Magnetohydrodynamic laminar boundary layer slip flow of a nanofluid over a moving plate with Newtonian heating boundary condition in the presence of heat generation-absorption effects is studied using Lie group analysis and a numerical method. The model used for the nanofluid includes the effects of Brownian motion and thermophoresis. The governing transport equations are non-dimensionalized and transformed into a set of similarity equations using similarity transformations generated by Lie group transformations. The transformed equations are then solved using the Runge-Kutta-Fehlberg fourth-and fifth-order numerical method in Maple 17, which is also used to generate relevant graphs and tables. The flow, heat, and nanoparticle volume fraction characteristics are shown to depend on a number of thermophysical parameters, namely, Brownian motion, thermophoresis, Lewis number, Prandtl number, linear momentum slip, magnetic field, suction-injection, Newtonian heating, and heat generation-absorption. The effects of these parameters on the dimensionless stream function, velocity, temperature, nanoparticle volume fraction, wall heat, and mass transfer rates are investigated. Comparisons of the present numerical solutions with published works show very good correlation. The study finds applications in nano-technological magnetic materials processing. Utilisant une analyse par groupe de Lie et une methode numerique de calcul, nous etudions l'ecoulement glissant de la couche limite laminaire magnetohydrodynamique d'un nano-fluide sur une plaque en mouvement avec condition limite de chauffage newtonien en presence de sources et de puits thermiques. Le modele utilise pour le nano-fluide inclut le mouvement brownien et la thermophorese. Les equations maitresses de transport sont rendues non dimensionnelles et transformees par transformations de similarite generees par un groupe de transformation de Lie. Les equations ainsi transformees sont solutionnees numeriquement en utilisant une Runge-Kutta-Fehlberg au quatrieme-cinquieme ordre dans Maple 17, qui sert aussi pour generer les graphiques et les tables pertinents. Nous montrons que les caracteristiques d'ecoulement, de chaleur et de fraction volumique des nano-particules dependent d'un certain nombre de parametres thermophysiques, comme le mouvement brownien, la thermophorese, le nombre de Lewis, le nombre de Prandtl, le glissement de l'impulsion lineaire, le champ magnetique, la succion-injection, le chauffage newtonien et la generation-absorption de chaleur. Objective The objective of this study was to identify polymorphism in olfactomedin like 3 (OLFML3) gene, and association analysis with meat quality, carcass characteristics, retail meat cut, and fatty acid composition in sheep, and expression quantification of OLFML3 gene in phenotypically divergent sheep. Methods A total of 328 rams at the age of 10 to 12 months with an average body weight of 26.13 kg were used. A novel polymorphism was identified using high-throughput sequencing in sheep and genotyping of OLFML3 polymorphism was performed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Among 328 rams, 100 rams representing various sheep genotypes were used for association study and proc general linear model was used to analyse association between genotypes and phenotypic traits. Quantitative real-time polymerase chain reaction (qRT-PCR) was used for the expression analysis of OLFML3 mRNA in phenotypically divergent sheep population. Results The findings revealed a novel polymorphism in the OLFML3 gene (g.90317673 C>T). The OLFML3 gene revealed three genotypes: CC, CT, and TT. The single nucleotide polymorphism (SNP) was found to be significantly (p<0.05) associated with meat quality traits such as tenderness and cooking loss; carcass characteristics such as carcass length; retail meat cut such as pelvic fat in leg, intramuscular fat in loin and tenderloin, muscle in flank and shank; fatty acids composition such as tridecanoic acid (C13:0), palmitoleic acid (C16:1), heptadecanoic acid (C17:0), ginkgolic acid (C17:1), linolenic acid (C18:3n3), arachidic acid (C20:0), eicosenoic acid (C20:1), arachidonic acid (C20:4n6), heneicosylic acid (C21:0), and nervonic acid (C24:1). The TT genotype was associated with higher level of meat quality, carcass characteristics, retail meat cut, and some fatty acids composition. - Abstract
Satellite cells take an indispensable place in skeletal muscle regeneration, maintenance, and growth. However, only limited works have investigated effects of dietary compounds on the proliferation of porcine satellite cells (PSCs) and related mechanisms. Sulforaphane (SFN) at multiple levels was applied to PSCs. The PSCs’ viability and HDAC activity were measured with a WST-1 cell proliferation kit and Color-de-Lys® HDAC colorimetric activity assay kit. Gene expression and epigenetics modification were tested with qRT-PCR, Western blot, bisulfite sequencing, and ChIP-qPCR. This study found that SFN enhanced PSC proliferation and altered mRNA expression levels of myogenic regulatory factors. In addition, SFN inhibited histone deacetylase (HDAC) activity, disturbed mRNA levels of HDAC family members, and elevated acetylated histone H3 and H4 abundance in PSCs. Furthermore, both mRNA and protein levels of the Smad family member 7 (SMAD7) in PSCs were upregulated after SFN treatment. Finally, it was found that SFN increased the acetylation level of histone H4 in the SMAD7 promoter, decreased the expression of microRNAs, including ssc-miR-15a, ssc-miR-15b, ssc-miR-92a, ssc-miR-17-5p, ssc-miR-20a-5p, and ssc-miR-106a, targeting SMAD7, but did not impact on the SMAD7 promoter’s methylation status in PSCs. In summary, SFN was found to boost PSC proliferation and epigenetically increase porcine SMAD7 expression, which indicates a potential application of SFN in modulation of skeletal muscle growth.

