Abstract

Phenalenyl is a radical nanographene with triangular shape that hosts an unpaired electron with spin S = 1/2. The open-shell nature of phenalenyl is expected to be retained in covalently bonded networks. Here, we study a first step in that direction and report the synthesis of the phenalenyl dimer by combining in-solution synthesis and on-surface activation and its characterization both on Au(111) and on a monolayer of NaCl on top of Au(111) by means of inelastic electron tunneling spectroscopy (IETS). IETS shows inelastic steps that, together with a thorough theoretical analysis, are identified as the singlet-triplet excitation arising from interphenalenyl exchange. Two prominent features of our data permit to shed light on the nature of spin interactions in this system. First, the excitation energies with and without the NaCl decoupling layer are 48 and 41 meV, respectively, indicating a significant renormalization of the spin excitation energies due to exchange with the Au(111) electrons. Second, a position-dependent bias-asymmetry of the height of the inelastic steps is accounted for by an interphenalenyl hybridization of the singly occupied phenalenyl orbitals that is only possible via third neighbor hopping. This hybridization is also essential to activate kinetic interphenalenyl exchange. Abstract

On the wake of the results obtained so far at the SPARC\_LAB test-facility at the Laboratori Nazionali di Frascati (Italy), we are currently investigating the possibility to design and build a new multi-disciplinary user-facility, equipped with a soft X-ray Free Electron Laser (FEL) driven by a $\sim$1 GeV high brightness linac based on plasma accelerator modules. This design study is performed in synergy with the EuPRAXIA design study. In this paper we report about the recent progresses in the on going design study of the new facility.

Abstract

Smith-Magenis syndrome (SMS) is a genetic neurodevelopmental disease characterized by neurological, psychiatric, anatomical, and motor symptoms. The disease is caused by deletions on chromosome 17p11.2, which may lead to the loss of up to 95 genes, depending on the length of the chromosomal rearrangement. One of these genes is retinoic acid-induced 1 (RAI1). Evidence that loss of RAI1 is responsible for several clinical manifestations of SMS came with the identification of patients carrying point mutations in this gene and presenting a phenotype overlapping with SMS. Rather, disease severity and some clinical presentations are associated with loss of genes other than RAI1. SMS patients are typically heterozygous for the mutation (RAI1 mutations and chromosomal deletions), indicating that loss of one functional allele of RAI1 is sufficient to cause disease. Interestingly, duplications of the same chromosomal region cause another neurodevelopmental disease with similar clinical manifestations, thus indicating that RAI1 (and possibly other genes nearby) are dosage-sensitive genes. RAI1 is a polyglutamine- and polyserine-containing factor induced by retinoic acid and with nuclear localization. However, its function in physiological conditions and how its haploinsufficiency causes SMS is not known. Here, we will review several aspects related to SMS, from diagnosis to clinical presentation. Moreover, we analyze in detail what is known about the RAI1 isoforms, expression pattern, native function, and the impact of different types of mutations (deletions, frameshift mutations that generate premature stop codons, and missense mutations that result in the production of the full-length protein). Finally, we review the animal and cell models available to date, focusing on those based on the immortalized pluripotent technology. Abstract

Smith-Magenis syndrome (SMS) is a neurodevelopmental disorder characterized by cognitive and behavioral symptoms, obesity, and sleep disturbance, and no therapy has been developed to alleviate its symptoms or delay disease onset. SMS occurs due to haploinsufficiency of the retinoic acid-induced-1 (RAI1) gene caused by either chromosomal deletion (SMS-del) or RAI1 missense/nonsense mutation. The molecular mechanisms underlying SMS are unknown. Here, we generated and characterized primary cells derived from four SMS patients (two with SMS-del and two carrying RAI1 point mutations) and four control subjects to investigate the pathogenetic processes underlying SMS. By combining transcriptomic and lipidomic analyses, we found altered expression of lipid and lysosomal genes, deregulation of lipid metabolism, accumulation of lipid droplets, and blocked autophagic flux. We also found that SMS cells exhibited increased cell death associated with the mitochondrial pathology and the production of reactive oxygen species. Treatment with N-acetylcysteine reduced cell death and lipid accumulation, which suggests a causative link between metabolic dyshomeostasis and cell viability. Our results highlight the pathological processes in human SMS cells involving lipid metabolism, autophagy defects and mitochondrial dysfunction and suggest new potential therapeutic targets for patient treatment.

