BRCA1 and BRCA2 are tumour suppressor genes whose mutant phenotypes predispose to breast and ovarian cancer. Screening for mutations in these genes is now standard practice for hereditary breast and ovarian cancer (HBOC) cases in Europe, and permits medical follow-up and genetic counselling adapted to the needs of individuals in such families. Currently, most laboratories performing diagnostic analysis of the BRCA genes use PCR of exons and intron-exon boundaries coupled to a pre-screening step to identify anomalous amplicons. The techniques employed for the detection of mutations and SNPs have evolved over time and vary in sensitivity, specificity and cost-effectiveness. As a variant for pre-screening techniques, we chose the recently developed Surveyor® heteroduplex cleavage method as a sensitive and specific technique to reveal anomalous amplicons of the BRCA genes, using only basic laboratory equipment and agarose gel electrophoresis. Here we present the detection of either mutations or SNPs within the BRCA1 exon 7, using heteroduplex analysis (HA) by mismatch-specific endonuclease, confirmed by dideoxy sequencing.

Barley is an important crop for both human and animal food use because of its high starch seed content. Germination, as a process of developing a new plant, or as part of making alcoholic beverages, make use of starch stored in barley seeds as food for the embryo. Amylases, who broke down the starch, are among the most important enzymes, mostly from human point of view. Analyzing and understanding the whole starch mobilizing process by amylases could be of high value for both the agricultural processes and for biotechnological ones. The amylases from germinating barley seeds have been investigated by electrophoresis for the presence of different isoenzymic forms. Both α and β amylases have been investigated and the results shows that there is more than one isoform in both types of amylases. [ABSTRACT FROM AUTHOR]

Prospective study aimed at establishing the incidence of sepsis with oro-maxillofacial portal of entry and of the causal relationship between the disease and the incriminated risk factors (factors in the living environment, collectivity, occupational environment, behavioral factors). The study group included 200 patients admitted to the Oro-Maxillofacial Surgery and ENT Clinics of the Iasi "St. Spiridon" Emergency Hospital in the interval 2012-2015. S. aureus was the main causal agent incriminated in the development of oro-dental sepsis, about ½ of the isolates strains being methicillin-resistant. Age under 50 years, male gender, urban, immunosuppression, recent medical history (previous hospitalization and antibiotic therapy) were significantly correlated with the oral-maxillofacial involvement, which draws attention on the outpatient follow-up of moderate and severe oral infections. The insidious onset described in 54.6% of patients did not raise the suspicion of a potentially life-threatening disease, such as sepsis, but the severe respiratory (43.1%) and neuromeningeal manifestations (25.7%) contributed to the early seeking of expert advice from an infectious diseases specialist. Over 92% of the study patients were at high risk for staphylococcal infection with multidrug-resistant (MDR) strains, significantly more common in men and in patients in whom MRSA was identified as the sepsis pathogen. Depending on the MDR agent involved in the causation of oral-maxillofacial sepsis, the risk of developing severe infections is attributable to methicillin-resistant Streptococcus mutans, Streptococcus anginosus, S. epidermidis, P. aeruginosa, Klebsiella, β-hemolytic streptococcus, and Streptococcus salivarius. Experimental investigation focused on influence of different CoFeO4 nanoparticles concentrations on soluble protein and electrophoretic pattern of Phanerochaete chrysosporium fungus. Number of electrophoretic fraction is higher in older mycelium P. chrysosporium 14-day-old compared to that 7-day-old. Moreover, there is an increase in staining intensity of polypeptides of mycelium 14-day-old fact confirmed also by the higher amount of soluble protein. According to the World Health Organization, cervical cancer ranks second in cancer in women, after breast cancer, as it represents 44% of the total feminine genital cancers. Every year 460,000 women all over the world are diagnosed with invasive cervical cancers and 200,000 women die by this illness. According to the data supplied by the Center for Health Statistics in Romania, over 66% of the new cases of cancer detected are between 35 and 55 years old, and about 