The budding yeastSaccharomyces cerevisiaeis commonly used as an expression platform to produce valuable compounds. Yeast-based genetics research can uniquely utilize use auxotrophy in transformant selection: auxotrophic complementation by an auxotrophic marker gene on exogenous DNA (such as plasmids). However, the number of auxotrophic nutrients required restricts the number of plasmids maintained by the cells. We therefore developed novel Δ10 strains that are auxotrophic for 10 different nutrients, and new plasmids with two multicloning sites and auxotrophic markers for use in Δ10 strains. We confirmed that Δ10 strains could maintain 10 types of these plasmids. Because each plasmid can express two different genes, this Δ10 strain-based expression system has the potential to co-express a maximum of 20 different proteins.

Background Elective orthopedic surgery, as well as the procedures of many surgical departments, have been severely curtailed by coronavirus disease 2019. Here, we aimed to analyze how all surgeons could safely perform essential procedures during the coronavirus disease 2019 pandemic. Methods A retrospective review of elective surgeries performed between May 15, 2020, and February 28, 2022, was conducted. A screening questionnaire was used, and reverse transcription-polymerase chain reaction testing was assessed in all admitted surgical department patients. Their positivity rate and the positivity rate in our fever outpatient clinic were analyzed. Results Of 6099 patients who tested for severe acute respiratory syndrome coronavirus-2 during the study period, eight (0.13%) tested positive. The positive results were seen in four patients undergoing orthopedic surgery, two undergoing respiratory surgery, one undergoing breast surgery, and one undergoing plastic surgery. The number of patients who visited the outpatient clinic for fever was 15,639, including 1640 positive cases (positive rate of 10.5%). The positive rate of preoperative reverse transcription-polymerase chain reaction testing for scheduled surgery was consistently low and did not coincide with the peak of the wave of infection, while the positivity rate of outpatients with fever demonstrated a wave consistent with the national infection situation. All 6091 patients, excluding the eight positive patients, underwent surgery; all patients who underwent surgery were discharged from the hospital without developing coronavirus disease 2019 symptoms. Conclusions Our findings suggest that the establishment of a universal reverse transcription-polymerase chain reaction testing system is essential for the safe performance of scheduled surgeries during the coronavirus disease 2019 pandemic.

This study aimed to investigate factors affecting discharge to an inpatient rehabilitation facility or home following total hip arthroplasty, using a clinical pathway in Japan.Five hundred hips with osteoarthritis who underwent unilateral total hip arthroplasty at our institution, with no deviation from the pathway, were included in this retrospective study. The variables were examined by univariate analysis. Multivariate logistic regression analysis was used to identify the independent factors that influenced the discharge outcome.Four hundred and thirty-four hips were discharged home directly, and 66 were discharged to an inpatient rehabilitation facility. Patients discharged to an inpatient rehabilitation facility were significantly older, shorter, lighter, and more likely to live alone. Additionally, the preoperative clinical score was significantly lower in the inpatient rehabilitation facility Group for all items. Logistic regression analysis showed a significant association between being discharged to an inpatient rehabilitation facility and higher age [odds ratio 3.87, 95% confidence interval 2.03-7.38, P 0.001], lower total score in the preoperative Japanese Orthopaedic Association hip score [odds ratio 2.42, 95% confidence interval 1.38-4.23, P = 0.002] and living alone [odds ratio 1.84, 95% confidence interval 1.01-3.35, P = 0.046].In this study, age, the preoperative Japanese Orthopaedic Association hip score, and living arrangement impacted the discharge destination after THA.

Although atelocollagen gel is used as a scaffold for culturing human articular cartilage-derived chondrocytes, little is known about cell–gel interactions. In this study, we investigated the mechanism via which atelocollagen gel affects human articular cartilage-derived chondrocytes. Two types of three-dimensional cultures of human articular cartilage-derived chondrocytes (i.e., with and without atelocollagen gel) were compared. While the amount of atelocollagen gel in culture gradually decreased with time, it promoted the expression of matrix metalloproteinases (MMPs) during the early stages of culture. Genome-wide differential gene expression analysis revealed that cell membrane- and extracellular matrix-related genes were highly ranked among up- and down-regulated groups in cells cultured in the presence of atelocollagen gel. Among the integrin family of genes, the expression of integrin subunit alpha 2 and integrin subunit alpha 10 was significantly increased in the presence of atelocollagen gel. Blocking α2β1 integrin with the specific inhibitor BTT 3033 had a significant effect on cell proliferation, MMP expression, and cell shape, as well as on the response to mechanical stimulation. Taken together, our findings indicate that the α2β1 integrin pathway plays an important role in the interaction of atelocollagen gel with human articular cartilage-derived chondrocytes and may be a potential therapeutic target for articular cartilage disorders.