Additive Manufacturing (AM) has provided a new potential for material customization through the reduction in geometric constraints of products. The freeform geometry produced using AM allows for the creation of optimized structural parts that reduce weight and material waste. In addition, AM reduces production time by providing a model to product workflow with limited requirements for tooling and machine setup in advance. However, current work in printing thermoset composite parts is limited. One of the confounding factors in the AM of thermoset composites involves in situ curing of the composite resin system. This research proposes the creation of an additive manufacturing process for thermoset composites based upon induction initiated thermoset curing. The use of induction-based curing will allow the production of parts without the normal constraint of either oven or autoclave curing. A rapid curing resin system will be reinforced using milled fiber reinforcement and doped with a ferromagnetic susceptor to induce homogenous heating and curing of the resin in situ. The resulting composite will be evaluated for mechanical performance versus oven cure samples. Scanning electron microscopy in tandem with energy dispersive x-ray spectroscopy will be used to validate homogenous particle dispersion. Lastly, differential scanning calorimetry will be used to compare the degree of polymer conversion between induction and conventional curing.

- Abstract
Circulating autoantibodies (auto-Abs) neutralizing high concentrations (10 ng/ml; in plasma diluted 1:10) of IFN-α and/or IFN-ω are found in about 10% of patients with critical COVID-19 (coronavirus disease 2019) pneumonia but not in individuals with asymptomatic infections. We detect auto-Abs neutralizing 100-fold lower, more physiological, concentrations of IFN-α and/or IFN-ω (100 pg/ml; in 1:10 dilutions of plasma) in 13.6% of 3595 patients with critical COVID-19, including 21% of 374 patients >80 years, and 6.5% of 522 patients with severe COVID-19. These antibodies are also detected in 18% of the 1124 deceased patients (aged 20 days to 99 years; mean: 70 years). Moreover, another 1.3% of patients with critical COVID-19 and 0.9% of the deceased patients have auto-Abs neutralizing high concentrations of IFN-β. We also show, in a sample of 34,159 uninfected individuals from the general population, that auto-Abs neutralizing high concentrations of IFN-α and/or IFN-ω are present in 0.18% of individuals between 18 and 69 years, 1.1% between 70 and 79 years, and 3.4% >80 years. Moreover, the proportion of individuals carrying auto-Abs neutralizing lower concentrations is greater in a subsample of 10,778 uninfected individuals: 1% of individuals 80 years. By contrast, auto-Abs neutralizing IFN-β do not become more frequent with age. Auto-Abs neutralizing type I IFNs predate SARS-CoV-2 infection and sharply increase in prevalence after the age of 70 years. They account for about 20% of both critical COVID-19 cases in the over 80s and total fatal COVID-19 cases. © 2021 The Authors, some rights reserved.

Inflammation is regulated by epigenetic modifications, including DNA methylation and histone acetylation. Sulforaphane (SFN), a histone deacetylase (HDAC) inhibitor, is also a potent immunomodulatory agent, but its anti-inflammatory functions through epigenetic modifications remain unclear. Therefore, this study aimed to investigate the epigenetic effects of SFN in maintaining the immunomodulatory homeostasis of innate immunity during acute inflammation. For this purpose, SFN-induced epigenetic changes and expression levels of immune-related genes in response to lipopolysaccharide (LPS) stimulation of monocyte-derived dendritic cells (moDCs) were analyzed. These results demonstrated that SFN inhibited HDAC activity and caused histone H3 and H4 acetylation. SFN treatment also induced DNA demethylation in the promoter region of the MHC-SLA1 gene, resulting in the upregulation of Toll-like receptor 4 (TLR4), MHC-SLA1, and inflammatory cytokines’ expression at 6 h of LPS stimulation. Moreover, the protein levels of cytokines in the cell culture supernatants were significantly inhibited by SFN pre-treatment followed by LPS stimulation in a time-dependent manner, suggesting that inhibition of HDAC activity and DNA methylation by SFN may restrict the excessive inflammatory cytokine availability in the extracellular environment. We postulate that SFN may exert a protective and anti-inflammatory function by epigenetically influencing signaling pathways in experimental conditions employing porcine moDCs.

