Abstract

The study of starburst and AGN feedback is crucial for understanding the regulation of star formation and the evolution of galaxies across cosmic time. Arp 220, the closest ultraluminous infrared galaxy (ULIRG), is in an advanced phase of a major merger with two distinct nuclei, and shows evidence of multi-phase (molecular, ionised, neutral) and multi-scale (from < 0.1 to > 5 kpc) outflows. Therefore, it represents an ideal system for investigating outflow mechanisms and feedback phenomena in detail. Using new JWST NIRSpec IFU observations, we investigate the spatially resolved gaseous (in both ionized and hot molecular phases) and stellar kinematics in the innermost 1 kpc. We decouple the different gas kinematic components through multi-Gaussian fitting, identifying distinct multi-phase outflows associated with the two nuclei, with velocities up to $\sim$ 1000 km/s. We compute the mass ($\sim 10^7$ M$_\odot$), mass outflow rate ($\sim 20$ M$_\odot$/yr) and energetics ($\dot E_{out}\sim 10^{42}$ erg/s) for each outflow, finding that the ionized and hot molecular outflowing gas contribute around 2-30% to the total mass and the energy of the outflows, as inferred from the combination of multi-wavelength information. We discuss the possible origin of the outflows, finding no compelling evidence to prefer a starburst or AGN driven scenario. Regardless of their nature, outflows in Arp 220 propagate in multiple directions from parsec to kiloparsec scales, potentially impacting a significant portion of the host galaxy. This contrasts with isolated systems where outflows typically follow a more collimated path, and do not affect the interstellar medium throughout the entire galaxy. Abstract

The Euclid ERO programme targeted the Perseus cluster of galaxies, gathering deep data in the central region of the cluster over 0.7 square degree, corresponding to approximately 0.25 r_200. The data set reaches a point-source depth of IE=28.0 (YE, JE, HE = 25.3) AB magnitudes at 5 sigma with a 0.16" and 0.48" FWHM, and a surface brightness limit of 30.1 (29.2) mag per square arcsec. The exceptional depth and spatial resolution of this wide-field multi-band data enable the simultaneous detection and characterisation of both bright and low surface brightness galaxies, along with their globular cluster systems, from the optical to the NIR. This study advances beyond previous analyses of the cluster and enables a range of scientific investigations summarised here. We derive the luminosity and stellar mass functions (LF and SMF) of the Perseus cluster in the Euclid IE band, thanks to supplementary u,g,r,i,z and Halpha data from the CFHT. We adopt a catalogue of 1100 dwarf galaxies, detailed in the corresponding ERO paper. We identify all other sources in the Euclid images and obtain accurate photometric measurements using AutoProf or AstroPhot for 138 bright cluster galaxies, and SourceExtractor for half a million compact sources. Cluster membership for the bright sample is determined by calculating photometric redshifts with Phosphoros. Our LF and SMF are the deepest recorded for the Perseus cluster, highlighting the groundbreaking capabilities of the Euclid telescope. Both the LF and SMF fit a Schechter plus Gaussian model. The LF features a dip at M(IE)=-19 and a faint-end slope of alpha_S = -1.2 to -1.3. The SMF displays a low-mass-end slope of alpha_S = -1.2 to -1.35. Abstract

The current standard model of cosmology successfully describes a variety of measurements, but the nature of its main ingredients, dark matter and dark energy, remains unknown. Euclid is a medium-class mission in the Cosmic Vision 2015-2025 programme of the European Space Agency (ESA) that will provide high-resolution optical imaging, as well as near-infrared imaging and spectroscopy, over about 14,000 deg^2 of extragalactic sky. In addition to accurate weak lensing and clustering measurements that probe structure formation over half of the age of the Universe, its primary probes for cosmology, these exquisite data will enable a wide range of science. This paper provides a high-level overview of the mission, summarising the survey characteristics, the various data-processing steps, and data products. 4. Growing a nuclear star cluster from star formation and cluster mergers: The JWST NIRSpec view of NGC 4654

Abstract

We present a detailed study of the centre of NGC4654, a Milky Way-like spiral galaxy in the Virgo cluster that has been reported to host a double stellar nucleus, thus promising a rare view of ongoing star cluster infall into a galaxy nucleus. Analysing JWST NIRSpec integral-field spectroscopic data and Hubble Space Telescope imaging of the inner 330 $\times$ 330 pc, we find that the nucleus harbours in fact three massive star clusters. Maps of infrared emission lines from NIRSpec show different morphologies for the ionised and molecular gas components. The emission from molecular hydrogen gas is concentrated at the NSC location, while emission from hydrogen recombination lines is more extended beyond the central cluster. The velocity fields of both gas and stars indicate that the three clusters are part of a complicated dynamical system, with the NSC having an elevated velocity dispersion in line with its high stellar mass. To investigate the stellar populations of the three clusters in more detail, we use surface brightness modelling to measure their fluxes from ultraviolet to mid-infrared wavelengths and fit their spectral energy distributions (SEDs). Two of the clusters are UV-bright and well described by single stellar populations with young ages ($\sim$ 3 and 5 Myr) and low masses ($M_\ast \sim 4 \times 10^{4} - 10^{5} M_\odot$), whereas the central cluster is much more massive ($3 \times 10^7 M_\odot$), and cannot be fitted by a single stellar population. Instead, we find that a minor young population ($\sim$ 1 Myr) embedded in a dominant old population ($\sim$ 8 Gyr) is needed to explain its SED. Given its complex composition and the close proximity of two young star clusters that are likely to merge with it within a few hundred million years, we consider NGC4654 a unique laboratory to study NSC growth from both in-situ star formation and the infall of star clusters., Comment: Main text: 13 pages, 8 figures, accepted for publication in A&A

