13. A latent profile analysis of shift work tolerance among Chinese male sailors during a prolonged non-24-h rotating shift schedule at sea.

Abstract

Shift work tolerance (SWT) refers to the ability to adapt to shift work without significant adverse consequences. The present study aimed to examine the individual differences in SWT and their predictors and outcomes. Latent profile analyses were conducted using cross-sectional data collected form 448 Chinese male sailors who experienced a prolonged (>30 d) non-24-h rotating shift schedule at sea. Depression, anxiety, sleep disturbance, fatigue, domestic disruption, job satisfaction, work engagement, digestive and cardiovascular symptoms were included as indicators of SWT. The results showed that there existed 2 latent profiles of SWT named as High SWT group and Low SWT group. High SWT group was characterized by low levels on all negative bio-psycho-social outcomes but high levels of work engagement and job satisfaction, while Low SWT group exhibited completely opposite characteristics compared to High SWT group. The level of hardiness could predict profile membership that those with higher level of hardiness were more likely to belong to High SWT group. However, there were no significant differences observed in job performance between two groups. In conclusion, hardiness can serve as a predictor of personnel selection for shift work and hardiness-based intervention programs should be encouraged among the shift workers. [ABSTRACT FROM AUTHOR]

Abstract

White bamboo is the swollen stem of Zizania latifolia parasitized by the smut fungus Ustilago esculenta. Five samples of galls were harvested at different stages of swelling, along with young seedlings of Z. latifolia and isolated colonies of U. esculenta. The inhibition capacity of boiling water or 50% ethanol extracts on NO release by RAW264 cells, which was stimulated with lipopolysaccharide, was evaluated as anti-inflammatory activity, while the NO induction capacity was evaluated as immunostimulatory activity. Total polyphenol (TPP) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity were also measured. The anti-inflammatory effect, as well as TPP and DPPH, was comprehensively detected in the ethanol extracts. The immunostimulating activity was observed in the boiled extracts at different levels, depending on the swelling stage, and was especially high in the top (apical) part. These data may indicate that functional components could be dynamically induced by interactions between Z. latifolia and U. esculenta.

30. The Mechanism Underlying of Long-Term Stable Indigo Reduction State in Indigo Fermentation Using Sukumo (Composted Polygonum tinctorium Leaves)

Abstract

Indigo fermentation fluid maintains its indigo-reducing state for more than 6 months under open-air. To elucidate the mechanism underlying the sustainability of this indigo reduction state, three indigo fermentation batches with different durations for the indigo reduction state were compared. The three examined batches exhibited different microbiota and consisted of two phases. In the initial phase, oxygen-metabolizing-bacteria derived from sukumo established an initial network. With decreasing redox potential (ORP), the initial bacterial community was replaced by obligate anaerobes (mainly Proteinivoraceae; phase 1). Approximately 1 month after the beginning of fermentation, the predominating obligate anaerobes were decreased, and Amphibacillus and Polygonibacillus, which can decompose macromolecules derived from wheat bran, were predominantly observed, and the transition of microbiota became slow (phase 2). Considering the substrate utilization ability of the dominated bacterial taxa, the transitional change from phase 1 to phase 2 suggests that this changed from the bacterial flora that utilizes substrates derived from sukumo, including intrinsic substrates in sukumo and weakened or dead bacterial cells derived from early events (heat and alkaline treatment and reduction of ORP) to that of wheat bran-utilizers. This succession was directly related to the change in the major substrate sustaining the corresponding community and the turning point was approximately 1 month after the start of fermentation. As a result, we understand that the role of sukumo includes changes in the microbial flora immediately after the start of fermentation, which has an important function in the start-up phase of fermentation, whereas the ecosystem comprised of the microbiota utilizing wheat bran underpins the subsequent long-term indigo reduction.

36. Microbial Diversity in the Edible Gall on White Bamboo Formed by the Interaction between Ustilago esculenta and Zizania latifolia

Abstract

type:Article, An edible gall is formed between the third and fourth nodes beneath the apical meristem near the base of Zizania latifolia shoots. This gall is harbored by and interacts with the smut fungus Ustilago esculenta. The gall is also a valuable vegetable called “white bamboo,” jiaobai or gausun in China and makomotake in Japan. Five samples of the galls harvested at different stages of swelling were used to isolate microorganisms by culturing. Isolated fungal and bacterial colonies were identified by DNA sequencing and matrix-assisted laser desorption/ionization–time-of-flight mass spectrometry, respectively. Several strains of U. esculenta as well as 6 other species of fungi and 10 species of bacteria were isolated. The microbiome was also evaluated by simple and outlined DNA profiling with automated rRNA intergenic spacer analysis (ARISA), and the amount of DNA of U. esculenta was determined by qPCR. At least 16 species of fungi and 40 species of bacteria were confirmed by ARISA of the overall sample. Interestingly, the greatest bacterial diversity, i.e., 18 species, was observed in the most mature sample, whereas the fungal diversity observed in this sample, i.e., 4 species, was rather poor. Based on qPCR, U. esculenta occurred in samples from all stages; however, the abundance of U. esculenta exhibited unique U-shaped relationships with growth. These results may explain why the interaction between U. esculenta and Z. latifolia also influences the unique microbial diversity observed throughout the growth stages of the swollen shoot, although the limited sample size does not allow conclusive findings.

46. Identification of Mushroom Species by Automated rRNA Intergenic Spacer Analysis (ARISA) and Its Application to a Suspected Case of Food Poisoning with Tricholoma ustale.

Abstract

Automated rRNA intergenic spacer analysis (ARISA), a method of microbiome analysis, was evaluated for species identification of mushrooms based on the specific fragment sizes. We used 51 wild mushroom-fruiting bodies collected in the centre of Hokkaido and two cultivated mushrooms. Samples were hot-air-dried and DNA were extracted by a beads beating procedure. Sequencing analysis of portions of the rRNA gene (rDNA) provided 33 identifications of mushrooms by genus or species. The results of ARISA identification based on the combination of the fragment sizes corresponding to two inter spacer regions (ITS2 and ITS1) of rDNA within±0.1% accuracy showed that 27 out of the 33 species had specific fragment sizes differentiated from other species. The remaining 6 species formed 3 pairs that showed overlapping fragment sizes. In addition, within-species polymorphisms were observed as 1 bp differences among 32 samples of 13 species. ARISA was applied to investigate a case of suspected food poisoning in which the mushroom was thought to be a toxic Kakishimeji. The morphological identification of the mushroom was ambiguous since the remaining sample lacked a part of the fruiting body. Further, yeast colonies had grown on the surface of the fruiting body during storage. The ARISA fragment size of the mushroom showed 7 bp difference from that of the candidate toxic mushroom. Although ARISA could be a useful tools for estimation of mushroom species, especially in case where the fruiting bodies have deteriorated or been processed, further studies are necessary for reliable identification. For example, it may be necessary to adopt more informative genes which could provide clearer species-specific polymorphisms than the ITS regions.
