Amaç: Arka bacak iskemisi/reperfüzyonun (İ/R) sıçan modellerinde, rapamisinin memelilerdeki hedefi (mTOR)/inhibitör-κB-α/nükleer faktör-κB p65 sinyal ileti yolu etkinliğinin, artan oksidatif/nitrozatif stres ve inflamatuvar yanıt yoluyla organ zedelenmelerine aracılık ettiğini daha önce göstermiştik. mTOR’un İ/R zedelenmesine katkıda bulunduğuna ilişkin önceki bulgularımızı referans alarak, bu çalışmada arka bacak İ/R'ye bağlı hedef ve uzak organ zedelenmelerinde mTOR ile MEK1/ERK1/2 yolu arasındaki olası etkileşime odaklanmayı amaçladık. Gereç ve Yöntem: Erkek Wistar sıçanlar dört gruba ayrıldı. Arka bacak İ/R, her iki arka ekstremitelerine turnikeler uygulanarak iskemi oluşturuldu. İskemiden 4 saat sonra turnikeler açılarak 4 saat reperfüzyon uygulandı. 4 saatlik reperfüzyondan sonra kan, böbrek ve gastroknemius kası izole edildi.Bulgular: Arka bacak İ/R uygulaması gastroknemius kasında, böbrekte ve/veya serumda rpS6, MEK1, ERK1/2, tümör nekroz faktörü-α, indüklenebilir nitrik oksit sentaz, gp91phox, p22phox ve nitrotirozinin fosforilasyonu ve/veya ekspresyonu ile birlikte nitrit düzeylerinde artışa neden oldu. Ayrıca, İ/R uygulanan sıçanların dokularında nikotinamit adenin dinükleotit fosfat oksidaz ve miyeloperoksidaz seviyeleri arttı. mTOR'un seçici inhibitörü olan rapamisin, sıçanlarda kas ve böbrek dokusunda İ/R'nin neden olduğu yukarıda bahsedilen tüm etkileri ortadan kaldırdı.Sonuç: Bu veriler, MEK1/ERK1/2 yolunun etkinliğinin, mTOR’un aracılık ettiği arka bacak İ/R’nin neden olduğu hedef ve uzak organ zedelenmesine katkıda bulunduğunu göstermektedir., Purpose: We have previously demonstrated that activation of the mammalian target of rapamycin (mTOR)/inhibitory-κB-α/nuclear factor-κB p65 signaling pathway mediates organ injuries through increased oxidative/nitrosative stress and inflammatory response in rat models of hind limb ischemia/reperfusion (HL I/R). Following up our previous findings regarding I/R injury through mammalian target of rapamycin (mTOR), we aimed to focus on the possible interaction between mammalian target of rapamycin (mTOR and mitogen-activated protein kinase kinase (MEK)1/extracellular signal-regulated kinase (ERK) 1/2 pathway in hind limb ischemia/reperfusion (HL I/R) resulting in target and remote organ injuries in the present study. Materials and Methods: Male Wistar rats were divided into four groups. HL I/R was induced by occluding with tourniquets of both hind limbs. Following 4 h, the tourniquets were removed following reperfusion for 4 h. After 4 h of reperfusion blood, kidney, and gastrocnemius muscle were collected.Results: HL I/R caused an increase in phosphorylation and/or expression of rpS6, MEK1, ERK1/2, tumor necrosis factor-α, inducible nitric oxide synthase, gp91phox, p22phox, and nitrotyrosine as well as nitrite levels in gastrocnemius muscle, kidney, and/or serum. Additionally, nicotinamide adenine dinucleotide phosphate oxidase and myeloperoxidase levels were increased in the tissues of rats subjected to HL I/R. Rapamycin, the selective inhibitor of mTOR, abolished all the effects mentioned above caused by HL I/R in the rat’s muscle and kidney. Conclusion: These data suggest that activation of the MEK1/ERK1/2 pathway contributes to mTOR-mediated HL I/R-induced target and remote organ injury.

