William Tucker’s passion for chemistry was inspired by his high school teacher Gary Osborn. He left Maine to pursue Chemistry at Middlebury College, and after graduating in 2010 he decided to pursue a PhD in Organic Chemistry at the University of Wisconsin-Madison. At the University of Wisconsin-Madison, he worked in the laboratory of Dr. Sandro Mecozzi, where he developed semifluorinated triphilic surfactants for hydrophobic drug delivery. After earning his PhD in 2015, he took a fellowship at Boston University as a Postdoctoral Faculty Fellow. There he co-taught organic chemistry while working in the laboratory of Dr. John Caradonna. In the Caradonna l boratory, he worked on developing a surface-immobilized iron-oxidation catalyst for the oxidation of C–H bonds using dioxygen from the air as the terminal oxidant. Throughout all of this work, his passion has always been for teaching and working with students both in and out of the classroom. He has been lucky for the past six years to work at Concord Academy, where his students have, through their questions, pushed him to think deeper and more critically about chemistry. Their curiosity inspires him, and their inquisitiveness inspired his writing.

Abstract

This open access book examines the implications of The Bell Curve for the social, economic, and political developments of the early 21st century. Following a review of the reception of The Bell Curve and its place in the campaign to end affirmative action, Professor Tucker analyses Herrnstein’s concept of the “meritocracy” in relation to earlier 20th century eugenics and the dramatic increase in economic inequality over the past 30 years. Tucker demonstrates how, contrary to The Bell Curve’s predictions, the reallocation of these huge sums was neither rational nor beneficial for society. The book moves on to situate The Bell Curve within contemporary politics and shows how it can be seen to have played a role in the 2016 US election. This compelling analysis will appeal to scholars and those with an interest in the history of scientific racism, the history of psychology and the sociology of knowledge and science. This is an open access book.

Abstract

Leber hereditary optic neuropathy (LHON) is an important example of mitochondrial blindness with the m.11778G>A mutation in the MT-ND4 gene being the most common disease-causing mtDNA variant worldwide. The REFLECT phase 3 pivotal study is a randomized, double-masked, placebo-controlled trial investigating the efficacy and safety of bilateral intravitreal injection of lenadogene nolparvovec in patients with a confirmed m.11778G>A mutation, using a recombinant adeno-associated virus vector 2, serotype 2 (rAAV2/2-ND4). The first-affected eye received gene therapy; the fellow (affected/not-yet-affected) eye was randomly injected with gene therapy or placebo. The primary end point was the difference in change from baseline of best-corrected visual acuity (BCVA) in second-affected/not-yet-affected eyes treated with lenadogene nolparvovec versus placebo at 1.5 years post-treatment, expressed in logarithm of the minimal angle of resolution (LogMAR). Forty-eight patients were treated bilaterally and 50 unilaterally. At 1.5 years, the change from baseline in BCVA was not statistically different between second-affected/not-yet-affected eyes receiving lenadogene nolparvovec and placebo (primary end point). A statistically significant improvement in BCVA was reported from baseline to 1.5 years in lenadogene nolparvovec-treated eyes: -0.23 LogMAR for the first-affected eyes of bilaterally treated patients (P < 0.01); and -0.15 LogMAR for second-affected/not-yet-affected eyes of bilaterally treated patients and the first-affected eyes of unilaterally treated patients (P < 0.05). The mean improvement in BCVA from nadir to 1.5 years was -0.38 (0.052) LogMAR and -0.33 (0.052) LogMAR in first-affected and second-affected/not-yet-affected eyes treated with lenadogene nolparvovec, respectively (bilateral treatment group). A mean improvement of -0.33 (0.051) LogMAR and -0.26 (0.051) LogMAR was observed in first-affected lenadogene nolparvovec-treated eyes and second-affected/not-yet-affected placebo-treated eyes, respectively (unilateral treatment group). The proportion of patients with one or both eyes on-chart at 1.5 years was 85.4% and 72.0% for bilaterally and unilaterally treated patients, respectively. The gene therapy was well tolerated, with no systemic issues. Intraocular inflammation, which was mostly mild and well controlled with topical corticosteroids, occurred in 70.7% of lenadogene nolparvovec-treated eyes versus 10.2% of placebo-treated eyes. Among eyes treated with lenadogene nolparvovec, there was no difference in the incidence of intraocular inflammation between bilaterally and unilaterally treated patients. Overall, the REFLECT trial demonstrated an improvement of BCVA in LHON eyes carrying the m.11778G>A mtDNA mutation treated with lenadogene nolparvovec or placebo to a degree not reported in natural history studies and supports an improved benefit/risk profile for bilateral injections of lenadogene nolparvovec relative to unilateral injections.