Reelin is an extracellular matrix protein that is mainly produced in Cajal-Retzius cells and controls neuronal migration, which is important for the proper formation of cortical layers in the developmental stage of the brain. In the adult brain, Reelin plays a crucial role in the regulation of N-methyl-D-aspartate receptor-dependent synaptic function, and its expression decreases postnatally. Clinical studies showed reductions in Reelin protein and mRNA expression levels in patients with psychiatric disorders; however, the causal relationship remains unclear. Reelin-deficient mice exhibit an abnormal neuronal morphology and behavior, while Reelin supplementation ameliorates learning deficits, synaptic dysfunctions, and spine loss in animal models with Reelin deficiency. These findings suggest that the neuronal deficits and brain dysfunctions associated with the down-regulated expression of Reelin are attenuated by enhancements in its expression and functions in the brain. In this review, we summarize findings on the role of Reelin in neuropsychiatric disorders and discuss potential therapeutic approaches for neuropsychiatric disorders associated with Reelin dysfunctions.

Alzheimer’s disease (AD) is an age-related and progressive neurodegenerative disorder. It is widely accepted that AD is mainly caused by the accumulation of extracellular amyloid β (Aβ) and intracellular neurofibrillary tau tangles. Aβ begins to accumulate years before the onset of cognitive impairment, suggesting that the benefit of currently available interventions would be greater if they were initiated in the early phases of AD. To understand the mechanisms of AD pathogenesis, various transgenic mouse models with an accelerated accumulation of Aβ and tau tangles have been developed. However, none of these models exhibit all pathologies present in human AD. To overcome these undesirable phenotypes, APP knock-in mice, which were presented with touchscreen-based tasks, were developed to better evaluate the efficacy of candidate therapeutics in mouse models of early-stage AD. This review assesses several AD mouse models from the aspect of biomarkers and cognitive impairment and discusses their potential as tools to provide novel AD therapeutic approaches.

Purpose The aim of this study was to evaluate the outcomes and early complications of obese patients who underwent total hip arthroplasty for osteoarthritis via an anterolateral approach in the supine position (ALS-THA) and compare these outcome with of a matched control group of non-obese patients. Patients and methods Thirty-one hips in 28 patients with obesity (BMI ≧ 30 kg/m2) were included in this study. As a control group, 31 hips of 31 patients with a normal weight (BMI between 20 and 25 kg/m2) were matched based on age, sex, and laterality. Clinical evaluations using the Merle d’Aubigne and Postel hip score, radiological evaluations and perioperative complications were compared in two groups. Results There were no significant differences between the groups in the operative time, period of hospitalization, clinical hip score, or cup positioning, although the position of the cup tended to deviate from the optimal safe zone in the obese compared with non-obese group (32.3 and 16.1%, respectively). There was no infection, dislocation, nerve palsy, or life-threatening event in either group. The rate of avulsion fractures of the greater trochanter in the obese group was 3 times higher compared to that in the non-obese group. Conclusions As the clinical outcome of ALS-THA for the obese group is not inferior to that for the non-obese group, obesity is not considered to be a contraindication for ALS-THA. However, obesity increases the risk of intraoperative greater trochanteric fracture. Thus, surgeons should be particularly careful when manipulating the femur in this class of patients, who should be informed of this risk.

S-allylcysteine (SAC), a major thioallyl compound contained in mature garlic extract (MGE), is known to be a neuroactive compound. This study was designed to investigate the effects of SAC on primary cultured hippocampal neurons and cognitively impaired senescence-accelerated mice prone 10 (SAMP10). Treatment of these neurons with MGE or SAC significantly increased the total neurite length and number of dendrites. SAMP10 mice fed MGE or SAC showed a significant improvement in memory dysfunction in pharmacological behavioral analyses. The expression of &alpha, amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor, N-methyl-d-aspartate (NMDA) receptor, and phosphorylated &alpha, calcium/calmodulin-dependent protein kinase II (CaMKII) in the hippocampal tissue of SAMP10 mice fed MGE or SAC was significantly suppressed, especially in the MGE-fed group. These findings suggest that SAC positively contributes to learning and memory formation, having a beneficial effect on brain function. In addition, multiple components (aside from SAC) contained in MGE could be useful for improving cognitive function by acting as neurotrophic factors.