70% of the deaths caused by cervical cancer is recorded for the women between 45 and 70 years old. The study was performed on a number of 1885 women aged between 20 and 85 years old, detected with cervical neoplasia in different stages in the County Hospital Galaţi. The cases studied showed a prevalence of cervical cancer of 37.3%. The profile of the patient showed that they come mainly from the urban area (77.3%), that about 80% of these women have a reduced educational level, and 23,6% are unemployed. The detected risk factors are: age, marital status, smoking, coffee consumption, alcohol, sex life debut, associated chronic diseases (high blood pressure, obesity), and contraceptives. In order to establish the diagnostic of cervical neoplasia the following examinations were performed: cytological smear, colposcopic examination, biopsy, histopathologic and complementary examinations. The comparison of the diagnostic methods allowed the doctors to establish how effective each of the conduct options is in the early detection of the serious changes, as these changes can progress towards cancer. They also wanted to see how acceptable every conduct option for the patients is; what is the cost/efficiency ratio for each option. The diagnostic certainty was accomplished through a histo-pathological examination of the material obtained by biopsy in comparison with colposcopy, with an accuracy of the method of 50.1%. We consider that patients with cervical cancer must not be abandoned, regardless of the stage of the cancer when they come to see the doctor, because sometimes, after a well-conducted complex treatment, the cases had a satisfactory evolution. The determination of enzyme activity in the organism's humours or in different tissues is an important means of detecting and following the evolution of numerous pathological states. In our study we have determined the activity of some cytolysis enzymes (lactate dehydrogenase, LDH; alanine-aminotransferase or glutamic-pyruvic transaminase, GPT; aspartate aminotransferase or glutamic-oxaloacetic transaminase, GOT ; creatine kinase, CK and gammaglutamyltranspeptidase, GGT) on patients with acute coronary syndromes : angina pectoris (AP), chronic painful ischemic heart disease (CPIHD), arterial hypertension (HTN), arterial hypertension with chronic painful ischemic heart disease (HTN+CPIHD) and acute myocardial infarction (AMI). The results show that the levels of activity of cytolysis enzymes are different in patients with acute coronary syndromes (ACS). The activity of LDH, GPT, GOT, CK and GGT do not have the same values for one and the same coronary disease measured in patients from different groups of age and sexes (male and female). Aim of this study was to establish a significant way to the prognosis improvement in trophoblastic pathology in patients with hydatidiform mole. Objective. A mole is a particularity of embryonic development where a pregnancy occurs paradoxically without an embryo (the mole pregnancy) and develops only its placental tissue. It is important to demonstrate that the clinical diagnostic should always be completed by a histopathological diagnostic and also by the description of the natural history of the disease until the time of diagnostic. Material and method. The cases included in the study (n=45) were hospitalized in "Elena Doamna" Clinical Hospital Iaşi, in the period of time between 2008 and 2013, and diagnosed with hydatidiform mole. The study was a retrospective one, case-control type. Starting from the already diagnosed illness and following the clinical and paraclinical parameters outlined in literature, the objective of the study was to establish the main risk factors that trigger the molar pregnancy. Results. The epidemiological characteristics show the following main risk factors for a molar pregnancy: age over 30, urban area, tobacco and alcohol consumption. The mean values of βHCG decreased significantly after the hydatidiform mole was removed, from 26,624 to 9,859 mUI/ml (p<0.05).Conclusions. Every woman with a history of hydatidiform mole has an increased risk of developing carcinoma. After the complete or partial removal of the hydatidiform mole, it is necessary to monitor the values of βHCG twice a month until they stabilize. [ABSTRACT FROM AUTHOR]

