Stenotrophomonas maltophilia is a gram-negative bacterium that can be routinely identified in the aquatic environment, and has become an emergent, multi-drug-resistant life-threatening organism in some circumstances. This bacterium was identified as a presumptive causative agent in cases of polysystemic granulomatous disease in one diamondback terrapin (Malaclemys terrapin) and one mata mata (Chelus fimbriata). Both chelonians presented with lethargy, hyporexia, and multifocal subcutaneous masses diagnosed by physical examination and in one individual on computed tomography (CT). The primary hematologic finding was moderate to severe heterophilic leukocytosis, and both turtles were treated with a combination of supportive care, systemic antibiotics, and/or surgical resection of subcutaneous masses. Medical and surgical management were unsuccessful, and one patient was euthanized and the other found deceased. Necropsies revealed multisystemic granulomas, and cultures identified growth of Stenotrophomonas maltophilia. A postmortem investigation into possible risk factors identified comorbidities including shell disease, repeated administration of injectable antibiotics, low environmental temperature, and aquatic pH changes as possible contributing factors. Organic pollutants have bioaccumulative effects and induce a high index of toxicity in aquatic ecosystems. This study reports on tomographic abnormalities found in free-living turtles (Geoffroy's toadhead turtle, Phrynops geoffroanus) from an urban area near the Capibaribe and Beberibe River basins in Brazil, which are known to be contaminated by heavy metals and solid waste. Clinical and tomographic evaluation of eight turtles was performed to assess their health status for subsequent release. Tomographic analysis revealed bone abnormalities, including demineralization, coarse trabecular pattern, subperiosteal bone resorption, deformity of dorsal vertebrae, and shell fracture. Considering that turtles are recognized as bioindicators of water quality, a high occurrence of bone abnormalities may suggest environmental pollution in the studied area and signal that conservation initiatives are needed to safeguard the aquatic ecosystem of the metropolitan region of Recife. We live in a time of accelerated biological extinctions that has the potential to mirror past mass extinction events. However, the rarity of mass extinctions and the restructuring of diversity they cause complicate direct comparisons between the current extinction crisis and earlier events. Among animals, turtles (Testudinata) are one of few groups that have both a rich fossil record and sufficiently stable ecological and functional roles to enable meaningful comparisons between the end-Cretaceous mass extinction (∼66 Ma) and the ongoing wave of extinctions. Here we analyze the fossil record of the entire turtle clade and identify two peaks in extinction rates over their evolutionary history. The first coincides with the Cretaceous-Paleogene transition, reflecting patterns previously reported for other taxa. The second major extinction event started in the Pliocene and continues until now. This peak is detectable only for terrestrial turtles and started much earlier in Africa and Eurasia than elsewhere. On the basis of the timing, geography, and functional group of this extinction event, we postulate a link to co-occurring hominins rather than climate change as the cause. These results lend further support to the view that negative biodiversity impacts were already incurred by our ancestors and related lineages and demonstrate the severity of this continued impact through human activities. Organisms whose early life stages are environmentally sensitive produce offspring within a relatively narrow range of suitable abiotic conditions. In reptiles, development rate and survival are often maximized if incubation temperatures remain under 31°C, though this upper bound may vary within and among species. We addressed this expectation by comparing responses to egg incubation at 30°C versus 33°C in congeneric turtle species pairs with broad syntopic geographic distributions. In the two softshell turtles (Apalone spp.), the greatest changes in development rate and phenotypic variance were observed in the northernmost population, which had a low survival rate (40%) at 33°C. The presumably suboptimal temperature (33°C) for northern populations otherwise yielded 76%–93% survival rates and fast swimming speeds in more southern populations. Still, in one species, northern hatchlings incubated at 33°C matched the elevated speeds of their southern counterparts, revealing a countergradient response. In northern populations of the two map turtles (Graptemys spp.), survival was also reduced (28%–60%) at 33°C and the development rate (relative to 30°C) increased by up to 75%. Our experiments on divergent taxa with similar nesting ecologies substantiate that the optimal thermal range for offspring production is variable. These findings encourage further work on how population‐ and species‐level differences relate to local adaptation in widely distributed oviparous species. Aims Microbiome composition is increasingly considered in species reintroduction efforts and may influence survival and reproductive success. Many turtle species are threatened by anthropogenic pressures and are frequently raised in captivity for reintroduction efforts, yet little is known about turtle microbiome composition in either wild or captive settings. Here, we investigated trends in microbiome composition of captive and wild IUCN-endangered Blanding's turtles (Emydoidea blandingii). Methods and results We amplified and sequenced the V4 region of the 16S rDNA locus from plastron, cloaca, and water samples of wild E. blandingii adults and two populations of captive E. blandingii juveniles being raised for headstarting. Plastron, cloaca, and water-associated microbiomes differed strongly from each other and were highly variable among captive sites and between captive and wild sites. Across plastron, cloaca, and water-associated microbial communities, microbial diversity changed over time, but not in a predictable direction between captive sites. Plastron beta diversity correlated with growth rate in captive samples, indicating that external microbiomes may correlate with individual fitness. Conclusions Our results indicate that external and internal microbiomes vary between captive and wild turtles and may reflect differences in fitness of captive-raised individuals. Testudines are a highly threatened group facing an array of stressors, including alteration of their sensory environment. Underwater noise pollution has the potential to induce hearing loss and disrupt detection of biologically important acoustic cues and signals. To examine the conditions that induce temporary threshold shifts (TTS) in hearing in the freshwater Eastern painted turtle (Chrysemys picta picta), three individuals were exposed to band limited continuous white noise (50–1000 Hz) of varying durations and amplitudes (sound exposure levels ranged from 151 to 171 dB re 1 μPa2 s). Control and post-exposure auditory thresholds were measured and compared at 400 and 600 Hz using auditory evoked potential methods. TTS occurred in all individuals at both test frequencies, with shifts of 6.1–41.4 dB. While the numbers of TTS occurrences were equal between frequencies, greater shifts were observed at 600 Hz, a frequency of higher auditory sensitivity, compared to 400 Hz. The onset of TTS occurred at 154 dB re 1 μPa2 s for 600 Hz, compared to 158 dB re 1 μPa2 s at 400 Hz. The 400-Hz onset and patterns of TTS growth and recovery were similar to those observed in previously studied Trachemys scripta elegans, suggesting TTS may be comparable across Emydidae species. OBJECTIVE To evaluate and compare the pharmacokinetic parameters of SC ceftazidime administered at 20 and 40 mg/kg to red-eared sliders. ANIMALS 8 adult red-eared sliders (Trachemys scripta elegans). METHODS In a sequential, 2-period study with a 3-week washout period between treatments, ceftazidime was administered SC to turtles at 20 and 40 mg/kg. Blood samples were collected from the subcarapacial sinus at 0, 24, 48, 72, 96, and 120 hours after ceftazidime administration. Plasma ceftazidime concentrations were quantified using reversedphase HPLC. RESULTS Mean plasma half-life after 20- and 40-mg/kg dosing was 39.75 ± 8.0 hours and 33.03 ± 6.56 hours, respectively. Mean maximum plasma concentration after 20- and 40-mg/kg dosing was 71.0 ± 15.93 µg/mL and 120.0 ± 30.62 µg/mL, respectively. Mean plasma ceftazidime concentrations remained ≥ 8 µg/mL, the theoretical MIC for various reptile pathogens for all time points. CLINICAL RELEVANCE Results indicate that ceftazidime dosed at either 20 or 40 mg/kg produces plasma concentrations exceeding the theoretical MIC of various reptile pathogens for at least 120 hours. An ideal dosing interval could not be determined, as all plasma concentrations remained above the threshold of interest for all time points. The current study focuses on exploring the effectiveness of Turtle Shell Powder (TSP) as a new and unique approach to use it as a filler material while fabricating kenaf/epoxy composites and investigating its potential application. The FRP composite of kenaf/epoxy reinforced with TSP filler was manufactured at various loading (1%, 3% and 5% by weight). The influence of TSP filled KFRP composite