plant iPGAM. In Arabidopsis thaliana, phosphoproteomics analyses have previously identified a iPGAM phosphopeptide where serine 82 is phosphorylated. Here, we show that this phosphopeptide is less abundant in dark-adapted compared to illuminated Arabidopsis leaves. In silico comparison of iPGAM protein sequences and 3D-structural modelling of AtiPGAM2 based on non-plant iPGAM enzymes suggest a role for phosphorylated serine in the catalytic reaction mechanism. This is confirmed by the activity (or the lack thereof) of mutated recombinant Arabidopsis iPGAM2 forms, affected in different steps of the reaction mechanism. We thus propose that the occurrence of the S82-phosphopeptide reflects iPGAM2 steady-state catalysis. Based on this assumption, the metabolic consequences of a higher iPGAM activity in illuminated versus darkened leaves are discussed.

Abstract: Trees are long-lived organisms that continuously adapt to their environments, a process in which epigenetic mechanisms are likely to play a key role. Via down regulation of the chromatin remodeler DECREASED IN DNA METHYLATION 1 (DDM1) in poplar (Populus tremula 9 Populus alba) RNAi lines, we examined how DNA methylation coordinates genomic and physiological responses to moderate water deficit.We compared the growth and drought response of two RNAi-ddm1 lines to wild-type (WT) trees under well-watered and water deficit/rewatering conditions, and analyzed their methylomes, transcriptomes, mobilomes and phytohormone contents in the shoot apical meristem. The RNAi-ddm1 lines were more tolerant to drought-induced cavitation but did not differ in height or stem diameter growth. About 5000 differentially methylated regions were consistently detected in both RNAi-ddm1 lines, colocalizing with 910 genes and 89 active transposable elements. Under water deficit conditions, 136 differentially expressed genes were found, including many involved in phytohormone pathways ; changes in phytohormone concentrations were also detected. Finally, the combination of hypomethylation and drought led to the mobility of two transposable elements. Our findings suggest major roles for DNA methylation in regulation of genes involved in hormone-related stress responses, and the maintenance of genome integrity through repression of transposable elements.

We characterized two related proteins with contrasting impact on ethylene production, ACC oxidase 1 (SlACO1) and SlE8. We found SlACO1 and SlE8 to be highly expressed during fruit ripening. To identify loss-of-function alleles, we analysed the tomato genetic diversity but we did not find any natural mutations impairing the function of these proteins. We also found the two loci evolving under purifying selection. To engineer hypomorphic alleles, we used TILLING (target-induced local lesions in genomes) to screen a tomato ethylmethane sulfonate-mutagenized population. We found 13 mutants that we phenotyped for ethylene production, shelf-life, firmness, conductivity, and soluble solid content in tomato fruits. The data demonstrated that slaco1-1 and slaco1-2 alleles could be used to improve fruit shelf-life, and that sle8-1 and sle8-2 alleles could be used to accelerate ripening. This study highlights further the importance of SlACO1 and SlE8 in ethylene production in tomato fruit and how they might be used for post-harvest fruit preservation or speeding up fruit maturation.

Abstract: International audience; The liver is a complex organ composed of several cell types organized hierarchically. Among these, liver sinusoidal endothelial cells (LSECs) are specialized vascular cells known to interact with hepatocytes and hepatic stellate cells (HSCs), and to be involved in the regulation of important hepatic processes in healthy and pathological situations. Protocols for the differentiation of LSECs from human induced pluripotent stem cells, hiPSCs, have been proposed and in-depth analysis by transcriptomic profiling of those cells has been performed. In the present work, an extended analysis of those cells in terms of proteome and metabolome has been implemented. The proteomic analysis confirmed the expression of important endothelial markers and pathways. Among them, the expression of patterns typical of LSECs such as PECAM1, VWF, LYVE1, STAB1 (endothelial markers), CDH13, CDH5, CLDN5, ICAM1, MCAM-CD146, ICAM2, ESAM (endothelial cytoskeleton), NOSTRIN, NOS3 (Nitric Oxide endothelial ROS), ESM1, ENG, MMRN2, THBS1, ANGPT2 (angiogenesis), CD93, MRC1 (mannose receptor), CLEC14A (C-type lectin), CD40 (antigen), and ERG (transcription factor) was highlighted. Besides, the pathway analysis revealed the enrichment of the endocytosis, Toll-like receptor, Nod-like receptor, Wnt, Apelin, VEGF, cGMP-PCK, and PPAR related signaling pathways. Other important pathways such as vasopressin regulated water reabsorption, fluid shear stress, relaxin signaling, and renin secretion were also highlighted. At confluence, the metabolome profile appeared consistent with quiescent endothelial cell patterns. The integration of both proteome and metabolome datasets revealed a switch from fatty acid synthesis in undifferentiated hiPSCs to a fatty oxidation in LSECs and activation of the pentose phosphate pathway and polyamine metabolism in hiPSCs-derived LSECs. In conclusion, the comparison between the signature of LSECs differentiated following the protocol described in this work, and data found in the literature confirmed the particular relevance of these cells for future in vitro applications.

