1. CHARACTERIZATION OF CCL2 AND CXCL10 GENE IN THARPARKAR AND VRINDAVANI CATTLE.

Discovery of SNPs and their association with immune response candidate genes is key process for identifying genetic markers. This research was done to find new SNPs in the ccl2 and cxcl10 genes of cattle from Tharparkar and Vrindavani. Total 12 samples (6 from Tharparkar and 6 from Vrindavani calves) of 5 ml blood were collected and using primers, desired genomic segment of genes was amplified. After gel purification, amplified products were sent to Sanger sequencing. Sanger sequenced data were analyzed for SNPs. The average size of consensus sequences of ccl2 products was 1155 bp and of cxcl10 was 945 bp. Two SNPs (1462 A>G and 1744 T>G) were found in ccl2 apmlicons with two genotypes (AA, AG) and (TT, TG) respectively. Both were in intronic region causing no effect on amino acid sequence. One SNP (394 G>A) was found in cxcl10 amplicons with genotypes (GG, AG) and it was present in 3' UTR. None of the locus was found in Hardy Weinberg equilibrium. This is a brief exploratory investigation involving fewer animals. The study's sample size has to be expanded further in order to make an appropriate assessment in this regard. [ABSTRACT FROM AUTHOR]
