1. Clinical Evaluation of the Torq Zero Delay Centrifuge System for Decentralized Blood Collection and Stabilization

Abstract

Blood sample collection and rapid separation—critical preanalytical steps in clinical chemistry—can be challenging in decentralized collection settings. To address this gap, the Torq™ zero delay centrifuge system includes a lightweight, hand-portable centrifuge (ZDrive™) and a disc-shaped blood collection device (ZDisc™) enabling immediate sample centrifugation at the point of collection. Here, we report results from clinical validation studies comparing performance of the Torq System with a conventional plasma separation tube (PST). Blood specimens from 134 subjects were collected and processed across three independent sites to compare ZDisc and PST performance in the assessment of 14 analytes (K, Na, Cl, Ca, BUN, creatinine, AST, ALT, ALP, total bilirubin, albumin, total protein, cholesterol, and triglycerides). A 31-subject precision study was performed to evaluate reproducibility of plasma test results from ZDiscs, and plasma quality was assessed by measuring hemolysis and blood cells from 10 subject specimens. The ZDisc successfully collected and processed samples from 134 subjects. ZDisc results agreed with reference PSTs for all 14 analytes with mean % biases well below clinically significant levels. Results were reproducible across different operators and ZDisc production lots, and plasma blood cell counts and hemolysis levels fell well below clinical acceptance thresholds. ZDiscs produce plasma samples equivalent to reference PSTs. Results support the suitability of the Torq System for remotely collecting and processing blood samples in decentralized settings.

Abstract

We present the complete next-to-next-to-leading order (NNLO) pure pointlike QED corrections to lepton-proton scattering, including three-photon-exchange contributions, and investigate their impact in the case of the MUSE experiment. These corrections are computed with no approximation regarding the energy of the emitted photons and taking into account lepton-mass effects. We contrast the NNLO QED corrections to known next-to-leading order corrections, where we include the elastic two-photon exchange (TPE) through a simple hadronic model calculation with a dipole ansatz for the proton electromagnetic form factors. We show that, in the low-momentum-transfer region accessed by the MUSE experiment, the improvement due to more sophisticated treatments of the TPE, including inelastic TPE, is of similar if not smaller size than some of the NNLO QED corrections. Hence, the latter have to be included in a precision determination of the low-energy proton structure from scattering data, in particular for electron-proton scattering. Abstract

We present the first calculation of the complete set of NNLO QED corrections for muon-electron scattering. This includes leptonic, non-perturbative hadronic, and photonic contributions. All fermionic corrections as well as the photonic subset that only corrects the electron or the muon line are included with full mass dependence. The genuine four-point two-loop topologies are computed as an expansion in the small electron mass, taking into account both, logarithmically enhanced as well as constant mass effects using massification. A fast and stable implementation of the numerically delicate real-virtual contribution is achieved by combining OpenLoops with next-to-soft stabilisation. All matrix elements are implemented in the McMule framework, which allows for the fully-differential calculation of any infrared-safe observable. This calculation is to be viewed in the context of the MUonE experiment requiring a background prediction at the level of 10 ppm. Abstract

We present an improved theoretical prediction of the positron energy spectrum for the polarised Michel decay $\mu^+\to e^+ \nu_e\bar{\nu}_\mu$. In addition to the full next-to-next-to-leading order correction of order $\alpha^2$ in the electromagnetic coupling, we include logarithmically enhanced terms at even higher orders. Logarithms due to collinear emission are included at next-to-leading accuracy up to order $\alpha^4$. At the endpoint of the Michel spectrum, soft photon emission results in large logarithms that are resummed up to next-to-next-to-leading logarithmic accuracy. We apply our results in the context of the MEG II and Mu3e experiments to estimate the impact of the theory error on the branching ratio sensitivity for the lepton-flavour-violating decay $\mu^+\to e^+ X$ of a muon into an axion-like particle $X$., Comment: 37 pages, 14 figures, published version