Disrupted-in-schizophrenia-1 (DISC1) has been widely associated with several psychiatric disorders, including schizophrenia, mood disorders and autism. We previously reported that a deficiency of DISC1 may induce low anxiety and/or high impulsivity in mice with disruption of exons 2 and 3 of the Disc1 gene (Disc1(Δ2-3/Δ2-3)). It remains unclear, however, if deficiency of DISC1 leads to specific alterations in distinct neuronal systems. In the present study, to understand the role of DISC1 in γ-aminobutyric acid (GABA) interneurons and mesocorticolimbic dopaminergic (DAergic) neurons, we investigated the number of parvalbumin (PV)-positive interneurons, methamphetamine (METH)-induced DA release and the expression levels of GABAA, DA transporter (DAT) and DA receptors in wild-type (Disc1(+/+)) and Disc1(Δ2-3/Δ2-3) mice. Female Disc1(Δ2-3/Δ2-3) mice showed a significant reduction of PV-positive interneurons in the hippocampus, while no apparent changes were observed in mRNA expression levels of GABAA receptor subunits. METH-induced DA release was significantly potentiated in the nucleus accumbens (NAc) of female Disc1(Δ2-3/Δ2-3) mice, although there were no significant differences in the expression levels of DAT. Furthermore, the expression levels of DA receptor mRNA were upregulated in the NAc of female Disc1(Δ2-3/Δ2-3) mice. Male Disc1(Δ2-3/Δ2-3) mice showed no apparent differences in all experiments. DISC1 may play a critical role in gender-specific developmental alteration in GABAergic inhibitory interneurons and DAergic neurons.

Increasing epidemiological evidence indicates that prenatal infection and childhood central nervous system infection with various viral pathogens enhance the risk for several neuropsychiatric disorders. Polyriboinosinic-polyribocytidilic acid (polyI:C) is known to induce strong innate immune responses that mimic immune activation by viral infections. Our previous findings suggested that activation of the innate immune system in astrocytes results in impairments of neurite outgrowth and spine formation, which lead to behavioral abnormalities in adulthood. To identify candidates of astrocyte-derived humoral factors that affect neuronal development, we analyzed astrocyte-conditioned medium (ACM) from murine astrocyte cultures treated with polyI:C (polyI:C-ACM) by two-dimensional fluorescence difference gel electrophoresis (2D-DIGE). Through a quantitative proteomic screen, we found that 13 protein spots were differentially expressed compared with ACM from vehicle-treated astrocytes (control-ACM), and characterized one of the candidates, matrix metalloproteinase-3 (Mmp3). PolyI:C treatment significantly increased the expression levels of Mmp3 mRNA and protein in astrocytes, but not microglia. PolyI:C-ACM was associated with significantly higher Mmp3 protein level and enzyme activity than control-ACM. The addition of recombinant Mmp3 into control-ACM impaired dendritic elongation of primary cultured hippocampal neurons, while the deleterious effect of polyI:C-ACM on neurite elongation was attenuated by knockdown of Mmp3 in astrocytes. These results suggest that Mmp3 is a possible mediator of polyI:C-ACM-induced neurodevelopmental impairment.

The lateral and anterior lateral approaches are the most commonly used for antegrade nailing of humerus fractures. However, the surgical exposure is restricted by the bony acromion. The iatrogenic injury to the rotator cuff can also cause post-operative pain and compromise shoulder function. This article describes a new rotator interval approach that we used for central entry point nailing. In this approach, the skin incision starts from the midpoint between the acromion and coracoid process. A trans-rotator interval split in front of the anterior border of the supraspinatus (SSP) tendon is then made to open the glenohumeral joint. With the SSP retracted laterally and the long head of the biceps (LHB) retracted medially, the humeral head is directly visualized. The entry point can thus be determined and confirmed by intra-operative fluoroscopy in both axial and AP planes. We recommend this rotator interval approach as an alternative nailing technique for 2-part humeral neck fractures and humeral shaft fractures.

Background: In elderly patients with femoral neck fracture, functional recovery is affected largely by preoperative underlying diseases and postoperative complications. The present study was designed to analyze the relationship between preoperative underlying diseases and perioperative complications of surgery for proximal femur fracture. Methods: The study involved 86 patients aged 75 years or older, who underwent surgery for proximal femur fracture. Each patient was examined for the presence of preoperative underlying diseases and preoperative biochemical test data. Moderate and severe complications, which were life-threatening and necessitated treated, were investigated during the perioperative period. A retrospective analysis was conducted on the relationship of preoperative underlying diseases and preoperative biochemical data with the moderate or severe complications that developed during the perioperative period. Results: Eighty-one patients developed a preoperative underlying disease, with hypertension being most frequent (42 cases). Complications during the perioperative period were observed in 58 patients, with anemia develops most frequently (23 cases). Moderate or severe complications during the perioperative period were observed in 16 patients, and infection was the most frequent complication (8 cases). In the statistical analysis, a significant difference in the incidence of moderate or severe perioperative complications was observed only in the users of anticoagulants/antiplatelet drugs. Discussion: Patients of preoperative use of anticoagulants/antiplatelet drugs should be closely managed medically so that perioperative medical complications can be managed.