was studied and compared with the results of mechanical properties. SEM was employed to conduct morphological traits of the tensile fracture for the K0 and K5 samples with this EDAX also analysed. A notable enhancement observed from the result is the 5% TSP reinforced composite (K5) compared to other samples. This enhancement was ascribed to the improved dispersion and interfacial bonding between the reinforcement and the matrix in the composites, leading to more efficient load transfer mechanisms. The performance of TSP particles dramatically improves the strength of the laminated composites, and the inclusion of ply orientation further strengthens the interfacial bonding between TSP, epoxy and kenaf components. A new Chengyuchelys latimarginalis (Xinjiangchelyidae) specimen is reported based on a well-preserved, nearly complete shell from the upper part of the Upper Jurassic Shangshaximiao Formation of Qijiang District, Chongqing Municipality, China. The Xinjiangchelyidae were widely distributed in Asia during the Jurassic and were the dominant turtle group in Asian freshwater ecosystems during the Late Jurassic. The Qijiang specimen adds a new element to the yet poorly known freshwater vertebrate fauna and extends the geographical distribution of C. latimarginalis. This paper discusses the intraspecific variation in C. latimarginalis for the first time. It provides a new insight for studying the evolution, habitat, and diversity of xinjiangchelyids. The lower Rio Grande and Pecos River of the southwest United States have been heavily modified by human activities, profoundly impacting the integrity of their aquatic wildlife. In this context, we focused our study on the population genomics of the Rio Grande Cooter (Pseudemys gorzugi), a freshwater turtle of increasing conservation concern, residing in these two rivers and their tributaries. The genetic data revealed two distinct populations: one in the Pecos and Black Rivers of New Mexico and another in the Rio Grande and Devils River of Texas, with admixed individuals identified at the confluence of the Rio Grande and Pecos River. In addition to having a smaller geographic range, we found lower observed heterozygosity, reduced nucleotide diversity, and a smaller effective population size (N e) in New Mexico population. Our results depict a significant isolation-by-distance pattern across their distribution, with migration being notably infrequent at river confluences. The Critically Endangered Cantor's giant softshell turtle Pelochelys cantorii is a freshwater turtle found in South and Southeast Asia. Its population is declining because of habitat destruction and alteration, and hunting for its meat. Data on the species' ecology, behaviour, population size, distribution and threats are limited, and previous surveys undertaken in India have failed to detect individuals in the wild. This lack of data hinders strategic conservation planning. Ecologists and conservationists increasingly utilize local ecological knowledge to determine the status and gain insights into the ecology of threatened and rare species that are difficult to detect in field studies. To examine the historical and current occurrence of Cantor's giant softshell turtle, we conducted community interviews along the Chandragiri River in Kerala, India. With data from these interviews, we identified multiple sites where the species continues to occur. Older respondents and those that used the river for fishing and irrigation were more likely to report sightings of the species. Our findings also improved knowledge about the turtles' seasonal and diel activity patterns. A network of key informants identified through the interviews provided information on turtle bycatch, sightings and nesting. This network is being utilized for the continued monitoring of the species and could help in devising evidence-based management strategies for softshell turtles in India. Turtle nesting habitat can be created as a restoration strategy to increase habitat availability or provide suitable habitat away from threats. Traditional nest habitat restoration consists of creating nesting mounds using a mix of sand and gravel. However, nesting mounds do not resemble natural turtle nesting habitat in a rock barren landscape where turtles nest in crevices and cracks in the bedrock. Therefore, our objective was to design and evaluate the success of a landscape‐appropriate design for turtle nesting habitat in a rock barren landscape. To evaluate success of the nest habitat design, we assessed the (1) survival of transplanted moss and lichen cover on created nest sites, (2) ecohydrological and physical conditions at created and natural sites, and (3) turtle egg hatching success at created and natural sites using a split‐clutch experiment. We found no difference in productivity between lichen transplants and natural sites, indicating that intact lichen transplants were successful. Moss transplant success was more variable due to moisture stress because transplants were conducted during dry conditions. In general, created nest habitat tended to have a more stable thermal and moisture regime compared to natural sites. When accounting for maternal effects, the odds of an egg hatching successfully was 6.6 times higher in a created site than a natural site. Overall, the success of our nest habitat design in the first few years suggests that this landscape‐appropriate design will be a useful restoration strategy for increasing turtle nesting habitat in rock barren landscapes. Reliable population estimates are important for making informed management decisions about wildlife species. Standardized survey protocols have been developed for monitoring population trends of the wood turtle (Glyptemys insculpta), a semi-aquatic freshwater turtle species of conservation concern throughout its distribution in east-central North America. The protocols use repeated active search surveys of defined areas, allowing for estimation of survey-specific detection probability (p) and site-specific abundance. These protocols assume population closure within the survey area during the survey period, which is unlikely to be met as wood turtles are a highly mobile species. Additionally, current protocols use a single-pass design that does not allow for separation of availability (pa) and detectability (pd). If there are systematic influences on pa or pd that are not accounted for in the survey design or data analysis, then resulting abundance estimates could be biased. The objectives of this study were to determine if pa is a random process and if pa and pd are influenced by demographic characteristics. We modified the wood turtle survey protocol used in the upper Midwest to include a double-pass design, allowing us to estimate pa and pd using a robust design capture-recapture model. The modified protocol was implemented at 14 wood turtle monitoring sites in Minnesota and Wisconsin between 2017 and 2022. Our results indicated that pa was non-random and that pd increased with turtle carapace length. Our study suggests that model assumptions for current wood turtle population models may be violated, likely resulting in an overestimation of abundance. Anoxia in the mammalian brain leads to hyper-excitability and cell death; however, this cascade of events does not occur in the anoxia-tolerant brain of the western painted turtle, Chrysemys picta belli. The painted turtle has become an important anoxia-tolerant model to study brain, heart, and liver function in the absence of oxygen, but being anoxia-tolerant likely means that decapitation alone is not a suitable method of euthanasia. Many anesthetics have long-term effects on ion channels and are not appropriate for same day experimentation. Using whole-cell electrophysiological techniques, we examine the effects of the anesthetic, Alfaxalone, on pyramidal cell action potential amplitude, threshold, rise and decay time, width, frequency, whole cell conductance, and evoked GABAA receptors currents to determine if any of these characteristics are altered with the use of Alfaxalone for animal sedation. We find that Alfaxalone has no long-term impact on action potential parameters or whole-cell conductance. When acutely applied to naïve tissue, Alfaxalone did lengthen GABAA receptor current decay rates by 1.5-fold. Following whole-animal sedation with Alfaxalone, evoked whole cell GABAA receptor current decay rates displayed an increasing trend with 1 and 2 hours after brain sheet preparation, but showed no significant change after a 3-hour washout period. Therefore, we conclude that Alfaxalone is a suitable anesthetic for same day use in electrophysiological studies in western painted turtle brain tissue. [ABSTRACT FROM AUTHOR]