Abstract

We provide an overview of the status of Monte-Carlo event generators for high-energy particle physics. Guided by the experimental needs and requirements, we highlight areas of active development, and opportunities for future improvements. Particular emphasis is given to physics models and algorithms that are employed across a variety of experiments. These common themes in event generator development lead to a more comprehensive understanding of physics at the highest energies and intensities, and allow models to be tested against a wealth of data that have been accumulated over the past decades. A cohesive approach to event generator development will allow these models to be further improved and systematic uncertainties to be reduced, directly contributing to future experimental success. Event generators are part of a much larger ecosystem of computational tools. They typically involve a number of unknown model parameters that must be tuned to experimental data, while maintaining the integrity of the underlying physics models. Making both these data, and the analyses with which they have been obtained accessible to future users is an essential aspect of open science and data preservation. Abstract

McMule is a framework for fully differential higher-order QED calculations of scattering and decay processes involving leptons. It keeps finite lepton masses, which regularises collinear singularities. Soft singularities are treated with dimensional regularisation and using FKS$^\ell$ subtraction. We describe the implementation of the framework in Fortran 95, list the processes that are currently implemented, and give instructions on how to run the code. In addition, we present new phenomenological results for muon-electron scattering and lepton-proton scattering, including the dominant NNLO corrections. Abstract

We present the heavy-to-light form factors with two different non-vanishing masses at next-to-next-to-leading order and study its expansion in the small mass. The leading term of this small-mass expansion leads to a factorized expression for the form factor. The presence of a second mass results in a new feature, in that the soft contribution develops a factorization anomaly. This cancels with the corresponding anomaly in the collinear contribution. Abstract

We provide an overview of the status of Monte-Carlo event generators for high-energy particle physics. Guided by the experimental needs and requirements, we highlight areas of active development, and opportunities for future improvements. Particular emphasis is given to physics models and algorithms that are employed across a variety of experiments. These common themes in event generator development lead to a more comprehensive understanding of physics at the highest energies and intensities, and allow models to be tested against a wealth of data that have been accumulated over the past decades. A cohesive approach to event generator development will allow these models to be further improved and systematic uncertainties to be reduced, directly contributing to future experimental success. Event generators are part of a much larger ecosystem of computational tools. They typically involve a number of unknown model parameters that must be tuned to experimental data, while maintaining the integrity of the underlying physics models. Making both these data, and the analyses with which they have been obtained accessible to future users is an essential aspect of open science and data preservation. It ensures the consistency of physics models across a variety of experiments.

16. Fully differential NLO predictions for rare and radiative lepton decays

Abstract

We present a general purpose, parton-level Monte Carlo program for the calculation of the radiative ($L\to\nu\bar\nu l+\gamma$) and rare ($L\to\nu\bar\nu l'+ l^+l^-$) muon and tau decays at NLO in the effective Fermi theory. In the case of muon, these processes are irreducible Standard Model backgrounds to searches for lepton flavour violation at the PSI experiments MEG and Mu3e as they become indistinguishable from the corresponding signals when the neutrinos carry little energy. Furthermore, we argue that fully differential NLO corrections are very important for the analysis of measurements aiming at the percent level or better. This is especally true if very stringent phase-space cuts are applied. To illustrate this, we use a tension between BaBar's recent measurement of the radiative tau decay and the Standard Model prediction as an example of such an analysis. Finally, we present the branching ratios of the rare tau decay $\tau\to\nu\bar\nu l' l^+l^-$ at NLO. Using the automation program GoSam, fully differential NLO corrections were obtained for the rare decay of the muon $\mu\to e\nu\bar\nu ee$. This process is an important Standard Model background to searches of the Mu3e collaboration for lepton-flavour violation, as it becomes indistinguishable from the signal $\mu\to 3e$ if the neutrinos carry little energy. With our NLO program we are able to compute the branching ratio as well as custom-tailored observables for the experiment. Abstract

We present the complete next-to-next-to-leading order (NNLO) pure pointlike QED corrections to lepton-proton scattering, including three-photon-exchange contributions, and investigate their impact in the case of the MUSE experiment. These corrections are computed with no approximation regarding the energy of the emitted photons and taking into account lepton-mass effects. We contrast the NNLO QED corrections to known next-to-leading order corrections, where we include the elastic two-photon exchange (TPE) through a simple hadronic model calculation with a dipole ansatz for the proton electromagnetic form factors. We show that, in the low-momentum-transfer region accessed by the MUSE experiment, the improvement due to more sophisticated treatments of the TPE, including inelastic TPE, is of similar if not smaller size than some of the NNLO QED corrections. Hence, the latter have to be included in a precision determination of the low-energy proton structure from scattering data, in particular for electron-proton scattering. For muon-proton scattering, the NNLO QED corrections are considerably smaller.

