Abstract: International audience; Farming systems can expose animals to chronic mild stress which is known to induce negative affective state. Affective state in animals, as in humans, can be assessed through behavioral cues. This study aimed to describe the effect of a chronic mild stress, known to induce a negative affective state, on sheep health through their response to vaccination. The study used 15 lambs subjected to a model of chronic mild stress for 15 weeks and 15 lambs reared under conventional farming as a control group. After 7 weeks of stressful treatment, the lambs were individually exposed to a judgment bias test to assess a putative stress-induced 'pessimism.' After 15 weeks of stressful treatment, antibody immune response was measured after an injection of a live vaccine challenge (Chlamydia abortus attenuated vaccine strain 1B). Stressed lambs displayed a pessimistic-like perception in the judgment bias test, revealing a negative affective state. Stressed and control animals showed different immunological reactions to vaccine challenge: stressed sheep had lower hemoglobin concentrations and higher platelet, granulocyte and acute-phase protein concentrations. Antibody response induced by the vaccine strain was not different between stressed and control sheep. Our results suggest that negative affective state induced by chronic stress treatment may induce a stronger inflammatory response to vaccine challenge in sheep. Improvement of animal health may be achieved through consideration of stressors that may affect the emotional and immunological state of sheep.

Abstract: International audience; Kidney transplant donors are not currently implicated in predicting BK polyomavirus (BKPyV) infection in kidney transplant recipients. It has been postulated, however, that BKPyV infection originates from the kidney allograft. Because BKPyV seroreactivity correlates with BKPyV replication and thus might mirror the infectious load, we investigated whether BKPyV seroreactivity of the donor predicts viremia and BKPyV-associated nephropathy (BKPyVAN) in the recipient. In a retrospective cohort of 407 living kidney donor-recipient pairs, pretransplantation donor and recipient sera were tested for BKPyV IgG levels and correlated with the occurrence of recipient BKPyV viremia and BKPyVAN within 1 year after transplantation. Donor BKPyV IgG level was strongly associated with BKPyV viremia and BKPyVAN (p < 0.001), whereas recipient BKPyV seroreactivity showed a nonsignificant inverse trend. Pairing of high-BKPyV-seroreactive donors with low-seroreactive recipients resulted in a 10-fold increased risk of BKPyV viremia (hazard ratio 10.1, 95% CI 3.5-29.0, p < 0.001). In multivariate analysis, donor BKPyV seroreactivity was the strongest pretransplantation factor associated with viremia (p < 0.001) and BKPyVAN (p = 0.007). The proportional relationship between donor BKPyV seroreactivity and recipient infection suggests that donor BKPyV seroreactivity reflects the infectious load of the kidney allograft and calls for the use of pretransplantation BKPyV serological testing of (potential) donors and recipients.

Abstract: International audience; To get new insights into the distribution and modes of acquisition of trichodysplasia spinulosa polyomavirus (TSPyV) infection, we performed a seroepidemiological study in two populations from Cameroon. Our results suggest that TSPyV infection occurs, through close contacts, during early childhood with intra-familial transmission, especially from mother to child.

Abstract: Objective. In 2015, we conducted at 44 healthcare facilities (HCFs) and 21 nursing homes (NHs) a three-month bloodstream infection (BSI) survey, and a one-day prevalence study to determine the rate of carriage of methicillin-resistant Staphylococcus aureus (MRSA) in 891 patients and 470 residents. We investigated the molecular characteristics of the BSI-associated and colonizing MRSA isolates, and assessed cross-transmission using double-locus sequence typing and pulsed-field gel electrophoresis protocol. Abstract: The spread of anthelmintic resistance in nematodes has become a major problem, so that there is an urgentneed to develop alternatives for parasite control. Here, we investigated the efficacy of a short-termconsumption of sainfoin, a tannin rich plant, or of extra proteins in naturally infected horses. 3 groups of 10individuals received for 18 days either (i) a „control‟ diet: 100% of UFC and MADC requirements, (ii) a„protein-rich‟ diet: 100% UFC, 230% MADC, (iii) a „sainfoin‟ diet: 100% UFC, 230% MADC, 3.6%DM ofcondensed tannins. Faecal egg counts did not reveal any difference between the three feeding regimes.However, coprocultures from the faeces pooled in each group at the end of the experiment suggested a lowerrate of larval development in the „sainfoin‟ group (8.1% vs. 30.5%: „protein-rich „ and 22.6%: „control‟). Invitro tests using sainfoin extracts confirmed the influence of the secondary metabolites on egg hatching(P, L‟émergence de résistances aux anthelminthiques chez les nématodes nécessite de développer des solutionsalternatives de lutte contre le parasitisme. Abstract: Pattern recognition receptors (PRRs) play a critical role in the detection of microorganisms and the induction of inflammatory and immune responses. Using PCR and Western-blot analysis, this study investigated the differential expression in the intestine of 14 PRRs and nine associated cytokines. Thirty-two pigs were used to determine the expression of these markers (1) along the proximal/distal axis of the small intestine (duodenum, jejunum, and ileum) and (2) between the intestinal segments and their respective lymphoid organs (Peyer's patches [PP] and mesenteric lymph nodes [MLN]). Six additional animals were used to quantify the expression of these genes along the crypt/villus axis of jejunum, using microdissected samples. Most genes showed increased expression (1) in the distal than in the proximal parts of the small intestine (TLR3, 5, RIG-I, IL-1β, IL-8, and IFN-γ); (2) in lymphoid organs (TLR1, 2, 6, 9, 10, IL-10, TNF-α), especially the MLN (TLR4, 7, 8, NOD1, NOD2, NALP3, IFN-α, IL-6, IL-12, and TGF-β), than in intestinal segments. The analysis along the crypt/villus identified: (1) genes with higher expression in lamina propria (TLR1, 2, 4, 9, NOD1, NOD2, IL-1β, IL-10, TGF-β, TNF-α) and (2) genes with higher expression in the villus (TLR3, 5, 6, RIG-I, IL-6). These results highlight the differential expression of PRRs and cytokines along the proximal/distal and the crypt/villus axis of the intestine, contributing to a fine analysis of the complex functional architecture of the small intestine and should be related to the gut microbiota.

