Here, we present a retrospective study on honey bee brood diseases in the USA. The Bee Disease Diagnostic Laboratory (BDDL) in the USDA-ARS Beltsville Bee Research Laboratory received a total of 4790 brood samples between 2015 and 2022 from U.S. State Apiary Inspectors and beekeepers. Samples from 49 states were analyzed by microscopy for the presence and prevalence of two bacteria, Melissococcus plutonius and Paenibacillus larvae, causing European foulbrood (EFB) and American foulbrood (AFB) diseases, respectively. Samples that tested positive for AFB were cultured and subjected to antibiotic susceptibility tests via the agar disc diffusion method to determine their resistance to tetracycline (Terramycin®) and tylosin (Tylan®) antibiotics. A comprehensive data analysis was conducted at multiple levels, including state, month, year, nationwide, and climate region. Among the 49 states examined, EFB was identified in 44 states, while AFB was found in 31 states. Infection levels vary across states, ranging from 0% to 63.6% for EFB and from 0% to 54.8% for AFB. The national average of EFB (19.2%) was significantly (p < 0.001) higher than AFB (8.6%). Contrary to AFB, significant yearly increases (p < 0.001) were identified for EFB from 2015 to 2022. Furthermore, significant monthly variations were recorded for both brood diseases, with the highest occurrence of EFB observed from April to July. States with the lowest infection rates of both diseases were NV, ND, MS, AK, and AZ. The South U.S. climate region, which comprises six states, had the highest and lowest EFB and ABF infections, respectively. Tylosin is an important macrolide antibiotic produced by Streptomyces fradiae. In the biosynthesis of tylosin, macrocin O-methyltransferase TylF catalyzes the conversion of the side-product tylosin C (macrocin) to the primary component tylosin A (C/A conversion). This conversion is the rate-limiting step in the biosynthesis of tylosin, and affects the quality of the end product. To find a high activity and environment-adapted TylF enzyme, a TylF variant pool has been constructed via protein evolution approach in our previous study (Fan et al., 2023 [41]). In this study, the TylF variants with higher C/A conversion rates were expressed in E. coli and purified. The variants TylFY139F, TylFQ138H, F232Y and TylFT36S, V54A were shown to have a higher C/A conversion rate at 30 °C than that of TylF at 38 °C. Moreover, they had a greater acid resistance and showed more adaptable to the pH change during fermentation. Further protein structural and substrate-binding affinity analyses revealed that the T36S, V54A, Q138H, Y139F, and F232Y mutations enlarged the volume of the substrate-binding pocket, thereby increasing the affinity of enzyme variants for their substrates of SAM and macrocin, and decreasing the inhibition of SAH. Three of the TylF variants were overexpressed in the industrial tylosin-producing S. fradiae strain, and the recombinant strains showed the highest C/A conversion at 30 °C without heating up to 38 °C during the last 24 h of fermentation. This is of great energy-saving significance for tylosin industrial production.

Abstract Mathematical manipulation technique has proven to be a very powerful tool for efficient processing and handling of highly overlapped spectra. This work introduced two green and sustainable approaches for the successful recovery of the ternary mixture, Tylosin tartarate (TYL), Sulfadimdine (SLD), and Trimethoprim (TRI). The approaches were constructed using three different methods, derivative ratio spectrum zero-crossing method (DRSZ), double divisor ratio spectra derivative method (DDRD), and factorized derivative ratio method coupled with spectrum subtraction (FDRM-SS). The two approaches succeeded in recovering the three drugs (Linearity range achieved were 0.5–5 µg/mL for TYL, 0.3–1.3 µg/mL for SLD, and 0.3–5 µg/mL for TRI), giving convenient standard deviations and satisfactory recovery percentages. The recommended methods have been verified in accordance with (ICH) guidelines. When the results were statistically compared to the official methods, no significant difference was found. Both AGREE—Analytical GREEnness Metric Approach and Software, and White Analytical Chemistry (WAC) RGB model gave scores of 0.93 and 97.2%, respectively, which proved that the approaches were eco-friendly and abiding by the sustainability principles.

Abstract Aim of the work The study was conducted to evaluate the influence of theophylline pre-treatment on serum pharmacokinetics and milk elimination of tylosin following single intramuscular (IM) administrations in lactating goats. Methods and results In a cross-over study, tylosin was injected via intramuscular (IM) at a single dose of 15 mg/kg b.wt. After a one-month washout period goats received theophylline at a daily IM dose of 2 mg/kg b.wt. for seven consecutive days then tylosin was injected IM dose of 15 mg/kg b.wt. two hours after the last theophylline dosing. Blood samples were collected before and at 0.25, 0.5, 0.75, 1, 2, 4, 6, 8, 10, 12, and 24 h post-injection. Samples were left to clot and then centrifuged to yield serum. Milk samples were collected before and at 0.5, 1, 2, 4, 6, 8, 10, 12, 24, 48, and 72 h post-injection from each goat by hand milking. Tylosin serum concentrations were determined by high-performance liquid chromatography (HPLC). Tylosin concentrations versus time were analyzed by a noncompartmental method. Tylosin Cmax significantly declined from 1.73 ± 0.10 to 1.01 ± 0.11 µg/ml, and attained Tmax values of 2 and 1 h, respectively in theophylline-pretreated goats. Moreover, theophylline pretreatment significantly shortened the elimination half-life (t1/2el) from 6.94 to 1.98 h, t1/2ka from 0.62 to 0.36 h and the mean residence time (MRT) from 8.02 to 4.31 h, also Vz/F and AUCs decreased from 11.91 to 7.70 L/kg and from 12.64 to 4.57 µg*h/ml, respectively, consequently, theophylline enhanced the clearance (Cl/F) of tylosin from the body. Similarly, tylosin milk concentrations were significantly lower in theophylline-pretreated goats than in goats that received tylosin alone and were detected up to 24 and 72 h in both groups, respectively. Moreover, the t1/2el and AUCs were significantly decreased from 14.68 ± 1.97 to 4.72 ± 0.48 h, and from 181 to 67.20 µg*h/ml, respectively. Conclusions The withdrawal period for tylosin in goat milk is at least 72 h. Theophylline pretreatment significantly decreases serum and milk tylosin concentrations to subtherapeutic levels, which could have serious clinical consequences such as failure of therapy. This means that after administering tylosin to goats, milk from these animals should not be consumed for at least 96 h to ensure that the milk is free from residues of the antibiotic.

