OBJECTIVE Titanium bridges are used to separate the thyroid ala during type 2 thyroplasty for adductor spasmodic dysphonia. Revision surgeries have adventitiously indicated that bridge wing failure occurs in the area of the medial hole in some cases. This study investigated the rate and cause of device malfunctions and developed an improved device. METHODS We conducted a questionnaire survey to determine the number of surgeries performed in Japan up to the end of 2014, and to obtain information about revision cases. In addition, damage analyses were performed on the fracture surfaces of recovered titanium bridges through use of scanning electron microscopy (SEM). RESULTS Between 2002 and 2014, titanium bridges were used in 385 patients. Revision surgery was performed in 19 cases. Revision surgeries revealed that in 11 cases breakage occurred in the wings of the device in the region of the medial hole. However, such fractures were not associated with any signs of recurrence or any adverse events. SEM analyses of fracture surfaces confirmed that fatigue fractures were caused by repeated bending stress in the area of the medial hole. Based on these results, the shape of the hole was changed from round to oval and the wing thickness was increased to prevent breakage. CONCLUSIONS The wings of titanium bridges may break without any associated signs, symptoms or tissue damage. Based on the malfunctions detected and analyses of the devices recovered following malfunction, changes to the specification were made for commercial development of the titanium bridge.

Objective The objective of this study was to evaluate safety and efficacy of regenerative treatment using gelatin sponge with basic fibroblast growth factor (bFGF) in patients with tympanic membrane perforation (TMP). Methods The current study was a prospective, multicenter, open-label, single-arm, and exploratory clinical trial to evaluate the safety and efficacy of the TM regeneration procedure (TMRP). Myringotomy was used to mechanically disrupt the edge of the TMP, and a gelatin sponge immersed in bFGF was then placed over the perforation. Fibrin glue was dripped over the sponge as a sealant. TMP closure was examined 4 weeks later and, if insufficient, TMRP was repeated a maximum of three more times. TMP closure and hearing improvement 12 weeks after the final TMRP as well as safety were evaluated. Results Of the 11 patients with TMP who participated in this study, one who fulfilled the exclusion criteria and did not undergo TMRP and one with cholesteatoma were excluded from the efficacy analysis. TMP closure and hearing improvement 12 weeks after the final TMRP were achieved in eight out of nine patients (88.9%). Mean bone conduction threshold significantly improved 12 weeks after the TMRP compared with baseline (35.7 ± 20.3 vs 29.4 ± 21.0 dB, P = 0.015). Six out of ten patients receiving TMRP experienced temporary adverse events: appendicitis (serious, severe), otorrhea (mild), otitis media (mild), and sudden hearing loss (mild). However, none were related to the protocol treatment. Conclusion TMP closure and hearing improvement were frequently confirmed following the TMRPs which were safely performed. These favorable outcomes were accompanied with significant improvement of the bone conduction threshold. These promising outcomes would encourage a large-scaled, randomized and pivotal clinical trial in the future. Plant breeding programs in local regions may have genetic and phenotypic variations that are desirable and shape adaptability during the establishment of local populations. Despite the characterization of genetic population structures in various kinds of populations, the effects of variations in phenotype on agro-economical traits currently remain unclear. In the present study, we evaluated phenotypic changes in 26 agro-economical traits among the local population during rice breeding programs in Hokkaido. Wide variations were observed in all 26 agro-economical traits with continuous distributions. In order to elucidate improvements in these agro-economic traits during rice breeding programs in Hokkaido, values were compared between genetic population structures. Traits were classified into four patterns based on the timing of significant differences. Patterns A and B showed significant differences once and twice, respectively. Pattern C gradually showed significant differences. Pattern D showed no significant differences for the desired directions. Based on the changes in phenotype observed in the present study and the genetic population structure for the local population in Hokkaido, a model of the artificial selection for phenotypes in genetic diversity among the local population during plant breeding programs has been proposed.

Quantitative trait loci (QTLs) associated with eating quality, grain appearance quality and yield-related traits were mapped in recombinant inbred lines (RILs) derived from closely related rice (Oryza sativa L. subsp. japonica) cultivars, Yukihikari (good eating quality) and Joiku462 (superior eating quality and high grain appearance quality). Apparent amylose content (AAC), protein content (PC), brown grain length (BGL), brown grain width (BGWI), brown grain thickness (BGT), brown grain weight per plant (BGW) and nine yield-related traits were evaluated in 133 RILs grown in four different environments in Hokkaido, near the northernmost limit for rice paddy cultivation. Using 178 molecular markers, a total of 72 QTLs were detected, including three for AAC, eight for PC, two for BGL, four for BGWI, seven for BGT, and six for BGW, on chromosomes 1, 2, 3, 4, 6, 7, 8, 9, 11 and 12. Fifteen intervals were found to harbor multiple QTLs affecting these different traits, with most of these QTL clusters located on chromosomes 4, 6, 8, 9 and 12. These QTL findings should facilitate gene isolation and breeding application for improvement of eating quality, grain appearance quality and yield of rice cultivars.