Abstract: Background Streptococcus agalactiae, or Group B Streptococcus, is a leading cause of neonatal infections and an increasing cause of infections in adults with underlying diseases. In an effort to reconstruct the transcriptional networks involved in S. agalactiae physiology and pathogenesis, we performed an extensive and robust characterization of its transcriptome through a combination of differential RNA-sequencing in eight different growth conditions or genetic backgrounds and strand-specific RNA-sequencing. Results Our study identified 1,210 transcription start sites (TSSs) and 655 transcript ends as well as 39 riboswitches and cis-regulatory regions, 39 cis-antisense non-coding RNAs and 47 small RNAs potentially acting in trans. Among these putative regulatory RNAs, ten were differentially expressed in response to an acid stress and two riboswitches sensed directly or indirectly the pH modification. Strikingly, 15% of the TSSs identified were associated with the incorporation of pseudo-templated nucleotides, showing that reiterative transcription is a pervasive process in S. agalactiae. In particular, 40% of the TSSs upstream genes involved in nucleotide metabolism show reiterative transcription potentially regulating gene expression, as exemplified for pyrG and thyA encoding the CTP synthase and the thymidylate synthase respectively. Conclusions This comprehensive map of the transcriptome at the single nucleotide resolution led to the discovery of new regulatory mechanisms in S. agalactiae. It also provides the basis for in depth analyses of transcriptional networks in S. agalactiae and of the regulatory role of reiterative transcription following variations of intra-cellular nucleotide pools. Abstract: Tick-borne pathogens cause potent infections. These pathogens benefit from molecules contained in tick saliva that have evolved to modulate host innate and adaptive immune responses. This is called “saliva-activated transmission” and enables tick-borne pathogens to evade host immune responses. Ticks feed on their host for relatively long periods; thus, mechanisms counteracting the inflammation-driven recruitment and activation of innate effector cells at the bite site, are an effective strategy to escape the immune response. Here, we developed an original in vitro model to evaluate and to characterize the immunomodulatory effects of tick saliva that prevent the establishment of a local inflammatory immune response. This model mimics the tick bite and enables the assessment of the effect of saliva on the inflammatory-associated dynamic recruitment of cells from the mononuclear phagocyte system. Using this model, we were able to recapitulate the dual effect of tick saliva on the mobilization of inflammatory monocyte-derived cells, i.e. (i) impaired recruitment of monocytes from the blood to the bite wound; and (ii) poor mobilization of monocyte-derived cells from the skin to the draining lymph node. This simple tool reconstitutes the effect of tick saliva in vivo, which we characterized in the mouse, and should enable the identification of important factors facilitating pathogen infection. Furthermore, this model may be applied to the characterization of any pathogen-derived immunosuppressive molecule affecting the establishment of the inflammatory immune response. Abstract: Farmers value autonomy in the management of their farm. Amongst organic farmers especially, autonomy is thought to contribute to the success of the farm. Wider integrated farming systems however oppose such autonomy by making farm management decisions remotely. One of the greatest threats to the success of meat sheep farms is the presence of parasitic gastrointestinal nematodes. This study questioned whether a greater level of farmer autonomy could be correlated to the better control of the gastrointestinal nematode infections and whether this could be associated with farmer specific health beliefs. Data was collected across 36 meat sheep farms with similar climatic variables but with different beliefs and attitudes to health care in farming. These farms fell within three groups: French organic, French conventional and Algerian conventional farms. Information regarding farmers' health beliefs and their level of autonomy in management was gathered using questionnaires to address autonomy in the following variables: agriculture production, husbandry, feed, therapeutics, commercialization, and farmers' education. The intensity of gastrointestinal nematode infections was also measured on each of these farms using faecal egg counts on composite sheep samples. Statistical and correlation analyses of autonomy variables to gastrointestinal nematode infection intensities were carried out. The results showed farmer autonomy was not associated with better gastrointestinal nematode control in any of the three groups of farms studied. Greater level of autonomy in husbandry and therapeutics specifically were associated with greater levels of infection. Farmers across the 36 farms studied were unanimously concerned about the threat gastrointestinal nematodes present to their flocks. Yet their current knowledge was clearly not sufficient to control infections irrespective of their health beliefs. This paper makes suggestions to facilitate communication with farmers and to improve the integration of gastrointestinal nematode control measures.

Abstract: DOSSIER : LA RÉSISTANCE AUX ANTHELMINTHIQUES CHEZ LES ÉQUIDÉS; Dans le contexte de l’émergence de la résistance aux anthelminthiques, le développement de stratégies alternatives semble indispensable.Ces stratégies restent, à l'heure actuelle, expérimentales et seront mises en pratique que dans quelques années.Les alternatives considérées portent sur la lutte biologique, par l'intermédiaire de champignons nématophages notamment.D'autres stratégies visant à augmenter la résistance de l'hôte, conférée par un vaccin ou par certains apports nutritionnels , ne semblent pas appropriées dans le cas des nématodoses gastro-intestinales.À l'interface de ces deux approches, le potentiel bioactif de certaines plantes pourrait conférer aux prairies et aux fourrages cultivés un rôle “pharmacologique” permettant de limiter la charge parasitaire des équidés.

Abstract: International audience; We conducted a survey including 3334 bloodstream infections (BSIs) due to E. coli diagnosed in 2005-2014 at a stable cohort of hospitals. Marked increases in incidence were observed for community-acquired (CA) BSIs in patients aged >75 years, CA-BSIs of digestive origin in patients aged 60-74 years, healthcare-associated BSIs, and BSIs associated with ESBL (extended-spectrum B-lactamase)-producing E. coli (ESBLEc). Using MLST, we studied the genetic diversity of 412 BSI isolates recovered during the 2014 survey: 7 major sequence type complexes (STCs) were revealed in phylogenetic group B2, 3 in group A/B1 and 2 in group D. Among the 31 ESBLEc isolates, 1/3 belonged to STC 131. We searched for possible associations between clonal groups, clinical determinants and characteristics of BSIs: isolates from groups B2 (except STC 131) and D were susceptible to antibiotics and associated with BSIs of urinary origin in patients