Verifying habitats, including the foraging and nesting areas for sea turtles, enables an understanding of their spatial ecology and successful planning of their conservation and management strategies. Recently, the observation frequency and bycatch of loggerhead (Caretta caretta) and green (Chelonia mydas) turtles have increased in the northern limit of their distribution range, in the northern part of the East China Sea and East (Japan) Sea. We conducted satellite tracking to investigate the habitat use of seven loggerhead and eight green turtles from June 2016 to August 2022 in this area, where little is known about their spatial ecology. We applied a 50 percent volume contour method to determine their main foraging areas and analyzed 6 environmental variables to characterize their habitats. Loggerhead turtles mainly stayed in and used the East China Sea as a foraging area during the tracking period, while two individuals among them also used the East Sea as a seasonal foraging area. Most green turtles also used the East China Sea as a foraging area, near South Korea and Japan, with one individual among them using the lower area of the East Sea as a seasonal foraging area. Notably, one green turtle traveled to Hainan Island in the South China Sea, a historical nesting area. Our results showed that the two sea turtle species included the East Sea as a seasonal foraging area, possibly owing to the abundance of food sources available, despite its relatively lower sea temperature. Considering that loggerhead and green sea turtles were observed using the northern part of the East China Sea and East Sea more frequently than previously known and that the sea temperature gradually increases due to climate change, conservation and management activities are required for sea turtles in these areas. A possible turtle swim track from the Third Member of the Late Triassic Xujiahe Formation (Norian-Rhaetian) represents the oldest ichnological evidence of the group from China, supplementing previous reports of more abundant tracks from the Middle Jurassic of the region. The specimen is younger than the known pantestudinate ('stem-turtle') body fossils from China but matches the stratigraphic distribution of testudinate skeletons in other regions, for example, in Thailand, Europe and North America. Caution should be used to distinguish turtle swim tracks from those of crocodylomorphs and theropod dinosaurs, especially where only small samples are available. Therefore, we do not completely exclude an affinity with these groups or other archosaurs, such as phytosaurs. Nevertheless, the morphology of the imprints is most similar to turtle tracks, and it is most likely that an early non-crown group representative was the responsible producer. Balancing urban development with environmental sustainability is a major challenge that is increasingly recognized in planning decisions. Urban development proposals are often approved with the expectation that deleterious impacts on native species will be constrained, but this assumption is rarely tested over sufficient timelines to confirm its validity for long‐lived, at‐risk species. We tracked changes in Blanding's turtle (Emydoidea blandingii) habitat availability and demography over 10 years near Ottawa, ON, Canada, to determine whether urban development and associated mitigation measures were sufficient to ensure long‐term population persistence. Suitable turtle habitat declined by 10% during the study, and wetland corridors were essentially lost. Habitat loss coincided with a marked reduction in adult turtle apparent survival, resulting in a 70% decline in population size. Adult females experienced the greatest decline, and despite wildlife fencing and culvert placement as conditions of project approval, turtle road mortality likely was the primary cause of the decline. Deterministic population viability analysis revealed that ~4 adult female road mortalities (of an initial 56 females) per year produced a comparable decline to that observed in our population estimates; at this rate, the population will likely breach its quasi‐extinction threshold (4 females) in under a decade. Accordingly, we infer that in our study area, approved urban development was not compatible with at‐risk turtle population viability. Our findings imply that urban development approval conditions, even when conducted in the context of seemingly robust species‐at‐risk protection, can be inadequate to ensure sustainability. We contend that if environmental sustainability is to be prioritized, urban development projects in areas occupied by at‐risk species must be subject to more stringent oversight during the planning, approval and implementation phases. The Convention on International Trade of Endangered Species (CITES) is the primary mechanism for controlling international wildlife trade and preventing overexploitation, but measuring its efficacy is difficult, which complicates management efforts. We examined the effects of CITES listings on the trade of freshwater turtles from the US and compared spatiotemporal trends and dynamics of trade across 16 genera using 20 years of export data, triangulated with mixed qualitative data. For each CITES‐listed genus, we analyzed the average changes, immediate changes, and changes in trends of export volumes and spatial diversity of trade. Boom‐and‐bust trends of exports for different taxa were driven by several factors, including CITES listings, but CITES had mixed effects on export volumes for both Appendix II and III species while the diversity of spatial trade networks consistently declined for CITES and non‐CITES listed genera. These results point to differences in demand and species' availability from captive breeders, farms, or wild harvest which are important to consider in future listing decisions. However, a lack of reliable data on the source of turtles limited our ability to understand these drivers as well as the efficacy of CITES listings in preventing overexploitation. Vital rates for small, non-breeding individuals are important components of population dynamics for many species, but often individuals of these sizes are difficult to locate, capture, and track. As such, biologists frequently lack reliable estimates of juvenile survival because sample sizes and recapture rates for this life stage are low. Long-lived animals often take many years to reach sexual maturity and spend much of this time in the smaller size classes, making them sensitive to changes in survival rates. We estimated the survival rates of all size classes for the northern map turtle (Graptemys geographica) using a mark-recapture dataset with >3,500 captures from 2019-2021 and 210 nests from 2018-2021. As turtle size increased, annual survival probability increased regardless of sex. Estimated annual survival probability for turtles >18 cm long (i.e., adult females >15 years) was about 0.95, over 4 times higher than turtles that were 3 cm long (i.e., hatchlings <1 year; 0.22 annual survival probability). Although we did not observe a difference in survival probability between sexes of any size class, adult females are nearly twice the size of adult males, leading to an increased annual survival probability for females of 0.95, compared to 0.80 for males. Changes in adult survival had the greatest influence on population estimates over time, with temporary decreases, such as those due to poaching or an environmental disaster, potentially leading to unrecoverable decreases in the overall population size. Our study provides detailed survival rates for all size classes in a long-lived turtle, which are necessary to assess population stability and can be used to determine the most effective conservation or management practices. Bellinger River virus (BRV) is a serpentovirus (nidovirus) that was likely responsible for the catastrophic mortality of the Australian freshwater turtle Myuchelys georgesi in February 2015. From November 2015 to November 2020, swabs were collected from turtles during repeated river surveys to estimate the prevalence of BRV RNA, identify risk factors associated with BRV infection, and refine sample collection. BRV RNA prevalence at first capture was significantly higher in M. georgesi (10.8%) than in a coexisting turtle, Emydura macquarii (1.0%). For M. georgesi, various risk factors were identified depending on the analysis method, but a positive BRV result was consistently associated with a larger body size. All turtles were asymptomatic when sampled and conjunctival swabs were inferred to be optimal for ongoing monitoring. Although the absence of disease and recent BRV detections suggests a reduced ongoing threat, the potential for the virus to persist in an endemic focus or resurge in cyclical epidemics cannot be excluded. Therefore, BRV is an ongoing potential threat to the conservation of M. georgesi, and strict adherence to biosecurity principles is essential to minimise the risk of reintroduction or spread of BRV or other pathogens. Background: The developmental biology of Kinosternon scorpioides is described, based on the phenotype. This species is important for the flora because they are excellent seed disseminators. In addition, basic embryological information is not yet fully clarified, and this research provides unprecedented information on the chelonian embryology of the Amazonian fauna. Aim: The present study aims to identify the embryology of K. scorpioides in captivity during different periods. Methods: Females were monitored throughout the reproductive cycle, by video monitoring, to identify nests and the presence of newly laid eggs. At regular weekly intervals, embryo samples were collected fixed in a 4% paraformol solution and preserved in 70% alcohol. For the embryonic characterization, we used a stereomicroscope and the scanning electron microscopy method. Results: We describe 15 embryonic stages for a 15-week (105-day) incubation process. Only at 42 days (6th week) was the morphological characterization of a chelonian observed and at the 12th week (Stage XII), the phenotypic characterization of the species K. scorpioides. Conclusion: In view of the evidence, we