Purpose: Ramipril is effective in treating inflammatory myocardial injury by reducing cytokines such as TNF-?, IL-6, and IL-8. In this study, we investigated the effect of cytokine gene polymorphisms on inflammatory response which might be reduced by ramipril. Material and Methods: Of 102 patients undergoing on-pump coronary artery bypass grafting surgery, 51 were the treatment group which received ramipril and the remaining 51 were the non-treated control group. TNF-?, IL-6, and IL-8 were measured with Enzyme-Linked Immuno Sorbent Assay (ELISA), before anesthesia induction (t1), at 20 min following cross clamping (t2), at the end of the operation (t3), and 24 hours after anesthesia (t4). Genotyping was performed with PCR method. Results: While TNF-? increase began with surgery, IL-6 and IL-8 increase began with cardiac arrest during cardiopulmonary bypass and continued until the end of the operation. In contrast, only IL-8 remained high in the control group during the postoperative period, while TNF-?, IL-6, and IL-8 began to decrease. The decreases in IL-6 at t3 and in TNF-? at t4 were significant. Conclusion: Ramipril might have a role in preventing inflammatory myocardial injury by reducing cytokine and TnT levels after cardiac arrest., Amaç: Ramipril, inflamatuar miyokardial hasarın tedavisinde TNF-?, IL-6 ve IL-8 gibi sitokin seviyelerini azaltarak etkili olmaktadır. Çalışmamızda, ramipril tedavisinin, koroner arter bypass cerrahisi esnasında oluşan inflamatuar yanıt ve miyokardiyal hasara yönelik etkisi üzerinde, sitokin gen polimorfizmlerinin herhangi bir rolü olup olmadığını araştırdık. Gereç ve Yöntem: Çalışmamıza, koroner arter bypass cerrahisi uygulanan hastalardan cerrahi öncesi ramipril alan 51, kontrol grubu olarak ise ramipril almayan 51 olmak üzere toplam 102 hasta dahil edildi. Her iki grupta, anestezi başlamadan hemen önce (t1), kros klemp sonrası 20. dakikada (t2), ameliyatın sonunda (t3) ve anestezinin başlangıcından 24 saat sonra (t4) alınan kan örneklerinden TNF?, IL-6, IL-8 ve TnT serum düzeyleri ELISA yöntemi ile, genotipleme ise PCR-RFLP yöntemleri ile belirlendi. Bulgular: Her iki grupta TNF-? artışı cerrahi ile başlarken, IL-6 ve IL-8 artışı kardiyopulmoner bypass sırasında kardiyak arrestle başlayıp ameliyat sonuna kadar devam etmiştir. Postoperatif dönemde ise sadece IL-8 kontrol grubunda yüksek kalırken; çalışma grubunda TNF-?, IL-6, IL-8 düşüşe geçmektedir. t3 zaman aralığında IL-6, t4 zamanında TNF? değerlerinde anlamlı azalma gözlenmiştir. Sonuç: Ramiprilin kardiyak arrest sonrası sitokin ve TnT düzeylerini azaltarak inflamatuar miyokardiyal hasarı önlemede yeri olabilir.