Abstract: The aim of this study was to analyse gene expression along the small intestine (duodenum, jejunum, ileum) and in the ileal Peyer's patches in four young pigs with no clinical signs of disease by transcriptome sequencing. Multidimensional scaling evidenced that samples clustered by tissue type rather than by individual, thus prefiguring a relevant scenario to draw tissue-specific gene expression profiles. Accordingly, 1,349 genes were found differentially expressed between duodenum and jejunum, and up to 3,455 genes between duodenum and ileum. Additionally, a considerable number of differentially expressed genes were found by comparing duodenum (7,027 genes), jejunum (6,122 genes), and ileum (6,991 genes) with ileal Peyer's patches tissue. Functional analyses revealed that most of the significant differentially expressed genes along small intestinal tissues were involved in the regulation of general biological processes such as cell development, signalling, growth and proliferation, death and survival or cell function and maintenance. These results suggest that the intrinsic large turnover of intestinal tissues would have local specificities at duodenum, ileum and jejunum. In addition, in concordance with their biological function, enteric innate immune pathways were overrepresented in ileal Peyer's patches. The reported data provide an expression map of the cell pathway variation in the different small intestinal tissues. Furthermore, expression levels measured in healthy individuals could help to understand changes in gene expression that occur in dysbiosis or pathological states.

Abstract: Le microbiote digestif est un symbionte dont on a démontré l’implication essentielle dans la régulation de la physiologie de l’hôte qui l’héberge pour la digestion, le métabolisme et l’immunité. Ce microbiote a en particulier un rôle majeur, et jusqu’ici sous-estimé, sur la santé de son hôte et participe à son adaptabilité à des environnements changeants. Les technologies de séquençage à haut débit devraient faciliter l’inclusion de cet écosystème complexe dans le phénotypage des animaux d’élevage. Il faut pour ce faire prendre en compte sa composition, sa diversité, sa stabilité au cours du temps et ses variations selon différents facteurs comme le génotype de l’hôte, l’alimentation et les conditions d’élevage. Ces connaissances devraient permettre de définir les propriétés d’un microbiote digestif « normal » associé à une bonne santé, d’en préciser les dysfonctionnements et de comprendre leurs répercussions sur l’état de santé de l’animal. Cela conduira à identifier des marqueurs de diagnostic et de pronostic de ces dysfonctionnements, fournissant des moyens de contrôle nouveaux de la santé en élevage, appréhendée en terme d’optimisation des rendements de production, de résistance aux maladies et de maintien du bien-être. A terme, le contrôle du microbiote digestif par l’alimentation, la génétique ou les pratiques d’élevage permettra de favoriser l’adaptation des animaux à leur environnement d’élevage en améliorant leur robustesse, de limiter l’utilisation d’antibiotiques et de préserver la sécurité sanitaire des aliments en évitant le développement de bactéries pathogènes pour l’Homme. In particular, the role of the gastrointestinal microbiota in maintaining host health is important and has been under-estimated until now. It is an inherent component of the animal that should be described when phenotyping animals. The rapid development of high throughput sequencing technologies facilitates the description of this complex ecosystem in livestock species. For each animal species, its description should include the measure of its composition, diversity and stability over time and the measure of its variability according to different factors such as genotype, nutrition and breeding conditions. This knowledge should allow defining the properties of a ‘normal’ gastrointestinal microbiota associated with health, identifying the microbiota dysfunctions and understanding their consequences on host physiology. It will lead to the identification of diagnostic and prognostic markers for gastrointestinal dysbiosis thus furnishing new tools for promoting health. Finally, the manipulation of the gastrointestinal microbiota will constitute a way to better adapt animals to their environment by improving their robustness, limit the use of antibiotics and improve food safety by preventing the development of bacteria threatening human health. Due to the high quantity of data to be acquired and analyzed, achieving these objectives for each species of interest remains a challenge.

Abstract: Solid-state properties of active pharmaceutical compounds are closely related to their dissolution behavior and bioavailability, and understanding their phase behavior leads to better control in the drug formulation process. L-Citrulline is an amino acid, which is known to exhibit polymorphism; however, only the structure of the alpha form is reported in the literature. The structure of the form has been elusive due to lack of good quality single crystals. It has been found to be orthorhombic P2(1)2(1)2(1) with the cell parameters a, b, and c respectively 14.895(5) angstrom, 9.852(2) angstrom, and 5.353(2) angstrom. The unit cell contains four molecules, and its volume is 785.5(4) angstrom(3). Both the alpha form and the delta form possess uniaxial negative thermal expansion along the direction of the charge exchange of the L-citrulline zwitterions. In mixtures containing solid and saturated aqueous, solution, the alpha form quickly transforms in a dihydrate, whereas the delta phase persists for over three weeks. Although the evidence is not conclusive, the delta phase is most likely the more stable form and most suitable for storage.

Abstract: Merkel cell carcinoma (MCC) is a rare, malignant primary neuroendocrine cancer of the skin, usually affecting elderly, white people in sun-exposed areas. This is a highly aggressive tumor with strong propensity to metastasize. Surgery and radiation therapy remain the mainstay of treatment, with no curative treatment in case of disseminated metastases. Until 2008, MCC was thought to be caused by the malignant transformation of resident Merkel cells, but no investigation of a predominant molecular pathway that could be involved in MCC pathogenesis was successful. A real revolution in MCC understanding and management occurred in 2008, when a new human polyomavirus (MCPyV) was found to be the main etiological agent of this skin cancer. Following the discovery of MCPyV, the association of MCPyV with MCC has been confirmed worldwide, with detection of MCPyV in about 80% of MCCs. At the same time it had been shown that MCPyV infection is almost ubiquitous in healthy subjects, and MCPyV is thought to be persistent resident of the skin microbiome although the route of transmission, the host cell, the viral cycle and/or latency remain unknown. Most studies suggest that there may be two subtypes of MCC: MCPyV-positive (80%) and MCPyV-negative (20%) MCCs, and various studies have reported a better prognosis associated with MCPyV infection. The discovery of MCPyV in MCC patients opens up new therapeutic insights. The necessity and persistence of expression of MCPyV oncoproteins during MCC development make these proteins promising therapeutic targets.

