Abstract

The formation of spores by the yeast Saccharomyces cerevisiae, in a medium rich in nutrients, requires the presence of L-lysine and Ca2+ and is independent of the cAMP concentration. Sporulation takes place only when the concentration of L-lysine becomes 0.2-0.5 mmol/g cells. Ca2+ is obtained by the movement of Ca2+ into the cell from the external medium. D-lysine is also able to induce sporulation. The cells forming spores increase in size and collect L-lysine and L-cysteine.

Abstract

1. In the [13C]-octanoic acid breath test, the time versus pulmonary [13CO2] excretion rate curve is analysed using mathematical curve-fitting techniques to calculate gastric emptying parameters. Thus, the goodness-of-fit highly influences the accuracy of the breath test. However, a double-peaked [13CO2] excretion curve, which occasionally develops owing to the presence of an interval of quiescent gastric emptying (the plateau phase), is likely to be fitted poorly. 2. In pharmacokinetics, the Wagner-Nelson method has been used to describe precisely the absorption kinetics of orally administered drugs and its reliability is independent of the nature of gastric emptying. A recent study has shown the potential of the Wagner-Nelson method to generate a realistic gastric emptying flow curve from [13CO2] excretion data. In the present report, we have demonstrated that the Wagner-Nelson method can visualize the plateau emptying phase responsible for the cases of double peaks. 3. Wagner-Nelson analysis applied to the breath test described precisely the characteristic emptying pattern of the two emptying phases being interrupted by the plateau phase. Conventional analysis for the breath test failed to detect the plateau phase. 4. The Wagner-Nelson method is a useful tool for analysing atypical double-peaking [13CO2] excretion curves. Abstract

Background. Cigarette smoking is associated with an increased risk of peptic ulcer and gastroesophageal reflux disease. Gastric emptying disorders may play a role in the development of these upper gastrointestinal diseases. Thus, studies examining a link between smoking and gastric emptying disorders have clinical relevance. This study was conducted to investigate the effect of smoking on gastric emptying of solids in Japanese smokers. Methods. The 13C-octanoic acid breath test was performed in eight male habitual smokers on two randomized occasions (either sham smoking or actively smoking). The time vs 13CO2 excretion rate curve was mathematically fitted to a conventional formula of y (t) = m*k*β*e-k*t*(1 - e-k*t)β-1, and the parameters of k and were determined: under the crossover protocol, a larger (smaller) β indicates slower (faster) emptying in the early phase, and a larger (smaller) k indicates faster (slower) emptying in the later phase. The half 13CO2 excretion time (t1/2b = -[1n(1 - 2-1/β)]/k) and the time of maximal 13CO2 excretion rate (tmax = [1nβ]/k) were also calculated. Between the two occasions, k, βt1/2b, and tmax were compared by the Wilcoxon signed-rank test. Results. After smoking, k was significantly increased. No significant differences were found in β, t1/2, and tmax between the two occasions. Conclusions. The increase in k suggests the acceleration of gastric emptying in the later phase. For the first time, this study has revealed that acute smoking speeds the gastric emptying of solids in Japanese habitual smokers. [ABSTRACT FROM AUTHOR]

40. Ca 2+/calmodulin-activated protein phosphatase

Abstract

Protein phosphatase (PP2B) whose activity is stimulated 12-20-fold by Ca 2+/calmodulin (CaM) was partially purified by CaM-Sepharose and heparin-agarose chromatographies from cell extract of the yeast Saccharomyces cerevisiae. PP2B activity was not detectable in a mutant in which two genes ( CMP1and CMP2) encoding homologs of mammalian PP2B catalytic subunit were disrupted. We have previously shown that the double gene disruption has no significant effect on the growth of yeast [1991, Mol. Gen. Genet. 227, 52–59]. The results indicated that CMP1and CMP2are the only genes that encode the PP2B catalytic polypeptide in S. cerevisiae,and PP2B activity is not essential for the growth of the yeast under normal conditions.