Abstract

Arp 220 is the nearest ULIRG; it shows evidence of 100 pc-scale molecular outflows likely connected with galaxy-scale outflows traced by ionised and neutral gas. The two highly obscured nuclei of Arp 220 are the site of intense star formation, with extreme star-formation rate surface densities (~ 10^3 Msun/yr/kpc2). Despite extensive investigations searching for AGN activity in the Arp 220 nuclei, direct evidence remains elusive. We present JWST/NIRSpec IFS observations covering the 0.9 - 5.1 um wavelength range of the innermost (5''x4'', i.e. 1.8x1.5 kpc) regions of Arp 220. The primary goal is to investigate the potential presence of AGN signatures in the nuclear regions by analysing the spectra extracted from circular apertures of radius 55 pc (0.15'') around each of the two nuclei. We identify ~ 70 ionised and ~ 50 molecular emission lines in the nuclear spectra of Arp 220; we use recombination line ratios to measure optical extinctions in the range AV ~ 11 - 14 mag. High ionisation lines are not detected, except the [Mg IV] line at 4.49 um which we interpret as due to shocks rather than to AGN ionisation. We identify broadening and multiple kinematic components in the HI and H2 lines caused by outflows and shocks, with velocities up to ~ 550 km/s. Significantly higher velocities (up to ~ 900 km/s) are detected in the off-nuclear regions; however, they do not conclusively represent evidence for AGN activity. Even with the unprecedented sensitivity of JWST/NIRSpec IFS, achieving an unambiguous identification or exclusion of the presence of an AGN in the Arp 220 system remains challenging, because of its extreme dust obscuration., Comment: submitted to A&A, comments welcome

Abstract

An increasing number of observations and simulations suggests that low-power (<10$^{44}$ erg s$^{-1}$) jets may be a significant channel of feedback produced by active galactic nuclei (AGN), but little is known about their actual effect on their host galaxies from the observational point of view. We targeted four luminous type 2 AGN hosting moderately powerful radio emission ($\sim$10$^{44}$ erg s$^{-1}$), two of which and possibly a third are associated with jets, with optical integral field spectroscopy observations from the Multi Unit Spectroscopic Explorer (MUSE) at the Very Large Telescope (VLT) to analyze the properties of their ionized gas as well as the properties and effects of ionized outflows. We combined these observations with Very Large Array (VLA) and e-MERLIN data to investigate the relations and interactions between the radio jets and host galaxies. We detected ionized outflows as traced by the fast bulk motion of the gas. The outflows extended over kiloparsec scales in the direction of the jet, when present. In the two sources with resolved radio jets, we detected a strong enhancement in the emission-line velocity dispersion (up to 1000 km s$^{-1}$) perpendicular to the direction of the radio jets. We also found a correlation between the mass and the energetics of this high-velocity dispersion gas and the radio power, which supports the idea that the radio emission may cause the enhanced turbulence. This phenomenon, which is now being observed in an increasing number of objects, might represent an important channel for AGN feedback on galaxies., Comment: 18 pages, 17 figures

Abstract

We present HOMERUN (Highly Optimized Multi-cloud Emission-line Ratios Using photo-ionizatioN), a new approach to modelling emission lines from photoionized gas that can simultaneously reproduce all observed line intensities from a wide range of ionization levels and with high accuracy. Our approach is based on the weighted combination of multiple single-cloud photoionization models and, contrary to previous works, the novelty of our approach consists in using the weights as free parameters of the fit and constraining them with the observed data. One of the main applications of HOMERUN is the accurate determination of gas-phase metallicities and we show that a critical point is to allow for a variation of the N/O and S/O abundance ratios which can significantly improve the quality of the fit and the accuracy of the results. Moreover, our approach provides a major improvement compared to the single-cloud, constant-pressure models commonly used in the literature. By using high-quality literature spectra of H ii regions where 10 to 20 emission lines (including several auroral lines) are detected with high signal-to-noise ratio, we show that all lines are reproduced by the model with an accuracy better than 10%. In particular, the model is able to simultaneously reproduce [O i]6300, 6363, [O ii]3726, 3729, [O iii]4959, 5007, [S ii]6717, 6731, and [S iii]9069, 9532 emission lines which, to our knowledge, is an unprecedented result. Finally, we show that the gas metallicities estimated with our models for HII regions in the Milky Way are in agreement with the stellar metallicities than the estimates based on the Te-method. Overall, our method provides a new accurate tool to estimate the metallicity and the physical conditions of the ionized gas. Abstract