Zymosan is a component produced by polysaccharide which is obtained from cell wall of Saccharomyces cerevisiae. Zymosan consists of cross-linked polysaccharides such as 1,3-ß glucan, 1,6-ß glucan, and ?-mannan. Zymosan plays a role in inflammmatory diseases such as fungal sepsis, non-septic shock, multiple organ dysfunction syndrome, acute peritonitis, irritable bowel syndrome and it is used in related experimental models. Toll-like receptor 2, dectin-1, mannose, and compleman receptors mediate zymosan’s effects on immune system cells such as monocytes, macrophages, and dendritic cells. Spleen tyrosine kinase (Syk) is a cytosolic, intracellular, 72-kDa protein tyrosine kinase. It is demonstrated that Syk is expressed in hematopoietic cells such as mast cells, neutrophils, macrophages, B cells, leukocytes, and platelets and in non-hematopoietic cells such as epithelial, fibroblast, neuronal cells, and vascular endothelial cells. Spleen tyrosine kinase is a critical kinase that mediates cellular responses through dectin-1 receptor by zymosan. In this review, the role of syk in is evaluated in the signal transduction of zymosan., Zimosan, Saccharomyces cerevisiae’nin hücre duvarından elde edilen, polisakkarit zincirlerinden oluşan bir bileşendir. Dalak tirozin kinazı (Dtk), 72 kDa ağırlığında reseptör ile kenetli olmayan bir intraselüler tirozin kinazdır. Dtk’nin, mast hücreleri, nötrofiller, makrofajlar ve trombositler gibi tüm hematopoetik hücrelerde ve fibroblastlar, epitel, sinir ve damar endotel hücreleri gibi hematopoetik olmayan hücrelerde eksprese edildiği gösterilmiştir. Dtk, zimosanın dektin-1 reseptörü aracılığıyla oluşturduğu hücresel yanıtlara aracılık eden önemli bir kinazdır. Bu derlemede, zimosan sinyal iletisinde Dtk’nin rolü değerlendirilmiştir.

The Effect of Inducible Nitric Oxide Synthase Inhibition on Increased Oxidative Stress in the Liver of Endotoxemic Rats Objective: In this study, we aimed to investigate the effect of nitric oxide produced by inducible nitric oxide synthase to oxidative stress in the liver of endotoxemic rats. Method: Male Wistar rats received either saline (4 ml/kg, i.p.) in control group, endotoxin (10 mg/kg, i.p.) in endotoxic shock group and selective inducible nitric oxide synthase inhibitor phenylene-1,3-bis[ethane-2-isothiourea] dihydrobromide (10 mg/kg, i.p.) one hour after saline or endotoxin injection in the other groups. Mean arterial blood pressure was measured within four hours. The liver was isolated from the rats for the measurement of nitrite as an index of nitric oxide production, malondialdehyde for lipid peroxidation and the level of a potent antioxidant reduced glutathione. The activities of the antioxidant enzymes superoxide dismutase and catalase and the activity of myeloperoxidase as an index of neutrophil infiltration were measured. Rho Kinase and Cardiovascular Diseases Arterial diseases such as arteriosclerosis and hypertension are characterized by proliferation and hypertrophy of smooth muscle cells. The common feature of these diseases is endothelial dysfunction, which regulates the growth and tone of vascular smooth muscle cells. Inflammatory mediators like tumor necrosis factor alfa, interleukin-1β cause accumulation and adhesion of monocytes to the vessel wall by up-regulation of monocyte chemotactic peptide-1 and adhesion molecules such as vascular cell adhesion molecule-1, intercellular cell adhesion molecule-1, as well as migration, proliferation of vascular smooth muscle cells, and vessel obstruction. Rho kinase, sub-effector of small G protein Rho has two isoforms named as ROKα/ROCK II and ROKβ/ROCK I. ROCK mediates various cellular functions such as smooth muscle contraction, actin cytoskeleton formation, cell adhesion and motility, proliferation, cytokinesis, and gene expression that play a role in pathogenesis of cardiovascular diseases. Preclinical and clinical studies with selective ROCK inhibitors, fasudil and Y-27632, have shown that ROCK has an important role in the pathogenesis of cardiovascular diseases such as coronary vasospasm, hypertension, pulmonary hypertension, angina, myocardial infarction, stroke and heart failure. Also, beneficial effects of prolonged inhibition of ROCK in cardiovascular diseases have been demonstrated. Overproduction of reactive oxygen and nitrogen species leads to oxidative stress and decreased total antioxidant capacity, which is responsible for high mortality from several diseases such as endotoxic shock. Nitric oxide (NO) produced by inducible NO synthase (iNOS) during endotoxemia is