Polyketides are natural products synthesized by polyketide synthases (PKSs), where acyltransferase (AT) domains play a crucial role in selection of extender units. Engineering of AT domains enables the site‐specific incorporation of non‐natural extender units, leading to generation of novel derivatives. Here, we determined the crystal structures of AT domains from the fifth module of tylosin PKS (TylAT5) derived from Streptomyces fradiae and the eighth module of spinosad PKS (SpnAT8) derived from Saccharopolyspora spinosa, and combined them with molecular dynamics simulations and enzyme kinetic studies to elucidate the molecular basis of substrate selection. The ethylmalonyl‐CoA‐specific conserved motif TAGH of TylAT5 and the MMCoA‐specific conserved motif YASH of SpnAT8 were identified within the substrate‐binding pocket, and several key residues close to the substrate acyl moiety were located. Through site‐directed mutagenesis of four residues near the active site, we successfully reprogrammed the specificity of these two AT domains toward malonyl‐CoA. Mutations in TylAT5 enhanced its catalytic activity 2.6‐fold toward malonyl‐CoA, and mutations in SpnAT8 eliminated the substrate promiscuity. This study investigated batch-fed vermicomposting of cow manure, with a specific focus on assessing the effects of tylosin on the weight of earthworms and the overall quality of the resulting manure. Five reactors, including three concentrations of tylosin (50, 100, and 150 mg/kg) and two control reactors, were employed. Residual tylosin concentrations were measured using high-performance liquid chromatography (HPLC). Quality parameters such as pH, temperature, volatile solids (VS), organic carbon content (OCC), electrical conductivity (EC), ash content, C/N ratio, total Kjeldahl nitrogen (TKN), and microbial content were evaluated. The toxicity and maturity of vermicompost were assessed by determining the germination index (GI). The study also monitored variations in the earthworm's weight. The results demonstrated a decreasing trend in VS, OCC, C/N, and fecal coliforms, along with increased pH, EC, ash content, and TKN during the vermicomposting process. Furthermore, investigations revealed significant reductions in the reactors with tylosin concentrations of 50, 100, and 150 mg/kg, resulting in the removal of 98%, 90.48%, and 89.38% of the initial tylosin, respectively. This result confirms the faster removal of tylosin in reactors with lower concentrations. Degradation of tylosin also conforms to first-order kinetics. The findings showed a significant influence of tylosin on the weight of Eisenia fetida earthworms and the lowest antibiotic concentration led to the highest weight gain. Microbial degradation of tylosin (TYL) is a safe and environmentally friendly technology for remediating environmental pollution. Kurthia gibsonii (TYL-A1) and Klebsiella pneumonia (TYL-B2) were isolated from wastewater; degradation efficiency of the two strains combined was significantly greater than either alone and resulted in degradation products that were less toxic than TYL. With Polyvinyl alcohol (PVA)—sodium alginate (SA)—activated carbon (AC) used to form a bacterial immobilization carrier, the immobilized bacterial alliance reached 95.9% degradation efficiency in 1 d and could be reused for four cycles, with > 93% degradation efficiency per cycle. In a wastewater application, the immobilized bacterial alliance degraded 67.0% TYL in 9 d. There were significant advantages for the immobilized bacterial alliance at pH 5 or 9, with 20 or 40 g/L NaCl, or with 10 or 50 mg/L doxycycline. In summary, in this study, a bacterial consortium with TYL degradation ability was constructed using PVA-SA-AC as an immobilized carrier, and the application effect was evaluated on farm wastewater with a view to providing application guidance in environmental remediation. Corynebacterium pseudotuberculosis causes infection in goats, sheep, cattle, and humans. The disease imposing huge economic losses such as reducing milk, meat, and wool production and downgrading and condemnation of affected carcass to goat-rearing industry. Despite all its devastating effects, no effective antimicrobial was suggested for the treatment of this pathogen due to its highly resistant nature. Development and utilization of nanoparticles has paved the way to revive the efficacy of conventional antibiotics. Encapsulation of antibiotics within liposome as a nanoparticle may present a potential approach for successful treatment of resistant bacterial infections. The aim of this research was therefore to evaluate the antibacterial activity of free and liposome-encapsulating tylosin against Corynebacterium pseudotuberculosis. The minimum inhibitory concentration of free and liposome-encapsulated tylosin was determined by microbroth dilution and resazurin colorimetric-based methods. The antibiofilm activity of free and liposomal tylosin was assessed by crystal violet and resazurin techniques. Acridine orange/ethidium bromide staining assay was performed to assess the effect of free and liposomal tylosin on pre-formed biofilm of Corynebacterium pseudotuberculosis. The findings demonstrated minimum inhibitory concentration value of 1 µg/mL and 32 µg/mL for free and liposomal tylosin, respectively. The biofilm inhibitory data showed that free tylosin inhibited the formation of Corynebacterium pseudotuberculosis at 2 µg/mL, while this concentration was 64 µg/mL for liposomal tylosin. The antibiofilm concentration was 1024 µg/mL and 2048 µg/mL for liposome-encapsulated and free tylosin, respectively. It can be concluded that liposome as a nanocarrier can offer an excellent system of drug delivery for enhancing the efficacy of antibiotics against resistant form of Corynebacterium pseudotuberculosis. Liver abscesses (LA) resulting from bacterial infection in cattle pose a significant global challenge to the beef and dairy industries. Economic losses from liver discounts at slaughter and reduced animal performance drive the need for effective mitigation strategies. Tylosin phosphate supplementation is widely used to reduce LA occurrence, but concerns over antimicrobial overuse emphasize the urgency to explore alternative approaches. Understanding the microbial ecology of LA is crucial to this, and we hypothesized that a reduced timeframe of tylosin delivery would alter LA microbiomes. We conducted 16S rRNA sequencing to assess severe liver abscess bacteriomes in beef cattle supplemented with in-feed tylosin. Our findings revealed that shortening tylosin supplementation did not notably alter microbial communities. Additionally, our findings highlighted the significance of sample processing methods, showing differing communities in bulk purulent material and the capsule-adhered material. Fusobacterium or Bacteroides ASVs dominated LA, alongside probable opportunistic gut pathogens and other microbes. Moreover, we suggest that liver abscess size correlates with microbial community composition. These insights contribute to our understanding of factors impacting liver abscess microbial ecology and will be valuable in identifying antibiotic alternatives. Abstract Background Toxoplasma gondii is an important protozoan pathogen with medical and veterinary importance worldwide. Drugs currently used for treatment of toxoplasmosis are less effective and sometimes cause serious side effects. There is an urgent need for the development of more effective drugs with relatively low toxicity. Methods The effect of tylosin on the viability of host cells was measured using CCK8 assays. To assess the inhibition of tylosin on T. gondii proliferation, a real-time PCR targeting the B1 gene was developed for T. gondii detection and quantification. Total RNA was extracted from parasites treated with tylosin and then subjected to transcriptome analysis by RNA sequencing (RNA-seq). Finally, murine infection models of toxoplasmosis were used to evaluate the protective efficacy of tylosin against T. gondii virulent RH strain or avirulent ME49 strain. Results We found that tylosin displayed low host toxicity, and its 50% inhibitory concentration was 175.3 μM. Tylsoin also inhibited intracellular T. gondii tachyzoite proliferation, with a 50% effective concentration of 9.759 μM. Transcriptome analysis showed that tylosin remarkably perturbed the gene expression of T. gondii, and genes involved in “ribosome biogenesis (GO:0042254)” and “ribosome (GO:0005840)” were significantly dys-regulated. In a murine model, tylosin treatment alone (100 mg/kg, i.p.) or in combination with sulfadiazine sodium (200 mg/kg, i.g.) significantly prolonged the survival time and raised the survival rate of animals infected with T. gondii virulent RH or avirulent ME49 strain. Meanwhile, treatment with tylosin significantly decreased the parasite burdens in multiple organs and decreased the spleen index of mice with acute toxoplasmosis. Conclusions Our findings suggest that tylosin exhibited potency against T. gondii both in vitro and in vivo, which offers promise for treatment of human toxoplasmosis. Graphical Abstract

ABSTRACTTylosin (TYL) is a kind of antibiotics which promotes the development of animal husbandry, but it poses potential threats to food safety and human health. Therefore, it is necessary to establish a highly sensitive method for the detection of TYL. Here, an enzyme-linked immunosorbent assay (ELISA) combined with biotin–streptavidin (Bio-SA) system and hybridization chain reaction (HCR) technique (Bio-SA-HCR-ELISA) was developed. In the absence of the TYL, a strong fluorescence was generated through double signal amplification as signal output, while in the presence of TYL, the fluorescence intensity decreased. The results showed that the limit of detection for TYL was 0.39 ng/mL, and this method had a good specificity for 9 common antiboics. Compared to the traditional indirect competitive ELISA, the sensitivity of this method was 6.8 times higher. Additionally, the established double signal amplification Bio-SA-HCR-ELISA was used for detecting TYL in milk and honey, the LOD of this method were 4.9 and 2.5 ng/mL, respectively.