Studying the feedback process of Active Galactic Nuclei (AGN) requires characterising multiple kinematical components, such as rotating gas and stellar disks, outflows, inflows, and jets. To compare the observed properties with theoretical predictions of galaxy evolution and feedback models and to assess the mutual interaction and energy injection rate into the interstellar medium (ISM), one usually relies on simplified kinematic models. These models have several limitations, as they often do not take into account projection effects, beam smearing and the surface brightness distribution of the emitting medium. Here, we present MOKA3D, an innovative approach to model the 3D gas kinematics from integral field spectroscopy observations. In this first paper, we discuss its application to the case of AGN ionised outflows, whose observed clumpy emission and apparently irregular kinematics are only marginally accounted for by existing kinematical models. Unlike previous works, our model does not assume the surface brightness distribution of the gas, but exploits a novel procedure to derive it from the observations by reconstructing the 3D distribution of emitting clouds and providing accurate estimates of the spatially resolved outflow physical properties (e.g. mass rate, kinetic energy). As an example, we demonstrate the capabilities of our method by applying it to three nearby Seyfert-II galaxies observed with MUSE at the VLT and selected from the MAGNUM survey, showing that the complex kinematic features observed can be described by a conical outflow with a constant radial velocity field and a clumpy distribution of clouds., Comment: 17 pages, 14 figures

Abstract

Abstract Background Factors to accurately stratify patients with early-stage non-small cell lung cancer (NSCLC) in different prognostic groups are still needed. This study aims to investigate 1) the prognostic potential of circulating cell-free (CF) and extracellular vesicles (EVs)-derived microRNA (miRNAs), and 2) their added value with respect to known prognostic factors (PFs). Methods The RESTING study is a multicentre prospective observational cohort study on resected stage IA-IIIA patients with NSCLC. The primary end-point was disease-free survival (DFS), and the main analyses were carried out separately for CF- and EV-miRNAs. CF- and EV-miRNAs were isolated from plasma, and miRNA-specific libraries were prepared and sequenced. To reach the study aims, three statistical models were specified: one using the miRNA data only (Model 1); one using both miRNAs and known PFs (age, gender, and pathological stage) (Model 2), and one using the PFs alone (Model 3). Five-fold cross-validation (CV) was used to assess the predictive performance of each. Standard Cox regression and elastic net regularized Cox regression were used. Results A total of 222 patients were enrolled. The median follow-up time was 26.3 (95% CI 25.4–27.6) months. From Model 1, three CF-miRNAs and 21 EV-miRNAs were associated with DFS. In Model 2, two CF-miRNAs (miR-29c-3p and miR-877-3p) and five EV-miRNAs (miR-181a-2-3p, miR-182-5p, miR-192-5p, miR-532-3p and miR-589-5p) remained associated with DFS. From pathway enrichment analysis, TGF-beta and NOTCH were the most involved pathways. Conclusion This study identified promising prognostic CF- and EV-miRNAs that could be used as a non-invasive, cost-effective tool to aid clinical decision-making. However, further evaluation of the obtained miRNAs in an external cohort of patients is warranted.

Abstract

The novel Gaia Multi Peak (GMP) technique has proven to be able to successfully select dual and lensed AGN candidates at sub-arcsec separations. Both populations are important because dual AGN represent one of the central, still largely untested, predictions of lamdaCDM cosmology, and compact lensed quasars allow to probe the central regions of the lensing galaxies. In this work, we present high spatial resolution spectroscopy of twelve GMP-selected systems. We use the the adaptive-optics assisted integral-field spectrograph MUSE at VLT to resolve each system and study the nature of each component. All the targets reveal the presence of two components confirming the GMP selection. We classify five targets as dual AGN, two as lensed systems, and five as a chance alignment of a star and and AGN. Having separations between 0.30" and 0.86", these dual and lensed systems are, to date, among the most compact ever discovered at z >0.3. Abstract

The Gaia-Multi-Peak (GMP) technique can be used to identify large numbers of dual or lensed AGN candidates at sub-arcsec separation, allowing us to study both multiple SMBHs in the same galaxy and rare, compact lensed systems. The observed samples can be used to test the predictions of the models of SMBH merging once 1) the selection function of the GMP technique is known, and 2) each system has been classified as dual AGN, lensed AGN, or AGN/star alignment. Here we show that the GMP selection is very efficient for separations above 0.15'' when the secondary (fainter) object has magnitude G<20.5. We present the spectroscopic classification of five GMP candidates using VLT/ERIS and Keck/OSIRIS, and compare them with the classifications obtained from: a) the near-IR colors of 7 systems obtained with LBT/LUCI, and b) the analysis of the total, spatially-unresolved spectra. We conclude that colors and integrated spectra can already provide reliable classifications of many systems. Finally, we summarize the confirmed dual AGNs at z>0.5 selected by the GMP technique, and compare this sample with other such systems from the literature, concluding that GMP can provide a large number of confirmed dual AGNs at separations below 7 kpc., Comment: 14 pages,A&A, in press