Background and purpose Akinetic mutism is thought to be an appropriate therapeutic end-point in patients with sporadic Creutzfeldt−Jakob disease (sCJD). However, prognostic factors for akinetic mutism are unclear and clinical signs or symptoms that precede this condition have not been defined. The goal of this study was to identify prognostic factors for akinetic mutism and to clarify the order of clinical sign and symptom development prior to its onset. Methods The cumulative incidence of akinetic mutism and other clinical signs and symptoms was estimated based on Japanese CJD surveillance data (455 cases) collected from 2003 to 2008. A proportional hazards model was used to identify prognostic factors for the time to onset of akinetic mutism and other clinical signs and symptoms. Results Periodic synchronous discharges on electroencephalography were present in the majority of cases (93.5%). The presence of psychiatric symptoms or cerebellar disturbance at sCJD diagnosis was associated with the development of akinetic mutism [hazard ratio (HR) 1.50, 95% confidence interval (CI) 1.14–1.99, and HR 2.15, 95% CI1.61–2.87, respectively]. The clinical course from cerebellar disturbance to myoclonus or akinetic mutism was classified into three types: (i) direct path, (ii) path via pyramidal or extrapyramidal dysfunction and (iii) path via psychiatric symptoms or visual disturbance. Conclusions The presence of psychiatric symptoms or cerebellar disturbance increased the risk of akinetic mutism of sCJD cases with probable MM/MV subtypes. Also, there appear to be sequential associations in the development of certain clinical signs and symptoms of this disease.

New measurement conditions for arsenic speciation analysis of rice flour were developed using HPLC-ICP-MS equipped with a reversed phase ODS column. Eight arsenic species, namely, arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMAA), dimethylarsinic acid (DMAA), trimethylarsine oxide (TMAO), tetramethylarsonium (TeMA), arsenobetaine (AsB) and arsenocholine (AsC), were separated and determined under the proposed conditions. In particular, As(III) and MMAA and DMAA and AsB were completely separated using a newly proposed eluent containing ammonium dihydrogen phosphate. Importantly, the sensitivity changes, in particular those of As(V) and As(III) caused by coexisting elements and by complex matrix composition, which had been problematical in previously reported methods, were eliminated. The new eluent can be applied to C8, C18 and C30 ODS columns with the same effectiveness and with excellent repeatability. The proposed analytical method was successfully applied to extracts of rice flour certified reference materials.

AIM: Some previous studies have shown a positive relation between geomagnetic disturbances and an increased incidence of suicide. If such a relation exists, stronger geomagnetic fields may affect the number of suicides, because stronger geomagnetic fields generally cause larger geomagnetic field disturbances. Therefore, we here investigated the relation between local geomagnetic field magnetic flux density and the standardized morbidity ratios (SMR) for suicide by each prefecture in Japan. METHODS: Monthly suicide data for each prefecture in the period January 1999 to December 2008 was obtained, and it was found that a total of 216 171 male individuals and 85 154 female individuals committed suicide during this period. A multiple linear regression analysis was carried out with a backward elimination procedure. The SMR for suicide by each prefecture was taken as the response variable and the explanatory variables were each prefecture's local geomagnetic field magnetic flux density (nT), north latitude (°), monthly mean unemployment rate (%), monthly mean air pressure (hPa), monthly mean air temperature (°C), monthly mean humidity (%), and monthly total day length (hours). Analyses were carried out separately for each sex. RESULTS: In the multiple linear regression analysis for male subjects, the local geomagnetic field magnetic flux density (nT), monthly mean unemployment rate (%), and monthly mean humidity (%) were associated with the incidence of suicide, but in the multiple linear regression analysis of female subjects, only north latitude was associated with that. Corrosion behavior is a key issue in the assessment of disposal performance for activated waste such as spent fuel assemblies (i.e., hulls and end-pieces) because corrosion is expected to initiate radionuclide (e.g., C-14) leaching from such waste. Because the anticipated corrosion rate is extremely low, understanding and modeling Zircaloy (Zry) corrosion behavior under geological disposal conditions is important in predicting very long-term corrosion. Corrosion models applicable in the higher temperature ranges of nuclear reactors have been proposed based on considerable testing in the 523−633 K temperature range.In this study, corrosion tests were carried out to confirm the applicability of such existing models to the low temperature range of geological disposal, and to examine the influence of material, environmental, and other factors on corrosion rates under geological disposal conditions. A characterization analysis of the generated oxide film was also performed.To confirm applicability, the corrosion rate of Zry-4 in pure water with a temperature change from 303 K to 433 K was obtained using a hydrogen measuring technique, giving a corrosion rate for 180 days of 8 × 10-3 μm/y at 303 K.To investigate the influence of various factors, corrosion tests were carried out. The corrosion rates for Zry-2 and Zry-4 were almost same, and increased with a temperature increase from 303 K to 353 K. The influence of pH (12.5) compared with pure water was about 1.4 at 180 days at 303 K.