Broad-spectrum veterinary antibiotic tylosin (TYL) is utilized in livestock and poultry. Due of its extensive usage and animal excretion in urine and feces, it affects the environment and human health. For determining the equilibrium time, the adsorption maximum capacity and the optimal isotherm model, adsorption batch tests have been conducted. Sips and Pseudo-second order models had R2 values of 0.9937 and 0.9764, respectively, and best matched experimental isotherm and kinetic data. The maximum TYL uptake onto soil was 2355.08 mg.kg−1. Using fixed bed adsorption experiments, the current study clarifies the issue of soil contamination in conditions that are close to natural phenomena. Fixed bed adsorption tests demonstrated that the inlet TYL concentration, soil bed height, and flow rate, influenced saturation and breakthrough times. Bohart-Adams, Thomas, and Yoon-Nelson models matched the experimental findings well in a variety of conditions. With 100 mg.L−1 concentration, 4 cm bed height, and 5 mL.min−1 flow rate, the maximal bed capacity of 1277.06 mg.kg−1 was achieved with R2 value of 0.9797. Experimental findings also indicated that the adsorption capacity in batch and dynamic tests are of the same range of magnitude. Based on this, tylosin is a probable soil contaminant. In a murine model, tylosin treatment alone (100 mg/kg, i.p.) or in combination with sulfadiazine sodium (200 mg/kg, i.g.) significantly prolonged the survival time and raised the survival rate of animals infected with T. gondii virulent RH or avirulent ME49 strain. Meanwhile, treatment with tylosin significantly decreased the parasite burdens in multiple organs and decreased the spleen index of mice with acute toxoplasmosis. The paper aims to develop a method to rapidly detect the content of tylosin tartrate. In Clark-Lubs buffer solution with pH 6.6, malachite green can induce strong resonance light scattering due to the construction of supermolecule aggregates, and tylosin tartrate can react with malachite green cations, resulting in the decomposition of supermolecule aggregates constructed by malachite green, which significantly weakens the resonance light scattering intensity of the system. The maximum detection wavelengths are at 285 nm and 332 nm. Due to the additive nature of the resonance light intensity ( I), the two wavelengths are superimposed for measurement. The linear range of tylosin tartrate is 0.2-0.6 mg/L, the linear regression equation is IRLS= 5 299 C-766.4, the correlation coefficient ( R) is 0.999 8, and the detection limit is 0.004 1 mg/L. A fluorescence polarization immunoassay method has been developed for the determination of antibiotic tylosin in honey. For this purpose, rabbits were immunized with a conjugate of tylosin and bovine serum albumin, and antisera were obtained. The conditions for the interaction of antisera in a competitive format with tylosin in the sample and with tylosin labeled with fluorescein were optimized. The developed method is characterized by a limit of detection for tylosin of 34.7 ng/mL and an analytical range of 65.2 to 564.0 ng/mL. High reproducibility of measurements using a portable detector was demonstrated, with relative standard deviation (RSD) values of 1.5–2.5%. To avoid false negative results due to the low cross-reactivity rate (CR) in rapid immunoassay, a group-specific antibody with homogeneous CR toward target compounds is needed for accuracy. In this study, tylosin (TYL) and tilmicosin (TM) were selected as model molecules. Firstly, two-dimensional similarity, electrostatic potential energy, spatial conformation and charge distribution of the haptens TYL-CMO, TYL-6-ACA, TYL-4-APA, TYL-CHO and DES-CMO and target compounds of TYL and TM were obtained using Gaussian 09W and Discovery Studio. The optimal hapten was DES-CMO because it is the most similar to TYL and TM. Subsequently, the mAb 14D5 cell line was obtained with IC50 values of 1.59 and 1.72 ng/mL for TYL and TM, respectively, and a CR of 92.44%. Finally, amorphous carbon nanoparticles (ACNPs) were conjugated with mAb 14D5 to develop an accurate lateral flow immunoassay (LFA) for detection of TYL and TM by the reflectance value under natural light. The recoveries of TYL and TM ranged from 77.18 to 112.04% with coefficient of variation < 13.43%. The cut-off value in milk samples was 8 ng/mL, and the limits of detection were 11.44, 15.96, 22.29 and 25.53 μg/kg for chicken muscle, bovine muscle, porcine muscle and porcine liver samples, respectively, and the results being consistent with HPLC–UV. Swine dysentery, caused by Brachyspira hyodysenteriae and the newly recognized Brachyspira hampsonii in grower-finisher pigs, is a substantial economic burden in many swine-rearing countries. Antimicrobial therapy is the only commercially available measure to control and prevent Brachyspira -related colitis. However, data on antimicrobial susceptibility trends and genetic diversity of Brachyspira species from North America is limited. We evaluated the antimicrobial susceptibility profiles of U.S. Brachyspira isolates recovered between 2013 and 2022 to tiamulin, tylvalosin, lincomycin, doxycycline, bacitracin, and tylosin. In addition, we performed multilocus sequence typing (MLST) on 64 B. hyodysenteriae isolates. Overall, no distinct alterations in the susceptibility patterns over time were observed among Brachyspira species. However, resistance to the commonly used antimicrobials was seen sporadically with a higher resistance frequency to tylosin compared to other tested drugs. B. hampsonii was more susceptible to the tested drugs than B. hyodysenteriae and B. pilosicoli. MLST revealed 16 different sequence types (STs) among the 64 B. hyodysenteriae isolates tested, of which 5 STs were previously known, whereas 11 were novel. Most isolates belonged to the known STs: ST93 (n = 32) and ST107 (n = 13). Our findings indicate an overall low prevalence of resistance to clinically important antimicrobials other than tylosin and bacitracin, and high genetic diversity among the clinical Brachyspira isolates from pigs in the United States during the past decade. Further molecular, epidemiologic, and surveillance studies are needed to better understand the infection dynamics of Brachyspira on swine farms and to help develop effective control measures. This work compared the antibacterial activity and in vivo effects of nitazoxanide and tylosin against infected broilers with antibiotic resistant Clostridium perfringens field strain to control necrotic enteritis disease. Both in vitro and in vivo assessment studies were used. Firstly, C. perfringens was isolated with a total rate of 46% from liver and intestine samples of diseased broilers. By using PCR molecular typing all tested isolates were type A (produced only alpha-toxin (cpa virulence gene)) and included Bla and tetK (resistant genes). Using antibiotic sensitivity test they showed multi-drugs resistance against amoxiclave, tetracycline, gentamicin, clindamycin, sulphamethoxazole-trimethoprim and cefoxitin. Minimal inhibitory and bactericidal concentrations (µg/ml) were 0.4 and 12 for nitazoxanide and 0.7 and 49 for tylosin respectively. Then, a total of 90 one-day-old-chicks were divided into 6 groups; G1: negative-control, G2: infected-control, G3: non-infected (nitazoxanide 15.4 mg/kg body weight), G4: non-infected (tylosin 20 mg/kg body weight), G5: infected-nitazoxanide (15.4 mg/kg), and G6: infected-tylosin (20 mg/kg). Treatments lasted for 5 days in drinking water. No adverse effects on liver or kidney parameters were recorded in non-infected treated groups. Both treatments overcome the infection signs, C. perfringens count and revealed a significant improvement in most of inflammatory and biochemical parameters to their normal levels especially, G5 reflected a significant increase in total protein, albumin, globulin while reduced alanine and aspartate aminotransferase and gamma-glutamyl transferase activities, C-reactive protein, uric acid, and creatinine levels than G2. This paper introduces a novel, sensitive, and rapid method for the quantification of oxytetracycline, tetracycline, tilmicosin, and tylosin residues in cow's milk. The method involves a two-step process of extraction and detection. The extraction process uses acetonitrile and salting-out assisted liquid-liquid extraction to isolate the antibiotics from the milk. The detection process employs Liquid Chromatography coupled with photo-diode array detector (PDA) to quantify the antibiotics. The method has been successfully applied to milk samples, demonstrating its effectiveness and potential for widespread use in residue analysis. • The calibration curves for the antibiotics were found to be linear within the range of 0.06–3.0 µg/mL to 0.1–3.0 µg/mL. • The limits of detection for oxytetracycline, tetracycline, tilmicosin, and tylosin were 0.03 µg/mL, 0.02 µg/mL, 0.04 µg/mL, and 0.02 µg/mL respectively. • The method demonstrated an average recovery rate of over 90% from milk samples with peak values reaching up to 0.100–0.200 µg/mL.

Four groups of 36 days old chicks were infected experimentally with mycoplasma ‎gallisepticun suspension while a control group was injected with nutrient broth and ‎another control group was left uninfected Thtee of the infected groups were treated with ‎either Tylosin Spiramycin or lincomycin.‎ Parameters of airsacculitis score, and weight gain food conversion were used to evaluate ‎the treatment.‎ Also, the residues of tylosin in blood plasm nd muscles and the level of spiramycin in blood ‎plasma was estimated by a microbiological assay.‎ The results showed significant differences between infected and non-infected ‎groups with all parameters used Treatment with tylosin and apramycin improved weight ‎gain and increased food consumption but did not reduce airsacculitis score. In the last few decades, sulfonated carbon materials have garnered significant attention as Brønsted solid acid catalysts. The sulfonation process and catalytic activity of sulfonated biochar can be influenced by the aromaticity and degree of condensation exhibited by biochar. However, the relationships between the aromaticity, sulfonating ability, and resultant catalytic activity are not fully understood. In this study, biochar samples pyrolyzed at 300–650 °C exhibiting different aromaticity and degrees of condensation were sulfonated and employed as sulfonate-bearing solid catalysts for hydrolytically removing tylosin. They exhibited excellent hydrolytic performance and their kinetic constants were positively correlated with the total acidity and negatively correlated with their aromaticity. This study has uncovered the relationship between the structure, properties, sulfonating ability, and subsequent hydrolytic performance of biochar samples. It was observed that the aromaticity of biochar decreased as the pyrolysis temperature increased. Lower pyrolysis temperatures resulted in a reduced degree of condensation, smaller ring size, and an increased number of ring edge sites available for sulfonation, ultimately leading to enhanced catalytic performance. These findings provide valuable insights into the fundamental chemistry behind sulfonation upgrading of biochar, with the aim of developing functional catalysts for mitigating antibiotics in contaminated water. Industrial wastewater from drug production is one of the contributors to water pollution. For drug wastewater treatment, photodegradation-based chemical technology has gained more attention because of the drug's microbicidal nature and stability. A zinc-chromium-nickel trimetallic–layered double hydroxide compounding with graphene oxide catalyst (ZnCrNi/GO) was synthesized and exhibited a clustered crumb sheet morphology. The prepared catalyst was characterized by a scanning electron microscope (SEM) equipped with an energy-dispersive X-ray spectrometer (EDS), Fourier transform infrared (FTIR) spectra, and X-ray photoelectron spectroscopy (XPS). The results of material analysis established the crystallographic structures of catalysts and evidenced the successful synthesis. The ZnCrNi/GO nanohybrid revealed a higher activity of approximately 90% degradation of tolysin under high-pressure mercury lamp irradiation. The optimized condition of the catalyst dosage of 500 mg/L and the natural pH of the solution at 7.0 under the tylosin concentration of 10 mg/L with high photocatalytic efficiency was explored. In addition, the main reactive species involved in this photocatalysis degradation were explored as the active cavity h+ and ·O2− to a certain extent by the radical trapping experiments. Reuse experiments have shown that as-prepared catalysts possessed the properties of high efficiency and long-lasting catalytic performance, which could meet pharmaceutical wastewater treatment. A three-metal–layered double hydroxide composed by the metal of Ni, Zn, and Cr was synthesized and attached onto graphene oxide. The catalytic materials obtained in this way have a significant catalysis efficiency to tylosin with the likely degradation mechanism of the active cavity h+ and the oxidative capacity of hydroxyl radials. Weaning is a critical period in raising pigs. Novel animal feed additives that promote gut health and regulate immune function of piglets without antibiotics are needed. In this study, we aimed to test the ability of mesobiliverdin IXα‐enriched microalgae (MBV IXα‐enriched microalgae) to eliminate reliance on antibiotics to promote intestinal health in piglets. Eighty 28‐day‐old weaned piglets were randomly allocated to four groups each with four replicate pens and five piglets per pen. The dietary treatments were a basal diet as control (NC), basal diet plus 0.05% tylosin (PC), basal diet plus 0.1% or 0.5% MBV IXα‐enriched microalgae as low (MBV‐SP1) or high (MBV‐SP2) dose respectively. All treated animals showed no significant differences in live weight, average daily gain and feed efficiency compared to control animals. Histological examination showed that MBV‐SP1 and particularly MBV‐SP2 increased the ratio of villus height to crypt depth in the jejunum and ileum compared to NC (p < 0.05). Similarly, tylosin treatment also increased villi lengths and the ratio of villus height to crypt depth in the jejunum and ileum compared to the NC (p < 0.05). MBV‐SP1 and particularly MBV‐SP2 reduced the levels of inflammatory cytokines interleukin‐6 and tumour necrosis factor‐alpha in the small intestine. MBV‐SP2 and tylosin similarly reduced the lipid peroxidation marker (TBARS value) in the duodenum and ileum. In conclusion, feed supplementation with MBV IXα‐enriched microalgae improved gut health by villus height and production of immunomodulators that correlated with down‐regulated secretion of inflammatory cytokines. Formulating a therapeutic strategy that can effectively combat concurrent infections