Polyglutamine tract-binding protein 1 (PQBP1) is an intrinsically disordered protein composed of a small folded WW domain and a long disordered region. PQBP1 binds to spliceosomal proteins WBP11 and U5-15kD through its N-terminal WW domain and C-terminal region, respectively. Here, we reveal that the binding between PQBP1 and WBP11 reduces the binding affinity between PQBP1 and U5-15kD. Our results suggest that the interaction between PQBP1 and WBP11 negatively modulates the U5-15kD binding of PQBP1 by an allosteric mechanism.

Study Objective To investigate the use of caudal epidural anesthesia for postoperative pain after total hip arthroplasty. Design Prospective study. Setting University-affiliated hospital. Patients 32 (4 men and 28 women) patients, aged 49 to 89 years, scheduled for total hip arthroplasty for osteoarthritis of the hip. Interventions and Measurements Patients were allocated to three groups: lumbar epidural anesthesia (EA group; n = 16) or caudal epidural anesthesia (CA group; n = 16) groups, which were case-matched according to patient demographics. Nine patients received general anesthesia only (GA group). We evaluated the level of postoperative pain using a 100-mm Visual Analog Scale (VAS) recorded at 3, 6, 9, 12, and 24 hours after surgery. Main Results Total requirement of diclofenac sodium suppositories was significantly larger in the GA group than in the EA or CA groups (444 ± 302 vs 188 ± 124 and 145 ± 130 mg). The number of days requiring analgesics was significantly prolonged in the GA group compared with the EA or CA groups (14 ± 9 vs 4 ± 3 and 4 ± 4 days). These items were similar between the EA group and the CA group. All VAS values for pain, rest, and movement in the postsurgical period over 24 hours were significantly higher in the GA group than in either the EA or CA groups. Conclusions Caudal epidural anesthesia provides effective postoperative analgesia similar to lumbar epidural anesthesia.

BACKGROUND Useful markers of bone resorption are needed for hemodialysis (HD) patients with renal osteodystrophy. The present study investigated the use of a new immunoassay for the crosslinked N-terminal telopeptide of type 1 collagen to assess bone loss in HD patients. METHODS Forty-seven male and 19 female patients were enrolled in this study. Their mean age was 64 years, and the mean duration they had been receiving HD was 110 months at the time of investigation. Their mean body mass index (BMI) was 21 kg/m2. Serum levels of N-terminal telopeptide of type 1 collagen (NTx) and of total calcium (Ca), phosphorus (P), alkaline phosphatase (Alp), and intact parathyroid hormone (PTH) were measured. Bone mineral density (BMD) of the distal one-third of the radius was examined by dual-energy x-ray absorptiometry. RESULTS Serum levels of NTx, Ca, P, Alp, and intact PTH were 68.9 nmol BCE/L, 4.5 mEq/L, 6.3 mg/dL, 248 IU/L, and 241 pg/mL, respectively. BMD was positively correlated with BMI (r = 0.24, p

The relationship between perioperative blood loss and intraosseous pressure of the ilium during cementless total hip arthroplasty has not previously been clarified. The aim of the present study was to evaluate whether blood loss in cementless total hip arthroplasty correlates with intraosseous pressure of the ilium or systemic arterial pressure. The subjects were 26 patients (5 men and 21 women) undergoing primary cementless total hip arthroplasty. Intraoperative and postoperative blood loss, intraosseous pressure of the ilium, and systemic arterial pressure were measured prospectively. Mean age of the subjects was 68.3 years. Intraoperative blood loss correlated significantly with mean arterial pressure (r=0.522, p=0.0079), but not with intraosseous pressure, age, or body weight. Postoperative blood loss correlated significantly with intraosseous pressure (r=0.399, p=0.0427), but not with mean arterial pressure, age, or body weight. Intraosseous pressure did not correlate with mean arterial pressure. These results suggest that intraosseous pressure of the ilium is a predictor of postoperative bleeding in primary cementless total hip arthroplasty.

We report a 14-year-old boy in whom isolated nonunion fracture of the posterior process of the talus. He underwent surgical repair with Herbert Whipple screw fixation and plaster immobilization. Osseous union was achieved 3 months after surgery, resulting in the resolution of symptoms and complete functional recovery. To our knowledge, the present report is the first to describe a successful outcome for surgical treatment of painful nonunion of fracture of the entire posterior process of the talus.

Measures for pain management after total hip arthroplasty (THA) are important for early improvement in the quality of life after operation and early postoperative rehabilitation. We investigated the analgesic effect of locally injected drugs around the total hip prosthesis.54 patients undergoing THA were randomized either to receive a periarticular intraoperative injection of a 30-ml mixture containing morphine hydrochloride 10 mg, 0.5% bupivacaine 20 ml, epinephrine 0.3 mg, and saline 8.7 ml or to receive no injection. The perioperative analgesic regimen was standardized. The evaluation items included assessment of pain using a 100-point visual analog scale (VAS) after the patients awoke on the day of the operation and on postoperative day 1, the dose of diclofenac sodium suppository, the number of days for acquiring assisted ambulation with a walking cane, and side effects.The VAS score on the day of the operation was significantly low in the injection group. No cardiac or central nervous system toxicity was observed.Intraoperative periarticular injection with multimodal drugs can significantly reduce pain on the day of the operation, with no apparent risks, following THA.