We previously showed that the agamid lizard Pogona vitticeps responded to an extremely low-frequency electromagnetic field (ELF-EMF, frequency: 6 and 8 Hz, peak magnetic field: 2.6 &micro, T, peak electric field: 10 V/m) with tail-lifting behavior. In addition, the tail-lifting response to ELF-EMF disappeared when the parietal eyes of the lizards were covered by small round aluminum caps. This result suggests that the parietal eye contributes to light-dependent magnetoreception. In the present study, we set up an ELF-EMF group to evaluate the long-term effect of the ELF-EMF on lizards&rsquo, behavior and examine our hypothesis that exposure to ELF-EMFs increases the magnetic field sensitivity in lizards. We therefore include the lunar phase (full moon/new moon) and K index as environmental factors related to the geomagnetic field in the analysis. The number of tail lifts per individual per day was the response variable while calendar month, daily mean temperature, daily mean humidity, daily mean atmospheric pressure, full moon, new moon, and K index were the explanatory variables. We analyzed an ELF-EMF group and a control group separately. In a multiple linear regression analysis, the independent determinants associated with the number of tail lifts were the full moon, the temperature, February, March, April, and May in the ELF-EMF group and March, April, May, and June in the control group. The P. vitticeps in the ELF-EMF group responded to the full moon whereas those in the control group did not. In addition, in the ELF-EMF group, the number of tail lifts was higher on days when the K index was higher (P = 0.07) in the first period whereas there was no such tendency in either period in the control group. There is the possibility that the exposure to ELF-EMFs may increase magnetic-field sensitivity in lizards.

The Ppd-A1 genotype of 240 Japanese wheat cultivars and 40 foreign cultivars was determined using a PCR-based method. Among Japanese cultivars, only 12 cultivars, all of which were Hokkaido winter wheat, carried the Ppd-A1a allele, while this allele was not found in Hokkaido spring wheat cultivars or Tohoku-Kyushu cultivars. Cultivars with a photoperiod-insensitive allele headed 6.9-9.8 days earlier in Kanto and 2.5 days earlier in Hokkaido than photoperiod-sensitive cultivars. The lower effect of photoperiod-insensitive alleles observed in Hokkaido could be due to the longer day-length at the spike formation stage compared with that in Kanto. Pedigree analysis showed that 'Purple Straw' and 'Tohoku 118' were donors of Ppd-A1a and Ppd-D1a in Hokkaido wheat cultivars, respectively. Wheat cultivars recently developed in Hokkaido carry photoperiod-insensitive alleles at a high frequency. For efficient utilization of Ppd-1 alleles in the Hokkaido wheat-breeding program, the effect of Ppd-1 on growth pattern and grain yield should be investigated. Ppd-A1a may be useful as a unique gene source for fine tuning the heading time in the Tohoku-Kyushu region since the effect of Ppd-A1a on photoperiod insensitivity appears to differ from the effect of Ppd-B1a and Ppd-D1a.

Corrosion behavior is a key issue for the waste disposal of irradiated metals, such as hulls and endpieces, and is considered to be a leaching source of radionuclides including C-14. However, little information about Zircaloy corrosion in anticorrosive conditions has been provided.In the present study, long-term corrosion tests of Zircaloy-4 and Zircaloy-2 were performed in assumed disposal conditions (dilute NaOH solution, pH 12.5, 303 K) by using the gas flow system for 1500 days. The corrosion rate, which was determined by measuring gaseous hydrogen and the hydrogen absorbed in Zircaloy, decreased with immersion time and was lower than the value of 2×10−2 μm/y used in performance assessment (1500-day values: 5.84×10−3 and 5.66×10−3 μm/y for Zircaloy-4, 1000-day values: 8.81×10−3 μm/y for Zircaloy-2). The difference in corrosion behavior between Zircaloy 4 and Zircaloy-2 was negligible. The average values of the hydrogen absorption ratios for Zircaloy-4 and Zircaloy-2 during corrosion were 91% and 94%, respectively.The hydrogen generation kinetics of both gas evolution and absorption into metal can be shown by a parabolic curve. This result indicates that the diffusion process controls the Zircaloy corrosion in the early corrosion stage of the present study, and that the thickness of the oxide film in this stage is limited to approximately 25 nm and may therefore be in the form of dense tetragonal zirconia.