of Actinobacillus pleuropneumoniae (A. pleuropneumoniae) and Pasteurella multocida (P. multocida) can be challenging. This study aimed to 1) establish minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), time kill curve, and post-antibiotic effect (PAE) of tylosin against A. pleuropneumoniae and P. multocida pig isolates and employ the MIC data for the development of epidemiological cutoff (ECOFF) values; 2) estimate the pharmacokinetics (PKs) of tylosin following its intramuscular (IM) administration (20 mg/kg) in healthy and infected pigs; and 3) establish a PK-pharmacodynamic (PD) integrated model and predict optimal dosing regimens and PK/PD cutoff values for tylosin in healthy and infected pigs. The MIC of tylosin against both 89 and 363 isolates of A. pleuropneumoniae and P. multocida strains spread widely, ranging from 1 to 256 µg/mL and from 0.5 to 128 µg/mL, respectively. According to the European Committee on Antimicrobial Susceptibility Testing (EUCAST) ECOFFinder analysis ECOFF value (=64 µg/mL), 97.75% (87 strains) of the A. pleuropnumoniae isolates were wild-type, whereas with the same ECOFF value (=64 µg/mL), 99.72% (363 strains) of the P. multicoda isolates were considered wild-type to tylosin. Area under the concentration time curve (AUC), T1/2, and Cmax values were significantly greater in healthy pigs than those in infected pigs (13.33 h × µg/mL, 1.99 h, and 5.79 µg/mL vs. 10.46 h × µg/mL, 1.83 h, and 3.59 µg/mL, respectively) (p < 0.05). In healthy pigs, AUC24 h/MIC values for the bacteriostatic activity were 0.98 and 1.10 h; for the bactericidal activity, AUC24 h/MIC values were 1.97 and 1.99 h for A. pleuropneumoniae and P. multocida, respectively. In infected pigs, AUC24 h/MIC values for the bacteriostatic activity were 1.03 and 1.12 h; for bactericidal activity, AUC24 h/MIC values were 2.54 and 2.36 h for A. pleuropneumoniae and P. multocida, respectively. Monte Carlo simulation lead to a 2 µg/mL calculated PK/PD cutoff. Managing coinfections can present challenges, as it often demands the administration of multiple antibiotics to address diverse pathogens. However, using tylosin, which effectively targets both A. pleuropneumoniae and P. multocida in pigs, may enhance the control of bacterial burden. By employing an optimized dosage of 11.94-15.37 mg/kg and 25.17-27.79 mg/kg of tylosin can result in achieving bacteriostatic and bactericidal effects in 90% of co-infected pigs. The removal of antibiotics from wastewater to prevent their environmental accumulation is significant for human health and ecosystems. Herein, iron (Fe)-atom-doped anatase TiO2 nanofibers (Fe-TNs) were manufactured for the photocatalytic Fenton-like decomposition of tylosin (TYL) under LED illumination. Compared with the pristine TiO2 nanofibers (TNs), the optimized Fe-TNs exhibited improved visible-light-driven photocatalytic Fenton-like activity with a TYL degradation efficiency of 98.5% within 4 h. The effective TYL degradation could be attributed to the expanded optical light absorption and accelerated separation and migration of photogenerated electrons and holes after the introduction of Fe. The photogenerated electrons were highly conducive to the generation of active SO4•− radicals as they facilitated Fe(III)/Fe(II) cycles, and to oxidizing TYL. Moreover, the holes could be involved in TYL degradation. Thus, a significant enhancement in TYL degradation could be achieved. This research verifies the use of iron-doped anatase nanofibers as an effective method to synthesize novel photocatalytic Fenton-like catalysts through surface engineering for wastewater remediation. Streptomyces fradiae sf106 is a type of actinomycete that can produce abundant secondary metabolites, making it a natural cell factory for drug synthesis. In order to comprehensively understand the genomic profile of Streptomyces fradiae sf106 and its potential for producing secondary metabolites, a combination of several methods was used to perform whole-genome sequencing of sf106. The results showed that sf106 is most closely related to Streptomyces xinghaiensis S187; the average nucleotide identity and average amino acid identity of sf106 and S187 were more than 96%. The genome size of sf106 is approximately 7300 kb, the GC content is greater than 72%, and more than 6700 coding sequences (CDS) were identified. Analysis of mobile genetic elements revealed the presence of a large number of horizontally transferred genes in Streptomyces fradiae sf106, which contribute to microbial diversity. Through antiSMASH prediction, 22 secondary metabolite gene clusters were obtained, which had great potential to generate polyketide metabolites. By examining the data, it was found that the genes contained in cluster 9 were similar to those involved in tylosin synthesis. Non-targeted metabolome sequencing revealed that a total of 1855 identifiable metabolites were produced in the fermentation broth, and the majority of metabolites showed highly significant differences in mean relative abundance between the groups. The identified metabolites were compared against the KEGG compound database to obtain metabolite classifications, mainly including Biological Roles, Phytochemical Compounds, Lipids, and Pesticides. One-way ANOVA indicated that the relative concentration of tylosin differed significantly across all the growth periods, except for the late-logarithmic and stabilization stages. This study provides important basic information on the secondary metabolite research of sf106, which will help us to understand and apply Streptomyces fradiae sf106 more comprehensively. Tylosin is a 16-membered macrolide antibiotic widely used in veterinary medicine to control infections caused by Gram-positive pathogens and mycoplasmas. To improve the fermentation titer of tylosin in the hyperproducing Streptomyces xinghaiensis strain TL01, we sequenced its whole genome and identified the biosynthetic gene cluster therein. Overexpression of the tylosin efflux gene tlrC, the cluster-situated S-adenosyl methionine (SAM) synthetase gene metKcs, the SAM biosynthetic genes adoKcs-metFcs, or the pathway-specific activator gene tylR enhanced tylosin production by 18%, 12%, 11%, and 11% in the respective engineered strains TLPH08-2, TLPH09, TLPH10, and TLPH12. Co-overexpression of metKcs and adoKcs-metFcs as two transcripts increased tylosin production by 22% in the resultant strain TLPH11 compared to that in TL01. Furthermore, combinational overexpression of tlrC, metKcs, adoKcs-metFcs, and tylR as four transcripts increased tylosin production by 23% (10.93g/L) in the resultant strain TLPH17 compared to that in TL01. However, a negligible additive effect was displayed upon combinational overexpression in TLPH17 as suggested by the limited increment of fermentation titer compared to that in TLPH08-2. Transcription analyses indicated that the expression of tlrC and three SAM biosynthetic genes in TLPH17 was considerably lower than that of TLPH08-2 and TLPH11. Based on this observation, the five genes were rearranged into one or two operons to coordinate their overexpression, yielding two engineered strains TLPH23 and TLPH24, and leading to further enhancement of tylosin production over TLPH17. In particular, the production of TLPH23 reached 11.35 g/L. These findings indicated that the combinatorial strategy is a promising approach for enhancing tylosin production in high-yielding industrial strains.

