Abstract

International audience; PremiseThe conifer Geinitzia reichenbachii was a common member of the Cretaceous Laurasian floras. However, the histology of G. reichenbachii leafy axes was never described in detail, and our knowledge of its paleoecology remains very limited. Using new and exquisitely preserved silicified material from the Upper Cretaceous of western France, we describe G. reichenbachii from the gross morphology to the cellular scale, then discuss paleoecological and taphonomical implications.MethodsWe examined specimens from two localities in western France (Claix and Moragne) using propagation phase-contrast X-ray synchrotron microtomography.ResultsThe cuticle and the inner tissues of leafy axes are preserved in three dimensions. Epidermis, hypodermis, palisade parenchyma, spongy parenchyma, transfusion tracheids, and most of tissues of the vascular cylinder are clearly discernible. The numerous resin ducts are sometimes filled by persisting resin. Additionally, surfaces of some leaves preserved drops and flows of resin.ConclusionsDepositional environmental context combined with histological features of G. reichenbachii suggest that this conifer was adapted to a range of marginal-littoral ecosystems including those open to the sea paleoenvironments and innermost ones influenced by strong continental inputs. Geinitzia reichenbachii was adapted to withstand intense sunlight, hot temperatures coupled with salty sea wind, and dry conditions. The frequent amber-bearing beds in the Cretaceous from western France with Geinitzia as a main component of the associated floras raises the possibility of a role for Geinitzia in the production of the “Charentese amber”.

Abstract

International audience; Stresses associated with changes in diet or environmental disturbances are common situations that fish encounter during their lifetime. The stability and ease of measuring microRNAs (miRNAs) present in biological fluids make these molecules particularly interesting biomarkers for non-lethal assessment of stress in animals. Rainbow trout were exposed for four weeks to abiotic stress (moderate hypoxia) and/or nutritional stress (a high-carbohydrate/low-protein diet). Blood plasma and epidermal mucus were sampled at the end of the experiment, and miRNAs were assessed using small RNA sequencing. We identified four miRNAs (miR-122-5p, miR-184-3p, miR-192-5p and miR-194a-5p) and three miRNAs (miR-210-3p, miR-153a-3p and miR-218c-5p) that accumulated in response to stress in blood plasma and epidermal mucus, respectively. In particular, the abundance of miR-210-3p, a hypoxamiR in mammals, increased strongly in the epidermal mucus of rainbow trout subjected to moderate hypoxia, and can thus be considered a relevant biomarker of hypoxic stress in trout. We explored the contribution of 22 tissues/organs to the abundance of circulating miRNAs (c-miRNAs) in blood plasma and epidermal mucus influenced by the treatments. Some miRNAs were tissue-specific, while others were distributed among several tissues. Some c-miRNAs (e.g., miR-210-3p, miR184-3p) showed similar variations in both tissues and fluids, while others showed an inverse trend (e.g., miR-122-5p) or no apparent relationship (e.g. miR-192-5p, miR-194a-5p. Overall, these results demonstrate that c-miRNAs can be used as non-lethal biomarkers to study stress in fish. In particular, the upregulation of miR-210-3p in epidermal mucus induced by hypoxia demonstrates the potential of using epidermal mucus as a matrix for identifying non-invasive biomarkers of stress. This study provides information about the tissue sources of c-miRNAs and highlights the potential difficulty in relating variations in miRNA abundance in biological fluids to that in tissues.