Abstract: The major challenge of animal phenotyping is to acquire systemic knowledge of farm animal robustness. This will allow responding to a double objective in terms of exploiting the variability of animal aptitudes: selection for major objectives in order to improve efficiency and robustness of genotypes and precision farming that exploits individual variability of animals for an improved resilience on the herd level or to improve the management for a specific genotype. Two major challenges of our knowledge exist which are interdependent and common to all animal breeding, and which must be met in order to improve animal robustness. The first is understanding andpredicting the compromise amongst vital functions. By this we mean understanding how priority changes are made under specific limited resources. The second is understanding and exploiting the temporal aspects of robustness during an animal’s life in order to allow the animal to maintain its level of performance under different environments without compromising its reproduction, health or welfare. Attaining these objectives requires accomplishing the following: i) defining complex phenotypes through the integration of data obtained on the molecular or tissular scales of an animal or of one of its functions; ii) developing high-throughput technologiesto characterize at a lower cost and the most precisely as possible the largest number of animals; iii) developing important data bases and reliable methods and computing tools necessary for the treatment of information for modeling and predictive biology., L’enjeu majeur du phénotypage animal est d’acquérir une connaissance systémique de la robustesse des animaux d’élevage pour répondre au mieux à une double finalité en termes d’exploitation de la variabilité des aptitudes animales : une sélection éclairée vers des objectifs majeurs pour améliorer l’efficience et la robustesse des génotypes, et un élevage de précision exploitant la variabilité individuelle des animaux pour gagner en résilience à l’échelle du troupeau, ou pour améliorer la conduite à génotype donné. Deux défis majeurs de connaissance, interdépendants et communs à toutes les filières animales, sont à relever pour améliorer la robustesse des animaux : i) comprendre et prédire les compromis entre fonctions vitales, c’est-à-dire les changements de priorités dans l’allocation de ressources limitées ; ii) comprendre et exploiter les aspects temporels de la robustesse au cours de la vie de l’animal pour lui permettre de maintenir son niveau de production dans une large gamme d’environnements sans pour autant compromettre sa reproduction, sa santé et son bien-être. Atteindre ces objectifs nécessite de lever les verrous techniques suivants : i) définir des phénotypes complexes à partir de l’intégration de données obtenues à différentes échelles moléculaires, tissulaires, de l’animal ou d’une de ses fonctions ; ii) mettre en oeuvre des technologies à haut débit pour caractériser à moindre coût et le plus précisément possible le plus grand nombre d’animaux ; iii) développer d’importantes bases de données, des méthodes et outils informatiques performants, nécessaires aux traitements des informations à des fins de modélisation et de biologie prédictive.

Abstract: Données de la recherche; Understanding of biology of the avian male gamete is essential to improve the conservation of genetic resources and performances in farming. In this study, the semen proteome of the main domestic avian species (Gallus gallus) and evaluation of the molecular phenotype related to sperm quality were investigated using GeLC–MS/MS approach and label-free quantitative proteomic based on Spectral Counting (SC) and extracted ion chromatograms (XIC) methods. Here we describe in details the peptide/protein inventory of chicken ejaculated spermatozoa (SPZ) and seminal plasma (SP). We also show differential analyses of chicken semen (SPZ and corresponding SP) from 11 males demonstrating different levels of fertilizing capacity and sperm motility. The interpretation and description of these data can be found in a research article published by Labas and colleagues in the Journal of Proteomics in 2014 [1]. This is a new resource for exploring the molecular mechanisms involved in fertilizing capacity and to reveal new sets of fertility biomarkers.

Abstract: Session : Innovations en élevageSession : Innovations en élevage; Ce projet consiste en une expérimentation "système" pluridisciplinaire : nous avons mis à l’épreuve du réel, pour les évaluer sur différents critères, deux systèmes herbagers ovins biologiques, productifs et autonomes, dont l’un propose le fractionnement des périodes de mises-bas comme élément de sécurisation face aux aléas. L’hypothèse initiale, formulée par simulation ex-ante, n’a pas été validée. Le système fractionné a permis une meilleure productivité animale, mais les coûts de production, la charge de travail et sa complexité y ont été plus élevés que dans le système autonome. A signaler notamment une moindre récolte de stocks fourragers dans ce système, notamment en 2011 (printemps sec), due à une plus grande difficulté de gestion du pâturage.

Abstract: Background/Aims: Deoxynivalenol (DON) is a mycotoxin produced by Fusarium species which is commonly found in temperate regions worldwide as a natural contaminant of cereals. It is of great concern not only in terms of economic losses but also in terms of animal and public health. The digestive tract is the first and main target of this food contaminant and it represents a major site of immune tolerance. A finely tuned cross-talk between the innate and the adaptive immune systems ensures the homeostatic equilibrium between the mucosal immune system and commensal microorganisms. The aim of this study was to analyze the impact of DON on the intestinal immune response. Methodology: Non-transformed intestinal porcine epithelial cells IPEC-1 and porcine jejunal explants were used to investigate the effect of DON on the intestinal immune response and the modulation of naive T cells differentiation. Transcriptomic proteomic and flow cytometry analysis were performed. Results: DON induced a pro-inflammatory response with a significant increase of expression of mRNA encoding for IL-8, IL-1 alpha and IL-1 beta, TNF-alpha in all used models. Additionally, DON significantly induced the expression of genes involved in the differentiation of Th17 cells (STAT3, IL-17A, IL-6, IL-1 beta) at the expenses of the pathway of regulatory T cells (Treg) (FoxP3, RALDH1). DON also induced genes related to the pathogenic Th17 cells subset such as IL-23A, IL-22 and IL-21 and not genes related to the regulatory Th17 cells (rTh17) such as TGF-beta and IL-10. Conclusion: DON triggered multiple immune modulatory effects which could be associated with an increased susceptibility to intestinal inflammatory diseases.

Abstract: Session : Elevage bioSession : Elevage bio; La plateforme INRA de recherches en production ovine allaitante biologique a été constituée en 1999 et est convertie en agriculture biologique (AB) depuis 2002. Elle associe trois approches complémentaires : des expérimentations de type système, des études plus analytiques sur des verrous identifies et le suivi technico-économique d'un réseau d'exploitations. Un outil de modélisation permet par ailleurs d'aider à la conception et au pilotage des expérimentations systèmes et a l'extrapolation de certains bilans. Plusieurs thématiques de recherche y sont développées : i) la conception de systèmes d'élevage AB cherchant a combiner de manière optimale le niveau de productivité animale et le niveau d'autonomie alimentaire, ii) la maîtrise durable du parasitisme a travers la gestion raisonnée du pâturage, le développement d'indicateurs pour repérer les animaux 'sensibles' ou 'sentinelles', et évaluation de l'efficacité de thérapeutiques alternatives, enfin iii) l'évaluation des qualités de la viande et de la carcasse d'agneau AB. Cette communication présente certains résultats obtenus, ainsi que les perspectives de recherche envisagées.