Tylosin is a commonly used antibiotic in animal medicine. However, it remains unclear how tylosin impacts the broader ecosystem once the host animal has excreted it. One of the main concerns is that it can lead to the development of antibiotic resistance. Therefore, there exists a need to develop systems that remove tylosin from the environment. Utilizing UV irradiation to destroy pathogens is one technique often deployed by scientists and engineers. However, for light-based techniques to be efficient, it is necessary to understand the spectral properties of the material being removed. Steady-state spectroscopy and density functional theory were used to analyze the electronic transitions of tylosin responsible for its strong absorbance in the mid-UV region. It was observed that the absorbance peak of tylosin stems from two transitions in the conjugated region of the molecule. Moreover, these transitions stem from an electronegative region of the molecule, which would allow them to be manipulated by changing solvent polarity. Finally, a polariton model has been proposed, which can be used to initiate the photodegradation of tylosin without the need for direct irradiation of the molecule with UV-B light. Microplastic pollution is becoming one of the most severe threats to the entire earth surface ecosystem; moreover, it has the potential to act as a carrier for other chemical pollutants, introducing these pollutants in diverse environments. To evaluate this threat, this study investigated the adsorption of two antibiotics, tylosin (TYL) and tetracycline (TC), onto three common environmental microplastics, polyethylene (PE), polystyrene (PS), and polyvinyl chloride (PVC), through batch adsorption experiments. Overall, the Freundlich model fitted the isothermal adsorption well, which indicated inhomogeneous distribution of adsorption sites involved in the adsorption. The fitted parameters indicated that the adsorption of TYL and TC onto PVC was highest relative to that onto the other two microplastics, which was attributed to the differences in functional groups and crystallinity of polymers. The adsorption efficiency decreased with increasing microplastic concentration. Owing to the salting-out effect, the increase in salinity promoted TYL adsorption onto the microplastics; salinity had no significant effect on TC adsorption onto microplastics. The results suggest that microplastics can be carriers of antibiotics in the environment, for which the physicochemical properties of microplastics and antibiotics as well as environmental factors are crucial influencing factors. The impact of in-feed use of tylosin in feedlot cattle on Gram-negative foodborne bacteria is unknown. We evaluated the effect of continuous in-feed tylosin use on the concentration and prevalence of tetracycline-resistant (TETr)-, third-generation cephalosporin-resistant (3GCr)-, and extended-spectrum β-lactamase–producing (ESBLs) E. coli in feedlot cattle. A cohort of weaned calves (10 animals/group) were randomized to receive a feed ration with or without tylosin. Fecal samples, regularly collected over the entire feeding period, and pen surface and feed samples, collected at the end of the feeding period, were cultured on selective media. Enumeration and binary outcomes were analyzed by mixed effects linear regression or logistic regression, respectively, using treatment and days on feed as fixed factors, and animal ID as a random variable. Tylosin supplementation did not affect the fecal concentrations of TETr E. coli or fecal prevalence of 3GCr E. coli. However, cattle in the tylosin group were 1.5 times more likely (Odds ratio = 1.5: 95% confidence interval: 1.1–2.0) to harbor ESBLs E. coli than the control cattle. Regardless of tylosin treatment, fecal concentrations of TETr E. coli and the prevalence of 3GCr- and ESBLs-E. coli increased over time. Tylosin-supplemented feed did not affect the prevalence of TETr E. coli; 3GCr and ESBLs-E. coli were not detected from the feed samples. Most of the 3GCr- and ESBLs-E. coli isolates carried the blaCTX-M-15 gene, widely detected among ESBLs-E. coli human isolates. In summary, although in-feed tylosin use in feedlot cattle did not select for TETr- and 3GCr-E. coli, it increased the likelihood of detecting ESBL-producing E. coli. Furthermore, the study indicated that the feedlot production setting gradually increases the levels of E. coli resistant to the critically and/or important antibiotics for public health, indicating an increased risk of their dissemination beyond the feedlot environment.