found that these phases are similar to the other turtles, with structural variations in the appearance and disappearance of structures due to the specific characteristics of the species. Aeromonas jandaei is a gram‐negative bacterium commonly found in aquatic environments and can induce illnesses in amphibians, reptiles and aquatic animals. In this study, a strain of bacteria was isolated from the diseased Chinese soft‐shell turtle (Pelodiscus sinensis), then named strain JDP‐FX. This isolate was identified as A. jandaei after analysis of morphological, physiological and biochemical characteristics, as well as 16S rRNA and gyrB gene sequences. Virulence genetic testing further detected temperature‐sensitive protease (eprCAI), type III secretion system (TTSS) (ascv), nuclease (nuc), cytotonic enterotoxin (alt) and serine proteinase (ser) in JDP‐FX. Compared with healthy Chinese soft‐shell turtle, the serum levels of total protein (TP), albumin (ALB) and globulin (GLB) were significantly decreased in the diseased Chinese soft‐shell turtle, while, the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased. Histopathological observations showed that multiple tissues, including intestinal mucosa, liver and kidney, were severely damaged in the diseased Chinese soft‐shell turtle. Moreover, the diseased Chinese soft‐shell turtle had significant cell degeneration, necrosis, sloughing and interstitial inflammatory cell infiltration. The pathogenicity of JDP‐FX was tested via artificial infection. The median lethal dosage (LD50) of the strain was 1.05 × 105 colony forming units (CFU/g) per weight of Chinese soft‐shell turtle. Drug susceptibility analysis revealed that JDP‐FX was susceptible to ceftazidime, minocycline, cefoperazone, ceftriaxone and piperacillin. In addition, JDP‐FX was resistant to doxycycline, florfenicol, sulfonamides, gentamicin, ampicillin and neomycin. Urbanization is a significant driver of the global biodiversity crisis. Turtles are particularly impacted by urbanization because of the vulnerability of riparian habitats to habitat loss and road mortality. Behaviour plays a crucial role in determining the success of urban animals. Behavioural responses to urbanization, however, are rarely studied in turtles even though many turtles are at‐risk and sometimes live in urban areas. Therefore, we evaluated behavioural changes in painted turtles (Chrysemys picta) living in wetlands surrounded by a gradient of urbanization. We tested the consistency of painted turtle behaviour in the laboratory and examined the behaviour of painted turtles from 24 wetland sites across an urbanization gradient in Ottawa, Ontario, Canada. We assessed: (i) aggression by measuring the number of active defensive behaviours the turtles performed in response to handling, (ii) boldness by measuring the amount of time the turtles took to emerge from their shells and move from their initial locations in a circular arena and (iii) activity by measuring the amount of time the turtles spent moving in the same circular arena. We found that all behaviours were consistent in the laboratory. We also found that as the level of urbanization increased, turtles were more aggressive and bolder. Urbanization affects painted turtle behaviour, but further research is required to understand the mechanisms responsible and the conservation implications. [ABSTRACT FROM AUTHOR]

Impressions of vertebrate bodies or their parts, such as trace fossils and natural molds of bones, are a valuable source of information about ancient faunas which may supplement the standard fossil record based on skeletal elements. Whereas trace fossils of animal activity are relatively common and actively studied within the field of ichnology, and natural impressions of internal or external surfaces are a frequent preservation mode in fossil invertebrates, natural molds of bones are comparatively rare and less extensively documented and discussed. Among them, internal molds (steinkerns) of turtle shells are a relatively well-known form of preservation, but the mechanisms and taphonomic prerequisites leading to their formation are poorly studied. External shell molds are even less represented in the literature. Herein, we describe a historic specimen of a natural external turtle plastron mold from the Triassic (Norian) Löwenstein Formation of Germany–a formation which also yielded a number of turtle steinkerns. The specimen is significant not only because it represents an unusual form of preservation, but also due to its remarkably large size and the presence of a potential shell pathology. Although it was initially interpreted as Proterochersis sp., the recent progress in the knowledge of proterochersid turtles leading to an increase in the number of known taxa within that group allows us to verify that assessment. We confirm that the specimen is morphologically consistent with the genus and tentatively identify it as Proterochersis robusta, the only representative of that genus from the Löwenstein Formation. We note, however, that its size exceeds the size observed thus far in Proterochersis robusta and fits within the range of Proterochersis porebensis from the Grabowa Formation of Poland. The marks interpreted as shell pathology are morphologically consistent with Karethraichnus lakkos–an ichnotaxon interpreted as a trace of ectoparasites, such as leeches. This may support the previously proposed interpretation of Proterochersis spp. as a semiaquatic turtle. Moreover, if the identification is correct, the specimen may represent a very rare case of a negative preservation of a named ichnotaxon. Finally, we discuss the taphonomy of the Löwenstein Formation turtles in comparison with other Triassic turtle-yielding formations which show no potential for the preservation of internal or external shell molds and propose a taphonomic model for the formation of such fossils. Freshwater fauna is facing an uphill task for survival in the Ganga Basin, India, due to a range of factors causing habitat degradation and fragmentation, necessitating conservation interventions. As part of the ongoing efforts to conserve the freshwater fauna of the Basin, we are working on rehabilitating rescued freshwater chelonians. We carry out various interventions to restore rescued individuals to an apparent state of fitness for their release in suitable natural habitats. Morphometric measurements are crucial to managing captive wild animals for assessing their growth and well-being. Measurements are made using manual methods like vernier caliper that are prone to observer error experience and require handling the specimens for extended periods. Digital imaging technology is rapidly progressing at a fast pace and with the advancement of technology. We acquired images of turtles using smartphones along with manual morphometric measurements using vernier calipers of the straight carapace length and straight carapace width. The images were subsequently processed using ImageJ, a freeware and compared with manual morphometric measurements. A significant decrease in the time spent in carrying out morphometric measurements was observed in our study. The difference in error in measurements was, however, not significant. A probable cause for this may have been the extensive experience of the personnel carrying out the measurements using vernier caliper. Digital image processing technology can cause a significant reduction in the stress of the animals exposed to handling during measurements, thereby improving their welfare. Additionally, this can be used in the field to carry out morphometric measurements of free-ranging individuals, where it is often difficult to capture individuals, and challenges are faced in obtaining permission to capture specimens. Simple Summary: Observing juvenile sea turtles at sea is challenging due to their small sizes and cryptic behaviors and is compounded by the vastness of the ocean. The first phases of sea turtle life history, commonly known as the "Lost Years", remain enigmatic and poorly understood, despite significant advances in animal ad ocean observation technologies. Our study focused on testing new prototypes of microsatellite tags and analyzing their performance on 160 juvenile sea turtles of four species in the North Atlantic. The results demonstrated that, despite challenges brought by miniaturization, the tags proved effective. However, the tracking periods were shorter than expected, limiting our ability to deeply study and understand the turtles' dispersal features. We found that tracking durations varied among species, indicating some limitations due to certain behaviors such as neritic interactions or diving activity by young sea turtles, and revealing that some of their behaviors may be too strenuous for small tags. Our findings have important implications for the bio-logging community, especially those studying marine animals such as sea turtles. Our study showcases recent technological advances and contributes to improving the effectiveness and durability of miniaturized satellite tags deployed within the marine environment. Our methodologies and findings have improved our understanding of the "Lost Years", promise to inform ongoing future technological advances and contribute to increasing the effectiveness of conservation efforts. After hatching, sea turtles leave the nest and disperse into the ocean. Many years later, they return to their natal coastlines. The period between