Abstract

Rhizobium–legume nitrogen-fixing symbiosis involves the formation of a specific organ, the root nodule, which provides bacteria with the proper cellular environment for atmospheric nitrogen fixation. Coordinated differentiation of plant and bacterial cells is an essential step of nodule development, for which few transcriptional regulators have been characterized. Medicago truncatula ETHYLENE RESPONSE FACTOR REQUIRED FOR NODULE DIFFERENTIATION (MtEFD) encodes an APETALA2/ETHYLENE RESPONSIVE FACTOR (ERF) transcription factor, the mutation of which leads to both hypernodulation and severe defects in nodule development. MtEFD positively controls a negative regulator of cytokinin signaling, the RESPONSE REGULATOR 4 (MtRR4) gene. Here we showed that that the Mtefd-1 mutation affects both plant and bacterial endoreduplication in nodules, as well as the expression of hundreds of genes in young and mature nodules, upstream of known regulators of symbiotic differentiation. MtRR4 expressed with the MtEFD promoter complemented Mtefd-1 hypernodulation but not the nodule differentiation phenotype. Unexpectedly, a nonlegume homolog of MtEFD, AtERF003 in Arabidopsis (Arabidopsis thaliana), could efficiently complement both phenotypes of Mtefd-1, in contrast to the MtEFD paralog MtEFD2 expressed in the root and nodule meristematic zone. A domain swap experiment showed that MtEFD2 differs from MtEFD by its C-terminal fraction outside the DNA binding domain. Furthermore, clustered regularly interspaced short palindromic repeats-CRISPR associated protein 9 (CRISPR-Cas9) mutagenesis of MtEFD2 led to a reduction in the number of nodules formed in Mtefd-1, with downregulation of a set of genes, including notably NUCLEAR FACTOR-YA1 (MtNF-YA1) and MtNF-YB16, which are essential for nodule meristem establishment. We, therefore, conclude that nitrogen-fixing symbiosis recruited two proteins originally expressed in roots, MtEFD and MtEFD2, with distinct functions and neofunctionalization processes for each of them.

International audience; In perennial grasses, the reproductive development consists of major phenological stages which highly determine the seasonal variations of grassland biomass production in terms of quantity and quality. The reproductive development is regulated by climatic conditions through complex interactions subjected to high genetic diversity. Understanding these interactions and their impact on plant development and growth is essential to optimize grassland management and identify the potential consequences of climate change. Here, we review the main stages of reproductive development, from floral induction to heading, i.e., spike emergence, considering the effect of the environmental conditions and the genetic diversity observed in perennial grasses. We first describe the determinants and consequences of reproductive development at individual tiller scale before examining the interactions between plant tillers and their impact on grassland perenniality. Then, we review the available grassland models through their ability to account for the complexity of reproductive development and genetic × environmental interactions. This review shows that (1) The reproductive development of perennial grasses is characterized by a large intraspecific diversity which has the same order of magnitude as the diversity observed between species or environmental conditions. (2) The reproductive development is determined by complex interactions between the processes of floral induction and morphogenesis of the tiller. (3) The perenniality of a plant is dependent on the reproductive behavior of each tiller. (4) Published models only partly explain the complex interactions between morphogenesis and climate on reproductive development. (5) Introducing more explicitly the underlying processes involved in reproductive development in models would improve our ability to anticipate grassland behavior in future growth conditions.

Abstract

International audience; The characterization of the largest worldwide representative data set of apricot (Prunus armeniaca L.) germplasm was performed using molecular markers. Genetic diversity and structure of the cultivated apricot genetic resources were analyzed to decipher the history of diffusion of this species around the world. A common set of 25 microsatellite markers was used for genotyping a total of 890 apricot accessions in different collections from the center of origin to the more recent regions of apricot culture. Using a Bayesian model-based clustering approach, the apricot genotypes can be structured into five different genetic clusters (F-ST = 0.174), correlated with the geographical regions of origin of the accessions. Accessions from China and Central Asia were clustered together and exhibited the highest levels of diversity, confirming an origin in this region. A loss of genetic diversity was observed from the center of origin to both western and eastern zones of recent apricot culture. Altogether, our results revealed that apricot spread from China and Central Asia, defined as the center of origin, following three major diffusion routes with a decreasing gradient of genetic variation in each geographical group. The identification of specific alleles outside the center of origin confirmed the existence of different secondary apricot diversification centers. The present work provides more understanding of the worldwide history of apricot species diffusion as well as the field of conservation of the available genetic resources. Data have been used to define an apricot core collection based on molecular marker diversity which will be useful for further identification of genomic regions associated with commercially important horticultural traits through genome-wide association studies to sustain apricot breeding programs.

Abstract

International audience; Understanding the phenology of grasses is crucial for grasslands management as it strongly affects the quantity and quality of the biomass. The floral transition (FT) of the apex is induced by the exposition to specific conditions of temperature and photoperiod. Under non-inductive climatic conditions, the apex is vegetative and continually produces leaves. After exposure to cold temperature and long photoperiod, the meristem completes its floral induction and starts floral development. Heading date (HD) constitutes a main marker of the reproductive development. Determinants of the HD identified are the advancement of floral induction, the final number of leaves and the development of the leaves and the spike. The aim of the present work is to present a model of FT for perennial grasses driven by environmental conditions. The HD is simulated from the phyllochrone and organ size, given by a functional-structural model of ryegrass. Model behaviour is assessed using a dataset of HD.