Mycoplasma gallisepticum (MG) is a contagious avian pathogen that causes financial losses to the poultry industry. Isolation of the pathogen is difficult and time-consuming, and therefore, far from a routine method. Serological testing methods to detect antibodies resistant to MG are widely used in routine diagnosis. Tylosin is a class of macrolide antibiotics tremendously administered in veterinary medicine for the treatment of mycoplasmosis and prophylaxis. This study aimed to detect MG by immunoassay testing, culture, and polymerase chain reaction (PCR) in commercial poultry farms and to investigate the tylosin susceptibility of the isolates. To verify the presence of antibodies resistant to MG, 750 blood samples were randomly collected from 38 broiler farms from 2019 to 2022 in Mazandaran and Golestan provinces, Iran, and rapid slide agglutination (RSA) assay was performed. Positive results were analyzed by the enzyme-linked immunosorbent assay (ELISA) for further investigation. Here, 920 swab samples were collected from 38 non-vaccinated commercial farms for culture, and PCR tests were performed for the isolated strains. The activities of tylosin were tested in vitro against these isolates using the broth microdilution method. The lowest antibiotic concentration that resulted in a color change was considered the minimum inhibitory concentration (MIC) value. Twenty-four (63.1%) farms were positive in the RSA test, and 21 (55.2%) farms were positive in the ELISA test. Nine (23.68%) of the farms grew on culture media, and 8 (21.05%) were detected as Gallisepticum species by PCR. The geometric mean of MIC for tylosin was 5.75 µg/ml, MIC50 was 4 µg/ml, and MIC90 was 8 µg/ml. The results indicated that commercial farms were infected with MG. Considering the ability of MG to spread and the probable use of the RSA test as a rapid and cheap method, it can be argued that ELISA and RSA serological tests can be used to find MG in poultry flocks, and the positive result should be confirmed by standard microbiological tests or PCR. It was also found that the isolated parts of MG changed their sensitivity to tylosin, indicating the need for routine testing to optimize treatment dose and efficiency. [ABSTRACT FROM AUTHOR]

Macrolides are currently a class of extensively used antibiotics in human and animal medicine. Tylosin is not only one of the most important veterinary macrolides but also an indispensable material for the bio- and chemo-synthesis of new generations of macrolide antibiotics. Thus, improving its production yield is of great value. As the key rate-limiting enzyme catalyzing the terminal step of tylosin biosynthesis in Streptomyces fradiae (S. fradiae), TylF methyltransferase’s catalytic activity directly affects tylosin yield. In this study, a tylF mutant library of S. fradiae SF-3 was constructed based on error-prone PCR technology. After two steps of screening in 24-well plates and conical flask fermentation and enzyme activity assay, a mutant strain was identified with higher TylF activity and tylosin yield. The mutation of tyrosine to phenylalanine is localized at the 139th amino acid residue on TylF (TylFY139F), and protein structure simulations demonstrated that this mutation changed the protein structure of TylF. Compared with wild-type protein TylF, TylFY139F exhibited higher enzymatic activity and thermostability. More importantly, the Y139 residue in TylF is a previously unidentified position required for TylF activity and tylosin production in S. fradiae, indicating the further potential to engineer the enzyme. These findings provide helpful information for the directed molecular evolution of this important enzyme and the genetic modification of tylosin-producing bacteria.

Carbon-based solid acids have been successfully employed as acidic catalysts for pollutant mitigation in wastewater. To fully tap the potentials of commercially viable carbons for the preparation of solid acids and enhance their catalytic performances is a challenging problem. In this work, three commercialized carbons including biochar, activated carbon and graphite were preprocessed (ball-milling, Hummer exfoliation, HNO3 soaking, and microwave heating in HNO3, etc.), sulfonated, and evaluated as solid-acid catalysts for tylosin mitigation. Graphite-originated solid acid performed the best through a balling-milling preprocess, while biochar-originated solid acids behaved well under all preprocessing treatments, in which 40 mg L−1 of tylosin was mitigated within 8 min by 1 g L−1 of biochar-originated solid acids. The biochar solid acid through the ball-milling preprocess presented high total acidity and large amounts of –SO3H groups, due to dramatically increased surface area and the rise of activation groups (hydroxyl, alkyl and alkoxy groups, etc.) facilitating electrophilic reaction. In addition, decreased particle size and aromaticity and increased structural defects also contributed. Theoretical calculation of average local ionization energy (ALIE) of condensed aromatic model molecules with substituted activation groups confirmed the promoting effects on sulfonation from strong to weak were 8.40–9.06 eV. These findings have deepened the knowledge in tuning carbon surface chemistry for better sulfonation, thus strengthening catalytic degradation of tylosin. The value of this study is in pulling a clear thread for maneuvering solid-acid catalysts using carbons, which holds a novel promise for rationally functionalizing biochar-based catalysts for the remediation of macrolide antibiotics in polluted water. Employing affordable and uncomplicated sample preparation techniques to recommend the most efficient antibacterial therapy could help reduce antibiotic-resistant bacteria. This study evaluated the suitability of immunoassays and microbiological assays as alternatives for liquid chromatography/mass spectrometry (LC/MS) in determining plasma tylosin concentrations after intramuscular administration at a dose of 20 mg/kg to both healthy and diseased pigs in clinical veterinary practice. The diseased pigs were confirmed using the target genes Actinobacillus pleuropneumoniae (apxIVA) and Pasteurella multocida (kmt1). The methods showed good linearity, precision, and accuracy. In both healthy and diseased pigs, a significant correlation was observed between LC/MS and the microbiological assay (Pearson correlation coefficient: 0.930, p < 0.001 vs. Pearson correlation coefficient: 0.950, p < 0.001) and between LC/MS and the enzyme-linked immunosorbent assay (ELISA) (Pearson correlation coefficient: 0.933; p < 0.001 vs. Pearson correlation coefficient: 0.976, p < 0.001). A strong correlation was observed between the microbiological assay and the ELISA in both healthy and diseased pigs (Pearson correlation coefficient: 0.911; p < 0.001 vs. Pearson correlation coefficient: 0.908, p < 0.001). A Bland-Altman analysis revealed good agreement between the methods, i.e., 95% of the differences were within the limits of agreement. • Tylosin was efficiently mitigated by SO 3 H-abundant polymer-based solid acids. • Co-existence of COD and HA did not obviously affect the hydrolytic efficiency. • Polymer-based solid acids presented reliable regeneration and reuse ability. • Pharmacophores in tylosin were hydrolytically removed through desugarization. • The cytotoxicity and ecotoxicity of the transformation products decreased. Antibiotic production wastewater usually contains high concentrations of antibiotic residues, which can cause instability and deterioration of biological wastewater treatment units and also domestication and proliferation of antibiotic-resistance bacteria. An effective pretreatment on antibiotics production wastewater is expected to selectively reduce the concentration of antibiotics and decrease the toxicity, rather than mitigate organic and other contaminants before further treatments. In this work, two polymer-based solid acids, PS-S and CPS-S bearing high concentrations of -SOH 3 groups (up to 4.57 mmol/g), were prepared and successfully used for hydrolytic mitigation of 100 mg/L tylosin within 