the major cause of vascular hyporeactivity, hypotension and multiple organ failure. We investigated whether NO-mediated oxidative stress in rat heart, aorta and mesenteric artery is involved in the attenuation of endotoxin-induced hypotension by inhibition of iNOS. In conscious male Wistar rats, injection of endotoxin (10 mg/kg, i.p.) caused a gradual fall in mean arterial pressure (MAP) for 4 hours and increased serum and tissue nitrite levels. These effects of endotoxin were prevented by selective inhibition of iNOS with phenylene-1,3-bis[ethane-2-isothiourea] dihydrobromide (1,3-PBIT) (10 mg/kg, i.p.; 1 hour after endotoxin). Myeloperoxidase (MPO) activity was increased in the heart and aorta and decreased in the mesenteric artery by endotoxin which was reversed by 1,3-PBIT. Endotoxin caused a decrease in products of lipid peroxidation in the tissues, which is prevented by 1,3-PBIT. These data suggest that NO-mediated decrease in MAP during endotoxemia is associated with decreased oxidative stress in the heart, aorta and mesenteric artery and that the beneficial effects of iNOS inhibitors on the endotoxin-induced hypotension may be due to restoration of total antioxidant capacity.

Bisphenol A (BPA) is an environmental endocrine-disrupting chemicals that is widely used in common consumer products. There is an increasing concern regarding human exposure to BPA owing to the potential adverse effects associated with its estrogenic activity. For assessing environmental exposure to BPA, it is essential to have a sensitive, accurate and selective analytical method, especially one that can detect low BPA levels in complex sample matrices. In this study, we developed and validated an accurate, sensitive, and robust liquid chromatography-tandem mass spectrometry method for simultaneous quantification of free BPA and BPA -d-glucuronide (BPA-gluc) concentrations in human urine with only a single injection. Calibration curves were linear over a concentration range of 1-100ng/mL for BPA and 10-1000ng/mL for BPA-gluc. The levels of the analytes were determined quantitatively with HPLC/ESI-MS/MS by using negative electrospray ionization in the select ion monitoring mode and a pentaflouraphenyl propyl column. Daha önce farede oluşturulan deneysel septik şok modelinde yaptığımız bir çalışmada, indometazinin (1, 10 veya 100 mg/kg, i.p.) endotoksin (10 mg/kg, i.p.) ile nitrik oksit (NO) ve eikozanoit oluşumundaki artışı önlediği, ancak endotoksin ile artan mortaliteyi etkilemediği gösterilmiştir. Bu çalışmanın amacı, endotoksin ile oluşan mortalite üzerinde indometazinin etkisine NO ve tümör nekroze edici faktör (TNF)- α’nın katkısını araştırmaktır. Farelere 9:00'da endotoxin (10 mg/kg, i.p.), indometazin (siklooksijenaz (COX) inhibitörü) (100 mg/kg, i.p.), aminoguanidin (indüklenebilir NO sentaz inhibitörü) (100 mg/kg, i.p.) ve/veya pentoksifilin (TNF- α oluşumu ve aktivitesi inhibitörü) (90 mg/kg, i.p.) uygulanmıştır. Madde uygulanmasından 15 saat sonra kan örnekleri alınmış ve serum örnekleri nitrit konsantrasyonları ölçümü için kullanılmıştır. Endotoksin uygulanmasından sonra artan serum nitrit düzeyleri indometazin ve aminoguandinin ile azalmış, pentoksifilin ile değişmemiştir.Endotoksin ile artan mortalite ise aminoguanidin ve pentoksifilin tamamen önlenirken, indometazin ile değişmemiştir. Indometazinin endotoksin ile oluşan mortalite üzerindeki etkisizliği aminoguanidin ve/veya pentoksifilin ile değişmemiştir. The aim of the present study was to investigate the contribution of NO and tumor necrosis factor (TNF)-α to the effect of indomethacin on endotoxin-induced mortality. Animals were injected with endotoxin (10 mg/kg, i.p.) alone or in combination with indomethacin [cyclooxygenase (COX) inhibitor] (100 mg/kg), aminoguanidine (inducible NO synthase inhibitor) (100 mg/kg), and/or pentoxifylline (TNF- α production and activity inhibitor) (90 mg/kg) at 9:00 a.m. The blood samples were collected 15 h after the drug administration and serum samples were used for the measurement of nitrite concentrations. The endotoxin-induced increase in serum nitrite levels was decreased by indomethacin and aminoguanidine, but not by pentoxifylline. Endotoxin-induced mortality was prevented by aminoguanidine and pentoxifylline, but not by indomethacin. The ineffectiveness of indomethacin on endotoxin-induced mortality was not changed by aminoguanidine and/or pentoxifylline treatment. These results suggest that the overproduction of NO and/or TNF-a. may not contribute to the effect of indomethacin on endotoxin-induced mortality.