Il décrit et définit les grands acteurs de la procédure d’évaluation des variétés de plantes de services : le CTPS, le GEVES et la CISPS. Il met en avant les difficultés rencontrées pour l’inscription au catalogue officiel des nouvelles variétés de plantes de services, en s’appuyant sur les expérimentations VATE locales de la station de Lusignan. Il montre également les subtilités administratives, réglementaires et techniques des nouveaux dossiers CTPS avec de nouvelles espèces à étudier, prenant la fétuque des près avec endophytes comme étude de cas. Enfin, il résume la communication faite autour des plantes de services pour diffuser les résultats et promouvoir ces cultures auprès des agriculteurs et du grand public, à travers notamment le salon des Culturales.

International audience; Variety registration of lucerne often fails because of the difficulty to pass the distinction test. This test is presently based on phenotypic traits. We proposed to evaluate if high-throughput genotyping of varieties with genotyping-by-sequencing markers could help to distinguish among 20 varieties and if the genetic distances were correlated to phenotypic ones. Genotyping was performed either on 40 individuals or on 4 bulks of 100 plants per variety. At the methodological level, we obtained a high number of polymorphic markers, a precise determination of the allele dosage on the individuals, a high reproducibility of allele frequency estimated on bulks and a good correlation between allele frequencies scored on individuals or on bulks. Each pair of the 20 varieties was significantly distinct when using the markers. In addition, the varieties grouped in a way that was consistent with their genetic origin. Genetic distances were correlated to phenotypic distances obtained from data collected in the distinction tests. We suggest that molecular markers could be useful to assist distinction tests during the official trials for registration of lucerne varieties.

International audience; Distinctness among lucerne varieties, evaluated in DUS tests, is a frequent obstacle to variety registration. Promising varieties may fail the distinction test despite an agronomic plus-value so the genetic progress is not delivered to farmers. The Genotyping By Sequencing (GBS) technology that enables to obtain a large number of markers on pools of individuals was used to evaluate the distinction among 20 lucerne varieties of the European Catalogue. A total of 40 000 polymorphic markers was obtained. All the varieties were statistically different from the others, and the structure was consistent with knowledge on the varieties. Perspectives to use GBS in DUS testing are given.

Abstract

International audience; Structural variation (SV) is a major driver of plant adaptation and genome evolution. It originates from transposable element insertion, as well as gene Copy Number (CNV) and Presence/Absence Variation (PAV). Maize is a crop species with a complex genome, and exhibits extensive SV among lines, as well as strong phenotypic differences. It is therefore a good model to explore the diverse molecular mechanisms leading to SV, and to investigate to what extent SV impacts phenotypic variation. Finally, the geographical origin of the different maize inbred lines is well described, allowing for linking SV to environmental adaptation. Here, we present whole genome assemblies from seven European and American maize lines of various geographical origins and phenotypes, and with contrasted genome size. This dataset allows unprecedented genome-wide comparisons and characterization of maize SV with high sequence accuracy, thus offering the opportunity to evaluate the prevalence of the molecular mechanisms underlying these variations and to characterize the features responsible for genome size variation. These seven maize lines together with B73 were cultivated under contrasted water conditions in the PHENOARCH phenotyping platform allowing precise characterization of growth and development together with precise measurements of environmental conditions. Thirteen different organs harvested at various developmental stages have been used for RNA-seq-based transcriptome analysis. This massive dataset will be used to evidence the possible role of SVs in quantitative responses to water deficit as well as the impact of SVs in gene regulation networks. Overall, this work will provide insights on the molecular origins and functional consequences of SV.