20 min. The co-existence of high concentrations of COD and humic substances did not affect the mitigation of tylosin obviously, while more than 500 mg/L of nitrogenous compounds suppressed the hydrolytic efficiency. Recycle and reuse experiments showed that the solid acids performed well in five cycles after regeneration. Three transformation products (P1, P2 and P3) were identified using UPLC-QTOF-MS/MS. Sugar moieties including mycarse, mycaminose, and mycinose detached and released simultaneously or in order from the 16-member lactone ring through desugarization, which led to a dramatic decrease in antibacterial activity as revealed by cytotoxicity evaluations using S. aureus. Ecotoxicity estimation indicated the acute toxicities of the hydrolyzed products to model species (e.g., fish, daphnid and green algae) were classified as "not harmful". This work suggested an effective and selective method to pretreat tylosin-contained production wastewater by using polymer-based solid acids. In this study, an immunochromatographic test (using the Charm QUAD2® Test) was used to screen for residual macrolides and lincosamides in raw cow's milk. The validation parameters (selectivity/specificity, detection capability (CCβ), and ruggedness) were in agreement with the requirements of[EC] 2021. The selectivity of the immunochromatographic test was verified by the negative results of microbiological tests. The false-positive rate was 0%. The CCβ values of the immunochromatographic test for various antibiotics in milk were as follows: erythromycin 0.02 mg/kg, spiramycin 0.1 mg/kg, tilmicosin 0.025 mg/kg, tylosin 0.05 mg/kg, lincomycin 0.15 mg/kg, and pirlimycin 0.15 mg/kg. The determined CCβ values were lower than the respective maximum residue limits (MRLs; regulatory limits in Japan) for milk, except for lincomycin (equal to the MRL). The presence of antibiotic groups other than macrolides and lincosamides did not interfere with the specificity of the test. It showed no significant difference in lot-to-lot repeatability. The results obtained by the two researchers showed no significant differences. Finally, the test was applied to milk samples obtained from a tylosin-treated cow. The outcome was positive and in agreement with the results of the chemical analytical and microbiological methods. Cytochromes P450 (P450s) are nature's catalysts of choice for performing demanding and physiologically vital oxidation reactions. Biochemical characterization of these enzymes over the past decades has provided detailed mechanistic insight and highlighted the diversity of substrates P450s accommodate and the spectrum of oxidative transformations they catalyze. Previously, we discovered that the bacterial P450 MycCI from the mycinamicin biosynthetic pathway in Micromonospora griseorubida possesses an unusually broad substrate scope, whereas the homologous P450 from tylosin-producing Streptomyces fradiae (TylHI) exhibits a high degree of specificity for its native substrate. Here, using biochemical, structural, and computational approaches, we aimed to understand the molecular basis for the disparate reactivity profiles of these two P450s. Turnover and equilibrium binding experiments with substrate analogs revealed that TylHI strictly prefers 16-membered ring macrolides bearing the deoxyamino sugar mycaminose. To help rationalize these results, we solved the X-ray crystal structure of TylHI in complex with its native substrate at 1.99-Å resolution and assayed several site-directed mutants. We also conducted molecular dynamics simulations of TylHI and MycCI and biochemically characterized a third P450 homolog from the chalcomycin biosynthetic pathway in Streptomyces bikiniensis These studies provided a basis for constructing P450 chimeras to gain further insight into the features dictating the differences in reaction profile among these structurally and functionally related enzymes, ultimately unveiling the central roles of key loop regions in influencing substrate binding and turnover. Our work highlights the complex nature of P450/substrate interactions and raises interesting questions regarding the evolution of functional diversity among biosynthetic enzymes.

Abstract Background Antibiotic‐responsive enteropathy (ARE) is diagnosed by excluding other causes of diarrhea and when there is a short‐term response to administration of antibiotics. Objectives To characterize the gut microbiota and clinical trend of dogs with suspected ARE and to evaluate the variation in microbiota before (T0), after 30 days (T30) of tylosin treatment, and 30 days after discontinuation of treatment (T60). A further objective was to evaluate whether changes in gut microbiota are related to relapses of diarrhea when the therapy is tapered. Animals Study sample (group A) was composed of 15 dogs with chronic diarrhea, group B was composed of 15 healthy dogs. Group A was given tylosin for 30 days. Methods A multicentric prospective study. Clinical Indexes, fecal score, and samples for microbiota analysis were collected at T0, T30, and T60 in group A and T0 and T30 in group B. The gut microbiota was analyzed via 16S ribosomal RNA gene. Qiime2 version 2020.2 was used to perform bioinformatic analyses, and Alpha‐ and Beta‐diversity were computed. Results Diarrhea recurred after T30 in 9 of 14 dogs, which were classified as affected by ARE. At T0, a difference was noted in the beta‐diversity between groups (Bray Curtis metric P = .006). A T0‐T30 difference in alpha‐diversity was noted in group A (Shannon index P = .001, Faith PD P = .007). Conclusions and Clinical Importance Although tylosin influences the microbiota of dogs with ARE, we failed to find any specific characteristic in the microbiota of dogs with ARE.

Hydrothermal pretreatment (HPT) followed by anaerobic digestion (AD) is an alternative for harvesting energy and removing organic contaminants from sewage sludge and animal manure. This study investigated the use, in an energetically sustainable way, of HPT and AD, alone or combined, to produce methane and remove tylosin and antimicrobial resistance genes (ARG) from poultry litter (PL). The results showed that HPT at 80 °C (HPT80), followed by single-stage AD (AD-1S), led to the production of 517.9 ± 4.7 NL CH4 kg VS−1, resulting in 0.11 kWh kg PL−1 of electrical energy and 0.75 MJ kg PL−1 of thermal energy, thus supplying 33.6% of the energy spent on burning firewood at a typical farm. In this best-case scenario, the use of HPT alone reduced tylosin concentration from PL by 23.6%, while the process involving HPT followed by AD-1S led to the removal of 91.6% of such antibiotic. The combined process (HPT80 + AD-1S), in addition to contributing to reduce the absolute and relative abundances of ARG ermB (2.13 logs), intI1 (0.39 logs), sul1 (0.63 logs), and tetA (0.74 logs), led to a significant removal in the relative abundance of tylosin-resistant bacteria present in the poultry litter. Antibiotic residues in meat products may have public health implications. The aim of this study was to determine the tylosin residue level in meat products(sausages, persian sausages, and hamburgers) in Tabriz. For this study 60 samples( 20 samples from each meat product) were collected from food supply stores from March to May 2021, randomly. The tylosin residue levels were detected by the HPLC method. Of all samples, 90 percent of sausages, and 100 percent of hamburgers had tylosin residue. The mean of tylosin residue in sausages, persian sausages(Kalbas), and hamburgers were 19.34±2.62, 22.2±2.62, and 39.2±2.62 µg/Kg, respectively. The mean of tylosin residue in hamburgers was higher than in other products significantly. In addition, the residual amount of tylosin in all samples was lower than the allowed limit of CodexAlimentarius(100 µg/Kg). Furthermore, the tylosin residue level in hamburgers with 90 percent meat was higher than hamburgers with 70 percent meat significantly. Even though the amount of tylosin residues is low in meat products in Tabriz, continuous control of antibiotic residues in meat products is recommended. [ABSTRACT FROM AUTHOR]
