Sprachduschen sind eine Methode zum Lernen einer neuen Sprache. Dabei wird durch Spiele und Aktivitäten ein Einblick in die neue Sprache gegeben und versucht, einen positiven Eindruck von verschiedenen Sprachen und Kulturen zu vermitteln. Sprachduschen sind besonders nützlich für die Entscheidungsfindung vor der Wahl einer Fremdsprache in der Schule, wie es im Zusammenhang mit dieser Untersuchung der Fall ist. In dieser Pro-Gradu-Arbeit geht um zwei Sprachduschen in zwei verschiedenen dritten Klassen, die die Sprachenwahl vor sich haben. Es wird gezeigt, wie Sprachduschen geplant werden, welche Aktivitäten man durchführen kann und was man beim Organisieren von Sprachduschen berücksichtigen soll. Außerdem ist es das Ziel dieser Arbeit herauszufinden, wie aktiv die Schüler an den Sprachduschen teilnehmen und was sie von solchen Aktivitäten halten. In diesem Zusammenhang wird auch untersucht, welche Faktoren die Meinungen der Schüler beeinflussen, beispielsweise ob die Begeisterung für die englische Sprache oder Sprachen allgemein einen Einfluss auf die Begeisterung für die deutsche Sprache und Sprachduschen hat. Diese Pro-Gradu Arbeit ist eine ethnographische Forschung, die Sprachduschen von ihrer Organisation über die Durchführung bis zur Evaluierung durch die Schüler beobachtet und analysiert. Aufgrund der Ergebnisse der Analyse der Videoaufzeichnungen und der Fragebögen kann behauptet werden, dass Sprachduschen ein effektiver Weg sind, um eine neue Sprache kennenzulernen und die Schüler zu motivieren, Sprachen zu lernen. Die Schüler nehmen mit Begeisterung und ohne Vorurteile an der Sprachdusche teil. Die allgemeine Sprachbegeisterung wirkt sich auch positiv auf die Begeisterung für Sprachduschen und auf die Motivation für das Lernen neuer Sprachen aus. Gleichzeitig haben die Schüler, die Sprachenlernen im Allgemeinen nicht mögen, auch eine negative Einstellung gegenüber den Sprachduschen. Die Mehrheit der Schüler jedoch lernt gerne Sprachen.Tiivistelmä. Kielisuihkut ovat menetelmä uuden kielen oppimiseen. Ne tarjoavat pelien ja aktiviteettien avulla tietoa uudesta kielestä ja pyrkivät antamaan myönteisen vaikutelman eri kielistä ja kulttuureista. Kielisuihkut ovat erityisen hyödyllisiä tehdessä päätöksiä ennen vieraan kielen valintaa, kuten tämän tutkimuksen yhteydessä. Tämä pro gradu -työ käsittelee kahta kielisuihkusta kahdella eri kolmosluokalla, joilla on edessään kielivalinta. Tässä tutkimuksessa esitellään, miten kielisuihkut suunnitellaan, mitä aktiviteetteja niissä voidaan toteuttaa ja mitä on otettava huomioon kielisuihkuja järjestäessä. Lisäksi tämän työn tavoitteena on selvittää, kuinka aktiivisesti oppilaat osallistuvat kielisuihkuihin ja mitä mieltä he ovat tällaisesta toiminnasta. Tässä yhteydessä tutkitaan myös, mitkä tekijät vaikuttavat oppilaiden mielipiteisiin, esimerkiksi vaikuttaako innostus englannin kieleen tai kieliin yleisesti innostukseen saksan kieltä ja kielisuihkuja kohtaan. Tämä pro gradu -työ on etnografinen tutkimus, jossa havainnoidaan ja analysoidaan kielisuihkuja niiden järjestämisestä niiden toteuttamiseen ja oppilaiden suorittamaan arviointiin. Videotallenteiden ja kyselylomakkeiden analyysin tulosten perusteella voidaan väittää, että kielisuihkut ovat tehokas tapa oppia uutta kieltä ja motivoida oppilaita kielten opiskeluun. Oppilaat osallistuvat kielisuihkuun innokkaasti ja ennakkoluulottomasti. Yleinen innostus kieliä kohtaan vaikuttaa myönteisesti myös innostukseen kielisuihkuja kohtaan ja motivaatioon oppia uusia kieliä. Samaan aikaan oppilaat, jotka eivät pidä kielten opiskelusta yleensä, suhtautuvat kielteisemmin kielisuihkuihin. Suurin osa tutkimukseen osallistuneista oppilaista kuitenkin pitää kielten opiskelusta.