There is some evidence that exposure of various animals to a power-frequency magnetic field can cause increases in body mass. However, the precise nature of the relationship is not clear, particularly for long-term exposure. To clarify the relationship between long-term power-frequency magnetic field exposure (2 years) and animal body mass, we conducted a pooled analysis of data in two studies to examine the impact of power-frequency magnetic field exposure on body mass as compared with sham-exposed rats of both sexes. Relative body mass was the response variable, and the study duration (in weeks), magnetic flux density ($\upmu$T), and daily exposure time (hours/day) were the explanatory variables with adjustment for the study. The effects of extremely low-frequency electromagnetic fields (ELF-EMFs) on blood pressure (BP) are controversial. In this double-blind, randomized, sham-controlled study, we examined the effects of repeated exposure to a 1-μT ELF-EMF on BP in 20 humans with mild-to-moderate hypertension. Subjects were randomly assigned to either the ELF-EMF group or the sham group. Subjects in the ELF-EMF group were exposed to an ELF-EMF (6- and 8 Hz, respectively, peak magnetic field 1 μT, peak electric field 10 Vm(-1)) for at least two 10- to 15-min sessions per week, over a period of 4 weeks. In the sham group, the EMF-generating apparatus was not active. We obtained systolic and diastolic BP (SBP and DBP, respectively) measurements at registration and before and after each ELF-EMF exposure session. Subjects in the ELF-EMF and sham groups had mean ages of 52.8 and 55.1 years, and were exposed to a mean of 9.9 and 9.0 sessions, respectively. There was a significant difference between the ELF-EMF and sham groups with respect to change in SBP value between baseline and the end of the exposure regimen (P=0.02), but not with respect to change in DBP (P=0.21). There were no adverse events other than mild paresthesia of the hands of two subjects in the ELF-EMF group. Our results suggest that repeated exposure to an ELF-EMF has a BP-lowering effect on humans with mild-to-moderate hypertension.

SUMMARY Animals from a wide range of taxa have been shown to possess magnetic sense and use magnetic compasses to orient; however, there is no information in the literature on whether lizards have either of these abilities. In this study, we investigated the behavioral responses of a diurnal agamid lizard (Pogona vitticeps) to a sinusoidal extremely low-frequency electromagnetic field (ELF-EMF; 6 and 8 Hz, peak magnetic field 2.6 μT, peak electric field 10 V m−1). Fourteen adult lizards were divided randomly into two groups (the EMF and control groups; each group had three males and four females). The EMF group received whole-body exposure to ELF-EMF and the control group did not. Lizards in the EMF group were exposed to ELF-EMF for 12 h per day (during the light period). The number of tail lifts was monitored beginning 3 days before exposure and ending after 5 days of exposure. For each individual, the average number of tail lifts per day was calculated. The average number of tail lifts per individual per day was greater in the EMF group than in the control group (20.7±6.3 and 9.1±4.5 tail lifts, respectively, N=7 each, P=0.02). We confirmed the reproducibility of this response by a cross-over trial. These results suggest that at least some lizards are able to perceive ELF-EMFs. Furthermore, when the parietal eye of the lizards was covered with a small round aluminum ‘cap’ which could block light, the tail-lifting response to ELF-EMF disappeared. Our experiments suggest that (1) lizards perceive EMFs and (2) the parietal eye may be involved in light-dependent magnetoreceptive responses.

放射性廃棄物を地下に処分したときの炭素鋼などの廃棄物含有金属と廃棄物に含有される NO3–の化学的相互作用のモデル化を，混成電位の概念に基づき行った．炭素鋼は処分施設の金属製構成要素および金属性放射性廃棄物の例として選定した．炭素鋼の腐食に伴う NO3–の還元反応は，NO3– → NO2– → NH3 なる逐次反応と見なし，この反応及び水の還元による電流が，炭素鋼がマグネタイトになる酸化電流とバランスするとしてモデル化した．この速度論的モデルの入力パラメータは，電気化学測定実験及び浸漬試験により決定した．本モデルを用いた解析により，NO3–共存下における浸漬実験の結果を説明することができた．

Objective: In a previous paper, we described the major health tragedy that occurred in Japan involving the use of gefitinib, and recommended steps to prevent such problems occurring again. In that paper, we argued that erlotinib should only be approved on the condition that all users are subject to surveillance. Erlotinib was subsequently approved for use in Japan on October 19, 2007. In the present paper, we examine whether our recommendations have been implemented in the safety measures established for erlotinib use. Methods: We comprehensively reviewed reports regarding erlotinib treatment published between 2007 and 2009 by regula- tory agencies and the manufacturer of the erlotinib-containing drug. We evaluated the safety measures established for erlotinib in view of three problems we identified with the treatment of gefitinib marketing: (1) the results of animal experiments and pre-marketing clinical trials, and reports of adverse drug reactions from other countries were not properly incorporated into the product information; (2) indications for the drug were expanded without strict evaluation of the external validity of pre- marketing clinical trials; and (3) despite many serious cases of interstitial lung disease being spontaneously reported, well- designed post-marketing surveillance was not conducted immediately after these problems became evident. Results: We found that appropriate measures were taken for erlotinib in relation to each of the three above-mentioned prob- lems. We found that there were fewer fatal adverse reactions to erlotinib after marketing relative to the pre-marketing period. Conclusions: Our recommendations following the health tragedy caused by gefitinib were implemented in the safety mea- sures for erlotinib, which probably explains the smaller number of fatal adverse reactions to erlotinib in post-marketing surveil- lance than in pre-marketing trials.