Summary The apparent reaction rate constants (k) of the reaction that generates reducing sugar (presented as mg maltose equivalents) from raw sweet potato starch by using β-amylase were determined by varying enzyme concentration, starch concentration and pH. The Arrhenius plot of the k-value reached a peak at approximately 86 °C; the activation energy and frequency factor were also determined. Moreover, the amount of reducing sugar produced in sweet potato heated by infrared irradiation was estimated by using the values of k and the kinetic parameters. The calculated amounts of reducing sugar produced generally agreed with the experimental values. These results indicate that the amount of reducing sugar produced in heat-treated sweet potato can be predicted by the methodology described in this study.

Background: Fractures of the hip are the cause of hospitalization in the geriatric population. In many cases, hip fractures are a sentinel event signaling a systemic decline in the patient’s health. Methods: A retrospective review of the cases of 88 patients aged 75 years or older with proximal femoral fracture was performed. Fracture types, surgical procedures, walking ability before the injury and at discharge, and waiting time from injury to surgery were obtained. Results: No significant difference was observed in walking ability before injury among patients with femoral neck fracture treated with bipolar hip arthroplasty (BHA) or open reduction and internal fixation (ORIF) and those with trochanteric fracture. The degree of deterioration of walking ability was 0.6 in patients treated with BHA, 1.2 in those with neck fracture treated with ORIF, and 1.1 in those with trochanteric fracture. Deterioration was significantly less severe in patients who received BHA, compared with those who received ORIF. Conclusions: Among the patients aged 75 years or older with femoral neck fracture, those who received BHA had a shorter time interval to the start of walking with parallel assist bars or with walking aids, and also had a higher rate of regaining walking ability.

Objective Rapidly destructive osteoarthritis (OA) of the hip is an uncommon subset of OA that affects mainly elderly women. Previous studies indicate that elevated levels of matrix metalloproteinases (MMPs) are produced within the tissue of patients with the condition. In the present study, we sought to determine whether serum and plasma levels of MMPs and tissue inhibitors of metalloproteinases (TIMPs) are also elevated. Methods Blood samples were obtained from 16 patients with rapidly destructive hip OA and from 20 patients with OA before total hip arthroplasty was performed. Synovial specimens were obtained during surgery. Synovial fibroblasts that had migrated sufficiently from explants were subcultured in vitro for 72 hours after confluency, and harvested supernatants were collected. Blood, tissue samples, and fibroblasts were assayed for MMPs 1, 2, 3, and 9, and TIMPs 1 and 2 by sandwich enzyme immunoassay. Results In blood samples, the levels of MMP-3 and MMP-9 in the group with rapidly destructive hip OA were significantly higher than the normal range and were also significantly higher than those in the OA group. In tissue samples, the levels of MMP-1, MMP-3, MMP-9, and TIMP-1 in the group with rapidly destructive hip OA were significantly higher than those in the OA group. Conclusion The results of this study show that serum and plasma levels of MMP-3 and MMP-9 are significantly increased in patients with rapidly destructive hip OA. Significantly large amounts of these MMPs produced in synovial tissues within the hip joint could contribute in part to elevation of blood levels. Detection of increased levels of MMP-3 and MMP-9 in patients with painful, disabling hip OA may be of diagnostic value for rapidly destructive hip OA.

Synaptic plasticity in hippocampal neurons has been thought to represent a variety of memories. Although accumulating evidence indicates a crucial role of BDNF/TrkB/Akt signaling in the synaptic plasticity of the hippocampus, the mechanism by which Akt, a serine/threonine kinase, controls activity-dependent neuronal plasticity remains unclear. Girdin (also known as APE, GIV, and HkRP1), an actin-binding protein involved both in the remodeling of the actin cytoskeleton and in cell migration, has been identified as a substrate of Akt. Previous studies have demonstrated that deficit of neuronal migration in the hippocampus ofGirdin-deficient (Girdin−/−) mice is independent on serine phosphorylation of Girdin at S1416 (Girdin S1416) by Akt. In the present study, we focused on the role of Girdin S1416 phosphorylation in BDNF/TrkB/Akt signaling associated with synaptic plasticity. We found that Girdin in the hippocampus was phosphorylated at S1416 in an activity-dependent manner. Phosphorylation-deficient knock-in mice (GirdinSA/SAmice), in which S1416 is replaced with alanine, exhibited shrinkage of spines, deficit of hippocampal long-term potentiation, and memory impairment. These phenotypes ofGirdinSA/SAmice resembled those ofGirdin+/−mice, which have 50% loss of Girdin expression. Furthermore, Girdin interacted with Src kinase and NR2B subunit of NMDA receptor, leading to phosphorylation of the NR2B subunit and NMDA receptor activation. Our findings suggest that Girdin has two different functions in the hippocampus: Akt-independent neuronal migration and Akt-dependent NR2B phosphorylation through the interaction with Src, which is associated with synaptic plasticity in the hippocampus underlying memory formation.