their leaving and their returning to natal areas, known as the "Lost Years", is poorly understood. Satellite tracking studies aimed at studying the "Lost Years" are challenging due to the small size and prolonged dispersal phases of young individuals. Here, we summarize preliminary findings about the performance of prototype microsatellite tags deployed over a three-year period on 160 neonate to small juvenile sea turtles from four species released in the North Atlantic Ocean. We provide an overview of results analyzing tag performance with metrics to investigate transmission characteristics and causes of tag failure. Our results reveal that, despite certain unfavorable transmission features, overall tag performance was satisfactory. However, most track durations were shorter than those observed on individuals of similar size in other studies and did not allow for detailed analyses of trajectories and turtle behavior. Our study further suggests that tracking durations are correlated with the targeted species, highlighting a lack of robustness against some neritic behaviors. Unprecedented diving data obtained for neonate sea turtles in this study suggest that the vertical behaviors of early juveniles are already too strenuous for these miniaturized tags. The anatomy of North American tortoises is poorly understood, despite a rich fossil record from the Eocene and younger strata. Stylemys nebrascensis is a particularly noteworthy turtle in this regard, as hundreds of specimens are known from Oligocene deposits, and as this species is one of the earliest fossil turtles to have been described in the scientific literature. Since its initial description based on a shell, many specimens with more complete material have been referred to Stylemys nebrascensis. Here, we review and confirm the referral of an important historic specimen to Stylemys nebrascensis, which includes shell, non-shell postcranial, and skull material. This allows us to document unique skull features of Stylemys nebrascensis (e.g., an unusual 'poststapedial canal' that connects the posterior skull surface with the cavum acustico-jugulare) and to refer another well-preserved skull to the species. Based on computed-tomography scanning of these two skulls, we provide a detailed description of the cranial and mandibular osteology of Stylemys nebrascensis. Stylemys nebrascensis has a combination of plesiomorphic skull characteristics (e.g., retention of a medial jugal process) and derived traits shared with extant gopher tortoises (e.g., median premaxillary ridge) that suggest it may be a stem-representative of the gopher tortoise lineage. This supports the hypothesis that extant and fossil tortoises from North America form a geographically restricted clade that split from Asian relatives during the Paleogene. [ABSTRACT FROM AUTHOR]

Numerous studies have identified traveling waves in the cortex and suggested they play important roles in brain processing. These waves are most often measured using macroscopic methods that are unable to assess the local spiking activity underlying wave dynamics. Here, we investigated the possibility that waves may not be traveling at the single neuron scale. We first show that sequentially activating two discrete brain areas can appear as traveling waves in EEG simulations. We next reproduce these results using an analytical model of two sequentially activated regions. Using this model, we were able to generate wave-like activity with variable directions, velocities, and spatial patterns, and to map the discriminability limits between traveling waves and modular sequential activations. Finally, we investigated the link between field potentials and single neuron excitability using large-scale measurements from turtle cortex ex vivo. We found that while field potentials exhibit wave-like dynamics, the underlying spiking activity was better described by consecutively activated spatially adjacent groups of neurons. Taken together, this study suggests caution when interpreting phase delay measurements as continuously propagating wavefronts in two different spatial scales. Longer distance water delivery culverts pose obstacles such as deposited silt, stones, and dead trees. In this paper, a crawling robot is designed to mimic the joint structure of a turtle using bionic design principles. The mechanism and gait of the robot are analyzed. The kinematics model of the robot is established using the D-H method and analytical approach, while the dynamics model is established using Lagrange's method. Based on kinematics and dynamics analysis theory, compound cycloid and cubic polynomial motion trajectories for the robot foot are planned along with a crawling gait resembling that of a turtle's abdomen. Simulation experiments and scale prototype experiments confirm that when gait parameters are identical, the energy consumption of compound cycloid trajectory exceeds that of cubic polynomial foot trajectory. When planning these two types of foot trajectories, it was observed that energy consumption ratio decreases with increasing step length but increases with increasing step height. Single‐visit surveys of plots are often used for estimating the abundance of species of conservation concern. Less‐than‐perfect availability and detection of individuals can bias estimates if not properly accounted for. We developed field methods and a Bayesian model that accounts for availability and detection bias during single‐visit visual plot surveys. We used simulated data to test the accuracy of the method under a realistic range of generating parameters and applied the method to Florida's east coast diamondback terrapin in the Indian River Lagoon system, where they were formerly common but have declined in recent decades. Simulations demonstrated that the method produces unbiased abundance estimates under a wide range of conditions that can be expected to occur in such surveys. Using terrapins as an example we show how to include covariates and random effects to improve estimates and learn about species‐habitat relationships. Our method requires only counting individuals during short replicate surveys rather than keeping track of individual identity and is simple to implement in a variety of point count settings when individuals may be temporarily unavailable for observation. We provide examples in R and JAGS for implementing the model and to simulate and evaluate data to validate the application of the method under other study conditions. [ABSTRACT FROM AUTHOR]

Four native chelonians species occur in Spain, three of which are severely threatened. In recent surveys, 25 species of alien turtles and tortoises were reported, some of which had established breeding populations and interfered with native species. In this study, we used ecological niche models to evaluate which regions are more vulnerable to colonization by alien turtles and tortoises and which alien chelonians show substantial environmental overlap with native species. The parameters of the ecological niche models were iteratively optimized using Akaike procedures, and we assessed the projection transferability between donor and recipient geographic regions using Mobility-Oriented Parity estimation. Our results indicated that the Mediterranean and some Atlantic rivers (Miño-Sil and Guadalquivir) are particularly vulnerable to colonization by semi-aquatic alien turtles, while southwestern Spain is exposed to colonization by alien tortoises. The aquatic species most susceptible to overlap with the four species of native turtles are those native to the cold and warm temperate regions of North America and China: Apalone spinifera, Chelydra serpentina, Chrysemys picta, Mauremys mutica, Mauremys reevesii, Mauremys sinensis, Pelodiscus sinensis, Sternotherus odoratus, and Trachemys scripta. Turtles and tortoises are among the most threatened vertebrate groups. Their life history is characterized by delayed sexual maturity and a long lifespan, making populations susceptible to decline following perturbations. Despite the urgent conservation need, we are missing estimates of basic demographic traits for many species and populations. The ornate box turtle Terrapene ornata is a species lacking crucial demographic data. Many populations are isolated in fragmented habitats, especially in the eastern portion of their range. We carried out long-term capture-mark-recapture surveys on two isolated populations in northern Illinois to estimate population vital rates and project population persistence with deterministic stage-based matrix models. Using 34 years of data, we estimated adult female survival = 0.974 (95% CI: 0.946-0.988) and juvenile survival = 0.867 (95% CI: 0.688-0.951) at our most intensively surveyed site. At a second site using eight years of data, we estimated adult female survival = 0.897 (95% CI: 0.783-0.954) and juvenile survival = 0.844 (95% CI: 0.551-0.960). Despite seemingly high annual survival rates, populations declined under population projections using mean vital rates. Population growth was most sensitive to adult survival, with increasing sensitivity under more pessimistic scenarios. Our results highlight the importance of long-term demographic studies for threatened species and demonstrate protecting adult female ornate box turtles is critical for ensuring populations persist at the northern edge of their distribution. Freshwater turtles comprise 81% of all chelonian species despite freshwater systems only occupying 1% of the earth's surface, and they are commonly exploited as pets and food resources. This contact between humans and turtles may put both sides at risk of disease transmission. Additionally, human impact on ecosystems can cause disease outbreaks in turtle populations. In this review, we focused on disease agents affecting freshwater turtles, intending to contribute to conservation and public health efforts. We analysed 423 articles and noted a post-SARS-COVID-19 peak, with most research originating from Asia, North America, and Europe. Emydidae was the most frequently studied family, and there was also a bias towards adults, live specimens, and native species. Since most of the studied turtles were wild-caught, we recommend that captive turtles should also be thoroughly studied since they can transmit diseases to other turtles and humans. We registered 2104 potential disease-causing agents, with Platyhelminthes dominating within Animalia, while Proteobacteria dominated bacterial agents. Viruses' representation was low, highlighting gaps in reptile virology. Fungi, Chromista, and Protozoa were also underrepresented, but this is changing with the development of molecular tools. This synthesis serves as a foundation for targeted health assessments, conservation strategies, and future research, essential to mitigate ecosystem and public health threats. [ABSTRACT FROM AUTHOR]