An empirical and analytical study was performed on the aqueous dissolution of silver iodide (AgI) to release iodine under reducing conditions with Fe2+ in order to understand the fundamental chemical and/or physical behavior of potential radioactive iodine waste forms under geological disposal conditions. Aqueous dissolution tests of AgI powder in FeCl2 solutions (10−6 M to 10−3 M) were performed in a glove box purged with a gas mixture (Ar + 5% H2). The test results showed that AgI dissolves to release iodine at extremely slow rates, being controlled by a diffusion process in any FeCl2 solution. The comparison with thermodynamic calculations based on redox equilibria suggested that the AgI dissolution proceeds by redox reaction between Ag+ and Fe2+; however, it was far from the thermodynamic equilibrium. These results suggested that the form of AgI itself has a potential to immobilize iodine for a long time even under the disposal conditions. Decomposition of 14 C containing organic molecules into an inorganic compound has been investigated by γ -ray irradiation experiments under simulated repository conditions for radioactive waste. Lower molecular weight organic acids, alcohols, and aldehydes leached from metallic waste are reacted with OH radicals to give carbonic acid. A decomposition efficiency that expresses consumption of OH radicals by decomposition reaction of organic molecules is proposed. Decomposition efficiency increases with increasing concentration of organic molecules (1×10 −6 –1×10 −3 mol dm −3 ) and is not dependent on dose rate (10–1000 Gy h −1 ). Observed dependence indicates that decomposition efficiency is determined by reaction probability of OH radicals with organic molecules.

Arsenic (As) uptake in human occurs via the food chain mainly. The Joint FAO/WHO Expert Committee on Food Additives has established the provisional tolerable weekly intake level for As as an inorganic As (iAs) value, because iAs in food is much more toxic than organic As. In this study, we studied an acid based partial-digestion method for the complete extraction of arsenicals from rice. HPLC/ICP-MS was used to determine the concentration of iAs selectively. The conditions adopted to extract arsenicals from a 0.5 g of finely ground rice sample were addition of 2 mL of 0.15 mol/L nitric acid and heating at 80 degrees C for 2 hr. The LOD and LOQ for iAs were 0.0024 and 0.0079 mg/kg dry weight, respectively. Recovery studies showed good accuracy. When the method was applied to ten short-grain brown rice samples, the iAs concentrations were 0.108-0.227 mg/kg dry weight and the total As concentrations were 0.118-0.260 mg/kg dry weight. Although dimethylarsinic acid was also detected in most samples, the percentage of iAs content in total As content was 62.2-96.3%. Thus, iAs was the principal As species in the short-grain brown rice samples tested.

Because there is a great difference between the toxicity of inorganic arsenic (As) and organic As in food, the JECFA has set a PTWI value for inorganic As (iAs) rather than for total As. The difference in As toxicity makes it necessary to extract iAs completely from food samples for toxicological analysis, but complete extraction of As from most foods including seaweed has not been achieved to date. We developed a partial-digestion method that uses nitric acid as a solvent in order to extract almost all arsenicals from the solid matrix of hijiki (Hizikia fusiforme, a brown alga) samples. In this method, organic As species were not converted into iAs. HPLC/ICP-MS was then used to determine the concentration of iAs. Total As was measured by hydride generation-atomic absorption spectrometry. The adopted conditions for 0.1 g of ground fine powder sample were: 2 mL of 0.3 mol/L nitric acid; heating, 80 degrees C for 1 hr. Intra-laboratory validation of the method showed good precision and accuracy. The repeatability and intermediate precision for iAs were 1.5% and 1.5%, respectively. The LOD and LOQ for iAs were 0.14 and 0.46 mg/kg dry weight, respectively. Recovery studies performed by spiking 0.5 mg/kg dry weight as the LOQ level and by spiking 3 mg/kg dry weight as the iAs concentration of an un-spiked hijiki sample showed good accuracy. The method was applied to hijiki samples after a water soaking process and a water soaking and simmering process. The results suggested that the As concentration in hijiki after both processes was lower than that before the treatments and that the water soaking and simmering process reduced the iAs concentration much more effectively than the water soaking process.

Aims To report the changes in survival over 20 years of 775 breast cancer women operated between 1982 and 2003 at the Kyoto University Hospital in Japan, reflecting changes in clinical practice over that period. Results Survival curves have significantly improved between the periods 1982–1989 and 1990–2003. The 5- and 10-year survival rates between these periods were 80.3% and 85.1%, and 67.5% and 75.0%, respectively. Moreover, there was a difference in overall survival curves of patients of stages II and III, of 35–54 ages, or of positive estrogen receptor (ER) status between these periods. Conclusion The present study presented the recent advance of the survival rates might be due to the rational development of breast cancer treatment, and suggested the possibility that the patients of stages II and III, of 35–54 ages, or of positive ER status were received benefits by these treatments.

In order to determine trace amounts of lead in copper gluconate by atomic absorption spectrometry (AAS), the authors investigated a separation and pre-concentration procedure using a co-precipitation technique with bismuth. After ashing 2.0 g of the sample by means of a dry process, the ash was dissolved in (1→100) nitric acid and 75 μg of bismuth was added. Lead was co-precipitated by using an ammonium solution controlled to pH 9.5-10.5. The precipitate was left at room temperature for over 15 minutes to age, and then washed with a (3→100) ammonium solution three times. The precipitate was dissolved in (1→100) nitric acid and then analyzed by AAS. The quantification limit of this method was 0.5 mg/kg, and the trueness, repeatability and intermediate precision were 99.6%, 4.2% and 4.2% at the spiked concentration of 0.5 mg/kg, and 94.4%, 2.8% and 4.0% at the spiked concentration of 5.0 mg/kg, respectively. Thus, the present method for trace analysis of lead in copper gluconate was validated.

