Silica-on-silicon is a major optical integration platform, while the emergent class of the integrated laser-written circuits’ platform offers additionally high customizability and flexibility for rapid prototyping. However, the inherent waveguides’ low core/cladding refractive index contrast characteristic, compared to other photonic platforms in silicon or silicon nitride, sets serious limitations for on-chip efficient coupling with single photon emitters, like semiconductor nanowires with quantum dots, limiting the applications in quantum computing. A new light coupling scheme proposed here overcomes this limitation, providing means for light coupling >50%. The scheme is based on the incorporation of an optical microsphere between the nanowire and the waveguide, which is properly optimized and arranged in terms of size, refractive index, and the distance of the microsphere between the nanowire and waveguide. Upon suitable design of the optical arrangement, the photonic nanojet emitted by the illuminated microsphere excites efficiently the guided eigenmodes of the input channel waveguide, thus launching light with high-coupling efficiency. The method is tolerant in displacements, misalignments, and imperfections and is fabricationally feasible by the current state of art techniques. A primary constraint in the major photonic integration platform of Silica-on-Silicon, especially when combined with fabrication approaches like Direct Laser Writing is the optical waveguides' low refractive index contrast, leading thus to limitations for efficient coupling with currently available state-of-the-art single photon emitters such as semiconductor nanowires with quantum dots (NWQD). We propose and demonstrate a novel approach to drastically enhance the light coupling between silica based Laser-written channel waveguides and NWQDs, by incorporating an optical microsphere in their intermediate space. It is demonstrated that the induced photonic nanojet action of a suitably designed microsphere illuminated by the NWQD, excites efficiently the channel waveguide's modes and can enable light coupling to a degree even above 50%. The proposed method is reasonably robust to imperfections and misalignments and could be implemented by current state-of-the-art micro/nano patterning techniques. The paper presents current design issues, limitations, and fabrication challenges towards the development of hybrid integrated optical circuits with embedded single photon sources, which are critical components for future quantum computing, sensing, and communication systems. Optical nanowires with embedded quantum dots (NWQD) are very promising single photon sources, with several reported results of integration in high index platforms like silicon or silicon nitride. Silica on silicon is a favorable platform for optical computing, providing the high degree of customization when combined with direct laser writing techniques for definition of optical structures with refractive index modification. However, the low ~10-3 achievable refractive index contrast imposes strict limitations on the compatibility with NWQD resulting in general in low coupling efficiency as shown in this work. We report on a miniaturized platform for superconducting infrared photon counting detectors. We have implemented a fibre-coupled superconducting nanowire single photon detector in a Stirling/Joule-Thomson platform with a base temperature of 4.2 K. We have verified a cooling power of 4 mW at 4.7 K. We report 20% system detection efficiency at 1310 nm wavelength at a dark count rate of 1 kHz. The detection of a single photon at 1270 nm wavelength allows the direct monitoring of Singlet Oxygen (1O2), making Singlet Oxygen Luminescence Detection (SOLD) a powerful dosimetry technique for photodynamic therapy in the treatment of cancer. However, the direct detection of 1O2 emission at 1270 nm wavelength is extremely challenging as the 1O2 → 3O2 transition in biological media has very low probability and short lifetime due to the high reactivity of singlet oxygen with biomolecules. Recent advances in single photon detection providing high detection efficiency, low noise single-photon detectors are an important innovation in the development of a practical SOLD system for eventual clinical use. In this thesis I present a compact fibre coupled SOLD system, using a supercontinuum pump source to precisely target exact photosensitizer absorption peak wavelengths and single-photon detectors for near-infrared detection by benchmarking a superconducting and a semiconductor photon counting detector. Both pump laser and detector are intrinsically fibre-coupled making them ideally suited for the development of practical singlet oxygen sensor head. The SOLD system was used to carry out a series of singlet oxygen time-resolved measurements in solution and in live cells. These measurements offer information on the photosensitized generation and deactivation of singlet oxygen generated by different photosensitizers and microenvironments at the 1270 nm wavelength and a first investigation of the 1590 nm singlet oxygen luminescence signal is presented.
