Abstract

DenseNets have been shown to be a competitive model among recent convolutional network architectures. These networks utilize Dense Blocks, which are groups of densely connected layers where the output of a hidden layer is fed in as the input of every other layer following it. In this paper, we aim to improve certain aspects of DenseNet, especially when it comes to practicality. We introduce ParaNet, a new architecture that constructs three pipelines which allow for early inference. We additionally introduce a cascading mechanism such that different pipelines are able to share parameters, as well as logit matching between the outputs of the pipelines. We separately evaluate each of the newly introduced mechanisms of ParaNet, then evaluate our proposed architecture on CIFAR-100.

Abstract

G protein–gated inwardly rectifying potassium (GIRK) channels are activated by the phospholipid phosphatidylinositol 4,5 bisphosphate (PIP2). Using functional and computational experiments, Lacin et al. reveal that PIP2 interacts with the tether helix of the neuronal GIRK channel in a dynamic way., G protein–gated inwardly rectifying potassium (GIRK) channels control neuronal excitability in the brain and are implicated in several different neurological diseases. The anionic phospholipid phosphatidylinositol 4,5 bisphosphate (PIP2) is an essential cofactor for GIRK channel gating, but the precise mechanism by which PIP2 opens GIRK channels remains poorly understood. Previous structural studies have revealed several highly conserved, positively charged residues in the “tether helix” (C-linker) that interact with the negatively charged PIP2. However, these crystal structures of neuronal GIRK channels in complex with PIP2 provide only snapshots of PIP2’s interaction with the channel and thus lack details about the gating transitions triggered by PIP2 binding. Here, our functional studies reveal that one of these conserved basic residues in GIRK2, Lys200 (6′K), supports a complex and dynamic interaction with PIP2. When Lys200 is mutated to an uncharged amino acid, it activates the channel by enhancing the interaction with PIP2. Atomistic molecular dynamic simulations of neuronal GIRK2 with the same 6′ substitution reveal an open GIRK2 channel with PIP2 molecules adopting novel positions. This dynamic interaction with PIP2 may explain the intrinsic low open probability of GIRK channels and the mechanism underlying activation by G protein Gβγ subunits and ethanol.

Abstract

A kill assessment system built into a High Energy Laser (HEL) Combat System will provide the U.S. Navy with a method to efficiently engage threats with an HEL effector, improve the weapon scheduling function, and help manage ship’s limited power resources. Near real-time Battle Damage Assessment (BDA) and Dwell Time determinations make up the new kill assessment system, which is simply called the BDA System. This system is a critical force-multiplier for ship survivability by limiting all HEL-target engagements to the minimum dwell time required for threat mitigation, while providing a mission kill interface to the Combat System for a calculated decision point to either re-engage the same threat or engage the next assigned target. This new BDA system concept for a shipboard HEL Combat System was analyzed in order to verify an expected increase in overall system efficiency and performance. The minimum desired increase of threat engagement efficiency was set at 25%. The proof of concept model developed for this project shows that adding a BDA system function to the HEL Combat System causes the system to exceed this threshold of efficiency.