7 pages; International audience; Since the end of the XXth century, the palaeontological site of Archingeay-Les Nouillers (Charente-Maritime, western France) continued to afford Albian-Cenomanian (ca 93–113 Ma) plant meso- and macroremains. The successive excavations provided numerous specimens useful for the reconstruction of mid-Cretaceous coastal forest ecosystems. However, one of the most remarkable features of this locality is the diversity of plant preservations. Unique in Europe from this point of view, the fossiliferous site of Archingeay-Les Nouillers yielded compressions with or without cuticle, impressions/imprints, and silicified permineralizations/petrifications. The quality of the silicified tissues and the cell preservation allow this locality to be considered as a plant Konservat-Lagerstätte.; Depuis la fin du XXe siècle, le site paléontologique d’Archingeay-Les Nouillers (Charente-Maritime, Ouest de la France) continue de livrer des méso- et macrorestes végétaux d’âge Albien-Cénomanien (environ 93–113 millions d’années). Les campagnes de fouilles successives ont fourni de très nombreux spécimens utiles à la reconstitution des écosystèmes forestiers côtiers du Crétacé moyen. Néanmoins, un des caractères les plus remarquables de ce gisement est la diversité des types de préservations végétales. Unique en Europe de ce point de vue, le site fossilifère d’Archingeay-Les Nouillers a livré des compressions avec ou sans cuticule, des impressions/empreintes, ou encore des perminéralisations/pétrifications siliceuses. La qualité de préservation des tissus et cellules silicifiées permet de placer ce gisement dans les Konservat-Lagerstätten à plantes.

Abstract

International audience; Powdery mildew (Blumeria graminis f. sp. tritici) is one of the major diseases of wheat (Triticum aestivum). Adult plant resistance (APR) to powdery mildew is considered more durable than resistance conferred by major race-specific resistance genes. The objective of the present study was a better understanding of the genetic basis of APR in RE714 by means of QTL analysis of several resistance scores along the growing season. A population of 160 recombinant inbred lines obtained from the cross between RE714 and Hardi (susceptible) was assessed for APR under natural infection conditions during 3 years and a genetic map with whole genome coverage was developed with microsatellite and AFLP markers in this population. Two major QTL on chromosomes 5D and 6A were detected each year, and 6 minor QTL were detected only in 1 or 2 years. The QTL on chromosome 5D was detected during all the growing season each year and its R 2 value varied between 8.5 and 56.3%, whereas the QTL on chromosome 6A was detected at 1–4 scoring dates in the 3 years, and its R 2 value varied between 6.1 and 20.5%. The two QTL explained between 24.4 and 52.1% of the phenotypic variance for AUDPC, depending on the year. The models including QTL and cofactors in the composite interval mapping explained between 29 and 72% of the variance. The molecular markers linked to the two major QTL could be used in marker-assisted selection for adult plant resistance to powdery mildew.

Cette synthèse a pour objectif de faire le point sur les différents facteurs de variation de la teneur en matières azotées totales (MAT) du maïs plante entière, au moment de la récolte et après ensilage, à partir d’une banque de données (16 368 parcelles de maïs plante entière sur pied) constituée des résultats d’essais conduits par le GEVES en vue de l’inscription de nouvelles variétés de maïs au catalogue officiel, et des données de la littérature. Dans cette banque de données, la teneur en MAT des maïs sur pied est en moyenne de 7,7 + 0,7 % MS et varie entre 4,7 et 10,5 % MS. Elle est liée à la teneur en matières minérales, ce qui traduit le fait que l’absorption de l’azote est étroitement liée à celle des éléments minéraux. La teneur en MAT des maïs a tendance à diminuer avec le stade de maturité, de 0,20 point entre le stade pâteux-vitreux et le stade vitreux dur. La variabilité due aux conditions de milieu est supérieure à la variabilité due à la variété, soit un écart type moyen de 0,65 point et de 0,47 point respectivement. La relation négative observée dans un certain nombre d’analyses entre la teneur en MAT et le rendement traduit un effet négatif indirect de la sélection des maïs sur leur teneur en MAT, mais cette relation n’est pas systématique. Le processus d’ensilage entraîne une augmentation passive de la teneur en MAT de 0,40 point en moyenne avec de profondes modifications de la composition biochimique des protéines., We used a database containing 16 368 data obtained on whole plant maize, and literature data to evaluate the extent and factors of variation of crude protein (CP) content. CP content of whole plant maize at harvest was 7.7 + 0.7 % DM on average, and varied between 4.7 and 10.5 % DM. CP and ashes contents were significantly correlated, involving a relationship between N and minerals absorption by plant. These variations in CP content had several causes. It decreased to a low extent with maturity stage, 0.20 percentage unit between dough-flint and flint stages. Variability due to ’milieu’ (location x year) was higher than variability due to variety (standard deviation of 0.65 and 0.47 percentage unit, respectively). The negative relationship between CP content and yield sometimes observed might express an indirect negative effect of selection on maize CP content, but this relationship was not systematic. Ensiling involved a passive increase in CP content (0.40 percentage unit) with large modifications of biochemical composition of proteins.