Abstract

Quasar feedback in the form of powerful outflows is invoked as a key mechanism to quench star formation in galaxies, although direct observational evidence is still scarce and debated. Here we present Early Release Science JWST NIRSpec IFU observations of the z=1.59 prototypical obscured Active Galactic Nucleus (AGN) XID2028: This target represents a unique test case for studying quasar feedback at the peak epoch of AGN-galaxy co-evolution because extensive multi-wavelength coverage is available and a massive and extended outflow is detected in the ionised and molecular components. With the unprecedented sensitivity and spatial resolution of the JWST, the NIRSpec dataset reveals a wealth of structures in the ionised gas kinematics and morphology that were previously hidden in the seeing-limited ground-based data. In particular, we find evidence of an interaction between the interstellar medium of the galaxy and the quasar-driven outflow and radio jet that produces an expanding bubble from which the fast and extended wind detected in previous observations emerges. The new observations confirm the complex interplay between the AGN jet, wind and the interstellar medium of the host galaxy, highlighting the role of low-luminosity radio jets in AGN feedback. Abstract

Background: The prevalence of pathogenic germline mutations in DNA damage repair (gDDR) genes in the Italian population is unknown. Objective: In this prospective multicenter cohort study, we evaluated the prevalence of gDDR alterations in the Italian population affected by metastatic prostate cancer (mPCa) and analyzed the impact on response to therapy, survival, and time to castration resistance. Design, setting, and participants: In an observational prospective trial, 300 consecutive Italian mPCa patients, enrolled in the Meet-Uro-10 trial from three academic Italian centers, were recruited between 2017 and 2019 and were screened for gDDR mutations in 107 genes. Outcome measurements and statistical analysis: The primary endpoint was to assess the prevalence of gDDR mutations in the Italian population of patients with mPCa. The secondary endpoints included the association of gDDR subgroups with metastatic onset, Gleason score, and time to castration resistance. Results and limitations: We identified 297 valuable patients. Forty-six patients had a pathogenic/likely pathogenic variant (15.5%, 95% confidence interval: 11.4–19.6): the more frequent was gBRCA2 found in nine cases (3%), followed by gATM in five cases (1.7%). In patients without mutations, longer median overall survival was observed with the sequence docetaxel-androgen receptor signaling inhibitor (ARSI) than with the sequence ARSI-docetaxel (87.9 vs 42 mo, p = 0.0001). In a univariate analysis, the median time to castration resistance in gDDR mutated patients was 19.8 mo, versus 23.7 mo in no mutated patients (p = 0.024). There were no associations of gDDR subgroups with metastatic onset and Gleason score ≥8. In our cohort, variants of unknown significance in gDDR genes were found in 80 patients and might have a prognostic relevance. Conclusions: The study reported the prevalence of gDDR in the Italian population. The presence of gBRCA2 mutations correlates with a shorter time to the onset of castration resistance disease. Patient summary: The prevalence of gBRCA2 in the Italian population is 3%, which is similar to that in the Spanish population, identifying similarities between people of the Western Mediterranean area.

(1) Background: BRAF mutations affect 4–5% of lung adenocarcinomas. This study aimed to analyze the clinicopathological features of lung carcinomas with BRAF mutations, focusing on V600E vs. non-V600E and the presence of co-mutations. (2) Methods: All BRAF-mutated lung carcinomas were retrieved from a molecular diagnostic unit (the reference unit for four different hospitals). The samples were analyzed using next-generation sequencing. Statistical analyses included log-rank tests for overall survival (OS) and progression-free survival (PFS). (3) Results: In total, 60 BRAF-mutated lung carcinomas were retrieved: 24 (40.0%) with V600E and 36 (60.0%) with non-V600E mutations, and 21 (35.0%) with other co-mutations and 39 (65.0%) with only BRAF mutations. Survival data were available for 54/60 (90.0%) cases. Targeted therapy was documented in 11 cases. Patients with V600E mutations exhibited a better prognosis than patients with non-V600E mutations (p = 0.008 for OS, p = 0.018 for PFS); this was confirmed in PFS (p = 0.036) when considering only patients who received no targeted therapy. Patients with co-mutations displayed no prognostic difference compared to patients carrying only BRAF mutations (p = 0.590 for OS, p = 0.938 for PFS). (4) Conclusions: BRAF-mutated lung carcinomas with V600E (40.0%) had a better prognosis than those without V600E. Concomitant co-mutations (35.0%) did not affect the prognosis.

Abstract

Introduction Chronic autoimmune (type 1 diabetes and coeliac disease) and metabolic/cardiovascular (type 2 diabetes, dyslipidaemia, hypertension) diseases are highly prevalent across all age ranges representing a major public health burden. Universal screening for prediction/early identification of these conditions is a potential tool for reducing their impact on the general population. The aim of this study is to assess whether universal screening using capillary blood sampling is feasible at a population-based level.Methods and analysis This is a low-risk interventional, single-centre, pilot study for a population-based screening programme denominated UNISCREEN. Participants are volunteers aged 1–100 who reside in the town of Cantalupo (Milan, Italy) undergoing: (1) interview collecting demographics, anthropometrics and medical history; (2) capillary blood collection for measurement of type 1 diabetes and coeliac disease-specific autoantibodies and immediate measurement of glucose, glycated haemoglobin and lipid panel by point-of-care devices; (3) venous blood sampling to confirm autoantibody-positivity; (4) blood pressure measurement; (5) fulfilment of a feasibility and acceptability questionnaire. The outcomes are the assessment of feasibility and acceptability of capillary blood screening, the prevalence of presymptomatic type 1 diabetes and undiagnosed coeliac disease, distribution of glucose categories, lipid panel and estimate of cardiovascular risk in the study population. Abstract