Ö Z E T Bu çalışmada, (1) makrofa] hücre kültürlerinde açığa çıkan NO miktarlarının, kolorimetrik olarak diazotizasyon ve izole kobay aortasında biyoesey yöntemlerinin kullanılmasıyla indirekt olarak belirlenmesi; (2) ölçülen NO miktarları bakımından bu yöntemler arasında karşılaştırma yapılması ve (3) aktif pulmoner tüberkülozlu hastaların kanlarından izole edilen monosit/makrofajların NO sentezleme yetenek lerinin, sağlıklı bireylerinki ile karşılaştırılması amaçlanmıştır. Diazotizasyon çalışmalarında (1) insan monosit/makrofaj kültürlerinin LPS ile uyarımdan sonra nitrit oluşumu bakımından 4 günden fazla süreyle izlenmesiyle kontrole göre anlamlı miktarlarda nitrit düzeyleri elde edilebilir; (2) LPS (25 ng/200 Iil) ile hücreler uyarıldığında anlamlı miktarlarda nitrit düzeyleri elde edilebilmektedir; (3) nitrit miktarları kültür süresince belli bir güne dek artmakta, daha sonra azalmak tadır; (4) nitrit miktarlarının maksimuma ulaştığı günler, bireyden bireye farklılık göstermektedir; (5) insanlar, monosit/makrofajları nitrit oluşturma yetenekleri bakımından, 10 ^M'dan daha düşük nitrit oluşturucular ve 10 nM'dan daha fazla nitrit oluşturucular olmak üzere iki gruba ayrılabilirler, (6) tüberkülozlu deneklerde görülen nitrit oluşumu sağlıklı deneklere göre daha azdır, (7) gerek sağlıklı gerekse tüberkülozlu deneklerde nitrit oluşumu L-NAME ile azaltıiabilmektedir. Biyoesey çalışmalarında (8) izole endotelsizleştirilmiş kobay aortası kul lanılarak, monosit/makrofajlardan sentezlenip salınan nitrit ve/veya nitrit-benzeri madde miktarları belirlenebilir, (9) kültür süpernatantlarıyla elde edilen gevşeme-145- yanıtlarından nitrit ve/veya nitrit-benzeri maddeler sorumlu gibi görünmektedir; (10) tüberkülozlu deneklerde, sağlıklılara göre daha fazla gevşeme yanıtları ve nitrit ve/veya nitrit-benzeri gevşetici madde miktarları elde edilebilmektedir; (11) sağlıklı deneklerde daha düşük nitrit miktarları elde edilmesinin nedeni, LPS uyarımı sonu cunda makrofajlardan salınan kastına maddelerin, geveşetici maddelerin etkisini baskılaması olabilir; (12) tüberkülozlularda daha fazla nitrit miktarları elde edilmesinin nedeni ise, NO-benzeri gevşetici aktivite gösteren maddeler ve sik- looksijenaz metabolitleri olabilir. SUMMARY In the present investigations, it was aimed that (1) indirect measurement of NO production by human cultured monocytes/macrophages stimulated with LPS by using two independent methods: a colorimetric measurement of the reaction of NO with Griess reagent and a bioassay for NO-mediated relaxation of preconstricted rings of guinea pig aorta; (2) to compare the nitrite amounts by the above men tioned methods and (3) to compare the ability of macrophage NO synthesis from healthy and active pulmonary tuberculous patients. In the diazotization studies it was concluded that (1) nitrite production can be observed up to four days for obtaining significant levels of nitrite; (2) LPS (25 ng/200 ^-induced cell stimulation caused significant levels of nitrite; (3) nitrite con centrations in the culture supematants gradually reached the peak levels at desig nated days following then to decrease; (4) nitrite production was reached a maxi mum at different time-points suggesting that this production was heterogeneous from donor to donor; (5) human monocytes can be classified