In dieser Arbeit geht es um Sprachduschen und ihren Nutzen und in dieser Arbeit werden die Meinungen zweier Lehrpersonen über die Wirkungen von Sprachduschen auf die Schüler behandelt. Dieses Thema ist gerade in Finnland sehr aktuell, weil nach dem neuen Curriculum (POPS 2019) die erste Fremdsprache, die sogenannte A1-Sprache, ab Januar 2020 in der ersten Klasse beginnt. Sprachduschen sind eine sehr gute Möglichkeit, die Schüler mit verschiedenen Sprachen bekannt zu machen. Das Ziel dieser Kandidatenarbeit ist also die Wirkung von Sprachduschen auf die Schüler nach der Meinung von Lehrern herauszufinden. Sprachdusche ist ein Begriff, der in Finnland entstanden ist. In der Sprachdusche werden neue Sprachen durch verschiedene Aktivitäten kennengelernt. Man lernt dabei die Grundkenntnisse einer Sprache. Das Hauptziel von Sprachduschen ist, sich für das Fremdsprachenlernen zu begeistern. Sprachduschen können entweder in der Schule oder auch schon im Kindergarten organisiert werden. In der Analyse werden zwei Lehrpersonen über die Wirkungen der Sprachduschen befragt und die Herausforderungen, die sich im Zusammenhang mit Sprachduschen ergeben, analysiert. Als Untersuchungsmethode wird Interview verwendet. Interview ist eine Methode, um Daten zu sammeln und durch Interviews versucht man die Meinung von Experten zu einem Thema herauszufinden. Das Material in dieser Arbeit baut sich auf die Interviews mit den zwei Lehrerinnen, bei denen eine Sprachdusche geführt wurde, auf. Durch die Ergebnisse kann gesagt werden, dass die Wirkungen der Sprachduschen auf die Schüler sehr positiv sind. Die Interviewten sind sich darüber einig, dass Sprachduschen wichtig und nützlich sind. Beide Interviewten sind der Meinung, dass Sprachduschen Motivation und Begeisterung für das Sprachenlernen geben.

We present FluxDirect, a novel computational framework for metabolic flux estimation from incomplete isotopomer data. In FluxDirect isotopomer measurements are first propagated in the metabolic network to obtain new constraints to the isotopomer distributions of metabolites in the network. The efficient propagation is facilitated by a fragment flow analysis of a metabolic network [1]. The method can utilize any measurement information produced by common (tandem) MS- and NMR-techniques [3]. After the propagation step the basic linear stoichiometric equation system bounding the fluxes is augmented with linear measurement constraints derived from the propagated isotopomer information. If the rank of the augmented linear equation system is full, the fluxes can be easily solved. If the rank of an equation system is less than full we can output upper and lower bounds for the fluxes. The general stability of an augmented equation system can be investigated by computing the condition number of the system. Unlike in most existing methods for flux estimation, techniques of nonlinear optimization are not required. Thus FluxDirect escapes the danger of convergence to local optima. Fragment flow analysis techniques introduced for efficient propagation of isotopomer measurement data also facilitate the experimental planning of isotopomer tracer experiments. As the development of measurement techniques for internal metabolites can be tedious, it is worthwhile to look for a small set of metabolites giving adequate flux information. We have shown that such small sets can be found for FluxDirect by applying computational set cover methods [2]. The flux estimation framework is empirically tested with a model of central carbon metabolism of S. cerevisiae.