Fatty acids (FA) in ruminants, especially unsaturated FA (USFA) have important impact in meat quality, nutritional value, and flavour quality of meat, and on consumer’s health. Identification of the genetic factors controlling the FA composition and metabolism is pivotal to select sheep that produce higher USFA and lower saturated (SFA) for the benefit of sheep industry and consumers. Therefore, this study was aimed to investigate the transcriptome profiling in the liver tissues collected from sheep with divergent USFA content in longissimus muscle using RNA deep-sequencing. From sheep (n = 100) population, liver tissues with higher (n = 3) and lower (n = 3) USFA content were analysed using Illumina HiSeq 2500. The total number of reads produced for each liver sample were ranged from 21.28 to 28.51 million with a median of 23.90 million. Approximately, 198 genes were differentially regulated with significance level of p-adjusted value <0.05. Among them, 100 genes were up-regulated, and 98 were down-regulated (p<0.01, FC>1.5) in the higher USFA group. A large proportion of key genes involved in FA biosynthesis, adipogenesis, fat deposition, and lipid metabolism were identified, such as APOA5, SLC25A30, GFPT1, LEPR, TGFBR2, FABP7, GSTCD, and CYP17A. Pathway analysis revealed that glycosaminoglycan biosynthesis- keratan sulfate, adipokine signaling, galactose metabolism, endocrine and other factors-regulating calcium metabolism, mineral metabolism, and PPAR signaling pathway were playing important regulatory roles in FA metabolism. Importantly, polymorphism and association analyses showed that mutation in APOA5, CFHR5, TGFBR2 and LEPR genes could be potential markers for the FA composition in sheep. These polymorphisms and transcriptome networks controlling the FA variation could be used as genetic markers for FA composition-related traits improvement. THO Complex 5 (THOC5) gene involves in lipid and fatty acid metabolism. The aim of this study was to analyse polymorphism of THOC5 gene and its association with fatty acid composition in sheep. A total of 120 rams at the age 12 month with the average body weight of 25–30 kg was used for identification of gene polymorphism using Polymerase Chain Reaction- Restriction Fragment Length Polymorphism (PCR-RFLP). 83 rams, representative from different genotypes of sheep were used for association study using a General Linear Model (GLM). The results identifed polymorphism in the THOC5 gene. The THOC5 gene showed two genotypes: CC and CT genotype. Association analysis revealed that THOC5 (g.68234589 C>T) was significantly (P<0.05) associated with fatty acid composition including unsaturated fatty acid: palmitoleic acid (C16:1), and saturated fatty acid: stearic acid (C18:0). The CC genotype was associated with higher level of unsaturated fatty acid and lower level of saturated fatty acid, while the CT genotype was vice versa. This result indicates that THOC5 gene (g.68234589 C>T) may contribute to fatty acid composition in sheep, as well as this polymorphism could be used as a candidate to select sheepmeat with high unsaturated fatty acid and low saturated fatty acid.

Bangladesh faces various types of natural hazards from its birth due to geographical location and physiographic conditions. Flood is the most common event among them. Northeastern part of Bangladesh faces flash flood almost every year with large scale of damage. Tanguar haor, (a famous Ramsar site of Bangladesh) located in Tahirpur upazila of Sunamgonj district. This wetland adjacent area is the most vulnerable zone in terms of flash flood hazard. About more than 80%, people are the direct victim of this hazard. The aim of this study was to assess the risk and resilience status of flash flood at community level. A risk and resilience assessment matrix leads to fulfill the assessment where only primary data were used. A comparative study between those statuses is computed based on Pearson correlation of co-efficient analysis that presents the difference of risk and resilience of the study area that helps to realize the emergence of flash flood risk reduction. The findings of this study will be a pathway for the policy makers, local administrators for the development of sustainable community level risk management, policy-making and its implementations.

- Abstract
Interindividual clinical variability in the course of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is vast. We report that at least 101 of 987 patients with life-threatening coronavirus disease 2019 (COVID-19) pneumonia had neutralizing immunoglobulin G (IgG) autoantibodies (auto-Abs) against interferon-w (IFN-w) (13 patients), against the 13 types of IFN-a (36), or against both (52) at the onset of critical disease; a few also had auto-Abs against the other three type I IFNs. The auto-Abs neutralize the ability of the corresponding type I IFNs to block SARS-CoV-2 infection in vitro. These auto-Abs were not found in 663 individuals with asymptomatic or mild SARS-CoV-2 infection and were present in only 4 of 1227 healthy individuals. Patients with auto-Abs were aged 25 to 87 years and 95 of the 101 were men. A B cell autoimmune phenocopy of inborn errors of type I IFN immunity accounts for life-threatening COVID-19 pneumonia in at least 2.6% of women and 12.5% of men.