into two different populations: a low nitrite-producing (< 10 M) and a high nitrite-producing (> 10 M) one; (6) in the tuberculous donors, nitrite production was observed but this produc tion was lower than the healthy donors and (7) L-NAME decreased the NO produc tion by macrophages isolated from healthy and tuberculous donors. In the bioassay studies it was concluded that (8) norepinephrine precon stricted deendothelised guinea-pig aorta ring can be used for the determination of-147- nitrite-like substances released from monocyte/macrophages; (9) these substances seems likely to be responsible for the relaxations obtained with supematants; (10) the reason of the increased nitrite levels obtained from tubereculous patients is that the supematants have nitrite like-activity and/or a nitrite-like relaxing activity in duced by other substances both elaborate relaxation with a masked effect on va soactive substances being found at the same time. As a result, substances having nitrite and/or nitrite-like relaxant activity ob tained from cultured human monocyte/macrophages are detectable by the diazoti- zation and the bioassay methods but the latter method is more sensitive than the diazotization method. In measuring the NO production sensitivity of the bioassay method can be specified by using inhibitors of nitrite-like relaxing factors during culture period and/or in the organ bath. 200

BAZI BRONKOKONSTRÎKTÖR MADDELERİN ETKİLERİNDE EP i TEL İN ROLÜ Bahar TUNÇTAN Doc- Dr. Nurettin ABACIOGLU (Tez Yöneticisi) Farmakoloji Anabilim Dalı (1992) ÖZET Bu çalışmada kobay trakeası ve SAK' tan oluşan ko-aksiyal biyoesey sisteminde 5-HT ve BK'nın etkilerinin epitelle ilişkisi ve kullanılan deneysel anaflaksi modelinde in vitro OA uygulamasından sonra bu ilişkide ne gibi bir derişiklik meydana geldiği incelenmiştir. Ko-aksiyal sistemde 5-HT düşük dozlarda gevşeme yüksek dozda kasılma seklinde bifazik cevap oluştururken, SAK tek basınayken ve epitelsizlestirilmis kobay trakeası içine yerleştirildiğinde doz-ba^ımlı kasılmalar oluşturması, oluşan deQisikliQin epitelden kaynaklandığını göstermektedir. Ko-aksiyal sistemde, imipramin ve parjilin muhtemelen 5-HT 'ye ait gevşemeleri, 5-HT'nin uptake ve parçalanmasını engelleyerek güçlendirmektedirler. Ko-aksiyal sistemde, ortamda İNDO bulunması halinde alınan sonuçlardan, trakea epi telinden SAK' ta inhibitor etkili bir SO ürününün yanısıra başka bir inhibitor maddenin de salmdiQi izlenimi edinilmiştir. Kullandığımız bir diŞer bronkokonstriktör madde olan BK SAK' ta doz-ba9ımlı gevşeme oluşturmuş, bu gevşemeler ko-aksiyal sistemde anlamlı olarak azalmış, epitelsizlestirmeden sonra ise yine anlamlı bir azalma görülmüştür. Bu olay epitelden salıverilen inhibitor maddelerin, gevşemelere katkıda bulunabilecek düzeyde olmadığını, ya da aksine BK tarafından epitel veya epitel-dısı dokulardan salıverilmesi stimüle edilen eksitatör maddeler nedeniyle gevşemelere katkısının maskelendiğini düşündürmektedir. îNDO ve kaptopril'in gevşemeleri anlamlı olarak etkilememesi ko-aksiyal 'sistemde BK'nın olusturduQu gevşemeleri prostanoid yapıda olmıyan eksitatör maddelerin azaltabileceğini ve BK'nın epitelde bulunan karboksipeptidaz dışındaki enzimler tarafından metabolize edildiğini göstermektedir. OA ile önceden aktif olarak duyarl ılastırı Imıs kobaylardan