For total hip arthroplasty (THA), minimally invasive surgery (MIS) has been developed to reduce incision length, muscle damage, and a shorter hospital stay. However, reduced exposure of anatomical landmarks may result in technical errors and inferior implant survivorships. The aim of this study was to report the short-term results and clinical complications of primary MIS THA in the supine position.A consecutive series of 103 patients who underwent MIS cementless THA with a modified Watson-Jones anterolateral approach (AL) were enrolled. Outcomes data were reviewed at a minimum of 12 months following the procedure. Clinical evaluations were made using the Merle d'Aubigne and Postel hip score. The results of these procedures were retrospectively compared with those of a historical series of 98 total hip arthroplasties that had been performed by the same surgeon with use of a posterolateral approach (PL).In the MIS AL THA group, intraoperative fracture was observed in 6 hips; 3 in greater trochanter and 3 in calcar femoral. One hip was subjected to irrigation because of postoperative infection was suspected. In the PL group, intraoperative fracture was demonstrated in 4 hips in calcar femoral. No postoperative dislocation and no pulmonary embolism or nerve paralysis was observed in both groups.The MIS AL THA did not show a clinically relevant superior outcome compared with the PL THA. When performing MIS AL THA, special attention should pay for prevention of greater trochanter fracture.

In laser processing, the heating, melting, and vaporization of work material occur continuously when high power laser energy is applied, and such phenomena make it difficult to control laser processing in an on-line mode. Hence, in the case of the material removal process, e.g., it is difficult to maintain the removal volume and the processing accuracy at a constant level without applying laser process control. The purpose of this study was to clarify the relationship between the strength of laser processing sound and the groove cross-sectional area per pulse when a processing condition and a work material is changed.

Technique for integrating optical waveguide circuits on electrical circuits is developed toward the application to opto-electronic multi-chip modules (O/E-MCM), which enables one to handle ultrafast lightwave pulses and electrical signals on a common circuit board. We use ceramic material as the substrate of O/E-MCM having a multi-layer structure. A siloxane polymer is utilized to form low loss optical waveguides on ceramic substrates by low temperature processes. The waveguide has shown high transparency around 1310 nm. The group velocity dispersion has been measured and shown to become almost zero at wavelength of 1310 nm and about 0.4 fs/cm nm at 1550 nm. An optical circuit has been formed on simple electrical circuit consisting of a thermometer film resistance layer. A Mach–Zehnder interferometer optical switch exhibiting the waveguide extinction ratio of 30 dB, control power of 5 mW, and switching speed of 6 ms has been demonstrated. Thermal management characteristics also have been demonstrated successfully. This single-mode optical waveguide can thus be formed readily on ceramic substrates involving electrical circuit layers; this result confirms its applicability to O/E-MCM.

In 78 patients undergoing a total hip replacement we examined the production of matrix metalloproteinases (MMP) by the fibroblasts from the connective tissue of the acetabulum; we then correlated these findings with clinical and radiological characteristics of the same patients. In 53 patients only MMP-2 was produced; in 15 cases MMP-1, -2 and -3 were present; and in 10 cases not only MMP-1, -2, -3 but also MMP-9. Significant differences among the clinical and radiological parameters were found in the 3 subsets. A positive correlation between the production of MMP-9 and a rapid destruction of the hip joint was found.

The friction behavior of Ni - -, SiO - 2 - and mica sodium silicate-based lubricant composites, which included BN, WS 2 and graphite as lubricants, were examined. A ring-on-disk apparatus, in which a solid lubricant composite disk was held against a rotating stainless ring, was used as the test configuration. The tests were run with a load from 62 to 250 N in temperatures from 20 to 800°C in the laboratory environment. The wear surface was characterized by scanning electron microscope and X-ray photo spectroscopy. The major findings were that both mica sodium silicate- and SiO - 2 -based composites failed at above 500°C due to severe wear and surface damage; in contrast, Ni - -based composite showed a stable friction coefficient and low wear from 20 to 800°C.