25. Vascular mimetics based on microfluidics for imaging the leukocyte--endothelial inflammatory response.

Abstract

We describe the development, validation, and application of a novel PDMS-based microfluidic device for imaging leukocyte interaction with a biological substrate at defined shear force employing a parallel plate geometry that optimizes experimental throughput while decreasing reagent consumption. The device is vacuum bonded above a standard 6-well tissue culture plate that accommodates a monolayer of endothelial cells, thereby providing a channel to directly observe the kinetics of leukocyte adhesion under defined shear flow. Computational fluid dynamics (CFD) was applied to model the shear stress and the trajectory of leukocytes within the flow channels at a micron length scale. In order to test this model, neutrophil capture, rolling, and deceleration to arrest as a function of time and position was imaged in the transparent channels. Neutrophil recruitment to the substrate proved to be highly sensitive to disturbances in flow streamlines, which enhanced the rate of neutrophil-surface collisions at the entrance to the channels. Downstream from these disturbances, the relationship between receptor mediated deceleration of rolling neutrophils and dose response of stimulation by the chemokine IL-8 was found to provide a functional readout of integrin activation. This microfluidic technique allows detailed kinetic studies of cell adhesion and reveals neutrophil activation within seconds to chemotactic molecules at concentrations in the picoMolar range.

Such benefits are thought to emerge partly from scaling effects that increase efficiency as group size increases.sup.3-5. In social insects and other taxa, the benefits of group living have been proposed to stem from division of labour.sup.5-8, which is characterized by between-individual variability and within-individual consistency (specialization) in task performance. However, at the onset of sociality groups were probably small and composed of similar individuals with potentially redundant--rather than complementary--function.sup.1. Self-organization theory suggests that division of labour can emerge even in relatively small, simple groups.sup.9,10. However, empirical data on the effects of group size on division of labour and on fitness remain equivocal.sup.6. Here we use long-term automated behavioural tracking in clonal ant colonies, combined with mathematical modelling, to show that increases in the size of social groups can generate division of labour among extremely similar workers, in groups as small as six individuals. These early effects on behaviour were associated with large increases in homeostasis--the maintenance of stable conditions in the colony.sup.11--and per capita fitness. Our model suggests that increases in homeostasis are primarily driven by increases in group size itself, and to a smaller extent by a higher division of labour. Our results indicate that division of labour, increased homeostasis and higher fitness can emerge naturally in social groups that are small and homogeneous, and that scaling effects associated with increasing group size can thus promote social cohesion at the incipient stages of group living.Experimental data from, and mathematical modelling of, colonies of the clonal raider ant Ooceraea biroi demonstrate that increases in group size generate division of labour among similar individuals, increased homeostasis and higher colony fitness.

Abstract

We present the first calculation of the complete set of NNLO QED corrections for muon-electron scattering. This includes leptonic, non-perturbative hadronic, and photonic contributions. All fermionic corrections as well as the photonic subset that only corrects the electron or the muon line are included with full mass dependence. The genuine four-point two-loop topologies are computed as an expansion in the small electron mass, taking into account both, logarithmically enhanced as well as constant mass effects using massification. A fast and stable implementation of the numerically delicate real-virtual contribution is achieved by combining OpenLoops with next-to-soft stabilisation. All matrix elements are implemented in the McMule framework, which allows for the fully-differential calculation of any infrared-safe observable. This calculation is to be viewed in the context of the MUonE experiment requiring a background prediction at the level of 10 ppm. Our results thus represent a major milestone towards this ambitious precision goal.