Abstract: International audience; Background : The lack of cystic fibrosis transmembrane conductance regulator (CFTR) function causes cystic fibrosis (CF), predisposing to severe lung disease, reduced growth and osteopenia. Both reduced bone content and strength are increasingly recognized in infants with CF before the onset of significant lung disease, suggesting a developmental origin and a possible role in bone disease pathogenesis. The role of CFTR in bone metabolism is unclear and studies on humans are not feasible. Deletion of CFTR in pigs (CFTR −/− pigs) displays at birth severe malformations similar to humans in the intestine, respiratory tract, pancreas, liver, and male reproductive tract. Methods : We compared bone parameters of CFTR −/− male and female pigs with those of their wild-type (WT) littermates at birth. Morphological and microstructural properties of femoral cortical and trabecular bone were evaluated using micro-computed tomography (μCT), and their chemical compositions were examined using Raman microspectroscopy. Results : The integrity of the CFTR −/− bone was altered due to changes in its microstructure and chemical composition in both sexes. Low cortical thickness and high cortical porosity were found in CFTR −/- pigs compared to sex-matched WT littermates. Moreover, an increased chemical composition heterogeneity associated with higher carbonate/phosphate ratio and higher mineral crystallinity was found in CFTR −/− trabecular bone, but not in CFTR −/− cortical bone. Conclusions : The loss of CFTR directly alters the bone composition and metabolism of newborn pigs. Based on these findings, we speculate that bone defects in patients with CF could be a primary, rather than a secondary consequence of inflammation and infection.

Abstract: International audience; Genotyping of Mycobacterium avium subsp. paratuberculosis (Map) is an indispensable tool for surveillance of this significant veterinary pathogen. For Map, multi-locus variable number tandem repeat analysis (MLVA) targeting mycobacterial interspersed repetitive units (MIRUs) and other variable number variable-number tandem repeats (VNTRs) was established using 8 markers. In the recent past this standard, portable, reproducible and discriminatory typing method has been frequently applied alone or in combinations with multi-locus short-sequence-repeat (MLSSR) sequencing. With the widespread use of these genotyping methods, standardization between laboratories needs to be managed, and knowledge of existing profiles and newly defined genotypes should be indexed and shared. To meet this need, a web application called "MAC-INMV-SSR database" was developed. This freely accessible service allows users to compare MLVA and MLSSR subtype data of their strains with those of existing reference strains analyzed with the same genotyping methods.

Abstract: The upper airway epithelium, which is mainly composed of multiciliated, goblet, club and basal cells, ensures proper mucociliary function and can regenerate in response to assaults. In chronic airway diseases, defective repair leads to tissue remodeling. Delineating key drivers of differentiation dynamics can help understand how normal or pathological regeneration occurs. Using single-cell transcriptomics and lineage inference, we have unraveled trajectories from basal to luminal cells, providing novel markers for specific populations. We report that: (1) a precursor subgroup of multiciliated cells, which we have entitled deuterosomal cells, is defined by specific markers, such as DEUP1, FOXN4, YPEL1, HES6 and CDC20B; (2) goblet cells can be precursors of multiciliated cells, thus explaining the presence of hybrid cells that co-express markers of goblet and multiciliated cells; and (3) a repertoire of molecules involved in the regeneration process, such as keratins or components of the Notch, Wnt or BMP/TGFβ pathways, can be identified. Confirmation of our results on fresh human and pig airway samples, and on mouse tracheal cells, extend and confirm our conclusions regarding the molecular and cellular choreography at work during mucociliary epithelial differentiation., Highlighted Article: Single-cell RNAseq data in fresh human airway epithelial tissues and air liquid cultures identifies novel cell populations and offers new insights into the molecular mechanisms occurring during mucociliary epithelium regeneration.

Abstract: International audience; DprA, a ubiquitous multidomain DNA-binding protein, plays a crucial role during natural transformation in bacteria. Here, we carried out the structural analysis of DprA from the human pathogen Helicobacter pylori by combining data issued from the 1.8 Å resolution X-ray structure of the Pfam02481 domain dimer (RF), the NMR structure of the carboxy-terminal domain (CTD) and the low-resolution structure of the full-length DprA dimer obtained in solution by SAXS. In particular, we sought a molecular function for the CTD, a domain that we show here is essential for transformation in H. pylori. Albeit its structural homology to winged helix DNA binding motifs, we confirmed that the isolated CTD does not interact with ssDNA nor with dsDNA. The key R52 and K137 residues of RF are crucial for these two interactions. Search for sequences harboring homology to either HpDprA or Rhodopseudomonas palustris DprA CTDs led to the identification of conserved patches in the two CTD. Our structural study revealed the similarity of the structures adopted by these residues in RpDprA CTD and HpDprA CTD. Abstract: International audience; Cytokinins (CK) have been extensively studied for their roles in plant development. Recently, they also appeared to ensure crucial functions in the pathogenicity of some bacterial and fungal plant pathogens. Thus, identifying cytokinin-producing pathogens is a prerequisite to gain a better understanding of their role in pathogenicity. Taking advantage of the cytokinin perception properties of Malus domestica CHASE Histidine Kinase receptor 2 (MdCHK2), we thereby developed a selective and highly sensitive yeast biosensor for the application of cytokinin detection in bacterial samples. The biosensor is based on the mutated sln1Δ Saccharomyces cerevisiae strain expressing MdCHK2. The biosensor does not require any extraction or purification steps of biological samples, enabling cytokinin analysis directly from crude bacterial supernatants. For the first time, the production of cytokinin was shown in the well-known plant pathogenic bacteria Erwinia amylovora and was also revealed in human pathogens Staphylococcus aureus and Streptococcus agalactiae. Importantly, this biosensor was shown to be an efficient tool for unraveling certain steps in cytokinin biosynthesis by micro-organisms since this it was successfully used to unveil the role of ygdH22, a LOG-like gene, that is probably involved in cytokinin biosynthesis pathway in Escherichia coli. Overall, we demonstrated that our biosensor displays several advantages including time- and cost-effectiveness by allowing a rapid and specific detection of cytokinins in bacterial supernatants These results also support its scalability to high-throughput formats.