Abstract Bevacizumab (Bev) plus chemotherapy is a standard first-line treatment in metastatic colorectal cancer (mCRC), however to date no predictive factors of response have been identified. Results of our previous analysis on patients enrolled in a randomized prospective phase III multicenter study (ITACa study) showed a predictive value of Vascular Endothelial Growth Factor (VEGF) polymorphism (VEGF + 936), a 27-nucleotide variable number tandem repeat (VNTR) of the endothelial nitric oxide synthase (eNOS) gene and eNOS + 894 polymorphism. mCRC patients, treated with Bev plus chemotherapy, were included in this prospective validation trial. eNOS + 894G > T was analyzed by Real time PCR, while the eNOS VNTR and VEGF + 936C > T were determined by standard PCR and direct sequencing analysis. These polymorphisms were assessed in relation to progression-free survival (PFS), overall survival (OS) and objective response rate (ORR). These three polymorphisms were not predictive of PFS (p 0.91, 0.59 and 0.09, respectively), and OS (p 0.95, 0.32 and 0.46, respectively). Moreover, the haplotype analyses did not confirm what was found in our previous study; patients bearing a specific haplotype of eNOS had not significantly improved outcomes. This prospective study failed to validate the predictive impact of eNOS and VEGF polymorphisms for response to Bev plus first-line chemotherapy in mCRC patients.

Abstract

Abstract Resting heart rate is associated with cardiovascular diseases and mortality in observational and Mendelian randomization studies. The aims of this study are to extend the number of resting heart rate associated genetic variants and to obtain further insights in resting heart rate biology and its clinical consequences. A genome-wide meta-analysis of 100 studies in up to 835,465 individuals reveals 493 independent genetic variants in 352 loci, including 68 genetic variants outside previously identified resting heart rate associated loci. We prioritize 670 genes and in silico annotations point to their enrichment in cardiomyocytes and provide insights in their ECG signature. Two-sample Mendelian randomization analyses indicate that higher genetically predicted resting heart rate increases risk of dilated cardiomyopathy, but decreases risk of developing atrial fibrillation, ischemic stroke, and cardio-embolic stroke. We do not find evidence for a linear or non-linear genetic association between resting heart rate and all-cause mortality in contrast to our previous Mendelian randomization study. Systematic alteration of key differences between the current and previous Mendelian randomization study indicates that the most likely cause of the discrepancy between these studies arises from false positive findings in previous one-sample MR analyses caused by weak-instrument bias at lower P-value thresholds. The results extend our understanding of resting heart rate biology and give additional insights in its role in cardiovascular disease development.

Abstract

The real-world, retrospective, NEROnE registry investigated the impact of next-generation sequencing (NGS) in advanced non-small-cell lung cancer (NSCLC) patients (pts) at three oncology units in the north of Italy between January 2020 and December 2022. We focused on the clinical characterization and outcomes of NSCLC with rare molecular alterations: EGFR exon 20 insertion, non-activating EGFR mutations, BRAF V600E and non-V600, ROS1 and RET rearrangements, MET, ErbB2, and FGFR mutations. Overall, these represented 6.4% (62/970) of the pts analysed with NGS in the daily practice. The most heavily represented rare alterations were ROS1 rearrangement (15 pts—24%) and MET exon 14 skipping mutation (11 pts—18%). No associations were found with the demographic and clinical features. Forty-nine pts received targeted therapies, of which 38.8% were first- and 9.8% were second-line. The remaining pts received chemotherapy and/or immunotherapy. In terms of the clinical outcomes, although not statistically significant, a tendency toward shorter OS was seen when therapies other than specific targeted therapies were used (HR: 1.84, 95% CI: 0.79–4.33, p = 0.158). The pts with co-mutations (19.4%) seemed to receive an advantage from the front-line chemotherapy-based regimen. Finally, an NLR score (a well-known inflammatory index) ≥ 4 seemed to be related to shorter OS among the pts treated with immunotherapy alone or in combination with chemotherapy (HR: 2.83, 95% CI: 1.08–7.40, p = 0.033). Prospective evaluations need to be performed to clarify whether these indexes may help to identify patients with oncogene-addicted NSCLC who could benefit from immunotherapy.

Abstract

Among the many ice polymorphs, ice I, that is present in nature at ambient pressure, occurs with two different structures, i.e. the stable hexagonal (Ih) or the metastable cubic (Ic) one. An accurate analysis of cubic ice Ic was missing until the recent discovery of an easy route to obtain it in a structurally pure form. Here we report a Raman spectroscopy study of the transformation of metastable ice Ic into the stable form Ih, by applying a very slow temperature ramp to the ice Ic samples initially at 150 K. The thermal behavior of the spectroscopic features originating from the lattice and OH-stretching vibrational modes was carefully measured, identifying the thermodynamic conditions of the Ic-Ih transition and the dynamics of both structure with remarkable accuracy. Moreover, the comparison of two determinations of the transformation kinetics allowed also to provide an estimation of the activation energy for this transformation process.