Abstract

The chicken major histocompatibility complex ( MHC) genes are organized into two genetically independent clusters which both possess class I and class IIbeta genes: the classical B complex and the Restriction fragment pattern- Y ( Rfp-Y) complex. In this study, we have examined the role of Rfp-Y genes in transplantation immunity. For this we used three sublines, B19H1, B19H2 and B19H3, derived from a line fixed for B19. Southern blots, PCR-SSCP assays using primers specific for Rfp-Y genes, and Rfp-Y class I allele-specific sequencing show that the polymorphisms observed in B19H1, B19H2 and B19H3 are due to the presence of three different Rfp-Y haplotypes. The Rfp-Y class I ( YF) alleles in these three haplotypes are highly polymorphic, and RT-PCR shows that at least two YF loci are expressed in each subline. The three sublines show Rfp-Y-directed alloreactivity in that Rfp-Y-incompatible skin grafts are rejected within 15 days, a rate intermediate between that seen in B-incompatible rejection (7 days) and that observed for grafts within the sublines (20 days). We conclude that Rfp-Y has an intermediate role in allograft rejection, likely to be attributable to polymorphism at the class I loci within this region.

Abstract

At present, registration and protection of rapeseed (Brassica napus L.) cultivars relies on a small number of morphological traits, As the number of cultivars increases, the ability to distinguish them on the basis of these traits becomes more difficult. New descriptors like molecular markers are required to maintain the efficiency of registration testing. The objectives of this study were to (i) evaluate the discrimination power of 17 amplified fragment length polymorphism (AFLP) primer combinations tested on a collection of 83 spring and winter rapeseed cultivars, (ii) assess the structure of the genetic diversity revealed by AFLPs, and (iii) compare three genetic distance estimators (Jaccard, Sokal, and Michener and Sokal and Michener weighted by the PIC). A total of 324 polymorphic markers were scored, with an average of 19.1 markers per primer combination. The use of only two primer combinations was sufficient to identify uniquely all the cultivars. The analysis of the genetic structure of the diversity by cluster and principal component analyses and AMOVAs (analyses of molecular variance) clearly delineated three significant factors: the cultivar type (winter or spring). the country of origin (France or Germany), and the breeding company. The three measures of genetic distance were highly correlated (P < 0.001, r = 0.96-0.98) and led to similar groupings. AFLPs were shown to be a powerful method for identifying cultivars and analyzing the genetic structure of the diversity in rapeseed.

Additionally, we present and discuss efforts to reducethe trade-off between reliability and analysis costs.

Abstract

Unauthorised GMOs (UGM) and derived materials are not uncommon in products found on the European market. At present there is zero tolerance for UGM in the EU. In most documented cases, the UGM concentration relative to the product in which the UGM material is found, was low. Low level presence will always represent a challenge to analytically based detection, in particular if the UGM is obscured by other GM material. Analytical ad hoc implementation of the zero tolerance for particular UGMs in feed is described in Regulation (EU) No 619/2011. In Europe, GM detection is predominantly achieved with polymerase chain reaction (PCR) derived methods targeting the transgenic construct and insertion site DNA sequences. The increase in number and divergence of GMOs developed and commercialised has gradually forced the GM detection laboratories to rationalise their analytical work, and most laboratories now apply initial PCR based screenings followed by (when appropriate) more specific PCR based identification and quantification. The detection of any GM is dependent on availability of suitable detection method(s) and control materials to verify the performance of the method(s). Other information, e.g. describing the novel trait, introduced genetic elements, etc. may also facilitate detection, verification and identification of the GM. For UGM, this is a major challenge, and the GMOs are therefore classified into four knowledge groups in the present document. This classification may facilitate stakeholder communication and decision making in analytical laboratories. A decision tree is presented, summarising the recommended principles of GM and UGM detection. Notably, the state-of-the-art of GMO analysis is not static, and it is expected that the guidelines and recommendations presented in this document will have to be modified on a regular basis. Finally, the document highlights a number of R&D priorities and points out the need for reinforced information sharing at the global level.