We have developed a set of methods and tools for automatic discovery of putative regulatory signals in genome sequences. The analysis pipeline consists of gene expression data clustering, sequence pattern discovery from upstream sequences of genes, a control experiment for pattern significance threshold limit detection, selection of interesting patterns, grouping of these patterns, representing the pattern groups in a concise form and evaluating the discovered putative signals against existing databases of regulatory signals. The pattern discovery is computationally the most expensive and crucial step. Our tool performs a rapid exhaustive search for a priori unknown statistically significant sequence patterns of unrestricted length. The statistical significance is determined for a set of sequences in each cluster with respect to a set of background sequences allowing the detection of subtle regulatory signals specific for each cluster. The potentially large number of significant patterns is reduced to a small number of groups by clustering them by mutual similarity. Automatically derived consensus patterns of these groups represent the results in a comprehensive way for a human investigator. We have performed a systematic analysis for the yeast Saccharomyces cerevisiae. We created a large number of independent clusterings of expression data simultaneously assessing the "goodness" of each cluster. For each of the over 52,000 clusters acquired in this way we discovered significant patterns in the upstream sequences of respective genes. We selected nearly 1,500 significant patterns by formal criteria and matched them against the experimentally mapped transcription factor binding sites in the SCPD database. We clustered the 1,500 patterns to 62 groups for which we derived automatically alignments and consensus patterns. Of these 62 groups 48 had patterns that have matching sites in SCPD database.

We have examined methods and developed a general software tool for finding and analyzing combinations of transcription factor binding sites that occur relatively often in gene upstream regions (putative promoter regions) in the yeast genome. Such frequently occurring combinations may be essential parts of possible promoter classes. The regions upstream to all genes were first isolated from the yeast genome database MIPS using the information in the annotation files of the database. The ones that do not overlap with coding regions were chosen for further studies. Next, all occurrences of the yeast transcription factor binding sites, as given in the IMD database, were located in the genome and in the selected regions in particular. Finally, by using a general purpose data mining software in combination with our own software, which parametrizes the search, we can find the combinations of binding sites that occur in the upstream regions more frequently than would be expected on the basis of the frequency of individual sites. The procedure also finds so-called association rules present in such combinations. The developed tool is available for use through the WWW.

We consider the problem of automatic discovery of patterns and the corresponding subfamilies in a set of biosequences. The sequences are unaligned and may contain noise of unknown level. The patterns are of the type used in PROSITE database. In our approach we discover patterns and the respective subfamilies simultaneously. We develop a theoretically substantiated significance measure for a set of such patterns and an algorithm approximating the best pattern set and the subfamilies. The approach is based on the minimum description length (MDL) principle. We report a computing experiment correctly finding subfamilies in the family of chromo domains and revealing new strong patterns.

Data mining techniques must be developed and applied to analyse the large public data bases containing hundreds to thousands of millions entries. The aim of this study is to develop methods for locating previously unknown stellar clusters from the UKIDSS Galactic Plane Survey catalogue data. The cluster candidates are computationally searched from pre-filtered catalogue data using a method that fits a mixture model of Gaussian densities and background noise using the Expectation Maximization algorithm. The catalogue data contains a significant number of false sources clustered around bright stars. A large fraction of these artefacts were automatically filtered out before or during the cluster search. The UKIDSS data reduction pipeline tends to classify marginally resolved stellar pairs and objects seen against variable surface brightness as extended objects (or "galaxies" in the archive parlance). 10% or 66 x 10^6 of the sources in the UKIDSS GPS catalogue brighter than 17 magnitudes in the K band are classified as "galaxies". Young embedded clusters create variable NIR surface brightness because the gas/dust clouds in which they were formed scatters the light from the cluster members. Such clusters appear therefore as clusters of "galaxies" in the catalogue and can be found using only a subset of the catalogue data. The detected "galaxy clusters" were finally screened visually to eliminate the remaining false detections due to data artefacts. Besides the embedded clusters the search also located locations of non clustered embedded star formation. The search covered an area of 1302 square degrees and 137 previously unknown cluster candidates and 30 previously unknown sites of star formation were found.

In this paper we develop a theory describing how the extended Burrows-Wheeler Transform (eBWT) of a collection of DNA fragments tends to cluster together the copies of nucleotides sequenced from a genome G. Our theory accurately predicts how many copies of any nucleotide are expected inside each such cluster, and how an elegant and precise LCP array based procedure can locate these clusters in the eBWT. Our findings are very general and can be applied to a wide range of different problems. In this paper, we consider the case of alignment-free and reference-free SNPs discovery in multiple collections of reads. We note that, in accordance with our theoretical results, SNPs are clustered in the eBWT of the reads collection, and we develop a tool finding SNPs with a simple scan of the eBWT and LCP arrays.