Background: Validated predictors of sensitivity or resistance to Bevacizumab (Bev) are not available, and Inflammatory Indexes (IIs) has been reported to be useful prognostic factors in various malignant solid tumours, including metastatic colorectal cancer (mCRC). Objectives: To explore the prognostic value of IIs in mCRC patients treated with first-line chemotherapy plus Bev. Design: One hundred and eighty-two patients diagnosed with mCRC and treated with first line chemotherapy plus Bev were considered for this prospective non-pharmacological study. Neutrophil, lymphocyte, platelet, aspartate transaminase (AST) and lactate dehydrogenase (LDH) tests were carried out at baseline and before each treatment cycle, according to clinical practice. Methods: Pre-treatment Systemic Immune-inflammation Index (SII), Colon Inflammatory Index (CII) and Aspartate aminotransferase-Lymphocyte Ratio Index (ALRI) were evaluated to assess a correlation with progression-free survival (PFS) and overall survival (OS). Results: In the overall population, PFS and OS were lower in patients with high SII (HR 1.64, p = 0.006 and HR 1.75, p = 0.004, respectively) and high ALRI (HR 2.13, p = 0.001 and HR 1.76, p = 0.02, respectively), but no difference was detected according to CII value. The multivariate analysis confirmed both SII and ALRI as independent prognostic factors for PFS (HR 1.64 and 2.82, respectively) and OS (HR 1.65 and 2.12, respectively). Conclusion: Our results demonstrate and confirm that IIs, and in particular SII and ALRI, are easy to measure prognostic markers for patient candidates to first line chemotherapy plus Bev for mCRC.

Background: Cyclin-dependent kinase 4 and 6 (CDK4 and CDK6) inhibitors have changed the therapeutic management of hormone receptor-positive (HR+) metastatic breast cancer (mBC) by targeting the cell cycle machinery and overcoming endocrine resistance. However, a large number of patients present disease progression due to cancer cells resisting CDK4/6 inhibitors. Our research considers which clinicopathological characteristics could be useful in identifying patients who might respond to CDK4/6 inhibitors by analyzing a retrospective case series of patients with HR+ mBC who were treated with hormone therapy plus CDK4/6 inhibitors. Methods: Approximately 177 mBC patients were enrolled, of whom 66 were treated with CD4/6 inhibitors plus letrozole and 111 were treated with CDK4/6 inhibitors and fulvestrant. A multistate model was used. Results: A low body surface area and older age were associated with an increased risk of developing neutropenia. A high Ki67 index, the presence of visceral metastases, and not having previously undergone adjuvant chemotherapy were prognostic factors of disease progression/death. As expected, some of the neutropenic patients who had previously undergone multiple lines of treatment were at a higher risk of disease progression/death. Furthermore, neutropenia status was associated with a more than doubled risk of progression/death compared to patients without neutropenia (HR = 2.311; p = 0.025). Conclusions: Having identified certain factors that could be associated with the development of neutropenia and considering that neutropenia itself is associated with an increased risk of progression, we believe that the baseline characteristics should be taken into account to reduce cases of neutropenia and disease progression.

Immune checkpoint inhibitor (ICI) therapy has revolutionized the treatment of cancer, in particular lung cancer, while the introduction of predictive biomarkers from liquid biopsies has emerged as a promising tool to achieve an effective and personalized therapy response. Important progress has also been made in the molecular characterization of extracellular vesicles (EVs) and circulating tumor cells (CTCs), highlighting their tremendous potential in modulating the tumor microenvironment, acting on immunomodulatory pathways, and setting up the pre-metastatic niche. Surface antigens on EVs and CTCs have proved to be particularly useful in the case of the characterization of potential immune escape mechanisms through the expression of immunosuppressive ligands or the transport of cargos that may mitigate the antitumor immune function. On the other hand, novel approaches, to increase the expression of immunostimulatory molecules or cargo contents that can enhance the immune response, offer premium options in combinatorial clinical strategies for precision immunotherapy. In this review, we discuss recent advances in the identification of immune checkpoints using EVs and CTCs, their potential applications as predictive biomarkers for ICI therapy, and their prospective use as innovative clinical tools, considering that CTCs have already been approved by the Food and Drug Administration (FDA) for clinical use, but providing good reasons to intensify the research on both.

Abstract

Summary: Stress-related psychiatric disorders and the stress system show prominent differences between males and females, as well as strongly divergent transcriptional changes. Despite several proposed mechanisms, we still lack the understanding of the molecular processes at play. Here, we explore the contribution of cell types to transcriptional sex dimorphism using single-cell RNA sequencing. We identify cell-type-specific signatures of acute restraint stress in the paraventricular nucleus of the hypothalamus, a central hub of the stress response, in male and female mice. Further, we show that a history of chronic mild stress alters these signatures in a sex-specific way, and we identify oligodendrocytes as a major target for these sex-specific effects. This dataset, which we provide as an online interactive app, offers the transcriptomes of thousands of individual cells as a molecular resource for an in-depth dissection of the interplay between cell types and sex on the mechanisms of the stress response.