The soft winter wheat variety, Kitahonami, shows a superior flour yield in comparison to other Japanese soft varieties. In order to map quantitative trait loci (QTL) associated with the flour-yield trait, association mapping was performed using panel lines in Kitahonami’s pedigree, along with leading varieties and advanced breeding lines. Using a mixed linear model corrected for kernel types and familial relatedness, 62 marker-trait associations were identified and classified into 21 QTLs. Five out of eight QTLs tested were validated by linkage analyses using three sets of doubled haploid populations from crosses in which Kitahonami was used as a parent. Among them, QTLs on 3B and 7A chromosome showed highly significant effects and consistency across the three populations. A joint linkage map of 3B showed that the QTL on this chromosome was located at the same interval across the populations. By applying a meta-analysis approach, we have succeeded in identifying QTLs with consistent contributions to high flour yield across various genetic backgrounds.

FK506, originally classified as an immunosuppressant, may also be implicated in some events in the central nervous system. FK506 elicits both neuroprotective and neurotrophic effects in vitro. FK506 is neuroprotective for focal cerebral ischemia, but it is not clear whether FK506 has neuroprotective effects for other brain diseases. In this study, we investigated possible neuroprotective effects of FK506 in experimental temporal lobe epilepsy (TLE) induced by kainic acid (KA) or trimethyltin (TMT). In rat models, we observed marked protection against seizures, abnormal behaviors, and accompanying delayed neuronal damage in the hippocampus by the systemic injection of FK506.

放射性廃棄物を地下に処分したときの炭素鋼などの廃棄物含有金属と廃棄物に含有されるNO3-の化学的相互作用を評価するために, 閉鎖系で炭素鋼とNO3-を共存させ, NO3-の化学的変遷挙動及び水素ガス発生挙動を検討した. なお, 検討するpH範囲は, セメント系材料中を想定し, 10.0～13.5とした.pHが12.5の条件での炭素鋼のカソード分極測定において, NO3-あるいはNO2-の共存する条件でのカソード反応は, Tafel型の電位依存性を示すことがわかった. NO2-濃度1.0mol/dm3の場合及びNO3-濃度1.0mol/dm3の場合とNO3-もNO2-も含まない場合の同一電位での電流密度を比較すると, それぞれおよそ2～3桁程度及び1～2桁程度大きいにもかかわらず, 炭素鋼の腐食速度は, 硝酸塩の濃度の影響は受けないかむしろ低下するという結果を得た. これは, 腐食反応系がアノード支配型であることを示唆している. 炭素鋼の腐食に伴うNO3-の還元過程は, NO3-→NO2-→NH3なる逐次反応と考えられる. また, アノード支配型の腐食反応におけるカソード過程として, NO3-の還元反応は, 水の還元反応と競合すると考えられる. NO3-濃度が高い場合 (e.g. 1.0mol/dm3) には, NO3-の共存しない場合と比較して, 腐食に伴う水素発生速度が1/100から1/500と非常に小さくなった. NH3の生成速度は, NO3-濃度が1.0×10-3～1.0mol/dm3の範囲でほとんど影響はなかった.

Objective: The aim of this study was to investigate the outcomes of gemcitabine-treated patients with inoperable biliary tract cancers. Methods: We conducted a retrospective study of consecutively treated 22 inoperable biliary tract cancer patients with gemcitabine (500–1,000 mg/m2 on days 1, 8, 15 every 4 weeks) as first-line, and 17 patients as second- or third-line treatment. Results: The response rate of patients treated with gemcitabine as first-line and second- or third-line treatment was 5.3 and 28.5%, respectively. The median overall survival time in the first-, and second- or third-line treatment groups was 8.3 and 17.0 months, and the 1-year survival rate was 44.0 and 50.9%, respectively. The present study also suggests the possibility that the prognosis of patients with high levels of C-reactive protein and total bilirubin, or a low level of albumin might be worse. Conclusions: Our results indicate that the treatment of inoperable biliary tract cancers with gemcitabine is feasible. There was no difference in the response rate and overall survival between biliary tract cancer patients in the first- and second- or third-line treatment groups. We also present the systematic review of literature of the recent treatment results of biliary tract cancers treated with gemcitabine.

Pre-harvest sprouting and high alpha-amylase activity are the most serious problems for the wheat production in Hokkaido, the northern most part of Japan. It has become more frequent for wheats to be exposed to continuous rain for several days in harvest time. In addition, low temperature below 15 °C, combined with rainfall, accelerates the damage of pre-harvest sprouting. Therefore, we started a breeding program in 1998 to develop wheat varieties with excellent resistant to sprouting damage in Hokkaido.