Abstract

In this manuscript, we report the outcome of the topical workshop: paving the way to alternative NNLO strategies (https://indico.ific.uv.es/e/WorkStop-ThinkStart_3.0), by presenting a discussion about different frameworks to perform precise higher-order computations for high-energy physics. These approaches implement novel strategies to deal with infrared and ultraviolet singularities in quantum field theories. A special emphasis is devoted to the local cancellation of these singularities, which can enhance the efficiency of computations and lead to discover novel mathematical properties in quantum field theories.

Abstract

BACKGROUND: Semen quality assessment in population-based epidemiologic studies presents logistical and financial challenges due to reliance on centralised laboratory semen analysis. The Trak Male Fertility Testing System is an FDA-cleared and validated at-home test for sperm concentration and semen volume, with a research use only sperm motility test. Here we evaluate the Trak System’s overall utility among men participating in Pregnancy Study Online (PRESTO), a web-based study of North American couples planning pregnancy. METHODS: US male participants aged ≥21 years with ≤6 months of pregnancy attempt time at study enrolment were invited to participate in the semen testing substudy after completing their baseline questionnaire. Consenting participants received a Trak Engine (battery-powered centrifuge) and two test kits. Participants shared their test results via smartphone images uploaded to online questionnaires. Data were then linked with covariate data from the baseline questionnaire. RESULTS: Of the 688 men invited to participate, 373 (54%) provided consent and 271 (73%) completed at least one semen test result. The distributions of semen volume, sperm concentration, motile sperm concentration, total sperm count, and total motile sperm count were similar to 2010 World Health Organization (WHO) semen parameter data of men in the general population. The overall usability score for the Trak System was 1.4 on a 5-point Likert scale (1 = Very Easy, 5 = Difficult), and 92% of participants believed they performed the test correctly and received an accurate result. Lastly, men with higher motile sperm count were more likely to report feeling “at ease” or “excited” following testing, while men with low motile sperm count were more likely to report feeling “concerned” or “frustrated.” Overall, 91% of men reported they would like to test again. CONCLUSIONS: The Trak System provides a simple and potentially cost-effective means of measuring important semen parameters and may be useful in population-based epidemiologic fertility studies.

Abstract

Blood sample collection and rapid separation—critical preanalytical steps in clinical chemistry—can be challenging in decentralized collection settings. To address this gap, the Torq™ zero delay centrifuge system includes a lightweight, hand-portable centrifuge (ZDrive™) and a disc-shaped blood collection device (ZDisc™) enabling immediate sample centrifugation at the point of collection. Here, we report results from clinical validation studies comparing performance of the Torq System with a conventional plasma separation tube (PST). Blood specimens from 134 subjects were collected and processed across three independent sites to compare ZDisc and PST performance in the assessment of 14 analytes (K, Na, Cl, Ca, BUN, creatinine, AST, ALT, ALP, total bilirubin, albumin, total protein, cholesterol, and triglycerides). A 31-subject precision study was performed to evaluate reproducibility of plasma test results from ZDiscs, and plasma quality was assessed by measuring hemolysis and blood cells from 10 subject specimens. The ZDisc successfully collected and processed samples from 134 subjects. ZDisc results agreed with reference PSTs for all 14 analytes with mean % biases well below clinically significant levels. Results were reproducible across different operators and ZDisc production lots, and plasma blood cell counts and hemolysis levels fell well below clinical acceptance thresholds. ZDiscs produce plasma samples equivalent to reference PSTs. Results support the suitability of the Torq System for remotely collecting and processing blood samples in decentralized settings.

Abstract

In this manuscript, we report the outcome of the topical workshop: paving the way to alternative NNLO strategies (https://indico.ific.uv.es/e/WorkStop-ThinkStart_3.0), by presenting a discussion about different frameworks to perform precise higher-order computations for high-energy physics. These approaches implement novel strategies to deal with infrared and ultraviolet singularities in quantum field theories. A special emphasis is devoted to the local cancellation of these singularities, which can enhance the efficiency of computations and lead to discover novel mathematical properties in quantum field theories.