Abstract: International audience; Modulation and fine-tuning of the strength of weak interactions to bind anions are described in a series of synthetic receptors. The general design of the receptors includes both a urea motif and a tetrazine motif. The synthetic sequence towards three receptors is detailed. Impacts of H-bond strength and linker length between urea and tetrazine on chloride complexation are studied. Binding properties of the chloride anion are examined in both the ground and excited states using a panel of analytical methods (NMR spectroscopy, mass spectrometry, UV/Visible spectroscopies, and fluorescence). A ranking of the receptors by complexation strength has been determined, allowing a better understanding of the structure-properties relationship on these compounds.

Abstract: Objectives EUCAST changed the definition of the ‘intermediate’ (I) category in 2019, now defined as ‘susceptible, increased exposure’. This new definition could lead to an increased prescription of antibiotics still reported as ‘S’, compared with those now reported as ‘I’. The objective of this study was to evaluate the influence of this definition on the use of overly broad-spectrum antibiotics for the treatment of infections caused by WT Pseudomonas aeruginosa. Methods A retrospective observational multicentre study was conducted, involving five hospitals. Two 15 month study periods were defined, before and after the implementation of the new definition. All patients with an infection caused by WT P. aeruginosa treated by β-lactams were included. The main endpoint was the proportion of patients treated by an overly broad-spectrum antibiotic treatment by meropenem or ceftolozane/tazobactam. Results Two hundred and ninety-one patients were included. No difference between groups was found, in terms of infection, microbiology or demographic characteristics. Two overly broad-spectrum antibiotic treatments by meropenem or ceftolozane/tazobactam were observed in Period 1 (1.2%), versus 13 in Period 2 (10.8%; P Conclusions This new definition can cause a negative impact on the use of overly broad-spectrum antibiotic treatment due to misunderstanding by clinicians. Its successful implementation requires adaptation of software for reporting antibiotic susceptibility, a sustained strong information campaign by microbiologists and support by an antimicrobial stewardship team.

Abstract: International audience; In this paper we present the modeling of an airship with a load suspended by a flexible heavy cable. The objective is to determine a control law that allows to stabilize the machine and its load under the effect of a gust of wind. In order to overcome the great inertia of the airship and the impossibility of using the accelerations of the latter to attenuate the oscillations of the suspended load, a new actuator was proposed at the exit of the cable in order to control its oscillations, namely a motorized cardan joint. The methodology of the study consisted of extracting a reduced dynamic model, which allowed us to build a robust control vector in order to stabilize the suspended load. The analysis of the stability of the zero dynamics of the system was carried out by means of the phase portraits of Poincaré. Numerical simulations were carried out and demonstrated the interest of our approach.

Abstract: International audience; Dichloromethane (DCM, methylene chloride) is a toxic halogenated volatile organic compound massively used for industrial applications, and consequently often detected in the environment as a major pollutant. DCM biotransformation suggests a sustainable decontamination strategy of polluted sites. Among methylotrophic bacteria able to use DCM as a sole source of carbon and energy for growth, Methylorubrum extorquens DM4 is a longstanding reference strain. Here, the primary 5′-ends of transcripts were obtained using a differential RNA-seq (dRNA-seq) approach to provide the first transcription start site (TSS) genome-wide landscape of a methylotroph using DCM or methanol. In total, 7231 putative TSSs were annotated and classified with respect to their localization to coding sequences (CDSs). TSSs on the opposite strand of CDS (antisense TSS) account for 31% of all identified TSSs. One-third of the detected TSSs were located at a distance to the start codon inferior to 250 nt (average of 84 nt) with 7% of leaderless mRNA. Taken together, the global TSS map for bacterial growth using DCM or methanol will facilitate future studies in which transcriptional regulation is crucial, and efficient DCM removal at polluted sites is limited by regulatory processes.