Abstract: The role of the recently described interleukin-32 (IL-32) in Staphylococcus aureus -induced mastitis, an inflammation of the mammary gland, is unclear. We determined expression of IL-32, IL-6, and IL-8 in S. aureus - and Escherichia coli -infected bovine mammary gland epithelial cells. Using live bacteria, we found that in S. aureus -infected cells, induction of IL-6 and IL-8 expression was less pronounced than in E. coli -infected cells. Notably, IL-32 expression was decreased in S. aureus -infected cells, while it was increased in E. coli -infected cells. We identified the staphylococcal phenol-soluble modulin (PSM) peptides as key contributors to these effects, as IL-32, IL-6, and IL-8 expression by epithelial cells exposed to psm mutant strains was significantly increased compared to that in cells exposed to the isogenic S. aureus wild-type strain, indicating that PSMs inhibit the production of these interleukins. The use of genetically complemented strains confirmed this observation. Inasmuch as the decreased expression of IL-32, which is involved in dendritic cell maturation, impairs immune responses, our results support a PSM-dependent mechanism that allows for the development of chronic S. aureus -related mastitis.

Abstract: International audience; Using a structure-based design approach, we have developed a new series of imidazo[1,2-b]pyridazines, targeting the calcium-dependent protein kinase-1 (CDPK1) from Toxoplasma gondii. Twenty derivatives were thus synthesized. Structure-activity relationships and docking studies confirmed the binding mode of these inhibitors within the ATP binding pocket of TgCDPK1. Two lead compounds (16a and 16f) were then identified, which were able to block TgCDPK1 enzymatic activity at low nanomolar concentrations, with a good selectivity profile against a panel of mammalian kinases. The potential of these inhibitors was confirmed in vitro on T. gondii growth, with EC50 values of 100 nM and 70 nM, respectively. These best candidates also displayed low toxicity to mammalian cells and were selected for further in vivo investigations on murine model of acute toxoplasmosis.

Abstract: International audience; Les phtalides sont des petites molécules de structure bicyclique de type 3H-isobenzofuran-1-ones isolées à partir de végétaux ou d’organismes marins (champignons, éponges…). Un grand intérêt est porté à cette classe de composés dû à leur large champ d’activités biologiques, dont antibactériennes. Parmi les phtalides naturels connus, la cytosporone E a particulièrement retenu notre attention. Isolée en 2000 d’un champignon endophyte, son potentiel antibactérien a été mis en évidence contre des bactéries Gram positif et négatif.Nous proposons une synthèse de la cytosporone E et d’analogues diversement fonctionnalisés en position 3. Leur activité antibactérienne est actuellement évaluée vis-à-vis d’une souche de Pseudomonas aeruginosa, une bactérie présentant une résistance croissante aux antibiotiques.

Abstract: It is a common view that assemblages of parasitic nematodes are influenced by climatic conditions; however, there are only a few articles available regarding those that infect farm animals. We investigated the relationship between climate variables and infection with abomasal trichostrongyles in 335 1-year-old rams grazed on steppe pastures in Eastern Algeria. Abomasa were collected from 12 local slaughterhouses located in four climatic areas (from humid to arid) and the worms extracted, identified and counted. The abundance was low and the fauna composed primarily of Teladorsagia circumcincta, Marshallagia marshalli and Trichostrongylus sp. The high percentage of M. marshalli is typical of steppe areas. Ostertagia ostertagi and Haemonchus contortus were present in low numbers. Rainfall was the most important climatic variable related to the main species. This relationship was not linear for M. marshalli but an optimal rainfall was detected (350–400 mm/year). The more complex climatic indicators used in the study did not demonstrate a more significant correlation than rainfall. The predictive value of rainfall on the abundance or proportion of species in the assemblage was modest but highly significant. The seasonality of assemblages was different between the two main sub-climates (sub-humid and semi-arid).

Abstract: We identified a Streptococcus agalactiae metabolic region (fru2) coding for a Phosphoenolpyruvate:carbohydrate phosphoTransferase System (PTS) homologous to the Frz system of extraintestinal pathogenic Escherichia coli strains. The Frz system is involved in environmental sensing and regulation of the expression of adaptation and virulence genes in E. coli. The S. agalactiae fru2 region codes three subunits of a PTS transporter of the fructose-mannitol family, a transcriptional activator of PTSs of the MtlR family, an allulose-6 phosphate-3-epimerase, a transaldolase and a transketolase. We demonstrated that all these genes form an operon. The fru2 operon is present in a 17494-bp genomic island. We analyzed by multilocus sequence typing a population of 492 strains representative of the S. agalactiae population and we showed that the presence of the fru2 operon is linked to the phylogeny of S. agalactiae. The fru2 operon is always present within strains of clonal complexes CC 1, CC 7, CC 10, CC 283 and singletons ST 130 and ST 288, but never found in other CCs and STs. Our results indicate that the fru2 operon was acquired during the evolution of the S. agalactiae species from a common ancestor before the divergence of CC 1, CC 7, CC 10, CC 283, ST 130 and ST 288. As S. agalactiae strains of CC 1 and CC 10 are frequently isolated from adults with invasive disease, we hypothesize that the S. agalactiae Fru2 system senses the environment to allow the bacterium to adapt to new conditions encountered during the infection of adults.

Abstract: Development of sub-unit mucosal vaccines requires the use of specific delivery systems or immune-modulators such as adjuvants to improve antigen immunogenicity. Nasal route for vaccine delivery by nanoparticles has attracted much interest but mechanisms triggering effective mucosal and systemic immune response are still poorly understood. Here we study the loading of porous nanoparticles (DGNP) with a total extract of Toxoplasma gondii antigens (TE), the delivery of TE by DGNP into airway epithelial, macrophage and dendritic cells, and the subsequent cellular activation. In vitro, DGNP are able to load complex antigens in a stable and quantitative manner. The outstanding amount of antigen association by DGNP is used to deliver TE in airway mucosa cells to induce a cellular maturation with an increased secretion of pro-inflammatory cytokines. Evaluation of nasal vaccine efficiency is performed in vivo on acute and chronic toxoplasmosis mouse models. A specific Th1/Th17 response is observed in vivo after vaccination with DGNP/TE. This is associated with high protection against toxoplasmosis regarding survival and parasite burden, correlated with an increased delivery of antigens by DGNP in airway mucosa cells. This study provides evidence of the potential of DGNP for the development of new vaccines against a range of pathogens.