Flat hydrodynamic shells likely represent an evolutionary trade-off between adaptation to an aquatic lifestyle and the instability of more rounded shells, thought beneficial for self-righting. Trade-offs often result in compromises, this is particularly true when freshwater turtles, with flatter shells, must self-right to avoid the negative effects of inverting. These turtles, theoretically, invest more biomechanical effort to achieve successful and timely self-righting when compared to turtles with rounded carapaces. This increase in effort places these hatchlings in a precarious position; prone to inversion and predation and with shells seemingly maladapted to the act of self-righting. Here, we examine hatchling self-righting performance in three morphologically distinct freshwater turtle species (Apalone spinifera, Chelydra serpentina and Trachemys scripta scripta) that inhabit similar environmental niches. We demonstrate that these hatchlings were capable of rapid self-righting and used considerably less biomechanical effort relative to adult turtles. Despite differences in shell morphology the energetic efficiency of self-righting remained remarkably low and uniform between the three species. Our results confound theoretical predictions of self-righting ability based on shell shape metrics and indicate that other morphological characteristics like neck or tail morphology and shell material properties must be considered to better understand the biomechanical nuances of Testudine self-righting. Background: Studies have elucidated the importance of gut microbiota for an organism, but we are still learning about the important influencing factors. Several factors have been identified in helping shape the microbiome of a host, and in this study we focus on two factors—geography and host. We characterize the fecal microbiota of the Big-headed Turtle (Platysternon megacephalum) and compare across a relatively fine geographic scale (three populations within an 8-km radius) and between two syntopic hosts (P. megacephalum and Sacalia bealei). Both species are endangered, which limits the number of samples we include in the study. Despite this limitation, these data serve as baseline data for healthy, wild fecal microbiotas of two endangered turtle species to aid in conservation management. Results: For geography, the beta diversity of fecal microbiota differed between the most distant sites. The genus Citrobacter significantly differs between sites, which may indicate a difference in food availability, environmental microbiota, or both. Also, we identify the common core microbiome for Platysternon across Hong Kong as the shared taxa across the three sites. Additionally, beta diversity differs between host species. Since the two species are from the same site and encounter the same environmental microbiota, we infer that there is a host effect on the fecal microbiota, such as diet or the recruitment of host-adapted bacteria. Lastly, functional analyses found metabolism pathways (KEGG level 1) to be the most common, and pathways (KEGG level 3) to be statistically significant between sites, but statistically indistinguishable between species at the same site. Conclusions: We find that fecal microbiota can significantly differ at a fine geographic scale and between syntopic hosts. Also, the function of fecal microbiota seems to be strongly affected by geographic site, rather than species. This study characterizes the identity and function of the fecal microbiota of two endangered turtle species, from what is likely their last remaining wild populations. We sequenced the mitochondrial genomes (mitogenomes) and ten nuclear loci of 20 Emydura samples from the Carson, Mitchell, and Ord River drainages to examine the taxonomic identity of turtles with variable facial coloration from the Mitchell River. We analysed our data together with previously published sequences, including mitogenomes of crucial name-bearing type material. Our results provide evidence for the occurrence of a variably coloured single species in the Mitchell River that harbours two deeply divergent mitochondrial lineages. One of these lineages could originate from an ancient hybridization and mitochondrial introgression from the E. subglobosa/E. tanybaraga complex. Our results and published evidence suggest that Emydura represents a speciation continuum and that the evolutionary history of the genus is characterized by multiple hybridization and introgression events. Based on comparison with previously published mitogenomes of type material and our present results, we conclude that E. victoriae (Gray, 1842) is a junior synonym of E. australis (Gray, 1841). OBJECTIVE To compare dexmedetomidine-ketamine (DK; 0.1 and 10 mg/kg, respectively) with midazolam (M; 1.0 mg/kg) or 0.9% sodium chloride (S; 0.2 mL/kg) administered IM in the forelimb (F) or hindlimb (H) in eastern box turtles (Terrapene carolina carolina). ANIMALS 20 clinically healthy, captive adult eastern box turtles. METHODS In a randomized, blinded, complete crossover study with 1-week washout periods, turtles were administered each of 3 treatments: F-DKS, F-DKM, or H-DKM. Palpebral reflex, muscle tone, and withdrawal responses were serially assessed and used to calculate cumulative sedation scores at each 5-minute time point. The ability to intubate was evaluated. At 60 minutes, atipamezole (1.0 mg/kg) and either flumazenil (F-DKM, H-DKM; 0.05 mg/kg) or 0.9% sodium chloride (F-DKS; 0.5 mL/kg) were administered IM. RESULTS All treatments resulted in clinically relevant anesthetic effects. F-DKM produced significantly higher sedation scores than H-DKM or F-DKS at all time points between 10 and 60 minutes (P < .05). Sedation score variability was observed with all treatments with significantly higher variability for H-DKM (P < .05). Intubation was successful in 32, 89, and 11% of turtles in F-DKS, F-DKM, and H-DKM, respectively. Median (range) recovery time was 10 (5-22), 16 (7-45), and 12 (4-28) minutes for F-DKS, F-DKM, and H-DKM, respectively. CLINICAL RELEVANCE In eastern box turtles, forelimb dexmedetomidine-ketamine resulted in clinically relevant anesthetic effects that were heightened with the addition of midazolam. Hindlimb administration of midazolam-dexmedetomidine-ketamine resulted in reduced and more variable anesthetic effects compared to forelimb administration, supporting a hepatic first-pass effect. The Critically Endangered Nubian Flapshell Turtle (Cyclanorbis elegans) is found in the White Nile River system in South Sudan and northern Uganda. Over the past few decades, its populations have sharply declined, primarily due to human-induced threats, leading to its near-extinction across almost its entire range. In this paper, we present the results of a comprehensive threat analysis undertaken to pinpoint the challenges confronting the species within its natural habitat, specifically in the only known sites where it currently exists. We aimed to develop a conceptual framework to understand causal relationships among driving forces, threats and the target species. This framework allows the visualisation and analysis of the web of intricate interactions that contribute to the species' decline and vulnerability. We convened a panel of experts who identified three primary direct threats to the Nubian Flapshell Turtle: (1) Sand mining (IUCN code 3.2); (2) Overfishing by native fishers and refugees (code 5.4) and (3) Habitat loss at nesting sites (code 7.3). Among these threats, overfishing had the largest geographical extension, the highest intensity and duration, while habitat loss occurred at the highest frequency. These threats varied significantly in intensity and frequency. Additionally, when assessing the overall magnitude of these threats, habitat loss and overfishing had the highest impact, with significantly higher scores compared to sand mining. Our experts also identified the underlying driving forces behind these threats: (1) Economic interests driving sand mining activities along the Nile; (2) The