Preeclampsia is an important problem of public health, leads among the main causes of mortality and morbidity worldwide. There are many patho-psychological abnormalities of preeclampsia, but its etiology is poorly known. Aim. The study intends to find out the possibilities and limitations in diagnosing and treating this disease. The objective of this study was to establish that the C reactive protein is an efficient marker of preeclampsia and it correlates significantly with the severity of this disease. Material and method. The patients were selected from the pregnant women who were hospitalized in "Cuza Vodă" Iaşi Clinical Hospital of Obstetrics and Gynecology (n=82), in the period of time between 2003 and 2012, with a gestational age of over 20 weeks, and who came here for a speciality consult. The study was retrospective and case-control type; it was focused on the clinical-progressive aspects of preeclampsia, by checking the correlation of inflammatory parameters with blood pressure for a lot of pregnant women with preeclampsia, in comparison with a lot of normal pregnant women. Results. The severe forms of preeclampsia can be found in older age (25-43 years old), with an average around 37 years old (p=0.02). According to the cases studied, the severity of preeclampsia was noticed for average values of blood pressure of about 155 mmHg (p<0.001). The average values of C reactive protein were significantly higher for women with severe preeclampsia (89.14 ± 51.31 mg/l) (p<0.001). Conclusions. The severe forms of preeclampsia represent a major cause for maternal morbidity and mortality, especially in the complicated cases with HELLP syndrome and acute pulmonary edema, cerebral hemorrhage, acute respiratory infections, cuoagulopathy. C-reactive protein (CRP) is an indirect risk factor in the acute coronary syndromes [2,13,21]. Its high levels may reflect one of the following cases: an inflammation of the coronary arteries as a response to the infectious agents; the severeness of the inflammatory response within the atherosclerotic vessels; inflammation expansion associated with myocardial ischemia; inflammation expansion associated with myocardial necrosis. Therefore, CRP indicates the cardiac lesions specific to various coronary heart diseases. The prognostic value of CRP augmentation appears to be more important in the patients affected by myocardial lesions [21]. Between 2009 and 2011 we monitored the clinical and paraclinical evolution of 109 patients for 24 months. Hormonal therapy in menopause proved its efficiency, especially for the improvement of the climacteric symptoms, as the long term effects on cardiovascular diseases is still being studied. The patients treated with Climara and Riselle showed, after 2 years of treatment, a libido and a decrease of hot flushes. No matter what the type of menopause you monitor, the LDL cholesterol shows a significant decrease for the lots that are treated with hormones. The average values of HDL cholesterol significantly increased for the patients who were under hormonal treatment. The average values of glycaemia decreased for all the lots under treatment. Our study observed the role that the n3 fat acids and the lignans (phytoestrogens) from the composition of flaxseed can have on the markers of endothelial dysfunction and on the metabolic lipid changes, under endocrine imbalance (surgical menopause) associated with a hyperlipidemic diet. The present work intends to determine the quantity of vWF antigen with the help of the immune-enzymatic "sandwich" technique for a lot of 48 female rats belonging to Wistar breed that got 3 types of diets: standard, hyperlipidemic and hyperlipidemic diet supplemented with lignans. The vWF antigen that is present in the sample gets attached from the anti vWF antibodies, at the surface of the microtiter plate. The sample concentration can be determined by extrapolation and by reporting it to the standard used. The ovariectomy, an experimental model of atherosclerosis that is induced by the estrogen deficiency, was characterized by the appearance of an endothelial dysfunction, which was shown by the increased values of vWF. The results of the present experimental research regarding the effects of an adjuvant therapy, that prevents the appearance of the endothelial dysfunction by nutritive supplements requires the study to be continued on humans, too, because of the multiple advantages of using flaxseeds. One of the most important roles of hormone replacement therapy is to remove the symptoms of menopause, especially the vaso-motor and urogenital tract appeared immediately after oophorectomy. The decreased risks of cardiovascular diseases are the long-term benefits. The cardiovascular protective effect of estrogen occurs by different mechanisms, all leading to reduction or slowing atherosclerotic processes and this improves cardiovascular performance.