Abstract: A common challenge in avian cell biology is the generation of differentiated cell-lines, especially in the keratinocyte lineage. Only a few avian cell-lines are available and very few of them show an interesting differentiation profile. During the last decade, mammalian embryonic stem cell-lines were shown to differentiate into almost all lineages, including keratinocytes. Although chicken embryonic stem cells had been obtained in the 1990s, few differentiation studies toward the ectodermal lineage were reported. Consequently, we explored the differentiation of chicken embryonic stem cells toward the keratinocyte lineage by using a combination of stromal induction, ascorbic acid, BMP4 and chicken serum. During the induction period, we observed a downregulation of pluripotency markers and an upregulation of epidermal markers. Three homogenous cell populations were derived, which were morphologically similar to chicken primary keratinocytes, displaying intracellular lipid droplets in almost every pavimentous cell. These cells could be serially passaged without alteration of their morphology and showed gene and protein expression profiles of epidermal markers similar to chicken primary keratinocytes. These cells represent an alternative to the isolation of chicken primary keratinocytes, being less cumbersome to handle and reducing the number of experimental animals used for the preparation of primary cells.

Abstract: International audience; An in vitro screening of the anti-apicomplexan activity of 51 compounds, stemming from our chemical library and from chemical synthesis, was performed. As a study model, we used Toxoplasma gondii (T. gondii), expressing β-galactosidase for the colorimetric assessment of drug activity on parasites cultivated in vitro. This approach allowed the validation of a new series of molecules with a biphenylimidazoazine scaffold as inhibitors of T. gondii growth in vitro. Hence, 8 molecules significantly inhibited intracellular replication of T. gondii in vitro, with EC50 < 1 μM, while being non-toxic for human fibroblasts at these concentrations. Most attractive candidates were then selected for further biological investigations on other apicomplexan parasites (Neospora caninum, Besnoitia besnoiti, Eimeria tenella and Plasmodium falciparum). Finally, two compounds were able to inhibit growth of four different apicomplexans with EC50 in the submicromolar to nanomolar range, for each parasite. These data, including the broad anti-parasite spectrum of these inhibitors, define a new generation of potential anti-parasite compounds of wide interest, including for veterinary application. Studies realized on E. tenella suggest that these molecules act during the intracellular development steps of the parasite. Further experiments should be done to identify the molecular target(s) of these compounds.

Abstract: Weaning has been described as one of the most stressful events in the life of horses. Given the importance of the interaction between the gut-brain axis and gut microbiota under stress, we evaluated (i) the effect of two different weaning methods on the composition of gut microbiota across time and (ii) how the shifts of gut microbiota composition after weaning affect the host. A total of 34 foals were randomly subjected to a progressive (P) or an abrupt (A) weaning method. In the P method, mares were separated from foals at progressively increasing intervals every day, starting from five min during the fourth week prior to weaning and ending with 6 h during the last week before weaning. In the A method, mares and foals were never separated prior to weaning (0 d). Different host phenotypes and gut microbiota composition were studied across 6 age strata (days −30, 0, 3, 5, 7, and 30 after weaning) by 16S rRNA gene sequencing. Results revealed that the beneficial species belonging to Prevotella, Paraprevotella, and Ruminococcus were more abundant in the A group prior to weaning compared to the P group, suggesting that the gut microbiota in the A cohort was better adapted to weaning. Streptococcus, on the other hand, showed the opposite pattern after weaning. Fungal loads, which are thought to increase the capacity for fermenting the complex polysaccharides from diet, were higher in P relative to A. Beyond the effects of weaning methods, maternal separation at weaning markedly shifted the composition of the gut microbiota in all foals, which fell into three distinct community types at 3 days post-weaning. Most genera in community type 2 (i.e., Eubacterium, Coprococcus, Clostridium XI, and Blautia spp.) were negatively correlated with salivary cortisol levels, but positively correlated with telomere length and N-butyrate production. Average daily gain was also greater in the foals harboring a community type 2 microbiota. Therefore, community type 2 is likely to cnfer better stress response adaptation following weaning. This study identified potential microbial biomarkers that could predict the likelihood for physiological adaptations to weaning in horses, although causality remains to be addressed. info:eu-repo/semantics/publishedVersion

Abstract: International audience; The current therapeutic arsenal for toxoplasmosis is restricted to drugs non-specific to the parasite which cause important side effects. Development of more efficient and specific anti-Toxoplasma compounds is urgently needed. Imidazo[1,2-b]pyridazines designed to inhibit the calcium-dependent protein kinase 1 of Toxoplasma gondii (TgCDPK1) and effective against tachyzoite growth in vitro at submicromolar ranges were modified into hydrochloride salts to be administered in vivo in a mouse model of acute toxoplasmosis. All protonated imidazo[1,2-b]pyridazine salts (SP230, SP231 and SP232) maintained their activity on TgCDPK1 and T. gondii tachyzoites. Rat and mouse liver microsomes were used to predict half-life and intrinsic clearance, and the pharmacokinetic profile of the most rapidly degraded imidazo[1,2b]pyridazine salt (SP230) was determined in serum, brain and lungs of mice after a single administration of 50 mg/kg. Compounds were then tested in vivo in a murine model of acute toxoplasmosis. Mice infected with tachyzoites of the ME49 strain of T. gondii were treated for 4, 7 or 8 days with 25 or 50 mg/kg/day of SP230, SP231 or SP232. The parasite burdens were strongly diminished (>90% reduction under some conditions) in the spleen and the lungs of mice treated with imidazo[1,2-b]pyridazine salts compared with untreated mice, without the need for pre-treatment. Moreover, no increases in the levels of hepatic and renal toxicity markers were observed, demonstrating no significant signs of short-term toxicity. To conclude, imidazo[1,2-b]pyridazine salts have strong efficacy in vivo on acute toxoplasmosis and should be further tested in a model of mouse congenital toxoplasmosis.