23. Advancing Personalized Learning through Effective Use and Protection of Student Data. Education Leaders Report. Volume 4, No. 2

Abstract

With roots in student-centered pedagogies that go back at least to the 1900s, personalized learning meets students' learning goals, needs, context, and pace while incorporating their interests and preferences. Personalized learning in today's classroom depends upon and creates an abundance of rich student data, which simultaneously fosters new opportunities for learning and creates fresh concerns over student data privacy. In this report, the authors argue that good student data privacy policies recognize the potential for personalized learning to accelerate student achievement while also guaranteeing safe, secure access to a predetermined, transparent set of student data. To advance personalized learning, state policymakers need to develop laws and policies that avoid key pitfalls that can hamper the efficient, effective use of data by school leaders, teachers, parents, and students. The report recommends actions state boards can take to foster personalized learning and data protection.

Abstract

In this work, we examine metal electrode-ionomer electrolyte systems at high voltage / negative surface charge and at high pH to assess factors that influence hydrogen production efficiency. We simulate the hydrogen evolution electrode interface investigated experimentally in Bates et al., Journal of Physical Chemistry C, 2015 using a combination of first principles calculations and classical molecular dynamics. With this detailed molecular information, we explore the hypotheses posed in Bates et al. In particular we examine the response of the system to increased bias voltage and oxide coverage in terms of the potential profile, changes in solvation and species concentrations away from the electrode, surface concentrations, and orientation of water at reactive surface sites. 25. Dissipation and plastic deformation in collisions between metallic nanoparticles

Abstract

Collisions between amorphous Fe nanoparticles were studied using molecular-dynamics simulation. For head-on collisions of nanoparticles with radii $R =$ 1.4 nm, $R =$ 5.2 nm, and $R =$ 11 nm, sticking was observed at all simulated velocities. The results were compared to the description provided by the JKR model. It was found that strong disagreement exists between the predictions of JKR and the results of the molecular-dynamics simulation due to the presence of additional dissipative processes which strengthen sticking behavior. First, it is demonstrated that very strong dissipation into atomic vibrations occurs during the collision. The dissipation is strong enough to prevent significant rebound of the nanoparticles. Additionally, the morphology of the adhered nanoparticles includes a ``neck'' that increases in radius with increasing collision velocity which results in amplified irreversibility and adhesion. Approximate calculation of the stress during the collision indicates that stress levels are well above typical yield stress values even for low velocity collisions, consistent with the observation of plastic deformation. Furthermore, it is shown that for nanoparticles with $R \leq$ 11 nm, the dominance of surface attraction results in large effective collision velocities and plastic deformation. By obtaining scaling relations for computed quantities, predictions are made for larger nanoparticles up to $R$ $\sim$ 1 $\mu$m. 29. Is subsurface soil a chemically more hostile zone for deep‐placed phosphorus fertilizer?

Abstract

The susceptibility of surface soil to drying when rainfall is scarce has prompted evaluation of deeper placement (20 cm) of fertilizers within the profile to where soil moisture is more likely to be stored. However, concerns arise regarding potential chemical challenges to P availability in the subsurface. This study examined differences in chemical conditions between the soil surface (0–10 cm) and subsurface (15–25 cm), and investigated the effect on the diffusion and extractability of banded P. Analysis of surface and subsurface samples from 15 sites revealed substantial differences in characteristics that influence P behavior between depths. The phosphorus buffering index (PBI) was largely explained by concentrations of amorphous Al and Fe (hydr)oxides and CaCO3. The measured diffusion distance of P from monoammonium phosphate (MAP) granules was found to be inversely related to PBI. Consequently, significant differences in diffusion distances were typically observed between soil layers characterized by marked PBI variations. The measured diffusion radii were used to calculate the mean P concentrations of surface‐banded and deep‐banded MAP in a field scenario, which were replicated in a 4‐week laboratory incubation. The resulting lability of applied P was determined through isotopic exchange, and although significantly higher P recoveries were measured from the subsurface at two sites where large pH differences existed between depths, no significant differences were measured between depths at all other sites. Abstract