Summary: Purpose: We analyzed the short- and long-term effects of adrenocorticotropic hormone (ACTH) therapy for patients with epileptic spasms (ESs) who did not meet the criteria of West syndrome (WS). Methods: The subjects were 30 patients, including 13 boys and 17 girls, who had received ACTH therapy between 1970 and 2003. We excluded patients with WS, but included those with a history of WS who no longer showed hypsarrhythmia at the period of ACTH therapy. The age at onset of ESs and at ACTH therapy ranged from 2 to 82 months with a median of 18 months, and from 11 to 86 months with a median of 29 months, respectively. Results: Excellent and poor responses were obtained in 19 (63%) and 11 (37%) patients, respectively, as a short-term effect. Although the patients could be subclassified into five subgroups according to the previous reports, no difference was seen in shortterm response to ACTH. Among 17 of the 19 patients with excellent short-term outcomes and a follow-up of >1 year after the ACTH therapy, eight patients have continued to be seizure free (29%; excellent long-term effect), whereas the remaining nine patients had a recurrence of seizures (complex partial seizures, four; generalized tonic seizures, three; ESs, two) at 9 months to 198 months (median, 49 months) after ACTH therapy. In addition, nine of the 17 patients demonstrated a localized frontal EEG focus after the ACTH therapy, although most of these had previously shown diffuse epileptic EEG abnormality. Conclusions: ACTH therapy is worth trying for patients with resistant ESs, even without features of WS. However, the longterm effect is uncertain because recurrences of various types of seizures, including focal, were frequently observed. Epileptic spasms (ESs) are the primary seizure manifestation not only in patients with West syndrome (WS) but also in patients with other forms of generalized or focal epilepsies (1‐4). The latter groups are subdivided largely into patients characterized by early onset of ESs without hypsarrhythmia, those with late onset of ESs and clinicoelectrical features of Lennox‐Gastaut syndrome, and those with focal epilepsy manifesting both partial seizures and ESs (2‐4). Detailed studies focusing on ESs in the latter groups have been conducted by several investigators, who demonstrated that the clinical and EEG characteristics of the attacks were identical to those of WS, whereas the interictal EEG pattern manifested no typical hypsarrhythmia (5‐8). The prognosis of the seizures has been reported to be poor, because ESs without hypsarrhythmia (ESwoH) have been resistant to medical treatment and also to the epileptic surgery attempted in some patients

The effect of blowing with a hydrogen–oxygen flame on the structure of an OH-free fused quartz tube was studied by microscopic spectroscopy. The number of OH groups increased within 1000 μm from the outside surface (OSS). The peak decomposition of the IR absorption at around 3600 cm−1 showed that most OH structures were ‘free SiOH’ without the hydrogen bond, and H2O molecules were also distributed throughout the cross section. The distribution of the fictive temperature, TF, in the cross section was measured from the peak position of infrared absorption at ≈2200 cm−1. TF before blowing was ≈1420 K, which is near the strain point, except in the region within 200 μm from the OSS, where TF steeply decreases approaching the OSS. After blowing, the values of TF became greater than those before blowing, increasing steeply from the OSS and having a maximum at ≈1000 μm. The steep change near the OSS must be caused by the shorter structural relaxation time due to the SiOH introduced by blowing and H2O vapor in the flame. The distribution of the intensities of Raman bands at 495 (D1) and 606 cm−1 (D2) derived from the planar four- and three-member rings was also measured.

Effects of heat treatment on absorption bands in fused quartz produced by melting natural quartz powder containing 2×1017 cm−3 (sample I) and 1.3×1019 cm−3 of OH (sample II) were studied. Both samples have an absorption peak at ≈5.2 eV. Fluorescent spectra and the results of Gaussian peak decomposition suggest that this absorption is associated with the B2β band with a peak at 5.15 eV and full width at half maximum of 0.42 eV. The 5.15 eV band and an absorption component at 7.5 eV in sample II were annealed out at 1423 K, whereas almost no change was observed in the absorption spectra of sample I. The decrement of the 5.15 and 7.5 eV bands was proportional to the decrement of the OH content. The annealing mechanism in sample II can be explained using a previously proposed model for OH-containing fused quartz wherein the B2β band is the twofold-oxygen-coordinated silicon (=Si:) coordinated with two ≡Si-OH structures. Trimethyltin (TMT) is an organic metal known to induce neuronal degeneration in the hippocampus, and abnormal behavior characterized by seizures, increased aggression and memory deficits. We administered TMT to rats and studied the changes of neuropeptide Y (NPY) and somatostatin (SOM) in the hippocampus. Phenobarbital (PB) was administered as an anticonvulsant to assess the effect of seizures on neuropeptide expressions in both dorsal and ventral hippocampus. Histochemically, NPY-immunoreactivity increased 4 days after TMT treatment in the hilus of the hippocampus, then progressively decreased and dropped to a level below control 16 days after TMT treatment. Detection of NPY mRNA by in situ hybridization preceded the detection of NPY by immunohistochemistry. NPY mRNA signals increased in the hilus 2 days after TMT treatment. SOM-immunoreactivity also increased in the hilus of the hippocampus 2 days after TMT treatment, then decreased rapidly to a normal level. Similar changes in SOM mRNA were demonstrated by in situ hybridization. PB treatment significantly inhibited changes of NPY in terms of both immunoreactivity and mRNA expression; however, the same treatment failed to affect changes in SOM expression. This suggests that NPY and SOM act by different mechanisms in TMT-induced neurodegeneration.