Abstract

Fruit growing occupies 139 794 ha in France, ie 1% of the French agricultural area. Apple orchards are the most important with 31.8% of the whole fruit growing area (44 547 ha), and peach orchards the second one with 11.3% (16 587 ha). Level 1 describes reasoned chemical protection, which is used on the largest part of the French fruit growing area. For this level, the total TFI (Treatment Frequency Index) varies from 35 to 43 in apple orchards depending on production region, and from 18 to 28 in peach orchards. The key apple pests and diseases are codling moth (Cydia pomonella) mainly in South-East of France and scab (Venturia inaequalis) mostly in Western France. The main pests in peach orchards are aphids (Myzus persicae and other species), oriental moth (Cydia molesta) and powdery mildew (Sphaerotheca pannosa). Mating disruption against codling moth is implemented on 50% of the French apple and pear orchards, and allows to save 5 treatments in an average-pressure year. Using scab-resistant varieties can help reducing TFI between 4 and 9 points in level 2b. Association of these 2 control methods in level 2c provides a total TFI from 25 to 34 depending on the apple production region. The total TFI for organic apple orchards is close to 26. Thanks to the combination of direct alternative control methods and prophylaxy, the use of early and less susceptible varieties and codling moth proof hail-nets, the total TFI could noticeably decrease, however without reaching a decrease of 50%. Moreover, apple scab resistance has already been overcome in some regions, and mating disruption is only efficient on large size orchards. A global fruit culture TFI decrease could only be obtained through new quality standards for fruits, or an intense re-design of the French orchard.; L’arboriculture fruitière représente 1% de la SAU française, soit 139 794 ha. Les vergers de pommes de table occupent 31.8% de cette surface (44 547ha) et ceux de pêchers 11.3% (16 587 ha). Le niveau 1, qualifiant l’agriculture raisonnée appliquée dans la majorité des vergers français, se traduit par des IFT totaux variant de 35 à 43 selon les bassins de production pour la pomme de table, et de 18 à 28 pour la pêche. Les principales pressions pour le pommier sont le carpocapse (Cydia pomonella), plus particulièrement dans le Sud-Est et la tavelure (Venturia inaequalis) plus particulièrement dans l’Ouest. Pour la pêche, il s’agit des pucerons (Myzus persicae et autres), de la tordeuse orientale (Cydia molesta) et de l’oïdium (Sphaerotheca pannosa). L’utilisation de la confusion sexuelle contre le carpocapse des pommes (qualifiant ici le niveau 2a) peut faire économiser 5 traitements en situation de pression moyenne. Elle est d’ores et déjà appliquée sur 50% du verger national de pommiers-poiriers. L’utilisation de variétés résistantes à la tavelure peut diminuer l’IFT de 4 à 9 points dans le niveau 2b. L’association de ces 2 techniques alternatives dans le niveau 2c permet d’obtenir des IFT totaux de 25 à 34 selon les bassins de production de pomme. En agriculture biologique (niveau 3), les IFT totaux s’élèvent à environ 26. Grâce à l’association de ces techniques alternatives à de la prophylaxie, à l’utilisation de variétés précoces et peu sensibles, à la pose de filets sur les vergers, ces IFT totaux pourraient sensiblement diminuer, sans toutefois atteindre une baisse de 50% par rapport au niveau 1. De plus, il faut tenir compte du fait que la résistance à la tavelure est déjà contournée dans certains bassins, et que la confusion sexuelle n’est pas efficace sur toutes les configurations de vergers. Une diminution de l’IFT global lié à l’arboriculture pourrait passer par l’adoption de nouvelles normes qualitatives des fruits, ou par une profonde re-conception du verger français.