izole edilen kobay trakeaları ile hazırlanan ko-aksiyal sistemde, 5-HT ile oluşan bifazik cevap in vitro OA uygulanmasından sonra tamamen kasılmalara dönüşmesi, diQer yandan BK ile elde edilen gevşemelerin ise in vitro OA uygulaması sonrasında yalnızca düşük dozlarda anlamlı olarak artmasının OA uygulanmasından sonra epitelde oluştuğu saptanan morfolojik ve fonksiyonel derişikliklere baQli olabileceği anlaşılmıştır. Geliştirdiğimiz modifiye ko-aksiyal sistemde prekontrak ti 1 madde olarak kullandığımız FE ile oluşan kasılmaların platoyu sürdüremiyerek gevşemeye dönüşmesi, ayrıca bu gevşemelerin epitelsizlestirilmesinden sonra azalması FE'nin oluşturduğu kasılmaların azalmasının inhibitor f aktör ( ler )' in bazal salıverilmesi nedeniyle meydana gelebileceğini düşündürmektedir.SUMMARY In the present study, it has been investigated that the relationship between epithelium and the effects of 5-HT and BK, and whether there was a difference in this relationship after in vitro OA challenge in the experimental anaphylaxis model used. 5- HT produced dose-dependent contractions on the precontracted RAM which was mounted alone and when it was mounted co-axial ly within epithelium-denuded GPT, while in the co-axial system with epithelium-intact trachea 5-HT produced biphasic response. This showed that alteration depends on guinea-pig tracheal epithelium. In the co-axial system, imipramine and pargyline caused a potentiation on the relaxations of 5-HT possibly by preventing uptake and degradation of 5-HT. In the co-axial system, results in the presence of INDO meant that 5-HT caused an inhibitory CO product and another inhibitory factor release from the guinea-pig tracheal epithelium. Another bronchoconstrictor agent which we have used, BK, produced dose-dependent relaxations on the RAM and these relaxations decreased significantly in the co-axial system with epithelium-intact 6PT. These relaxations decreased again, after epithelium removal. These results suggested that inhibitory factors released from the guinea-pig tracheal epithelium did not contribute to the relaxations or, on the contrary, this contribution masked by excitatory factors which released from epithelial or nonepithelial tissue by BK. In the co-axial system INDO and captopril did not affect the relaxations caused by BK significantly. This situation implied that nonprostanoid excitatory factors might reduce the relaxations and enzymes different from carboxypeptidase might responsible from degradation of BK. In the co-axial assemlies with epithelium-intact 6PT isolated from sensitized guinea-pigs, biphasic response which produced by 5-HT reversed completely to the contractions after in vitro OA challenge. In addition BK-induced relaxations increased significantly (especially at the low doses of BK) after in vitro OA administration. These results suggested that epithelial alterations (morphological and functional) due to OA challenge might contribute to these alterations. In the modified co-axial system which we have developed, Phe-induced contractions did not produce plateau and conversion if these contractions to the relaxations and also, reduction of the precentage of fade after epithelium removal, implied that basal release of inhibitory factor(s) might responsible from the fade of Phe-induced contractions.