Abstract: International audience; We report the diagnostic sensitivity of 3 EU-approved rapid tests (ELISAs; 1 from IDEXX and 2 from Bio-Rad) for the detection of transmissible spongiform encephalopathy diseases in goats. Ninety-eight goat brainstem samples were tested. All the rapid tests had 100% specificity and ≥80% sensitivity, with the IDEXX test significantly more sensitive than the 2 Bio-Rad tests. All tests detected 100% of samples from goats with clinical scrapie, but missed 8% (IDEXX) to 33% (Bio-Rad SG) of samples from preclinical goats. Importantly, only IDEXX picked up all samples from clinical bovine spongiform encephalopathy (BSE)-infected goats, whereas the other 2 rapid tests missed 15% (Bio-Rad SG) to 25% (Bio-Rad SAP). These results show that a fraction of preclinical scrapie infections are likely missed by EU surveillance, with sensitivity of detection strongly dependent on the choice of the rapid test. Moreover, a significant proportion of clinical BSE infections are underestimated by using either Bio-Rad test. Assuming that the same sensitivity on preclinical goats would also occur in BSE-infected goats, our data suggest that IDEXX is likely the most sensitive test for detecting preclinical field cases of BSE infection in goats, although with an 8% failure rate. These results raise some concerns about the reliability of current EU surveillance figures on BSE infection in goats.

They were reported as carriers of extended-spectrum β-lactamase (ESBL) genes, and recently of carbapenemases, mostly carried by the Salmonella genomic island 1 (SGI1) and Proteus genomic island 1 (PGI1). Proteae have also lately become an increasing cause of UTIs in companion animals, but antimicrobial susceptibility data in animals are still scarce. Seventeen P. mirabilis strains (3.6%) were positive for SGI1/PGI1 and 18 Proteae (3.8%) were resistant to extended-spectrum cephalosporins (ESC). The 28 isolates carrying SGI1/PGI1 and/or ESC-resistance genes were isolated from cats, dogs, and horses. ESBL genes were detected in six genetically related P. mirabilis harboring blaV EB-6 on the SGI1-V variant, but also independently of the SGI1-V, in 3 P. mirabilis strains (blaVEB-6 and blaCTX-M-15) and 1 Providencia rettgeri strain (blaCTX-M-1). The AmpC resistance genes blaCMY -2 and/or blaDHA-16 were detected in 9 P. mirabilis strains. One strain presented both an ESBL and AmpC gene. Interestingly, the majority of the ESBL/AmpC resistance genes were located on the chromosome. In conclusion, multiple ESC-resistance genetic determinants are circulating in French animals, even though SGI1-V-carrying P. mirabilis seems to be mainly responsible for the spread of the ESBL gene blaVEB-6 in dogs and horses. These results are of public health relevance and show that companion animals in close contact with humans should be regarded as a potential reservoir of ESC-resistant bacteria as well as a reservoir of ESC-resistance genes that could further disseminate to human pathogens.

Abstract: National audience; Principles, constraints and research questions related to several perspectives of therapeutic alternatives to antimicrobials are presented. Different approaches are not new such as phytotherapy and phagotherapy that relies on the lytic properties of phages infecting bacteria, but their use in practice would require further research on quality, efficacy and security of the products, as well as regulatory developments. Other approaches are more recent and require further research before considering their application in the field. This is the case for host defence peptides, molecules of innate immunity that are mainly produced by epithelial cells and phagocytes; they have large spectrum antimicrobial properties and immunity regulation capacity. There are few industrial developments due to technical problems despite a high number of candidates. Another approach concerns quorum sensing, based on signalling molecules produced by bacteria, that allows coordinated changes of the bacteria properties above a certain threshold, such as producing virulent factors or biofilms. Quorum quenching might allow regulating these properties.

Abstract: National audience; Reversible posterior leukoencephalopathy syndrome (RPLS) is characterised by clinical neurological features of sudden onset and brain MRI findings such as T2/Flair white matter hyperintensities. RPLS can occur in autoimmune diseases, and rarely in systemic vasculitis. We report a case of RPLS in a woman presenting granulomatosis with polyangiitis (Wegener's granulomatosis). A 22-year-old female patient was treated with methylprednisolone pulses for granulomatosis with polyangiitis and neurological impairment. A few hours after the second pulse, the patient had seizures, blindness and confusion associated with high blood pressure and acute renal failure. MRI revealed a high-intensity area on T2-Flair weighted images of the occipital-temporal lobes. The patient was treated with antiepileptic and antihypertensive medications, oral steroids and cyclophosphamide; the clinical and radiological findings proved reversible over the ensuing days. The occurrence of RPLS in systemic vasculitis is rare. Six cases of RPLS associated with granulomatosis and polyangiitis have been reported. It appears important to screen for high blood pressure in patients recently treated with corticosteroids for vasculitis as this condition may represent a precipitating factor for RPLS.; Le syndrome d’encéphalopathie postérieure réversible (PRES) associe des symptômes neurologiques d’apparition brutale à des hypersignaux de la substance blanche à l’IRM cérébrale en T2 Flair. Le PRES peut survenir au cours de maladies auto-immunes, rarement au cours de vascularites systémiques. Nous rapportons un cas de PRES au cours d’une granulomatose avec polyangéite (ex-maladie de Wegener).Une patiente de 22ans était traitée par bolus de méthylprednisolone pour une granulomatose avec polyangéite avec atteinte neurologique. Dans les suites du deuxième bolus, la patiente présentait des crises convulsives, une cécité, un syndrome confusionnel associés à une hypertension artérielle et une insuffisance rénale aiguë. L’IRM cérébrale montrait des hypersignaux T2 Flair occipitotemporaux bilatéraux. Les signes cliniques et radiologiques étaient rapidement résolutifs suite à un traitement associant antiépileptiques, antihypertenseurs, corticothérapie per os et cyclophosphamide.La survenue de PRES au cours de vascularites systémiques est rare et six cas de PRES associés à une granulomatose avec polyangéite ont été antérieurement rapportés. Il semble important de dépister l’existence d’une hypertension artérielle chez un patient traité par corticothérapie pour une vascularite systémique, car ceci pourrait constituer un facteur favorisant de survenue du PRES.