Objective REFLECT is the first randomised, double-masked, placebo-controlled multicentre phase 3 clinical trial that evaluated the efficacy and safety of bilateral intravitreal (IVT) injection of lenadogene nolparvovec in subjects with Leber hereditary optic neuropathy carrying the m.11778G>A mutation.Methods and analysis A total of 98 subjects were enrolled with vision loss of ≤12 months. The subjects were randomised to one of two treatment arms with all subjects receiving an intravitreal (IVT) injection of lenadogene nolparvovec in their first affected eye and the second-affected eye randomised to receive IVT of either lenadogene nolparvovec or placebo.Results The majority of subjects were male with a mean duration of vision loss of 8.3 months. All but one subject experienced bilateral loss of vision at the time of injection. The mean best-corrected visual acuity of first-affected eyes was worse compared with second/not-yet-affected eyes. Analysis of retinal anatomical parameters showed increased thinning in the first-affected eyes when compared with the second/not-yet-affected eyes with both treatment arms showing significant changes compared with unaffected individuals.Conclusion The REFLECT trial is the third and the largest phase 3 clinical study evaluating lenadogene nolparvovec in m.11778G>A Leber hereditary optic neuropathy (LHON) subjects. The observed demographics in REFLECT are consistent with previous reports in LHON subjects in the acute and dynamic phases of LHON disease. Combined with the visual function and anatomical parameters obtained in the previous RESCUE and REVERSE trials, REFLECT has provided a uniformly collected data set that should help direct future LHON clinical trials.

35. Ineluctable modality of the other : the ethical excess in the selected works of William Gaddis, Thomas Pynchon, and David Foster Wallace

Abstract

In his seminal study of postmodern literature, Brian McHale contends that the primary concerns of postmodern fiction tend to be ontologically, rather than epistemologically, dominant. This thesis will challenge McHale's claim by analyzing how three American post-war novelists self-reflexively adopt and subvert various epistemological approaches to understanding the world in order to reveal the pitfalls associated with the reductive impulse to categorize disparate data and make it present-at-hand within epistemic frameworks. The ultimate aim of these efforts is to highlight an ineffable alterity-the ethical excess of the Other-that eludes codification. In doing so these authors indicate how ontological questions and their implications are predicated on the question of the Other, which is not primarily an ontological query but equally-or even more so-an epistemological one. The first chapter will explore how William Gaddis was one of the earliest post-war American authors to negotiate epistemic closure in favor of an alterity that cannot be totalized within his proto-postmodern novel, The Recognitions (1955). Through his aesthetic vision espousing a return to the primordial first idea as well as an ethics of indeterminacy in order to foster an agape with the unthematizable Other, Gaddis promotes the ethical imperative of the "self-who-can-do-more" who consequently attempts to "make negative things do the work of positive ones" by maintaining a responsibility to the ineffable Other in a modern world devoid of absolutes. Gaddis's quixotic attempt to redeem a culturally-vacuous post-war society that is ultimately more ignorant than malicious will then transition to the second chapter exploring Thomas Pynchon's more cynical view in Gravity's Rainbow (1973) of a world dominated by malicious epistemic systems of control where active resistance against these systems can become totalizing acts themselves. Pynchon therefore attempts to combat determinate modes of thought through his notion of "illogical negativism" designed to maintain a non-committal, liminal position in order to remain truly open to an "Other Order of Being" by avoiding rationalizing the problem of alterity altogether. The final chapter will analyze how David Foster Wallace's exploration of a "cohesion-renewing Other" in Infinite Jest (1996) attempts to renew cohesion between the different positions previously adopted by Gaddis and Pynchon. Conscious of both the need to adopt an ethical imperative for the Other while also being mindful of the necessity of having to engage with totalizing epistemic frameworks at the expense of alterity in order to convey meaning, Wallace adopts a neopragmatic approach that maintains a faith in language but nevertheless repudiates epistemic foundationalism and acknowledges the radical contingency of language and selfhood as communal constructs dependent on the Other.