influence of civil conflict and refugee populations, which contribute to the demand for bushmeat; (3) the Chinese expatriates, that lead to overfishing; (4) The removal of bank vegetation serves as a catalyst for habitat loss at nesting sites. The red-eared slider (Trachemys scripta elegans) is renowned for its remarkable adaptations, yet much of its complex biology remains unknown. In this pioneering study, we utilized a combination of gross anatomy, scanning electron microscopy (SEM), light microscopy, and immunofluorescence techniques to examine the tongue's omnivorous adaptation in this species. This research bridges a critical knowledge gap, enhancing our understanding of this intriguing reptile. Gross examination revealed a unique arrowhead-shaped tongue with a median lingual fissure and puzzle-piece-shaped tongue papillae. SEM unveiled rectangular filiform, conical, and fungiform papillae, with taste pores predominantly on the dorsal surface and mucous cells on the lateral surface of the papillae. Histologically, the tongue's apex featured short rectangular filiform and fungiform papillae, while the body exhibited varying filiform shapes and multiple taste buds on fungiform papillae. The tongue's root contained lymphatic tissue with numerous lymphocytes surrounding the central crypt, alongside lingual skeletal musculature, blood and lymph vessels, and Raffin corpuscles in the submucosa. The lingual striated muscle bundles had different orientations, and the lingual hyaline cartilage displayed a bluish coloration of the ground substance, along with a characteristic isogenous group of chondrocytes. Our research represents the first comprehensive application of immunofluorescence techniques to investigate the cellular intricacies of the red-eared slider's tongue by employing seven distinct antibodies, revealing a wide array of compelling and significant findings. Vimentin revealed the presence of taste bud cells, while synaptophysin provided insights into taste bud and nerve bundle characteristics. CD34 and PDGFRα illuminated lingual stromal cells, and SOX9 and PDGFRα shed light on chondrocytes within the tongue's cartilage. CD20 mapped B-cell lymphocyte distribution in the lingual tonsil, while alpha smooth actin (α-SMA) exposed the intricate myofibroblast and smooth muscle network surrounding the lingual blood vessels and salivary glands. In conclusion, our comprehensive study advances our knowledge of the red-eared slider's tongue anatomy and physiology, addressing a significant research gap. Viruses from a new species of piscichuvirus were strongly associated with severe lymphocytic meningoencephalomyelitis in several free-ranging aquatic turtles from 3 coastal US states during 2009-2021. Sequencing identified 2 variants (freshwater turtle neural virus 1 [FTuNV1] and sea turtle neural virus 1 [STuNV1]) of the new piscichuvirus species in 3 turtles of 3 species. In situ hybridization localized viral mRNA to the inflamed region of the central nervous system in all 3 sequenced isolates and in 2 of 3 additional nonsequenced isolates. All 3 sequenced isolates phylogenetically clustered with other vertebrate chuvirids within the genus Piscichuvirus. FTuNV1 and STuNV1 shared ≈92% pairwise amino acid identity of the large protein, which narrowly places them within the same novel species. The in situ association of the piscichuviruses in 5 of 6 turtles (representing 3 genera) with lymphocytic meningoencephalomyelitis suggests that piscichuviruses are a likely cause of lymphocytic meningoencephalomyelitis in freshwater and marine turtles. Simple Summary: The Asian yellow pond turtle (Mauremys mutica) is widely traded in China, and its artificial breeding has now become a major industry. However, the insufficient offspring supply and reproductive decline of farmed turtles make wild turtles more vulnerable. The present study was mainly designed to quantify the fecundity of M. mutica and attempt to screen for good reproductive performance in females. The parent–offspring relationships of all offspring in four consecutive years were confirmed using sixteen microsatellite loci. We also summarised the reproductive results of all females and counted the annual number of offspring and the variation in the number of offspring. The females were then divided into three types (stable, undulating and levelling off) according to the continuity. This study can provide the basis and materials for the creation of a good reproductive group and the study of the reproductive biology of turtles in M. mutica aquaculture. We selected seven females with good reproductive ability, which provided 16.94% of the annual contributions, while there were two females that had no offspring in four years. We also analysed the possible reasons for this difference and the importance of carrying out a family survey. Simple Summary: Turtles and tortoises are routinely presented to veterinarians for medical and surgical treatment. In many of these cases, sedation is required to properly examine the animal and perform necessary diagnostic tests. Unfortunately, there is a dearth of evidence-based research available to guide veterinarians working with these animals. In addition, the available publications show that there are differences between species. Therefore, with nearly 350 species of chelonians in the world, it is important for us to evaluate protocols on a species-by-species basis. In this study, six eastern mud turtles (Kinosternon subrubrum) were given 10 mg/kg alfaxalone via intramuscular (IM) and intravenous (IV) routes using a cross-over study design that allowed us to compare the results between and within turtles. As expected, the IV route led to a faster induction and recovery. This study demonstrated that alfaxalone 10 mg/kg IV or IM can be used to provide safe and effective sedation in eastern mud turtles. Current sedation protocols for chelonians can pose a challenge to clinicians because of prolonged induction and recovery times, difficulties in gaining venous access, and natural species variation. This study evaluated the sedative and physiologic effects of intramuscular (IM) and intravenous (IV) alfaxalone in six wild-caught adult eastern mud turtles (Kinosternon subrubrum). The turtles received alfaxalone 10 mg/kg IM and IV in a randomized cross-over design. A 10-day washout period occurred between trials. Baseline parameters (heart rate, respiratory rate, temperature, and reflexes) were assessed prior to injection and every 5 min post-injection until recovery. Three venous blood gas samples were also collected and analyzed over the course of each trial (baseline, induction, and recovery). Intravenous alfaxalone resulted in a significantly faster induction (p = 0.016; median: 1.5 min, 25–75%: 1–7.5, minimum–maximum: 1–21) and a shorter total sedation time (p = 0.041; median: 52 min, 25–75%: 34.5–62.5, minimum–maximum: 33–87) when compared with IM alfaxalone (induction, median: 20 min, 25–75%: 15–22.5, minimum–maximum: 15–25; total, median: 70 min, 25–75%: 65–82.5, minimum–maximum: 65–90). Blood gas and physiologic parameters were not significantly different between groups; however, the pH (p = 0.009) and glucose (p = 0.0001) significantly increased, and partial pressure of carbon dioxide (p = 0.024) significantly decreased over time. Although the fossil record of turtles in the Cretaceous of Patagonia is diverse and abundant, the record from the Austral-Magallanes Basin (Santa Cruz Province, Argentina, and Magallanes Region, Chile) is scarce and fragmentary. This contribution presents a new record of a turtle from the Piedra Clavada Formation (early Cenomanian) found near Tres Lagos Village, Santa Cruz Province, Argentina. This new fossil is characterized by a unique combination of character states: medium-sized carapace (length circa 50 cm), shell with a smooth surface, having narrow vertebral scutes, regular neural bones longer than wide, the tips of thoracic ribs seen in dorsal view, posterior marginals reaching costal bones, and not having costo-peripheral fontanelles. Furthermore, based on paleolatitudinal reconstructions, the new turtle from the Piedra Clavada Formation lived in the circumpolar region (54º S), representing the oldest southernmost record of turtles in the Late Cretaceous of South America. Considering this unique combination of characters and the comparison with other Early--early Late Cretaceous turtles from Gondwana, we can suggest that MSV103 does not only belong to a new species but a new, previously unrecognized lineage of turtles in southern Patagonia [ABSTRACT FROM AUTHOR]

Saxochelys gilberti is a baenid turtle from the Late Cretaceous Hell Creek Formation of the United States of America known from cranial, shell, and other postcranial material. Baenid turtles are taxonomically diverse and common fossil elements within Late Cretaceous through Eocene faunas. Detailed anatomical knowledge is critical to understanding the systematics and morphological evolution of the group. This is particularly important as baenids represent an important group of continental vertebrates that survived the mass extinction event associated with the Cretaceous/Paleogene boundary. High-resolution micro-computed tomography scanning of the holotype skull reveals additional anatomical details for the already well-known Saxochelys gilberti. This includes the revision of some anatomical statements from the original description, but also detailed knowledge on internal anatomical features of the braincase and the description of a well-preserved axis (cervical vertebra 2). Our new detailed description and previous work on the shell and postcrania make Saxochelys one of the best-described, nearly complete baenid turtles, which are often only known from either isolated shell or cranial material. A revised phylogenetic analysis confirms the position of Saxochelys gilberti as a derived baenid (Eubaeninae) more closely related to Baena arenosa than to Eubaena cephalica. [ABSTRACT FROM AUTHOR]