Abstract

Familial Hypocalciuric Hypercalcemia (FHH1) is a rare autosomal dominant disease with low penetrance, caused by inactivating mutations of the calcium-sensing receptor (CaSR) gene, characterized by significant hypercalcemia, inappropriately normal serum PTH levels and a low urinary calcium level. Human induced pluripotent stem cells (hiPSCs) from a patient carrying a previously identified heterozygous mutation, a p.T972M amino acid substitution in cytoplasmic tail of CasR, were produced using a virus, xeno-free and non-integrative protocol.

Abstract

While many efforts are being currently spent to forge reliable damage laws based on the physics of the materials to be studied, damage modeling is still addressed numerically too naively in many situations. This article highlights some topical conceptual aspects that have been up to now dealt with too superficially by comparing the performances of different numerical algorithms in solving Karush–Kuhn–Tucker conditions for a simple linearly softening Hooke’s spring. It is concluded that even such a primitive model, because of the multiplicity of solutions satisfying simultaneously equilibrium, damage law and irreversibility conditions, actually requires well-established numerical algorithms to face unexpected challenges. A comparison between different numerical strategies, beyond highlighting critical behaviors of traditional algorithms, permitted to observe an appealing robustness shown by an iterative strategy based on the fixed-point theorem. As a closure remark, evidences collected within this contribution naturally lead to the following question, which is left open for future studies: Is it possible to envisage the formulation of a criterion– possibly an energetic one, like that distinguishing stable and unstable solutions in elasticity—to establish which solution should be considered as valid in a given situation?

Abstract

Background: Currently, no data are available on the burden of morbidity and mortality in people with HIV-1 (PWH) harboring a 4-class drug-resistant (4DR) virus (nucleoside reverse transcriptase inhibitors, non-nucleoside reverse transcriptase inhibitors, protease inhibitors, integrase strand transfer inhibitors). The study aimed to assess the incidence of clinical events and death in this population. Methods: This was a cohort study on PWH from the PRESTIGIO Registry with a documented 4DR virus. Burden of disease was defined as the occurrence of any new event including an AIDS-defining event (ADE) or non-AIDS-defining event (NADE) or death from any cause after 4DR evidence (baseline). Cox regression models evaluated factors associated with the risk of new clinical events/death. Results: Among 148 PWH followed for a median (interquartile range) of 47 (32-84) months after 4DR evidence, 38 PWH had 62 new events or died from any cause (incidence rate, 9.12/100 person-years of follow-up; 95% CI = 6.85-11.39): 12 deaths (6 AIDS-related and 6 non-AIDS-related), 18 ADEs, 32 NADEs; 20 of the 38 NADEs (45%) of the incident clinical events were malignancies. The 4-year cumulative incidence of death was 6% (95% CI, 3%-13%), and that of ≥1 event or death was 22% (95% CI, 16%-31%). A higher risk of new clinical events/death was more likely in PWH with previous clinical events (adjusted hazard ratio [aHR], 2.67; 95% CI, 1.07-6.67) and marginally associated with lower baseline CD4+/CD8+ ratio (aHR, 0.82; 95% CI, 0.65-1.02). Conclusions: PWH harboring 4DR have a high burden of disease with a worrying incidence of malignancies, strongly advising for close prevention and monitoring interventions as well as access to innovative therapeutic strategies, especially in people with a history of clinical events and low CD4+/CD8+ ratio.