Senescence-accelerated mouse prone 8 (SAMP8) is a model of aging characterized by the early onset of learning and memory impairment and various pathological features of Alzheimer's disease (AD). Our recent studies have demonstrated that nobiletin, a polymethoxylated flavone from citrus peels, ameliorates learning and memory impairment in olfactory-bulbectomized mice, amyloid precursor protein transgenic mice, and NMDA receptor antagonist-treated mice. Here, we present evidence that this natural compound improves age-related cognitive impairment and reduces oxidative stress and tau phosphorylation in SAMP8 mice. Treatment with nobiletin (10 or 50mg/kg) reversed the impairment of recognition memory and context-dependent fear memory in SAMP8 mice. Treatment with nobiletin also restored the decrease in the GSH/GSSG ratio in the brain of SAMP8 mice. In addition, increases in glutathione peroxidase and manganese-superoxide dismutase activities, as well as a decrease in protein carbonyl level, were observed in the brain of nobiletin-treated SAMP8 mice. Furthermore, nobiletin reduced tau phosphorylation in the hippocampus of SAMP8 mice. Together, the markedly beneficial effects of nobiletin represent a potentially useful treatment for ameliorating the learning and memory deficits, oxidative stress, and hyperphosphorylation of tau in aging as well as age-related neurodegenerative diseases such as AD.

Measures for pain management after total knee arthroplasty (TKA) are important for early improvement in the quality of life after operation and early postoperative rehabilitation. We investigated the benefits and safety analgesic effect of locally injected drugs around the total knee prosthesis.60 patients undergoing TKA for osteoarthritis were divided randomly into three groups.Group A (20 knees; control group), this group did not receive multimodal drug cocktailtherapy; group B (21 knees), received intra-articular injection of a multimodal drug cocktail; and group C (19 knees), received localperiarticular injection of a multimodal drug cocktail. All analgesics administered in the first 24 h after surgery were recorded. The evaluation items included assessment of pain using a 100-point visual analogue scale (VAS) after the patients awoke on the day of the operation and on postoperative day 1, the dose of diclofenac sodium suppository, the number of days for acquiring assisted ambulation with a walking cane, and side effects. Assessment of flexion angles was conducted at postoperativeweek 1 and at theconclusion of the study.The VAS scores on the day of surgery and the amounts of diclofenac sodium used indicated good pain relief in groups B and C; the level of pain control was higher in group C than in group B. No cardiac or central nervous system toxicity was observed.Periarticular injection with multimodal drugs can significantly reduce the requirements for analgesia, with no apparent risks, following TKA.

There were 56 consecutive patients enrolled in our study from the outpatient clinic of our institution, mean age of 59 years, who had been on hemodialysis for 2–26 years. The duration of hemodialysis was less than 10 years for 36 patients (short-term group) and 10 years or more for 20 patients (long-term group) with a mean duration of 11 years at time of the investigation. In the short-term group, plain radiography revealed bone cysts in 9 hips (17%) and joint space narrowing in 10 hips (19%), while in the long-term group, there were bone cysts in 19 hips (48%) and joint space narrowing in 6 hips (15%). Bone mineral density as determined by dual-energy X-ray absorptiometry was 0.621 g/cm2 for patients in the short-term group and 0.503 g/cm2 for patients in the long-term group. Hip arthroplasties were performed in 3 patients suffering from femoral neck fracture due to bone cysts. All of them showed marked bone loss (mean 0.380 g/ cm2). In conclusion, plain radiography and dual-energy X-ray absorptiometry are useful for evaluating the hip arthropathy of hemodialysis patients, especially those who have undergone long-term hemodialysis.

Platinum(IV) [Pt(IV)] complex, satraplatin, is currently in clinical trials for the treatment of various cancers. As a key step of the anti-cancer effect exertion, satraplatin is supposed to be reduced by endogenous reductants to platinum(II) [Pt(II)] complex. In this study, we investigated the interaction of DNA, Pt(IV), and the endogenous reductants such as ascorbic acid (AsA) and glutathione (GSH). As a model Pt(IV) compound, cis-diammine-tetrachloro-Pt(IV) [cis-Pt(IV)], which is a prodrug of cisplatin [cis-diammine-dichloro-Pt(II), cis-Pt(II)], was incubated with calf thymus DNA in the presence of AsA or GSH. In the presence of AsA, cis-Pt(IV) induced oxidative DNA damage. Hydroxyl radical scavengers suppressed the AsA-associated oxidative damage, thereby suggesting that hydroxyl radicals are involved in the DNA oxidation. cis-Pt(II)-like CD spectral change and crosslink formation in calf thymus DNA were also observed during this DNA oxidation, suggesting cis-Pt(IV) reduction by AsA and DNA conformational change induced by the newly formed cis-Pt(II) binding to DNA. GSH did not induce oxidative DNA damage likely due to its own hydroxyl radical scavenging ability. Further, GSH suppressed the Pt(II)-mediated DNA conformational change and crosslink formation, suggesting that GSH sequesters the cis-Pt(II) away from DNA by GSH-cis-Pt(II) complex formation.