For over 70 million years, during the Paleozoic, the placoderms (Greek for 'plated skin'), an extinct group of armoured fishes, were the most abundant and diverse vertebrates on our planet. Some of the first placoderm fossils discovered — such as Bothriolepis with its bone-covered pectoral fins — seemed so bizarre that they were thought to represent turtles or ancient beetles. All placoderms bear thick overlapping dermal plates around the head (called the 'head shield') and an area of the body covered in similar overlapping plates enveloping the pectoral to anal region (called the 'trunk-shield'). Placoderm fossils (Figure 1) are known from every continent on Earth ranging from the early Silurian (∼438 million years ago) to the end Devonian (∼359 million years ago) when they became extinct. Male and female Chinese soft-shelled turtles (Pelodiscus sinensis) have sex-dimorphic growth patterns, and males have higher commercial value because of their larger size and thicker calipash. Thus, developing sex-specific markers is beneficial to studies on all-male breeding in P. sinensis. Here, we developed an accurate and efficient workflow for the screening of sex-specific sequences with ZW or XY sex determination systems. Based on this workflow, female and male P. sinensis reference genomes of 2.23 Gb and 2.26 Gb were obtained using de novo assembly. After aligning and filtering, 4.01 Mb female-specific sequences were finally identified. Subsequently, the seven developed sex-specific primer pairs were 100% accurate in preliminary, population, and embryonic validation. The presence and absence of bands for the primers of P44, P45, P66, P67, P68, and P69, as well as two and one bands for the PB1 primer, indicate that the embryos are genetically female and male, respectively. NR and functional annotations identified several sex-determining candidate genes and related pathways, including Ran, Eif4et, and Crkl genes, and the insulin signaling pathway and the cAMP signaling pathway, respectively. Collectively, our results reveal that a ZW-type sex-determination system is present in P. sinensis and provide novel insights for the screening of sex-specific markers, sex-control breeding, and the studies of the sex determination mechanism of P. sinensis. Designing monitoring with adequate certainty to evaluate management actions can be challenging, especially for elusive species in relatively inaccessible habitats. The diamond‐backed terrapin (Malaclemys terrapin) is considered a high priority species for management in all states within its range. Among key threats to terrapin populations is bycatch mortality in crab pots, prompting states throughout the species' range to consider crabbing regulations and creating a need for a monitoring approach able to evaluate the success of any new crabbing regulations to benefit terrapins. Because terrapins occupy extensive and often difficult to access estuarine habitat, it is hard to collect sufficient data to estimate population responses accurately and precisely for management. To assist state managers with decisions regarding monitoring bycatch reduction regulations, we modeled 12 years of terrapin capture‐recapture data. We used estimates from those models to simulate capture‐recapture data pre‐ and post‐implementation of bycatch reduction that resulted in a 0.20 absolute increase in mean apparent survival probability. Results indicated weak reliability of a monitoring approach using only manual seining of tidal creeks to detect a real management effect, with a positive management effect detected at the 95% certainty level only 34% of the time. When we considered 85% and 75% certainty thresholds, we detected a positive effect on survival among 61% and 75% of simulations, respectively. Low within‐year recapture probability and the effect of tide amplitude on terrapin availability indicate there is low feasibility of improving monitoring precision at a single site, requiring monitoring of more sites to improve confidence in the detection of the management effect. The number of sites monitored depends on the acceptable level of certainty. We recommend that researchers and management entities that use seining assess the level of certainty they require to evaluate management actions and increase the number of sites sampled to meet that level of certainty. Additionally, the use of multiple monitoring methods and integrated models should be explored to reduce uncertainty and to allow for easier monitoring of more populations over broader spatial scales. Fire is a vital management tool for maintaining prairie ecosystems. Prescribed burns control invasive species, regulate succession, stimulate plant growth, and are a cheap and effective method for removing excess biomass; however, fire can also inadvertently cause wildlife mortality, placing land managers in a challenging situation. Turtles are especially at risk of mortality from fire because of their low mobility and population sensitivity to reductions in adult survival. We studied ornate box turtles (Terrapene ornata) at 3 sites in Illinois, USA, from 2019–2022 to determine the best predictors of above‐ground activity so land managers can conduct prescribed burns when turtles are underground. We used turtle shell temperature, air temperature, soil temperature, and precipitation data to develop a predictive model of above‐ground activity. The best model for predicting above‐ground activity included an interaction between day of year and current air temperature. Earlier in spring and later in fall, above‐ground activity is more likely at higher air temperatures compared to later in spring and earlier in fall when the same likelihood of above‐ground activity is predicted at lower air temperatures. In spring, we recommend burning in Illinois ornate box turtle habitat before 1 April when air temperature is <10°C and in fall after 1 November when air temperature is <15°C. Maladaptive movement behavior is a leading cause of failure for reptile reintroductions, thus characterizing movement for translocated populations is important to project success. From 2014–2016 we radio‐tracked 183 immature reintroduced alligator snapping turtles (Macrochelys temminckii) using very high frequency telemetry in a stream in southern Illinois, USA. We assessed environmental, temporal, morphometric, and microhabitat factors influencing movement, including post‐release dispersal from reintroduction sites, movement probability, and step length. We used directional statistics to investigate the effects of precipitation and age on directional movement in the stream. Numerous factors influenced active movement, including ambient temperatures, time since release, use of log jams, turtle size, and age. Precipitation had an age‐biased effect on movement. Directional analyses suggested that movement was largely passive as turtles were swept downstream with increased discharge. Overall, the behavior of reintroduced alligator snapping turtles was similar to that reported in wild populations. Passive downstream transport has implications for reintroduced and natural populations as a force for unintentional emigration in channelized streams under current conditions and future high flow regimes. Headstarting involves ex situ rearing of vulnerable life stages, then releasing individuals into the wild once they are larger and less vulnerable to predation. Sometimes, headstarted animals display underdeveloped behaviors that may lead to an acclimation period of reduced survival and growth after release. Using data from a 6‐year headstarting program, we tested whether the early release condition affected survival, body condition, and somatic growth rate in 2 groups of headstarted Blanding's turtles (Emydoidea blandingii) released into Rouge National Urban Park (RNUP) in Toronto, Ontario, Canada. The first group included turtles released directly into the wild (i.e., hard release). The second group included turtles released into an in situ enclosure in which individuals remained for a week without food supplementation before being fully released into the wild (i.e., delayed release). Release condition did not affect survival or growth rate. In the delayed‐release group, body condition initially declined rapidly and remained low for up to 1 year after release. Given the lack of wild juveniles in RNUP, we compared body condition of headstarted turtles at various time points since release to similar‐sized wild juveniles from 2 other Ontario populations, one from Algonquin Provincial Park (APP) and one near Lake Erie (LE). Body condition of headstarted turtles was similar to those of wild APP turtles regardless of release method, and higher than those of wild LE turtles. Our results indicate that delayed release did not improve post‐release outcomes for headstarted turtles in an urban landscape and headstarted turtles sustain similar health metrics as wild turtles.