Abstract

The NMDARs are key players in both physiological and pathologic synaptic plasticity because of their involvement in many aspects of neuronal transmission as well as learning and memory. The contribution in these events of different types of GluN2A-interacting proteins is still unclear. The p140Cap scaffold protein acts as a hub for postsynaptic complexes relevant to psychiatric and neurologic disorders and regulates synaptic functions, such as the stabilization of mature dendritic spine, memory consolidation, LTP, and LTD. Here we demonstrate that p140Cap directly binds the GluN2A subunit of NMDAR and modulates GluN2A-associated molecular network. Indeed, in p140Cap KO male mice, GluN2A is less associated with PSD95 both in ex vivo synaptosomes and in cultured hippocampal neurons, and p140Cap expression in KO neurons can rescue GluN2A and PSD95 colocalization. p140Cap is crucial in the recruitment of GluN2A-containing NMDARs and, consequently, in regulating NMDARs’ intrinsic properties. p140Cap is associated to synaptic lipid-raft (LR) and to soluble postsynaptic membranes, and GluN2A and PSD95 are less recruited into synaptic LR of p140Cap KO male mice. Gated-stimulated emission depletion microscopy on hippocampal neurons confirmed that p140Cap is required for embedding GluN2A clusters in LR in an activity-dependent fashion. In the synaptic compartment, p140Cap influences the association between GluN2A and PSD95 and modulates GluN2A enrichment into LR. Overall, such increase in these membrane domains rich in signaling molecules results in improved signal transduction efficiency.

Abstract

This paper presents a macroeconomic agent based model with endogenous innovation-driven growth and knowledge accumulation which aims to analyze the underlying causes of the recent increase in market concentration, by focusing on the interplay of technical change and market power, and the resulting macroeconomic consequences in terms of higher inequality and lower growth. The source of concentration lies in the fact that heterogeneous firms do not have equal access to capital-embodied innovations, as we assume that this depends on the “knowledge gap”, i.e., the difference between the degree of capital good's technical advancement and the firm's accumulated technological knowledge. The analysis shows that, in the absence of consistent knowledge spillovers and as long as capital goods remain considerably different from each other, technical progress generates systematic differences in productivity across firms, leading to a reallocation of market shares towards more productive firms. Consequently, as the newly-emerging large firms seek to translate the enhanced market power into higher mark-ups, the resulting shift in the income distribution from wages to profits eventually undermines aggregate demand and growth. Yet, simulation experiments reveal that the evolution of market concentration over time as well as its macroeconomic effects crucially depend on the presence (or lack thereof) of legal entry barriers, which, by influencing the process of diffusion of technological innovations, reinforce (or attenuate) the large firms’ ability to consolidate their dominant position and thus exploit their market power.

Abstract

BackgroundCurrently, no data are available on the burden of morbidity and mortality in people with HIV-1 (PWH) harboring a 4-class drug-resistant (4DR) virus (nucleoside reverse transcriptase inhibitors, non-nucleoside reverse transcriptase inhibitors, protease inhibitors, integrase strand transfer inhibitors). The study aimed to assess the incidence of clinical events and death in this population.MethodsThis was a cohort study on PWH from the PRESTIGIO Registry with a documented 4DR virus. Burden of disease was defined as the occurrence of any new event including an AIDS-defining event (ADE) or non-AIDS-defining event (NADE) or death from any cause after 4DR evidence (baseline). Abstract

Litho- and biostratigraphic data are provided of 5 stratigraphic sections in Romania covering the “Badenian” marine flooding that occurred in the Central Paratethys during the middle Miocene (Langhian). The dataset includes stratigraphic logs and descriptions of the profiles, and biostratigraphic analyses on calcareous nannofossils and foraminifera. In addition, characteristic stratigraphic features and representative fossils, including tiny Streptochilus foraminifera in the Campiniţa section in the SE Carpathian Foredeep, are presented in photographs. The data show that the flooding is characterized by the sudden abundance of Langhian calcareous nannofossils and foraminifera with a strong Mediterranean affinity.