Abstract

International audience; Powdery mildew (Blumeria graminis f. sp. tritici) is one of the major diseases of wheat (Triticum aestivum). The objective of the present study was to describe the quantitative resistance to powdery mildew of the winter wheat line RE714 at the seedling stage and to identify microsatellite markers tightly linked to the RE714 resistance QTL, which could be used in marker-assisted selection. A population of 160 recombinant inbred lines obtained from the cross between RE714 (resistant) and Hardi (susceptible) was genotyped with microsatellite and AFLP markers. Fifteen powdery mildew isolates were used to test the resistance of these lines at the seedling stage. QTL analysis enabled us to identify three major QTLs controlling powdery mildew resistance in RE714: a QTL located on chromosome 2A, corresponding to the Pm4b gene, explaining 76-93% of the phenotypic variance for resistance to six isolates; two QTLs located on chromosomes 5D and 6A, each explaining 20-67%of the phenotypic variance for resistance tofive isolates.AminorQTLfor resistance to four of the six isolates revealing Pm4b was detected in the same region as the 5D QTL. Other minor QTLs were detected on chromosomes 2A and 6B, explaining, respectively, 10.9 and 11.5% of the phenotypic variance for resistance to isolate 96-27. The maps around the three major QTLs were enriched with microsatellite markers that could be used in marker-assisted selection of these QTLs.

Abstract

International audience; Among fruit species, apricot (Prunus armeniaca L.) is characterized by a large increase in the number of registered and/or granted cultivars over the last decades, and by an effective interest at the European level. At the same time, an evolution concerning fruit quality attributes was observed globally among registered cultivars. In order to characterize this tendency, 172 cultivars in collection were observed. Representative fruit samples have been collected and their quality attributes such as fruit weight, firmness, colour, soluble solids content, and titratable acidity were considered. The impacts of genetic and geographic origin factors were evaluated. A significant increase in fruit firmness and colour, as well as a decrease in fruit acidity, was observed. No evolution on sugar content was noticed.

Abstract

In order to explore genetic variability of wool production and other quantitative traits, an 8-cohort divergent selection experiment for total fleece weight (TFW) was carried out in French Angora rabbits. Studies were made on the wool production of 669 female rabbits born between 1994 and 2001 and having produced wool from the third to 12th harvests. The aim of the selection experiment was to obtain two divergent lines (low and high) on total fleece weight. The studied traits included total fleece weight, weight of the two qualities of wool (WAJ1 and WAW1), homogeneity (HOM), live body weight at ages of 4 (LW4), 8 (LW8), 12 (LW12), 16 (LW16), and 20 (LW20) weeks and then 9 weeks before each harvest (9LW). A preliminary analysis of non-genetic factors was done with the GLM procedure. The genetic parameters and genetic trends were analysed using a BLUP animal model. Heritability estimates for TFW, WAJ1, WAW1, HOM, LW4, LW8, LW12, LW16, LW20 and 9LW were 0.38, 0.30, 0.10, 0.06, 0.30, 0.09, 0.14, 0.32, 0.39 and 0.45, respectively. Genetic and phenotypic correlations between TFW and WAJ1 were high (0.98 ± 0.01 and 0.89 ± 0.01, respectively). There was a low genetic correlation between TFW and 9LW (0.26 ± 0.12). After eight cohorts of selection, the divergence between the lines was approximately three genetic standard deviations. Selection for total fleece weight had a generally beneficial effect on fleece quality.

Abstract

International audience; The aim of this study, led by the GEVES (Research and Control Group for Varieties and Seeds), was to suggest indicators to assess the diversity available to farmers since the French Official Catalogue for Plant Varieties and Species was initiated. The largest datasets of 1990 inbred maize lines and 578 pea lines from the last 50 years were analysed using morphological and enzymatic parameters. Lines were grouped into three to five periods. Genetic diversity was estimated in each period from morphological and enzymatic markers by computing numerous indices, such as the number of classes of scores for each characteristic, allelic richness or genetic diversity index (H (e) ). Population differentiation parameters (G(ST), G(ST)', F-ST, Q(ST)) were also estimated between periods. While genetic diversity computed from distinction, uniformity, stability traits was more marked for maize (0.66) than for garden peas (0.35) or feed peas (0.29), the opposite trend was observed with enzymes, resulting in a genetic diversity of 0.43, 0.35 and 0.22 for garden peas, feed peas and maize, respectively. However, no significant changes in genetic diversity were observed over time, and genetic differentiation was slight between periods. All our results demonstrated that no significant reduction in the diversity available to farmers had been observed since initiation of the French Catalogue. The H (e) was a good indicator providing a quantitative estimate of genetic diversity, but it should be interpreted alongside a more precise indicator such as allelic richness or the number of classes for morphological characteristics