An analytical method to speciate two inorganic As forms [arsenite, As(III) and arsenate, As(V)] in indica and japonica types of rice (both husked and polished) and determine the inorganic As concentration as the sum of these two was internationally validated. The method can additionally determine two organic As compounds, monomethylarsonic acid and dimethylarsinic acid, in rice as separate LC peaks. The method is based on LC separation and inductively coupled plasma (ICP)-MS detection. The method was evaluated through the International Union of Pure and Applied Chemistry/International Organization for Standardization/AOAC harmonized protocol. Sixteen laboratories from four countries participated in the study, and 13 laboratories returned valid data. Twenty test portions of 10 blind duplicates of indica and japonica type rice samples (both husked and polished) were used in this study. Repeatability RSD (RSDr) and reproducibility RSD (RSDR) were calculated at five concentrations of total inorganic As between 0.03 and 0.68 mg/kg. The RSDr was in a range of 3.8 to 7.7% and the RSDR was in a range of 10 to 36%. These performance characteristics were found to be sufficient for determination of inorganic As at or higher than 0.03 mg/kg. Applicability of the method was estimated to be in a range of 0.02–2.0 mg/kg.

SUMMARYThe objective of this study was to examine temporal and regional variations of sporadic Creutzfeldt–Jakob disease (sCJD) in a retrospective study using Japanese national surveillance data from 2001 to 2010. We calculated the incidence of sCJD by age and sex, derived the standardized incidence in each of the 47 prefectures, and performed spatial disease clustering analysis. The average annual incidence of sCJD was 1·026 per million in men (637 patients) and 1·132 per million in women (733 patients), a significant sex difference after adjustment for age (P = 0·001). The ratios of familial CJD to sCJD apparently increased between 2001–2005 and 2006–2010, possibly as a result of the nationwide introduction of genetic testing after 2006. Based on the data of 2006–2010, certain geographical clusters of sCJD were identified. The incidence of sCJD was higher in several specific prefectures compared to the national average. Thus, sCJD appears to have regional variations, suggesting the existence of genetic or region-specific factors affecting the incidence of the disease.

The concentrations of 16 elements in 10 rice flour samples and the distribution of the elements in the rice grains from which the flour were made were determined by ICP-MS and ICP-OES after microwave-assisted digestion of the samples. Arsenic speciation analysis was carried out by HPLC-ICP-MS following heat-assisted extraction of the sample. The concentrations of inorganic As (As(III) and As(V)), monomethylarsonic acid (MMAA) and dimethylarsinic acid (DMAA) and their distribution in the rice grains were determined. Portions of the brown rice were polished/milled to different degrees to yield milled off samples and polished rice samples. All samples were powdered and analyzed for 16 elements and for As species. The recoveries and mass balances for all elements in all samples showed good agreements with the starting materials. As(III), As(V), MMAA and DMAA were detected, and the sums of the concentrations of all species in the extract were 86-105% of the total As concentration in each case.

The effects of polishing, cooking, and storing on total arsenic (As) and As species concentrations in rice were studied adopting typical Japanese conditions. Total and inorganic As levels in three white rice samples polished by removing 10% of bran by weight were reduced to 61-66% and 51-70% of those in brown rice. The As levels in the white rice after three washings with deionized water were reduced to 81-84% and 71-83% of those in raw rice. Rinse-free rice, which requires no washing before cooking because bran remaining on the surface of the rice was removed previously, yielded an effect similar to that of reducing As in rice by washing. Low-volume cooking (water:rice 1.4-2.0:1) rice to dryness did not remove As. The As content of brown rice stored in grain form for one year was stable.

According to our previous study, trimethyltin (TMT), a neurotoxicant, induces the loss of pyramidal neurons in the rat hippocampus, which is preceded by a transient increase in plasma corticosterone concentration. To address whether this transient activation of the hypothalamopituitary-adrenocortical axis is related to neuronal loss in the hippocampus, we evaluated the effects of bilateral adrenalectomy (ADX) and the chronic supplemental treatment of glucocorticoid receptor agonists after ADX on TMT-induced hippocampal damage. Peroral administration of a single dose of TMT (9 mg/kg body wt) induced the extensive loss of CA3 pyramidal neurons and reactive astrocytosis in the hippocampus, as evidenced by results of vimentin and glial fibrillary acidic protein immunohistochemistry, and the effects were profoundly exacerbated by bilateral adrenalectomy. Prolonged administration of corticosterone not only attenuated the exacerbating effects of adrenalectomy but also partially reversed the TMT-induced neuronal loss and reactive astrocytosis. Dexamethasone, but not aldosterone, could be substituted for corticosterone, suggesting a novel neuroprotective action of type II glucocorticoid receptors in the hippocampus.