Abstract

Among the over eighteen different forms of water ice, only the common hexagonal phase and a cubic phase are present in nature on Earth. The existence of these two polytypes, almost degenerate in energy, represents one of the most important and unresolved topics in the physics of ice. It is now widely recognised that all the samples of "cubic ice" obtained so far are instead a stacking-disordered form of ice I (i.e. ice Isd), in which both hexagonal and cubic stacking sequences of hydrogen-bonded water molecules are present. Here we describe a new method to obtain cubic ice Ic in large quantities, and demonstrate its unprecedented structural purity from two independent neutron diffraction experiments performed on two of the leading neutron diffraction instruments in Europe.

Abstract

The electrocardiographic PR interval reflects atrioventricular conduction, and is associated with conduction abnormalities, pacemaker implantation, atrial fibrillation (AF), and cardiovascular mortality. Here we report a multi-ancestry (N = 293,051) genome-wide association meta-analysis for the PR interval, discovering 202 loci of which 141 have not previously been reported. Variants at identified loci increase the percentage of heritability explained, from 33.5% to 62.6%. We observe enrichment for cardiac muscle developmental/contractile and cytoskeletal genes, highlighting key regulation processes for atrioventricular conduction. Additionally, 8 loci not previously reported harbor genes underlying inherited arrhythmic syndromes and/or cardiomyopathies suggesting a role for these genes in cardiovascular pathology in the general population. We show that polygenic predisposition to PR interval duration is an endophenotype for cardiovascular disease, including distal conduction disease, AF, and atrioventricular pre-excitation. These findings advance our understanding of the polygenic basis of cardiac conduction, and the genetic relationship between PR interval duration and cardiovascular disease.

Abstract Stress exposure impairs brain structure and function, resulting in cognitive deficits and increased risk for psychiatric disorders such as depression, schizophrenia, anxiety and post-traumatic stress disorder. In particular, stress exposure affects function and structure of hippocampal CA1 leading to impairments in episodic memory. Here, we applied longitudinal deep-brain optical imaging to investigate the link between changes in activity patterns and structural plasticity of dorsal CA1 pyramidal neurons and hippocampal-dependent learning and memory in mice exposed to stress. We found that several days of repeated stress led to a substantial increase in neuronal activity followed by disruption of the temporal structure of this activity and spatial coding. We then tracked dynamics of structural excitatory connectivity as a potential underlying cause of the changes in activity induced by repeated stress. We thus discovered that exposure to repeated stress leads to an immediate decrease in spinogenesis followed by decrease in spine stability. By comparison, acute stress led to stabilization of the spines born in temporal proximity to the stressful event. Importantly, the temporal relationship between changes in activity levels, structural connectivity and activity patterns, suggests that loss of structural connectivity mediates the transition between increased activity and impairment of temporal organization and spatial information content in dorsal CA1 upon repeated stress exposure.

Biliary tract cancers (BTCs) have poor prognosis and limited therapeutic options. The impact of O6‐methylguanine‐DNA methyltransferase (MGMT) inactivation in advanced BTC patients is not established. We investigated the prevalence, prognostic, and predictive impact of MGMT inactivation in two multicenter cohorts. MGMT inactivation was assessed through PCR and immunohistochemistry (IHC) in an Italian cohort; the results were then externally validated using RNA sequencing (RNA‐seq) data from the BTC subcohort of the Molecularly Aided Stratification for Tumor Eradication Research (MASTER) precision oncology program of the National Center for Tumor Diseases Heidelberg and the German Cancer Consortium. Abstract

Non-small cell lung cancer (NSCLC) is one of the deadliest diseases worldwide. Tissue biopsy is the current gold standard for the diagnosis and molecular profiling of NSCLC. However, this approach presents some limitations due to inadequate tissue sampling, and intra- and intertumour heterogenicity. Liquid biopsy is a noninvasive method to determine cancer-related biomarkers in peripheral blood, and can be repeated at multiple timepoints. One of the most studied approaches to liquid biopsies is represented by circulating tumour cells (CTCs). Several studies have evaluated the prognostic and predictive role of CTCs in advanced NSCLC. Despite the limitations of these studies, the results of the majority of studies seem to be concordant regarding the correlation between high CTC count and poor prognosis in patients with NSCLC. Similarly, the decrease of CTC count during treatment may represent an important predictive marker of sensitivity to therapy in advanced NSCLC. Furthermore, molecular characterization of CTCs can be used to provide information on tumour biology, and on the mechanisms involved in resistance to targeted treatment. This review will discuss the current status of the clinical utility of CTCs in patients with advanced NSCLC, highlighting their potential application to prognosis and to treatment decision making.