Cobalt oxide thin films were prepared by a low-temperature atmospheric-pressure chemical vapor deposition method. The raw material was cobalt (II) acetate which is non-toxic and easy to handle. Polycrystalline films were obtained at a reaction temperature above 300°C. From near-normal reflection measurements it follows that the films have solar absorptance α = 0.73 and thermal emittance ϵ = 0.089.

The transverse-rupture strength of WC-(10, 20)%Co alloys oxidized in air at the temperature of 973-1173 K for the time of 0.3-1.8 ks was studied as a function of Co content, carbon content of alloys, WC grain size and r-domain size. It was found that the strength of the alloy decreased owing to the surface oxidation and the strength decrease became larger with rising temperature and increasing time. It was noted that the strength decrease was enhanced with increasing r-domain size. It was also found that the rate of oxidation was faster in the domain boundary than inside the domain, so that the domain boundary after oxidation acted as a notch during transverse-rupture test, leading to the strength decrease.

High carbon WC-(10, 20)mass%Co micro-grained alloys containing no Cr3C2 or 5.0 mass%Cr3C2 in binder, were prepared by solid-phase sintering, that is, vacuum sintering at 1473-1553K and then HIP-treatment at 1523 K. Some properties such as microstructure and mechanical properties were studied. It was found that the alloys prepared by solid-phase sintering, for instance, sintering at 1523K for 10.8ks and then HIP-treatment, reached full density and showed excellent mechanical properties. As to the transverse-rupture strength, the alloys, having grain size of 0.5-0.6 μm, exhibited the value so high as about 3.5 GPa or more, independent of binder content or Cr3C2 addition. It was noted that WC grains recrystallized during solid-phase sintering.

The wavelength response of a waveguide volume grating coupler (WVGC) is analyzed for coupling light from a slab waveguide into the superstrate. A leaky-mode approach is used in conjunction with rigorous coupled-wave analysis. A quantitative theoretical study of the effect of index modulation, waveguide index, and grating thickness on the wavelength bandpass of a WVGC is also presented. The FWHM wavelength bandpasses found for high-efficiency couplers range from 173 to 525 nm. The various Bragg conditions that can be used in designing a WVGC are also presented and compared. The use of the propagation constant of the mode being outcoupled as the incident wave vector in the Bragg condition is shown to produce the highest coupling efficiency.

Proximal bone resorption around the femoral stem is one of the major complications of cementless total hip arthroplasty (THA). The potential complications resulting from proximal bone resorption include femoral fracture and late stem loosening. The purpose of this study was to evaluate the effects of cyclic therapy with etidronate on periprosthetic bone mineral density (BMD) after cementless THA. Fifty-two patients who had undergone cementless THA were randomized for this study. Group A consisted of 30 hips in 29 patients without osteoactive drugs. Group B consisted of 23 hips in 23 patients with cyclic therapy with etidronate (i.e., 400 mg/day of oral etidronate for 2 weeks followed by 12 weeks of 500 mg/day of calcium lactate and repeated every 14 weeks), one of whom was excluded from the study because of side effects attributed to the drug. Periprosthetic BMD in seven regions of interest based on the zones of Gruen et al. (Clin. Orthop. 141 (1979), 17-27) was measured with dual energy X-ray absorptiometry (DXA) at 3 weeks, 6 months, and 12 months postoperatively. The postoperative decreases of BMD in group B were significantly lower than those in group A in zones 1 and 7 (P0.05 and P0.05, respectively) at 6 months and in zones 1, 2, 6, and 7 (P0.05, P0.05, P0.05, and P0.001, respectively) at 12 months. The BMD change appeared to be stabilized at 6 months in all zones in group B, while in group A there was a progressive decrease of average BMD (6.1%) in zone 7 between 6 months and 12 months. These findings suggested that cyclic therapy with etidronate may help to reduce the resorptive changes in the proximal part of the femur after cementless THA. Further follow-up study with larger populations will be required to define the potential efficacy of intermittent cyclic etidronate therapy on postoperative bone loss.

Transparent conductive fluorine‐doped indium oxide thin films were prepared by an atmospheric‐pressure chemical vapor deposition method. The raw material was [C7H15COO]2 In[H(CF2)4COO]. The polycrystalline films were obtained at a reaction temperature in the range of 350–450 °C. This chemical vapor deposition method incorporates fluorine atoms into a crystalline structure and consequently leads to a high carrier concentration, yielding fluorine‐doped indium oxide films of low resistivity. For the 320‐nm‐thick film deposited at 380 °C, the resistivity was 4.4×10−4 Ω cm, and the sheet resistance was 13.8 Ω/⧠.