Abstract

Background The introduction of targeted therapies for the treatment of BRAF-mutated metastatic melanoma was associated with different cutaneous adverse events (AEs). Objectives To describe the type, frequency and severity of cutaneous AEs related to vemurafenib; to understand the association between AEs and vemurafenib efficacy in terms of median overall survival (OS) and median progression-free survival (PFS); to identify molecular characteristics of long-term responders. Methods This observational, retrospective, monocentric study included all consecutive patients with unresectable stage III or stage IV melanoma and BRAF V600E mutation that started treatment with vemurafenib between May 2012 and May 2014. Results 62 patients with a median age of 56 years (range 26–82) were enrolled and received vemurafenib for a median period of 7.9 months (range 0.8–63.7). Among them, 45 patients presented at least one skin AE, 12 reduced the dosage due to cutaneous toxicity, and only one firstly reduced and after stopped the therapy. No specific molecular biomarkers were detected in long-term survivors. Conclusions Among long-term survivors, skin AEs seem to be less frequent and less severe. Results on multivariable analysis revealed that the presence of at least one G2 toxicity is a protective factor considering PFS, but not in terms of OS.

Abstract

Tumors are complex and heterogeneous diseases characterized by an intricate milieu and dynamically in connection with surrounding and distant tissues. In the last decades, great efforts have been made to develop novel preclinical models able to recapitulate the original features of tumors. However, the development of an in vitro functional and realistic tumor organ is still utopic and represents one of the major challenges to reproduce the architecture of the tumor ecosystem. A strategy to decrypt the whole picture and predict its behavior could be started from the validation of simplified biomimetic systems and then proceed with their integration. Variables such as the cellular and acellular composition of tumor microenvironment (TME) and its spatio-temporal distribution have to be considered in order to respect the dynamic evolution of the oncologic disease. In this perspective, we aim to explore the currently available strategies to improve and integrate in vitro and in vivo models, such as three-dimensional (3D) cultures, organoids, and zebrafish, in order to better understand the disease biology and improve the therapeutic approaches.

Subpopulations of neurons display increased activity during memory encoding and manipulating the activity of these neurons can induce artificial formation or erasure of memories. Thus, these neurons are thought to be cellular engrams. Moreover, correlated activity between pre- and postsynaptic engram neurons is thought to lead to strengthening of their synaptic connections, thus increasing the probability of neural activity patterns occurring during encoding to reoccur at recall. Therefore, synapses between engram neurons can also be considered as a substrate of memory, or a synaptic engram. One can label synaptic engrams by targeting two complementary, non-fluorescent, synapse-targeted GFP fragments separately to the pre- and postsynaptic compartment of engram neurons; the two GFP fragments reconstitute a fluorescent GFP at the synaptic cleft between the engram neurons, thereby highlighting synaptic engrams. In this work we explored a transsynaptic GFP reconstitution system (mGRASP) to label synaptic engrams between hippocampal CA1 and CA3 engram neurons identified by different Immediate-Early Genes: cFos and Arc. We characterized the expression of the cellular and synaptic labels of the mGRASP system upon exposure to a novel environment or learning of a hippocampal-dependent memory task. We found that mGRASP under the control of transgenic ArcCreERT2 labeled synaptic engrams more efficiently than when controlled by viral cFostTA, possibly due to differences in the genetic systems rather than the specific IEG promoters.

Abstract Background Cabazitaxel improves overall survival (OS) in metastatic castration-resistant prostate cancer (mCRPC) patients progressing after docetaxel. In this prospective study, we evaluated the prognostic role of CTC gene expression on cabazitaxel-treated patients and its association with plasma androgen receptor (AR) copy number (CN). Methods Patients receiving cabazitaxel 20 or 25 mg/sqm for mCRPC were enrolled. Digital PCR was performed to assess plasma AR CN status. CTC enrichment was assessed using the AdnaTest EMT-2/StemCell kit. CTC expression analyses were performed for 17 genes. Data are expressed as hazard ratio (HR) or odds ratio (OR) and 95% CI. Abstract

The present high-resolution inelastic neutron scattering experiment on ice XVII, containing molecular hydrogen with different ortho/para ratio, allows to assign the H$_2$ motion spectral bands to rotational and center-of-mass translational transitions of either {\it para}- or {\it ortho}-H$_2$. Due to its structure, ice XVII confines H$_2$ molecules to move in spiral channels of molecular size. Reported data demonstrate that H$_2$ molecules rotate almost freely in these nanometric channels, though showing larger perturbation than in clathrate hydrates, and perform a translational motion exhibiting two low frequency excitations. The good agreement between experimental spectra and corresponding molecular dynamics results clearly enables to portray a picture of the confined motions of a hydrophobic guest within a metastable ice framework, i.e. ice XVII., Comment: 5 pages, 4 figures

Abstract

Lung cancer (LC) is the deadliest malignancy worldwide. In an operable stage I–III patient setting, the detection of minimal residual disease (MRD) after curative treatment could identify patients at higher risk of relapse. In this context, the study of circulating tumor DNA (ctDNA) is emerging as a useful tool to identify patients who could benefit from an adjuvant treatment, and patients who could avoid adverse events related to a more aggressive clinical management. On the other hand, ctDNA profiling presents technical, biological and standardization challenges before entering clinical practice as a decisional tool. In this paper, we review the latest advances regarding the role of ctDNA in identifying MRD and in predicting patients’ prognosis, with a particular focus on clinical trials investigating the potential of ctDNA, the technical challenges to address and the biological parameters that influence the MRD detection.
